以下のゼラチン/カルシウム水溶液を作製した。平均分子量100,000(分布が20,000~300,000)アルカリ処理ゼラチン(ゼラチン濃度:20wt%)/0.5Mカルシウム水溶液。このゼラチン水溶液を用いて、トランスグルタミナーゼによる架橋試験を実施した。
トランスグルタミナーゼ(TG)は、活性86U/g、濃度6mg/ml、多糖類濃度600mg/mlのもの(名称:TGA、商品名:KS-OT(味の素社製))を使用し、含まれる多糖類を除去した後、以下の5種類のデキストランをそれぞれ600mg/ml添加した。架橋試験の結果をそれぞれ右に示す。
(1)DE値38、平均分子量4000、多孔質有り:架橋反応が5分以内
(2)DE値16、平均分子量15000、多孔質有り:架橋反応が30秒以内
(3)DE値10、平均分子量25000、多孔質有り:架橋反応した
(4)DE値10、平均分子量23000、多孔質無し:架橋試験が行えなかった
(5)DE値12、平均分子量20000、多孔質無し:架橋試験が行えなかった
また、止血機能について、架橋試験が行えなかったものを1、架橋反応したものを2、架橋反応が5分以内のものを3、架橋反応が2分以内のものを4、架橋反応が30秒以内のものを5と評価し、DE値と止血機能との関係を図2(A)、平均分子量と止血機能との関係を図2(B)に示す。
以上の結果より、止血機能の観点から、トランスグルタミナーゼとともに、DE値が10以上25以下、平均分子量が5000より大きく25000以下のデキストランを含むことが好ましいことがわかった。また、トランスグルタミナーゼとともに、DE値が25以上、平均分子量が5000以下のデキストランを含むものも好ましいことがわかった。
(Test 4)
The following gelatin / calcium aqueous solution was prepared. An aqueous solution of alkali-treated gelatin (gelatin concentration: 20 wt%) / 0.5 M calcium having an average molecular weight of 100,000 (distribution is 20,000 to 300,000). A cross-linking test with transglutaminase was carried out using this aqueous gelatin solution.
The transglutaminase (TG) used has an activity of 86 U / g, a concentration of 6 mg / ml, and a polysaccharide concentration of 600 mg / ml (name: TGA, trade name: KS-OT (manufactured by Ajinomoto Co., Inc.)) and contains polysaccharides. Was removed, and 600 mg / ml of each of the following five types of dextran was added. The results of the cross-linking test are shown on the right.

From the above results, it was found that from the viewpoint of hemostatic function, it is preferable to contain dextran having a DE value of 10 or more and 25 or less and an average molecular weight of more than 5000 and 25,000 or less together with transglutaminase. It was also found that those containing dextran having a DE value of 25 or more and an average molecular weight of 5000 or less are preferable together with transglutaminase.

The liquid medical material according to any one of claims 1 to 6, wherein the material is an arterial hemostatic material.
