Abstract
Description
本発明は、粒子検出で用いる流体に関する。 The present invention relates to a fluid used in particle detection.
粒子を個々に1つずつ測定することで、マイノリティーな粒子群を正確に検出可能な技術として、電気的検出を用いるコールター法(電気的検知帯法;以下ESZと記載)(例えば、非特許文献1参照)が知られている。この手法は、各検出手段から得られる情報(シグナル)が、各粒子に対して1対1で対応しているため粒子個々の評価をすることが可能であり、数的に含まれる割合の少ない粒子でも正確に測定できる。 The Coulter method (electrical detection band method; hereinafter referred to as ESZ) using electrical detection is a technique capable of accurately detecting a minority particle group by measuring each particle one by one (for example, non-patent literature). 1) is known. In this method, since the information (signal) obtained from each detection means has a one-to-one correspondence with each particle, it is possible to evaluate each particle individually, and the ratio contained numerically is small. Even particles can be measured accurately.
ESZ法ではアパーチャに粒子を通過させた際に発生する電気的シグナルを用いて粒子径を算出するが、一般にそのダイナミックレンジはアパーチャ径の2〜60%といわれている。ESZ法の欠点であるダイナミックレンジの狭さを解消するため粒子を分級し、異なるアパーチャ径を持つESZ法で検出する方法が開発されている(例えば、特許文献1)。 In the ESZ method, the particle size is calculated using an electrical signal generated when particles are passed through the aperture, and the dynamic range is generally said to be 2 to 60% of the aperture diameter. In order to eliminate the narrow dynamic range, which is a drawback of the ESZ method, a method of classifying particles and detecting them by the ESZ method having different aperture diameters has been developed (for example, Patent Document 1).
As a technique for enabling continuous separation as in
The types of particles that can be measured by the apparatus described in
本発明の課題は、PFFとESZの組み合わせによる粒子検出において、高感度な粒子検出が可能な流体を提供することにある。 An object of the present invention is to provide a fluid capable of highly sensitive particle detection in particle detection by a combination of PFF and ESZ.
本発明者らは上記課題を鑑み、粒子を希釈懸濁する流体および分離に使用する流体に含まれる界面活性剤濃度を最適化することで高感度な粒子検出が可能となることを見出した。
すなわち、本発明は、Pinched Flow Fractionation法と電気的検知帯法の組み合わせによる粒子検出において用いることができる、電解質を含み且つ界面活性剤濃度0%以上10%(V/V)未満であることを特徴とする流体である。
In view of the above problems, the present inventors have found that highly sensitive particle detection is possible by optimizing the concentration of the surfactant contained in the fluid for diluting and suspending the particles and the fluid used for separation.
That is, the present invention contains an electrolyte and has a surfactant concentration of 0% or more and less than 10% (V / V), which can be used in particle detection by a combination of the Pinked Flow Fractionation method and the electrical detection band method. It is a characteristic fluid.
本発明により、Pinched Flow Fractionation法と電気的検知帯法の組み合わせによる粒子検出において、高感度な粒子検出が可能な流体を提供することができる。 INDUSTRIAL APPLICABILITY According to the present invention, it is possible to provide a fluid capable of highly sensitive particle detection in particle detection by a combination of a Pinked Flow Fractionation method and an electrical detection band method.
以下、本発明を実施するための形態について、図面を用いて詳細に説明する。但し本発明は異なる形態による実施が可能であり、以下に示す実施形態、実施例の例示にのみ限定されるものでは無い。 Hereinafter, embodiments for carrying out the present invention will be described in detail with reference to the drawings. However, the present invention can be implemented in different embodiments, and is not limited to the examples of the embodiments and examples shown below.
本発明による粒子検出装置の装置構成の一実施例を示す模式図である図1をもとに本発明の実施形態についての詳細を説明する。 The details of the embodiment of the present invention will be described with reference to FIG. 1, which is a schematic diagram showing an embodiment of the device configuration of the particle detection device according to the present invention.
As shown in FIG. 1, the fluid 100P containing particles and the fluid 100N not containing particles are introduced from the
The inside of the particle detection unit 103 and the outlet 104 are filled with a fluid containing an electrolyte, and the concentration of the surfactant thereof is 0 wt% or more and less than 10%. Preferably, it is 0% or more and less than 5%. Electrolytes are essential and salts can be used. As an example, salts of sodium chloride, potassium chloride, lithium chloride, sodium acetate, potassium acetate, lithium acetate and amino acids such as histidine hydrochloride, arginine hydrochloride and glycine hydrochloride can be used. Sodium chloride is particularly preferred.
The fluid may be mixed with a buffer or an additive. As an example of the buffer, phosphoric acid, citric acid, acetic acid, succinic acid, trishydroxymethylaminomethane, histidine, glycine, arginine and salts thereof can be used, and a buffer solution of phosphoric acid is preferable. Examples of additives include surfactants such as Tween 20 and Tween 80, amino acids such as arginine, histidine and glycine, sugars such as sucrose, mannitol, sorbitol and trehalose, preservatives such as sodium azide, and antibiotics such as streptomycin and penicillin. You may. Water is preferable as the solvent, but an organic solvent, oil, or oil may be used as long as the electrolyte is soluble.
粒子を含む流体100Pは、測定対象とする粒子を含んだ流体である。本発明における粒子とは、粒径が1nm〜1000μm、好ましくは10nm〜100μmの範囲にあり、例えば、核酸、タンパク質、小胞、リポソーム、細胞外小胞、無機粒子、金属コロイド、高分子粒子、ウイルス、細胞、細胞塊、前記粒子の凝集体や会合体、融合体などが含まれる。 The fluid 100P containing particles is a fluid containing particles to be measured. The particles in the present invention have a particle size in the range of 1 nm to 1000 μm, preferably 10 nm to 100 μm, and include, for example, nucleic acids, proteins, vesicles, liposomes, extracellular vesicles, inorganic particles, metal colloids, and polymer particles. Includes viruses, cells, cell clumps, aggregates and aggregates of the particles, fusions and the like.
インレット14aは、粒子を含むサンプルを保持できる構造であればよく、凹型構造であることが好ましい。また材質としては、溶出物が少ない金属やガラス、セラミクスを用いてもよいが、安価に製造するために高分子材料で形成されることが好ましい。
The
The liquid feeding unit may use a method of feeding liquid by a pressure gradient such as a syringe pump, a perista pump, or a pressure feeding pump, or an electroosmotic flow pump in order to suppress a non-uniform velocity distribution in the flow path cross section of the
送液部の流量は、流路の断面積やアパーチャの断面積により任意の値に設定することが好ましく、一例として、0.1μL/hourから1mL/hourの間に設定することが好ましい。 The flow rate of the liquid feeding unit is preferably set to an arbitrary value depending on the cross-sectional area of the flow path and the cross-sectional area of the aperture, and as an example, it is preferably set between 0.1 μL / hour and 1 mL / hour.
Two or more particle recovery flow paths 102 are provided from the viewpoint of expanding the measurable particle size range (dynamic range). Here, the particle detection unit 103 is provided inside or downstream of the particle recovery flow path 102, and is used to detect the particles that have flowed into the particle recovery flow path 102. At least one particle detection unit 103 needs to be provided for one particle recovery flow path 102, and two or more particle detection units 103 or two or more apertures may be provided. As shown in FIG. 2, the particle detection unit 103 is composed of two apertures, the downstream of each aperture is connected to an outlet, and the electrode 54 is immersed in each outlet to function as an electrode insertion port 59. Particle detection by ESZ may be performed in this way.
The particle detection unit 103 includes an
The
図2のように粒子検出部103のアパーチャが複数設けられている場合、各々の断面積または体積が同じであれば両アパーチャから得られるシグナルは凡そ同一となる。すなわち、両アパーチャへ流れてきた粒子全てを同様に検出することが可能であり、濃度の定量的な測定という観点で好ましい。 When a plurality of apertures of the particle detection unit 103 are provided as shown in FIG. 2, if the cross-sectional areas or volumes of the apertures are the same, the signals obtained from both apertures are substantially the same. That is, it is possible to detect all the particles flowing into both apertures in the same manner, which is preferable from the viewpoint of quantitative measurement of the concentration.
電源57は直流または交流電源を用いられるが、測定の際によりノイズが影響しにくいものを選択する方が好ましく、コスト面からは、例えば乾電池等の安価で低ノイズである直流電源を用いる方が好ましい。また、電極の材料は電気抵抗が小さい材質であれば制限はなく、金属、無機化合物、有機化合物を用いることができるが、耐久性とコストの面から金属であることが好ましい。
A DC or AC power supply is used as the
The electric detector of the particle detection unit 103 mainly includes an electrode 54, an electric measuring instrument 56 connected to the electrode 54 via a
The analysis unit 61 can include an arithmetic unit for calculating the measurement result and a recording medium for recording the measurement result or the calculation result derived from the measurement result. Alternatively, these arithmetic units and recording media may be integrated with the electric measuring instrument 56, or may be an external device that can be connected to the electric measuring instrument 56. The data recorded on the recording medium includes the sampled current value, the change in the current value generated when the particles pass, and the particle size, number of particles, particle concentration, detection time or measurement calculated from the change in the current value. The elapsed time from the start is included.
粒子検出装置の製造
本発明による粒子検出方法を実施するための粒子検出装置の実施形態を備えたマイクロチップ10は、一般的なフォトリソグラフィーとソフトリソグラフィー技術を用いて作製した。具体的な手順を以下の通り示す。
4インチベアシリコンウェハ(株式会社フィルテック)上へ、フォトレジストSU−8 3005(Microchem社)を滴下後、スピンコーター(MIKASA社)を用いてフォトレジスト薄膜を形成した。この時、目的膜厚に応じて、SU−8 3005へ希釈剤Cyclopentanone(東京応化工業社)を添加した。
Manufacture of Particle Detection Device The
A photoresist SU-83005 (Microchem) was dropped onto a 4-inch bare silicon wafer (Filtech Co., Ltd.), and then a photoresist thin film was formed using a spin coater (MIKASA). At this time, a diluent Cyclopentanone (Tokyo Ohka Kogyo Co., Ltd.) was added to SU-8 3005 according to the target film thickness. Subsequently, a channel pattern is formed on the photoresist film using a mask aligner (Ushio, Inc.) and a chrome mask having an arbitrary pattern formed, and the channel pattern is developed using SU-8 Developer (Microchem). As a result, a mold for the flow path to be used was prepared.
Subsequently, uncured LSR7070FC (Momentive Performance) was poured into the prepared mold and heated at 80 ° C. for 2 hours to prepare polydimethylsiloxane (PDMS) having the shape of the flow path transferred. The cured PDMS was peeled off from the mold, molded to an arbitrary size with a cutter, and then the inlet and outlet of the flow path were formed using a puncher. The peeled PDMS and the slide glass (Matsunami Glass Co., Ltd.) were surface-treated with an oxygen plasma generator (Meiwaforsis Co., Ltd.), and then the PDMS and the slide glass were bonded to each other to prepare a
