本明細書に記載の活性化可能抗CD166抗体は、抗体治療、特にin vivoで少なくともある程度毒性であることが知られている抗体治療の制限を克服する。標的を介した毒性は、治療用抗体の開発にとって大きな制限となる。本明細書で提供される活性化可能抗CD166抗体は、従来の治療用抗体による正常組織の標的の阻害に関連する毒性に対処するように設計されている。これらの活性化可能抗CD166抗体は、疾患部位でタンパク質分解的に活性化されるまでマスキングされたままである。親治療用抗体としての抗CD166抗体から始めて、本発明の活性化可能抗CD166抗体は、プロテアーゼ基質(CM)を組み込んだリンカーを介して抗体を阻害マスク(マスキング部分MM)に連結することにより改変させた。

実施例1:CD166に結合する結合型活性化可能抗体の産生および試験
以下に示される実施例で使用されるAAは、本明細書で提供され、その内容全体が参照により本明細書に組み込まれるPCT公開番号WO2016/179285に開示されている方法を使用して生成し、かつ特徴を明らかにした。
本試験では、高いCD166発現腫瘍(乳癌、肺癌、前立腺癌、卵巣癌、子宮内膜癌、頭頸部癌、胆管癌)の単剤療法として投与される活性化可能抗CD166抗体薬物コンジュゲートの安全性の一次エンドポイント、最大耐量(MTD)、推奨フェーズ2用量(RP2D)、用量制限毒性、および予備的な抗腫瘍活性を評価する。
この実施例は、7614.6−3001−HuCD166を投与されたマウスの血漿および異種移植腫瘍試料中で活性化されている、および完全な抗CD166活性化可能抗体7614.6−3001−HuCD166を検出する能力について記載する。
この実施例は、SPDBリンカーを介してメイタンシノイド毒素DM4に結合した、活性化された完全な抗CD166活性化可能抗体を検出する能力について記載する(組み合わせ55)。
この実施例は、SPDBリンカー(組み合わせ55)を介してメイタンシノイド毒素DM4に結合した完全抗CD166活性化可能抗体の投与が、対象において部分的応答をもたらすことを実証している。
この実施例は、ヒト対象への投与後、SPDBリンカー(組み合わせ55)を介してメイタンシノイド毒素DM4に結合した合計の完全抗CD166活性化可能抗体の薬物動態を実証している。
