第1基板の表面上にレジストを塗布した後、電子ビームによりレジストの一部を除去しレジストにパターンを形成した。その後、レジストをマスクとしてエッチングを行い第1基板の一部を除去し溝を形成した。このようなエッチング処理により、第1基板に扁平流路、第1流路、第1連通流路、第2流路、第1収集チャンバ、第2連通流路などとなる溝を形成した。また、フォトリソグラフィとエッチング技術により、第2基板に導入流路及び排出側流路となる溝を形成した。第1連通流路の流路幅Waは140nmとし、第2連通流路の流路幅Wcは110nmとした。また、第1及び第2流路の深さ、第1及び第2連通流路の深さ、並びに第1収集チャンバの深さは、250nmとした。
その後、洗浄した第1基板及び第2基板を接触させて真空炉内に入れ、1060℃で熱処理することにより第1基板と第2基板とを融着させた。このようにして、第1基板と第2基板との間に導入流路、扁平流路、第1流路、第1連通流路、第2流路、第1収集チャンバ、第2連通流路、排出側流路などが形成された粒子収集装置を製造した。

図8はHT−29細胞由来のエクソソームの粒径分布を示すグラフであり、図9はエクソソーム収集実験1の結果を示すグラフである。図9のグラフのように、第1収集チャンバに収集される粒子量は、送液圧力を大きくするほど多くなることがわかった。
図8のグラフのように、HT−29細胞由来のエクソソームは、110nm(Wc)以上140nm(Wa)以下の粒径範囲内のものが多いため、送液圧力の増加に伴い第1収集チャンバに収集される粒子量が多くなったと考えられる。

図10はHEK−293T細胞由来のエクソソームの粒径分布を示すグラフであり、図11はエクソソーム収集実験2の結果を示すグラフである。図11のグラフのように、第1収集チャンバに収集される粒子量は、毛管現象で第1流路などに液体を流したときよりも、シリンジポンプを利用して第1流路などに流したときの方が多くなったが、シリンジポンプの送液圧力を増加させても第1収集チャンバに収集される粒子量はほぼ一定であった。
図10のグラフのように、HEK−293T細胞由来のエクソソームは、110nm(Wc)以上140nm(Wa)以下の粒径のものが少ないため、送液圧力を増加させても第1収集チャンバに収集される粒子量は多くならなかったと考えられる。
