【発明の属する技術分野】
本発明は、滅菌装置に関し、詳しくは、片面が透明フィルムであり、他の片面が不織布である容器に内包された医療材料等を、容器ごと安全に、かつ効率よく滅菌できる滅菌装置に関する。

【従来の技術】
医療材料や食料品などの、高度の信頼性を要する滅菌方法としては、酸化エチレンガス等の殺菌剤を用いる方法、ガンマ線や電子線等の放射線を照射する方法、オートクレーブによる高圧蒸気を用いる方法等が知られている。

酸化エチレンガス等の殺菌剤を用いる殺菌方法は、使用する殺菌剤そのものが毒性を有することが多い。そのため、密閉系で処理しなければならず、処理装置自体が大型となる。さらに、被処理物に殺菌剤が残存する虞もある。

ガンマ線や電子線等の放射線を照射する方法は、前記殺菌剤を用いた場合の如き殺菌因子が残存する虞はない。しかし、放射線照射により被処理物の機械的強度を低下させたり、物品が樹脂である場合には、樹脂が分解等して悪臭を発生したり、変色する等の問題点がある(特公平3−73309号公報参照)。

また、オートクレーブを用いる方法は、殺菌が高圧、高温度下で行われるため、被処理物に制限があった。

これら従来技術が有する課題を解決できる殺菌方法としてプラズマを用いる方法が知られている(特開平5−229530号)。この方法は、例えば、複合酸化物からなるエネルギー変換体に電磁波を照射し、励起したエネルギー変換体と希ガス等を接触させたプラズマ状態とし、プラズマ状になった希ガス等を被殺菌体と接触させるものである。このプラズマを用いる方法は、包装材料等の被処理物を安全で、容易に殺菌でき、かつ殺菌した物品を変質させることが少なく、優れた方法といえる。本発明者らはプラズマを用いる方法について検討を進め、先に、より小型の装置で実施しうる、安全性の高い殺菌方法を見出し、出願を行った(特開平8−168516号)。本発明は、その殺菌方法を実用に供するための好適な装置に関するものである。

【発明が解決しようとする課題】
即ち、本発明の目的は、医療材料包装体等の高度の信頼性を要する物品の滅菌に好適に使用し得る、安全で、被処理物を変質させることが少なく、信頼性の高い滅菌を効率よく行い得る滅菌装置を提供することにある。

【課題を解決するための手段】
本発明は、大気圧プラズマを応用した滅菌装置であり、本発明の滅菌装置は、大気圧でプラズマを発生させるプラズマ発生器を備え、該大気圧プラズマ発生器により生成される殺菌因子を貯留する第1のチャンバーと、該第1のチャンバーとの間が連結管で連結され、且つ、内部に被処理物を配置し得ると共に、内部を真空状態にできるように耐圧構造とされた第2のチャンバーと、開閉自在に該連結管に配置され、且つ、開放によって真空とされる第2のチャンバー内に第1のチャンバー内から殺菌因子を供給しうる開閉バルブと、を有することを特徴とする。

通常は滅菌に有効な量の殺菌因子を被処理物に供給するために相当の時間を要するものであるが、被処理物を滅菌するチャンバーとは別に殺菌因子を貯留するためのチャンバーを備え、両者を開閉バルブを備えた連結管で連結することにより、滅菌に必要な殺菌因子の貯留と、被処理物の乾燥等の前処理とを平行して行うことができ、さらに、相当量の殺菌因子を短時間で被処理物に接触させることができるため、有効な滅菌処理を効率よく行い得る。

【発明の実施の形態】
以下に本発明の実施の形態について詳細に説明する。

本発明の滅菌装置に用いる大気圧でプラズマを発生させるプラズマ発生器は、電界を発生させるためにパルス電圧を用い、気体と液体との混合物の少なくとも一部を電離させる機能を有している。本発明の滅菌装置においては、この気体と液体との混合物を電離させて得た殺菌因子を被処理物に接触させて滅菌することが特徴である。

図1は本発明の滅菌装置の一態様を示す概略断面図である。滅菌装置10は、プラズマ発生器12を備えた第1のチャンバー14と、耐圧構造の第2のチャンバー18とそれらの間を連結するとともに、第1のチャンバーから第2のチャンバーへ供給する殺菌因子を含んだ気体の流れを制御する開閉自在の開閉バルブ20及び加圧装置であるコンプレッサー22を備えた連結管24を主要部として構成される。第2のチャンバーには、第2のチャンバー内の気体を吸排することにより内部の気圧を一定に保つ圧力センサーと開閉バルブとからなる圧力調整器16が連結されている。

第1のチャンバー14(殺菌因子貯留タンク)は特に耐圧構造を必要としないため、ステンレス鋼(sus304)などの通常の肉厚を有する鋼板で成形することができる。この第1のチャンバー14に備えられたプラズマ発生器12においては、パルス電源を用いて気体と液体の混合物の少なくとも一部を電離させることができ、かくして得られた電離混合物が殺菌因子となる。

ここで、気体と液体の混合物の少なくとも一部を電離させるために印加する電界は、例えば、大気圧プラズマ発生器12内に少なくとも1対の高圧電極と接地電極とを用い、この電極間に一定以上の電圧を与えることで発生させることができる。

図2は、本発明の滅菌装置10に好適に用いられる大気圧プラズマ発生器12の一例を示す概略断面図である。石英被覆電極26と金属電極27によって筒状の接地電極28を構成しており、その筒状の接地電極28の略中央部には棒状の金属電極30が配置されて高圧電極を構成する。電流を印加することにより、この接地電極28と高圧電極30との間に電界が形成される。ここに気体又は気体と液体の混合物の導入管(パイプ32)及び気体と液体の混合物の導入管(パイプ34)が配置されている。

気体と液体との混合物を直接電離させる場合には、気体を過酸化水素等の液体を満たしたネブライザーに通して得られる霧状の混合物をパイプ32から電界中に供給する。接地電極28と高圧電極30の間を通過したガスは、少なくとも一部が電離し、殺菌因子を形成する。この場合には、パイプ34は使用しない。また、気体を電離させ、次いで電離した気体と、気体と液体との混合物を混合する場合には、電離用の気体をパイプ32から導入し、パイプ34からは、気体を過酸化水素等の液体を満たしたネブライザーに通して得られる霧状の混合物を導入する。接地電極28と高圧電極30との間を通過した気体は、少なくとも一部が電離し、この電離気体はパイプ34から供給される霧状の混合物と混合され、殺菌因子を形成する。

このような電界の発生装置は、例えばコロナ放電等に用いられる高圧電極と接地電極とをそのまま用いることができ、高圧電極及び接地電極の少なくともいずれか一方の表面が固体誘電体で被覆されているものを用いることができる。なお、固体誘電体には特に制限はないが、例えば石英等のセラミックスやハイパロンラバー、ポリエチレンテレフタレート等のポリエステルの積層体等を用いることができる。また、高圧電極30及び接地電極28のいずれもが、金属電極であることもできる。

また、大気圧プラズマ発生器12の高圧電極30と接地電極28の数及び形状等には特に制限はなく、発生させた電界内を通過する気体又は気体と液体の混合物をどの程度電離させる必要があるか否かにより適宜決定できる。例えば、気体又は気体と液体の混合物の流量が多い場合は 電離の方法には、電界中に気体と液体の混合物を通して、前記混合物の少なくとも一部を電離させる方法と、電界中に気体を通して得られる少なくとも一部を電離させた気体と、気体と液体の混合物とを前記電界外で混合して少なくとも一部が電離した気体と液体の混合物を得る方法とがある。

これら、殺菌因子の原料となる気体、液体を一定以上の割合で電離させる目的で、電界中における滞在時間が長くなるように調整することができ、その手段としては、高圧電極と接地電極を並列に複数設けたり、或いは高圧電極と接地電極の少なくとも一方を帯状の形状にすることもできる。また、局部放電を防止する目的で、高圧電極の表面積を大きくするために、電極に突起や凹凸等を設けることもできる。

本発明の滅菌装置に用いられる大気圧プラズマ発生装置において、殺菌因子を発生させるための、パルス電圧の立ち上がり速度は、いずれも0.01kV/ns〜10kV/nsの範囲にあることが適当である。パルス電圧の立ち上がり速度が0.01kV/ns未満では、滅菌効果も低下する傾向がある。パルス電圧の立ち上がり速度が10kV/nsを超えても、滅菌効果に悪影響はないが、電圧発生が困難となる。

パルス電圧の立ち上がり速度の好ましい範囲は0.1kV/ns〜1kV/nsの範囲である。

また、上記パルス電圧のパルス幅は10-9秒〜10-1秒の範囲にあることが適当である。パルス幅が滅菌効果には大きな影響はないが、発振可能なパルス幅は上記の範囲である。パルス幅の好ましい範囲は10-8秒〜10-6秒である。

パルス電圧のピーク電圧は1kVp〜100kVpの範囲にあることが適当である。ピーク電圧が1kVp未満では、電界強度が小さく、ピーク電圧が100kVpを超えると装置を大型化する必要がでてくる等の問題がある。ピーク電圧の好ましい範囲は8〜50kVpである。

パルス電圧の周波数は1Hz〜100kHzの範囲であることが適当である。周波数が1Hz未満では、滅菌効率が低下し、100kHzを超えると電界内のガスの温度が大幅に上昇する。パルス電圧の周波数の好ましい範囲は、50Hz〜500Hzの範囲である。

殺菌因子である電離混合物発生のために、大気圧プラズマ発生器内で電界を通過させる原料となる気体は、前記の電界中で電離可能な気体である。そのような気体として、例えば、酸素、窒素、希ガス(アルゴン、ヘリウム及びネオン)、水素、空気等を挙げることができる。希ガス中でも、アルゴンは電離し易く、コスト的に優れているので好ましい。また、ヘリウムは電離が連続的になり易いという観点から好ましい。特に、アルゴンは、ヘリウムよりも比重が空気により近く、大気圧下での取扱が容易であるため、より好適に使用することができる。また、上記気体の2種以上を混合して併用することもできる。

また、液体は、例えば、水、過酸化水素、過酸化水素水、エタノール、エタノールと水との混合物等であることができる。

過酸化水素水を用いる場合、過酸化水素の濃度は、市販され、入手が容易であるという観点からは、例えば過酸化水素濃度50%以下のものであることが適当である。それ以下の濃度においては、滅菌条件等を考慮して、市販の過酸化水素水を水で希釈して適宜濃度を調整することができる。但し、滅菌効果を考慮すると1%以上の過酸化水素水を用いることが好ましい。

上記液体は、霧状であることが好ましく、霧状の液体は、液体の供給源と接続しているネブライザーに上記気体をキャリアーガスとして通すことにより発生させることができる。また、霧状の気体は、これらにキャリアーガスをバブリングさせることによっても発生させることができる。また、気体の一部をキャリアーガスとし、気体とキャリアーガスとで得られた霧状物を残りの気体と混合することによっても調製することができる。

気体(キャリアーガスも含む全ての気体)と液体との割合は、特に制限はないが、気体1リットル当たり1mg〜100mgの範囲とすることが、放電持続と被処理物への圧力と言う観点から適当である。

電界中を通過させる気体又は気体と液体の混合物は、前記の如く少なくとも一部が電離して殺菌因子を形成することが必要である。そこで、気体又は気体と液体の混合物の流量、電界発生のため投入する電圧及び電流(電力)量、電極の数及び形状、等は、気体又は気体と液体の混合物は、少なくとも一部が電離できるように適宜決定する。又、ガス圧は、通常は大気圧付近であることが、操作が容易であることから好ましい。

このプラズマ発生器12を作動させて、気体又は気体と液体の混合物を電離して得られた殺菌因子を第1のチャンバー14内に蓄積する。有効量の殺菌因子を蓄積するために、プラズマ発生器12の電離混合物生成能力から予め設定された所定時間、プラズマ発生器12を作動させる。

一方、滅菌を行う第2のチャンバー18は、被処理物36を予め乾燥させる目的で内部を真空状態とする必要があり、耐圧構造を有している。第2のチャンバー18は従って、例えば、肉厚2m/mのステンレス鋼(sus304)等の肉厚材料で成形されることが好ましい。

被処理物36は、絶乾状態としてから滅菌処理することが処理効率の観点から好ましいため、第2のチャンバー18内に配置された被処理物36を配置する棚38上に被処理物36を適宜配置した後、チャンバー18を密閉し、排出パイプ40の経路に配置された真空ポンプ42を作動させて真空状態となし、通気バルブ46を開放して常圧にする操作を数回、好ましくは2〜3回繰り返して、被処理物36を十分乾燥させておく。この乾燥処理が終了したのち、第1のチャンバー14内に蓄積した殺菌因子を、連結管24の開閉バルブ20を開放して真空状態である第2のチャンバー18内に導入する。

こうして殺菌因子を被処理物36と接触させることにより被処理物36の滅菌が行われる。この第2のチャンバー18内が大気圧よりやや加圧状態(大気圧より最大1気圧までの陽圧)になるようにして操作することが、滅菌効果を高め、特に、厚みのある被処理物36の内部まで滅菌することができるという観点から好ましい。従って、必要に応じて連結管24に配置されたコンプレッサー22を作動させて第2のチャンバー18内に導入する殺菌因子を含む気体の圧力を高めたり、第2のチャンバー18に配置された圧力調整器16を作動させて第2のチャンバー18内の気圧が所定の範囲に保持されるように調整する。

このように被処理物36を配置するチャンバー18内は、前記のように大気圧よりやや加圧状態(本発明ではこの大気圧よりやや加圧状態とした状態を陽圧状態と称する)、例えば、2〜10mmH2 O、好ましくは2.5〜5mmH2 O程度の微陽圧として処理を行うことが、滅菌効果の向上、特に、厚みのある被処理体の内部まで滅菌することができるという観点から好ましい。また、チャンバー18内を陽圧にすることにより、チャンバー18内の無菌状態を維持することが容易となる。

処理が終了した後、第2のチャンバー18内に残存する気体の排気は、圧力調整器16を介して排出パイプ40により排出されるが、排気中に残存する殺菌因子による環境への影響を低減するため、排出パイプ40には、排気ガス分解装置(排気処理装置)44が配置され、オゾンなどの有害物質が分解処理された後、外気へと排出される。

本発明の滅菌装置で滅菌しうる被処理物には特に限定はないが、例えば、カテーテル、エクステンションチューブ、注射針などの医療材料包装体や食料品、医薬品など、高度の信頼性を要する被処理物に対して、好適に使用することができる。さらに、食品用又は薬品用包装のシートまたはロール、若しくは容器トレイ、ボトル等、天然繊維または合成樹脂繊維からなる織布または不織布、及び紙製品及びこれらの素材よりなる衣服類、ガーゼ、マスク、綿等の厚みのある物品などの滅菌処理にも有効である。

医療材料を包装する包装袋の構成としては、片面が中身を透視し得る透明フィルムであり、片面が不織布である包装袋が好適に用いられ、ここで使用される不織物の透気度が1000秒以下のグレートが滅菌効率上好ましい。

被処理物が包装材料である場合には、その形態は、例えば、袋、自立袋、成形容器、成形シート、ボトル等であることができる。本発明の方法は、食品、薬品等の無菌を要求する、例えばアセブチック用分野、及び衛生的に無菌を要求する分野へと応用範囲は広い。

滅菌できる細菌にも特に限定はない。本発明の方法によれば、例えば、大腸菌(E.coli)、サルモネラ・ティフィ(Sal.typhi)、枯草菌(B.subtilis)、黄色ブドウ球菌(Staphylococcus.aureus)、アスペルギルス・ニガー(Asp.niger)等の菌を滅菌することができる。

前記態様においては、第1のチャンバー14と第2のチャンバー18の容積はほぼ同じであるが、第1のチャンバー14の容積を第2のチャンバー18の容積の2倍以上とすることもできる。このように第1のチャンバー14の容積を大きくすることにより、連結管24の開閉バルブ20を開放すれば、第2のチャンバー18内に直ちに十分な量の殺菌因子が供給されるため、特にコンプレッサーなどの加圧装置を必要とせず、簡易な構成で好適な滅菌条件を達成することができる。

また、第1のチャンバー14も耐圧構造となし、且つ、被処理物36を配置するための棚38を設け、第2のチャンバー18にもプラズマ発生器12を備えることにより、例えば、第1のチャンバー14と第2のチャンバー18とで、殺菌因子の貯留と、予備乾燥処理、滅菌処理を交互に行うことができるため、処理効率を一層向上することができる。

前記した態様においては、殺菌因子が残存する排気は排気ガス分解装置(排気処理装置)44により処理された後、外気へと排出されるが、この殺菌因子が残存する排気を排出せずに、移送管を設けて第2のチャンバー18から第1のチャンバー14内へ移送して再利用することもできる。このように、殺菌因子が残存する排気を循環、再利用することは、環境及び処理効率の観点から好ましい。

【実施例】
以下本発明を実施例によりさらに説明する。
(実施例1〜3)
図1に示す滅菌装置を用いて、医療用エクステンションチューブの滅菌処理を行った。第1のチャンバー(殺菌因子貯留タンク)14は肉厚2m/mのステンレス鋼(sus304)で、半径0.25m、長さ0.6m、容積約0.12m3 のサイズの横円筒形に成形されている。第2のチャンバー(滅菌室)18は、耐圧構造を要するため、肉厚2m/mのステンレス鋼(sus304)で、第1のチャンバー14と同様の半径0.25m、長さ0.6m、容積約0.12m3 のサイズの横円筒形に成形されている。

プラズマ発生器12として、高電圧方形波パルス発生器(ピーク電圧16.4kV、波形:方形波、周波数:240Hz〜328.5Hz)を用い、さらに気体と液体の混合にはネブライザーを用い、パイプAから気体(O2 、供給量200リットル/分)と液体(35%H2 O2 )の混合物を電界内に供給した。また、液体の供給量は18g/hrであった。

第2のチャンバー18内には、被処理物36を配置し、予め2〜3回真空に引いて充分乾燥しておく。プラズマ発生器12を10分間作動させて第1のチャンバー14内に殺菌因子を貯留した後、連結管24の開閉バルブ20を開放して、真空にした第2のチャンバー18内に殺菌因子を移送する。第2のチャンバー18内が大気圧になった時点で加圧装置(コンプレッサー)22と圧力調整器16を稼働させ、第2のチャンバー18内の圧力を微陽圧2.5〜3.5mm/H2 Oに調整しながら滅菌処理をスタートする。

この状態で10分間保持した後、連結管24の開閉バルブ20を閉鎖し、加圧装置22を停止して、排気バルブ40に配置された真空ポンプ42を作動して排気し、第2のチャンバー18内を真空状態とした後、通気バルブ46を開放して大気圧に戻し、滅菌処理工程を終了する。

処理が終了した後の排気は、排気ガス分解装置(排気処理装置)44によって排気中に残存する殺菌因子や有害なオゾン等を除去された後、外気へと排出される。

本実施例では、被処理物としては、片面が透明フィルムで他の片面が不織布(透気度:1000秒以下のグレート)の包装体内に、枯草菌(バシルス・スブチリス:B.subtilis)の芽胞子(endspore)を1ピース当たり1×106 個になるように付着させたエクステンションチューブを密閉包装したものを用いた。同様の条件にて3つの被処理物を準備し、それぞれ同様の滅菌処理を行い、実施例1〜3とした。

滅菌処理後の被処理物に残存する菌数を以下の方法で算出した。結果を下記表1に示す。

第2のチャンバー18内が大気圧になった時点で加圧装置(コンプレッサー)22を稼働させず、第2のチャンバー18内の圧力を1気圧に保持した他は実施例1〜3と同様にして滅菌処理を行った。

(実施例5)
第2のチャンバー18内が大気圧になった時点で加圧装置(コンプレッサー)22を稼働させず、第2のチャンバー18内の圧力を1気圧に保持し、滅菌処理条件で15分間保持した他は実施例1〜3と同様にして滅菌処理を行った。

評価方法(残存胞子数検査)
殺菌試験に供した被処理物を、滅菌した界面活性剤0.2%トゥイーン80(Tween80:商品名、東京化成工業(株)製)を含む生理食塩水10mlに1時間浸漬後攪拌して、残存胞子を抽出した。得られた残存胞子抽出液を、標準寒天培地を用いて、35℃で72時間培養した。培養後、出現したコロニー数から1ピース当たりの残存胞子を算出した。結果を表1に示す。なお、表1中、未処理品とは、滅菌処理を行わず、同じ時間常温にて放置したものであり、残存胞子数は1.1×106 (胞子数/ピース)であった。

表1に明らかな如く、本発明の滅菌装置を用いることにより、滅菌処理開始後10分間という短時間で、実施例1〜3においては所望の滅菌処理が完全に達成された。また、実施例4においては大幅に細菌数が減少しているが、医療用具の滅菌度の観点からするとやや不完全なレベルであった。実施例4と同様の条件で滅菌処理時間を5分間延長した実施例5においては実施例1〜3と同様に所望の滅菌処理が完全に達成された。さらに、いずれの実施例においても被処理物であるエクステンションチューブの変質は見られなかった。

この結果より、本実施例における如き短時間の滅菌処理では、低いレベルの滅菌を必要とする被処理物については滅菌用の第2のチャンバーの内圧は特に制御を要さないが、医療用材料などの高度な信頼性を要する被処理物については、滅菌用の第2のチャンバーを陽圧にすることが好ましいことがわかった。

【発明の効果】
本発明の滅菌装置によれば、医療材料包装体等の高度の信頼性を要する物品の滅菌に好適に使用でき、安全で、被処理物を変質させることが少なく、信頼性の高い滅菌を効率よく行い得るという効果を奏する。
