The present invention relates to immunological detection methods and reagents for SARS-CoV-2.
SARS coronavirus-2 (SARS-CoV-2) is the causative virus of COVID-19, a novel coronavirus infection, classified in the genus Betacoronavirus of the family Coronaviridae. SARS-CoV-2 is a virus that is closely related but different from the SARS coronavirus (SARS-CoV) that causes severe acute respiratory syndrome (SARS) in China in the 2000s.
A coronavirus is an enveloped virus having a positive single-stranded RNA as the viral genome. The nucleocapsid protein (N protein) in virus particles forms a dimer with a C-terminal domain (CTD), which in turn forms a tetramer. Furthermore, it is believed that genomic RNA binds to the N-terminal domain (NTD) of a nucleocapsid protein (N protein) to form a nucleocapsid. A nucleocapsid has a structure covered by an envelope consisting of a lipid bilayer membrane. The coat is bound by the S, E and M proteins through their transmembrane sites. The N protein binds to the M protein, which constitutes the coat, to form an enveloped virus particle.
Although there are no reports on the molecular properties of the N protein in specimens, it is generally believed that (i) it is contained in the nucleocapsid of virus particles and (ii) cells infected with the virus are involved in the proliferation of the virus. Alternatively, it is thought that there are substances that are released by being crushed by the immune response of the infected host.
A method using an antibody against the SARS-CoV N protein (full length: 422 amino acids) has been reported as an immunoassay method for SARS coronavirus.
An object of the present invention is to provide a technology that can immunologically detect SARS-CoV-2.
As a result of intensive studies, the present inventors found that a combination of two specific antibodies against two epitopes in the C-terminal region of the SARS-CoV-2 N protein, that is, (1) SARS-CoV-2 N protein and (2) a second antibody against a second epitope in the 365-419 amino acid region in the SARS-CoV-2 N protein. It was found that SARS-CoV-2 can be detected with high sensitivity by doing so. The present inventors also found useful antibodies that are easy to use in combination with other antibodies and that can achieve more sensitive detection of SARS-CoV-2 when used in combination with other antibodies. found to be a combination.
Although
Based on the above findings, the present inventors have successfully developed an immunological detection method and reagent for SARS-CoV-2, and have completed the present invention.

In the sandwich method, the N protein is detected, for example, by treating the sample with a solid-phase antibody to allow the N protein in the sample to bind to the solid-phase antibody (e.g., contacting the sample with the solid-phase antibody); This may be done by detecting the N protein bound to the phase antibody with a labeled antibody. Detection of N protein may further comprise a step of removing N protein that is not bound to the solid phase antibody (e.g., (B/F separation or washing step). Also, in the sandwich method, detection of N protein may include: For example, a step of treating the specimen with a labeled antibody to bind the N protein in the specimen to the labeled antibody (e.g., a step of contacting the specimen with the labeled antibody), further treating the N protein with a solid-phase antibody, and immobilizing the N protein. It may be carried out by the step of binding to the phase antibody, and the step of detecting the N protein bound to the solid phase antibody and the labeled antibody with the labeled antibody. In addition, the solid phase antibody may be an antibody that has been immobilized on a solid phase in advance. Alternatively, an antibody immobilized on a solid phase may be used in the detection of the N protein. For example, a step of treating the sample with an antibody that can be immobilized on a solid phase to allow the N protein in the sample to bind to the antibody (e.g., contacting the specimen with the solid phase antibody), binding the N protein bound to the antibody that can be immobilized on the solid phase to the labeled antibody, immobilizing the antibody that can be immobilized on the solid phase to the solid phase, and It can also be carried out by the step of detecting the N protein bound to the immobilized antibody (solid phase antibody) and the labeled antibody with the labeled antibody. As the binding method of the antibody immobilized on the solid phase and the solid phase, the above-mentioned affinity can be used.
In another particular embodiment, the methods of the invention may be performed by immunochromatographic methods. Immunochromatographic methods are preferred as rapid and convenient qualitative methods. Immunochromatographic methods and equipment used therefor are well known. As such a device, for example, an immunochromatography cartridge (which may further include a developing solution reservoir) comprising a zone containing a solid-phase antibody and a zone containing a labeled antibody can be used. As a preferred example of immunochromatography, an outline of a lateral flow immunochromatography method and apparatus will be described below with reference to FIG.
In FIG. 1, an
At the time of use, the sample is added to the

Example 1: Antibody production An antibody was produced according to the method described in WO2005/042579. Specifically, it is as follows.
(1) Production and Purification of Recombinant N Protein A SARS coronavirus (SARS-CoV) nucleocapsid protein (N protein) gene was inserted into an expression plasmid to prepare a plasmid pWS-N. Escherichia coli was transformed with this to obtain ampicillin-resistant transformant Escherichia coli. The nucleotide sequence and amino acid sequence of N protein are shown in SEQ ID NOS: 1 and 2, respectively.
The resulting transformant was cultured at 37° C. in 2 ml of LB medium containing 50 μg/ml ampicillin. This transformant was precultured to grow to a density of about 0.7 in OD at 600 nm, and then 0.4 mM IPTG was added to induce expression. After culturing for 18 hours, centrifugation was performed to recover E. coli. A 20 mM Tris-HCl buffer solution (pH 8.0) containing 0.1 mM PMSF was added to the collected E. coli, and sonication was performed under ice-cooling. After centrifugation, ammonium sulfate was added to the soluble fraction to collect 20-40% ammonium sulfate fraction. This ammonium sulfate fraction was applied to SP Sepharose Fast Flow (manufactured by Amersham) equilibrated with 0.1 M NaCl, 8 M urea, 20 mM phosphate buffer (pH 6.9), and 0.2 M NaCl, 8 M urea, 20 mM Purification was performed by elution with a phosphate buffer (pH 6.9). The eluted fraction was dialyzed against 0.2 M NaCl, 20 mM Tris-HCl buffer (pH 8.0) to obtain a purified recombinant N protein (CoV) solution. SDS-PAGE and Western blotting confirmed the purity of the recombinant N protein, which showed a single band.
(2) Production of Monoclonal Antibody Anti-N protein monoclonal antibody was produced by immunizing a mouse with the recombinant N protein (CoV) produced above and fusing the mouse's splenic lymphocytes and myeloma cells. That is, BALB/C mice were first immunized with 50 to 100 μg/mouse of recombinant N protein emulsified with Freund's complete adjuvant, and after 2 to 3 weeks, 50 to 100 μg/mouse of the same antigen emulsified with Freund's incomplete adjuvant. Mice were boosted. The antibody titer was checked by solid-phase ELISA using a 96-well ELISA plate coated with recombinant N protein. 25 to 100 μg of free recombinant N protein was intravenously administered to mice in which an increase in antibody titer was observed, and 3 to 4 days later, spleens were removed from the mice to prepare splenocytes. Mouse myeloma cells (P3U1) that had been previously cultured in RPMI-1640 medium and splenocytes were mixed at a ratio of 1:2 to 1:5, and cell fusion was performed using PEG. After the fused cells floated in HAT medium, they were dispensed into 96-well culture plates and cultured in a CO 2 incubator at 37°C.
Antibody screening was performed by solid-phase ELISA as described above. That is, the recombinant N protein was dispensed into 96-well ELISA plates at a concentration of 1 μg/mL at a concentration of 50 μL/well and allowed to stand overnight at 4° C. for adsorption. After blocking the wells with 1% skimmed milk, the wells were washed three times with a washing buffer (PBS containing 0.05% Tween), 50 μL of the culture supernatant of the cell-fused plate was added, and the mixture was reacted at 37° C. for 1 hour. let me After washing three times with washing buffer in the same manner, a POD-labeled anti-mouse immunoglobulin antibody (manufactured by DACO) was added and allowed to react at 37° C. for 1 hour. After washing four times with washing buffer, the substrate ABTS was added and wells showing color development were selected. Next, the cells in the selected wells were transferred to a 24-well culture plate, cultured in a CO 2 incubator at 37° C., and single cloned by the limiting dilution method to obtain each monoclonal antibody.
Also, instead of recombinant N protein (CoV), mice were immunized with a KLH conjugate of a synthetic peptide (N5 peptide: GQTVTKKSAAEASKKPRC: SEQ ID NO: 3) as an immunogen to similarly obtain an anti-N protein monoclonal antibody.
(3) Confirmation of monoclonal antibody reactivity The reactivity of each established monoclonal antibody to the native antigen (SARS-CoV-derived N protein) was confirmed by Western blot (WB) using a concentrated virus suspension as a sample. . VeroE6 cells were infected with SARS virus Hanoi strain, cultured in a CO 2 incubator for 48 hours, and then centrifuged at 2,000 rpm for 15 minutes to prepare a virus culture supernatant (TCID50 was 7.95×10 6 /mL). . After the culture supernatant was inactivated at 56° C. for 90 minutes, 31.5 mL of the culture supernatant was centrifuged at 30,000 rpm for 3 hours using a Hitachi ultracentrifuge (40T rotor). 0.3 mL of Tris-NaCl-EDTA buffer was added to the resulting precipitate and pipetting was performed to prepare a concentrated virus suspension. An equal amount of sample treatment solution for electrophoresis was added to this suspension, followed by heat treatment to prepare a sample for analysis. After performing SDS-PAGE using a 12.5% gel, the sample was transferred to a nitrocellulose membrane to prepare a WB transfer membrane (antigen-transferred WB membrane). After blocking the transfer membrane with skimmed milk, each monoclonal antibody was shaken with the antigen-transferred WB membrane at room temperature for 1 hour, reacted, and then washed three times with washing buffer (washing with shaking for 5 minutes). Next, a POD-labeled anti-mouse immunoglobulin antibody was added and reacted at room temperature for 1 hour. After washing 4 times with washing buffer (washing with shaking for 5 minutes), a 4-chloronaphthol solution was added as a substrate to confirm the band. selected.
The selected monoclonal antibody is further subjected to WB using the SARS-CoV-2 recombinant N protein prepared in the same manner as in (1) above, and the common coronavirus N protein, and the SARS-CoV-2 recombinant N protein. Those that bound but did not bind to the common coronavirus N protein were designated as anti-N protein monoclonal antibodies. In the following examples, the N protein of SARS-CoV-2 was used as the N protein unless otherwise specified.
Example 2: Identification of antibody reactive region (part 1)
The epitopes of the multiple anti-N protein monoclonal antibodies obtained in Example 1 above were identified by the WB method using a recombinant antigen containing a partial sequence of the N protein.
First, the SARS-CoV-2 N protein full length (419 amino acids), NTD (44th to 180th amino acids), CTD (247th to 364th amino acids) and N protein prepared in the same manner as in Example 1 An equal amount of the sample treatment solution for electrophoresis is added to a solution containing a recombinant antigen of the C-terminal peptide (330th to 419th amino acids), and then subjected to overheating to obtain a sample for analysis, as described in Example 1 (3) above. SDS-PAGE and WB were performed in the same manner as in , and the reactivity of each N protein monoclonal antibody was confirmed.
As a result, 6 antibodies (anti-N protein antibodies N1 to N6) reacted with the full-length antigen and NTD antigen, and 6 antibodies (anti-N protein antibodies C1-C6) reacted with the full-length antigen and CTD antigen. This was confirmed, and multiple antibodies reacting with NTD and CTD were obtained. In addition, antibodies (anti-N protein antibodies C12 and C13) that recognize the region contained in the 365th to 419th amino acid sequences, which react with the full-length antigen and the C-terminal peptide but do not react with the NTD or CTD antigen, were also obtained. rice field. Furthermore, as an immunogen, an anti-N protein monoclonal antibody that recognizes the 243rd to 259th amino acid regions of the N protein obtained by immunizing a mouse with a KLH conjugate of a synthetic peptide (N5 peptide: GQTVTKKSAAEASKKPRC: SEQ ID NO: 3) is also available. Multiple (P1, P2) were obtained.

(manufactured by Fujirebio) to obtain solid-phase antibodies (antibody-immobilized magnetic particles).
Example 4: Preparation of labeled antibody (alkaline phosphatase-labeled anti-N protein monoclonal antibody) The anti-N protein monoclonal antibody obtained above and pepsin were mixed in 0.1 M citrate buffer (pH 3.5) and heated at 37°C. The mixture was allowed to stand at 1 hour for pepsin digestion. After stopping the reaction, purification by gel filtration was performed to obtain an antibody from which the Fc region was removed. Thiolation was then carried out by adding 2-mercaptoethylamine hydrochloride (2-MEA). Further, this was desalted to obtain a Fab' fragment.
N-(4-maleimidobutyryloxy)-succinimide (GMBS)-treated alkaline phosphatase and Fab' fragments prepared from each anti-N protein monoclonal antibody were mixed at a molar ratio of 1 to 3:1, performed the coupling. 2-Mercaptoethylamine hydrochloride and iodoacetamide were added to the coupling solution to terminate the reaction. Further, peaks with a molecular weight of Fab' and ALP of 1 to 3:1 were pooled by gel filtration to obtain a labeled antibody (ALP-labeled anti-N protein monoclonal antibody).

Example 6: Immunochromatography Method As shown in FIG. 1, an aqueous solution containing an anti-N protein monoclonal antibody is positioned 15 mm from the end of the developing solution absorption zone side of a matrix consisting of a nitrocellulose membrane having a width of 3.7 mm and a length of 50 mm. 0.5 μL was spotted on a nitrocellulose membrane and dried to create a detection zone. Further, an anti-alkaline phosphatase (ALP) antibody solution (sodium carbonate buffer containing 0.15 M NaCl) was spotted at a position 12 mm from the end of the developing solution absorption zone and dried to form a development confirming portion. In addition, 0.9 μL of a 5-bromo-4-chloro-3-indolyl-phosphate disodium salt (BCIP) solution was spotted in a line as a substrate to form a substrate zone. Next, 3 μL of a solution containing an ALP-labeled anti-N protein monoclonal antibody (labeled antibody) was spotted on the matrix and dried to form a labeling reagent zone consisting of an enzyme-labeling reagent pad.
The matrix, developing solution pad, enzyme-labeled reagent pad and absorbent pad (highly water-holding filter paper) are stacked as shown in FIG. 1 and fixed with adhesive tape. An immunochromatographic cartridge for protein detection was manufactured.
A solution containing the recombinant N protein or 20 μL of the sample and 100 μL of the treatment liquid were mixed and allowed to stand at room temperature for about 5 minutes. The N protein in the resulting mixture was detected using the manufactured immunochromatography cartridge. Specifically, 20 μL of the mixed liquid was dropped onto the labeled antibody zone of the immunochromatography cartridge, and then the developing liquid tank was broken, and the developing liquid was supplied to the developing liquid pad and the upper end of the matrix to start the measurement. Thirty minutes after the start of the measurement, the development of the developing solution was confirmed by the color development of the development confirmation part 10, and then the color development of the detection zone was visually measured.
Example 7: Identification of antibody reactive region (Part 2)
Recombinant N proteins (SARS-CoV-2 or SARS-CoV) were measured according to the sandwich immunoassay method of Example 5 above, and grouped according to the reactive region of each N protein monoclonal antibody. Specifically, antibody-immobilized magnetic particles (solid-phase antibodies) and ALP-labeled antibodies were N1 to N6 antibodies, C1 to C6 antibodies, and C12 antibodies, respectively, and the recombinant N protein was measured.
As a result, when the recombinant N protein (0, 10 ng/mL) was measured using the N1 antibody and the N2 antibody as the solid-phase antibody and the labeled antibody, respectively, 10 ng/ It was confirmed that the luminescence ratio of mL was very low and that both reacted in overlapping regions (Table 1). Similarly, when measuring recombinant N protein (0, 1 ng/mL) using N4, N5, and N6 antibodies (N6 antibody group) as solid-phase antibodies and labeled antibodies, respectively, the amount of light emitted from 1 ng/mL to 0 ng/mL The luminescence difference after subtracting the luminescence in mL was very low, confirming that these antibodies recognize overlapping regions (Table 2). On the other hand, when the N1 antibody and the N3 antibody were used as the solid-phase antibody and the labeled antibody, the difference in the amount of light emission was smaller than in the combination with other antibodies, confirming partial competition. Compared to the N3 antibody, the N1 antibody has higher reactivity in sandwich immunoassays with other NTD antibodies, so in sandwich immunoassays using multiple antibodies, the N1 antibody group (N1, N2 antibodies) was considered more useful than the N3 antibody.
Similarly, when C1 to C6 antibodies were also examined, it was found that the C1 antibody and the C2 antibody recognize the overlapping region, the C3 antibody and the C4 antibody recognize the overlapping region, and the C5 antibody and the C6 antibody overlap the region. Recognition was confirmed (Tables 3-5). However, both C5 and C6 antibodies showed low luminescence in any combination, confirming that antibodies against the region recognized by the C5 antibody group (C5 and C6 antibodies) have low reactivity in measurements by sandwich immunoassay. was done.
Example 8: Examination of combination of antibodies (Part 1)
For the antibodies grouped in Example 7 above and the C12 antibody, combinations of solid-phase antibodies and labeled antibodies were examined. Using each combination of antibodies shown in Table 6, the recombinant N protein was measured in the same manner as in the sandwich immunoassay method of Example 5 above. Each was measured with N=2.
Table 6 shows the results. Table 6 shows the ratio (S/N ratio) of the average luminescence amount from 1 ng/mL specimens to the average luminescence amount from 0 ng/mL specimens.
As a result, as a combination of a solid-phase antibody and a labeled antibody for sandwich immunoassay, both N1 antibody and C3 antibody, N1 antibody and C12 antibody, N4 antibody and C3 antibody, N4 antibody and C12 exhibiting high S/N ratios. It was confirmed that the antibody, the combination of the C3 antibody and the C12 antibody, is preferable.
Example 9: Examination of combination of antibodies (Part 2)
For the antibodies grouped in Example 7 above, the P1 antibody, and the C12 antibody, combinations of solid-phase antibodies and labeled antibodies were examined. Samples containing 10 ng/mL of recombinant N protein were assayed using the antibody combinations shown in Tables 7 and 8 by the immunochromatographic method of Example 6 above. Further, using each combination of antibodies shown in Tables 7 and 8, 30 minutes after the start of measurement (Table 7) or 60 minutes (Table 8), after confirming the development of the developing solution by the color development of the development confirmation part 10 , the color development of the detection zone was measured visually. The results are shown in Tables 7 and 8. Tables 7 and 8 show the color intensity of the detection zone under each condition. Higher numbers indicate darker coloration and "w" indicates weaker. That is, the intensity of color development decreases in the order of 4.5>4>3>2>1>w. Empty columns indicate non-implementation.
As a result, similar to the sandwich immunoassay using particles, the combination of N1/N2 antibodies and C3 antibodies, N1 antibody and C12 antibody, N4 antibody and C3 antibody, N4 antibody and C12 antibody, and C3 antibody and C12 antibody showed strong luminescence. Strength confirmed. Furthermore, the N1 antibody/N2 antibody and N4 antibody also showed relatively high luminescence intensity.
Furthermore, when the combination of the C1 antibody and the P1 antibody with other antibodies was also evaluated, the C1 antibody showed relatively strong luminescence intensity in combination with the C3 antibody. On the other hand, the P1 antibody showed strong luminescence intensity in combination with the C3 antibody, and showed relatively strong luminescence intensity in combination with the N1 and N4 antibodies.
Example 10: Examination of combination of antibodies (Part 3)
Examples 8 and 9 above discussed the combination of a single solid phase antibody and a single labeled antibody. Although a measurement system with sufficiently high sensitivity could be constructed even with such a combination, it is more preferable to use a combination of solid-phase antibodies recognizing different regions and/or labeled antibodies recognizing different regions. Therefore, an immunoassay system was constructed using a labeled antibody as one of the antibodies for which an effect was observed alone in Examples 8 and 9 above, and a plurality of other antibodies as solid-phase antibodies, in the same manner as in Example 8 above. A sample containing 10 ng/mL of recombinant N protein (SARS-CoV) was evaluated by the method of . Solid-phase antibodies were prepared by simultaneously chemically bonding multiple antibodies and particles according to the production method of Example 3 above. Table 9 shows the results.
As a result, when the mixture of C3 antibody and C12 antibody was used, excellent improvement in luminescence (count at 1000 pg/ml) and S/N ratio was observed. In particular, when the N1 antibody or N4 antibody is used as the labeled antibody, the amount of luminescence when a mixture of the C3 antibody and the C12 antibody is used as the solid phase antibody is higher than when the C3 antibody and the C12 antibody are used alone as the solid phase antibody. increased, and the S/N ratio also increased. On the other hand, when C3 antibody or C12 antibody was used as the labeled antibody, even if a mixture of N1 antibody and N4 antibody was used as the solid-phase antibody, neither the count value nor the S/N ratio increased. From this, it was confirmed that the combined use of the C3 antibody and the C12 antibody is effective in improving the sensitivity of the sandwich immunoassay, rather than simply using a plurality of antibodies for the solid-phase antibody.
Example 11: Identification of antibody reactive region (Part 3)
For the antibodies confirmed to be effective in Examples 8 to 10 above, the epitope of N protein (SARS-CoV-2) was analyzed.
For the N1 antibody group (N1, N2 antibodies) and N6 antibody group (N4 to N6 antibodies) that recognize NTD, the NTD N-terminal region (44th to 112th amino acids) prepared in the same manner as in Example 1 above, Central region of NTD (79th to 147th amino acids), recombinant antigen consisting of NTD C-terminal region (113th to 180th amino acids), and 1st to 119th, 110th to 229th, 220th to 339th of N protein, The reaction region was analyzed by WB using recombinant antigens (trNP1, trNP2, trNP3, trNP4 antigens) consisting of the 330th to 419th amino acid regions. As a result, the N1 antibody group was shown to recognize the 44th to 78th amino acid regions of the N protein. Also, the N6 antibody group was shown to recognize the 120th to 147th amino acid region of the N protein.
Similarly, for the C3 antibody group and C1 antibody group that recognize CTD, the CTD N-terminal region (247th to 305th amino acids) prepared in the same manner as in Example 1 above, the CTD central region (276th to 334th amino acids ), WB and sandwich assays using recombinant antigens consisting of the C-terminal region of CTD (amino acids 306-364) and trNP1, trNP2, trNP4 and trNP4 antigens, and reactivity with synthetic peptides and other antibodies, The reaction area was analyzed. As a result, the C3 antibody group was shown to recognize the 260th to 305th amino acid region of the N protein. Also, the C1 antibody group was shown to recognize the 306th to 339th amino acid region of the N protein.
Example 12: Examination of combination of antibodies (Part 4)
In Example 10 above, it was shown that it is useful to use a mixture of C3 antibody and C12 antibody as the solid-phase antibody when using the N1 antibody or the N4 antibody. In order to confirm that these antibody combinations are similarly useful for SARS-CoV-2, using the combinations of solid-phase antibodies and labeled antibodies listed in Table 10, recombinants were used in the same manner as in Example 5 above. A solution containing 100 pg/mL of N protein was measured.
Specifically, 100 μL of a solution containing a recombinant N protein and 20 μL of a treatment solution (Tris buffer, 150 mM NaCl, 0.1% NaN 3 , 2.5% C16APS) were diluted with a particle suspension containing a solid-phase antibody. It was added to 50 μL of the liquid and reacted at 37° C. for 8 minutes. After the reaction, B/F separation was performed with a magnet, and after washing with a lumipulse washing solution, 50 μL of the labeled body fluid containing the ALP-labeled antibody was added and allowed to react at 37° C. for 8 minutes. After B/F separation with a magnet and washing with a Lumipulse washing solution, 200 μL of a Lumipulse substrate solution containing AMPPD was added, an enzymatic reaction was performed at 37° C. for 4 minutes, and the amount of luminescence at a wavelength of 463 nm was measured to determine the amount of light emitted from each sample. N protein was measured. Solid-phase antibodies were prepared by chemically binding each antibody and particles according to Example 3 above, and then mixing them at a ratio of 1:1. Labeled antibodies are GMBS-treated ALP and Fab' fragments prepared from a plurality of anti-N protein monoclonal antibodies at the same time so that the molar ratio of each Fab' fragment to ALP is 1: 1 according to Example 4 above. Mixed and coupled to prepare ALP-labeled antibody.
Table 10 shows the results. As a result, when the mixture of C3 antibody and C12 antibody was used, excellent improvement in luminescence (count at 100 pg/ml) and S/N ratio was observed. It was confirmed that the combined use of the C3 antibody and the C12 antibody is also effective in improving the sensitivity of the SARS-CoV-2 sandwich immunoassay.
Example 13: Examination of combination of antibodies (Part 5)
In Examples 10 and 12 above, it was shown that it is useful to use a mixture of C3 antibody and C12 antibody as the solid-phase antibody when N1 antibody or N4 antibody is used. As described above, SARS-CoV-2 may mutate, so it is more preferable to use a combination of antibodies that recognize different regions. Therefore, it is an antibody of a group different in reactivity from the group of N1/N2 antibody, N4/N6 antibody, C3/C4 antibody, and C12/C13 antibody, and reacts with N1 antibody, N4 antibody, and C3 antibody in Example 10 above. A combination with the P1/P2 antibody, which is highly potent, was evaluated. Combination with C1/C2 antibody was also evaluated.
Specifically, combinations of solid-phase antibodies and labeled antibodies shown in Tables 11-1 to 11-3 were used and evaluated in the same manner as in Example 8 above. Solid-phase antibodies were prepared by simultaneously chemically coupling multiple antibodies and particles according to Example 3 above. Labeled antibodies are Fabs prepared from GMBS-treated ALP and multiple anti-N protein monoclonal antibodies according to Example 4 above, such that the molar ratio of each Fab' fragment to alkaline phosphatase (ALP) is 1:1. The 'fragments were mixed together and coupling was performed to prepare the ALP-labeled antibody.
As a specimen, SARS-CoV-2 negative saliva specimen, recombinant N protein or SARS-CoV-2 positive patient's
The results are shown in Tables 11-1 to 11-3. In Tables 11-1 to 11-3, the light emission amount ratio (S/N ratio) when 10 ng/mL of recombinant N protein is measured to the light emission amount when 0 ng/mL of recombinant N protein is measured, and 0 ng of recombinant N protein. The ratio of the amount of luminescence when measuring each SARS-CoV-2 positive sample to the amount of luminescence when measuring /mL is shown.
As a result, SARS-CoV-2 in specimens could be measured with very high sensitivity by using C3 antibody and C12 antibody as solid-phase antibodies or labeled antibodies and using N6 antibody on the other hand. That is, an antibody that recognizes the 260th to 305th amino acid region and an antibody that recognizes the 365th to 419th amino acid region is used as a solid phase antibody or labeled antibody, and the other is an antibody that recognizes the 120th to 147th amino acid region. can be used to measure SARS-CoV-2 in specimens with very high sensitivity.
Furthermore, SARS-CoV-2 in specimens could be measured with higher sensitivity by combining the N1 antibody and the P1 antibody. That is, in addition to the combination of the above antibodies, further combining an antibody that recognizes the 44-78th amino acid region and an antibody that recognizes the 243-259th amino acid region, SARS in the specimen with higher sensitivity - Confirmed that CoV-2 can be measured.
In addition, SARS-CoV-2 in specimens could be measured with sufficiently high sensitivity by adding C1 antibody to the combination of these antibodies. In other words, it was confirmed that combining an antibody that recognizes a different CTD region from these antibodies is also effective.
Example 14: Examination of surfactants used in the reaction solution A SARS-CoV-2 negative saliva sample was added with a nasal swab of a SARS-CoV-2 positive patient (Boca Biolistics, LLC) as a sample. did. 100 μL of this sample and a treatment solution containing surfactants shown in Table 12 (Tris buffer, 150 mM NaCl, 1% BSA, 0.1% NaN 3 , concentration of each surfactant in treatment solution 2.5%, sample and the concentration of 0.19% after mixing with the particle suspension) was added to 150 μL of the particle suspension diluent containing the solid-phase antibody, and reacted at 37° C. for 8 minutes. After the reaction, B/F separation was performed with a magnet, and after washing with Lumipulse washing solution (manufactured by Fujirebio), 150 μL of the labeled body fluid containing the ALP (alkaline phosphatase)-labeled antibody prepared in Example 4 above was added, and the mixture was heated at 37°C. React for 8 minutes. After B/F separation with a magnet and washing with Lumipulse washing solution, AMPPD (3-(2′-spiroadamantane)-4-methoxy-4-(3″-phosphoryloxy) phenyl-1,2-dioxetane・2 200 μL of Lumipulse substrate solution containing sodium salt) was added, enzyme reaction was performed at 37° C. for 4 minutes, and the amount of light emitted at a wavelength of 463 nm was measured to measure the N protein of each sample. N6 antibody and P1 antibody were used as solid-phase antibodies, and N1 antibody, C3 antibody and C12 antibody were used as labeled antibodies.
Table 12 shows the results. In addition to the measured luminescence values, Table 12 also shows the ratio (%) of the luminescence value under each surfactant addition condition to the luminescence value under the condition where no surfactant was added.
As a result, when using amphoteric surfactants (C12APS and C14APS) containing a hydrocarbon chain as the hydrophobic part, a very high luminescence value was obtained in the positive sample, and the N protein could be detected with high sensitivity. . From the above, it was shown that a zwitterionic surfactant containing a hydrocarbon chain as a hydrophobic part is effective for measuring SARS-CoV-2 N protein contained in patient specimens.
Example 15: Examination of surfactants used in sample treatment liquid (Part 1)
In Example 14 above, it was found to be useful in measuring the SARS-CoV-2 N protein to mix a zwitterionic surfactant containing a hydrocarbon chain as the hydrophobic part with the sample. Therefore, the N protein was detected by the immunochromatographic method of Example 6 above using a specimen treated with a zwitterionic surfactant containing a hydrocarbon chain as a hydrophobic moiety.
As specimens, purchased nasopharyngeal specimens (negative specimens, positive specimens, Boca Biolistics, LLC) were used. A Tris buffer solution (150 mM NaCl, 1% BSA, 0.1% NaN 3 ) containing each surfactant was used as the sample treatment solution. P1 antibody and N6 antibody were used as solid-phase antibodies, and N1 antibody, C3 antibody and C12 antibody were used as labeled antibodies. Table 13 shows the color development state of the detection zone under each condition. Higher numbers indicate darker coloration and "w" indicates weaker. In other words, the coloring intensity decreases in the order of 1>1w>1ww. As a result, as shown in Table 13, it was confirmed that the N protein could be detected with sufficient sensitivity.
Example 16: Investigation of Surfactant for Specimen Treatment Liquid (Part 2)
In Examples 14 and 15 above, it was found to be useful in measuring the SARS-CoV-2 N protein to treat the specimen with a zwitterionic detergent containing a hydrocarbon chain as the hydrophobic moiety.
Next, zwitterionic surfactants containing hydrocarbon chains as hydrophobic moieties and combinations with other surfactants were investigated by sandwich immunoassay methods using particles.
As specimens, 3 mL of virus transport solution SGVTM-3R (Sugiyamaken) was added with 6 μL of SARS-CoV-2 positive nasopharyngeal specimen (Boca Biolistics, LLC). Among the purchased SARS-CoV-2-positive specimens (Boca Biolistics, LLC), specimens that were found to have different viral amounts by RT-PCR were selected and used as low-value specimens and medium-value specimens.
Surfactants shown in Table 14 and 20 μL of a sample treatment solution containing 0.25% C16APS (Tris buffer, 150 mM NaCl, 1% BSA, 0.1% NaN 3 ) were added to 100 μL of the sample and heated at 37° C. Incubated for 6.5 minutes. After that, 50 μL of a diluted particle suspension solution containing a solid-phase antibody was added to the treated sample, and reacted at 37° C. for 8 minutes. After the reaction, B/F separation is performed with a magnet, and after washing with Lumipulse washing solution (manufactured by Fujirebio), 50 μL of the labeled body fluid containing the ALP-labeled antibody prepared in Example 4 above is added and allowed to react at 37° C. for 8 minutes. rice field. In addition, standard solutions (0, 100, 5000, 10000 pg/mL) containing known concentrations of recombinant N protein were measured in the same manner to create a standard curve. The concentration of N protein in each sample was determined from the amount of light emitted from each sample and the standard curve. The results are shown in Table 14.
As a result, by further adding deoxycholic acid or cholic acid to the treatment liquid, an increase in the amount of luminescence was observed for both the low value sample (LS) and medium value sample (MS). In particular, the additional addition of deoxycholic acid significantly increased the amount of luminescence at all added concentrations.
Addition of a component such as deoxycholic acid causes the solution to gel. Samples can be treated with a gelled solution, but for easier handling, adding a nonionic surfactant such as Tween 80, Tween 20 or NP-40 to the treatment solution will gel the solution. It was found that a high light emission amount can be obtained without
Example 17 Destruction of Capsid by Specimen Treatment The efficiency of capsid destruction by specimen treatment was examined.
As a method, genomic RNA extracted from virus particles was semi-quantified by RT-PCR to quantify the extracted RNA. At that time, by adding RNase in advance to the sample treatment solution or the sample diluent used as a control, if the sample treatment solution destroys the capsid of the virus, the RNA will be removed before the RNA extraction treatment. decomposed. The amount of RNA extracted from the particles by the treatment solution can be estimated by comparison with the amount of RNA before adding the RNase treatment. This allows us to estimate the efficiency of capsid disruption.
Specifically, sample diluent (Tris buffer, 150 mM NaCl, 1% BSA), 0.25% C16APS containing sample treatment solution (Tris buffer , 150 mM NaCl, 1% BSA, 0.1% NaN 3 ), each concentration of surfactant listed in Table 16, and 0.25% C16APS. LLC) was added to the specimen prepared by adding the diluent to the specimen, and after mixing, the mixture was allowed to stand at room temperature for 5 minutes. Then, RNA was extracted using an RNA extraction kit (GeneJET Viral DNA/RNA Purification kit (Thermo Fisher)) according to the attached manual. The obtained RNA was measured using an RT-PCR kit (SARS-CoV-2 Direct Detection RT-qPCR Kit (TaKaRa)), and the Ct value of RT-PCR was calculated. A higher Ct value indicates that RNA is degraded and the efficiency of capsid disruption is higher. The results are shown in Tables 15 and 16.
As a result, the Ct value increased by 4.9 by treating the specimen with the specimen treatment liquid (no addition) to the specimen diluent. That is, since the amount of RNA decreased to about 1/30, it was found that the extraction efficiency of RNA increased 30 times.
In addition, when 0.5% deoxycholic acid was added to the sample treatment solution, the Ct value did not change significantly, but the Ct value increased by 3.8 and 4.9 as it was added to 1% and 2%. . That is, it was found that the amount of RNA decreased by about 1/14 and 1/30, respectively, and the efficiency of RNA extraction increased by 14 times and 30 times, respectively.
Furthermore, when a nonionic surfactant (Tween 20) was added to the specimen treatment solution, the Ct value increased by 1. That is, the amount of RNA was reduced by 1/2, resulting in an increase in RNA extraction efficiency.
These results indicate that C16APS is effective for capsid extraction, and that addition of 1% or more deoxycholic acid and a nonionic surfactant enhances the RNA extraction effect, that is, the capsid destruction efficiency. It turned out to be.
