Description
バキュロウイルスベクターは1)ウイルス遺伝子は130kbpもあり、大きな(<15kbp)外来遺伝子を挿入できる、2)ウイルスの遺伝子は全く哺乳動物細胞では発現しないため、細胞傷害性がほとんどなく有害な免疫応答の誘導もない、3)組換えウイルスを短時間で作製できる、4)ヒトにはバキュロウイルスに対する中和抗体が存在しない等の点で、ヒトへの遺伝子導入ベクターとして、極めて優れた性質を有する。
発明者らは、バキュロウイルスのgp64遺伝子を欠損させ、その代わりに目的のタンパク質をウイルス粒子表面に効率よく取り込ませる方法を開発したことに基づき、本発明に至った。
<1> 少なくとも、細胞表面に発現可能なタンパク質をコードする遺伝子を含むプラスミドと、野生型、変異型及び組換えバキュロウイルスDNAのいずれかとを昆虫細胞にコトランスフェクトするコトランスフェクション工程を含み、かつ、該バキュロウイルスDNAの少なくとも一部を含み、該細胞表面に発現可能なタンパク質を被ったシュードタイプバキュロウイルスが生成され、
該コトランスフェクション工程の後に、細胞表面に発現可能なタンパク質をコードする遺伝子を含むプラスミドが発現する細胞に、前記コトランスフェクション工程により生成された第一のシュードタイプバキュロウイルスを感染させ、ウイルスを増幅させ、第二のシュードタイプバキュロウイルスを生成する増幅工程をさらに含むことを特徴とするバキュロウイルス製造方法である。
<2> バキュロウイルスDNAが、組換えバキュロウイルスDNAであり、多角体遺伝子と第一の外来遺伝子との相同組換えと、gp64タンパク質と第二の外来遺伝子との相同組換えがなされたバキュロウイルスDNAである前記<1>に記載のバキュロウイルスベクター製造方法である。
<3> バキュロウイルスDNAが、組換えバキュロウイルスDNAであり、多角体遺伝子と第一の外来遺伝子との相同組換えがなされたバキュロウイルスDNAである前記<1>に記載のバキュロウイルスベクター製造方法である。
<4> コトランスフェクション工程において、第二の外来遺伝子を含むベクターを同時にコトランスフェクトし、gp64タンパク質と第二の外来遺伝子との相同組換えが起こり、外来遺伝子を含むバキュロウイルスDNAを含み、かつ、該細胞表面に発現可能なタンパク質を被ったシュードタイプバキュロウイルスが生成される前記<3>に記載のバキュロウイルスベクター製造方法である。
<5> 第一の外来遺伝子が導入遺伝子及びマーカー遺伝子の少なくともいずれかである前記<2>から<4>のいずれかに記載のバキュロウイルスベクター製造方法である。
<6> 第二の外来遺伝子が導入遺伝子及びマーカー遺伝子の少なくともいずれかである前記<2>及び<4>のいずれかに記載のバキュロウイルスベクター製造方法である。
<7> 細胞表面に発現可能なタンパク質が、昆虫細胞に感染不可能なタンパク質である前記<1>から<6>のいずれかに記載のバキュロウイルスベクター製造方法である。
<8> 細胞表面に発現可能なタンパク質が、昆虫細胞に感染可能なタンパク質である前記<1>から<6>のいずれかに記載のバキュロウイルスベクター製造方法である。
<9> バキュロウイルスDNAがgp64遺伝子を欠損した組換えバキュロウイルスDNAであり、前記コトランスフェクション工程において、更に、導入遺伝子を有するベクターをコトランスフェクトする前記<1>に記載のバキュロウイルスベクター製造方法である。
本発明者等はこれまでにバキュロウイルス粒子のgp64タンパク質が細胞表面のフォスファチジールイノシトール(PI)を認識することが、バキュロウイルスの哺乳動物細胞への侵入に重要であることを明らかにした(Tani et al., 2001)。PIは各種細胞に広く分布することから、gp64タンパク質を保持する限りバキュロウイルスベクターに細胞特異性を付与することは不可能である(図4)。
また、前述のように、外被タンパク質をウイルスDNAに組換える方法によれば、外被タンパク質が昆虫細胞に感染性を保持することが必須であり、外被タンパク質の種類が限られ、細胞に普遍的な感染性があるようなタンパク質しか選択することができなかった。
これらの組換えウイルスは一回だけgp64タンパク質を介して昆虫細胞に感染できるため、あらかじめ目的のリガンド分子を細胞表面に発現している昆虫細胞にこのウイルスを感染させると、目的のリガンド分子を被ったウイルス粒子が細胞表面から出芽してくることになる(図6及び7)。なお、プラスミドを用いた外被タンパク質の発現により一過性にタンパク質を被らせることは、遺伝子導入効率の点から困難であると考えられていた。
細胞表面に発現可能なタンパク質としては、昆虫細胞に感染不可能なタンパク質であっても良いし、昆虫細胞に感染可能なタンパク質であっても良い。ここでいう昆虫細胞に感染可能なタンパク質とは、バキュロウイルスエンベロープ上に発現させた場合に、昆虫細胞にウイルスの侵入を可能ならしめるタンパク質をいう。このベクターをそのままヒト等へのトランスフェクションに用いる場合には、昆虫細胞への感染性は必要ないが、このベクターを最終ベクター製造の中間体として用いる場合には、昆虫細胞に感染させて増殖させるため、昆虫細胞に感染可能であることが必要である。
昆虫細胞に感染不可能なタンパク質としては、細胞表面に発現可能なタンパク質であれば特に制限はないが、生体内の遺伝子導入したい細胞又は組織に特異的なレセプターや抗原に対応するリガンドやレセプターであることが好ましく、例えば、癌特異抗原や正常細胞とは異なる細胞表面分子を認識できる分子や、エイズウイルス等のウイルス感染細胞に特異的に発現した抗原に対するレセプター分子等が挙げられる。
細胞表面に発現可能なタンパク質をコードする遺伝子を含むプラスミドとしては、公知のプラスミドの中から適宜選択し、所望のタンパク質をコードする遺伝子を挿入することによって、作製することが出来る。例えば、下記の実施例のpIB/gp64、pA3Fb/gp64等が挙げられる。
また、プラスミド中のプロモーターは、gp64プロモーター以外で、昆虫細胞中でよく発現するプロモーターが好ましく、アクチンプロモーターが特に好ましい。
また、バキュロウイルスDNAが、組換えバキュロウイルスDNAであり、多角体遺伝子と第一の外来遺伝子との相同組換えがなされたバキュロウイルスDNAであることが好ましく、この場合には、コトランスフェクション工程において、第二の外来遺伝子を含むベクターを同時にコトランスフェクトし、gp64タンパク質と第二の外来遺伝子との相同組換えが起こるものであることが好ましい。
相同組換えされた外来遺伝子は、ターゲットとなる細胞に導入される目的の遺伝子である導入遺伝子、単離や導入の指標等に用いられるマーカー遺伝子のいずれかであることが好ましく、第一の外来遺伝子が導入遺伝子であることがさらに好ましい。また、相同組換えされた外来遺伝子は、多角体プロモーター等昆虫細胞内でよく発現するプロモーターを有することが好ましいが、宿主に適応する最終ベクターにする場合には、導入遺伝子のプロモーターは宿主で発現するプロモーターを用いる。
CMVプロモーター、RSVプロモーター等が好ましい。
また、導入する外来遺伝子としては、前記プロモーターで発現できるものであれば特に制限はないが、有用性の観点から、各種遺伝疾患に関与する欠損遺伝子や、サイトカイン類、神経栄養因子類、非自己抗原遺伝子、ウイルス抗原等をコードするヌクレオチド配列、癌抑制遺伝子、癌遺伝子であるRas等のアンチセンス配列、又はチミジンキナーゼのような自殺遺伝子等であることが好ましい。
前記バキュロウイルスベクター製造方法は、前記工程の前に、該gp64タンパク質を粒子表面に被ったgp64遺伝子を欠損するバキュロウイルスを作製する工程である、gp64タンパク質を一過性に発現させた昆虫細胞にgp64遺伝子を欠損させたウイルスDNAをトランスフェクトすることにより、一代のみgp64タンパク質を粒子表面に被ったバキュロウイルスを発芽させる工程をさらに含んでいてもよい。
その他、昆虫細胞の取り扱い方、遺伝子組換え、コトランスフェクションの一般的な手法については、周知の昆虫細胞の組換えウイルス作成方法と同様の手法を用いることができる。(松浦善治、タンパク核酸酵素、37,211−222,1992、松浦善治、バキュロウイルス発現系、「遺伝子導入と発現・解析法」横田崇、新井賢一編、洋土社、松浦善治、細胞33(2)30−34,2001)
Sf9細胞(1×107個/10cm ディッシュ)にストックウイルス(通常1×107−8pfu/ml)を0.5〜1.0ml接種し、感染4日後に培養上清を回収し、軽く遠心(6000g,10分間)して細胞片を除いて保存しておく。この上清をSW28ローター(Beckman)で25000rpm,60分間4℃で遠心し、得られたウイルスペレットを1mlのPBSに懸濁する。これを10〜60%のショ糖勾配にのせ、SW41ローター(Beckman)で25000rpm,60分間4℃で遠心しウイルスバンドで回収する。これをPBSに懸濁後、SW41ローターで25000rpm、60分間4℃で遠心し、得られた精製ウイルスのペレットを1mlのPBSに懸濁し4℃で遮光保存する。精製ウイルスの感染価はSf9細胞を用いてプラックアッセイで測定することができる。通常250−500mlの培養上清から0.2×1010pfu/mlの精製ウイルスが得られる。
また、本発明の他のバキュロウイルスベクターは、バキュロウイルスDNAがgp64遺伝子を欠損し、昆虫細胞に感染可能なタンパク質を被ったシュードタイプウイルスであり、該タンパク質が該バキュロウイルスDNAから発現されたタンパク質ではないバキュロウイルスベクターである。前記バキュロウイルスベクターとしては、後述のAcΔ64/GFP/LacZ*64(図11及び図12)のように導入遺伝子を含まずにgp64タンパク質を一過性に被っているものであってもよいし、AcΔ64/GFP/CAGluc*64(図13)のように導入遺伝子が組込まれていてgp64タンパク質を一過性に被っているものであってもよいし、また導入遺伝子が組込まれていてgp64タンパク質以外の細胞表面に発現可能なタンパク質であって、昆虫細胞に感染可能なタンパク質を一過性に被っているものであってもよい。
また、前記昆虫細胞に感染不可能なタンパク質を被ったシュードタイプバキュロウイルスベクターは、ヒトを含む生体内及び生体外のいずれかの細胞に遺伝子を導入するための最終的なベクターとして用いることができる。
前記バキュロウイルスベクターは、生体に投与する場合には、経口的又は非経口的(例えば、静脈内、筋肉内、腹腔内、皮下又は皮内等への注射、直腸内投与、経粘膜投与、経気道投与、臓器内投与など)に投与することができる。
投与量、投与回数は、投与対象の体重や導入遺伝子に併せて、適宜調節することが出来る。
生体への遺伝子発現は補体による不活化が障害となる場合があるため、補体系のタンパク分解阻害剤のフサンを同時に適用することにより不活化を回避できる。
本発明の遺伝子導入方法は、前記本発明のバキュロウイルスベクターを用いて、生体内(ヒトを除く)及び生体外のいずれかの細胞に遺伝子を導入することを特徴とする。
以下に本発明のバキュロウイルスベクター、及びその製造方法並びにこれを用いた遺伝子導入方法について、実施例より更に詳しく説明するが、本発明はこれに限定されるものではない。
(実施例1)
gp64遺伝子ノックアウトベクターの作製
バキュロウイルスのgp64遺伝子領域を多角体プロモーターと緑色蛍光タンパク質(GFP)遺伝子とで置換することによってその発現を消失させるため、ノックアウトベクターpUCΔ64/GFPを図8に示す手順で作製した。
バキュロウイルス(AcNPV)のgp64遺伝子領域を含むEcoRI-SmaI断片(nt 107,325-112,049)を、pUC18の同様の制限酵素サイトに組み込み、pUCgp64locusを作製した。次に多角体プロモーターの下流にGFP遺伝子及び宮崎らによって開発されたCAGプロモーター (Niwa et al., 1986) の下流にルシフェラーゼ遺伝子を組み込んだトランスファーベクター pAcGFP-CAGluc(Tani et al., 2001)をEcoRVとSnaBIで切断して、多角体プロモーターとGFP遺伝子を含む断片を回収し、pUCgp64locusをSpeI(nt 109,761)とBglII(nt 108,039)で切断後klenow fragmentで平滑末端化した部位に組み込んで、gp64遺伝子のノックアウトベクター、pUCΔ64/GFPを作製した。
gp64遺伝子を欠損させたバキュロウイルスAcΔ64/GFP/LacZの作製(図9)
多角体プロモーターの下流にβ-ガラクトシダーゼ(LacZ)遺伝子を組み込んだ組換えバキュロウイルス(AcLacZ)の感染性ウイルスDNAを抽出し、gp64遺伝子のノックアウトベクター、pUCΔ64/GFPと共に、Sf-9細胞にコトランスフェクトすると、昆虫細胞の核内で、ウイルスDNAとpUCΔ64/GFPが相同組換えを起こし、ウイルスDNAのgp64遺伝子領域が多角体プロモーターとGFP遺伝子に置換された組換えウイルスAcΔ64/GFP/LacZが生じる。このウイルスはgp64遺伝子とそのプロモーター領域が欠損しており、エンベロープタンパク質を完全に欠損するため、感染性を全く示さない。そこで、一過性にgp64タンパク質をAcΔ64/GFP/LacZに被せて、一回だけ感染できるように改変する。
gp64タンパク質を一過性に昆虫細胞で発現できるプラスミドの作製(図10)
昆虫由来のSf-9細胞内でgp64タンパク質を発現できるプラスミドを作製するため、pUCgp64locusからgp64遺伝子をSpeIとBglIIで切りだし、その断片を同じ制限酵素で切断したpUC18に組み込み、pUCgp64を作製した。さらにpUCgp64からgp64遺伝子を含む断片をHindIIIとEcoRIで切り出し、同じ酵素で切断したpIB/V5-His(Invitrogen)に組み込み、発現ベクターpIB/gp64を作製した。pIB/V5-Hisのバキュロウイルス初期遺伝子プロモーター(IEプロモーター)の下流に挿入された遺伝子は昆虫細胞で効率よく発現させることが可能である。また、昆虫細胞で高率に転写されるアクチンプロモーターを利用した同様の発現ベクターを作製した。まず、pUCgp64locusを鋳型としたPCR法によって、gp64遺伝子を増幅した。primerには、gp64-Fw(Bgl):AAAGATCTACCatggtaagcgctattgttt(配列番号1)と、gp64-Rv(Sal):TTGTCGACttaatattgtctattacggttt(配列番号2)を用いた。増幅させたgp64遺伝子をBglIIとSalIで切断し、pA3FbのBamHIとSalIサイトに組み込み、pA3Fb/gp64を作製した。
gp64タンパク質を一過的に被ったgp64遺伝子欠損ウイルスAcΔ64/GFP/LacZ*64の作製(図11)
上記のAcLacZの感染性ウイルスDNA、pUCΔ64/GFP、及び、pIB/gp64あるいはpA3Fb/gp64を共に、Sf-9細胞にコトランスフェクトする。これにより昆虫細胞の核内で、ウイルスDNAとpUCΔ64/GFPが相同組換えを起こし、ウイルスDNAのgp64遺伝子領域が多角体プロモーターとGFP遺伝子に置換された組換えウイルスAcΔ64/GFP/LacZが生じる。このウイルスはgp64遺伝子とそのプロモーター領域を欠損するため、通常の昆虫細胞では増殖できながコトランスフェクトしたpIB/gp64あるいはpA3Fb/gp64によりトランスにgp64タンパク質が細胞表面に供給されるため、gp64遺伝子を欠損しているにもかかわらず、gp64タンパク質を被って感染性を保持して出芽できる。さらに組換えウイルスはGFPの発現を指標に、親のAcLacZと識別可能である。即ち組換えウイルスはLacZとGFPの両方を発現する。実際のウイルス分離はトランスフェクト後4日目の培養上清を、前日にpIB/gp64あるいはpA3Fb/gp64をトランスフェクトしたSf-9細胞を用いて、通常のプラックアッセイ法で相同組換えを起こしたと思われるGFPとLacZの両方が陽性のプラックを回収し、プラックアッセイを4回繰り返して純化して、gp64遺伝子領域を多角体プロモーターとGFP遺伝子に置換し、一過的にgp64タンパク質を被って感染性を保持したAcΔ64/GFP/LacZ*64を得た。
AcΔ64/GFP/LacZ*64の増幅(図12)
上記で単離したAcΔ64/GFP/LacZ*6は、一過性にgp64タンパク質を被っているが、gp64遺伝子を欠失していることから、通常のSf-9細胞には一回しか感染できず増幅はできない。しかしながら、予めpIB/gp64あるいはpA3Fb/gp64をトランスフェクトしてgp64タンパク質を細胞表面に発現させたSf-9細胞に感染させることによって、高力価のストックウイルスを容易に調整することが可能である。
各種リポーター遺伝子を発現する組換えウイルスの作製
遺伝子導入する標的哺乳動物細胞内で導入遺伝子を発現させるため、CAGプロモーターの下流に3種のレポーター遺伝子、ホタルのルシフェラーゼ遺伝子、赤色蛍光タンパク質(DsRed)遺伝子、GFP遺伝子をそれぞれ組み込んだトランスファーベクター、pAcCAGluc、pAcCAGDsRed、及びpAcCAGGFPを用いた。これらのベクターは多角体遺伝子領域で相同組換えを起こすように設計されている。pAcCAGlucの作製は既に報告している(Tani et al., 2001)。pDsRed2-N1(Clontech)をKpnIで切断し、DsRed遺伝子を回収し、同じ制限酵素で切断したpAcCAGMCS2(Shoji et al., 1997)に組み込み、pAcCAGDsRedを作製した。pIRES2-EGFP(Clontech)をNcoIで切断し、GFPを含む断片をklenow fragmentで平滑化後、SmaIとNotIで切断し、klenow fragmentで平滑末端化したpAcCAGMCS2に組み込んでpAcCAGGFPを作製した。
図13にpAcCAGlucを用いた組換えウイルスの作製手順を示す。AcΔ64/GFP/LacZ*64からDNAを抽出し、LacZ遺伝子内に存在するユニークなBsu36Iサイトで切断してウイルスDNAを直鎖状にした。この直鎖ウイルスDNA、pAcCAGluc、及び、pIB/gp64あるいはpA3Fb/gp64を共にSf-9細胞にコトランスフェクトして、同様の手法でGFP陽性でLacZ陰性のプラークを回収した。プラックアッセイを4回繰り返して純化して、ルシフェラーゼ遺伝子を持つ組換えバキュロウイルスAcΔ64/GFP/CAGluc*64を作製した。このウイルスを予めpIB/gp64あるいはpA3Fb/gp64をトランスフェクトしてgp64タンパク質を発現しているSf-9細胞に感染させることによって、高力価のストックウイルスを調整した。
水疱性口内炎ウイルスのGタンパク質を被ったシュードタイプウイルスの作製(図14)
導入遺伝子の一例として、水疱性口内炎ウイルスのGタンパク質遺伝子について検証した。pIB/V5-His及びpA3Fbに水疱性口内炎ウイルスのGタンパク質遺伝子を挿入したpIB/VSVG及びpA3Fb/VSVGを作製した。これらのいずれかのプラスミドをSf-9細胞にトランスフェクトし、24時間後にAcΔ64/GFP/CAGluc*64を感染させることによって、gp64タンパク質の代わりに水疱性口内炎ウイルスのGタンパク質を被ったシュードタイプウイルスAcΔ64/GFP/CAGluc*VSVGを作製した。これらのウイルスがgp64タンパク質を欠損し、水疱性口内炎ウイルスのGタンパク質を保持していることを、精製ウイルスの抗gp64抗体及び抗水疱性口内炎ウイルス抗体を用いた免疫ブロットで確認した(図15)。また、前記水疱性口内炎ウイルスのGタンパク質を一過性に被ったシュードタイプウイルスAcΔ64/GFP/CAGluc*VSVGは、前記gp64を一過性に被ったシュードタイプウイルスAcΔ64/GFP/CAGluc*64同様各種哺乳動物細胞に効率よく感染することが分かった。図16は、AcΔ64/GFP/CAGluc*64とAcΔ64/GFP/CAGluc*VSVGとを各々293Tcell(ATCC社製)へ細胞あたりの感染価(moi)10、30及び50で接種した結果を表す。培養上清を除去してウイルス接種し、60分間吸着した後、培地を加え37℃で培養した。感染後24から48時間で細胞を回収し、luciferase遺伝子の発現をBright-Glo luciferase Assay system(Promega社製)により測定した数値を表す。
このことから、本発明のバキュロウイルス製造方法によれは、水疱性口内炎ウイルスのGタンパク質の昆虫細胞への感染性を全く用いることなく、該タンパク質を被ったバキュロウイルスベクターを増殖して得ることができることが分かった。したがって、所望のエンベロープのタンパク質を、その昆虫細胞への感染の有無にかかわらず、自由に被せることができることが実証された。また、この方法によれば、遺伝子導入される細胞に感染した後には、再度そのエンベロープタンパク質が発現されることはなく、安全性の高いバキュロウイルスベクターを作成することができる。
二つの蛋白質を粒子表面に発現するバキュロウイルス:麻疹ウイルスの宿主特異性を規定する二つのエンベロープ蛋白質を持つバキュロウイルスの作製
麻疹は強い感染性を示す急性ウイルス感染症で、発展途上国を中心に毎年100万人の命を奪っている。麻疹ウイルスは粒子表面にHとFの二つのエンベロープ蛋白質を持ち、H蛋白質が感染の組織特異性を規定している。即ち、実験室で継代された麻疹ウイルス株(Edmonston株)ではCD46あるいはSLAM(Signaling lymphocyte activating molecule; CDw150)をリセプターとして利用できるのに対し、野外からの新鮮分離株はSLAMのみをリセプターとすることが明らかにされている。そこで、Edmonston株あるいは新鮮分離株由来のHとF蛋白質をそれぞれ粒子表面に発現した組換えバキュロウイルスを作製し、麻疹ウイルスの感染特異性をバキュロウイルスで再現できるか否かを検討した。
実施例7のpA3Fb/VSVGの替わりに、Edmonston株あるいは新鮮分離株由来の麻疹ウイルスのHおよびF遺伝子を組み込んだプラスミドを、AcΔ64/GFP/CAGlucとコトランスフェクトし、AcΔ64/GFP/CAGluc*MV-H/Fを作製した。次に、これらのバキュロウイルスの感染特異性をSLAMあるいはCD46を発現しているハムスター由来細胞株(CH0細胞)で検定した。その結果、Edmonston株由来のHおよびF蛋白質を被ったバキュロウイルスベクターはSLAMを発現するCHO細胞及びCD46を発現するCHO細胞への感染が確認された。一方、新鮮分離株由来のHおよびF蛋白質を被ったバキュロウイルスはSLAMを発現するCHO細胞にのみに遺伝子導入が可能であった。陽性対照ウイルスのAcΔ64/GFP/CAGluc*VSVGはいずれのCHO細胞株にも感染し、陰性対照ウイルスのgp64蛋白質を欠損したAcΔ64/GFP/CAGlucは全く感染性を示さなかった。
この成績は、バキュロウイルスの粒子表面に二つのウイルス蛋白質を生物活性を保持した形で提示できることを示すものである。二つの蛋白質を粒子表面に提示することで、より精度の高いターゲッティングが可能となると思われる。また、ウイルスのエンベロープ蛋白質だけでなく、逆にウイルスのリセプター分子や癌抗原に対する単鎖抗体を粒子表面に提示すれば、ウイルスに感染してエンベロープ蛋白質を発現している細胞や癌細胞だけにチミジンキナーゼ等の自殺遺伝子を導入し、プロドラッグとの併用によって目的の細胞だけを生体から排除することが可能となる。特に、エイズウイルスは慢性持続感染し、感染細胞表面にエンベロープ蛋白質を高度に発現するので、エイズウイルスのリセプターとコリセプターを提示させた組換えバキュロウイルスにより、生体から感染細胞だけを排除することが期待される。
Claims (9)
- 少なくとも、細胞表面に発現可能なタンパク質をコードする遺伝子を含むプラスミドと、野生型、変異型及び組換えバキュロウイルスDNAのいずれかとを昆虫細胞にコトランスフェクトするコトランスフェクション工程を含み、かつ、該バキュロウイルスDNAの少なくとも一部を含み、該細胞表面に発現可能なタンパク質を被ったシュードタイプバキュロウイルスが生成され、
該コトランスフェクション工程の後に、細胞表面に発現可能なタンパク質をコードする遺伝子を含むプラスミドが導入され、かつ、前記プラスミドにより前記細胞表面に発現可能なタンパク質が発現する細胞に、前記コトランスフェクション工程により生成された第一のシュードタイプバキュロウイルスを感染させ、ウイルスを増幅させ、第二のシュードタイプバキュロウイルスを生成する増幅工程をさらに含むことを特徴とするバキュロウイルス製造方法。 - バキュロウイルスDNAが、組換えバキュロウイルスDNAであり、多角体遺伝子と第一の外来遺伝子との相同組換えと、gp64タンパク質と第二の外来遺伝子との相同組換えがなされたバキュロウイルスDNAである請求項1に記載のバキュロウイルスベクター製造方法。
- バキュロウイルスDNAが、組換えバキュロウイルスDNAであり、多角体遺伝子と第一の外来遺伝子との相同組換えがなされたバキュロウイルスDNAである請求項1に記載のバキュロウイルスベクター製造方法。
- コトランスフェクション工程において、第二の外来遺伝子を含むベクターを同時にコトランスフェクトし、gp64タンパク質と第二の外来遺伝子との相同組換えが起こり、外来遺伝子を含むバキュロウイルスDNAを含み、かつ、該細胞表面に発現可能なタンパク質を被ったシュードタイプバキュロウイルスが生成される請求項3に記載のバキュロウイルスベクター製造方法。
- 第一の外来遺伝子が導入遺伝子及びマーカー遺伝子の少なくともいずれかである請求項2から4のいずれかに記載のバキュロウイルスベクター製造方法。
- 第二の外来遺伝子が導入遺伝子及びマーカー遺伝子の少なくともいずれかである請求項2から4のいずれかに記載のバキュロウイルスベクター製造方法。
- 細胞表面に発現可能なタンパク質が、昆虫細胞に感染不可能なタンパク質である請求項1から6のいずれかに記載のバキュロウイルスベクター製造方法。
- 細胞表面に発現可能なタンパク質が、昆虫細胞に感染可能なタンパク質である請求項1から6のいずれかに記載のバキュロウイルスベクター製造方法。
- バキュロウイルスDNAがgp64遺伝子を欠損した組換えバキュロウイルスDNAであり、前記コトランスフェクション工程において、更に、導入遺伝子を有するベクターをコトランスフェクトする請求項1に記載のバキュロウイルスベクター製造方法。
