【発明の属する技術分野】
本発明は、大腸菌群判定装置、特に、被試験水中における大腸菌群の存否を判定するための大腸菌群判定装置に関する。

【従来の技術とその課題】
飲料水や食品の衛生管理においては、大腸菌群の存否の検査が必要不可欠となっている。ここで、「大腸菌群」とは、好気性または通性嫌気性のグラム陰性無芽胞の桿菌であり、乳糖を分解して酸とガスとを生じるか、β−ガラクトシダーゼをもつ細菌群をいい(例えば、社団法人日本水道協会発行、「上水試験方法2001 解説」845頁参照)、「大腸菌」そのものとは異なる概念である。因みに、飲料水中に大腸菌群が含まれている場合、当該飲料水は、単に汚物で汚染されているということだけではなく、病原菌類も含んでいる可能性があることを示唆することになるため、水道法上の水質基準では飲用に不適なものと判定されることになる。

ところで、大腸菌群の指標となる性状は乳糖発酵性であり、それに関与する酵素はβ−ガラクトシダーゼ(β−galactosidase)である。したがって、飲料水等の被試験水を細菌の培養環境に設定し、そこからβ−ガラクトシダーゼを検出することができると、間接的に大腸菌群の存在を証明することができる。そこで、このような大腸菌群の性状を利用した、飲料水等の被試験水中における大腸菌群の存否を迅速に判定するための方法として、合成酵素基質培地法が知られている。合成酵素基質培地法は、発色物質または発光物質を結合させた酵素基質を培地に使用し、目的とする細菌がもつ特異酵素により当該酵素基質が加水分解されて発色または発光することを利用した検査方法(判定方法)であり、大腸菌群の検出用の合成酵素基質培地法として、被試験水の変色の有無により大腸菌群の存否を判定するMMO−MUG法やXGal−MUG法が知られている。

このうち、XGal−MUG法では、発色合成酵素基質である5−ブロモ−4−クロロ―3−インドリル−β−D−ガラクトピラノシド(通称X−Gal)、大腸菌群の栄養素となるペプトン等およびピルビン酸ナトリウム等の炭素源、塩類、界面活性剤並びにpH調整剤を所定の濃度で含む合成酵素基質培地を一定量の被試験水中に所定量添加し、36±1℃で合成酵素基質培地の種類に応じて24〜48時間培養する。ここで、被試験水中に大腸菌群が含まれている場合は、大腸菌群が栄養素により培養され、β−ガラクトシダーゼが生成する。生成したβ−ガラクトシダーゼは、発色合成酵素基質であるXGalを加水分解し、青〜青緑色を呈する5,5−ジブロモ−4,4−ジクロロインジゴを生成させる。これにより、被試験水は青〜青緑色に変色するので、大腸菌群を含むものと判定することができる。一方、被試験水中に大腸菌群が含まれていない場合は、上述の合成酵素基質培地を添加しても大腸菌群が培養されることはないので、β−ガラクトシダーゼが生成することはなく、被試験水は上述のような青〜青緑色に変色しない。これにより、被試験水には大腸菌群が含まれないものと判定することができる。

ところが、上述のような合成酵素基質培地法は、経験を積んだ検定者の手作業により実施され、被試験水の変色は当該検定者が目視で判断しているため、作業が煩雑である。

本発明の目的は、被試験水中における大腸菌群の存否を合成酵素基質培地法に基づいて自動的に判定できるようにすることにある。

【課題を解決するための手段】
本発明に係る大腸菌群判定装置は、被試験水中における大腸菌群の存否を判定するためのものであり、被試験水を貯留するための容器と、容器内に貯留された被試験水の温度を調節するための温度調節装置と、容器内に貯留された被試験水に対し、β−ガラクトシダーゼと反応して発色する5−ブロモ−4−クロロ−3−インドリル−β−D−ガラクトピラノシドを発色合成酵素基質として含む合成酵素基質培地を供給するための試薬供給装置と、合成酵素基質培地の供給後における、発色合成酵素基質とβ−ガラクトシダーゼとの反応による被試験水の変色を判定するための判定装置とを備えている。判定装置は、被試験水における赤色光の透過率の変化に基づいて被試験水の変色を判定している。

ここで、合成酵素基質培地は、例えば、大腸菌群を検出するためのXGal−MUG培地である。

この大腸菌群判定装置において用いられる合成酵素基質培地の濃度は、通常、被試験水中における大腸菌群の存否を判定するための合成酵素基質培地法において用いられる合成酵素基質培地の規定濃度の少なくとも2倍に設定されている。

なお、本発明の大腸菌群判定装置は、例えば、容器に被試験水を供給するための供給路と、容器内に貯留された被試験水を容器の外部に排出するための排出路と、供給路を通じて容器に洗浄液を供給するための洗浄液供給装置とをさらに備えていてもよい。この場合に用いられる洗浄液は、例えば殺菌剤を含んでいる。

また、本発明の大腸菌群判定装置は、例えば、被試験水に対して試薬供給装置が合成酵素基質培地を供給する前に、容器の洗浄状態を確認するための洗浄確認装置をさらに備えていてもよい。

【発明の実施の形態】
図1を参照して、本発明の実施の一形態に係る大腸菌群判定装置を説明する。図において、大腸菌群判定装置1は、XGal−MUG法により被試験水中の大腸菌群の存否を自動的に判定するためのものであり、容器2、温度調節装置3、試薬供給装置4、洗浄液供給装置5、吸光度測定装置6および制御装置7を主に備えている。

容器2は、大腸菌群の存否の判定対象となる被試験水を貯留するためのものであり、例えば、石英ガラスなどの透光性を有する無色透明の材料を用いて形成された、底部を有する円筒状のもの(例えば試験管)である。容器2の上端部は、栓27により気密に封止されている。また、この容器2は、被試験水を内部に供給するための供給路20と、容器2に貯留された被試験水を外部に排出するための排出路21とを備えている。供給路20は、被試験水を供給するための供給ポンプ22と、被試験水の流通を制御するための第1電磁弁23とを備えており、栓27を通じて容器2内の下方に向けて延びている。一方、排出路21は、容器2内に貯留された被試験水を排出するためのものであり、栓27から容器2の外部に向けて延びている。

上述の容器2は、マグネチックスターラー25上に配置されており、また、内部の底部には磁石を内蔵した攪拌子26が配置されている。

温度調節装置3は、容器2内に貯留された被試験水の温度を大腸菌群の培養に適した温度に調節するためのものであり、容器2の上部外周に配置された、例えばアルミニウム等の良伝熱性材料からなるブロック30と、このブロック30を加熱するためのヒーター31とを備えている。

試薬供給装置4は、容器2内に貯留された被試験水に対してXGal−MUG法において用いられる合成酵素基質培地を供給するためのものであり、当該合成酵素基質培地を貯蔵するための試薬タンク40と、試薬タンク40から容器2内に延びる試薬供給路41とを備えている。試薬供給路41は、試薬タンク40内の合成酵素基質培地を容器2内に送り出すための試薬ポンプ42と、逆止弁43とを有している。

洗浄液供給装置5は、容器2および被試験水供給路20を洗浄するためのものであり、洗浄液を貯蔵するための洗浄液タンク50と、洗浄液タンク50から延びる洗浄液供給路51とを備えている。洗浄液供給路51は、洗浄液タンク50内の洗浄液を送り出すための洗浄液ポンプ52と、洗浄液の流量を制御するための第2電磁弁53とを有しており、供給路20において、供給ポンプ22と第1電磁弁23との間に連絡している。

吸光度測定装置6は、主として、容器2を通過する可視紫外光の吸光度を測定するためのものであり、容器2の下部近傍に配置された光源61と、容器2を挟んで光源61と対向する受光装置62とを主に備えている。受光装置62は、光源61からの可視紫外光のうち、後述する特定の波長の光を選択するためのフイルタを備えている。

制御装置7は、大腸菌群判定装置1の動作を制御するためのものであり、図2に示すように、中央処理装置(CPU)70、大腸菌群判定装置1の動作プログラムを記憶している読出し専用メモリー(ROM)71、各種の電子情報を記憶するためのランダムアクセスメモリー(RAM)72および入出力ポート73を備えている。入出力ポート73の入力側には、オペレーターが大腸菌群判定装置1に対して各種の動作指令を入力するためのスイッチ類74、受光装置62およびその他の装置が接続されている。一方、入出力ポート73の出力側には、光源61、各電磁弁23,53、各ポンプ22,42,52、マグネチックスターラー25、ヒーター31、大腸菌群の存否の判定結果を表示するための表示装置75および当該判定結果を印字するためのプリンタその他の装置が接続されている。

なお、この制御装置7に記憶された動作プログラムは、吸光度測定装置6で測定された被試験水の吸光度に基づいて被試験水の変色の有無を判定する判定プログラム(判定装置の一例)、および吸光度測定装置6で測定された容器2の吸光度に基づいて容器2の洗浄状態を確認するための洗浄確認プログラム(洗浄確認装置の一例)を含んでいる。

上述の大腸菌群判定装置1において用いられる合成酵素基質培地、すなわち、試薬タンク40内に貯蔵される合成酵素基質培地は、基本的には、XGal−MUG法において用いられる合成酵素基質培地(XGal−MUG培地)である。より具体的には、例えば、社団法人日本水道協会発行、「上水試験方法2001解説」842〜843頁の表に挙げられたピルビン酸添加XGal−MUG培地であり、1リットル中において次のような酵素基質、大腸菌群培養のための栄養成分、塩類、界面活性剤およびpH調製剤を含みかつpHが7.1±0.2に調整されたものである。

β−ガラクトシターゼと反応して発色する発色合成酵素基質であるXGal(5−ブロモ−4−クロロ―3−インドリル−β−D−ガラクトピラノシド)を0.10g、大腸菌群酵素誘導剤であるIPTG(1−イソプロピル−β−D−1−チオガラクトピラノシド)を0.10gおよび大腸菌検出のための酵素基質であるMUG(4−メチルアンベルリフェリル−β−グルクロニド)を0.10g。
大腸菌群培養のための栄養成分
ペプトンを5.0gおよびその他の炭素源としてピルビン酸ナトリウムを1.0g。
塩類
塩化物として塩化ナトリウムを5.0g、硝酸塩として硝酸カリウムを1.0g。
界面活性剤
ラウリル硫酸ナトリウムを0.10g。
pH調整剤
リン酸二水素カリウムを1.0g、リン酸水素二カリウムを4.0g。

なお、上述のようなピルビン酸添加XGal−MUG培地は市販されており、一例として日水製薬株式会社の商品名“ECブルー”を挙げることができる。

但し、この大腸菌群判定装置1では、上述の合成酵素基質培地として、各成分の濃度が規定濃度の少なくとも2倍、好ましくは少なくとも6倍、より好ましくは少なくとも11倍に設定されているものを用いるのが好ましい。ここで、規定濃度とは、例えば上述の「上水試験方法2001 解説」842〜843頁の表等において規定された、大腸菌群検出のための合成酵素基質培地法において用いる合成酵素基質培地の濃度をいう。因みに、ピルビン酸添加XGal−MUG培地の規定濃度は、上述のような各成分の含有量により規定されている。

なお、上述のような高濃度の合成酵素基質培地は、所要の成分をその含有量が規定濃度の少なくとも2倍になるよう水中に溶解するか、あるいは、規定濃度の既存の合成酵素基質培地を液量が少なくとも1/2になるまで濃縮すると調製することができる。

因みに、規定濃度の合成酵素基質培地は、上述のような栄養素を含んでいるため、試薬タンク40での貯蔵中において各種の雑菌が繁殖し易く、これらの雑菌により短時間のうちに汚染される可能性が極めて高い。したがって、大腸菌群判定装置1において規定濃度の合成酵素基質培地を試薬タンク40で貯蔵しながら用いると、被試験水における大腸菌群の存否の判定が不正確で信頼性を欠く可能性がある。これに対し、上述のような高濃度の合成酵素基質培地は、高濃度の塩類を含むため、試薬タンク40内で貯蔵しても雑菌の繁殖が抑制されるので、すなわち、雑菌により汚染される可能性が小さくなるので、被試験水における大腸菌群の存否を正確に判定することができる。

なお、上述のような高濃度の合成酵素基質培地を用いると、規定濃度のものを用いる場合に比べて試薬タンク40の容量を小さく設定することができるので、大腸菌群判定装置1を小型化することもできる。

一方、上述の大腸菌群判定装置1において用いられる洗浄液、すなわち、洗浄タンク50に貯蔵される洗浄液は、容器2内および被試験水の供給路20を洗浄するためのものであり、通常、界面活性剤、酸類(有機酸類若しくは無機酸類またはこれらの任意の混合物)、アルカリ類(アルカリ金属やアルカリ土類金属の水酸化物、炭酸塩、リン酸塩若しくはケイ酸塩またはこれらの任意の混合物)および酵素(アミラーゼ、プロテアーゼ若しくはリパーゼまたはこれらの任意の混合物)のうちの一つまたは二つ以上を洗浄剤として含む水溶液である。特に、この洗浄液は、被試験水供給路20および容器2内が各種の細菌類により汚染されていない清浄な状態に維持するために、殺菌剤を含むのが好ましい。ここで利用可能な殺菌剤は、特に限定されるものではないが、通常、次亜塩素酸ナトリウム等の塩素系殺菌剤、過酸化水素、逆性石鹸および両性石鹸のうちの一つまたは二つ以上のものが好ましく用いられる。

なお、殺菌剤として特に好ましいものは、次亜塩素酸ナトリウム等の塩素系殺菌剤である。塩素系殺菌剤は、各種の菌類に対して殺菌作用を示すだけではなく、漂白剤としても機能するため、後述する容器2の洗浄工程において、容器2の透明性を高めることができる。特に、合成酵素基質培地として上述のようなピルビン酸添加XGal−MUG培地を用いると、被試験水中に大腸菌群が含まれる場合は当該被試験水が青〜青緑色に変色するため、容器2の内面がそのような色で着色する場合があるが、塩素系殺菌剤を用いると、そのような着色も洗浄工程において容易に除去することができる。

次に、図3〜図5に示す動作プログラムのフローチャートに基づいて、大腸菌群判定装置1の動作を説明する。
オペレータが大腸菌群判定装置1の電源をONにすると、プログラムは、ステップS1において、第1電磁弁23を開放状態に設定し、また、第2電磁弁53を閉鎖する等の初期設定動作を実施する。

次に、プログラムは、ステップS2において、オペレータが判定開始スイッチをONしたか否かを判断する。ここで、オペレータが判定開始スイッチをONにしない場合、プログラムはそのまま待機状態を維持する。一方、オペレータが判定開始スイッチをONにすると、プログラムはステップS3に移行し、制御装置7の判定開始識別フラグをONに設定する。

次に、ステップS4において、プログラムは、被試験水の採水工程を実施する。ここでは、供給ポンプ22が動作し、目的とする被試験水が供給路20を通じて容器2内に供給される。そして、所定量の被試験水(通常は50ml)が容器2内に供給されると、第1電磁弁23が閉鎖すると共に供給ポンプ22が停止し、被試験水の供給は停止する。

ステップS4の終了後、プログラムはステップS5に移行し、試薬供給工程を実施する。ここでは、試薬供給装置4において、試薬ポンプ42が作動する。この結果、試薬タンク40内に貯留されている合成酵素基質培地が試薬ポンプ42により送り出され、試薬供給路41を通じて容器2内に貯留されている被試験水中に注入される。ここで、合成酵素基質培地は、上述のように濃縮されている場合、被試験水中に溶解したときの濃度が被試験水に対して規定濃度の合成酵素基質培地を規定量注入した場合の濃度と等しくなるよう注入量が設定される。この注入量は、試薬ポンプ42の動作により制御される。また、このような合成酵素基質培地の注入時には、同時にマグネチックスターラー25が作動し、容器2内の被試験水が攪拌子26により攪拌される。この結果、注入された合成酵素基質培地は、被試験水中において均等に分散することになる。所定量の合成酵素基質培地が被試験水に注入されると、試薬ポンプ42が停止し、試薬供給工程は終了する。また、マグネチックスターラー25が停止し、被試験水の攪拌が停止される。

次に、プログラムはステップS6に移行し、培養設定工程を実施する。ここでは、温度調節装置3においてヒーター31が作動し、ブロック30を加熱する。この結果、容器2がブロック30により加熱され、容器2内の被試験水は温められる。さらに、マグネチックスターラー25が作動し、被試験水が加熱撹拌される。そして、次のステップS7では、被試験水の温度が大腸菌群の培養に適した温度、例えば36±1℃に設定されたか否かを判断する。被試験水の温度が当該温度に設定されると、プログラムは、ステップS8において内部タイマーを作動させ(すなわち、経過時間tを0に設定し)、続いてステップS9において大腸菌群の培養に必要な所要の経過時間、例えばピルビン酸添加XGal−MUG培地を用いる本実施の形態の場合は24時間が経過したか否か(すなわち、t=24時間になったか否か)を判断する。これにより、被試験水は、上述の合成酵素基質培地の存在下で24時間培養されることになる。

t=24時間になると、プログラムはステップS10に移行し、判定設定工程を実施する。ここでは、ヒーター31およびマグネチックスターラー25の作動を停止し、培養を停止する。また、吸光度測定装置6を作動させ、光源61から受光装置62に向けて可視紫外光を照射する。そして、プログラムは、次のステップS11〜ステップS13において、被処理水中に大腸菌群が含まれているか否かを判定する。

ここで、先ず、大腸菌群の存否の判定原理を説明する。被試験水は、大腸菌群を含む場合(ケース1)、上述のような合成酵素基質培地(すなわち、XGalを含む合成酵素基質培地)の存在下で培養処理が施されると、培養された大腸菌群に由来のβ−ガラクトシダーゼを含むことになる。このβ−ガラクトシダーゼは、合成酵素基質培地中に含まれるXGalと反応し、被処理水を青〜青緑色に変色させる。したがって、被試験水の可視紫外光吸収スペクトルは、図6に実線で示すように、青〜青緑色に対応する650nm付近に吸収ピークを有することになる。これに対し、被試験水が大腸菌群および大腸菌群以外の細菌を含まない場合(ケース2)、被試験水は、上述のような培養処理後であっても青〜青緑色には変色しないことになる。従って、この場合における被試験水の可視紫外光吸収スペクトルは、図6に点線で示すように高波長側に向けてなだらかに減少し、特定の波長での吸収ピークを示さない。

一方、被試験水が大腸菌群を含まず、大腸菌群以外の細菌を含む場合(ケース3)、被試験水は、上述のような培養処理後に青〜青緑色には変色しないが、大腸菌群以外の細菌の増殖のために白濁する。このため、この場合における被試験水の可視紫外光吸収スペクトルは、図6に一点鎖線で示すように高波長側に向けてなだらかに減少し、ケース2の場合と同じく特定の波長での吸収ピークを示さないが、全波長領域における吸光度がケース2の場合に比べて上昇することになる。

したがって、培養処理後の被試験水は、青〜青緑色に対応する650nm付近の吸光度の測定結果からだけでは、大腸菌群の存在を示す青〜青緑色に変色しているのか、大腸菌群以外の細菌により単に白濁しているだけなのかの判別が困難である。すなわち、被試験水は、ケース1に該当するのかケース3に該当するのかの判別が困難である。しかし、ケース1の吸収スペクトルは650nm付近に吸収ピークを有しているのに対し、ケース3の吸収スペクトルは当該波長付近において吸収ピークを有していない。したがって、ケース1の場合、650nm付近の高波長側の吸光度に比べ、当該波長付近の吸収ピークの低波長側の裾にあたる550nm付近の吸光度が小さくなる。これに対し、ケース3の場合、被試験水は、650nm付近の吸光度が550nm付近の吸光度よりも小さくなる。これによると、550nm付近の吸光度と650nm付近の吸光度とを比較し、前者が後者に比べて小さい場合は、被試験水が青〜青緑色に変色しているものと判断することができる。逆に、前者が後者に比べて大きい場合は、被試験水が青〜青緑色に変色していないものと判断することができる。

そこで、プログラムは、ステップS11において、受光装置62のフイルタを選択して550nmの波長の光の吸光度を測定し、そのデータを記録する。また、次のステップS12において、受光装置62の他のフイルタを選択して650nmの波長の光の吸光度を測定し、そのデータを記録する。そして、ステップS13において、記録した550nmの光の吸光度と650nmの光の吸光度とを比較する。

ここで、550nmの吸光度が650nmの吸光度に比べて小さい場合、プログラムはステップS14に移行し、制御装置7において洗浄識別フラグがONか否かを判断する。洗浄識別フラグがOFFの場合、プログラムはステップS15に移行し、被試験水中に大腸菌群が存在していることを示す「大腸菌群陽性」の旨を表示装置75に表示すると共にその旨をプリンタで印字する。

一方、550nmの吸光度が650nmの吸光度に比べて大きい場合、プログラムはステップS13からステップS16に移行し、制御装置7において判定開始識別フラグがONか否かを判定する。判定開始識別フラグがONの場合、プログラムはステップS17に移行し、被試験水中に大腸菌群が存在しないことを示す「大腸菌群陰性」の旨を表示装置75に表示すると共にその旨をプリンタで印字する。

ステップS15またはステップS17の後、プログラムは、ステップS18において、ステップS15またはステップS17から一定時間(例えば、数分)が経過したか否かを判断する。一定時間が経過すると、プログラムは、ステップS19において制御装置7の判定開始識別フラグをOFFに設定し、続くステップS20において制御装置7の洗浄識別フラグをONに設定する。その後、プログラムは、ステップS21に移行し、殺菌設定工程を実施する。ここでは、ヒーター31およびマグネチックスターラー25を作動させ、容器2内の被試験水を再度加熱撹拌する。

そして、次のステップS22において、プログラムは、被試験水の温度が大腸菌群の殺菌に適した温度、例えば80℃に設定されたか否かを判断する。被試験水の温度が80℃に設定されると、プログラムはステップS23において内部タイマーを作動させ(すなわち、経過時間tを0に設定し)、続いてステップS24において大腸菌群の殺菌に必要な所要の経過時間、例えば30分が経過したか否か(すなわち、t=30分になったか否か)を判断する。t=30分になると、プログラムは次のステップS25に移行する。なお、このような殺菌工程により、容器2は、被試験水中での培養により生育した高濃度の大腸菌群による汚染が防止されることになる。

ステップS25において、プログラムは、被試験水の排出工程を実施する。ここでは、供給ポンプ22を一定時間作動させて容器2に被試験水を再度供給する。これにより、容器2内の被試験水は、新たに供給される被試験水により押出され、排出路21を通じて外部に排出される。この結果、容器2内の被試験水は、新たに供給される被試験水により置換されることになる。
