標的分子への抗体の結合に関して、特定の抗原(例えば、ポリペプチド標的)または特定の抗原上のエピトープ「と結合する」、「と特異的結合すること」、「に特異的に結合する」、「に特異的である」、「と選択的に結合する」、及び「に選択的である」という用語は、(例えば、非標的分子との)非特異的または非選択的相互作用とはある程度異なる結合を意味する。特異的結合は、例えば、標的分子への結合を測定し、それを非標的分子への結合と比較することにより測定することができる。特異的結合は、標的分子上で認識されるエピトープを模倣する対象分子との競合により決定することもできる。その場合、標的分子への抗体の結合が対象分子により競合的に阻害される場合、特異的結合が示される。

調製方法

一態様では、本出願は、非刺激性骨髄細胞を、ヒト抗体などの抗TREM2抗体と接触させる方法を提供し、これにより、非刺激性骨髄細胞の無効化がもたらされる。

クローン#237920のヒト化
マウス及びヒトTREM2に特異的なモノクローナルラットIgG2Bクローン#237920(R&D Systemsカタログ番号MAB17291)を、配列決定及びヒト化に使用した。簡単に言うと、抗体のジスルフィド結合をジチオスレイトール(DTT)で還元し、遊離のスルフヒドリル基をヨードアセトアミドでアルキル化した。アルキル化された抗体を配列決定グレードのエンドプロテイナーゼで消化させ、スピンカラムを使用して精製し、配列をLC-MS/MS分析で決定した。配列は、以下に示される。

mAb配列決定分析のために、モノクローナル抗体(mAb)の溶液中エンドプロテイナーゼ消化を実施した。50μgの抗体をDTTで還元し、ヨードアセトアミドを使用してアルキル化し、アセトン沈殿させ、1μg/μLの濃度で、水中で再構成した。製造者の使用説明書に従って、5つの個々の酵素消化:Asp-N、キモトリプシン、エラスターゼ、トリプシン、及びペプシンを使用して、抗体試料の溶液中消化を実施した。次に、試料を、凍結乾燥し、0.1%のTFAに再懸濁し、C18 Zip-Tipを使用して精製した。次に、試料を、真空遠心分離で乾燥させ、質量分析まで凍結保存した。
インタクト質量測定
mAbの試料を、変性させ、還元し、酸性化した。次に、Waters QToF Ultima Global質量分析計(LC-ESI-TOF MS)に接続されたAgilent 1100 HPLCを使用して、タンパク質を分析した。Waters MassLynx4.1ソフトウェアを使用して、適切なLC-MSスペクトルを処理(結合、減算、平滑化、デコンボリューション)した。
精製されたペプチドを0.1%のギ酸中で再懸濁し、消化物のそれぞれの半分を、ナノスプレー源及びEASY-nLC 1000システム(Thermo Fisher Scientific)が装備されたOrbitrap分析計(Q-Exactive,Thermo Fisher Scientific)で分析した。800バールの圧力で、PepMap(登録商標)RSLC 2μmのC18樹脂(Thermo Fisher Scientific)が充填された50cm(75μmの内径)EASY-Sprayカラムに、ペプチドをロードした。60分かけて、0.1%のギ酸中の0%~30%のアセトニトリルに設定された勾配を使用して、250mL/minの速度で、ペプチドを溶出させた。ナノエレクトロスプレーイオン源により、Q-Exactive質量分析計(Thermo Fisher Scientific)に、ペプチドを導入した。計器方法は、自動利得制御(AGC)標的が1E6であり、最大イオン注入時間が120msであり、解像度が70000であるOrbitrap質量分析計内の1つのMSフルスキャン(400~1600m/z)、続いて、解像度が17500であり、AGC標的が5E5であり、最大イオン時間が100msである10データ依存性MS/MSスキャン、及び1つのマイクロスキャンからなる。MS/MSスキャンを起動させる強度しきい値を、1.0%のアンダーフィル比に設定した。正規化されたコリジョンエネルギーが30に設定されたHCDコリジョンセル中で、フラグメンテーションが発生した。8秒のセッティングを使用して、動的除外を適用した。
抗体の産生及び特性決定
標準的な方法を使用して、標準的なタンパク質発現ベクターをHEK293にトランスフェクトし、その後、細胞を7日間成長させ、採取した。HEK293に加えて、CRISPR/Cas9編集(Alexander Weiss、トロント大学)により、哺乳動物のa1,6フコシルトランスフェラーゼ(FUT8)を欠損させた293細胞においても、抗体を産生した。上清のpHを、1MのHepes pH7.4で調整し、微生物の成長を防止するために、アジ化ナトリウムを添加した。KanCap A樹脂を使用して、タンパク質を捕捉し、1Mの塩化ナトリウムを含有するPBS及びPBSで抗体を洗浄した後に、50mMのクエン酸 pH3.5、100mMのNaClで溶出させた。溶出後直ちに、0.5Mのアルギニンを含有する1MのTris(pH8)で溶液を中和した。標準的技術を使用して、PBSにバッファー交換されたタンパク質に対する生物物理学的特性決定を行った。タンパク質をOD280で定量化し、計算された吸光係数を使用して、量及び濃度を決定した。還元及び非還元SDS-PAGE(Biorad基準トリス/グリシン/SDS、4~20%)またはPerkin Elmer GXIIキャピラリー電気泳動システムを使用して、純度及びおおよその分子量を決定した。Sepax Zenix-C SEC-300、3um、300Å、4.6*150mmサイズ排除カラム及びPBSランニング緩衝液を使用する、280nmでの検出を用いるHPLCで、凝集状態を決定した。
アミンカップリングによりチップに直接固定化されたヒトTREM2 His(Sino Biological、中華人民共和国北京)または抗Hisキャプチャーを通したSeries S CM5 chips上で捕捉されたヒトまたはTREM2ヒトIgG1 Fc融合タンパク質(95%超純度に精製された社内SEC)を用いる、Biacore T200(GE Healthcare、英国)を使用した表面プラズモン共鳴で、結合動力学を決定した。示された抗体の連続希釈液を30ul/分で2分間注入した。次に、PBSまたはシステム緩衝液を30ul/分で400秒間注入して、解離を観察した。ブランクフローセルの応答を減算することにより、結合応答を修正した。速度論的分析では、kon値及びkoff値のグローバルフィッティングの1:1ラングミュアモデルを使用した。Kd値を、kon及びkoffの比から決定した。
細胞結合(EC50測定):
ヒトまたはマウスTREM2を過剰発現する100,000~500,000のExpi293親細胞またはExpi293細胞を、96ウェルプレートにプレーティングし、死細胞を、Zombie Near Infrared(Biolegend)で染色した。示された非コンジュゲート抗体の滴定を、8~10点にわたって、1:3の希釈範囲で、0ug/ml~10ug/mlの範囲内で、これらの細胞とともにインキュベートした。それらのアイソタイプ(hIgG1またはmIgG2a)に応じて、これらの1次非コンジュゲート抗体を、Alexa Fluor 647コンジュゲート抗ヒトFcまたは抗マウスFc2次抗体(Jackson Immunoresearch)で検出した。Alexa Fluor 647シグナルを、フローサイトメトリー(BD Fortessa X-14、BD Biosciences)で測定した。Graphpad Prism(Graphpad Software)で、HEK293親細胞から生じたバックグラウンド蛍光を介して、過剰発現細胞に結合する抗体から生じたシグナルをカーブフィッティングすることにより、EC50値を計算した。
材料及び方法
CT26.WT(CRL-2638)細胞を、American Type Culture Collection(ATCC)から購入した。in vivoでの使用のための抗体を全て、エンドトキシンについて試験し、0.2EU/mg以下のタンパク質を使用した。クローンRMP1-14(Absolute Antibody Inc.、カタログ番号Ab00813-7.1)由来の抗マウスPD-1抗体のアミノ酸配列を、質量分析(LC-MS/MS)で決定した。文献(Nimmerjahn and Ravetch2005 Science 310:1510-15121)に記載されるように、FcgRへの結合を排除するために、マウスIgG1バージョンのRMP1-14抗体のFc領域に、単一点変異[D265A]を導入した。マウスIgG1[クローンMOPC-21]、及びマウスIgG2a[クローンC1.18.4]アイソタイプ対照を、BioXCellから購入した。PI-7012及びAfuc-PI-7012(PI37012のCDR配列を有し、マウスIgG2aフォーマットでマウス化された)を、それぞれ、マウスIgG2aフォーマットのExpi293細胞(Thermo Fisher Scientific)または293/FUT8ノックアウト細胞(University of Toronto)で産生させ、MabSelectプロテインAレジン(GE Life Sciences)を使用して精製した。抗体を、0.1Mのクエン酸緩衝液(pH3.0)で溶出させ、使用前に緩衝液を交換した。
糖鎖工学を介して特定のFcgRに結合するmAbの親和性(すなわち、抗TREM2 mAbのアフコシル化バージョンを生成することによる)は、抗腫瘍活性を増加させ得るかどうかを、本発明者らは判定した。CT26腫瘍モデルにおいて、PI-7012及びafuc-PI-7012を抗PD-1と組み合わせて試験した。PI-7012及びafuc-PI-7012は、類似のレベルの腫瘍成長阻害を示した(79%対88%のTGI)。afuc-PI-7012を用いる処置は、30%の治癒率をもたらした。図1Aにみられるように、afuc-PI-7012は、抗PD-1と組み合わせると、PI-7012よりも抗腫瘍活性を増加させている。抗腫瘍活性に対するPI-7012のアフコシル化の影響は、個々のマウス腫瘍体積の分析で、より明確にみられた(図1B及び1C)。これは、抗TREM2抗体のアフコシル化が、コアフ-シル化抗体よりも有意な治療上の利点を提供することを示す。
材料及び方法
上記の例で処置されたマウス由来の組織(肺、肝臓、脳、腎臓、及び心臓)を、10%の中性緩衝ホルマリン中に少なくとも24時間保存し、組織学のために規定通りに処理し、5~6μmで切断し、切片をヘマトキシリン及びエオシンで染色した。染色されたスライドを、低倍率(40~100×)光学顕微鏡を使用して試験し、画像をHisto Wizにより得た。抗CD68抗体(AbD Serotec)を使用して、CD68陽性細胞を検出し、光学顕微鏡を使用して、40×切片の8~9つの視野を定量化した。
処置後のマウス組織(肺、肝臓、心臓、腎臓、及び脳)のH&E染色による全体的な形態学的分析は、アイソタイプ対照で処置されたマウスと比較して、PI-7012、afuc-PI-7012、及び抗PD-1の組み合わせで処置されたマウスにおいて、いかなる形態学的変化も認められなかった(図3は、肺組織の染色を示す)。
材料及び方法
全ての動物試験は、the Murigenics Animal Studies Committeeにより承認された。C57BL/6J-Trem2em2Adiuj/J(以下、TREM2KOと呼ばれる)及び対照C57BL/6Jマウスは、Jackson研究所のものである。完全な肺、脾臓、及び骨を採取し、フローサイトメトリーのためにすぐに処理した。並行して、血液を心臓穿刺で採取した。Miltenyi MACS組織解離キットを使用して、組織を単一細胞懸濁液に処理した。1xの赤血球溶解緩衝液(Biolegend)を使用して、赤血球を溶解させた。細胞表面染色のための処理の前に、細胞を、Fixable Viability Dye(ThermoFisher Scientific)で染色した。抗マウス免疫表現型抗体は、Fcブロックとともに、FACS緩衝液(2%のFBS、2mMのEDTA、1×PBS)で希釈し、氷上で30分間染色した。染色後、細胞をFACS緩衝液で2回洗浄し、次に、PBS中の2%のパラホルムアルデヒドで15分間固定した。全てのデータをLSR Fortessaフローサイトメーター(BD)またはAttuneフローサイトメーター(Thermo Fisher)で収集し、FlowJoソフトウェアを使用して分析した。TREM2KO細胞染色は、灰色のプロットで示され、野生型細胞染色は、白抜きのプロットで示されている。
TREM2は、活性化マクロファージ、未成熟樹状細胞、破骨細胞、及びミクログリア上に発現される2,3。高レベルのTREM2を発現する細胞は、免疫監視、細胞-細胞相互作用、組織破片のクリアランス、及び潜在炎症反応の消失に関与すると考えられる4。遺伝子ノックダウンまたはノックアウトによるこれらの細胞上のTREM2発現の不存在は、細胞の破片を貪食する能力を損なわせ、また、制御性サイトカインの産生を増加させる5。生理学的設定では、FACSプロットでみられるように、末梢血、脾臓、肝臓、または肺でのTREM2の発現は、非常に少ない乃至検出されない(図5)。しかし、肺または肝臓に存在するマクロファージが単離され、純粋な細胞集団としてTREM2について染色される場合、TREM2発現は検出可能になる。
材料及び方法
腫瘍組織を処理して、標準的な方法で単一細胞懸濁液を単離した。要約すると、腫瘍をかみそりの刃で細かく刻み、Miltenyi MACS解離キットの酵素を含有するRPMI-1640培地中で消化した。腫瘍を、製造業者の推奨によりGentleMACsで処理し、37℃で約40分間インキュベートした。消化混合物を、2mMのEDTA及び2%のウシ胎児血清を含有するPBSでクエンチした。次に、単一細胞懸濁液を70umのフィルターに通し、次に、細胞をFACS緩衝液ですすいだ。遠心分離後、細胞ペレットを、FACS緩衝液に再懸濁し、腫瘍関連マクロファージ及び他の免疫細胞集団を同定するために、抗体カクテルで染色した6。TREM2KO細胞染色は、灰色のプロットで示され、野生型細胞染色は、白抜きのプロットで示されている。
T細胞、B細胞、NK細胞、及び他の非骨髄性細胞集団、ならびにCD45陰性細胞は、細胞表面に検出可能なTREM2発現を発現しない。しかし、腫瘍関連マクロファージ(TAM)及び骨髄由来抑制細胞(MDSC)を含む骨髄細胞サブセットは、細胞表面に様々な程度でTREM2を発現する。腫瘍微小環境においてTREM2について陽性である細胞型のうち、TAM上の受容体発現の密度は、腫瘍の起源に関係なく、他の細胞型よりも有意に高かった(図6に示されるCT26及びMC38)。
材料及び方法
ボランティアの正常なヒトから得られた末梢血単核細胞(PBMC)及び負に選別されたCD14+単球がAllCells Inc.により提供された。標準プロトコールを使用して、CD14+単球をin-vitroで分化させた5。2mMのL-グルタミン、1ml当たり100μgのストレプトマイシン、1ml当たり100Uのペニシリン、及び10%の熱不活化FBSが補充されたRPMI-1640培地からなる完全培地で、CD14+単球を培養した。マクロファージへの分化を引き起こすために、50ng/mLのM-CSFを培地に加えた。培地を2~3日ごとに補充した。7日後、マクロファージをピペッティングにより採取し、後続のトリプシン処理により、接着細胞を収集した。次に、細胞を遠心分離し、抗生物質、2%のFBS及び組み換えヒトIFN-g及び100ng/mLのLPSが補充されたRPMI-1640に再懸濁した。細胞サブセットにおけるTREM2の細胞表面染色を評価するために、標準的な骨髄性カクテルを使用して、これらのマクロファージをPBMCと同時に表面染色した。対照mAbで染色された細胞は、灰色のプロットで示される。抗TREM2mAbで染色された細胞は、白抜きのプロットで示される。
図7にみられるように。ex-vivo分化型マクロファージは、評価されたPBMCベースの細胞型と比較して、TREM2の細胞表面受容体密度が著しく高いことを示す。文献で報告された観察と同様に、単球及び一部の好中球は、より低いレベルのTREM2を発現する。
材料及び方法
ヒト腫瘍組織を、Cooperative Human Tissue Network(CHTN)から入手した。Miltenyi MACS分離キット及びgentleMACSプロトコールを使用して、新しいヒト腫瘍組織を単一細胞懸濁液に解離させた。事前検証済みのマルチカラーFACSパネルを使用して、ヒト腫瘍組織の単一細胞懸濁液を表面染色した。全てのデータをLSR Fortessaフローサイトメーター(BD)またはAttuneフローサイトメーター(Thermo Fisher)で収集し、FlowJoソフトウェアを使用して分析した。数字は、各集団の染色指数を示し、これは、抗TREM2染色からアイソタイプ対照染色を差し引いたものとして定義される。
腫瘍微小環境内で、TREM2発現は、他の細胞と比較して、TAM上で高レベルに差次的に発現され(図8)、それをTAMの翻訳関連マーカーにする。粘液性腺癌の種々の細胞集団におけるTREM2抗体染色(白抜き)またはアイソタイプ対照染色(灰色)の代表的なヒストグラムを示す。一括して、このデータは、TREM2標的化薬が特定のTAMの枯渇を促進し、末梢細胞または他の組織常在免疫サブセットに対する付随的な影響が比較的少ない乃至全くないという仮説をサポートする。
材料及び方法
CT26.WT(CRL-2638)、Py8119(CRL-3278)、4T1(CRL-2539)、及びEMT6(CTL-2755)細胞を、the American Type Culture Collection(ATCC)から購入した。Panc-02細胞を、AJES Life Sciences(Stony Brook、NY)で使用した。in vivoで使用される抗体は全て、0.2EU/mg以下のタンパク質であった。クローンRMP1-14の抗マウスPD-1抗体のアミノ酸配列を、質量分析(LC-MS/MS)で決定した。FcgRへの結合を排除するために、単一の点変異D265Aを、RMP1-14抗体のFc領域に導入した。マウスのIgG1、クローンMOPC-21、及びマウスのIgG2a、クローンC1.18.4は、アイソタイプ対照を、BioXCellから購入した。PI-7012及びafuc-PI-7012(双方ともにマウスIgG2aとして)を、それぞれ、Expi293細胞(Thermo Fisher Scientific)または293/FUT8ノックアウト細胞で産生させ、次に、MabSelectプロテインAレジン(GE Life Sciences)を使用して精製した。mAbを0.1Mのクエン酸緩衝液(pH3.0)で溶出させ、緩衝液を使用前に交換した。

材料及び方法
実施例9に記載の抗TREM2 mAb及び抗PD-1 mAb処置後の、以前の研究からの腫瘍がないBALB/cマウスに、1×106のCT26腫瘍細胞で3ヶ月後に再チャレンジした。腫瘍体積を、移植後25日間測定した。同齢の処置ナイーブマウスに、同等数のCT26細胞を投与し、研究期間中に腫瘍の成長について追跡した。研究期間中、マウスに追加の処置を提供しなかった。
抗TREM2 mAb afuc-PI-7012及び抗PD-1 mAbの組み合わせでの処置後に、CT26腫瘍が治癒したマウスは、有効な抗腫瘍記憶応答を確立した(図10)。治癒したマウスは、追加の治療がなくても、新しい腫瘍の成長を拒絶することができ、これは、元の移植された腫瘍に対する長期的な免疫記憶を示す。この形態の長期免疫記憶は、活発なCD8+エフェクター記憶応答の維持を利用する。
材料及び方法
in vivoでの使用のための抗体を全て、エンドトキシンについて試験し、0.2EU/mg以下のタンパク質を使用した。抗PD-1(クローンRMP1-14)をAbsolute Antibody Incから購入するか、または、PionyrにおいてマウスIgG1 D265Aフォーマットとして組み換え産生した。マウスIgG1(クローンMOPC-21)及びマウスIgG2a(クローンC1.18.4)アイソタイプ対照を、BioXCellから購入するか、または、PionyrにおいてHEK293細胞で組み換え産生した。抗TREM2抗体のキメラマウスIgG2aバージョン(PI-7012)を、PionyrにおいてHEK293細胞で産生し、SEC及びCE-SDSならびにエンドトキシン試験で単分散及び純度について評価した。
図11に示されるように、抗TREM2抗体のみの処置は、腫瘍量の大幅な低減をもたらし、TREM2抗体処置が単独療法の有効性を有することを示す。抗PD1抗体処置は、同等の腫瘍量の低減をもたらし、抗TREM2抗体及び抗PD1抗体の組み合わせも同様であった。
材料及び方法
組織の取得
異なる腫瘍の適応症及び疾患グレードにわたる腫瘍マイクロアレイ(TMA)を、Reveal Biosciences(カルフォルニア州サンディエゴ)から購入し、重複した48患者の症例または2mmのコアとしての96を超える単独症例が含まれる。10%の中性緩衝ホルマリンで24時間固定し且つ同一のSOPを使用して処理した組織を取得することによって、Reveal BiosciencesでのTMAを作製した。切片をSuperfrost Plus or Startfrost Adhesiveスライド上に採取し、全てのTMAを発注時に4μmの切片に新たに切断し、4℃で保存した。
ホルマリン固定パラフィン包埋(FFPE)組織でTREM2陽性細胞を検出するIHCアッセイを、TMA染色全てについて標準化した。最初に、スライドを60℃のオーブンで45分間焼き、続いて、キシレンで5分間ずつ3回脱パラフィン処理した。次に、スライドを100%~70%のエタノールの、一連のエタノール勾配で再水和し、最後に蒸留水で洗浄した。デクローキング圧力チャンバー(Biocare)を、pH6.0のクエン酸ナトリウム緩衝液(Sigma、C9999)で、110℃で15分間、熱誘導性抗原回復ステップに使用した。内因性ペルオキシダーゼ活性をブロックするために、ブロッカー溶液(Vector Labs)をスライドに15分間適用し、続いて、Tweenを含有するPBSバッファー(Alfa Aesar-J63596)ですすいだ。ヤギ血清を含有するブロッキング溶液(Vector Labs)とともに、組織切片を4℃で一晩インキュベートすることによって、非特異的結合をブロックした。組み換え抗TREM2抗体クローンEPR20243(Abcam、ab209814)としても知られるPIT2Dを、1次抗体として、PBS中5μg/mlの濃度、室温で60分間使用した。次に、スライドをそれぞれ、PBS-Tweenで5分間2回洗浄した後、1:500希釈のHRPポリマーコンジュゲート抗ウサギ2次抗体(Vector LabsのMP-7500検出キット)で20分間インキュベートした。スライドをそれぞれ、PBS-Tweenで5分間2回洗浄した後、検出ステップに進んだ。DAB基質を製造者の使用説明書(Abcam、ab64238)に従って調製し、スライドに3分間適用し、続いて、蒸留水で十分にすすいだ。ヘマトキシリンを使用して、スライドを30秒間対比染色し、続いて、流水ですすぎ、70%~100%のエタノールの一連のエタノール勾配で脱水し、キシレンで乾燥させた。染色したスライドを、最終的に、培地に載せ、一晩乾燥するためにカバーガラスで蓋をした。
明視野設定を使用したVectra顕微鏡(Perkin Elmer-Akoya Biosciences)上で、TMAスライドをイメージングした。スライド全体を10×でスキャンした後、以下のIHCスコアリングシステムを使用して、TREM2+細胞の定量を評価した。0:コア内の間質領域において染色がない、1:コア内の間質領域に約25%の陽性細胞、2:コア内の間質領域に50%陽性細胞、3:コア内の間質領域に75%の陽性細胞。これらの群間にある陽性細胞のパーセンテージを、0.5(約12%)、1.5(約37.5%)、2.5(約62.5%)、及び3.5(90%)としてスコアリングした。TREM2染色の強度を、低(スコア1)、中(スコア2)、及び強(スコア3)と決定した。面積の半分以上が変形した、失われたまたは折りたたまれたコアを、分析から除去し、スコアリングしなかった。各コアを同時に2人のオペレータがチェックし、各患者の症例のスコア(利用可能な場合、重複コアを平均した)をプリズムファイルにプロットした。面積の半分以上が変形した、失われたまたは折りたたまれたコアを、分析から除去し、スコアリングしなかった。

雌のBALB/cマウスに、1×106のEMT6細胞を注射して、実施例11に記載の皮下腫瘍を確立した。平均腫瘍体積が200立方mm超に達すると、マウスに、30mg/kgのアイソタイプ抗体またはafuc-PI-7012を5日間隔で2回投与した。腫瘍体積を上述のように計算した。2回目の投与の2日後に腫瘍を採取した。脂肪及び繊維状物質を切除によって腫瘍から除去した。次に、腫瘍を計量し、機械的解離及び酵素的解離の組み合わせによって単一細胞懸濁液用に処理した。組織を細かく切り刻んだ後、gentleMACS Octo Dissociator(Miltenyi Biotec、130-095-937)内のgentleMACS Cチューブ(Miltenyi Biotec、130-093-235)を用いて、最適化され酵素カクテル(Miltenyi Biotec、Tumor Dissociation Kit、mouse 130-096-830)を使用して、腫瘍を酵素的に消化させた。解離後、試料をフィルターに適用して、あらゆる残留する大きな粒子を除去した。フローサイトメトリーのために、抗体のパネルを使用して、腫瘍組織の単一細胞懸濁液を表面染色した。骨髄サブセットを評価するための抗体パネルは、CD45、XCR1、F4/80、CD64、CD11c、Ly6C、CD11b、Ly6G、CD24、MHCクラスII、及びダンプ・チャネル(CD45R、CD90.2、CD3e、NKp46、CD19、Siglec F)内の系統マーカーに特異的な抗体を含んだ。リンパ系サブセットを評価するための抗体パネルは、CD45、CD4、CD25、B220、NKp46、CD44、CD90.2、CD8a、CD11b、及びCD49bに特異的な抗体を含んだ。全てのデータを、Attuneフローサイトメーター(Thermo Fisher)で収集し、FlowJoソフトウェアを使用して分析した。
図13Aに示されるように、抗TREM2抗体処置は、in vivoで腫瘍サイズの減少をもたらした。加えて、抗TREM2抗体療法は、アイソタイプ抗体単独と比較して、MHCIIlowTAMの数を減少させ、MHCIIhighTAMの数を増加させた(図13B)。抗TREM2抗体療法は、また、単独のアイソタイプ抗体と比較して、CD8+T細胞及びNKp46+NK細胞の数を増加させた(図13C)。

firehose_getを使用して、胃癌試料のThe Cancer Genome Atlasのコホートのレベル2、RNAseqデータを、Broad Instituteからダウンロードした。腫瘍試料及び隣接する正常試料の両方からTREM2のRSEM値を、100万当たり、log2数に変換し、Rでプロットした。
材料及び方法
285人の卵巣患者に対する正規化されたTREM2発現プロファイル及び関連臨床データを、NCBIのGEO Webサイト(アクセッションGSE9899)からダウンロードした。発現プロファイルを、TREM2発現の中央値、三分位数、及び四分位数のレベルに基づいて3つのコホートに分けた。各コホートのKaplan-Meier生存曲線をプロットし、Rの生存パッケージ及びsurvminerパッケージを使用して関連するログランク検定を実行した。三分位数コホートは、下位33%のTREM2発現と比較した上位33%のTREM2発現を有するコホートであり、四分位数コホートは、下位25%のTREM2発現と比較した上位25%のTREM2発現である。

単一のヒト卵巣患者の解離腫瘍細胞(DTC)を、Discovery Life Sciencesから購入した。細胞は、免疫細胞を濃縮するためにCD45陽性についてフローサイトメトリーで選別し、10Xゲノミクスクロムコントローラを使用して、単一細胞RNA配列決定のためにカプセル化した。10XのCellRangerプログラム及びRプログラミング言語のSeuratモジュールを使用して、得られた生データを順次処理した。細胞型特異的マーカー遺伝子を使用して、得られたt分布型確率的近傍埋め込み法(tSNE)の次元削減の細胞集団に手動で注釈を付け、TREM2発現をプロットした。TREM2+細胞を、TREM2発現を有する任意の細胞として定義した。
