血管内皮増殖因子(Vascular Endothelial Growth Factor、VEGF)は、非常に特異的な血管内皮細胞増殖因子であり、VEGFは、内皮細胞膜上のその受容体である血管内皮増殖因子受容体(Vascular Endothelial Growth Factor Receptor、VEGFR)に結合し、受容体の自己リン酸化を引き起こすことにより、マイトジェン活性化プロテインキナーゼ(MAPK)を活性化し、マイトジェン特性を達成し、内皮細胞増殖を誘導する。それが血管新生の特性を有するため、VEGFは、血液循環が不十分な時に組織への酸素供給を回復することができる。組織中のVEGFが過剰発現される場合、疾患症状を引き起こす可能性がある。例えば、VEGFの過剰発現は、糖尿病性網膜病等の網膜の血管疾患を引き起こす可能性がある。さらに、固形腫瘍は、成長に必要な栄養素を得るのに十分な血管の供給がない場合、一定のサイズ制限を超えて成長することはできないため、当該制限を克服するために、固形腫瘍は、VEGFを発現して、成長および転移を可能にする。

ヒトVEGFに特異的な単一ドメイン抗体を取得するために、まず、哺乳動物細胞HEK293Fを使用して、ヒトVEGFAタンパク質を一過性に発現させ、アフィニティー精製後にラクダ免疫に使用される。具体的な方法については、特許CN2018101517526実施例1および実施例2の方法の説明を参照する。簡単に説明すると、精製されたVEGFAタンパク質で2匹の新疆フタコブラクダを免疫し、7回の免疫後にラクダ末梢血からトータル(total)RNAを単離し、その後、逆転写およびPCRによってVHH遺伝子を増幅し、VHH遺伝子をバクテリオファージベクターpMECSにクローニングし、TG1に形質転換してバクテリオファージディスプレイライブラリーを構築する。構築されたライブラリーのライブラリー容量は、それぞれ6.4×108CFUおよび5.5×108CFUであり、挿入率は、それぞれ91.7%および95.8%である。その後、ライブラリーをスクリーニングし、二つのライブラリーをそれぞれ6ラウンドおよび5ラウンドスクリーニングして、抗体遺伝子を含むバクテリオファージの濃縮を取得する。各ライブラリーから300クローンを選択して、PE―ELISA同定を実施し、得られた陽性クローンをシーケンシングし、異なる配列の単一ドメイン抗体をFcと融合して発現させ、HEK293Fシステムを用いて、抗体を一過性に発現させ、発現方法は、特許CN2018101517526の実施例3の方法の説明を参照する。
ELISA法を使用して、ヒトVEGFAとVEGFR2との間の相互作用を遮断できる単一ドメイン抗体をスクリーニングする。(1)VEGFR2タンパク質をELISAプレート(1ug/mL、100uL/ウェル)にコーティングし、4℃下で一晩インキュベートし、(2)PBSTで5回洗浄した後、300uLの1%BSAブロッキング溶液を加え、37℃下で2時間インキュベートし、(3)PBSTで5回洗浄した後、勾配希釈した50uLの抗体サンプル(40ug/mLから2倍勾配希釈する)を加え、各ウェルに50uLの0.08ug/mLビオチン化VEGFAタンパク質を加え、37℃下で1時間インキュベートし、(4)PBSTで5回洗浄した後、100uLのSA―HRP(1:100000希釈)を加え、37℃下で1時間インキュベートし、(5)PBSTで5回洗浄した後、100uLのTMB発色溶液を加え、37℃下で10分間発色させ、50uL/ウェルの2M H2SO4を加えて反応を停止し、マイクロプレートリーダーで450nmの波長下で吸光度を測定する。結果は、図1に示されるように、Nb24の遮断活性が、対照抗体アバスチンの遮断活性よりも有意に優れている(IC50 Nb24=0.0149ug/mL、IC50 アバスチン=0.2172ug/mL)。
簡単に説明すると、方法は、次のとおりである。(1)よく成長したHUVEC細胞をトリプシン消化し、培地全体を中和し、PBSで1回洗浄し、3E4/mLの濃度で再懸濁し、100uL/ウェルを96ウェルプレートに分配し、37℃下で、5%CO2で20時間培養する。(2)翌日、2%FBSのDMEMでVEGFAを100ng/mLに希釈し、抗体を10000ng/mL、5000ng/mL、2500ng/mL、1250ng/mL、312.50ng/mL、78.13ng/mL、39.06ng/mL、9.77ng/mL、2ng/mLに勾配希釈する。(3)別の96ウェルプレートに60uLのVEGFAおよび等量の希釈好的抗体を加えて均等に混合し、37℃下で2時間共培養し、各混合液は三つのデュープリケートウェルにある。(4)インキュベーターから細胞培養プレートを取り出し、上清を吸引し、次に対応するウェルにそれぞれ100ul(3)の混合液を加え、37℃下で72時間インキュベートする。(5)72時間後に、10ul/ウェルのCCK8溶液を加え、2時間発色させ、発色終了後、マイクロプレートリーダーでOD450波長での吸光値を読み取る。結果は、図2に示されるように、HUVEC細胞の増殖に対する候補抗体Nb24の阻害効果は、対照抗体アバスチンの阻害効果よりも強い(IC50 Nb24=29.58ng/mL,IC50 アバスチン=72.28ng/mL)。
可変領域を変更せずに候補抗体をヒト化し、四つのフレームワーク領域の配列に対してヒト化設計を実行し、変更方法は、特許CN2018101517526の実施例4の方法を参照する。ヒト化後の抗体huNb24配列をpFUSEベクターに構築して、ヒト化された単一ドメイン抗体をFc配列と融合させ、HEK293Fシステムを使用して発現し、発現後のタンパク質は、後続の検証に使用されることができる。変更後の配列は、以下の表2に示されたようである。
上記ヒト化抗体を二価の形態に構築し、リンカーGGGGSGGGS(SEQ ID NO.:18)を使用して接続し、接続後のアミノ酸配列は、SEQ ID NO.:16に示されたとおりであり(対応するそのコーディングヌクレオチド配列は、SEQ ID NO.:17に示されたようである)、その後、ピキアパストリスによって発現される。簡単に説明すると、発現方法は、次のとおりである。(1)SEQ ID NO.:16に示される単一ドメイン抗体の二価体配列をpPICZaAベクターに構築し、(2)Sac I制限エンドヌクレアーゼでpPICZaA―Nb24―Nb24を線形化した後、X―33コンピテント細胞に電気形質転換し、(3)電気形質転換したサンプルを、様々な濃度のブレオマイシン耐性を含むYPDプレート培地にそれぞれコーティングし、30℃のインキュベーターで3~4日培養し、具体的な実施形態については、Invitrogen会社によって提供されるpPICZaAベクター説明書を参照することができ、(4)プレート培地でシングルクローンが成長した後、様々な濃度平のプレート上でシングルクローンを選択して、BMGY培地に入れ、BMGY培養液のOD値が約20に達する場合、菌体を収集した後BMMY培地に交換し、28℃下で、250rpmで培養し、(5)その後24時間ごとにサンプリングし、最終体積が1%であるメタノールを加えてサンプリングし、サンプルを12000rpmで5分間遠心分離し、上清を採集し、-20℃下で保存し、5日間連続して誘導し、培養を終了し、(6)採集したサンプルをSDS―PAGEで検出し、同時にサンプルを収集し、硫酸アンモニウム沈殿法により標的抗体を精製する。結果は、図3に示されたとおりであり、発現上清から硫酸アンモニウム沈殿によって精製された単一ドメイン抗体の二価体抗体hu bi―Nb24(Y)を、SEC―HPLCで検出したその純度は、94.11%であり、後続の研究に使用されることができる。
検出方法は、実施例2と同じであり、結果は、図4に示されるように、ヒト化抗体の遮断活性は、ヒト化前の抗体の遮断活性と同等であり(IC50 Nb24=0.045ug/mL、IC50 huNb24=0.038ug/mL)、従って、当該ヒト化の変更が成功したことを示す。ヒト化した単一ドメイン抗体と酵母で発現させた二価抗体との遮断活性を比較し、上記の実験を繰り返し、結果は、図5に示されるように、酵母で発現させた単一ドメイン抗体の二価体hu bi―Nb24(Y)の遮断活性は、3倍以上増加し(IC50 huNb24=0.044ug/mL、IC50 hu bi―Nb24(Y)=0.013ug/mL)、対照抗体アバスチンの遮断活性よりも有意に高かった(IC50 アバスチン=0.331ug/mL)。上記の実験方法を使用して、候補抗体と市販の類似製品との遮断活性を検出および比較し、結果は、図6に示されるように、酵母で発現させたヒト化二量体抗体hu bi―Nb24(Y)の遮断活性は、明らかな利点を有する(IC50 hu bi―Nb24(Y)=0.022ug/mL、IC50 アイリーア=0.085ug/mL、IC50 コンバーセプト=0.088ug/mL、IC50 アバスチン=0.439ug/mL)。これは、酵母で発現させたヒト化二量体が、既存の市販の製品よりも有意に優れた遮断活性を有することを示す。
検出方法は、実施例3と同じであり、結果は、図7に示されるように、HUVEC細胞の増殖に対する酵母で発現させたヒト化二価抗体hu bi―Nb24(Y)の阻害効果は、同様の市販の対照製品よりも優れる(IC50 hu bi―Nb24(Y)=53.59ng/mL、IC50 アイリーア=65.96ng/mL、IC50 コンバーセプト=129.7ng/mL、IC50 アバスチン=254.7ng/mL)。
ELISAを使用して、候補抗体がVEGF相同タンパク質と交差反応できるかどうかを検証する。(1)1ug/mLの試験抗体を4℃下で、100uL/ウェルのELISAプレートに一晩コーティングし、(2)PBSTで5回洗浄した後、各ウェルに300uLの1% BSAを加えて、室温下で2時間ブロックし、(3)PBSTで5回洗浄した後、100uLの1ug/mLビオチン(Biotin)―hVEGFA、ビオチン―hVEGFB、ビオチン―hVEGFC、ビオチン―hVEGFDを加えて、37℃下で1時間インキュベートし、(4)PBSTで5回洗浄した後、100uLの希釈したSA―HRP(1:5000希釈)を加えて、37℃下で1時間インキュベートし、(5)PBSTで5回洗浄した後、100uLのTMB発色溶液を加え、37℃下で10分間発色させ、50uL/ウェルの2M H2SO4を加えて反応を停止し、マイクロプレートリーダーで450nmの波長下で吸光度を測定する。結果は、図8に示されるように、ヒト化二価抗体hu bi―Nb24(Y)は、VEGFB、VEGFC、VEGFD等の他の相同タンパク質と交差反応せずに、ヒトVEGFAと反応することができる。
(1)VEGFR1タンパク質をELISAプレートにコーティングし(1ug/mL、100uL/ウェル)、4℃下で一晩インキュベートし、(2)PBSTで5回洗浄した後、300uLの1%BSAブロッキング溶液を加え、37℃下で2時間インキュベートし、(3)PBSTで5回洗浄した後、50uLの勾配希釈した抗体サンプルを加え(20ug/mLから2倍勾配希釈する)、各ウェルに50uLの0.08ug/mLビオチン化VEGFAタンパク質を加え、37℃下で1時間インキュベートし、(4)PBSTで5回洗浄した後、100uLのSA―HRP(1:100000希釈)を加え、37℃下で1時間インキュベートし、(5)PBSTで5回洗浄した後、100uLのTMB発色溶液を加え、37℃下で10分間発色させ、50uL/ウェルの2M H2SO4を加えて反応を停止し、マイクロプレートリーダーで450nmの波長下で吸光度を測定する。結果は、図10に示されるように、候補抗体hu bi―Nb24(Y)は、ヒトVEGFAとVEGFR1との間の相互作用を遮断することができ(IC50 hu bi―Nb24(Y)=0.168ug/mL)、遮断活性は、対照抗体アバスチンの遮断活性よりも優れる(IC50 アバスチン=0.967ug/mL)。
(1)血管新生を研究するための古典的なマウスモデル-OIRモデルを使用する。7日齢の雄C57BL/6Jマウスを75%酸素濃度の密閉容器で12日間飼育し、この期間中に、酸素ボックス内の酸素濃度を定期的にモニターリングして75%に維持し、その後、正常な空気環境下で5日間飼育する。P7(7日齢)―P12(12日齢)は、網膜血管閉塞期間に対応し、P12―P17(17日齢)は、低酸素症―異常な血管増殖期間に対応し、P17―P21(21日齢)は、異常な増殖血管回復期間に対応する。異常な血管新生の観察店としてP17を選択し、P12の場合、OIRモデル群のマウスの硝子体に異なる濃度の1uLの抗体薬物hu bi―Nb24(Y)を注射し(1.5mg/mL、1.0mg/mL、0.5mg/mL)、対照抗体アイリーアの濃度は、40mg/mLであり、その後、マウスを正常な空気に移して、P17まで飼育する。
(2)FITC―IB4網膜血管内皮細胞を網膜プレーティングのために染色し、OIRモデルにおいてP17の場合の異常な血管新生クラスターおよび網膜非灌流領域を観察する:C57BL/6Jマウスを犠牲にした後、マウスの眼球を直ちに取り出し、4%のパラホルムアルデヒドで1~2時間固定し、手術顕微鏡下で、角膜輪部に沿って球面を切断し、水晶体と硝子体とを除去し、網膜神経線維層および色素上皮を分離し、鞏膜、脈絡膜、網膜色素上皮層を除去し、網膜神経線維層をPBSですすぎ、残留硝子体を除去し、1mg/mLのBSAおよび0.3%TX―100を含むPBSで1~2時間ブロックし、FITC―IB4溶液(1:50希釈)に4℃下で48時間加え、取り出した後PBSで15分間×3回すすぎ、網膜神経線維層をスライドガラス上にプレーティングし、視神経乳頭を中心として放射状で切開し、封入剤を封入し、蛍光顕微鏡および共焦点顕微鏡下で観察および写真撮影しかつ統計する。
1mg/mLの濃度の候補抗体hu bi―Nb24(Y)を、それぞれ-20℃(凍結融解を繰り返す)、4℃、25℃、40℃の条件下で10mMのPB溶液に入れ、異なる時点でサンプルを採取してSEC―HPLC検出する。Adcance Bio SEC 130A 2.7um 7.8*300mmカラム(column)を使用し、検出波長は、280nmであり、室温下でタッセルは、0.5mL/minであり、200mMのpH7.0PB溶液を移動相として30分間イソクラティック溶離する。
Claims (16)
- 抗VEGF単一ドメイン抗体VHH鎖の相補性決定領域(CDR)であって、
前記VHH鎖の相補性決定領域CDRは、SEQ ID NO.:1に示されるCDR1、SEQ ID NO.:2に示されるCDR2、およびSEQ ID NO.:3に示されるCDR3を含むことを特徴とする、前記抗VEGF単一ドメイン抗体VHH鎖の相補性決定領域(CDR)。 - 抗VEGF単一ドメイン抗体のVHH鎖であって、
前記VHH鎖は、フレームワーク領域FRおよび請求項1に記載の相補性決定領域CDRを含み、好ましくは、前記フレームワーク領域FRは、
(a)SEQ ID NO.:4に示されるFR1、SEQ ID NO.:5に示されるFR2、SEQ ID NO.:6に示されるFR3、およびSEQ ID NO.:7に示されるFR4、または
(b)SEQ ID NO.:10に示されるFR1、SEQ ID NO.:11に示されるFR2、SEQ ID NO.: 12に示されるFR3、およびSEQ ID NO.:13に示されるFR4を含むことを特徴とする、前記抗VEGF単一ドメイン抗体のVHH鎖。 - 抗VEGF抗体であって、
前記抗VEGF抗体は、請求項2に記載のVHH鎖を有することを特徴とする、前記抗VEGF抗体。 - 前記抗VEGF抗体は、SEQ ID NO.:8またはSEQ ID NO.:14に示されるようなアミノ酸配列を有する一つまたは複数のVHH鎖を含むことを特徴とする、

(a)単一ドメイン抗体の生成に適した条件下で、請求項8に記載の宿主細胞を培養することにより、前記抗VEGF単一ドメイン抗体を含む培養物を得る段階と、
(b)前記培養物から前記抗VEGF単一ドメイン抗体を単離または回収する段階と、および
(c)任意選択で、精製および/または修飾して段階(b)のVEGF単一ドメイン抗体を得る段階とを含むことを特徴とする、前記抗VEGF単一ドメイン抗体を生成する方法。 - 免疫コンジュゲートであって、
当該免疫コンジュゲートは、
(a)請求項2に記載の抗VEGF単一ドメイン抗体のVHH鎖、または請求項3に記載の抗VEGF抗体と、および
(b)検出可能なマーカー、薬物、毒素、サイトカイン、放射性核種、酵素、金ナノ粒子/ナノロッド、磁性ナノ粒子、ウイルスコートタンパク質またはVLP、またはその組み合わせからなる群から選択されるカップリング部分とを含むことを特徴とする、前記免疫コンジュゲート。 - 請求項2に記載の抗VEGF単一ドメイン抗体のVHH鎖、請求項3に記載の抗VEGF抗体の用途であって、
(a)血管新生を阻害するための薬物と、
(b)VEGFに関連する疾患または病症を治療するための薬物の調製に使用されることを特徴とする、前記用途。 - 医薬組成物であって、
前記医薬組成物は、
(a)請求項1に記載の抗VEGF単一ドメイン抗体VHH鎖の相補性決定領域(CDR)、請求項2に記載の抗VEGF単一ドメイン抗体のVHH鎖、または請求項3に記載の抗VEGF抗体、または請求項10に記載の免疫コンジュゲートと、および
(b)薬学的に許容されるベクターとを含むことを特徴とする、前記医薬組成物。 - 抗体であって、
前記抗体は、請求項2に記載の抗VEGF単一ドメイン抗体の一つまたは複数のVHH鎖を含むことを特徴とする、前記抗体。 - 組換えタンパク質であって、
前記組換えタンパク質は、
(i)請求項2に記載のVHH鎖または請求項3に記載の抗VEGF抗体と、および
(ii)発現および/または精製に協力する任意選択のタグ配列とを有することを特徴とする、前記組換えタンパク質。 - サンプル中のVEGFAタンパク質を検出する方法であって、
前記方法は、
(1)サンプルを、請求項2に記載のVHH鎖、請求項3に記載の抗体または請求項10に記載の免疫コンジュゲートと接触させる段階と、
(2)抗原―抗体複合体が形成されるかどうかを検出する段階とを含み、ここで、複合体の形成は、サンプル中にVEGFAタンパク質が存在することを示すことを特徴とする、前記サンプル中のVEGFAタンパク質を検出する方法。 - VEGFAタンパク質検出試薬であって、
前記検出試薬は、
(i)請求項2に記載のVHH鎖、請求項3に記載の抗体、または請求項10に記載の免疫コンジュゲートと、および
(ii)検出学的に許容されるベクターとを含むことを特徴とする、前記VEGFAタンパク質検出試薬。
