本発明は、ステージ上に載置される標本を加温するための加温プレートを設けた顕微鏡に関するものである。

例えば、生物組織や卵細胞などの標本を顕微鏡により観察するような場合、倒立型顕微鏡とマイクロマニピュレータなどを用いて卵巣などから細胞や卵の切り出しなど細かな操作を行い、さらに、細かな顕微鏡観察に邪魔な、種々の別の細胞、血液、粘膜などを取り除いた後、実体顕微鏡を使用して、細胞や卵の変化の過程を観察することなどが行われている。

この場合、顕微鏡観察に用いられる実体顕微鏡は、1〜4倍程度の倍率の低いもので、このため観察範囲を大きく取れることから、標本を載置するステージは固定のものを用い、この固定ステージ上で必要に応じて標本の位置を手で移動させることにより所望する観察部位を決定している。

ところで、顕微鏡観察される細胞や卵などの標本は、ステージ上でも細胞の活性を落とすことなく、本来の姿に近い状態で観察することが必要であり、このため、これら標本を保温することが重要である。

そこで、従来、このような目的のために、例えば、特願昭61−177413号公報に開示されるような顕微鏡の水平方向に移動可能なステージの中座部分に、中心部に観察用の開口を形成した円形の中座ヒータを取り付けたものや、特開昭62−135803号公報に開示されるように透明なガラス板の間に透明な導電膜を設けてなるヒータステージを加温可能にしたものなどが提案されており、これらの加温手段を実体顕微鏡の固定ステージ上に組み込むことが考えられる。

ところで、最近、実体顕微鏡は、高性能、高倍率になっており、10倍程度から20倍程度の倍率での観察も可能になっている。そして、このような高性能、高倍率の実体顕微鏡が上述した細胞観察にも使用されるようになっている。また、このような性能の向上とともに、実体顕微鏡上での作業も高度な内容のものが可能となり、高倍での細胞観察も考えられている。

【発明が解決しようとする課題】
ところが、これまでの実体顕微鏡のように、固定ステージ上に加温手段を組み込み、固定ステージ上で標本位置を手で移動させる方法では、標本を微小かつスムーズに移動させることが困難になるため、高倍の実体顕微鏡の使用により観察範囲が小さくなると、標本移動により、所望する部位が顕微鏡の観察範囲からはみだしてしまうことがあるなど安定した作業ができない。特に、生物組織や卵細胞などの標本は、シャーレなどの培養容器内の培養液の中に浸されており、卵細胞については、培養溶液中に浮遊した状態で観察されることが多いが、手操作では、培養容器の移動をスムーズに行うことが難しく、培養溶液中に浮遊している卵細胞が揺れて観察に支障をきたすという問題があった。

本発明は、上記事情に鑑みてなされたもので、ステージ上の標本を最適に加温できるとともに、標本を精度よく移動させることができる顕微鏡を提供することを目的とする。

【課題を解決するための手段】
請求項1記載の本発明は、少なくとも固定ステージを有する顕微鏡において、前記固定ステージ上に移動可能に載置されるとともに、標本が載置される発熱体を有する透明加温プレートと、前記透明加温プレートを前記固定ステージ面上に沿って移動させる加温プレート駆動手段と、前記透明加温プレートと前記加温プレート駆動手段の駆動部を接続する接続手段を具備し、前記接続手段は、中間に段部を有する板ばねからなり、前記板ばねは透明加温プレートの移動平面と垂直方向に板厚方向を向け、高所に位置する端部を前記加温プレート駆動手段の上面に、低所に位置する端部を前記透明加温プレートの上面にそれぞれ固定しており、それによって、前記加温プレート駆動手段に対する前記透明加温プレートの剛性を、加温プレート面に対して水平な方向には高くし、垂直な方向には弱くして自由度を強くできている。

以下、本発明の一実施の形態を図面に従い説明する。
図1、図2は、本発明が適用される実体顕微鏡の概略構成を示し、図1は、側面図、図2は、上面図を示している。

図において、1は顕微鏡基台で、この顕微鏡基台1上に支柱2を直立して設け、この支柱2先端に実体顕微鏡本体3を設けている。
実体顕微鏡本体3は、顕微鏡基台1上の後述する固定ステージ5に対向して観察光軸上に配置される対物レンズ31と、この対物レンズ31より得られた標本像を観察する接眼部32を設けている。また、実体顕微鏡本体3は、焦準ハンドル4により上下方向に移動可能になっていて、対物レンズ31のピント調整を可能にしている。

顕微鏡基台1は、対物レンズ31と対向する上面を固定ステージ5に形成している。この固定ステージ5は、そのほぼ中央部に透過照明用孔部51を形成している。そして、固定ステージ5上に、透明な加温プレート6を載置している。

この加温プレート6は、図3(a)(b)に示すように樹脂で形成された矩形枠状の加温プレート筐体61と、この加温プレート筐体61により周縁部を保持される透明加温部62から構成している。この場合、透明加温部62は、第1の透明板621と第2の透明板622を一対のスペーサ623を介して積層し、これら第1の透明板621と第2の透明板622の間に絶縁性透明材料624を設けている。第1の透明板621の内側面に、透明発熱体625を設けている。この透明発熱体625は、相対向する一方の側縁に向かい合うように一対の発熱体電極626a、626bを設けたものである。また、この透明発熱体625と絶縁性透明材料624との間に温度センサ627を設けている。この温度センサ627は、透明発熱体625の発熱体電極626a、626bでの発熱による温度を加温プレート6の温度として検出するものである。そして、これら発熱体電極626a、626bおよび温度センサ627は、ケーブル628により図示しない加温コントローラに接続している。

また、加温プレート6は、図4に示すように加温プレート筐体61の底面に沿って摩擦抵抗の小さい、例えば軸受けに用いられる樹脂などからなる板状の摺動部材7を複数個設け、固定ステージ5上での加温プレート6の摺動性を高めるようにしている。

この場合、摺動部材7は、図示例では矩形枠状からなる加温プレート筐体61底面の4か所の角部と、相対向する2組の側枠のうち一方組の側片の中間部の2か所の合計6か所に配置しているが、さらに残りの組の側枠の中間部の2か所にも配置して、合計8か所としてもよい。勿論、摺動部材7は、矩形枠状の加温プレート筐体61底面の4か所の角部のみのに設けてもよいし、底面の全体に設けるようにしてもよい。さらに、摺動部材7の形状についても、板状に限らずL形状、三角形などが考えられる。さらに、摺動部材7は、後述する十字動装置8による加温プレート6のXY移動の際に、固定ステージ5上から脱落するのを防止するため、十字動装置8による加温プレート6の移動方向と距離を考慮して長さ寸法を設定している。

一方、顕微鏡基台1上には、固定ステージ5と隣接して加温プレート駆動手段として十字動装置8をネジ80により固定している。この十字動装置8は、図5に示すように直角形状を有する固定部81と、この固定部81の一方の側縁に沿って図示しないアリ溝を介して移動自在にY方向駆動部82を設けている。このY方向駆動部82は、L形状をなすもので、固定部81の一方側縁に沿った一方側片821に対し他方側片822を固定部81の一方側縁に直交する他方側縁に沿った方向に位置している。また、Y方向駆動部82は、一方側片821内部にラック831とピニオン832からなる駆動力伝達機構を設けていて、このうちピニオン832に操作ノブ84より回転操作力を伝えることで、固定部81の一方側縁に沿った移動を可能にしている。また、Y方向駆動部82の他方側片822に沿って図示しないアリ溝を介して移動自在にX方向駆動部85を設けている。このX方向駆動部85は、その内部にリードネジ(雄)861とリードネジ(雌)862からなる駆動力伝達機構を設けていて、このうちリードネジ(雄)861に操作ノブ87より回転操作力を伝えることで、Y方向駆動部82の他方側片822に沿った移動を可能にしている。つまり、操作ノブ84の操作によりY方向駆動部82が固定部81に沿って図示Y方向に直線移動すると共に、操作ノブ87の操作によりX方向駆動部85がY方向駆動部82に沿って図示Y方向に直交する図示X方向に直線移動するようになっている。

そして、このような十字動装置8のX方向駆動部85に接続手段として板ばね9を介して加温プレート6を接続している。この場合、板ばね9は、加温プレート6の移動平面と垂直方向に板厚方向を向けてX方向駆動部85と加温プレート6に固定している。また、板ばね9は、図6に示すように段部91を有するとともに、高所に位置する端部をX方向駆動部84に、低所に位置する端部を加温プレート6にそれぞれネジ92により固定している。

まず、固定ステージ5上の加温プレート6を図示しない加温コントローラにより所定の温度に調整したのち、この加温プレート6上に顕微鏡観察を行う生物組織や卵細胞などの標本12を載置する。

次いで、透過照明用光源10を点灯すると、透過照明用光源10からの透過照明光は、顕微鏡基台1内部のミラー11で反射され、固定ステージ5の透過照明用孔部51より透明な加温プレート6を介して標本12を照明する。

また、ここでは、十字動装置8と加温プレート6の接続に板ばね9を使用しているが、この板ばね9を用いることにより次の効果が期待できる。つまり、加温プレート6は、その形状が大きく十字動装置8による十字動作により加温プレート6先端で生じる抵抗によってモーメントが生じる。このことから、十字動装置8との接続の剛性をよほど高めない限り加温プレート6は、十字動装置8によるXY方向に沿って真っ直に動かない。しかし、剛性接続にすると、加温プレート6先端が固定ステージ5面から浮くか押し付けられるかして、加温プレート6にスムーズな動きが得られない。そこで、これら間の接続に板ばね9を用いると、十字動装置8に対するXY方向の剛性を高くできるが、Z方向の剛性を弱くできるので、Z方向の誤差を吸収でき、十字動装置8のXY方向の動きを確実に加温プレート6に伝えることができる。また、加温プレート6の加温プレート筐体61底面に摩擦抵抗の小さい摺動部材7を設け、固定ステージ5との摩擦抵抗を軽減するようにもしている。

これにより、十字動装置8によるXY方向の駆動により、固定ステージ5上での加温プレート6の動きをスムーズにできるとともに、加温プレート6上の標本12位置を精度よく移動することができるので、従来の標本移動を手操作で行うため、移動精度が得られず所望する部位が顕微鏡の観察範囲からはみだし観察不能に陥るだけでなく、培養溶液中に浮遊している卵細胞が揺れて観察に支障をきたおそれのあるものと比べ、標本12の所望部位を確実に顕微鏡の観察範囲で移動できるとともに、スムーズな移動により培養溶液中に浮遊している卵細胞が揺れるようなこともなくなり、常に安定した顕微鏡観察を実現できる。

なお、上述した実施の形態では、固定ステージ5上での加温プレート6の摺動性を高めるため、加温プレート筐体61の底面に摺動部材7を設けたが、この摺動部材7に代えて、ベアリングなどを用いた車輪などを使用することもできる。また、上述では、加温プレート駆動手段として十字動装置8を使用したが、十字動装置の構成は、顕微鏡基台1上に固定できるものならば、図示した構成によるものでなくとも構わない。さらに、上述では、十字動装置8と加温プレート6の接続手段として板ばね9を用いたが、XY方向に比べ、Z方向、つまり加温プレート6面に対し垂直な方向に自由度の強い結合部材、例えば、ゴム、ゴム系接着剤、両面テープを用いても、同様な効果が得られ、さらには、蝶番を用いても同様な効果が得られる。さらにまた、上述では、一貫して実体顕微鏡について述べたが、固定ステージを有するものならば、他の顕微鏡にも本発明を適用することもできる。
