Description
The present invention relates to a filter that can be used for separating a biopolymer sample such as DNA according to size, a manufacturing method thereof, and a microchip using the filter .
DNA size separation is a widely used technology such as DNA diagnosis, DNA typing, and DNA testing by determining DNA sequences and detecting SNPs (single nucleotide polymorphisms). Therefore, easy handling and high speed are important issues.
For size separation of DNA, gel electrophoresis using gels and polymers such as agarose and polyacrylamide that has been conventionally electrophoresed and the movement speed is limited by the network structure depends on the size of the DNA. Electrophoresis was mainstream.
However, it is generally necessary to run for a long time, change the composition of the gel or polymer according to the size to be separated, or because tremendous time is required to separate large DNA, There was a problem that it was difficult to use.
Conventionally, nanopillars have been produced in quartz glass or resin materials by X-ray lithography, Deep-RIE, nanostamping, or the like. However, since the nanopillars obtained by conventional nanostamping are made of resin, there are problems with sterilization treatment at high temperatures, and in the case of nanopillars made of quartz, such as Deep-RIE, the organic solvent resistance is low. Also, there are problems such as causing deterioration with respect to alkalis and acids (hydrofluoric acid) (see Non-Patent Document 2).
An object of the present invention is to provide a highly resistant nanopillar, a method for producing the nanopillar, and a chip using such a nanopillar.
Filter of the present invention is a filter comprising nanopillar containing random pillar structure in which a plurality of shapes are mixed with metal.
The metal is a metal that self-assembles, and an example thereof is gold or platinum, which has heat resistance, sterilization, organic solvent resistance, acid resistance, alkali resistance, or chemical resistance.
Here, “self-organization” refers to the property of order formation in a dissipative system.
The microchip of the present invention is characterized in that a channel is formed inside a substrate, and the filter according to the present invention is provided by blocking at least a part of the channel.
The filter manufacturing method of the present invention is a method of forming a random nanopillar structure on a substrate by vapor-depositing a metal thin film that self-assembles on the substrate and then heat-treating the substrate to self-assemble the metal. .
There is a difference that conventional nanopillars are arranged in a uniform shape, whereas the nanopillars of the present invention are random pillars. Therefore, it becomes a thing suitable for uses, such as filtration (filtration).
In the conventional method, nano pillars are formed on a resin material by nano stamping or the like. However, when DNA is extracted, the resin material chip cannot withstand 200-degree sterilization and may be deformed.
In addition, in the nanopillar chip using quartz, there is a possibility that the separation function may be impaired due to degradation of the nanopillar when it is exposed to a solvent such as an acid (hydrofluoric acid) or an alkali, There was a problem with chemical resistance.
On the other hand, since metal is used as the material of the nanopillar in the present invention, it has higher temperature (200 degrees) sterilization treatment and higher organic solvent resistance, acid resistance, alkali resistance, chemical resistance, and heat resistance compared to conventional products. Very effective in terms of analysis and durability.
Particularly when gold or platinum is used as the metal, the chemical resistance is particularly excellent.
Such a microchip can be used as a biopolymer separator.
According to the production method of the present invention, a random nanopillar structure can be easily formed by simply depositing a metal thin film on a substrate and then performing a heat treatment.
An embodiment of the nanopillar according to the present invention will be described together with its manufacturing method.
As shown in FIG. 1A, in the microchip for forming the nanopillar, a microchannel (depth 150 nm) is formed on the lower quartz substrate 10 serving as a base, for example, by lithography and etching.
Gold is deposited in the flow path of the substrate 10 to a thickness of 20 nm. At this time, gold is vapor-deposited only on the flow path portion 20 by using a mask such as a metal mask.
Next, the substrate 10 is heat-treated at 600 ° C. for 1 hour under a nitrogen atmosphere to form a random nanopillar structure in the flow path of the substrate 10 by gold self-organization.
Random nanopillars as shown in the enlarged view (B) of the flow path 20 were formed on the substrate 10.
A cover glass 16 on which the sample inlet 12 and the outlet 14 were formed was closely bonded to the flow path forming surface of the substrate 10 by hydrofluoric acid bonding to obtain a microchip substrate.
The sample inlet 12 and outlet 14 to the cover glass 16 were formed by a sandblast method or the like.
The production conditions of the present invention are not limited to the contents described above. For example, even when the thickness of the gold thin film is 10 nm and the heat treatment temperature is 400 ° C., nanopillars are formed by gold self-assembly. However, if the deposited film is too thick, nanopillars are not formed. For example, when the thickness of the gold thin film is 85 nm, nanopillars are not formed even when the heat treatment temperature is 600 ° C.
As the formation conditions of the nanopillar, the thickness of the gold thin film is preferably 50 nm or less, and the heat treatment temperature tends to require a higher temperature as the thickness of the gold thin film increases.

The pillars in the figure have a random nanopillar structure with a width of 300 to 500 nm, a height of 20 to 200 nm, and irregular shapes and sizes.
FIG. 3 shows AFM data of the surface shape after depositing a gold thin film to a thickness of 20 nm and then heat-treating at 600 ° C. for 1 hour. The scale of FIG. 8 is displayed such that the vertical and horizontal dimensions are 0 to 1.25 μm and the height is 0 to 10 nm.

9 to 11, it can be seen that the height of the formed metal is 0 to 3 nm, no pillar is formed, and the surface is close to a flat surface.
Since gold pillars are formed in the flow path of the microchip substrate of the present invention, the microchip substrate has properties such as heat resistance, sterilization, organic solvent resistance, acid resistance, and alkali resistance. For this reason, there is no problem with the sterilization effect of the microchip due to the high temperature.
Utilizing this property, when a microchip substrate is incorporated into a biopolymer device and used as a filter in a flow path, it can be applied to μTAS (Micro Total Analysis Systems), electrophoresis chips, etc. For example, DNA or protein It is possible to provide a device capable of performing separation and the like.
In the description of the examples, gold vapor deposition was performed when forming metal nanopillars, but even when the same operation was performed with platinum, random nanopillars self-assembled on the substrate, and the same results as when performed with gold were obtained. Obtained.
