Abstract
Description
The present invention relates to a detection method for Salmonella infection using BamA protein, and an antigen reagent and kit for use in the detection method.
Salmonella spp. are Gram-negative bacilli subdivided into over 2,600 serotypes. Salmonellosis (livestock and poultry) and poultry salmonella infections are designated as notifiable and legally designated infectious diseases under the Act on Domestic Animal Infectious Diseases Control, respectively, and preventive measures against these infections are important. Infection of livestock and poultry with Salmonella spp. is characterized by fever, anorexia and lethargy, and causes acute sepsis and chronic diarrhea. However, the outbreak of salmonellosis in domestic and foreign livestock and poultry has not been completely controlled due to the limitations of scientific methods. In addition, salmonellosis-affected farms may have difficulty cleaning up the fungus.
Efforts to cleanse the bacteria include isolating and culturing the bacteria from the host and monitoring by measuring antibodies in serum. By estimating the timing of infection with Salmonella spp. by periodically measuring antibodies in serum, it becomes possible to take appropriate infection control measures.
On the other hand, when salmonellosis occurs on a farm, a vaccine may be administered to prevent the spread of infection. Salmonella vaccines currently inoculated to cattle and chickens in Japan are inactivated by formalin treatment, and are a very effective means for preventing infection.
However, when anti-Salmonella antibodies are measured in the sera of vaccinated individuals, they show a positive reaction, similar to Salmonella-infected individuals. In other words, vaccination results in a scientific flaw that makes it impossible to distinguish Salmonella-infected individuals from non-infected individuals. Therefore, it is desired to develop a test method capable of distinguishing Salmonella-infected individuals from vaccinated individuals.
As an inspection method capable of such discrimination, for example, an inspection method using a protein that constitutes flagellar antigens of Salmonella spp. is known (Patent Document 1).
The present invention has been made in view of such circumstances, and an object of the present invention is to provide a new method capable of detecting individuals infected with Salmonella spp. by distinguishing them from individuals vaccinated with an inactivated vaccine. is to provide Another object of the present invention is to provide reagents and kits for use in this detection.
As a result of intensive studies to solve the above problems, the present inventors found that BamA protein, which is a cell membrane protein of Salmonella spp. It has a high affinity for anti-Salmonella antibodies produced by individuals infected with the genus Salmonella, and by using the protein as an antigen for detecting anti-Salmonella antibodies, it can detect Salmonella infection with high accuracy. found that it is possible to On the other hand, when proteins (SseG protein and SseF protein) secreted from the type III secretion apparatus of Salmonella spp., cell membrane proteins (LptD protein) and LPS (Lipopolysaccharide) are used as antigens, , no significant difference in affinity to the two anti-Salmonella antibodies was observed. From the above, it was found that the specificity exhibited by the BamA protein is not a common phenomenon among the molecules that constitute Salmonella.
Based on the above findings, the present invention relates to a detection method for Salmonella infection using BamA protein, reagents and kits for use in the detection method, and more specifically provides the following.
(1) A method for detecting infection with Salmonella, which comprises immunologically detecting an anti-Salmonella antibody in a sample using BamA protein or a functional fragment thereof as an antigen.
(2)BamAタンパク質またはその機能的断片を含む、サルモネラ属菌感染を免疫学的に検出するための抗原試薬。 (2) an antigen reagent for immunologically detecting Salmonella infection, comprising a BamA protein or a functional fragment thereof;
(3) A kit for immunologically detecting Salmonella infection, comprising the antigen reagent according to (2).
INDUSTRIAL APPLICABILITY According to the present invention, it has become possible to detect Salmonella infection simply and with high accuracy using the BamA protein. Salmonella is a zoonotic pathogen that infects livestock and poultry, affecting their productivity and causing economic losses, as well as causing food poisoning in humans. By using the present invention, it becomes possible to efficiently prevent Salmonella infection in animals such as livestock and poultry, and in humans. In particular, in chickens, the serum of vaccinated individuals may show false positives against the rapid-aggregating brood diarrhea (poultry salmonella infection) diagnostic fluid used to diagnose brood dysentery (poultry salmonella infection). makes diagnosis difficult. The present invention can also be used as an adjunct to such existing diagnoses.
The present invention provides a method for detecting Salmonella spp. infection, which comprises immunologically detecting anti-Salmonella antibodies in a sample using BamA protein or a functional fragment thereof as an antigen.
Examples of "Salmonella spp." to be detected in the detection method of the present invention include Salmonella enterica subsp. Salmonella Typhimurium, Salmonella Choleraesuis, Salmonella Dublin, Salmonella Enteritidis, Salmonella Gallinarum, Salmonella Pullorum, etc. belonging to the genus Salmonella enterica, but these is not limited to
In the present invention, the "sample" used for detection of Salmonella infection is not particularly limited as long as it can contain anti-Salmonella antibodies. Generally, blood samples such as serum, plasma, and whole blood taken from individuals that can be infected with Salmonella spp. The organism from which the sample is derived is not particularly limited as long as it can be infected with Salmonella spp., and includes humans and other animals. The animal may be a domestic animal, a companion animal (pet), an experimental animal, or an animal for other purposes. Examples of animals include, but are not limited to, chickens, ducks, geese, ducks, quails, pheasants, pigeons, turkeys, guinea fowls, cows, pigs, horses, sheep, goats, dogs, cats, and the like.
These samples may be diluted or suspended in a diluent if necessary. Examples of the diluent include phosphate buffer, Tris buffer, Good's buffer, borate buffer, acetate buffer, citrate buffer, glycine buffer, succinate buffer, phthalate buffer, and the like. Buffers are included. In addition, the sample in the present invention may be appropriately pretreated as necessary. Examples of the pretreatment include treatments such as freezing, concentration, fractionation, and purification; It may be one or a combination of two or more.
In the present invention, an anti-Salmonella antibody in a sample is detected as an indicator of Salmonella infection. An "anti-Salmonella antibody" is an antibody that can specifically bind to the Salmonella spp., more specifically, an antibody produced in the body of an individual from whom a sample is derived. The isotype of the anti-Salmonella antibody may vary depending on the species, and may be, for example, IgY, IgG, IgM, IgA, or a combination of two or more of these. Moreover, each isotype may be of any subclass. The time of appearance and duration of anti-Salmonella antibodies in an individual's body after infection with Salmonella spp. can vary depending on the type of antibody. Accordingly, Salmonella infections detected in the present invention include current and past infections with Salmonella.
The present invention is characterized by using a BamA protein or a functional fragment thereof as an antigen for detecting anti-Salmonella antibodies. "BamA protein" is a cell membrane protein of Salmonella spp. A typical amino acid sequence of BamA protein is shown in SEQ ID NO: 2, and a typical nucleotide sequence of DNA encoding the protein is shown in SEQ ID NO: 1. However, it should be understood that in nature, the amino acid sequence of the protein may change due to genetic mutation. A "functional fragment" of the BamA protein can also be used as the antigen. Here, the functional fragment means a fragment of BamA protein having binding activity to anti-Salmonella antibody. The binding activity of the BamA protein fragment to the anti-Salmonella antibody can be evaluated by an immunological technique described below. The BamA protein or a functional fragment thereof may be modified such as mutagenesis as long as the binding activity to the anti-Salmonella antibody is not impaired.
A BamA protein or functional fragment thereof can be prepared as a recombinant or natural protein by methods well known in the art. Recombinant protein is obtained by inserting DNA encoding BamA protein (eg, DNA containing the nucleotide sequence of SEQ ID NO: 1) or DNA encoding a fragment of the protein into an appropriate expression vector, It can be prepared by introducing into a suitable host cell and purifying from an extract or culture supernatant of the host cell. Purification means include, but are not limited to, ion exchange chromatography, reverse phase chromatography, gel filtration, and the like. Also, when expressing the BamA protein or a fragment thereof in a host cell (e.g., E. coli, etc.) as a recombinant protein with multiple histidines or as a fusion protein with a glutathione-S-transferase protein, the expressed combination Recombinant proteins can be affinity purified using nickel or glutathione columns, respectively. After purification of the fusion protein, regions other than the target protein or fragment can be removed by cleaving with thrombin or factor Xa as necessary. The native protein can be prepared by methods known to those skilled in the art, for example, by contacting the Salmonella extract with an affinity column conjugated with antibodies against the BamA protein.
As the detection principle of the immunoassay method used in the present invention, the sandwich method is suitable in that a highly sensitive detection system can be constructed. In the sandwich method, an anti-Salmonella antibody is captured by a immobilized capture molecule, recognized by a detection molecule bound to a labeling substance, washed, and then detected according to the type of labeling substance. Examples of the solid phase include plates such as plastic plates, fibrous substances such as nitrocellulose, and particles such as magnetic particles and latex particles.
In the sandwich method, BamA protein or a functional fragment thereof is used as an antigen for at least one of the capturing molecule and the detecting molecule of the anti-Salmonella antibody. The other molecule is not particularly limited as long as it can bind to an anti-Salmonella antibody. For example, an antibody (secondary antibody) to an anti-Salmonella antibody or other protein that binds to an anti-Salmonella antibody can be used.
A labeling substance is usually bound to the detection molecule. Labeling substances are not particularly limited as long as they are detectable substances. and rhodamine isothiocyanate (RITC), fluorescent proteins such as allophycocyanin (APC) and phycoerythrin (R-PE), radioactive isotopes such as 125 I, gold particles, avidin, biotin, and latex. be done. When an enzyme is used as a labeling substance, various detections can be performed depending on the substrate by adding a chromogenic substrate, a fluorescent substrate, a chemiluminescent substrate, or the like.
When quantifying an anti-Salmonella antibody in a sample, a sandwich method such as ELISA using microplate wells or beads as a solid phase is preferred. Identification of the amount of anti-Salmonella antibody from the obtained measured value can generally be carried out by comparison with the measured value of a standard sample. In this case, for example, the amount of anti-Salmonella antibody in the sample can be determined by examining where the actual measured value is positioned on a standard curve created based on the measured value of the standard sample.
On the other hand, immunochromatography is suitable for simple and rapid detection of anti-Salmonella antibodies. To explain one aspect of the immunochromatographic device, the device comprises a detection zone in which capture molecules are immobilized in a line on a matrix made of a porous material such as a nitrocellulose membrane, and upstream of the detection zone, It includes a labeled reagent zone carrying labeled detection molecules. Typically, the labeled reagent zone consists of a porous pad carrying labeled detection molecules. A developing liquid tank storing a developing liquid is provided at the upstream end of the matrix. In addition, usually downstream of the detection zone, a development confirming section in which a molecule capturing the detection molecule is linearly immobilized in order to confirm the development of the labeled molecule for detection, and further downstream thereof, a development A developer absorption zone is provided with porous absorbent pads for absorbing liquid. Furthermore, when the label is an enzyme label, a substrate zone carrying a substrate for the labeling enzyme is provided upstream of the labeling reagent zone.
In use, the developer pad is inserted into the developer reservoir by adding a sample to the labeling reagent zone, pressurizing the indentation to move the protrusion, and supplying the developer to the matrix through the developer pad. When the developing liquid passes through the substrate zone, the substrate is eluted into the developing liquid, and the developing liquid containing the substrate flows. When the developing solution passes through the labeling reagent zone, the labeled detection molecules and the sample are eluted into the developing solution, and the developing solution containing the substrate, the labeled detection molecules and the sample flows. When an anti-Salmonella antibody is contained in the sample, the anti-Salmonella antibody and the labeled detection molecule bind to form a complex, and when this complex reaches the detection zone, the capture molecule on the detection zone and the anti-Salmonella antibody in the complex. As a result, the detectable molecule labeled via the anti-Salmonella antibody is immobilized in the detection zone. Anti-Salmonella antibodies are thus detected by measuring the label in the detection zone. If the sample does not contain anti-Salmonella antibodies, the labeled detection molecule will not be immobilized in the detection zone and will migrate further downstream, so no label will be detected in the detection zone. In addition, since the molecules that capture the detection molecules are immobilized in the developing solution confirming portion downstream of the detection zone, the labeled detecting molecules are immobilized in the developing solution confirming portion. Therefore, when the label is detected in the developing liquid confirming portion, it means that the developing liquid has been correctly developed. The developing solution is eventually absorbed by the downstream absorbent pad.
According to the detection method of the present invention, individuals infected with Salmonella can be distinguished from individuals vaccinated with inactivated Salmonella. Inactivated Salmonella used as a vaccine can be prepared by, for example, heat treatment of Salmonella, treatment with chemical disinfectants, irradiation with radiation or ultraviolet rays, treatment with a mutagen. Heat treatment or treatment with formalin, which is a chemical disinfectant, is preferably used as the inactivation treatment. The inactivated Salmonella may be a mixture of different Salmonella (eg, different species or different serotypes of Salmonella).

The antigen reagent may consist of only the antigen BamA protein or a functional fragment thereof, or may be one bound to a labeling substance, or one immobilized with a labeling substance. Moreover, the composition containing these may be used. Other components contained in the composition are not particularly limited, but include, for example, sterile water, physiological saline, surfactants, preservatives, etc., and even one of these may be used in combination of two or more. It may be a combination.
The present invention also provides a kit for immunologically detecting Salmonella infection, comprising the antigen reagent. In addition to the antigen reagent, the kit may further include components that should be included in the immunological detection method. Examples of such a configuration include, but are not limited to, standard samples (such as anti-Salmonella antibodies at various concentrations), control reagents, reaction buffers, diluents, washing solutions, and the like. When the labeling substance is an enzyme, it may further contain a substrate, a reaction stop solution, and the like necessary for detection and quantification of the labeling substance.
In the case of adopting the sandwich method for detection, when using the solid phase to which the BamA protein or its functional fragment is bound as a molecule for capturing the anti-Salmonella antibody (one aspect of the antigen reagent of the present invention), the kit of the present invention may further comprise a secondary antibody to which a labeling substance is bound as a molecule for detection of the anti-Salmonella antibody. Next, the PCR product was cloned using the TOPO cloning method, and then each Salmonella protein described in (1) was highly expressed in E. coli. pET151/D-TOPO was used as plasmid for protein expression. A His tag (6x) is added to the N-terminus of the protein expressed using this plasmid.
(3) Antigen Purification The Salmonella spp. was precultured (static culture) overnight in 5 mL of LB medium (containing ampicillin). Then, the mixture was cultured with shaking in 100 mL of LB medium until the absorbance (600 nm) reached 0.3, added with Isopropyl-β-D-thiogalactopyranoside, and further cultured for 6 hours. After culturing, the cells were recovered and disrupted by ultrasonic treatment. Then, the disrupted cells were dissolved in a urealysis buffer, various proteins were captured and purified using a nickel column, and eluted with an imidazole elution buffer.
LPS was purified from Salmonella spp. cultured overnight at 37° C. in 5 ml of LB medium using an LPS extraction kit (iNtRON).
(4) Chicken serum Chickens (white leghorn) were inoculated twice with Salmonella enterica serovar Typhimurium or inactivated vaccine (killed heat-inactivated bacteria) at 1 and 3 weeks after birth, and serum was taken.
In the Salmonella-infected group, the Salmonella enterica serovar Typhimurium aroA mutant strain (1×10 7 CFU/ml/200 μl PBS) was intravenously inoculated twice.
In the vaccinated group, Salmonella enterica serovar Typhimurium aroA mutant (1× 10 7 CFU/ml/200 μl PBS) inactivated by heat treatment at 100° C. for 5 minutes was intravenously inoculated twice.
(5) ELISA method An antigen (recombinant salmonella protein) adjusted to 0.5 µg/ml was allowed to react at 37°C for 1 hour to immobilize the antigen, and then blocked at room temperature for 1 hour using Blocking One (Nacalai Tesque). . Next, the sera of the Salmonella-infected group and the vaccinated group were each allowed to react with the immobilized proteins at 37°C for 1 hour. was reacted for 1 hour. Then, TMB (3,3′,5,5′-tetramethylbenzidine) was added as a substrate to develop the HRP label, and chicken IgY was quantified using an absorbance plate reader (450 nm).
B. Results (1) ELISA method using recombinant BamA protein in chickens as an antigen As a result of performing ELISA on chicken serum using BamA protein, which is a cell membrane protein, as an antigen, in the Salmonella spp. The serum antibody titer was higher than in the inoculated group (Fig. 1A). On the other hand, when the SseG protein and SseF protein secreted from the type III secretion apparatus, the LptD protein which is a membrane protein, and the LPS derived from Salmonella enterica serovar Typhimurium x3306 are used as antigens, respectively, the Salmonella-infected group and the vaccine No significant difference was observed in serum antibody titers between the inoculated groups (FIGS. 1B to 1E). From the above results, it was found that the BamA protein is an antigen capable of distinguishing between vaccinated individuals and Salmonella-infected individuals.
INDUSTRIAL APPLICABILITY According to the present invention, individuals infected with Salmonella can be identified by distinguishing them from individuals vaccinated with inactivated vaccines.
