【0005】

(See Kaihei 9-59254).
[0002]
[Prior art]
As a method for producing 2-sulfonylpyridines from sulfonylcyanide, (1) a method of reacting sulfonylcyanide and 1-acyloxy-1,3-butadiene [JP-A-9-118666, JP-A-10-87624] No. 1, JP-A-10-87626 and JP-A-10-139760], (2) a method of reacting sulfonylcyanide and 1-ethoxy-2-methyl-1,3-butadiene [Synthesis] 623 (1989)], (3) Method of reacting sulfonylcyanide with 1,3-butadiene [Recueil des Travaux Chimiques des Pays-Bas, Vol. 12, 92 No., page 1343 (1973)], (4) sulfonylcyanide and tetraphenyl A method of cycloaddition of pentadienone and elimination of carbon monoxide from the obtained cyclized adduct [Tetrahedron Letters, No. 12, 971 (1970)], (5) sulfonylcyanide A method of reacting an α, β-unsaturated carbonyl compound [see JP-A-11-269147] is known.
[0003]
[Problems to be solved by the invention]
In the above methods (1) and (2), it is essential to use 1,3-butadienes having an oxygen functional group, which are thermally unstable and difficult to handle. In the method (3), the reaction requires about In addition to requiring a long period of 3 days, the yield of 2-sulfonylpyridines is as low as about 20%. In the method (4), a high temperature of 175 ° C. is required for the cycloaddition reaction, and it is not easy to obtain tetraphenylcyclopentadienone as a substrate. In method (5), an α, β-unsaturated carbonyl compound that is relatively easily available is used, and an α, β-unsaturated carbonyl compound having a substituent at the 1-position or 4-position thereof is used as a substrate. In some cases, the corresponding 2-sulfonylpyridines cannot be obtained in satisfactory yields.
[0004]
An object of the present invention is to provide a method by which 2-sulfonylpyridines can be easily produced in good yield and industrially advantageously.

The reaction of acetal (II) and sulfonylcyanide (III) in the present invention is preferably carried out in the presence of a solvent. The solvent is not particularly limited as long as it does not adversely affect the reaction. For example, aliphatic hydrocarbons such as hexane, heptane, and octane; benzene, toluene, xylene, ethylbenzene, mesitylene, chlorobenzene, trifluoromethylbenzene, dichlorobenzene, etc. Aromatic hydrocarbons; Tetrahydrofuran, diisopropyl ether, tert-butyl methyl ether, 1,2-dimethoxyethane, 1,4-dioxane, diglyme and other ethers; Acetonitrile, benzonitrile and other nitriles; Ethyl acetate, butyl acetate and other esters Halogenation of chloroform, carbon tetrachloride, 1,2-dichloroethane, etc. Aliphatic Hydrocarbon; dimethylformamide, dimethyl sulfoxide and the like. Of these, aromatic hydrocarbons such as benzene, toluene, xylene, ethylbenzene, mesitylene, chlorobenzene, trifluoromethylbenzene, and dichlorobenzene are preferable, and toluene is particularly preferable. A solvent can be used individually or in combination of 2 or more types.
[0040]
The amount of acetal (II) used is preferably in the range of 0.1 to 10 equivalents, more preferably in the range of 0.5 to 3 equivalents, relative to the sulfonylcyanide (III).
[0041]
The reaction temperature is preferably in the range of 50 to 200 ° C, more preferably in the range of 80 to 150 ° C. The reaction can be performed under normal pressure, increased pressure, or reduced pressure.

The concentration of acetal (II) to be dropped is not particularly limited, but it is preferably diluted with a solvent used for the reaction. When the reaction is carried out while continuously removing the generated alcohol out of the reaction system, it is preferable to successively replenish the solvent lost from the reaction system with the operation of removing the alcohol.
[0044]

[0045]
Both acetal (II) and sulfonylcyanide (III) used as raw materials can be easily produced by known methods. Acetal (II) can be produced, for example, by reacting the corresponding α, β-unsaturated carbonyl compound with orthoesters in the presence of an acid catalyst [Organic Synthesis Collective Volume, 4, 21 (1963)], and can also be produced by condensing acetal of saturated aldehyde and vinyl ether in the presence of an acid catalyst [Synthesis, 137 (1981)]. The sulfonylcyanide (III) can be produced, for example, by reacting a corresponding metal alkylsulfinate or arylsulfinate metal salt with a cyanide halide [Organic Synthesis Collective Volume, Volume 6 727 (1988)].

After completion of the dropping, heating for 4 hours and distillation of the liquid containing methanol were continued, and finally 1.97 g of liquid containing methanol was distilled from the top of the Vigreux column. The reaction mixture was concentrated and purified by silica gel chromatography to obtain 2-benzenesulfonyl-5-bromo-3-methylpyridine having the following physical properties as a light brown waxy solid (2.54 g, benzenesulfonyl cyanide). Based on the yield of 78%).

Comparative Example 2
Synthesis of 2-benzenesulfonyl-4,6-dimethylpyridine (comparative example to Example 3)
9.80 g (100 mmol) of mesityl oxide (4-methyl-3-penten-2-one) was added to a three-necked flask with an internal volume of 50 ml equipped with a thermometer, a magnetic stirrer, a Dean-Stark moisture meter and a condenser. ) And 8.35 g (50.0 mmol) of benzenesulfonylcyanide, 15 ml of toluene and 1.5 ml of butanol were added as solvents, followed by 1.15 g (5.00 mmol) of tributyl borate, and then in a nitrogen atmosphere. The mixture was stirred at an internal temperature of 116 ° C., and heated and refluxed for 10 hours while separating and removing generated water. After cooling this solution to room temperature, low boiling components such as a solvent were concentrated by a rotary evaporator, and cooled in an ice bath to precipitate crystals. The crystals were filtered through a glass filter, washed with 10 ml of toluene cooled to 5 ° C. or lower, and then vacuum-dried with a vacuum pump for 2 hours. As colorless crystals, 2-benzenesulfonyl-4,6-dimethyl having the following physical properties was obtained. 6.14 g of pyridine was obtained (purity 99%, yield 49% based on benzenesulfonylcyanide).
