筋萎縮性側索硬化症(ALS)は、急速に進行する疾患であるので、薬剤の継続的投与による長期的な影響を考慮せずに構成的栄養因子の放出で十分であり得る。対照的に、より長い寿命を有する他の多くの神経学的疾患は、持続的な成長因子分泌を必要としない場合があり、または長期間にわたる送達が有害である場合がある。実際に、尾状核、尾状核被殻でGDNFを一時的に受けただけのパーキンソン病患者の死後分析により、GDNF送達停止後の維持された効果を説明し得る持続的な神経発芽が示された。さらに、構成的成長因子の放出は、禁忌の場合における中止が可能ではない。例えば、試験より、GDNFを受けた一部の患者がLhermitte症状を発症したことが報告されている。加えて、持続したGDNF発現は、標的化ニューロンの異常な発芽及び脱感作を引き起こし得る。重要なことに、これらの影響は、GDNFの停止時に後退した。全体として、これらのタイプの臨床用途の成長因子の制御に対する明確な理論的根拠、ならびにこれらの目標を達成する組成物及び手法の当該技術分野に大きいニーズがある。

細胞培養物または動物へのiPSC及びiNPCのDox添加
全ての実験では、Dox溶液を遮光して4℃に維持した。細胞培養では、dox(Clontech 631311)を100ng/mlで培地に添加した。in vivo研究では、1mlのシリンジ(BD 309659、Franklin Lakes,NJ)に取り付けられた先端が柔らかい給餌針(Instech FTP−20−30、Plymouth Meeting,PA)を使用して、経口強制飼養(例えば、20gの動物に対して60μl)で3〜4日毎に、動物にdox(15μgのdox/g体重)を投与した。
移植
BFPについてFACSを2回受けたヌクレオフェクトiNPCを、6ヶ月齢のNOD−SCIDマウス(Nod.cb17−Prkdcscid/J,Jackson Lab番号1303、Bar Harbor,ME)の線条体に移植した(各半球に100,000細胞/μlの細胞を2μl)。移植の3日前に、細胞のサブセットをdoxで処置した。移植のために、細胞を解離し、馴化培地中で最終濃度100,000細胞/μlに希釈し、氷上に維持した。動物をイソフルランで麻酔し、定位フレームに置いた。前頂部に対して吻側に0.7mm、外側に+/−2.0mmの両方に、細胞を注射した。PBSで充填した5μlのハミルトンマイクロシリンジに、2μlの細胞溶液を充填し、深さ−4.00mmに挿入した。次に、マイクロシリンジを−3.50mmに持ち上げ、細胞溶液を1μl/分で注射した。細胞注射後、マイクロシリンジを−3.00mmに持ち上げ、2分間保持し、徐々に引き上げた。毎日、動物をチェックした。
レポーター遺伝子及びGDNFの制御された発現を提供する新しいベクター
Tet−Onシステムは、rtTA−V10トランスアクチベーター及びdox応答TRE−Bi(Tet応答因子双方向性)プロモーターの構成的発現に依存する。トランスアクチベーター及び誘導性プロモーターが異なるプラスミドで発現し且つ細胞に共トランスフェクトされた場合、細胞が増殖中にドリフトし、二重トランスジェニック集団に対して選択され、その結果、非誘導性細胞がもたらされる(データは示さず)ことを、本発明者らは観察した。単一のシステムに、これらの因子及びレポーター遺伝子を組み込むには、大きなベクターが必要となり、これは、本発明者らが新たに作成した、「pB−RTP−Tet−GDNF/memClover−FLuc」[piggyBac−Reverse transactivator/TagBFP2nls/PacR−Tet inducible−GDNF/membrane Clover−Firefly Luciferase](図1A)と呼ばれるベクターで達成された。ベクターは、piggyBac末端反復(PB TR)が隣接しており(図1A1)、これは、pBase酵素が一過性に発現された時に、安定したゲノム取り込みを可能にする。
ヌクレオフェクトiPSC由来NPCの免疫細胞化学
細胞接着を促進するために、カバーガラスをポリオルニチン(Sigma、P4638)でコーティングし、続いて、25μlのラミニン(50μg/ml、Sigma−Aldrich L2020)を用いて37℃で1時間処理した。次に、コーティングを除去した。TrypLE中で5分間インキュベートすることにより、6ウェルプレートからヌクレオフェクト細胞を取り出した。等量の培地を加えて、酵素反応を中和し、次に、解離細胞をカウントし、150rcfでペレット化し、1,000細胞/μlで培地に再懸濁し、25μlの細胞をカバーガラスのラミニンコーティング領域にピペッティングした。細胞接着させるために、37℃で4〜6時間インキュベートした後、ウェルを500μlの成長培地で静かに満たした。半分の体積の培地を24時間毎に交換した。
ELISA
ELISAに使用される培地は、EGF及びFGFを含まず、1%の抗生物質−抗真菌剤が補充されたDMEM:F12(70:30)、及びビタミンAを含まない2%のB27からなる。24時間または7日間のインキュベーション期間中に培地を添加する前に、古い培地を吸引し、ウェルを滅菌PBSで2回洗浄した。収集された培地を−80℃で保存した。製造者の使用説明書に従って、GDNF ELISAを実施した(R&D Systems DY212,Minneapolis,MN)。
イメージング
免疫細胞化学及び免疫組織化学蛍光染色後、色を順番にイメージングし、シグナルクロストークを回避する適切な設定を有するNikon A1R倒立レーザー走査型共焦点顕微鏡で、共焦点画像を収集した。露出及び彩度の測定を利用して、最大ダイナミックレンジをキャプチャした。通常、露出が設定された後、群内の後続の試料に、同一の設定を再利用した。共焦点Zスタックを操作するか、または後続のAdobe Photoshop CS6での編集用の単一のZスライスから個々のチャネルを分離するために、最初に、ND2画像ファイルをImageJにインポートした。Photoshop CS6でダイナミックレンジ及び露出の一貫性のために画像曲線を調整し、トリミングし、次に、最終的な画像を準備するためにAdobe Illustrator CS6にインポートした。
移植組織のGDNFスコア付け
Leica DM200 LED顕微鏡を使用して、免疫染色された切片を撮影した。実験条件(dox対doxなし)を盲検化された観察者が、DAB GDNF染色の染色強度に基づいて、画像をスコア付けした。
レポータータンパク質の産生は、in vitroで可逆的に制御される。
治療関連細胞でのdox制御システムを評価するために、ヒトiNPCをpB−RTP−Tet−GDNF/memClover−FLuc及びpBaseベクターでヌクレオフェクトした。容易に維持し、急速に増殖させることができる浮動性スフィア(EZ sphereと呼ばれる)として、ヌクレオフェクト細胞を成長させた。doxの不存在下では、スフィアは、BFP+を構成的発現し、doxの添加から24時間以内に、スフィアは、BFP+及びGFP+の両方を発現した。これは、TRE−Biプロモーターがdoxに応答したことを示す(図1D)。
GDNF産生は、in vitroで可逆的に制御される。
本発明者らのシステムが治療関連分子を確実に制御できるようにするために、次に、GDNFの生成を、ヌクレオフェクトiNPCで評価した。免疫蛍光法により、GDNFが、doxの不存在下で細胞では見えないことが明らかになった。これにより、野生型細胞がGDNFを産生しないという本発明者らの以前の報告が確認される。レポータータンパク質であるClover及びルシフェラーゼもdoxの不存在下で生成されなかったが、BFPは構成的プロモーターの下で生成された(図2A)。doxの存在下では、BFP産生の維持に加えて、GDNFならびにClover及びルシフェラーゼが検出された(図2B)。その後にdoxが除去された時、GDNF及びレポーター遺伝子の産生は全て停止したが、BFPは維持された(図2C)。
成体脳へのiNPC移植において、レポーター導入遺伝子発現は、誘導可能及び可逆的である。
このシステムがin vivoでのタンパク質の発現を制御する能力を調査するために、ヌクレオフェクトiNPCを増殖させ、成体NOD−SCIDマウス脳に移植した(図3A)。in vitroでの増殖中に、細胞は、核BFPについて2回のFACSを受け、親細胞集団は、15.6%のBFP+を含有しており、次に、これを、ニューロスフィアとして7週間増殖させ、2回目にBFPについて選別した。これは、32.4%のBFP+細胞を生じた。第2の選別の3週間後、細胞を、2群に分け、一方の細胞群は、培養中、移植前の3日間doxを受け、他方の細胞群は、doxを受けなかった。6ヶ月齢のNOD−SCIDマウス(n=14)に200,000のiNPCを各線条体半球に移植し、10匹の動物がdoxで前処置された細胞を受け、4匹の動物が未処置の細胞を受けた。動物は、移植前にdoxを受けなかった。
iNPC移植において、GDNF発現は、誘導可能及び可逆的である。
移植細胞でのGDNF発現の制御を確認するために、最後のイメージングセッション後に動物を安楽死させ、蛍光免疫組織化学により、GDNF、Clover、及びBFP産生を評価した(図4)。オンdoxで試験を終了した動物(図4A)及びオフdoxで試験を終了した同腹仔(図4B)における移植により、線条体領域にBFP+細胞が示されたが、Clover及びGDNFは、dox動物でのみ検出された。高い増幅度により、dox処置動物では移植部位に隣接する宿主細胞がGDNFを吸収し得るのに対して(図4C、ピンクの矢印)、オフdox動物ではこれが観察されなかった(図4D)ことが示された。まとめると、これらのデータにより、GDNF分泌が移植の数か月後のiNPCにおいて誘導及び後退させることができることが表された。
考察
このレポートは、ヒト神経前駆細胞の操作により産生されたGDNFは、in vivoで複数サイクルにわたって厳密に制御することができるという概念実証を提供する。この技術は、他の成長因子にも適用することができ、種々の疾患で損傷したニューロンを保護する有益な手段を提供する。pB−RTP−Tet−GDNF/memClover−FLucを使用して、doxが、誘導可能なTRE−Biプロモーターからの導入遺伝子活性化を可能にするレベルまで、実質的な移植部位に浸透することを、本発明者らは観察した。本発明者らはまた、導入遺伝子活性が、複数サイクルで誘導及び後退させることができることを報告する。従って、疾患の進行中に患者が副作用、脱感作、または他の導入遺伝子関連現象を発症した場合、GDNF投与を減弱または停止することができる。あるいは、薬物レジメンは、宿主受容体及びシグナル伝達経路の再感作を可能にするように変化させることができる。これは、構成的遺伝子発現を伴うex vivo遺伝子治療とは著しく対照的である。重要なことに、遺伝子制御に必要なdox濃度の量(15μgのdox/g重量)は、抗菌用量未満であり、dox耐性菌の存在を増加させないか、または腸内細菌叢に悪影響を及ぼさない。
ヒトiPSC細胞株におけるAAVS1「安全なランディング部位」からのヒトGDNFの誘導可能な発現
治療環境に上記構築物を適用すると、有害な影響を回避するための「安全な」組み込みから大いに利益を得るであろう。これは、任意に、制御可能な発現を含む、ヒトGDNFを産生するDNAベクターの開発を含み、これは、ヒトAAVS1遺伝子座への組み込みを目的とすることができる。
iPSC細胞におけるGDNFの発現
蛍光タンパク質であるCloverの発現に基づく、AAVS1−teton−hGDNF含有細胞の同定を容易にするために、培養物をテトラサイクリンアナログであるドキシサイクリンの存在下に維持する。「緑色」コロニーを、増殖のために蛍光顕微鏡下で厳選した。十分な数の細胞が生成されると、クローンをFACS選別によりさらに精製した。同時に、培地をこれらのクローンから収集して、ELISAアッセイによりGDNF産生を確認した。
GDNF発現プラスミド(TREBi及びTRB3誘導性プロモーター)の単一コピー多様体のクローン
上述の別の例としては、GDNF発現プラスミドの単一コピー多様体が挙げられる。TREBi及びTRB3で誘導性プロモーターを使用して、そのようなベクターは、eGFP及びLuc2などのレポートを含み得る。例としては、図10に示されるpDonor−Teton3g−2a−TagBFP−V5−nls−p2a−puroR WPRE_隔離型 mpclover−2a−luc2pest−2a−gdnf wpre[配列番号3]が挙げられる。そのようなベクターとしては、相同組み換え配列が挙げられる。ここでは、内因性遺伝子座のAAVS1標的化は、ヒトPPP1R12C遺伝子のエクソン1及びエクソン2間にある。レシピエント「ランディング部位」は、上流のPPP1R12Cに連結されているスプライスアクセプターにより駆動されるレポーター/選択カセット(蛍光及び抗生物質選択のためTagBFP2及びPuroR)を含み、LoxP部位及びFRT部位に隣接する、構成的CAGプロモーターで駆動されるtd−Tomato赤色蛍光シストロンは、安定的に組み込まれた。続いて、これらのレポーターについて安定的に選択した時に、FlpO及びCreを発現するプラスミドで、これらの細胞をリポフェクトし、このドナープラスミドは、LoxP及びFRT隣接選択/レポーターカセットを含み、ならびにdoxで誘導可能なmpClover/Luc2p/GDNFシストロン含有プラスミドでリポフェクトした。

- 複数の人工多能性幹細胞由来神経前駆細胞(iNPC)を、神経変性障害に罹患している個体に投与することを含む、前記個体の中枢神経系におけるグリア由来神経栄養因子(GDNF)レベルを増加させる方法であって、
