Abstract
Description
老齢人口の拡大に伴い、神経系の軸索損傷の発生が増加している(DeVivo,M.J.,and Chen,Y.(2011).Archives of Physical Medicine and Rehabilitation 92,332-338(非特許文献1);Singh et al.,(2014).Clinical Epidemiology 6,309(非特許文献2))。残念ながら、軸索再生能および修復過程は、加齢と共に衰退するため、機能的回復が弱まり、長期的な能力障害が増加する(Nagano,A.(1998).Journal of Orthopaedic Science 3,71-80(非特許文献3);Pestronk et al.,(1980).Experimental Neurology 70,65-82(非特許文献4);Tanaka and deF.Webster,(1991).Journal of Comparative Neurology 308,180-187(非特許文献5);Vaughan,1992.Journal of Comparative Neurology,323,219-237(非特許文献6);Verdu et al.,(2000).Journal of the Peripheral Nervous System 5,191-208(非特許文献7))。
本発明は、加齢依存性の再生能衰退を有する対象における末梢神経再生の促進のための、CXCL13を中和する結合分子、例えば、抗体およびその抗原結合性断片の使用に関する。
加齢依存性の再生能衰退を有する対象において末梢神経再生を促進するためにCXCL13結合分子を使用する方法が、本明細書に開示される。本明細書において例示される開示の局面によると、CXCL13と特異的に結合し、CXCL13の効果を阻害し、抑制し、防止し、逆転させ、または減速させる、有効量の単離された結合分子を、対象へ投与する工程を含む、末梢神経損傷および加齢依存性の再生能衰退を有する対象における末梢神経再生を改善する方法が、提供される。
I.定義

CXCL13は、Bリンパ球(およびT細胞のサブセット)の遊走を促進することが本明細書において示される小さいケモカインであり(図2A、2B、および5D)、それは、バーキットリンパ腫受容体1(BLR-1)を発現する細胞へのカルシウム流入およびBLR-1を発現する細胞の走化性を刺激することによると考えられる。従って、それは、Bリンパ球の嚢胞へのホーミングにおいて機能することができる。
若齢哺乳動物において、軸索は、末梢神経損傷後に容易に再生する。細胞体から切り離された軸索の遠位部分は、ウォラー変性を受ける。この活発な過程は、軸索の断片化および崩壊をもたらす。破片は、膠細胞、例えば、シュワン細胞、およびマクロファージによって除去される。次いで、近位軸索が再生し、標的を神経再支配し、機能の回復を可能にすることができる。しかしながら、軸索修復過程および軸索再生能は、加齢過程と共に衰退する。加齢は、末梢神経系(PNS)のいくつかの形態学的特色および機能的特色に深く影響する。老齢動物においては、損傷後のウォラー変性が遅延し、マクロファージに蓄積される髄鞘の残骸が、若齢動物より大きくなる。高齢対象においては、若齢対象より、シュワン細胞と再生軸索との相互作用に時間がかかり、反応性シュワン細胞および標的器官によって分泌される栄養因子および刺激(tropic)因子の量が少なくなる。老齢動物においては、軸索再生の速度が遅くなり、再生軸索の密度が減少する。加齢は、再生された線維の終末および側枝の出芽の低下を決定して、標的神経再支配および機能的回復の能力をさらに制限する。これらの加齢に関連した変化は、年齢と共に直線的に進行するのではなく;軸索再生および神経再支配の能力は、生涯を通して維持されるが、加齢と共に遅延し、効果が低くなる傾向がある(E Verdu,et al.,J Peripher.Nerv Syst.2000 Dec;5(4):191-208;Fenrich K.et al,Can J Neurol Sci.2004 May;31(2):142-56)。これらの観察は、加齢が、損傷特異的な細胞シグナルと組み合わされた時、ニューロンを再生不能にする、独特の分子機序および細胞機序の発生をもたらすことを示唆している。

本開示の抗CXCL13結合分子、例えば、抗体、またはその抗原結合性断片、バリアント、もしくは誘導体を調製し、それを必要とする対象へ投与する方法は、当業者に周知であり、または当業者によって容易に決定される。抗CXCL13結合分子、例えば、抗体、またはその抗原結合性断片、バリアント、もしくは誘導体の投与経路は、例えば、経口、非経口、吸入、または局所であり得る。非経口という用語には、本明細書において使用されるように、例えば、静脈内、動脈内、腹腔内、筋肉内、皮下、直腸、または膣への投与が含まれる。これらの投与形態は、全て、本開示の範囲内にあると明確に企図されるが、投与のための形態の例は、注射用溶液、具体的には、静脈内、腹腔内、または動脈内への注射または点滴のための溶液であろう。一般的に、注射用の適当な薬学的組成物は、緩衝液(例えば、酢酸緩衝液、リン酸緩衝液、またはクエン酸緩衝液)、界面活性剤(例えば、ポリソルベート)を含み、任意で、安定剤(例えば、ヒトアルブミン)等を含み得る。しかしながら、本明細書中の教示に適合する他の方法において、本開示の抗CXCL13結合分子、例えば、抗体、またはその抗原結合性断片、バリアント、もしくは誘導体を、末梢神経損傷の部位へ直接送達し、それによって、損傷を負った組織の治療剤への曝露を増加させることもできる。
本開示は、対象が抗CXCL13結合分子による処置から恩恵を受けるか否か、例えば、対象が加齢依存性の神経再生能衰退を経験しているか否かを決定するための、対象における末梢神経損傷の診断のために有用な方法をさらに提供する。本法は、例えば、坐骨神経損傷などの末梢神経損傷を有するか、または有すると推測される個体に由来する、損傷を負った末梢神経のDRGもしくはその近傍の組織、またはその他の細胞もしくは体液において、CXCL13タンパク質または転写物の発現レベルを測定する工程、および測定された発現レベルを、若齢対象の正常な組織または体液における標準CXCL13発現レベルと比較する工程を含み、ここで、標準と比較された試料における発現レベルの増加は、対象が本明細書に開示される抗CXCL13抗体による処置から恩恵を受けるであろうことを示す。ある特定の態様において、本開示の抗CXCL13抗体、またはその抗原結合性断片、バリアント、および誘導体、例えば、MAb 5261、MAb 5378、MAb 5080、MAb 1476、3D2、または3C9は、加齢依存性の神経再生能衰退の存在の診断において使用される。
以下の材料、方法、およびプロトコルを、以下の実施例において適用した。
8~10週齢の野生型C57/BL6Jマウスは、Charles Riverから得られ、老齢マウス(20~22ヶ月齢)は、Charles RiverまたはJucker's laboratory,HIH(Tuebingen)のいずれかから供給された。B57/BL6の遺伝的背景を有するOT Iマウスは、Botto's laboratory,Imperial Collegeによって提供された。全ての動物手法が、UK Animals Scientific Procedures Act(1986)に従って実行され、Imperial College Londonの倫理委員会によって承認された。
24穴プレートにおいて、無菌15mmカバーガラスを、H2Oで希釈された0.003%ポリ-L-オルニチン(Sigma)によって37℃で1時間コーティングした。水で3回洗浄した後、カバーガラスを、PBS(Invitrogen)で調製された1μg/mlラミニン(Millipore)によって室温で3時間コーティングした。PBSでの2回の洗浄工程の後、ウェルにDMEMを充填し、細胞播種まで37℃に置いた。DRGを解剖し、氷上のハンクス平衡塩溶液(HBSS)(Invitrogen)に回収した。次いで、HBSSを除去し、5分毎にチューブを軽く叩きながら、37℃の水浴中で40分間、500μlの消化溶液(5mg/mlディスパーゼII(Sigma)、DMEM(Invitrogen)中の2.5mg/mlコラゲナーゼII型(Worthington))に交換した。消化後、800rpmでの2分間の遠心分離によって、DRGを回収し、温かいDRG培地(DMEM/F12(Invitrogen)、1×B27(Invitrogen)、10%ウシ胎仔血清(FBS)(Sigma))で1回洗浄した。次いで、DRGを、1mlの温かいDRG培地に再懸濁させ、ファイアポリッシュされたSigmacote(Sigma)でコーティングされたガラスピペットによる15回のピペッティングによって破砕した。未解離組織を100μmセルストレーナー(BD Falcon)で濾過し、血球計数器によって明視野顕微鏡下で細胞を計数した。細胞計数後、細胞をスピンダウンし、温かいDRG培養培地(DMEM/F12(Invitrogen)、1×B27(Invitrogen)、1%ペニシリンおよびストレプトマイシン(Invitrogen))に再懸濁させ、播種した(1ウェル当たり4000細胞)。DRG細胞を37℃で5%CO2中で24時間培養した。
技術的3回反復および生物学的3回反復によって、各条件について少なくとも100個の細胞(ランダムに選択されたカバーガラス1枚当たり5つの視野)を用いて、Neurolucidaソフトウェア(MBF Bioscience)を使用することによって、培養されたDRG細胞の平均神経突起長を測定した。
培養されたDRG細胞を、1×PBS中の氷冷4%パラホルムアルデヒド(PFA)(Sigma)によって氷上で30分間固定し、1×PBSで3回洗浄した。細胞を、1×PBS、0.1%トリトンX、5%正常ヤギ血清(Abcam)で、室温で1時間ブロッキングした後、一次抗体を含む1×PBS、0.1%トリトンX、2%正常ヤギ血清と共に4℃で一晩インキュベートした。次いで、細胞を1×PBSで3回洗浄し、二次抗体を含む1×PBS、0.1%トリトンと共に暗所で室温で2時間インキュベートした。細胞を1×PBSで3回洗浄し、4-6-ジアミジノ-2-フェニルインドール(DAPI、Molecular Probes、1:5000)を含む1×PBS、0.1%トリトンXと共に10分間インキュベートした後、PBSで2回洗浄した。細胞を下向きにカバーガラスをスライドに移し、Antifade封入剤(Vectashield、H-1000)によって封入した。
以前に記載されたプロトコルに従って、坐骨神経に挫滅を与えた(Bauder and Ferguson,(2012).JoVE(Journal of Visualized Experiments),e3606)。1L/min酸素中の2%イソフルランによってマウスを深麻酔下に置き、鎮痛のため、0.1mg/kgブプレノルフィンおよび5mg/kgリマダイル(rimadyl)を皮下(s.c.)注射した。正中線を横切って皮膚を切開し、筋膜面を開き、大殿筋と大腿二頭筋の前頭(anterior head)との間を小さくより深く切開することによって、坐骨神経を露出させた。
シャムまたは坐骨神経損傷の24時間後に、若齢または老齢の坐骨DRG組織全体に由来するRNAを使用して、RNA配列決定を実施した。具体的には、1試料当たり1匹のマウスから坐骨DRGを解剖し、RNAlater安定化溶液(ThermoFisher)で保存した。組織をRNaseフリーマイクロペッスルで均質化し、製造業者のプロトコルに従い、RNeasyキット(Qiagen)を使用してRNAを抽出した。DNAを除去するため、RNaseフリーDNaseセット(Qiagen)によって、オンカラムDNase消化を15分間実施した。RNAの濃度および純度を、Agilent2100 Bioanalyzer(Agilent)を使用して測定した。RNAインテグリティナンバー(integrity number)(RIN)が7.5を超えるRNAをライブラリ調製に使用した。ライブラリは、TruSeq mRNA Sample Preparationキット(Illumina)を使用して、Ospedale San Raffaele(Milan)で調製され、Illumina HiSeq 2500、100サイクル、ペアエンドシーケンシングを使用して配列決定された。
FDR(偽発見率)補正P値は、Benjamini-Hochberg(BH)補正によって実行された。データセット内の全ての発現遺伝子をバックグラウンドとして用いて、DAVID v6.7(Huang et al.,Systematic and integrative analysis of large gene lists using DAVID Bioinformatics Resources.Nature Protoc.2009;4(1):44-57;Huang et al.,Bioinformatics enrichment tools:paths toward the comprehensive functional analysis of large gene lists.Nucleic Acids Res.2009)を使用して、3つの比較(それぞれ、SNI若齢、シャム高齢、SNI高齢と、シャム若齢との比較)のディファレンシャルに発現された(DE)遺伝子(FDR<0.05)を、遺伝子オントロジー(GO)およびKEGGパスウェイについて分析した。アップレギュレーションおよびダウンレギュレーションの両方について、FDR<0.01によって、GOおよびKEGGパスウェイのカテゴリを選択した。ベン(Venn)図を、Biovennを使用して生成した(BioVenn - a web application for the comparison and visualization of biological lists using area-proportional Venn diagrams,T.Hulsen,J.de Vlieg and W.Alkema,BMC Genomics 2008,9(1):488)。GOおよびKEGGパスウェイから収集された免疫応答に関与するSNI高齢対シャム若齢からのディファレンシャルにアップレギュレートされた遺伝子を、STRINGによって確立されたタンパク質-タンパク質ネットワークについて分析した(Szklarczyk et al.,STRING v11:protein-protein association networks with increased coverage,supporting functional discovery in genome-wide experimental datasets,Nucleic Acids Res.2019)。実験およびデータベースのアクティブな相互作用ソース、および必要とされる最低相互作用スコアの最も高い信頼度を使用して、ストリンジェントな基準を選択した。ネットワークは、Cytoscapeによって可視化された(Shannon et al.,Cytoscape:a software environment for integrated models of biomolecular interaction networks,Genome Research 2003 Nov;13(11):2498-504)。
シャムまたは坐骨神経損傷の3日後に1試料当たり2匹の若齢または老齢のマウスから解剖された坐骨DRGを収集し、プロテアーゼ阻害剤(Roche)を含むマウスCXCL13 ELISAキット(ThermoFisher)から供給された150μLアッセイ希釈液Aにおいて均質化した。均質化後、Triton X-100を最終濃度1%で添加した。試料を液体窒素で凍結させ、解凍した後、破片を除去するため、10,000×gで5分間、遠心分離した。タンパク質濃度をBCAタンパク質アッセイキット(ThermoFisher)を使用して定量化し、100μLの細胞溶解物を、製造業者のプロトコルに従って、マウスCXCL13 ELISAキットを使用して、CXCL13 ELISAアッセイに使用した。CXCL13 ELISA測定値は、450nmに設定されたELISAプレートリーダーで吸光度を測定し、マイクロプレートの光学的欠陥を補正するため、550nmの値を差し引くことによって得られた。細胞溶解物中のCXCL13濃度は、標準曲線に従って計算された。
腹腔内(i.p.)注射を介して、ケタミン/キシラジン混合物(最大80mg/kg体重のケタミンおよび10mg/kg体重のキシラジン)によって、マウスを深麻酔下に置き、5ml/minの流速で、20mlの氷冷PBS、続いて、4%PFAで経心灌流した。後肢のDRG、坐骨神経、または無毛の皮膚を切り出し、氷上で4%PFAで2時間後固定し、30%ショ糖(Sigma)に3日間移した後、OCT化合物(Tissue-Tek)で包埋した。DRGを10μmに、坐骨神経を12μmに、指間足蹠を30μmに、クライオスタット(Leica)によって、皮膚表面に対して垂直に切片化した。切片を、10%正常ヤギ血清または正常ロバ血清(Abcam)および0.3%Triton X-100を含むPBSを含有するブロッキング溶液で室温で1時間処理し、4℃で一晩、一次抗体によって染色した。一次抗体は、抗Tuj1(1:500、Novus、NB100-1612)、抗CXCL13(10μg/ml、R&D SYSTEMS、AF470)、抗ニューロフィラメント200(1:100、Sigma、N4142)、抗CGRP(1:200、Abcam、ab36001)、Alexa Fluor 488コンジュゲートGS-IB4(1:100、ThermoFisher、121411)、抗SCG10(1:500、NOVUS、NBP1-49461)、抗CD8(1:100、ThermoFisher、14-0081-82)、抗CD3(1:100、Abcam、ab16669)、抗CXCR5(1:100、Abcam、ab133706)、抗CD68(1:200、Abcam、ab125212)、抗B220(1:100、Biolegend、103228)、抗MHC-I(1:100、Abcam、ab15681)、抗活性カスパーゼ3(1:200、Cell Signaling、#9664)、抗リン酸化AKT(1:200、Cell Signaling、#4060)、抗リン酸化S6(1:200、Cell Signaling、#5364)、抗パーフォリン(1:100、NOVUS、NBP1-97512)、抗グランザイムB(1:100、Abcam、ab4059)、抗PGP9.5(1:200、Proteintech、14730-1-AP)、抗リン酸化NFκB2(1:100、Abcam、ab194919)、抗ルシフェラーゼ(1:200、Fitzgerald、70R-12141)、抗GFP(1:500、Abcam、ab13970)であった。切片を1×PBSで3回洗浄した後、Alexa Fluor 488ヤギ抗ニワトリ、Alexa Fluor 488ロバ抗ニワトリ、Alexa Fluor 488ヤギ抗ラット、Alexa Fluor 488ヤギ抗ウサギ、Alexa Fluor 568ロバ抗ヤギ、Alexa Fluor 568ヤギ抗ウサギ(1:1000、ThermoFisher)によって、室温で2時間、二次抗体インキュベーションを行った。次に、切片を、DAPI(Molecular Probes、1:5000)で室温で15分間染色し、Antifade封入剤(Vectashield、H-1000)で封入した。DRGは、20倍の倍率を使用して、Nikon Eclipse TE2000顕微鏡によって画像化された。坐骨神経(20倍の倍率)および表皮(オイルレンズで40倍の倍率)は、Leica TCS SP8共焦点レーザー走査型顕微鏡によって画像化された。
ImageJソフトウェア(National Institute of Health、USA)によって任意蛍光強度を測定し、バックグラウンドの強度を除外した後、対照群に対して正規化された蛍光強度によって相対倍率変化を計算した。切断カスパーゼ3、パーフォリン、およびグランザイムBの発現について陽性と示された細胞は、バックグラウンドより少なくとも2倍高い蛍光強度を示した。SCG10で染色された坐骨神経の縦切片を、軸索再生について分析した。神経に沿ってSCG10の任意強度を測定し、挫滅の近位部位から0.5mm毎の強度の百分率を定量化した。強度が50%レベルに低下する近位損傷部位からの距離を計算することによって、再生指数も評価した。表皮神経再支配については、皮膚の矢状切片からのPGP9.5染色によって示される表皮内神経線維(IENF)の単位体積当たりの数を測定した。
インビボでの細胞特徴決定のため、若齢または老齢の未処置動物または坐骨神経損傷動物に、3μgの抗CD45-APC(I3/2.3、Biolegend)を、3分間、静脈内(i.v.)注射した。深麻酔後に、それぞれ、2mM EDTAおよびRPMI-1640培地(ThermoFisher)を含む10ml PBSに、末梢血またはDRGを収集した。RTでの5分間の300gでの遠心分離によって血球を回収し、赤血球を1×RBC溶解緩衝液(Biolegend)を使用して溶解した。細胞をPBSで3回洗浄し、凝集塊を除去するため、70μmセルストレーナー(Corning)で濾過し、染色のため、新鮮なFACS緩衝液(0.5%BSA、2mM EDTAを含むPBS)に再懸濁させた。DRGを1×PBSで1回洗浄し、消化緩衝液(0.5mg/ml CLSPA(Worthington、#LS005273)、7.5μg/ml DNase I(Roche、#10104159001)を含むRPMI-1640)によって、37℃で30分間、解離させた。消化後、DRG組織を分散させ、単一細胞懸濁液を収集するため、70μmセルストレーナーで濾過し、洗浄緩衝液(5%FBSを含むRPMI-1640)で洗浄した。遠心分離後、ペレットを、DNase溶液(250μg/ml DNase I、1×DNase緩衝液(1.21g/L Tris Base、0.5 g/L MgCl2、および0.073g/L CaCl2を含むRPMI-1640溶液))に、RTで30分間、再懸濁させた。細胞を染色のためFACS緩衝液で洗浄した。
インビボのCD8 T細胞を枯渇させるため、22~24ヶ月齢のマウスに、坐骨神経挫滅損傷前の1週間に、1日おきに3回、200μgのInVivoMAbラットIgG2bアイソタイプ対照(LTF-2、BioXcell、BE0090)またはInVivoMAb抗マウスCD8α(YTS 169.4、BioXcell、BE0117)を注射した。坐骨神経挫滅の後、動物が麻酔から完全に回復した時に、1回の追加の注射を実施した。神経損傷の3日後に、DRGおよび神経を解剖し、続いて、FACS、免疫染色、または再生アッセイを行った。CD4 T細胞の枯渇は、InvivoMAb抗マウスCD4(YTS 191、BioXcell、BE0119)およびラットIgG2bのアイソタイプ対照を使用して、CD8 T細胞枯渇のために使用されたのと同じ計画によって実施された。B細胞の枯渇は、以前に記載された戦略に従って実施された(Keren et al.,2011)。150μgのInvivoMAb抗マウスCD19(1D3、BioXcell、BE0150)、150μgのInvivoMAb抗マウスB220(RA3.3A1/6.1、BioXcell、BE0067)、150μgのInvivoMAb抗マウスCD22(Cy34.1、BioXcell、BE0011)の抗体カクテルを老齢マウスにi.p.注射した。48時間後、二次抗体InvivoMAb抗ラットカッパ(MAR18.5、BioXcell、BE0122)を、マウス1匹当たり150μgの用量で、マウスに注射した。全ての注射を、坐骨神経損傷前の1週間に2回、損傷の翌日にもう1回、実施した。ラットIgG2a(2A3、BioXcell、BE0089)、ポリクローナルラットIgG(BioXcell、BE0094)、マウスIgG1(MOPC-21、BioXcell、BE0083)、およびマウスIgG2a(C1.18.4、BioXcell、BE0085)を使用して、使用された抗体と同じアイソタイプの対照IgGを注射した。
DRGエクスビボ培養のため、若齢マウスおよび老齢マウスを、1週間に3回、30mg/kgの対照IgGまたは抗CXCL13抗体(抗CXCL13モノクローナル抗体(mAb 5378-41)および対照IgG2a(mAb 2510))によってi.p.処理した後、DRG解剖および初代細胞培養を行った。単離されたDRG細胞を5%CO2中で37℃で24時間インキュベートした後、神経突起伸長測定を行った。インビボでのCXCL13中和のため、坐骨神経挫滅の4時間後および3日目の屠殺まで毎日、30mg/kgの対照IgGまたは抗CXCL13抗体を、若齢または老齢マウスにi.p.注射した。慢性神経再生研究のため、30mg/kgの対照IgGまたは抗CXCL13抗体を、損傷の4時間後から、動物を屠殺するまで、週3回、i.p.注射した。
坐骨神経挫滅および対照IgGまたは抗CXCL13抗体の慢性投与を受けたマウスを、行動査定に使用した。坐骨神経および後肢の左側に損傷を与え、査定した。行動評価に参加した研究者には、群および処置が知らされなかった。
マウスを、順応のため、試験チャンバーに30分間置いた後、行動査定を開始した。機械感受性は、0.4g~4gの範囲の較正されたフォンフライフィラメントによって左後肢の足底表面をプロービングすることによって決定された。基線に戻るのに十分な時間を感覚受容器に与えるため、各トライアルの間隔は少なくとも30秒であった。迅速な後肢退避を陽性反応と見なした。各マウスについて5回のトライアルを実行し、3回の陽性退避反応からの潜時の値を平均化した。閾値は、モノフィラメントの力の最低レベルに設定された。
熱感受性については、以前に記載されたように、ハーグリーブス試験を実施した(Chen et al.,(2014).Nature Communications 5,5331;Hargreaves et al.,(1988).Pain 32,77-88)。簡単に説明すると、マウスをガラスの床に置き、プラスチックチャンバーで分離した。順応させるため、マウスに30分間与えた。熱刺激(赤外線放射源、強度=50)を後肢の足底表面の下に、10秒未満、注意深く置いた。後肢退避時間を自動的に記録した。各肢に対して5回のトライアルを行い、最長および最短の退避時間を含め、平均値を計算した。
小さい粘着刺激(1/4インチの丸い粘着ラベル)をマウスの左後肢に置き、両前肢または口で最初に触れるまでの時間、および粘着物を完全に除去するまでの時間を記録した。神経損傷前の1週間に、マウス1匹につき毎日10分間のトレーニングを実施した。各マウスは3回のトライアルを受けた。全ての試験が、動物のホームケージにおいて盲検的に実施された。
特記しない限り、データは、Graphpad Prism 8.0(Graphpad Software Inc.,La Jolla,CA)によって統計的に分析され、平均値±SEMとして表された。2つの群についての統計的比較は、対応のないスチューデントt検定を使用して分析された。複数の群は、事後チューキー補正による一元配置分散分析(ANOVA)、またはチューキーもしくはシダックの検定による二元配置分散分析を使用して分析された。統計的有意性をp<0.05と見なした。
後根神経節(DRG)における遺伝子発現に対する加齢の効果を、神経損傷前(シャム)および神経損傷後の8~10週齢(若齢)マウスおよび20~22週齢(老齢)マウスから得られた坐骨DRGからのRNA配列決定(RNA seq)によって査定した。X匹の若齢マウスおよび老齢マウスの坐骨神経に、鉗子で神経に圧迫を加えることによって、挫滅を与えた。各マウスは、前記のように、坐骨神経に、ほぼ同じ時点で、損傷またはシャム処置を受けた。24時間後、坐骨神経損傷(SNI)またはシャム損傷の部位付近のDRGを取り出し、そこからRNAを抽出し、配列決定した。DRGからの遺伝子発現の変化を、若齢シャム動物における基線発現と比較して、老齢シャム、SNI後の若齢および老齢において査定した。有意にディファレンシャルに発現された(DE)遺伝子(FDR<0.05)を、機能的分類のため、遺伝子オントロジー(GO)およびKEGGによって分析した。典型的には、神経新生、神経伝達物質、イオン輸送、G共役シグナル伝達、およびシグナル伝達に関与する、若齢動物においてSNI後にアップレギュレートされたGOクラスは、SNI後を含め、老齢DRGにおいてはディファレンシャルにエンリッチされ得なかった(データは示されない)。しかしながら、GO分析による最も顕著な所見は、老齢DRGが、SNIの前後の両方で、T細胞シグナル伝達および部分的なB細胞シグナル伝達を含む適応免疫応答について極めて有意なエンリッチメントを示したことであった。KEGG分析は、免疫応答およびシグナル伝達ならびにサイトカイン/ケモカインシグナル伝達の年齢および損傷に関連する顕著な変化の存在をさらに強調した。損傷後の老齢DRGにおけるDE遺伝子の大部分(66.6%のアップレギュレーション、65%のダウンレギュレーション)が、加齢依存性であった。
CXCL13のインビボニューロン発現がCXCR5+T細胞を誘引するか否かを決定するため、若齢マウスの坐骨神経にAAV-GFPまたはAAV-CXCL13-GFPを感染させることによって、DRGニューロンにおいてCXCL13を過剰発現させた。感染の6週間後、それぞれ、MHC-Iの提示を誘導し、血液関門透過性を有利にするため、インターフェロンγ(IFNγ)およびマンニトールを全身送達し、その直後に坐骨神経挫滅を実施し、損傷後3日目の坐骨DRGにおいてFACSによってCXCR5+T細胞を測定した(データは示されない)。興味深いことに、CXCR5+CD8+T細胞は、GFP対照と比較して、CXCL13過剰発現DRGにおいて有意に増加することが見出された(図2AおよびB)。
CD8 T細胞が老齢坐骨DRGニューロンの軸索再生を損なうか否かの分析を行った。坐骨神経挫滅の1週間前に老齢マウスに全身的にi.p.注射された抗CD8モノクローナル抗体を使用して、CD8 T細胞の枯渇を実施し、一方、対照動物は、正常アイソタイプ一致IgGを受容した。SCG10陽性坐骨神経軸索の分析は、抗CD8モノクローナル抗体が、神経再生を有意に促進すること(図3A~B)、免疫蛍光分析によって示されたように、DRG内のCD8 T細胞の有意な低下をもたらすことを示した(図3C)。全く対照的に、同じ実験的損傷モデルにおいて、抗CD4モノクローナル抗体によるCD4 T細胞の枯渇またはモノクローナル抗CD19/抗B220/抗CD22抗体によるB細胞の枯渇は、坐骨神経損傷後の加齢依存性の再生能衰退を改変しなかった(示されない)。
CXCL13の中和が老齢動物における軸索再生、表皮神経支配、および機能的回復を促進するか否かを、図5Aの模式図に概説された戦略を使用して調査した。最初の実験において、CXCL13に対するモノクローナル抗体または対照IgGを、坐骨神経挫滅損傷後に3日間、毎日、若齢マウスまたは老齢マウスに前記のように腹腔内注射した。軸索再生を、前記のように抗SCG10免疫染色によって測定した。抗CXCL13抗体は、加齢依存性の再生障害を阻止し、若齢における軸索再生に対しては効果を及ぼさなかった(図5B~C)。CXCL13中和は、老齢DRGにおけるCXCR5+T細胞およびB細胞の数を有意に低下させた(図5D)。別の実験において、機能的回復に対する抗CXCL13抗体の長期送達を試験した。前記のように、CXCL13に対するモノクローナル抗体または対照IgGを、損傷の4時間後から開始して、5週間以上にわたり、週3回、若齢動物または老齢動物に送達した。フォンフライによって機械刺激に対する反応を測定し、テープ除去試験によって接触に対する反応を測定し、ハーグリーブスによって熱侵害受容に対する反応を測定する、マルチモーダル感覚査定を実施した(図5A)。データ分析は、老齢マウスの回復が若齢と比較して有意に遅いことを示し、そして、重要なことに、CXCL13中和が、老齢マウスにおいて選択的に調査された感覚モダリティの全てにおいて、老齢マウスの回復の有意な加速を誘導することを示した(図5E~J)。CXCL13中和が表皮神経支配の改善をもたらすか否かを、抗CXCL13または対照IgGの送達後の若齢マウスおよび老齢マウスの両方における、坐骨神経損傷の18日後の後肢の無毛指間区域からのPGP 9.5免疫染色によって測定した。CXCL13中和は、老齢マウスにおいて有意な表皮神経支配を促進することが見出された(図5K、L)。

(B)それぞれSEQ ID NO:18、21、および24に記載の重鎖相補性決定領域1(VH-CDR1)、VH-CDR2、およびVH-CDR3のアミノ酸配列を含むVH;ならびに、それぞれSEQ ID NO:27、30、および33に記載の軽鎖相補性決定領域1(VL-CDR1)、VL-CDR2、およびVL-CDR3のアミノ酸配列を含むVL;
(C)それぞれSEQ ID NO:19、22、および25に記載の重鎖相補性決定領域1(VH-CDR1)、VH-CDR2、およびVH-CDR3のアミノ酸配列を含むVH;ならびに、それぞれSEQ ID NO:28、31、および34に記載の軽鎖相補性決定領域1(VL-CDR1)、VL-CDR2、およびVL-CDR3のアミノ酸配列を含むVL;または

を含む、請求項23~26のいずれか一項記載の使用のための抗体またはその抗原結合性断片。 - 前記抗体またはその抗原結合性断片のVHおよびVLが、(i)SEQ ID NO:6およびSEQ ID NO:7;(ii)それぞれSEQ ID NO:8およびSEQ ID NO:9;(iii)それぞれSEQ ID NO:10およびSEQ ID NO:11;(iv)それぞれSEQ ID NO:12およびSEQ ID NO:13;(v)それぞれSEQ ID NO:14およびSEQ ID NO:15;ならびに(vi)それぞれSEQ ID NO:16およびSEQ ID NO:17からなる群より選択されるVHおよびVLの配列と同一のアミノ酸配列を含む、請求項23~26のいずれか一項記載の使用のための抗体またはその抗原結合性断片。
- 前記単離された抗体またはその抗原結合性断片がMAb 5261、またはMAb 5091、Man 1476、またはMAb 1758である、請求項23~28のいずれか一項記載の使用のための抗体またはその抗原結合性断片。
- 対象が末梢神経損傷を有する、請求項23~29のいずれか一項記載の使用のための抗体またはその抗原結合性断片。
- 末梢損傷が坐骨神経損傷、腕神経叢損傷、脊髄副神経損傷、または腓骨神経損傷である、請求項30記載の使用のための抗体またはその抗原結合性断片。
- 末梢神経損傷が坐骨神経損傷である、請求項31記載の使用のための抗体またはその抗原結合性断片。
