Abstract
Description
(1)システイン残基を含むAPTEDB及びMal-PEG2000-DSPEを有機溶媒に1:2モル比で混合して重合反応を誘導する工程、
(2)前記(1)工程の混合溶液からAPTEDB-PEG2000-DSPEポリマーを得る工程、
(3)水、PBS、HBS、及びHBGからなる群から1種以上選択される溶媒に、前記APTEDB-PEG2000-DSPEポリマーとPEG2000-DSPE重合脂質とを混合してマイセル構造体形成を誘導する工程、及び
(4)前記(3)工程の混合溶液を濾過し、マイセル構造体を精製する工程。

全ての遺伝子発現プロファイルは、Oncopressionから得ることができ、EDB-FN関連データは、FN又はED-Bとしても知られているフィブロネクチン1(fibronectin 1、FN1、Entrez Gene ID:2335)を研究することによって収集した。FN1遺伝子の変異体は、ClinVar(https://www.ncbi.nlm.nih.gov/clinvar)を用いて調べ、126個の変異体が確認された。悪性神経膠腫における発現レベルをスクリーニングするために、トランスクリプトミクスデータベース分析を行ったため、変異体の分析は本発明で実行されなかった。EDB-FNのトランスクリプトミクス発現レベルは、Single Channel Array Normalization and Universal exPression Codes(SCAN.UPC)package of Rで正規化し、UPC値で表した。より具体的には、Oncopressionデータのために単一サンプル正規化方法を使用し、全てのサンプルは、Affymetrix Human Genome U133 Plus 2.0(GPL570 or A-AFFY-44)platformから受け取った。発現値は、0.0から1.0で表され、1.0は、完全に転写活性化した場合を示す。正常細胞に対する癌細胞の比率(cancer-to-normal ratio)は、癌細胞における発現値を正常細胞における平均発現値で割って算出した。生存分析のために、発現プロファイル及び患者予後情報を含む脳腫瘍データセットを収集した後、30個以下のサンプルを分析から除外した。全ての遺伝子発現値は、データセットによってクオンタイル正規化(quantile normalization)した。Z値は、各データセットでログランク検定(log-rank test)して算出し、体重として各データセットの患者数の平方根を用いてLiptak methodで平均を出した。
患者組織サンプルは、手術及び病理学的診断の後、パラフィンブロック(paraffin block)に保管され、患者は、成人21人で、年齢18歳から75歳であり、多形性膠芽腫と診断された。各患者別に、3~4種類の他の腫瘍部位から合計65個の組織サンプルを含む組織マイクロアレイスライドを製造した。組織コア(tissue core)は、直径2mmで各ブロック別に45個の孔を含む被移植者パラフィンブロック(recipient paraffin block)で移植された。パラフィンで満たされたブロックを3μmの厚さに切り、スライド上に置いた。その後、組織を後述する免疫組織化学(immunohistochemistry)部分に記載したように染色した。免疫染色後、病理学者から盲検レビュー(blind review)を介してEDG-FNの存在及びレベルに対する読み取りを受けた。染色強度は、none又は「1+」(非常に弱い陽性)から「4+」(非常に強い陽性)で表した。本発明において、「1+」及び「2+」は、低発現群(low-expression group)に、「3+」及び「4+」は、高発現群(high-expression group)で分類した。EDB-FN発現と患者の予後予測との間の相関関係を分析するために、患者組織マイクロアレイにおけるEDB-FN発現レベルの結果と各患者の臨床データとを結びつける作業を行った。変数として無進行生存期間(progression-free survival、PFS)及び全生存期間(overall survival、OS)を使用して患者予後を分析した。
ポリエチレングリコール(2000)-1、2-ジステアロイル-sn-グリセロ-3-ホスホエタノールアミン(アンモニウム塩)(PEG2000-DSPE)、DSPE-N-(リサミンローダミンBスルホニル)(アンモニウム塩)(Rh-DSPE)、及びN-マレイミド-PEG2000-DSPE(アンモニウム塩)(Mal-PEG2000-DSPE)は、Avanti Polar Lipids社(CA、USA)から購入した。EDB-FN-特異的ペプチドN’-CSSPIQGSWTWENGK(C)WTWGIIRLEQ-C’は、Anygen社(光州、大韓民国)で注文製造した。マウス抗-EDB-FN抗体(mouse anti-EDB-FN antibody)、及び抗マウスフルオレセインイソチオシアネート(FITC)-コンジュゲート二次抗体(anti-mouse fluorescein isothiocyanate(FITC)-conjugatedsecondary antibody)は、アブカム社(Abcam)(Cambridge、UK)から購入した。ドセタキセル(DTX)及びセファロース(Sepharose)CL-4Bカラムは、シグマアルドリッチ社(Sigma-Aldrich)(MO、USA)から購入し、封入液(mounting solution)は、Dako Diagnostics社(Glostrup、Denmark)から購入し、アラマーブルーアッセイキット(Alamar Blue assay kit)は、サーモフィッシャーサイエンティフィック社(Thermo Fisher Scientific)(MA、USA)から購入した。全ての試薬は、ラボラトリグレード(laboratory grade)であり、提供されたとおり使用した。
U87MG、U251MG、U373MG、MCF-7、PC3、B16F10、及びB16F1細胞は、アメリカ培養細胞系統保存機関(American Type Culture Collection)(VA、USA)で購入した。全ての細胞は、湿度5%のCO2環境で37℃を保持した。細胞は、10%のウシ胎児血清(FBS;fetal bovine serum)(Gibco、IL、USA)、100U/mLのペニシリン(penicillin)(Gibco)、及び100μg/mLのストレプトマイシン(streptomycin)(Gibco)を添加して基礎培地(minimal essential medium)(MCF-7)、RPMI(PC3、B16F10、B16F1)、又はダルベッコ変法イーグル培地(Dulbecco’s modified Eagle’s medium)(U87MG、U251MG、U373MG)で培養した。全ての細胞培養培地は、Gibcoで購入した。
3D回転楕円体培養のために、全ての3つの悪性神経膠腫細胞株(U87MG、U251MG、and U373MG)をウェル(well)当たり1~5×103細胞(cell)でNunclon Sphera Microplate 96-well round bottom plate(サーモフィッシャーサイエンティフィック社)で培養した。プレート(plate)を2分間200xgで遠心分離した後、37℃、5%CO2培養基に置いた。細胞は、6日間培養し、3日目に培地の半分を変更した。画像化のために、形成された回転楕円体を4%(w/v)パラホルムアルデヒド(paraformaldehyde)(Wako、VA、USA)で固定された8-well chambered coverglass slide(サーモフィッシャーサイエンティフィック社)に移し、後述する免疫細胞化学(immunocytochemistry)が実行された。
脇腹皮下異種移植(flank subcutaneous xenograft)マウスモデル(n=1 mouse per group)を形成するために、BALB/cヌードマウスの右脇腹にマウス当たり5×106細胞で、U87MG、U251MG、及びU373MG細胞を注入した。腫瘍が80~120mm3の体積になると、マウスを犠牲にして、切り出した腫瘍の免疫細胞化学を行った。
細胞を収集して、RNAアイソレーションキット(RNA isolation kit)(GeneAll、ソウル、大韓民国)を使用してRiboExでリボ核酸(ribonucleic acid、RNA)を抽出した。各サンプルから合計1μgのRNAを使用して逆転写(reverse transcription)によって相補的DNA(complementary DNA、cDNA)を合成した。その結果、以下の遺伝子が推定された。EDB-FN遺伝子(順方向プライマー:5’-AACTCACTGACCTAAGCTTT-3’;逆方向プライマー:5’-CGTTTGTTGTGTCAGTGTAG-3’);及びグリセルアルデヒド3-リン酸脱水素酵素遺伝子(順方向プライマー:5’-AATCCCATCACCATCTTCCA-3’;逆方向プライマー:5’-TGGACTCCACGACGTACTCA-3’)。ポリメラーゼ連鎖反応プロトコルは、以下の通りである。94℃で5分間の最初の変性工程(initial denaturation step);94℃で1分間の変性、55℃で1分間のプライマー結合(annealing)、及び72℃で2分間の伸長(extension)を30回繰り返す工程、及び72℃で7分間の最終の伸長をする工程。4μLの超純水(ultrapure water)に1μgのcDNAを添加し、リアルタイムポリメラーゼ連鎖反応(real-time polymerase chain reaction)(Qiagen社、東京、日本)を実施する前に5μLのSYBR Green real-time mixture(Takara社、東京、日本)を添加した。2-ΔΔCt methodを使用して各遺伝子のメッセンジャーRNA(messenger RNA、mRNA)レベルを定量化し、グリセルアルデヒド3-リン酸脱水素酵素のメッセンジャーRNAで正規化した。
8.1.免疫細胞化学(immunocytochemistry)
7つの異なる細胞株を2D染色するために、染色前に8-well chamberedカバーガラススライドに24時間、10,000個の細胞を培養した。3D染色のために、回転楕円体(spheroids)を培養皿から8-well chamberedカバーガラススライドに移した。培養された細胞は、冷リン酸緩衝食塩水(cold phosphate buffered saline)(PBS;Welgene社、Gyeongsan、大韓民国)で3回洗浄した後、室温で15分間4%(w/v)パラホルムアルデヒド(paraformaldehyde)で固定し、PBSで洗浄し、0.1%(v/v)Triton X-100(Amresco社、PA、USA)/PBSで10分間透過し、4℃で一晩中1%(w/v)bovine serum albumin(BSA;Millipore社、MA、USA)/0.1%(v/v)Triton X-100/PBSで、primary antibody against EDB-FN(ab154210、1:500 for 2D、1:100 for 3D;Abcam)で培養した。PBSで洗浄した後、免疫標識されたタンパク質を、蛍光標識二次抗体(fluorescence-conjugatedsecondary antibodies)を室温で60分間処理して可視化した。細胞は、PBSで洗浄した後、4’,6-ジアミジノ-2-フェニルインドール(DAPI)封入剤(mounting medium)(Vector laboratories社、CA、USA)で封入し、カバースリップ(cover slips)で封止した後、共焦点レーザ走査顕微鏡(confocal laser scanning microscope)(LSM 700;Carl Zeiss社、NY、USA)を使用して検査した。
EDB-FNの染色程度は、患者組織サンプルに対して組織マイクロアレイを、皮下異種移植動物組織サンプルに対して免疫組織化学を適用して分析した。組織マイクロアレイスライドを55℃で30分間加熱して蜜蝋を除去した後、5分間3回洗浄して、100、95、及び80%(v/v)のエタノールで5分間洗浄して連続的に純粋な蒸留水になるように再水化(rehydration)した。10mMのクエン酸ナトリウム(sodium citrate)(pH 6.0)で95℃、30分間、区間を加熱することによって抗原回収(antigen retrieval)した。3%(w/v)の過酸化水素(hydrogen peroxide)で30分間培養することによって内因性ペルオキシダーゼ活性(endogenous peroxidase activity)を阻害した。室温で30分間universal blocking serum(Dako Diagnostics社)を使用して基本反応性(background reactivity)を除去した。スライドにEDB-FNに特異的な抗体(orb227981、1:50;Biorbyt社、Cambridge、UK)を処理して1時間培養した後、ビオチン標識二次抗体(biotin-labeledsecondary antibody)を処理して30分間培養した。Streptavidin-peroxidase(Dako Diagnostics社)を使用して開発した。ヘマトキシリン(hematoxylin)を処理して少し対照染色(counterstaining)した後、顕微鏡観察のためにスライドを脱水し、カバースリップで封入した。
9.1.APTEDB-conjugated PEG2000-DSPEの合成
追加のシステイン残基を含む(cysteinylated)EDB-FN-特異的アプタマー類似ペプチド(aptamer-like peptide)(APTEDB、Anygen社)をジメチルスルホキシド(Sigma-Aldrich社)に溶解し、Mal-PEG2000-DSPEをクロロホルム(chloroform)(Sigma-Aldrich社)に溶解した。APTEDB:Mal-PEG2000-DSPEのモル比を1:2とし、不活性条件において周辺温度(ambient temperature)で12時間コンジュゲーション(conjugation)反応を行った。APTEDB-conjugated PEG2000-DSPE(APTEDB-DSPE)を逆相高速液体クロマトグラフィー(reversed-phase high-performance liquid chromatography)で精製し、マトリックス支援レーザ脱離イオン化法/イオン化飛行時間質量分析(matrix-assisted laser desorption/ionization time-of-flight mass spectrometry)を用いてコンジュゲーション効率を決定した。コンジュゲーションされていないペプチドを除去するために、透析膜(dialysis membrane)(Spectrum Chemical社、NJ、USA)を使用して分子量3.5kDaに基づいて透析を行った。48時間後、コンジュゲーションを減圧下で凍結乾燥した。
原液(stock solution)から2mg/mLのPEG2000-DSPEの陰イオン脂質膜(anionic lipid film)を製造した。蛍光標識されたマイセルナノ-DDSを製造するために、PEG2000-DSPE溶液(solution)に0.5wt%のローダミンBフルオロフォアで標識されたRh-DSPEを添加した。製剤に1.0、2.5、又は5.0wt%の濃度でAPTEDB-DSPEを添加した。例えば、1.0wt%のAPTEDB-DSPEの活性標的APTEDB-DSPEマイセルナノ-DDS(APTEDB-conjugated)を形成するために、0.5wt%のRh-DSPEを含むPEG2000-DSPE溶液の2mg/mLに20μgのAPTEDB-DSPEコンジュゲート(conjugate)を添加した。陰性対照群として、受動的/非標的(passive/nontargeting)PEG2000-DSPEマイセルナノ-DDS(APTEDB-unconjugated)を合成した。全ての成分をガラスバイアルに入れ、真空下で乾燥し、一晩中減圧した後に凍結乾燥して残ったクロロホルムを全て除去した。最終的に2mg/mLの濃度のマイセル溶液を得るために、再水化の間、脂質膜に1mLの超純水(Welgene)を添加した。均一なサイズのマイセルを形成するために、毎分1,000回転(revolution)の一定の撹拌下で再水化を行った。Amicon(登録商標)ウルトラ15遠心式フィルタ(Ultra-15 centrifugal filter)3kDa Units(Merck社、Darmstadt、Germany)を使用して、マイセルを溶解した溶液をPBSバッファに変えた。超過サイズのナノ粒子又は凝集体を除去するために、溶液を0.1μmの膜(Millipore)で濾過し、サイズ排除クロマトグラフィー(CL-4B column;Merck社)で精製した。製造後、PBSで全てのマイセル製剤のサイズ及びゼータ電位を動的光散乱(dynamic light scattering、DLS)及びゼータ電位分析(zeta potential analysis)で分析した。2mg/mLマイセルの1mLを透明なキューベット(transparent cuvette)に移し、Zetasizer Nano ZS90(Malvern Instruments社、Malvern、UK)を使用して各マイセルの流体力学的直径(hydrodynamic diameter)及びゼータ電位を測定した。
受動的/非標的DSPE-DTX(APTEDB-unconjugated)及び活性標的APTEDB-DSPE-DTX(APTEDB-conjugated)は、従来に開示された方法を適用して開発された(Chong K et al.,Chem Commun(Camb).2013;49:11476-8)。先ず、APTEDB-DSPE脂質(lipid)を得るために、cysteinylated APTEDBをPEG2000-DSPE脂質(lipid)のマレイミド基(maleimide group)とコンジュゲートした。DTXをロードするために、DTXをクロロホルムに溶解し、再水化の間に最終的な濃度が50g/mLになるようにマイセル脂質膜に添加した。本発明において、DTX-ロードされたPEG2000-DSPEマイセルナノ-DDS及びAPTEDB-DSPEマイセルナノ-DDSをそれぞれ「DSPE-DTX」及び「APTEDB-DSPE-DTX」と命名した。DSPE-DTX及びAPTEDB-DPSE-DTXを0.1μmの膜で濾過し、サイズ排除クロマトグラフィーで精製した。
10.1.APTEDB-DSPEマイセルナノ-DDSのインビトロ細胞の取り込み(cellular uptake)
APTEDB-DSPEマイセルナノ-DDSの細胞の取り込み効率は、EDB-FN-陽性U87MG及びU251MG細胞に各マイセル製剤を処理することによって決定した。U87MG及びU251MG細胞は、5,000細胞/ウェル(cells/well)において96ウェルプレート(well plate)で培養した。細胞を殺菌されたカバースリップにおいてコンフルエンス(confluence)で培養した後、細胞をAPTEDB-DSPEを1.0、2.5、又は5.0wt%の濃度で処理し、100μg/mLのPEG2000-DSPEマイセルナノ-DDSs又はAPTEDB-DSPEマイセルナノ-DDSsと共に1時間37℃で培養した。細胞をPBSで洗浄し、4%(w/v)のパラホルムアルデヒド(paraformaldehyde)で固定し、nuclear dye DAPI(Invitrogen社、CA、USA)で対照染色した後、glass slidesで封入し、ローダミンB(rhodamine B)-標識されたマイセルの取り込み率を確認するために、共焦点レーザ走査顕微鏡で確認した。
EDB-FN高発現細胞(U87MG及びU251MG)及びEDB-FN低発現細胞(MCF-7及びB16F1)を約80%の合流点(confluence)に達するまでガラスカバースリップに培養した。細胞を様々な濃度(100μg/ml及び500μg/ml)の自由APTEDBペプチドで30分間前処理した。その後、ローダミン(rhodamine)標識されたAPTEDB-DSPEを添加し、細胞をさらに30分間培養した。その後、細胞をPBSで洗浄し、4%(w/v)パラホルムアルデヒド(paraformaldehyde)で固定した後、共焦点顕微鏡下で見るために顕微鏡スライドに封入した。
EDB-FN特異的siRNA及びスクランブル対照群siRNA(scrambled control siRNA)は、Bioneer社(Daejeon、大韓民国)から購入した。RNA干渉に使用されるEDB-FN siRNAの標的配列は、以下の通りである。sense;ACAGUCCCAGAUCAUGGAG、antisense;CUCCAUGAUCUGGGACUGU。Lipofectamine 2000(Invitrogen社、CA、USA)を形質導入するために、細胞を一晩中成長させた後、60~70%の合流点(confluence)で96ウェル又は6ウェルプレートで培養した。製造業者の説明に従ってLipofectamine-siRNA複合体を製造した。形質導入有効性は、48時間後に分析した。
EDB-FN発現量に応じた細胞内取り込みを確認するために、U87MG細胞に上述した対照群siRNA又はEDB-FN siRNAを形質導入した。形質導入した細胞に100μg/mLのPEG2000-DSPEマイセルナノ-DDS又はAPTEDB-DSPEマイセルナノ-DDSを1.0wt%の濃度で処理し、1時間37℃で培養した。APTEDB-DSPEは、EDB-FNに対抗する抗体を使用して免疫細胞化学によって染色し、DAPIを含む封入液(mounting solution)で封入した。時間依存的細胞の取り込みを確認するために、U87MG細胞に1.0wt%の濃度のAPTEDB-DSPEで100μg/mLのAPTEDB-DSPEマイセルナノ-DDSを5分、15分、30分、1時間、及び4時間処理した。
悪性神経膠腫の分子標的としてEDB-FNの有用性及び価値を決定するために、上述した方法でDTXをPEG2000-DSPE及びAPTEDB-DSPEマイセルナノ-DDSsの中心にカプセル化した。24時間、各ナノ-DDS製剤を段階的に希釈して細胞と共培養した。製剤を除去した後、細胞をAlamar Blue assay(Bio-Rad社、CA、USA)の前にさらに24時間培養した。各製剤のIC50値は、プロビット分析(ProBit analysis)によって決定した。
インビボ異種移植モデルで、APTEDB-DPSEマイセルナノ-DDSの取り込みを評価するために、BALB/cヌードマウス(n=3 mice per group)の右脇腹に5×106細胞/マウスにU87MG細胞を注入した。3週間後、腫瘍成長を測定し、この時点で腫瘍体積は80~120mm3であった。その後、200μgのPEG2000-DSPEマイセルナノ-DDS in PBS又はAPTEDB-DSPEマイセルナノ-DDS in PBSを各マウスに注入し、予め指定された時間(15、30、60、及び120分)に、IVIS in vivo imaging system(PerkinElmer社、MA、USA)を使用してローダミンB-標識されたマイセルの腫瘍取り込みを比較した。
APTEDB-DPSEマイセルナノ-DDSインビボ組織取り込みを評価するために、U87MG細胞を5×106の細胞/マウスでBALB/cヌードマウスの右脇腹に注入した(n=3 mice per group)。3週間後、腫瘍成長を測定し、腫瘍体積は、80~120mm3に決定した。その後、200μgのPEG2000-DSPEマイセルナノ-DDS又はAPTEDB-DSPEマイセルナノ-DDSを各マウスに注入し、予め指定された時間(6、12、24、及び48時間)に、IVISインビボ画像システム(PerkinElmer社、MA、USA)を使用してローダミンB-標識されたマイセルの腫瘍取り込みを比較した。
スライドガラスに取り付けられた皮下異種移植モデルの脳スライスを冷PBSで2回洗浄し、ブロッキングした後、ブロッキングバッファ(blocking buffer)(PBS containing 0.3% Triton X-100 and 2% BSA)で1時間透過できるようにした。EDB-FNに特異的な主要抗体(ab154210 Abcam、MA、USA)をブロッキングバッファで1:100に希釈し、組織と共に4℃で一晩中培養した。PBSで洗浄した後、ブロッキングバッファで1:200に希釈したAlexa Fluor 488コンジュゲート二次抗体(A11001;Invitrogen社、CA、USA)を処理し、室温で1時間培養した。組織を、4’,6-ジアミジノ-2-フェニルインドール(DAPI、Invitrogen社、NY、USA)で対照染色し、カバースライドで覆った後、共焦点レーザ走査顕微鏡及びスライドスキャナ(Axio Scan.Z1;Carl Zeiss社、NY、USA)を使用して分析した。
脇腹皮下異種移植マウスモデルを製造するために、BALB/cヌードマウスの右脇腹に5×106の細胞/マウスでU87MG細胞を注入した。腫瘍体積が80~120mm3になると、マウスに塩分を5mg/kgのDTX濃度で(代表的な腫瘍組織摘出のためのn=1 mouse per groupを含み、n=5 mice per group)DPSE-DTX in PBS、又はAPTEDB-DSPE-DTX in PBSを静脈注射した。以前の研究プロトコルに基づいて、各製剤を2日ごとに3回静脈注射した。腫瘍のサイズは、摘出するまで3日に1回測定した。以下の式を使用して腫瘍体積を算出した。(長さ×幅×高さ)×0.5。以下の式を使用して腫瘍抑制率を算出した。[1-{(TDayE-TDay1)/TDay1×CDay1/(CDayE-CDay1)}]=100(TDayE=実験群の最終腫瘍体積;TDay1=実験群の最初(1日)の腫瘍体積;CDayE=対照群の最終腫瘍体積;CDay1=対照群の最初の腫瘍体積)。
ImageJ software(http://rsb.info.nih.gov/ij/)を使用して画像プロセッシング及びデータ分析を行った。全てのデータは、GraphPad Prism 7 software(GraphPad,La Jolla社、CA、USA)を使用して分析し、means±standard errors(std.errors)で示すIC50値を除いて、means±standard deviations(std.devs.)で示した。群間の比較は、主に正規分布データの場合、対応のない2標本t検定(unpaired two-tailed t test)with Welch’s correction(ウェルチのt検定)、非正規分布データの場合、マン・ホイットニーのU検定(Mann-Whitney test)を行った。P values of<0.05は、統計的に有意であるとみなされ、以下のように表示された。p<0.05(*)、p<0.01(**)、p<0.001(***)、及びp<0.0001(****)。
動物実験のために全ての適用可能な国際、国内、及び/又は機関のガイドラインを遵守した。本発明における動物に関する全ての実験は、KAIST動物実験倫理委員会(KAIST Institutional Animal Care and Use Committee)及び高麗大学医科大学(Korea University College of Medicine)(IACUC No.KA2013-13及びKOREA-2019-0123)の倫理的基準に準拠する。患者サンプル研究は、高麗大学九老病院機関監査委員会(Korea University Guro Hospital Institutional Review Board)(IRB No.2017GR0330)で承認されたガイドライン及びプロトコルを遵守した。
本発明者らは、EDB-FNが様々な癌に対してマーカーであり、標的として実現可能であるかを評価するために、正規化されたビッグデータ及びEDB-FN発現レベルの患者組織サンプルを分析した。さらに、正常細胞に対する癌細胞におけるEDB-FNの発現率が最も高い癌の1つである悪性神経膠腫の診断マーカーであり、薬物伝達標的としてEDB-FNの有用性を検証した。
様々なヒト癌細胞株におけるEDB-FN発現レベルを決定するためにスクリーニングを行った。乳癌細胞株MCF7、前立腺癌細胞株PC3、黒色種細胞株B16F1及びB16F10、及び悪性神経膠腫細胞株U373MG、U87MG、及びU251MGが実験に使用された。EDB-FNタンパク質の発現パターンは、2D単層培養(two-dimensional monolayer culture)中に定量的分析によって確認した(図1A)。PC3細胞株及びU373MG細胞株では、EDB-FN発現が弱いと検出されたが、U87MG細胞株では、明確な発現が観察された。また、EDB-FN発現の定量的分析のためにmRNAを抽出することによって、qRT-PCRを行った(図1B)。その結果(MCF7、47.8±7.9;PC3、12.1±2.1;B16F1、0.6±0.5;B16F10、23.3±1.4;U373MG、2.6±0.8;U251MG、643.0±31.0;U87MG、1430.3±61.4)、U251MG細胞株において統計学的にかなりの過剰発現が観察され(p<0.001、他の全ての細胞株に対して、ウェルチのt検定)、最も高い発現は、U87MG細胞株で観察された(p<0.001、他の全ての細胞株に対して、ウェルチのt検定)。上記の結果を通して、他の癌に比べて悪性神経膠腫においてEDB-FNが過剰発現されたことを確認した。
マイセルの動的光散乱(DLS)分析の結果、11.5±1.9nm for the PEG2000-DSPEマイセルナノ-DDSに対して、11.5±1.9nmの直径、1.0、2.5、及び5.0wt%の濃度のAPTEDB-DSPEを処理したAPTEDB-DSPEマイセルナノ-DDSに対して、各々8.2±1.3nm、10.5±1.9nm、及び10.3±1.3nmの直径を示した(図2A及びB)。PEG2000-DSPEマイセルナノ-DDSと比較してAPTEDB-DSPEマイセルナノ-DDSのDLS-測定のサイズが減少したことと、1.0wt%の濃度で同じDDSと比較して2.5及び5.0wt%の濃度のAPTEDB-DSPEにおいてAPTEDB-DSPEマイセルナノ-DDSの測定されたサイズが増加したことは、マイセルの外面にAPTEDBの存在がマイセルナノ-DDSにおいて形態学的変化に影響を及ぼすことを示した。各ナノ-DDSのゼータ電位(ナノ粒子の表面電荷を推定することができる)を測定した。PEG2000-DSPEマイセルナノ-DDSのゼータ電位は、-9.3±1.1mVであり、1.0、2.5、及び5.0wt%の濃度のAPTEDB-DSPEを処理したAPTEDB-DSPEマイセルナノ-DDSのゼータ電位は、各々-9.5±0.7mV、-10.4±1.3mV、及び-12.1±0.7mVであった(図2C)。
最適化された標的配位子密度を決定するために、脂質膜形成前に、1.0、2.5、及び5.0wt%の濃度でPEG2000-DSPEを混合したRh-DSPEのクロロホルム溶液にAPTEDB-DSPEを添加した。全ての条件下で、癌細胞における活性標的APTEDB-DSPEマイセルナノ-DDSの取り込み効力(uptake efficacy)は、消極的/非標的PEG2000-DSPEマイセルナノ-DDSの取り込み効力よりも高かった(図4A)。興味深いことに、2.5wt%の濃度のAPTEDB-DSPEを処理したAPTEDB-DSPEマイセルナノ-DDSは、U251MG細胞で最も高い細胞の取り込みを示した(PEG2000-DSPE、44.5±9.6;1.0% APTEDB-DSPE、76.4±8.1;2.5% APTEDB-DSPE、128.8±12.1;5.0% APTEDB-DSPE、53.4±5.1)、一方、1.0wt%の濃度のAPTEDB-DSPEを処理したAPTEDB-DSPEマイセルナノ-DDSは、U87MG細胞で最も高い細胞の取り込みを示した(PEG2000-DSPE、6.8±5.1;1.0% APTEDB-DSPE、165.7±3.7;2.5% APTEDB-DSPE、93.2±6.7;5.0% APTEDB-DSPE、56.7±7.9)。APTEBD-DSPEの濃度が増加するにつれてより負の方向にあり(図2C)、5.0wt%の濃度のAPTEDB-DSPEを処理したAPTEDB-DSPEマイセルナノ-DDSの減少した細胞の取り込みは、ナノ-DDSにおける負電荷によって部分的に説明可能である。しかし、U87MG及びU251MG細胞の両方が悪性神経膠腫細胞であり、同じ標的配位子が使用されたにもかかわらず、最も高い細胞の取り込み効力を有するAPTEDB-DSPE濃度が細胞株ごとに異なった。従って、配位子密度は、細胞株及び細胞型によって異なり、それによって細胞の取り込みも異なることが分かった。
4.1.APTEDB-DSPE-DTXのインビトロ毒性
EDB-FNの悪性神経膠腫の分子標的として価値及び有用性を決定するために、PEG2000-DSPE及びAPTEDB-DSPEマイセルナノ-DDSの中心にDTXをカプセル化した。各マイセルナノ-DDSのローディング能力は10wt%、カプセル化有効性は約95%で算出した。細胞生存率(cell viability)は、DSPE-DTX及びAPTEDB-DSPE-DTXを用いて評価した(図5B)。DSPE-DTX及びAPTEDB-DSPE-DTXシステムにおいて、DTXがU251MG及びU87MG細胞の生存率を阻害するにもかかわらず、阻害の程度は異なった。U87MG細胞におけるIC50値は、DSPE-DTXの場合、87.38±6.87nM、APTEDB-DSPE-DTXの場合、23.15±1.67nMであり、APTEDB-DSPE-DTXがDSPE-DTXより約3.8倍低かった(p<0.0001、ウェルチのt検定)。U251MG細胞におけるIC50値は、DSPE-DTXの場合、43.16±7.05nM、APTEDB-DSPE-DTXの場合、26.20±1.53nMであり、APTEDB-DSPE-DTXがDSPE-DTXより約1.6倍低かった(p<0.05、ウェルチのt検定)(図5C)。U87MG及びU251MG細胞におけるインビトロ細胞毒性データは、優れた癌標的能力を暗示し、薬物の取り込みの増加は、消極的/非標的に比較してEDB-FN活性標的によって達成できると考えられる。DSPE-DTX及びAPTEDB-DSPE-DTXのIC50値の有意な差がU87MG細胞で観察されたが、U87MG細胞をインビボモデリング及び悪性神経膠腫の分子標的としてEDB-FNを評価するために使用することにより選別した。
U87MG脇腹異種移植マウスモデルにおけるリアルタイムIVIS画像の研究の結果、APTEDB-DSPEマイセルナノ-DDSは、消極的/非標的PEG2000-DSPEマイセルナノ-DDSと比較して腫瘍局在化(tumor localization)のかなりの増加を示した。腫瘍局在化の増加は、15分から120分の間持続的に観察された(図6A)。15分で、最小のナノ粒子蓄積が観察された。30分後、APTEDB-DSPEマイセルナノ-DDS群でより高い蓄積が開始された。60分後、APTEDB-DSPEマイセルナノ-DDS群は、PEG2000-DSPEマイセルナノ-DDS群より腫瘍部位でより多くの蓄積を示した。また、DDSが腫瘍より他の器官に影響を与えるか否かを決定するために、マイセルナノ-DDSの組織取り込みを48時間測定した(図7A)。消極的/非標的PEG2000-DSPEマイセルナノ-DDSは、徐々に48時間内に体内全般に拡散した。しかし、APTEDB-DSPEマイセルナノ-DDSは、安定的かつ限定された腫瘍部位として残った。上記の結果は、APTEDB-DSPEマイセルナノ-DDSの安定性及び高い悪性神経膠腫標的能力を示す。APTEDB-DSPEが高いEDB-FN標的能力を有することで、高い親和性でEDB-FNに結合することにより、悪性神経膠腫で非常に増加したAPTEDB-DSPEマイセルナノ-DDSの保持時間(retention time)を示し、それにより、増加した腫瘍における薬物の生物学的利用の可能性を有した。
インビボ抗癌効果は、U87MG皮下異種移植動物モデルで検証した。有効性は、経時的な対照群、DSPE-DTX及びAPTEDB-DSPE-DTX群における腫瘍成長の傾向を比較することによって評価した(図6B)。対照群では、腫瘍体積は、16日内(17日)に初日(1日)より約5.9倍増加した(1日:91.5±7.4mm3対17日:538.0±115.9mm3;p<0.01、ウェルチのt検定)。DSPE-DTX又はAPTEDB-DSPE-DTXを処理することによって腫瘍成長を著しく阻害した。上述のように、腫瘍阻害のパーセントを算出した結果、DSPE-DTXが腫瘍成長を約54.8%抑制した(1日:87.9±12.6mm3対17日:281.7±29.4mm3;p<0.001、ウェルチのt検定)、一方、APTEDB-DSPE-DTXは、腫瘍成長を約97.6%にかなり抑制することを確認した(1日:87.5±12.6mm3対17日:97.8±2.6mm3;p<0.20、ウェルチのt検定)。初日、腫瘍体積は群間に有意な差はなかった。しかし、ナノ-DDSを3回服用した2日目、4日目、及び6日目に、群間の腫瘍体積の差は、時間が経つにつれて顕著となった(図6C及びD)。APTEDB-DSPE-DTX群は、対照群と比較して7日目からかなりの腫瘍抑制を示し(p<0.001、ウェルチのt検定)、DSPE-DTX群と比較した場合、17日目から差があった(p<0.01、ウェルチのt検定)。
図7Bに示すように、マイセルナノ-DDSを体内投与した結果、心臓、肝臓、脾臓、肺臓、及び腎臓などの主要臓器では何の副作用も観察されず、実験が終了するまでマウスの体重には大きな変化がなかった(1日、対照群=20.4±0.6g、DSPE-DTX=21.4±0.2g、APTEDB-DSPE-DTX=18.9±0.6g;17日、対照群=19.8±1.6g、DSPE-DTX=20.7±1.2g、APTEDB-DSPE-DTX=19.5±0.9g)(図7C)。これは、APTEDB-DSPEマイセルナノ-DDSが毒性をほどんど有さず、生体適合性を有することを示している。
脳腫瘍の治療標的として、その実現の可能性を評価するために、U87MG同所異種移植動物モデルを作製した。図6Eに示すように、塩分(saline)を対照群として、DSPE-DTX及びAPTEDB-DSPE-DTXを細胞移植後に7日間静脈注射により注入した(n=4 mice per group)。2週間後、脳を抽出し、各群の腫瘍体積を比較した。H&E染色を使用してスライスされた脳組織の正常及び腫瘍部位の局在化(tumor localization)を行い、各モデルにおける最も大きい腫瘍のスライスに基づいて腫瘍のサイズを測定した(図6F及びG)。その結果、EDB-FN標的マイセルナノ-DDS処理により腫瘍成長が著しく阻害されることが分かった。対照群と比較して(116.9±21.0mm3)、DSPE-DTX(86.7±28.7mm3)及びAPTEDB-DSPE-DTX(46.5±27.0mm3)群は、各々25.8%(p<0.15、ウェルチのt検定)及び60.2%(p<0.01、ウェルチのt検定)の腫瘍成長を抑制した。統計学的に大きな差が見られなかったが、APTEDB-DSPE-DTX群は、DSPE-DTX群(p<0.09、ウェルチのt検定)より約34.4%さらに高い腫瘍成長抑制を示した。実験中、全ての群でかなりの体重変化が観察されることはなかった(1日、対照群=22.7±1.3g、DSPE-DTX=23.3±2.5g、APTEDB-DSPE-DTX=23.3±1.6g;21日、対照群=24.5±1.3g、DSPE-DTX=25.1±1.7g、APTEDB-DSPE-DTX=25.5±1.5g)(図8C)。上記の結果は、本発明のEDB-FN-標的マイセルナノ-DDSが悪性神経膠腫を治療する潜在力を有し、EDB-FNが薬物治療のための有用な標的であることを示している。
本発明は、癌細胞の表面及び細胞外基質内に位置するEDB-FNに関する。より具体的には、表面及び細胞外基質内のタンパク質は、薬物伝達体(drug delivery systems、DDSs)のための有用な標的だけでなく、癌診断バイオマーカーとして活用することができる。
Claims (20)
- PEG2000-DSPE重合脂質及びAPTEDB-PEG2000-DSPEポリマーを含むマイセル構造の薬物伝達体であって、
前記APTEDBは、フィブロネクチンエキストラドメインB(EDB-FN)遺伝子(Entrez Gene ID:2335)に特異的な結合力を示すアプチドである、
薬物伝達体。 - 前記薬物伝達体は、PEG2000-DSPE重合脂質100重量部基準でAPTEDB-PEG2000-DSPEポリマーを1から2.5重量部含む、請求項1に記載の薬物伝達体。
- 前記薬物伝達体の直径は、5.0から12.0nmであり、
ゼータ電位は、-8.0から-11.0である、請求項1に記載の薬物伝達体。 - 前記薬物伝達体は、血液脳関門(BBB)又は脳血管腫瘍障壁(BBTB)を通過して脳腫瘍細胞に特異的に薬物を送達する、請求項1に記載の薬物伝達体。
- 薬物がロードされた請求項1の薬物伝達体を有効成分として含む、脳腫瘍予防又は治療用の医薬組成物。
- 前記薬物は、造影剤又は抗癌剤であり、
前記抗癌剤は、ドセタキセル、ハラベン、ビンクリスチン、シスプラチン、ビンブラスチン、ビノレルビン、パクリタキセル(Paclitaxel)、エトポシド、トポテカン、イリノテカン、ダクチノマイシン、ドキソルビシン、ダウノルビシン、マイトマイシン、グリベック、カルボプラチン、バルルビシン、フルタミド、ゲムシタビン、ブレオマイシン、テモゾロミド、プロカルバジン、ロムスチン(CCNU;1-(2-chloroethyl)-3-cyclohexyl-1-nitrosourea)、ビンクリスチン及びカルムスチン(BCNU)からなる群から選択されるいずれか1つ以上であることを特徴とする、請求項5に記載の医薬組成物。 - 前記脳腫瘍は、星状細胞腫、多形性膠芽腫、乏突起細胞腫、乏突起星細胞腫、上衣細胞腫、血管芽細胞腫、血管芽細胞腫、髄膜腫、下垂体腺腫、頭蓋咽頭腫、及び脈絡叢乳頭腫からなる群から選択されるいずれか1つ以上であることを特徴とする、請求項5に記載の医薬組成物。
- 薬物がロードされた請求項1の薬物伝達体を個体に投与することを含む、脳腫瘍診断又は治療方法。
- 前記薬物は、造影剤又は抗癌剤であり、
前記抗癌剤は、ドセタキセル、ハラベン、ビンクリスチン、シスプラチン、ビンブラスチン、ビノレルビン、パクリタキセル(Paclitaxel)、エトポシド、トポテカン、イリノテカン、ダクチノマイシン、ドキソルビシン、ダウノルビシン、マイトマイシン、グリベック、カルボプラチン、バルルビシン、フルタミド、ゲムシタビン、ブレオマイシン、テモゾロミド、プロカルバジン、ロムスチン(CCNU;1-(2-chloroethyl)-3-cyclohexyl-1-nitrosourea)、ビンクリスチン及びカルムスチン(BCNU)からなる群から選択されるいずれか1つ以上であることを特徴とする、請求項8に記載の脳腫瘍診断又は治療方法。 - 前記脳腫瘍は、星状細胞腫、多形性膠芽腫、乏突起細胞腫、乏突起星細胞腫、上衣細胞腫、血管芽細胞腫、血管芽細胞腫、髄膜腫、下垂体腺腫、頭蓋咽頭腫、及び脈絡叢乳頭腫からなる群から選択されるいずれか1つ以上であることを特徴とする、請求項8に記載の脳腫瘍診断又は治療方法。
- (1)システイン残基を含むAPTEDB及びMal-PEG2000-DSPEを有機溶媒に1:2モル比で混合して重合反応を誘導する工程、
(2)前記(1)工程の混合溶液からAPTEDB-PEG2000-DSPEポリマーを得る工程、
(3)水、PBS、HBS、及びHBGからなる群から1種以上選択される溶媒に、前記APTEDB-PEG2000-DSPEポリマーとPEG2000-DSPE重合脂質とを混合してマイセル構造体形成を誘導する工程、及び
(4)前記(3)工程の混合溶液を濾過し、マイセル構造体を精製する工程、
を含む脳腫瘍特異的薬物伝達体の製造方法であって、
前記(1)工程のAPTEDBは、フィブロネクチンエキストラドメインB(EDB-FN)遺伝子(Entrez Gene ID:2335)に特異的な結合力を示すアプチドである、製造方法。 - 前記(1)工程の有機溶媒は、クロロホルム、ジメチルスルホキシド、ジメチルホルムアミド、エタノール、メタノール、テトラヒドロフラン、トルエン、キシレン及びヘキサンからなる群から選択される1種以上であることを特徴とする、請求項11に記載の脳腫瘍特異的薬物伝達体の製造方法。
- 前記(1)工程は、常温で12時間、不活性条件保持下で実行されることを特徴とする、請求項11に記載の脳腫瘍特異的薬物伝達体の製造方法。
- 前記(2)工程は、液体クロマトグラフィーを使用して混合溶液からAPTEDB-PEG2000-DSPEポリマーを得ることを特徴とする、請求項11に記載の脳腫瘍特異的薬物伝達体の製造方法。
- 前記(3)工程は、APTEDB-PEG2000-DSPEポリマーとPEG2000-DSPE重合脂質とを溶媒に混合し、音波破砕を行って水和させてマイセル構造体形成を誘導することを特徴とする、請求項11に記載の脳腫瘍特異的薬物伝達体の製造方法。
- 前記(4)工程は、0.025~0.1μmの膜で混合溶液を濾過し、サイズ排除クロマトグラフィーを介してマイセル構造体を精製することを特徴とする、請求項11に記載の脳腫瘍特異的薬物伝達体の製造方法。
- 請求項11の方法で製造された脳腫瘍特異的薬物伝達体であって、
前記薬物伝達体は、5.0から12.0nmの直径を有し、-8.0から-11.0のゼータ電位であるマイセル構造体であることを特徴とする、薬物伝達体。 - 請求項11の(1)~(4)工程を含み、
前記(3)工程は、APTEDB-PEG2000-DSPEポリマー及びPEG2000-DSPE重合脂質が溶解した溶液に抗癌剤をさらに混合することによりマイセル構造体形成を誘導するものである、脳腫瘍治療用の薬剤の製造方法。 - 前記抗癌剤を最終濃度が50g/mLになるように混合することを特徴とする、請求項18に記載の脳腫瘍治療用の薬剤の製造方法。
- 請求項18に記載の方法で製造された脳腫瘍治療用の薬剤であって、
前記薬剤は、血液脳関門(BBB)又は脳血管腫瘍障壁(BBTB)を通過して脳腫瘍内部に蓄積され、腫瘍細胞内部に陥入されることを特徴とする、薬剤。
