【発明の属する技術分野】
本発明は、育毛剤原料及び育毛剤組成物に係り、その目的は、優れた育毛効果を有し、しかも頭皮に対してアレルギー反応を起こすことのない安全な育毛剤原料及び育毛剤組成物を提供することにある。

【従来の技術】
従来より、育毛・養毛の目的で使用される頭髪用化粧品や医薬品、医薬部外品が多数創出されており、一般的な育毛剤組成物には、毛根に浸透して血管を拡張、血行を促進、さらには、毛乳頭を刺激し、毛髪の生成を促進する育毛成分、清涼感を与える成分、殺菌作用を有する成分、フケやカユミを防止する成分等が配合されている。
具体的には、育毛成分として、女性ホルモン、ビタミンE、パントテン酸、トウガラシチンキ、ショウキョウチンキ、センブリエキス、セファランチン、感光素等が、清涼感を与え殺菌作用を有する成分としてエタノール等が、フケ防止成分として、レゾルシン、サリチル酸、ジンクピリジオン等が、更にカユミ防止成分として抗ヒスタミン等が配合されたものが多数存在する。

【発明が解決しようとする課題】
しかしながら、上記したような従来の育毛剤組成物では十分な育毛効果が得られず、また、頭皮に対して好ましくない刺激を与えてしまうこともあった。
そこで、優れた育毛効果を有しており、しかも頭皮に対して好ましくない刺激を与えることのない安全な育毛剤原料及び育毛剤組成物の創出が望まれており、本発明者らは、特願平8−153371号において、タカサブロウ(Eclipta alba (L.)Hassk)の有機溶媒による抽出物が優れた育毛効果を有しており、しかも頭皮に対して安全であることを開示している。更に、特願平11−013342号においては、クサギ(Clerodendron trichotomum Thunb. )、ノトパナックススクテラリウム(Notopanax scutellarium Mer. )、ククイノキ(Aleuritesmoluccana Willd. )、カユプテ(Melaleuca leucadendra L.)及びツルグミ(Elaeagnus glabra Thunb. )もまた、優れた育毛効果を有することを提案している。

本発明者らは、優れた育毛効果を有し、しかも天然物由来で頭皮に対する安全性の高い育毛剤原料に関する鋭意研究を更に続けたところ、パンダナスアマリリフォリウス(Pandanus amaryllifolius folia )、ジンギバーゼルベット(Zingiber zerumbet (L.) E. Sm.)、カルメグ(Andrographis paniculata Nees )、ビャクダン(Santalum album L. )、更にはこれらの抽出物が優れた育毛効果を有することを見出し、本発明の完成に至った。

本発明に係る育毛剤原料には、パンダナスアマリリフォリウス(Pandanus amaryllifolius folia )、ジンギバーゼルベット(Zingiber zerumbet (L.) E. Sm. )、カルメグ(Andrographis paniculata Nees )、ビャクダン(Santalum album L. )より選択された1種以上の植物が用いられる。

パンダナスアマリリフォリウス(Pandanus amaryllifolius folia )はタコノキ属タコノキ科に属する単子葉植物である。本発明においては、パンダナスアマリリフォリウス(Pandanus amaryllifolius folia )の地上部及び地下部の全部位が使用可能で、その全草を用いても、或いは葉部や根部、茎部などの各部位を単独で、或いは適宜混合して用いてもよい。特に葉部には有効成分が多く含まれるため好ましく用いられる。また、乾燥状態のもの、非乾燥状態のものいずれも好適に用いられる。

カルメグ(Andrographis paniculata Nees )は、キツネノマゴ科に属する一年生草本で、苦味物質を多く含み、薬として用いられている。本発明においては、カルメグ(Andrographis paniculata Nees )の地上部及び地下部の全部位が使用可能で、その全草を用いても、或いは葉部や根部、茎部などの各部位を単独で、或いは適宜混合して用いてもよい。また、乾燥状態のもの、非乾燥状態のものいずれも好適に用いられる。

ビャクダン(Santalum album L. )はビャクダン科に属する常緑の高木で、インド南部、インドネシアに分布し、仏像、美術彫刻等に用いられ、また薫香、抹香、線香の材料とされている。
本発明においては、ビャクダン(Santalum album L. )の地上部及び地下部の全部位が使用可能で、その全草を用いても、或いは葉部や根部、茎部などの各部位を単独で、或いは適宜混合して用いてもよい。また、乾燥状態のもの、非乾燥状態のものいずれも好適に用いられる。

上記植物の使用形態は特に限定されず、植物の粉砕物や乾燥粉末等をそのまま育毛剤原料としてもよい。また、本発明においては、上記した5種の植物の抽出物を育毛剤原料とすることもでき、更に、抽出物を濃縮や乾燥により軟エキス、乾燥エキスとしたものを用いることもできる。

具体的には、メタノール、エタノール、イソプロピルアルコール、n−プロピルアルコール、イソブタノール、n−ヘキサノール、メチルアミルアルコール、2−エチルブタノール、n−オクタノール等の炭素数1〜8の一価アルコール、エチレングリコール、プロピレングリコール、1、3−ブチレングリコール、ヘキシレングリコール、エチレングリコールモノメチルエーテル、エチレングリコールモノエチルエーテル、プロピレングリコールモノメチルエーテル、プロピレングリコールモノエチルエーテル等の炭素数2〜6の多価アルコール或いはその誘導体、アセトン、メチルアセトン、エチルメチルケトン、イソブチルメチルケトン、メチル−n−プロピルケトン等の炭素数3〜6のケトン、酢酸エチル、酢酸イソプロピル等の炭素数4〜5のエステル、エチルエーテル、イソプロピルエーテル、n−ブチルエーテル等の炭素数4〜8のエーテルや石油エーテル、n−ブタン、n−ペンタン、n−ヘキサン、n−オクタン等の炭素数4〜8の脂肪族炭化水素、四塩化炭素、クロロホルム、ジクロロエタン、トリクロロエチレン等の炭素数1〜2の脂肪族炭化水素のハロゲン化物、ベンゼン、トルエン等の炭素数6〜7の芳香族炭化水素のうちの1種、或いは2種以上が含まれてなる無水或いは含水有機溶媒を好ましい例として挙げることができる。

上記抽出溶媒により得られた抽出液から溶媒を留去して得られた抽出物を育毛剤原料とすることができるが、抽出溶媒としてエタノールを用いた場合には、エタノールは皮膚に対する安全性が高いため、抽出液から溶媒を留去することなく育毛剤原料とすることができる。

本発明に係る育毛剤組成物には、上記育毛剤原料のうちの1種以上が配合される。
育毛剤原料の配合量は特に限定されないが、少なすぎると育毛剤原料配合による効果が十分発揮されないため、全組成物中0.01〜100重量%とするのが望ましく、植物の乾燥粉末や植物の抽出物のみを育毛剤組成物とすることも可能である。

本発明に係る育毛剤組成物には、上記育毛剤原料以外に、育毛・養毛成分として、例えば、ビタミンE及びその誘導体、センブリエキス、ニンニクエキス、セファランチン、塩化カルプロニウム、アセチルコリン等の血行促進剤、トウガラシチンキ、カンタリスチンキ、ショウキョウチンキ、ノニル酸バニルアミド等の局所刺激剤、サリチル酸、レゾルシン、乳酸などの角質溶解剤、プラセンタエキス、ペンタデカン酸グリセリド、パントテニルエチルエーテル、ビオチン、ヒノキチオール、アラントイン等の代謝賦活剤、グリチルリチン酸、グリチルレチン酸等の消炎剤、イソプロピルメチルフェノール、トリクロサン、ジンクピリチオン、ヒノキチオール等の殺菌剤、メントール、カンフル等の清涼剤、その他女性ホルモン等を適宜配合することも可能である。

乾燥したパンダナスアマリリフォリウス(Pandanus amaryllifolius folia )の葉の粉砕物100gにメタノール500mlを加えて還流抽出し、抽出液を冷却、濾過後、溶媒を留去して得られた抽出物3.1gを実施例2の試料とした。

乾燥したジンギバーゼルベット(Zingiber zerumbet (L.) E. Sm. )の根茎の粉砕物100gにメタノール500mlを加えて還流抽出し、抽出液を冷却、濾過後、溶媒を留去して得られた抽出物2.5gを実施例3の試料とした。

〔試験例1:マウスの発毛に対する効果〕
(試験方法)
(1)試料溶液の調製
実施例〜5の各試料を、その濃度が5%(w/v)となるように99.9%エタノールに溶解したものを検体溶液とした。また、99.9%エタノールを比較例1の検体溶液とした。

(2)マウスへの塗布
C3H/HeN Crjマウス(8週齢、体重21〜26g)を一週間以上馴化飼育を行ったあと、異常のなかったものについて、背部被毛を電気バリカンで、2cm×4cmの広さに毛刈りし、さらに電気シェーバーにて除毛し、検体溶液の投与部位とした。除毛してから3日後、実施例〜5及び比較例1の検体溶液を各10匹のマウスに連続19日間、100μlずつ、1日1回午前中に塗布した。試験期間中、C3H/HeN Crjマウスは、温度22±2℃、相対湿度55±15%、換気回数20回/時、照射時間を午前6時から午後6時に設定した飼育室で、プラスチックケージ(14.5cm×26cm×12.5cm)を用いて5匹ずつ飼育した。

〔試験例2:マウス毛包由来細胞の増殖に対する効果〕
(試験方法)
(1)試料溶液の調製
まず、実施例〜5の試料を、培養液に添加したときの最終濃度が後記表2〜6に示す濃度となるように、99.9%エタノールに溶解した。また比較例2としては、近年優れた育毛効果を有する物質として注目されているミノキシジルを培養液に添加したときの最終濃度が70μg/mlとなるように、99.9%エタノールに溶解したものを用いた。

(2)毛根の回収
生後4日齢のC3H/HeSlc系新生仔マウスの皮膚を無菌的に採取し、10%FBS−DMEM培地で数回洗浄した。筋組織を取り除き、皮膚片を約1mm幅の短冊状に切り、毛包下部が現れるよう真皮結合組織を剥離した。出来るだけ多くの完全な毛球が得られるよう、メスで真皮組織をさらに細かく分け、0.2%コラゲナーゼDMEM培養液(カルシウム、マグネシウム不含)で60分間、37℃でインキュベートした後、5℃に冷し、10%FBS−DMEM培地を加え反応を止め、毛球を回収した。

得られた毛根をトリプシン処理し、毛球部分の細胞(主に毛母細胞)を得、この細胞を10%FBS−DMEM培地に分散させコラーゲンコートした96ウェルマイクロプレートに播種した。5%CO2 ,37℃条件下で24時間培養した後、培養液を試験培地(MCDB153培地に、5μg/mlのインシュリン、5ng/ml のEGF,0.5μg/mlのHydrocortisone及び、35μg-protein/ml-Bovine Pituitary Extract を添加したもの)に試験物質又は99.9%エタノールのみを1/100容添加したものに交換し、引き続き同じ条件で4日間培養した後、細胞数を測定した。また、対照例として、99.9%エタノールのみを添加したものについても同様に培養し、細胞数を測定した。尚、比較例及び対照例の試料を用いた試験は、実施例〜5それぞれについて、実施例と同じ条件下で行った。
