【発明の属する技術分野】
本発明は、フローサイトメータに代表される粒子分析装置及び複数の焦点位置を有するレンズ要素を一体化した複合レンズに関するもので、より具体的には前方散乱光の散乱角度情報に基づいて細胞や血球等の粒子(試料)を分類することのできる装置に関する。

【従来の技術】
フローサイトメータは、セルソータやセルアナライザ、及び、それらを内蔵する粒子分析装置(血液分析装置)等があり、フローセル内を流れる粒子や、フローセルから落下する粒子に対して光を照射させ、そのときに発生する前方散乱光や側方散乱光等を検出し、その検出した光情報に基づいて細胞や血球等の粒子を分類するようになっている。

ところで、前方散乱光の中でも、粒子の種類によって散乱角度が異なることが知られている。そこで、前方散乱光を散乱角度の異なる複数の領域に分けてそれぞれ集光して測定することにより粒子の分類を行う装置が提案されている。一例をあげると、特開平8−271509があり、これは、白血球に含まれる各細胞を光学的に検出して含有率の少ない細胞の分類を精度良く行うための装置である。この公報に開示された装置における散乱光を検出する構造を示すと、図15及び図16,図17のようになっている。

図16,図17に示されるフローサイトメータに使用されるフローセルの断面図のように、フローセル2の内部は鞘状に流れるシース液2aの中心部分に試料(粒子)の流入する試料管2cが位置し、その先端部分でシース液に包まれる様に粒子を含んだサンプル流2bとなって細管部分へ順次供給される構造となっている。その細管部分ではレーザー光線aが照射されているため、そこを通過した粒子7が散乱源となって、前方散乱光b,c,d、側方散乱光e,f,g、(図の背面方向のため図示されていない)及び後方散乱光h,jを生じさせる。

図15で示されるように、光源1から出射されたレーザー光はフローセル2を透過後、移動不能に設置されたオブスキュレータ11で遮光される。そして前方散乱光b、c、d等は、フローセル2の前方に設置されたコリメータレンズ3により平行光束にされ、前方のミラー4に照射される。ミラー4には、中央に孔部4aが形成されており、前方散乱光のうち散乱角度が小さい光は、その孔部4a内を通過し、第1集光レンズ5により第1検出器6に集光される。また、前方散乱光のうち散乱角度が大きい光は、ミラー4の周囲に照射されそこにおいて反射されて光路が90度変更され、第2集光レンズ5′により第2検出器6′に集光される。それら第1,第2検出器6,6′は、フォトダイオード等の光電変換素子により構成され、受光した光の強度に応じた電気信号に変換し、分析装置9に送る。

つまり、第1検出器6は、散乱角度の小さい前方小角散乱光強度データを検出し、第2検出器6′は、散乱角度の大きい前方大角散乱光強度データを検出する。そして、分析装置9では、入力された各検出信号から前方小角散乱光強度データ及び前方大角散乱光強度データを算定して記憶し、かつ前方小角及び前方大角散乱光強度の2次元座標データを算定する。また、分析装置9は、算定された前方小角散乱光強度データと前方大角散乱光強度データを、順次外部の表示装置(図示せず)に出力し、画面上に2次元座標の光強度散乱分布図(スキャッタグラム)として表示させる。

なお、レーザー光照射時に散乱する散乱光には以下のような方向性が知られている。すなわち、粒子の大きさがレーザー光の波長に対して大きい場合は前方散乱光となり、粒子の大きさがレーザー光の波長と同程度の場合は前方及び側方散乱光となり、粒子の大きさがレーザー光の波長に対して小さい場合は前方、側方及び後方散乱光となることが知られている。

従って、実際の装置では粒子の分類をより正確に行うため、図16,図17に示すように前方散乱光b,c,dに対しては、コリメータレンズ3,孔部付きのミラー4,集光レンズ5をそれぞれ有する光学系8と各光強度を検出する検出器6を有している。また同様に側方散乱光e,f,gに対しても光学系8及び検出器6を有し、後方散乱光h,iに対しても光学系8と検出器6を有している。

また、別の装置としては、図18に示すようなものもある。図示する装置は、上記した孔部付きのミラー4により散乱角度の異なる複数の領域に分けるのではなく、光ファイバー10を用いて分離するようにしている。つまり、フローセル2内の粒子から発せられる前方散乱光が、光ファイバー10の受光面に照射されるようにしている。この受光面は、同図(B)に示すように、中心の前方小角散乱光受光用の光ファイバー群10aと、周囲の前方大角散乱光受光用の光ファイバー群10bを配置する。そして、同図(A)に示すように、光ファイバー群10aの出射側は、第1検出器6に接続し、光ファイバー群10bの出射側は第2検出器8に接続する。これにより、光ファイバー10の受光面に照射された散乱光は、散乱角度に応じて所定の光ファイバー群により、いずれかの検出器6,8に供給される。なお、第1,第2検出器6,8の出力は、図6と同様に分析装置に送られ、所定の信号処理が行われる。

【発明が解決しようとする課題】
しかしながら、上記した従来の装置では、以下に示す問題があった。すなわち、図15に示す装置では、各検出器6,6′に受光させるための光学系としてコリメータレンズ3、孔部付きミラー4並びに集光レンズ5が必要となり、部品点数が多いとともに、光学系の調整が高度で煩雑になる。さらに、専有面積も大きくなり、装置全体の小型化を阻害する。また、図16,図17のように分割する領域を増やし、さらに、側方散乱光及び後方散乱光を検出する装置では、「検出する散乱光の数−1」だけの孔部付きミラー、検出する散乱光の数と同じ集光レンズが必要となり、部品点数の増大を招き、上記した問題がより顕著となる。

一方、図18に示す装置では、コリメータレンズや集光レンズが不要となり、部品点数としては光ファイバーだけでよい。しかも、分割する領域が増えても、ファイバー束の数が増えるだけで、図15,図16,図17のように部品点数の増加や専有面積の増大もあまりない。しかし、各光ファイバー束の受光面を同心円上に配置するとともに、反対側の出射口では光ファイバー束単位で纏めなければならないので、光ファイバーの引き回し処理が煩雑で、分割数に限度がある。また、そもそも光ファイバーの場合には、ロスが大きい。つまり、光ファイバーを通過中のロスはもちろんのこと、クラッド部分に照射された光は伝送されないため、結局図2(B)に示す光ファイバー10の受光面に照射された光のうち、最終的に検知器に到達する光は50%以下となり、光の利用効率が悪い。

本発明は、上記した背景に鑑みてなされたもので、その目的とするところは、上記した問題を解決し、簡単な構成で異なる散乱角度の光を分離することができ、光学系の調整も容易に行え、光の利用率も高く、しかも分割する領域数が増えても、光を分離する光学系の部品点数も増えず専有面積も大型化を招くことがなく検出器の増加に伴うスペースの増加にとどめることができ、しかも、本発明の要部の構成となる複合レンズの姿勢あわせを容易かつ正確に行うことができ、測定精度の高い粒子分析装置及びそれに用いるのに適した複数の焦点位置を有するレンズ要素を一体化した複合レンズを提供することにある。

【課題を解決するための手段】
上記した目的を達成するために、粒子に光を照射することにより発生する散乱光のうち、異なる散乱角度のものを分離して検出することにより前記粒子の分類を行うようにした粒子分析装置において、前記散乱光の光路を遮るようにして配置された、焦点位置の異なる複数のリング状のレンズ要素を同心円上に位置させるとともに、中央部に位置決め要素を設けた状態で一体化された複合レンズと、前記複数のレンズ要素の焦点位置に応じた前記散乱光の結像位置にそれぞれ配置された各散乱角度検出用の検出器(実施の形態では第2,第3検出器17,18)と、前記位置決め要素を通過した光を検出可能な位置決め用検出器(実施の形態では、第1検出器16)と、前記複数の検出器の検出出力に基づいて、前記粒子の分類を行う分析手段とを備え、かつ、前記位置決め要素は、少なくとも前記複合レンズの中心と光軸とが一致するとともに、その光軸に対して前記複合レンズが直交している時に、前方に設置した前記位置決め用検出器の所定位置に、所定の光量の光を受光させることができるように構成した(請求項1)。

なお、各検出器を設置する焦点位置に応じた結像位置とは、必ずしも焦点位置ではない。すなわち、複合レンズに照射される散乱光は、平行光束ではないので、焦点位置に結像はしないからである。但し、散乱光の発生位置は、フローセル内を流れている粒子であったり、フローセルから落下する粒子であり、いずれにしても複合レンズとの相対位置関係が既知であるので、結像位置は一義的に決められる。

このようにすると、散乱光は、複合レンズの所定位置に照射される。すなわち、散乱角度の小さい前方小角散乱光は、複合レンズの中央側に位置するリング状のレンズ要素に照射され、散乱角度の大きい前方大角散乱光は、複合レンズの周囲に位置するレンズ要素に照射される。そして、各レンズ要素は、焦点位置が異なるので、各レンズ要素に照射された光は、それぞれ異なる位置に結像され、検出器で検出される。

さらに請求項1に記載の発明では、複合レンズの中心に位置決め要素を設けたため、複合レンズの姿勢が所望の状態になったか否かは、位置決め用検出器の出力を監視することにより、簡単に判別できる。よって、位置決め・姿勢制御も容易に行える。

なお、所定の光量とは、一定の基準以上の光量をいい、係る一定の基準とは、位置決め要素としてピンホールを用いたもののように、基準値自体が0より大きい所定の値の場合もあれば、検出器にピンホール付きのフォトダイオードを用い、かつ、位置決め要素として凸レンズを用いた場合のように、基準値が0となりその0よりも大きいか否か(受光があるか否か)を判断する場合も含む。

そして、上記した各粒子分析装置の各構成要素をより具体的に限定するとすれば、以下のようにすることができる。すなわち、まず、位置決め要素としては、請求項1の条件を満たすものであれば各種のものを用いることができるが、例えば凸レンズとしたり、ピンホールとすることができる。

そして、前記複合レンズは、通常のレンズでもよいが、フレネルレンズから構成するようにすると、設計並びに製造が容易に行え、取り扱いも簡便となるので好ましい。

また、焦点位置を異ならせるための構造としては、例えば焦点距離の異なる複数のレンズ要素を接合するようにしたり、主軸の異なる複数のレンズ要素を接合するようにしたり、それらを組み合わせるようにするなどの他、各種の構造を採ることができる。

さらに、前記複数のレンズ要素の境界部分及びまたは位置決め要素とレンズ要素の境界部分を環状のマスク材で覆うようにしてもよい。このようにマスク材で覆うと、安定した組成のレンズ要素の部分の光に基づいて集光されるので、検出器で検出した信号に雑音成分が乗りにくくなり、より高精度な測定が可能となる。

さらにまた、前記粒子が供給されるフローセルと、前記複合レンズの間の所定位置に、光源から出射した直接光を遮光する遮光部材を設け、かつ、前記遮光部材は、前記位置決め要素を用いて前記複合レンズの調整を行う際には待避可能としてもよい。つまり、測定時には光量の大きい光源からの直接光と前方散乱光が一緒に検出器に受光されると、前方散乱光の情報が打ち消されてしまう。そこで通常フローサイトメータでは、所定位置に遮光部材を設置し直接光が複合レンズに照射しないようにしている。一方、複合レンズの姿勢を調整している時には、複合レンズの中央の位置決め要素に光を照射しなければならないが、上記遮光部材を設置しておくと位置決め要素に光を照射できない。そこで、遮光部材を待避可能にし、複合レンズの調整時には、遮光部材を待避させ、位置決め要素に光を照射可能とすることにより、位置決めと測定をともに確実に行えるようにするとよい。

なお、この待避は、例えば遮光部材をスライドレールなどに取り付け、そのスライドレールに沿って移動させるようにしてもよい。この場合には、モータなどの駆動手段に連結し、自動的に行うことができる。また、手動により設置位置から取り外すようにしてもよい。この手動による取り外しの一例を示すと、例えば遮光部材を複合レンズの装着用のホルダに着脱自在に取り付け可能とし、位置決め時にはホルダから取り外し、測定時にはホルダに装着することにより対応できる。なお、本発明では、直接光の光路上には、位置決め要素が位置しているので、直接光の広がりが位置決め要素よりも小さい場合には、レンズ要素には直接光が照射されることなく前方散乱光が照射されることがないので、たとえ直接光の光強度が大きい場合であっても必ずしも遮光部材が必要となるわけではない。

また、本発明に係る複合レンズとしては、例えば、主軸が平行にずれている複数のレンズ要素を同心円状に配置し一体化することを前提とし、前記複数のレンズ要素が同心円状に配置された中央部に位置決め要素を設けるようにしたり(請求項2)、前記レンズ要素と前記位置決め要素の境界部分をマスク材で覆うようにしたり(請求項3)、それらを組み合わせて構成したり(請求項4)するとよい。

【発明の実施の形態】
図1は、本発明に係る複合レンズ15の一例を示している。同図に示すように、本例では、中心に位置決め要素としての凸レンズ15aを有し、その周囲に同心円状に2つのリング上の第1,第2のレンズ要素15b,15cを位置させた状態で接合し一体化することにより複合レンズ15が形成されている。

本例では、両レンズ要素15b,15cは、いずれもフレネルレンズを適宜形状に切断して形成している。そして、係る凸レンズ15a並びに両レンズ要素15b,15cを嵌め合わすとともに、接合面を接着一体化することにより、1枚の複合レンズ15を形成するようにしている。さらに、凸レンズ15aと第1,第2レンズ要素15b,15cは、それぞれ焦点位置が異なるようにしている。これにより、同一の物点から発せられた光は、凸レンズ15a並びに第1,第2レンズ要素15b,15cにて集光されるが、結像される位置は異なる。そして、焦点位置を異ならせる具体的な構造としては、各レンズ要素15a〜15cを異なる焦点距離のものを用いたり、主軸が異なるようにすることにより実現できる。

図2は、上記した複合レンズを用いた本発明に係る粒子分析装置(フローサイトメータ)の前方散乱光に関する一実施の形態を示している。同図に示すように、レーザー光源1から出射された光の光路上にフローセル2,複合レンズ15の順に配置している。なお、図示省略するが、レーザー光源1の出射側には、集光レンズが配置され、フローセル2の設置位置で収束するようになっている。また、フローセル2内の粒子にレーザー光が照射されると、前方散乱光が発生する。そして、その前方散乱光の進路の中心(光軸)と複合レンズ15の中心とを一致させている。しかも、その光軸と、複合レンズ15とは直交するように配置する。

さらに、複合レンズ15を構成する各要素15a〜15cは、その焦点距離が中央の凸レンズ15aが最も短く、最外周の第2レンズ要素15cが最も長くなるようにしている。但し、各要素15a〜15cの主軸は一致させている。これにより、同一の物点から出射された光は、各要素15a〜15cを通過することにより集光されるが、その結像位置は異なる。具体的には、いずれも複合レンズ15の中心線(光軸)上に結像されるが、凸レンズ要素15aの結像位置が最も複合レンズ15側になり、第2レンズ要素15cが複合レンズ15から最も離れた位置に結像される。

一方、複合レンズ15の前方には、第1検出器16,第2検出器17,第3検出器18の順に配置している。この時、各検出器16〜18並びに複合レンズ15の中心が同一線上に位置するようになっている。つまり、レーザー光源1から出射された光の光路の中心軸上に、各検出器16〜18の受光面の光軸が一致するように配置されている。そして、第1検出器16は、焦点距離が最も短い凸レンズ要素15aを通過した光の結像位置に配置される。第2検出器17は、焦点距離が中間の第1レンズ要素15bを通過した光の結像位置に配置される。第3検出器18は、焦点距離が最も長い第2レンズ要素15cを通過した光の結像位置に配置される。

さらに、各検出器16〜18は、フォトダイオード(PD)その他の光電変換素子により構成される。そして、各検出器16〜18の出力は、分析装置9に与えられ、そこにおいて粒子の分析が行われる。さらに、第1検出器16は、受光面の中心に一致するようにピンホールを設け、中心からずれた光は検出しないようにしている。また、第2,第3検出器17,18の出力に基づく分析結果が、出力装置19に出力される。なお、異なる散乱角度からの信号に基づいて行う分析・分類処理は従来と同様に行えるため、その詳細な説明を省略する。

次に、上記したフローサイトメータを用いた測定手順について説明する。測定に先立ち、複合レンズ15の姿勢を調整する位置決め処理と、その位置決め処理が終了後に行う実際の分析・分類処理がある。そして、位置決め処理は、測定の都度行ってもよい。また、何回かに1回の割合で定期的あるいは不定期に調整を行うようにしてもよい。

*位置決め処理
位置決めは処理は、フローセル2に散乱源となる試料を供給しない状態で行う。散乱源がないので、レーザー光源1から出射されるレーザー光は、フローセル2をそのまま通過し、その多くは複合レンズ15の中央の位置決め要素たる凸レンズ15aに照射され、所定位置に結像する。この時の結像位置は、複合レンズ15がフローセル2,レーザー光源1に対して前後方向にずれた位置にあると、第1検出器16の受光面で結像しない。また、前後方向の位置があっていても、光が複合レンズ15(凸レンズ15a)の中心に照射されないと、第1検出器16の設置位置では結像するものの、複合レンズ15のずれ方向・距離に応じて第1検出器16の受光面の中心から位置ずれをした位置に結像する。すると、本形態では、第1検出器16の受光面の前面には、ピンホールが設けられており中心軸に一致する光のみが通過することができるので、上記のように位置ずれをした光はピンホールを通過できず、第1検出器16に受光面には到達しない。同様に、光が複合レンズ15(凸レンズ15a)の中心に照射されたとしても、複合レンズ15が傾いているとすると、光は凸レンズ15aの中心に斜めに照射されることになり、そこで屈折して第1検出器16の受光面の中心からずれた位置に結像する。よって、ピンホールにより遮光され第1検出器16の受光面に照射されない。

従って、複合レンズ15が、その中心が光軸上にあり、かつ、直交するように設置されており、しかも、前後方向の位置が所望の状態になるように設置されると、凸レンズ15aで結像される光が、第1検出器16の受光面に照射される。その結果、第1検出器16の出力を監視しながら、複合レンズ15の姿勢を調整する。そして、第1検出器16の出力がON(受光あ)の時が、複合レンズ15が正しい状態で設置されたことになるので、その状態に複合レンズ15の姿勢を保持し、位置決め処理を終了する。

そして、図面を書く上での便宜上、凸レンズ15aの径を比較的大きく表示したが、位置決め処理を行うためには、第1検出器16の中心軸上に光が結像するか否かを判断できればよいので、凸レンズ15aの径も小さくすることができる。そして、後述する実際の測定の際には、試料の吸光度を測定する場合を除き、その凸レンズ15aの部分に照射される光は使用されないので、測定時の光の有効利用の観点からも凸レンズ15aの径は小さい方が好ましい。

なお、実際には複合レンズ15は、フローサイトメータ内の所定位置に設置されたホルダーなどに固定されるため、おおよその位置並びに姿勢はあっており、しかも、少なくとも1つの方向(例えば光軸に対する前後方向など)の位置決めはできている場合がある。係る場合に、残りの方向に対する調整を行うことになる。

また、上記した実施の形態では、第1検出器16として、ピンホール付きのフォトダイオードを用いた例を説明したが、本発明はこれに限ることはなく、例えば、位置検出フォトダイオード(PSD:position sensitive detector)等を用い、受光面の中心で結像されているか否かを判断するようにしてもよい。

なおまた、本形態では、試料を供給しなくても位置決めができるため、あえて試料を供給しないで行ったが、試料を供給しながら位置決めしてももちろんかまわない。なお、上記した説明の方法を用いた方が、散乱がないため、より多くの光を位置決め用として利用できるので好ましい。

分析処理をする際には、第2,第3検出器17,18の出力を用いて行う。つまり、図からも明らかなように、散乱角度の小さい前方小角散乱光(例えば1,5度から10度)は、第1レンズ要素15bに照射され、そこにおいて集光されて複合レンズ15から2番目に位置する第2検出器17に受光される。さらに、散乱角度の大きい前方大角散乱光(例えば10度から15度)は、最外周の第2レンズ要素15cに照射され、そこにおいて集光されて複合レンズ15に最も遠い第3検出器18に受光される。

このように、1つの複合レンズ15により複数の異なる散乱角度の領域を分離して集光することができる。つまり、従来であれば、孔部付きのミラーやコリメートレンズや複数個の集光レンズにより構成された光学系が、1個の複合レンズで構成することができ、構成が簡略化されるとともに、占有面積も小さくてすみ、しかも調整も容易となる。しかも、分離する領域の数が増加しても、レンズ要素の数を増やすだけでよいので、大型化することもない。さらに、レンズで集光するだけであるので、前方散乱光のほぼ全部を各検出器に受光させることができ、光の利用率も高く、微小な光であっても確実に検出することができる。

図4は前方散乱光に加えて、側方散乱光及び後方散乱光を利用したフローサイトメータに本発明を実施した態様を示している。同図においてレーザー光源1から発せられた照射光aは、後方散乱光を分離するための複合レンズの中心部分を通過してフローセル2の散乱源である粒子7に照射され、前方散乱光b、c、d、側方散乱光e、f、g及び後方散乱光h、iを得ている。各散乱光は、それぞれの方向に1つずつ配置された複合レンズ15により分離され、それぞれの検出器6により光の強度が検出される構造となっている。またレーザー光源1からフローセル2を通過した直接光は位置決めのための検出器6aにて検出されるごとく構成されている。本実施の形態は、図16,図17に示したフローサイトメータの従来例に本発明の複合レンズ15を適用した例である。前記従来例では検出すべき散乱光の数が増加するにつれてコリメータレンズ3、ミラー4及び集光レンズ5からなる光学系8の数が増加し、装置の小型化を阻害する要因となっていたが、本発明の複合レンズ15を用いるならば、光学系8を1つの複合レンズに置き換えることが可能となり、小型で安価なフローサイトメータの供給を可能とすることができる。さらには従来はコリメータレンズ3、ミラー4及び集光レンズ5それぞれの光軸を調整する必要があったものが、複合レンズ15単体の調整を行えばすみ、装置を用いた測定が極めて容易となるメリットを有している。さらには複合レンズをモールドで製作する場合には、1枚の複合レンズの中でのレンズ要素ごとの焦点位置のばらつきを本質的に無くすことができる。

図5は、上記した第2の実施の形態で用いられる複合レンズ15の製造方法の一例を示している。第2の実施の形態では、第1,第2レンズ要素15b,15cの焦点距離は等しくし、主軸のみが異なるようにしている。従って、図中二点鎖線で示すように、大きなフレネルレンズからなる母レンズ20を用意し、その母レンズ20の主軸から所定距離ずらした位置に中径のリング状に繰り抜くことにより、第1レンズ要素15bを形成する。この第1レンズ要素15bの主軸は、母レンズ20の主軸であるので、リングの中心O1 に対して図中左側にずれた位置となる。また母レンズ20の主軸から上記第1レンズ要素15bと反対側に所定距離ずらした位置に大径のリング状に繰り抜くことにより、第2レンズ要素15cを形成する。この第2レンズ要素15cの主軸は、母レンズ20の主軸であるので、リングの中心O2 に対して図中右側にずれた位置となる。従って、第1レンズ要素15bと第2レンズ要素15cの主軸はそれぞれ中心から所定距離だけ離れた位置となり、しかも両レンズ要素15b,15cのずれ方向は逆になる。このように1枚の大きなフレネルレンズの適宜位置を繰り抜くことにより、複数のレンズ要素を製造することができ、それらを接合することにより容易に複合レンズを製造できる。そして、係る製造プロセスは、接合するレンズ要素の数が増加するにつれて、同一の焦点距離のレンズ要素を作る上で効果的となる。なお、このように必ずしも1枚のフレネルレンズから各レンズ要素を製造する必要がないのはもちろんである。

すなわち、このレンズは所定の面積を有する複数の領域に照射された光をそれぞれその領域ごとに焦点させる機能を持たせることを目的とするため、フレネルレンズなど、装置の設計に合った焦点距離を有する複数のレンズを複数枚用意し、それらのレンズから所定の形状・面積を有するように複数枚切断またはくり抜いてレンズ要素を成形し、それらのレンズ要素を一体的に、平面状に接合させることで製造することができる。用意した複数のレンズの焦点距離はそれぞれ異なる距離であってもよく、レンズ要素の主軸は、一致させる設計としても、ずらした設計であってもよい。このような成形により、限られたスペースにおいてコンパクトに光学系ユニットを製造することができる。

なお、複合レンズ15の製造方法としては、上述の方法のほか、予め各レンズ要素ごとあるいはそれらを組み合わせて一体にした金型の表面を作成し、その金型を用いて圧縮成型、ICM成形(インジェクション・コンプレッション・モールド)あるいは射出成形などの技法で製造することもできる。図6〜図8により、その製造の一例を次に示す。

図9は、本発明の別の実施の形態であり、複合レンズ15の改良を図ったものである。すなわち、この形態でも第1,第2レンズ要素15b,15cの2つの領域を分離検出するものである。そして、隣接するレンズ要素間の接合領域並びに凸レンズ15aと第1レンズ要素15b間の接合領域を、リング状のマスク材22で覆うようにしている。このマスク材22は、各要素15a〜15cを接合した後、その接合部位を光学的に不透明な材質を装着することにより形成される。そして、具体的には、所定の薄膜を蒸着・スパッタした後パターニングしたり、所定のインク等を塗布したり、テープ材等を張り付けることにより実現できる。

つまり、マスク材22がないと、接合部分では、光が乱反射などして所定の検出器に正しく集光できなかったりして雑音成分となるおそれがある。しかし、本形態のように、マスク材22を設けることにより、正常なレンズ要素部分のみを光が透過するので、所望の状態で検出器に受光させることができる。そして、係るマスク材22を設けた複合レンズ15は、上記したいずれの実施の形態にも適用できる。

なお、上記した各実施の形態では、位置決め要素として凸レンズ15aを用いた例を説明したが、本発明はこれに限ることはなく、例えば複合レンズ15の中心にピンホールを設け、そのピンホールを位置決め要素として使用するようにしてもよい。

すなわち、ピンホールにすると、例えば複合レンズ15の中心とレーザー光源1から出射される光の光軸とが一致しないと、係る光はピンホールを通過できず、第1検出器の受光面に照射されない。また、複合レンズ15が斜めに配置されると、ピンホールを通過する光の光量が減少し、第1検出器16の出力が小さくなったり、さらには傾斜角度が大きくなると、ピンホールを通過できなくなる。よって、第1検出器の出力を監視することにより、複合レンズ15の姿勢制御が行える。具体的には、光量が一定の値以上となった時や、光量が最大になった時に位置決めが行われたと判定することができる。但し、このピンホールを用いた場合には、光軸方向に沿った前後方向の位置ずれには対応できない。よって、係る前後方向の位置決めが不要な場合には、複合レンズの中央にピンホールを設けるといった簡単な構成で形成できるというメリットが生じる。

また、上記した実施の形態では、いずれも2個のレンズ要素を組み合わせて複合レンズを構成したが、使用するレンズ要素は3個以上でもよい。また、レンズ要素ひいては複合レンズは、上記した各実施の形態では、フレネルレンズを用いて構成したが、本発明はこれに限ることはなく、凸レンズなどの通常のレンズを用いて構成することもできる。

さらにまた、図示省略するが、上記した各実施の形態において、フローセルと複合レンズの間の所定位置に、遮光部材たるオブスキュレータ(図15に示した符合11と同様の機能のもの)を設置し、レーザー光源1から出射した直接光をそこにおいて遮光するようにしてもよい。但し、そのオブスキュレータは、移動可能となっており、通常の測定の際には、直接光の光路上に位置するが、複合レンズの姿勢の調整を行う際には、その光路上から待避させ、レーザー光源1からの直接光が、位置決め要素15aに入射可能としている。また、遮光部材としては、従来のオブスキュレータのようなものに限らず、複合レンズ15に直接または間接的に取り付けるものであってもよく、その構造は任意である。

【発明の効果】
以上詳細に説明したように、本発明では、焦点位置の異なる複数のレンズ要素を接合一体化して1個の複合レンズを製造し、その複合レンズによって散乱角度の異なる光を分離し、各検出器に振り分けることができる。しかも、1枚の複合レンズという単体で行えるので、部品点数の削減並びに光学系の調整も容易に行える。さらに、レンズで集光されるだけであるので、光の利用率も高くなる。さらにまた、分割する領域数が増えても、レンズ要素が増えるだけで、部品としては1個の複合レンズでよく、光を分離する光学系の部品点数も増えず専有面積も大型化を招くことがなく検出器の増加に伴うスペースの増加にとどめることができる。

しかも、複合レンズの中心に、凸レンズやピンホールなどの位置決め要素を設けた場合には、その位置決め要素を光が正常に通過するか否かにより、複合レンズが正しい位置・姿勢になっているか否かを判断でき、位置決め処理を簡単かつ正確に行うことができる。

The present invention relates to a compound lens in which a particle analyzer represented by a flow cytometer and a lens element having a plurality of focal positions are integrated, and more specifically, based on scattering angle information of forward scattered light. The present invention relates to an apparatus capable of classifying particles (samples) such as blood cells.

Flow cytometers include cell sorters, cell analyzers, and particle analyzers (blood analyzers) that incorporate them. The flow cytometer irradiates particles flowing in the flow cell or particles falling from the flow cell with light. Forward scattered light, side scattered light, and the like generated in the above are detected, and particles such as cells and blood cells are classified based on the detected light information.
