Description

【発明の詳細な説明】 本発明はニコチン含有流動媒質を含む経口投与用カプ
セルに関する。
このタイプのカプセルは既に知られている（***特許
公開公報第3,606,892号）。このカプセルの狙いは、カ
プセルを服用した人の口中で、その内容物が浸透圧によ
りカプセル壁の微細な穴を通って徐々に、かつ連続的に
浸出するようにすることであるが（第13頁、第１節参
照）、これには成功していない。
この公報はカプセル中のニコチン量を記載しておら
ず、添加剤についても、媒質のpHについても記載してい
ない。
本発明の目的は、カプセルを服用した人に喫煙した時
と同様な量及び速度で、ニコチンを吸収させるところ
の、上記したタイプのカプセルを提供することにある。
本発明によれば、咀嚼でカプセルを破壊し、ニコチンの
量を0.1〜10mg/個、好ましくは0.1〜3mg/個、特に２〜5
mg/個とし、風味及び耐久安定性を改善する添加剤を含
む媒質である溶液を使用し、さらに媒質のpHを６〜10、
好ましくは７〜９、特に６〜８にすることにより、上記
の目的が達成される。
このタイプのカプセルを服用して噛み砕くと、その内
容物は口腔粘膜に流れ出し、ニコチンは前記粘膜を経て
体内に入る。カプセルを服用した人は、ニコチンが迅速
に脳に達するので、速くて強いニコチン投与を体験す
る。吸収されるニコチンの量は、予め設定されたカプセ
ルのニコチン含量及び媒質のpHによって決まる。このニ
コチン含量は喫煙者が喫煙によって吸収するニコチン量
に匹敵する。
添加剤はニコチンの不快な収斂味を改善することを保
証する。
ニコチン含有流体が口中に暫時留まることは重要であ
る。このことにより、3.0mgのニコチンを含有し、pHが
8.0である溶液1.5mlから、1.5mgまでのニコチンが５分
以内に吸収される。この量は普通の強さのタバコを禁煙
した時の吸収量にほぼ相当する。溶液に残るニコチン
は、カプセルを服用した人が決める一定時間後に、吐き
出されるか飲み込まれる。飲み込んだ場合には、口腔内
でのニコチンが吸収される場合とは対照的に、血中及び
組織内のニコチンが付加的に徐々に増大し、その結果、
初期効果が増進する。
***特許公開公報第3,241,437号は、口腔用のニコチ
ン含有スプレーを開示する。しかし、スプレーによるニ
コチンの投与は余り快適でない。
英国特許第1,528,391号は、口腔粘膜に作用するニコ
チンの投与量が0.1〜10.0mgの範囲にあることを開示す
る。しかし、この量は口内に噴霧させるエーロゾルに関
係する。また、風味及び耐久安定性を改善するために添
加剤も使用されておらず、媒質のpHについても全く開示
されていない。
***特許公開公報第3,401,763号は、鼻に投与するニ
コチン含有媒体を開示する。鼻の粘膜は口の粘膜より敏
感であるので、鼻への投与は不快感を伴う。加えて、媒
体が鼻孔に残ることを保証しない。
***特許公開公報第3,438,284号は、ニコチンを放出
するデポプラスターを開示する。しこし、これでは、ニ
コチンを迅速に投与するとができない。また、ニコチン
の吸収は制御できず、極めて緩慢にしか生起しない。
「皮膚を通してのニコチンの投与」［「薬物及びアル
コール依存症」第13号（1984年）、第209〜213頁参照］
には、皮膚を介して作用するニコチン媒質が記載されて
いるが、この場合もニコチンの迅速な吸収は実現できな
い。
所望の薬剤を含有するカプセルはまた公知である（F.
グスティナー著、「薬品成形技術の手引き」1973年、第
177〜181頁参照）。しかし、この場合のカプセルはニコ
チンを含有していないい。
噛むことのできるカプセルは、B.ヘルウィッグ著「最
近の薬」第815頁（1980年）にも記載されているが、こ
のカプセルもニコチンを含んでいない。
喫煙に際して吸収されるニコチンが病原性でないこと
は、科学的に認定されており、特に所謂喫煙者疾病の進
行に、ニコチンが寄与しないことが認められている。本
発明の媒質は確実に禁煙を実行させるために使用するこ
とができる。
ニコチンは神経系及び精神植物性機能に対して様々な
影響を及ぼす。こうしたニコチンの精神組織栄養的及び
向神経的特性は、今日まで喫煙者だけに活用され、医薬
には利用されていない。この本質的理由は、恐らく、喫
煙するだけで、あるいは噛タバコを服用するだけで、か
なり多量のニコチンが容易に吸収されるためである。チ
ューイングガム、皮膚用プラスター又は鼻用スプレー
に、ニコチンを導入する試みは、禁煙したいと望む喫煙
者向けに限定されている。しかし、この試みはニコチン
の吸収が遅すぎるとか、血中のニコチン濃度が適当な値
に達しないとか、あるいはニコチンの投与に不快な刺激
や面倒を伴うために、満足な成果を収めていない。本発
明のカプセルは簡単な手段で血中のニコチン濃度を喫煙
した時のようなレベルに到達させることを初めて可能に
するが、このことはカプセルを治療に使用可能ならしめ
るための本質的な前提条件である。ニコチンは下記の症
候に対する医薬として使用できる。
肥満症 パーキンソ症候群 アルツハイメル病 不安神経症 脳硬変症 潰瘍性大腸炎 本発明は0.1〜10mg/個、好ましくは0.1〜3mg/個、特
に２〜5mg/個の放出量でニコチンを溶液として含有し、
その溶液のpHが６〜10、好ましくは７〜９、特に６〜８
の範囲にあり、加えて収斂味を改善する添加剤を含有す
る経口投与用の噛むことができるカプセルを、禁煙用に
使用することを包含する。
カプセルの内容物以外で重要なのは、媒質である溶液
のpHである。溶液のpHはニコチンの吸収速度を左右する
からである。個々のカプセルに含まれる媒質のpHを異な
らせ、カプセルを服用する人々が彼らのニコチン要求量
に応じた適当なpHのカプセルを選択できるようにするこ
とは、価値のあることと思われる。ニコチン要求量には
個人差があることが知られている。

    (57)【特許請求の範囲】
  1. 【請求項１】ニコチン含有流動媒質を含み、口内で咀嚼
    されるカプセルに於て、前記ニコチン量が0.1〜10mg/個
    の範囲にあり、前記媒質が風味及び耐久安定性を改善す
    る添加剤を含有し、前記媒質のpHが６〜10の範囲にある
    ことを特徴とするニコチン含有流動媒質を含む経口投与
    用カプセル。
  2. 【請求項２】禁煙用の特許請求の範囲第１項記載のカプ
    セル。
  3. 【請求項３】個々のカプセルに含まれる前記媒質のpHが
    前記の範囲内で異なることを特徴とする特許請求の範囲
    第１項または第２項記載のカプセル。
  4. 【請求項４】前記カプセルの内容量が0.1〜2.5mlである
    ことを特徴とする特許請求の範囲第１項記載のカプセ
    ル。
  5. 【請求項５】溶解又は咀嚼される大カプセル中に複数個
    収納されることを特徴とする特許請求の範囲第４項記載
    のカプセル。
