形質導入Day−2に末梢血単核球(PBMCs)はCD3/CD28ynabeads(Invitrogen)を25μL/1×106cellsで刺激し、リコンビナントIL−2 30U/mLを添加したRPMI1640培地(10%FCS)で培養を開始した。形質導入Day1に24穴ノントリーメントプレートに1wellあたりPBMCs5×105cells/mlを1ml加えて形質導入した。なお、形質導入しない細胞、CD8CARを形質導入する細胞、CD26CARを形質導入する細胞は各2wellずつ形質導入した。適宜リコンビナントIL−2 30U/Lを含むRPMI1640培地(10%FCS)を添加し、Day3に2wellずつ形質導入していた細胞を回収し、1つのフラスコに拡大し細胞培養した。以後、適宜培地を足して拡大し、Day6にCD3/CD28Dynabeads(Invitorogen)を1サンプルにつき25μL加えた。CD26CAR導入後の細胞増殖は、Day10に細胞数をカウントして比較した。CD26CAR2ndとCD26CAR3rdのいずれも、CD8CAR2ndやCDCAR 3rdと同様に増殖した(図4A)。

リンパ球の活性化に伴って分泌されるIFN−γは、Human IFN−gamma Duo Set(R&D System)を使用し、プロトコールに沿ってELISA法で確認した。まず、CAR導入後Day10のPBMCsを1×105cells/100μL、CD26強制発現Jurkat細胞(CD26−Jurkat)を1×105cells/100μLにRPMI1640培地で調整し、96穴プレートに、それぞれ1wellあたり100μL加えて12時間共培養した。また、ELISA前日にプレートに4μg/mlになるようにPBSで調整したCapture Ab 100μL を加えて室温で一晩静置した。上清を除去した後、1%牛血清アルブミン(BSA)を含むPhosphate−buffered saline(PBS)300μLを加えてブロッキングを行い、室温で1時間静置した後、1%BSA/PBSで洗浄した。プレートに細胞培養後の上清100μLを加え、2時間静置した後、1%BSA/PBSで洗浄した。1%BSA/PBSで400ng/mlとなるよう調整したDetection Abを100μL加えた後1時間室温で静置し、1%BSA/PBSで洗浄した。Streptavidin−HRPを100μL加え、30分室温で静置した後、1%BSA/PBSで洗浄し、TMB Solution 100μLを加えた。30分室温で静置後、TMB stop solution50μLを加えて、O.D.450nmで測定した。CD26CAR2ndとCD26CAR3rdのいずれにおいてもIFN−γの分泌が認められた(図6A)。
