0.5 mL of fixation/membrane permeabilization agent (eBioscience Foxp3/Transcription Factor Fixation/Permeabilization Concentrate and Diluent, manufactured by Invitrogen) was added to the sample after centrifugation, stirred, and allowed to stand at room temperature for 30 minutes. 1 mL of a washing solution (eBioscience Permeabilization Buffer (10X), manufactured by Invitrogen) was added to the sample and stirred. The samples were centrifuged at 400 G for 5 minutes at room temperature and the supernatant was removed. After centrifugation, 1 mL of the washing solution (same as above) was added to the sample and stirred, followed by centrifugation at 400 G for 5 minutes at room temperature, and the supernatant was removed and stirred.

0.3 mL of PBS containing 0.5% BSA was added to the sample and stirred. The obtained sample was measured using a commercially available flow cytometer FACS Canto II (manufactured by Becton Dickinson) to detect Teff cells (CD62L low and CD4+ T cells) and Treg cells (CD25+, Foxp3+, and CD4+ T cells). Whole blood samples collected in heparin blood collection tubes were dispensed into centrifuge tubes. Red blood cells in the blood were hemolyzed by adding 2 mL of a hemolytic agent (CyLyse; manufactured by Sysmex Corporation) to a centrifuge tube and allowing it to stand at room temperature for 10 minutes. After stirring the centrifuge tube with a vortex mixer, 2 mL of PBS was added to the centrifuge tube. The tubes were centrifuged at 300G for 5 minutes at room temperature and the supernatant was removed. 2 mL of PBS was added to the centrifuge tube, and the centrifuge tube was again centrifuged at 300 G for 5 minutes at room temperature, the supernatant was removed, and a comparative sample was obtained. Regarding the sample of the comparative example, the same operation as in the example was performed, and measurement was performed using a flow cytometer.

ヘパリン採血管に採取した5mlの全血検体を、5mLの液体培地(10mM HEPES/PRMI-1640)で希釈した。10mLのリンパ球分離用媒体(Ficoll-Paque Plus、Cytiva社製)を50mLチューブに入れた。このチューブに、希釈された全血検体を入れて重層した。Ficoll Paque Plusの添付文書に記載のインストラクションにしたがってチューブ内でPBMCの分離を行い、CELLBANKER2(日本全薬工業社製)を用いてPBMCを1x10^6 cells/mLとなるように懸濁し、ディープフリーザーで凍結した。凍結処理後、PBMCを24時間以降1週間以内に液体窒素(液相下)へ移管した。解凍した細胞を、10% FBS/RPMI1640で洗浄後、10% FBS/RPMI1640を1x10^6 cells/mLとなるように添加した。2mLの細胞懸濁液を24穴プレート(コーニング社製) に入れ、48時間培養した。培養後、細胞培養液を15mL チューブに移し、400Gで5分間遠心分離して、対照例の試料を得た。対照例の試料について、実施例と同様の操作を行い、フローサイトメータによる測定を実施した。
