Abstract
【解決手段】 バイオマス処理方法に関し、バイオマスを、水素生成菌による水素発酵処理をして水素を回収する。さらに、水素発酵処理後に発生する発酵液をメタン菌によるメタン発酵処理をしてメタンを回収する。この場合において、発酵液には少なくとも有機酸が含まれている。
Description
以下、本発明の第1の実施の形態に係るバイオマス処理方法について、詳しく説明する。しかしながら、この出願の発明は、以下の実施の形態によって、限定されるものではない。
(1)水素生成菌の前培養
300mlの三角フラスコに0.1%のブドウ糖を加えたPY培地100mlに水素生成菌を接種し、37℃の嫌気性グローブボックス(米国ホーマー社製、形式1024)内で一晩培養した。なお、PY培地の組成は、1Lの水で、10g peptone、5g yeast extract、500mg L-cystein HCl、8mg CaCl、8mg MgSO4 、40mg KH2 PO4 、400mg NaHCO3 、80mg NaClであり、炭素源は含まない。
3000mlの三角フラスコに水素発生源の澱粉またはブドウ糖を7.5g含むPY培地1400mlと水素生成菌の前培養菌液100mlを加え、ガスの排出口、pHコントローラ(東京理科機製、形式FC-10)の差込口、pH調整用NaOH液流入口等をつけたゴム栓をした後、嫌気性グローブボックスの外に出した。37℃の恒温水槽内で保温し、マグネティクスターラで攪拌をした。
発生した気体(ガス)は、10% NaOH液に通して二酸化炭素、その他のNaOH液溶解性ガスを除いたガスを水上置換法でメスシリンダに集め定量した。高速ガスクロマトグラフィによる分析と燃焼試験の爆発音から水素ガスであることを確認した。
(1)メタン発酵処理装置
メタン発酵処理は、UASB(upflow anaerobic sludge blanket)法で行った。実験に使用した処理装置は、水深930mmで内容量10Lの円筒状で底部に緩やかに攪拌できる攪拌装置が付属されている。この処理装置の下部には、流入口が設けられていて、また上部には越流堰が設けられていて、発生ガスは処理装置の上部から取り出せる構造になっている。
生活廃水処理施設の返送汚泥7.6Lをメタン発酵装置に投入し、定量ポンプで流入口から水素発酵後の発酵液(廃液)を流入させメタン発酵処理を開始した。発酵液(廃液)の流入量は、0.3L/日、または1L/日に設定し、1日に2回、1回あたり2時間定量ポンプを稼動させて間欠流入させた。処理開始後約5ヶ月間連続して処理を行った。この間、15日経過した時点でガス発生量がほぼ安定したことを確認したので、この時点からほぼ毎日ガス発生量を測定した。また、3ヶ月経過した時点からほぼ1週間おきに処理水(最終廃液)を採取して分析を行った。
メタン発酵処理の効果を判定するために、水素発酵処理後の発酵液(廃液)と、メタン発酵処理後の処理水(最終廃液)における、生化学的酸素要求量(BOD)、化学的酸素要求量(COD)、全有機性炭素(TOC)、無機性炭素(IC)、ケルダール窒素(kje-N)、全りん(T-P)、蒸発残留物(TS)、強熱減量(VM)、溶解性物質(DM)、浮遊物質(SS)、有機性浮遊物質(VSS)、アルカリ度、pHおよび有機酸をそれぞれ測定した。これらの測定は、従来の下水試験方法にしたがって行った。CODは、酸性条件下で、100℃における過マンガン酸カリウムによる酸素消費量を求めた。また、TOCおよびICの測定には、TOC計5000A(株式会社島津製作所)を、有機酸の測定には、ガスクロマトグラフィGC-14B(株式会社島津製作所)を使用した。
(1)スターチ(澱粉)の水素発酵処理
澱粉を7.5g含むPY培地1400mlと水素生成菌(Clostridium beijerinkii AM21B 株)の前培養菌液100mlを3000mlの三角フラスコに加え、37℃の恒温水槽内で培養を開始した。保温を開始して2時間から3時間経過したころから発泡が観察され、7時間から8時間頃に発生するガス量は最高に達した。10数時間でガスの発生は収束した。発生した水素ガスは、総計で3750mlであった。水素発酵後の培養液である発酵液(廃液)は、そのまま無処理でメタン発酵槽に注入した。
内容量10Lの円筒状のUASB法のメタン発酵処理装置に、水素発酵後の発酵液(廃液)を流入させて処理を開始した。流入量は、0.3L/日に設定し、1日に2回、1回あたり2時間定量ポンプを稼動させて間欠流入させた。したがって、滞留時間は33日となった。
上記(1)の水素発酵処理で発生した発酵液(廃液)の水質性状と、(2)のメタン発酵処理で発生した処理水(最終廃液)との水質性状をそれぞれ比較して表1に示す。表1に示す通り、水素発酵処理で発生した発酵液(廃液)にメタン発酵処理を行うことで、上記の通り水素ガスおよびメタンガスを回収できると共に、TOCやBOD、COD等を効果的に除去することもできることを確認した。このため、最終廃液は、下水放流の基準値を下回るため、処理の手間を減らすことができた。
(1)ブドウ糖の水素発酵処理
PY培地をイオン交換水で5倍に希釈した希釈PY培地にブドウ糖7.5gを溶かした培養液1400mlと水素生成菌(Clostridium beijerinkii AM21B株)の前培養菌液100mlを3000mlの三角フラスコに加え、37℃の恒温水槽内で培養を開始した。保温を開始して2時間経過した頃から泡が発生し、7時間頃に発生するガス量は最高に達し、10数時間でガスの発生は終息した。発生した水素ガスは、総計で5750mlであった。水素発酵後の発酵液(廃液)は、そのまま無処理でメタン発酵槽に注入した。
水素発酵後の発酵液(廃液)の注入量は、1L/日に設定し、1日に2回、1回あたり2時間定量ポンプを稼動させて間欠流入させた。したがって、滞留時間は10日であった。
上記(1)の水素発酵処理で発生した発酵液(廃液)の水質性状と、上記(2)のメタン発酵処理で発生した処理水(最終廃液)の水質性状をそれぞれ比較して表2に示す。表2に示す通り、水素発酵処理で発生した発酵液(廃液)にメタン発酵処理を行うことで、上記の通り水素ガスおよびメタンガスを回収できると共に、TOCやBOD、COD等を効果的に除去することもできることを確認した。このため、最終廃液は、河川放流の基準値および下水放流の基準値共に下回るため、最終廃液の処理の手間を減らすことができた。
以下、本発明の第2の実施の形態について、図1から図3に基づいて説明する。図1は、本実施の形態に用いられる水素生成装置10の構成を示す側断面図である。この図において、水素生成装置10は、反応容器11を有している。反応容器11には、水素生成用の材料を投入するための材料投入口12aが設けられている。なお、図示はしていないが、投入する材料が固形物の場合、材料投入口12aの部分に粉砕器が設けられる。また、本実施の形態では、上方に開口する材料投入口12aが、径の大きな配管状部材12の上端に形成されている。この配管状部材12は、反応容器11の上端面に接続されている。有機材料は、配管状部材12から反応容器11の内部に投入可能としている。
以下、本発明の第3の実施の形態について、図4および図5に基づいて説明する。なお、本実施の形態においては、上述の第2の実施の形態で述べたものと同一の構成については、同一の符号を用いて説明する。
以下、本発明の第4の実施の形態について、図6および図7に基づいて説明する。なお、本実施の形態は、上述の第3の実施の形態で述べた構成に加えて、メタン生成機構41を備えた、水素生成装置40に関するものである。以下、上述の第2の実施の形態、および第3の実施の形態で述べたのと、同一の構成については、同一の符号を用いて説明する。また、本実施の形態では、メタン生成機構41以外にも、後述するように、固体排出物を処理するための固形物最終処理槽44を具備した構成について説明する。

Claims (7)
- バイオマスを、Clostridium beijerinkii AM21B株、Clostridium sp.No.2株、Clostridium sp.X53株のうちの少なくとも1つを含む水素生成菌により水素発酵処理をして水素を回収し、さらに水素発酵処理後に発生する発酵液をメタン菌によるメタン発酵処理をしてメタンを回収すると共に、上記発酵液には少なくとも有機酸が含まれていることを特徴とするバイオマス処理方法。
- 前記バイオマスには、食品関連廃棄物および/または生活関連廃棄物が含まれていることを特徴とする請求項1記載のバイオマス処理方法。
- 前記水素発酵処理により回収された前記水素は、圧縮処理により液体状に設けられることを特徴とする請求項1または2記載のバイオマス処理方法。
- 前記水素発酵処理により回収された前記水素は、カーボンナノチューブによって吸蔵されることを特徴とする請求項1または2記載のバイオマス処理方法。
- 前記水素発酵処理により回収された前記水素は、水素吸蔵合金によって吸蔵されることを特徴とする請求項1または2記載のバイオマス処理方法。
- 前記メタン発酵処理により回収された前記メタンは、圧縮処理により液体状に設けられることを特徴とする請求項1から5のいずれか1項に記載のバイオマス処理方法。
- 前記メタン菌として、Methanobacterium属、Methanococcus属、Methanosarcina属、Methanosaeta属、Methanohalophillus属に属する細菌のうちの少なくとも1つを用いて前記メタン発酵処理を行うことを特徴とする請求項1から6のいずれか1項に記載のバイオマス処理方法。
