Abstract
Description

CARを介した T 細胞認識は抗体領域によって定義され、 MHC の発現とは無関係です。 この認識は、抗体が使用可能 なあらゆるターゲットにまで拡張されます( CHMIELEWSKI 、 et al. 2013 )。 この相互作用は、腫瘍上の標的分子の構造と密度、および scFv と T 細胞膜間の配列が重要であり、柔軟性を提供する必要があることを示すエピトープの位置によって強く影響さ れます (HUDECEK 、 et at. 2015)。 スペーサの長さはターゲット分子によって異なる場合 があり、この論理的思考はビー固有CARの 2 つの認識部位間の領域にも適用できます( HUDECEK 、 et at. 2015; GRADA 、 et al. 2013 、 HEGDE 、 et al. 2016 )。 また、スペーサの特性は、トランスジェニック細胞表現型、活性化状態、移動能力、および腫瘍認識の変調において、CARの能力に影響を与える重要な要素となっています(WATANABE, et al. 2016; NORELLI 、 et al. 2016 )。
GRADA ( GRADA 、 et al e 2013 ) は CD19 と HER2 用のタンデムCARを構築しました。 2 つの認識モチーフの間のスペーサ / リンカーは 、一連の血糖 とセリン アミノ酸であった。 タンデムリピートは 、残基が 2,000 未満のタンパク質の少なくとも 14% で発生し、 αヘリックスやβシート などの標準的な二次構造を形成しません(MATSUSHIMA, et al. 2008 )。 Gly-Ser タンデムリピートは柔軟性が高く、切断可能でないため、CARサブユニットの動きがほぼ自由であることを示しています( MATSUSHIMA, et al. 2008 )。 タンデムリピート は、金属イオンやその他のタンパク質 を含む様々なリガンドとの相互作用を可能にする構造的柔軟性を可能にしているようです(MATSUSHIMA, et al. 2008 )。
IgG1 は最も豊富な免疫グロブリンクラスであり、例えば、CAR構造のスペーサと広く適用されています( VIDARSSON 、 et al 2014 )。 スペーサは、 IgG1 FC 領域全体(CH2CH3 )、 FC 領域とヒンジ、またはヒンジのみで構成されます。 ヒンジは CH1 領域とCH2 領域間の 15 個のアミノ酸 (VIDARSSON, et al 2014 )。 Guest et al は、 CD19 CAR の最適な機能 に IgG1 Fc 領域スペースが不要であることを示した( GUEST et al. 2005 ) およびは、鼠における CD19 CAR-T 細胞の有効性を廃止させることができます (ALMASBAK 、 et al 2015 )。 また、 Hombach( HOMBACH 、 et al 2000 ) は、抗 CD30 モデルにおける細胞外 IgG1 領域が抗原依存性細胞活性化を損なうすることを示しました。 一方で、いくつかの著者は IgG1 Fc 領域を用いた効率的なCAR構造を持っ ています。例えば CAR-PSCA や CAR-MUC1 (ATHAPAN et al. 2014 )、 NGFR間隔されたCD44v6 、 NGFR間隔されたCD19 、および NGFR間隔されたCEA ( CASUCCI 、 et al 2015 、 CASUCCI 、 et al 2018 )、 CAR-PSA ( WATANABE et al. 2016 )。 Moritz ( Moritz 、 et al 1995 ) は、機能CARの領域間でヒンジ領域を使用して、さまざまな構造をテストしました。 IgG1 Fc適応の懸念の 1 つは、この領域が補数カスケード 活性化の原因で あり、抗体 Fab 領域が削除 されると強化されることです( WANG 、 et al 2016 )。 免疫活性化における FC チェーンの推定効果にもかかわらず、 IgG1 を使用した実験結果は良好なスペーサ候補である可能性があることを示していますが、その影響は実験アッセイでのみ検出される可能性があります。
IgG4 クラスは IgG1 よりも豊富ではなく 、ヒンジ領域サイズに非常に類似した構造を持っています( 12aa )( VIDARSSON 、 et al 2014 )。 IgG4 抗体は、非感染性の環境で抗原に繰り返しまたは長期機関暴露された後に形成されることがよくあります。 ( VIDARSSON 、 et al 2014 )。 IgG4 は機能的に一価であり、望ましくない架橋にはあんまり適していないことを示し ます( AALBERSE 、 et al 2002 )。 これらの特性により、 IgG4 Fc およびヒンジは既にCAR構造に適用されています。 一般的に 、 IgG4 配列はヒンジ および Fc CH2CH3 領域( Qin 、 et al 2017 )適用されるか または他の IgG からの配列で構成されています。 IgG4 ( APEFLG )のCH2 領域の最初の 6 つのアミノ酸を、 IgG2 ( APPVA )の対応する 5 つのアミノ酸に入れ替えることは、 Fc 受容体への結合を無効にし、in vivoでの腫瘍認識に必要である ( HUDECEK 、 et al 2015 )。 IgG4 CH2 領域の変化の必要性は他の著者から報告され、 2 つの部位( L235E ; N297Q )を行うことでCH2 領域を変異させ、また削除が組み込まれているため抗原特異的溶解(JONNALAGADDA et al. 2015)を媒介することなく、Fc受容体結合を低減することができます。 IgG4 CH2 部分の欠失および突然変異は細胞毒性を21026廃止し、IgG4による補体活性化を大幅に減少させた。( Montano 、 et al 2002 、 DORAI 、 et al 1992 )。
CD4 、 CD8 および CD28 膜貫通型およびヒンジは、CAR構造に広く適用されている( NORELLI 、 et al 2016 )。 CD19 CARで は、 CD8 α または CD28のヒンジ領域および膜貫通型領域は鼠で同様の機能を持ちますが、 CD8 α は 炎症性部位カイン産生および活性化誘発細胞死 のレベルが低いように思われます( REN-HEIDENREICH 、 et al 2000 、 ALABANZA 、 et al 2017 )。 これらのモチーフは 通常、単一特異的なCARの scFv と T 細胞膜の間にスペーサとして適用されます。

今回の発明は、 RfuCAR と名付けられたタンデム受容体CARの二重得意を提案することを目的として います。 これには、腫瘍細胞( CD33 、 CD123 、または CD19 、 Mesothelin 、 BCMA などの他の腫瘍ターゲット)および IL-1 受容体タイプ 2 ( IL-1R2 )上の表面分子を認識してリガンド化する scFv と、腫瘍微小環境を変調する方法が含まれます。
構造体と機構は、細胞シグナル形質導入経路だけでなく、マクロファージやその他の細胞から分泌される分子にも腫瘍微小環境を制御するという新しいアプローチです。 In silico および in vitro 試験は 、Celluris RfuCAR アプリケーションの利点を実証するために実施されています。
今回の発明は、 RfuCAR と名付けられたタンデム受容体CARにおける二重特異性を記述 しています。 これには、腫瘍細胞( CD33 、 CD123 、CD19に似ているけど限定されてない 、 Mesothelin 、 BCMA などの他の腫瘍ターゲット)および IL-1 受容体タイプ 2 ( IL-1R2 )上の表面分子を認識してリガンド化する scFv と、腫瘍微小環境を変調する方法が含まれます。 さらに、このような機構は、規制安全スイッチを構成する両方の受容体エピトープペプチドの投与によってオフにすることができます(図 1 A および B )。
RfuCAR の細胞外推定構造 は、 IntFold の 2 つの構造予測オンライン機能で試験された(MCGUFFIN et al. 2010 ; MCGUFFIN 、 et al 2018 ; MCGUFFIN 、 et al 2015 ; BUENAVISTA、et al 2012 ; Roche 、 et al 2012 )と RaptorX ( K A LLBERG, et al 2012 、 MA 、 et al. 2012 ; Peng 、 et al. 2011 ; Peng 、 et al 2011 ; MA 、et al. 2013 )。 これらの機能は、 PDB データベース内の 2 次および 3 次構造の特性を持つ提出された一次構造と比較して推定構造を予測し、類似性を示すスコアを与えます。 また、無秩序領域と推定的結合部位も提供しています。
・ RaptorX
- スコア:は位置合わせスコアは 0 から(領域)シーケンス長までの間にあり、 0 は最悪を示します。 実際には、スコアは推定エラーのためにシーケンス長をわずかに超えてしまうことがあります。
- uSeqID と SeqID :はアライメント内の同一の残留物の数です。 SeqID は 、タンパク質(または領域)シーケンス長によって正規化されたuSeqID で、 100 を掛けたものです。 uSeqID ( SeqID )が高いほど、より良い結果が得られます。 SeqID が 30% を超え、タンパク質(または領域)の残基が 200 を超える場合、通常は予測モデルの正しい折り目があることを示します。
- uGDT および GDT : uGDT は、
1 * N( 1 )+ 0.75 * N( 2 )+ 0.5 * N( 4 )+ 0.25 * N( 8 )として定義された正規化されていない GDT (グローバル距離試験)スコアです。 N( x )は x より小さいモデリングエラーが予測される残留の数( Åに )です。 GDT は 、 uGDT をタンパク質(または領域)の長さで割って計算 され、 100 を掛けた値です。 uGDT ( GDT )は絶対モデルの品質を測定します。 残基が 100 を超えるタンパク質の場合は、 uGDT >50 が良好な指標となります。 残基が 100 以下のタンパク質の場合、 GDT >50 は良好な指標です。 モデルに適切な uGDT (>50) があっても GDT (<50) が正しくない場合は、モデルの小さい部分だけが適切である可能性があることを示します。
- P 値: は予測モデルがこのタンパク質(または領域)に対してランダムに生成される一連のモデルのベストよりも悪い可能性があるため、 P 値はモデルの相対的な品質を評価します。 P 値が小さいほど、モデルの品質が高くなります。 主にαタンパク質の場合、10^-3 未満の P 値 は良好な指標となります。 マンリーベータタンパク質の場合、10^-4 未満の P 値 は良好な指標となります。
結果テーブルは、グローバルモデル品質スコアの減少に従ってランク付けされます。 グローバルモデル品質スコアの範囲は 0 ~ 1 です。 一般的 に、 0.2 未満のスコアは領域のモデル化が誤っている可能性があることを示し、 0.4 を超えるスコアは一般に、ネイティブ構造と非常に似た完全で信頼性の高いモデルを示します。 各モデルに は、 p 値に応じて色分けされた信頼レベルも割り当てられます。
- RasMol 温度カラーリングスキームに従って、残留誤差で色分けされたモデルの 3D カートゥーンビュー。
- 障害予測 - この画像は、シーケンス内のNo.付きアミノ酸( x 軸上)ごとに、障害の確率( y 軸上)のプロットを示しています。無秩序 /秩序確率のしきい値は、プロット上に破線で表示されます。 しきい値を超える残留物は、ほとんどが順不同以下であると考えられますが、このしきい値はユーザーを誘導するためにのみ使用されます。
- 領域境界予測 - イメージには、予測される領域を示すように色分けされた予測される最上位 3D モデルが表示されます。
- 色の変化は、領域境界の可能性を示します。
- 結合部位予測 - 画像には、推定上の結合部位の残基を示す注釈が付けられた、予測される最上位 3D モデルが表示されます。 モデルのカートゥーンビューは青で表示 され、結合部位はラベル付きの残留物が付いた青い棒で表示されます。 結合残留物のリストは、最も可能性の高い(多数の)リガンド、予測結合ポケットの中心に最も近いリガンド、相互作用する可能性のあるリガンドのリスト、および関連するテンプレート構造で識別されたリガンドの数とともに提供されます。

- SEQ IDNo.1~6で構成されるRfuCAR抗CD33及び、SEQ IDNo.7~12で構成されるRfuCAR抗CD123であることを特徴とするタンデム受容体CARにおける二重特異性。
- SEQ IDNo.2で構成されるRfuCAR抗CD33であることを特徴とする請求項1に記載のタンデム受容体CARにおける二重特異性。
- タンデム受容体CARにおける二重特異性は、主にSEQ ID No. 2で表されるモデル2で特徴づけられ、50 、 227 、 228 、 229 、 272 、および 360のRfuCAR抗CD33の推定結合部位と位置であることを特徴とする請求項2に記載のタンデム受容体CARにおける二重特異性。
- 各部位で最も可能性の高いリガンドはILE, FULである特徴づけられている;各部位の中心点リガンドは SER657 、 FUL641 、でクラスタ内の全てのリガンドはGLY-1 、 TRP-1 、 GLU-2 、 ILE-3 、 PRO-3 、 SEL-2 、 THR-2 、 TYR-3 、 ASP-1 、 LEU-1 、 ASN-1 、 FUL-1 、 FUC-1;そしておそらく 中心点リガンドは ILE650 、 FUL641 であることを特徴とする請求項2に記載のタンデム受容体CARにおける二重特異性。
- SEQ IDNo.8で構成されるRfuCAR抗CD123であることを特徴とする請求項1に記載のタンデム受容体CARにおける二重特異性。
- タンデム受容体CARにおける二重特異性は、主にSEQ ID No. 8で表されるモデル8で特徴づけられ、33 、 235 のRfuCAR抗CD123の推定結合部位と位置であることを特徴とする請求項5に記載のタンデム受容体CARにおける二重特異性。
- 各部位で最も可能性の高いリガンドは TYR;各部位の中心点リガンドは PRO656 、クラスタ内のすべてのリガンドは GLY-3 、 TRP-2 、 GLU-2 、 ILE-3 、 PRO-4 SER-3 、 ARG-1 、 THR-2 、 TYR-5 、 ASP-1 、 LEU-1 、 ASN-4 、 ALA-2 、 CYS-1であることが特徴づけられ、中心点リガンドは TYR672であることを特徴とする請求項5に記載のタンデム受容体CARにおける二重特異性。
- CD33、CD123などの腫瘍細胞上の表面分子や、CD19、Mesothelin、BCMAなどの別の腫瘍標的、IL-1R2などのIL-l受容体2型を認識して結合するscFvを含むことを特徴とする、請求項1から7のいずれか1項に記載のタンデム受容体CARにおける二重特異性。
- 分泌された IL-1 β が 、IL-1R1 および IL-1 シグナリング形質導入への結合を阻害し、このような阻害のIL-1 経路の活性化が減少させ、腫瘍細胞増殖の変調が促進される、RfuCAR の IL-1R に結合を閉じ込められることを特徴とする腫瘍状微小環境を変調する方法。
- 微小物環境における IL-1 含有量の規制は、腫瘍環境における他の部位カイン活性化を変調し、CRS および神経毒性イベントで観察されるサイトカインの過剰分泌を予防することを特徴とする、請求項9に記載の腫瘍状微小環境を変調する方法。
- RfuCAR 細胞の作用は、有毒なCAR-T効果が患者で検出された場合、IL-1R 、 IL-1-IL-1R 、および/または scFv エピトープにリンクする異なるペプチドの投与によって一時的にスイッチオフおよび/または調整できることを特徴とする、請求項9に記載の腫瘍状微小環境を変調する方法。
- 腫瘍微小環境が急性骨髄性白血病、急性リンパ芽球性白血病、膵臓、肺癌、および卵巣癌など主に急性骨髄性白血病のような任意の種類の癌で構成されていることが特徴とする請求項9~11のいずれか1項に記載の方法。
