Example 1
Compound XII was produced from compound Xj by carrying out the reaction shown in the following formula.
窒素雰囲気下、丸底フラスコに化合物Xj(230mg crude,0.39mmol)を加え、CH2Cl2(1mL)で溶解し、0℃に冷却した。 Compound Xj (230 mg crude, 0.39 mmol) was added to a round-bottom flask under nitrogen atmosphere, dissolved in CH 2 Cl 2 (1 mL), and cooled to 0° C. Then, boron trifluoride diethyl ether complex (BF 3 Et 2 O) (96 μL, 0.78 mmol) was added slowly over 1 min, and stirred at 0° C. for 5 min. Then, triethylsilane (124 μL, 0.78 mmol) was added slowly over 1 min. Then, the reaction was allowed to proceed with stirring at room temperature for 4 h. After TLC confirmed that the starting material was completely consumed, the reaction mixture was diluted with methyl tert-butyl ether (MTBE), quenched with saturated aqueous NaHCO 3 (10 mL), the two phases were separated, and the organic layer was washed with brine (10 mL), dried over Na 2 SO 4 , and concentrated on a rotary evaporator. The resulting crude product was purified by silica gel column chromatography (ethyl acetate/n-hexane=3/7) to obtain Compound XII (55 mg, yield from Compound Xj: 25%) as a colorless oily substance (diastereomer ratio (β form:α form)=3:1, Rf value: 0.5). Note that the β form was the target substance.
