Fc−hRLX2−4−15AAおよびFc−hRLX2−15−15AA遺伝子配列は、GeneArt(登録商標)(Regensburg、Germany)によって合成し、プラスミドDNAとして供給した。遺伝子配列を、XbaIおよびNotIによる制限酵素消化によってGeneArt(登録商標)プラスミドから取り出し、同じ制限消化部位を使用して、pHOEベクターの中に別々にクローニングし、それぞれpOE−Fc−hRLX2−4−15AAプラスミドおよびpOE−Fc−hRLX2−15−15AAプラスミドがもたらされた。pHOEベクターは、ヒトIgG1の重鎖部分を発現するように使用する。このクローニング戦略により、ベクター抗体配列はすべて除去したが、シグナルペプチドおよびFc配列の間の介在配列は保存した。

インビトロにおける細胞ベースのアッセイは、実施例1において生成したリラキシン2融合ポリペプチドの活性を測定するために使用した。アッセイにより、融合ポリペプチド(Fc−hRLX2−4−15AAおよびFc−hRLX2−15−15AA)がcAMP産生を刺激する能力を測定した。
試験した構築物の生物学的活性を、表2および図16(1回の実験)および17(繰り返しの実験)において提供する。
リラキシンファミリーペプチドは、4つのGタンパク質共役受容体(GPCR)のグループを活性化することによって、それらの生理学的効果をもたらし、これらは、それらのリガンドによって刺激する(Gs)または阻害する(Gi)ことができる。リラキシン2は、受容体RXFP1に特異的に結合する。

をDiscoverX(Fermont、CA)から購入した。
結果を図22に示す。試験したリラキシン2融合ポリペプチドは、RXFP2発現細胞株において最少の活性しか有しておらず、他の関係のあるRXFP発現細胞株(RXFP3およびRXFP4)において活性でない。予想通り、ADCYAP1R1 Gs/Gq細胞株において活性は観察されなかった。
リラキシン2融合ポリペプチドが、リラキシン2の既知の下流の標的であるVEGFの発現を誘発する能力について(Xiao et al.,2013)、アッセイした。THP−1細胞(カタログ番号TIB−202、ATCC)におけるVEGF RNAの発現は、定量リアルタイムPCRによって分析した。合計106細胞/mlを、400μl試験培地(フェノールレッド、0.5% FBS、1% Pen/Strep、および0.05mMの2−メルカプトエタノールなしのRPMI−1640)において24ウェル平底プレート(カタログ番号353226、Corning)に接種した。37℃、5%CO2での24時間のインキュベーションの後に、ヒトリラキシン−2(カタログ番号3956−RN、R&D systems)またはFc−hリラキシン−2融合ポリペプチドを37℃で2.5時間、プレート中の細胞に追加した。THP−1細胞からのRNA単離および精製は、メーカーのプロトコールに従って、QiagenのQiaShredder(カタログ番号79656、Qiagen)およびRNA Plus Mini Kit(カタログ番号74134、Qiagen)を使用して実行した。mRNA濃度は、NanoDrop(Thermofisher)によって測定し、サンプルは、同一の出発濃度(RT−PCR設定;50〜20ng/μlの間の範囲)に対して標準化した。RT−PCRサンプルは、Express One−Step Superscript qRT−PCR kit(カタログ番号11791−200、Invitrogen)を使用して調製し、プライマー/プローブセットは以下のとおりとした:VEGFAヒト(Hs00900055_m1、カタログ番号4331182、ThermoFisher Scientific)およびGAPDHヒト(Hs02758991_g1、カタログ番号4331182、ThermoFisher Scientific)。RT−PCR反応は、7900HT Fast Real−Time PCR machineを使用して実行し、サイクルの設定は以下のとおりとした:cDNA合成のための15分間50℃での1サイクル、Taq活性化における20秒間95℃での1サイクル、ならびにqPCRのための1秒間95℃および20秒間60℃の2つの温度での40サイクル。VEGF mRNAレベルの相対的倍数変化は、GAPDH発現を標準化に使用して、比較Ct法によって計算した。結果は、治療なしコントロールに対する倍数変化として計算した。実験は、n>2で三通り実行した。データは、GraphPad、V6を用い、対応のある両側t検定によって分析した。
図23(上)に示されるように、5ng/ml、50ng、および80ng/mlの濃度で、偽「治療なし」コントロールと比較して、試験したリラキシン2融合ポリペプチドは、THP−1細胞においてVEGF転写物を有意に刺激する。同様に、市販で入手可能な組換えヒトリラキシン2(R&D systems)もまた、0.01、0.06、および10ng/mlの濃度で、「治療なし」コントロール細胞と比較して、THP−1細胞においてVEGF転写物を有意に増加させた。ヒトリラキシン2と比較するために、Fcリラキシン2融合ポリペプチド濃度は、分子量の差(6×)およびcAMPアッセイにおいて見られた活性の低下(10〜15倍)に基づいて調整した。
本発明のいくつかのリラキシン2融合ポリペプチドの薬物動態学的(PK)プロファイルをリラキシンELISAアッセイを使用して決定した。ラットインビボPKプロファイルを以下の融合ポリペプチドについて決定した:Fc_hRLX2_4−15AAおよびFc_hRLX2_15−15AA。
図24は、ラットにおける皮下(SC)または静脈内(IV)投与後のFc_hRLX2_4−15AAのインビボにおけるPKプロファイルを示す。SC投与後、ピーク血漿濃度は、8〜24時間以内に実現した。ノンコンパートメント解析から推測した平均終末相半減期は、IV用量グループについては2.46日であり、SC用量グループについては3.21日であった。
Fc融合ポリペプチド中に存在するYTE突然変異(M252Y/S254T/T256E)によって与えられる長期の半減期を、社内で生成したFcRnトランスジェニックマウスにおいて6mg/kgの用量で静脈内にまたは皮下に投与するFc−YTE−hRLX2−15−15AA、Fc−YTE−FQQ−hRLX2−15−15AA、およびFc−YTE−TM−hRLX2−15−15AA融合ポリペプチドについて実行するPKプロファイル研究によって確認する。長期の半減期は、Fc捕捉およびリラキシン検出ELISAアッセイを使用して判断した(R&D Systems Relaxin detection ELISA kit、カタログ番号DRL200のポリクローナル抗体を使用)。
実施例1において記載されるように産生した本発明のFc−リラキシン−2融合ポリペプチドのいくつか(Fc_hRLX2_15−15AA, Fc−TMΔTHTΔK_hRLX2_21−15AA, Fc−TMΔTHTΔK_hRLX2(BA)_21−15AA, hRLX2−Fc−TMΔTHTΔK_15−24AA, hRLX2(BA)−Fc−TMΔTHTΔK_15−24AA)を以下の方法によって、生物学的活性、たとえば、1つ以上の細胞の受容体応答の刺激について試験した。
試験した融合ポリペプチドの生物学的活性を表3に提供し、それぞれのポリペプチドについての1つの代表的なアッセイの結果を図28に示す。
インビトロにおける細胞ベースのcAMP刺激アッセイのばらつきのために、組換えヒトリラキシン−2を、ポジティブコントロールとしてそれぞれのアッセイにおいて常に使用する。
−リラキシン融合ポリペプチドのN−末端にまたはリラキシン融合ポリペプチドのC−末端に付けられたFc成分を含む試験したリラキシン−2融合ポリペプチドは、生物学的に活性である。

−RXFP1およびRXFP2の間の選択性は、試験したすべてのFc−リラキシン−2融合ポリペプチドについて保持される。

- A−L−BまたはB−L−Aを含む融合ポリペプチドであって、
Aは、リラキシンA鎖ポリペプチドまたはその変異体であり;
Bは、リラキシンB鎖ポリペプチドまたはその変異体であり;かつ
Lは、少なくとも15のアミノ酸を含むリンカーポリペプチドであり;
前記融合ポリペプチドは、リラキシン活性を有し、かつ半減期を延長する成分をさらに含む、融合ポリペプチド。 - 前記リラキシンA鎖は、リラキシン2 A鎖であり、前記リラキシンB鎖は、リラキシン2 B鎖である、請求項1に記載の融合ポリペプチド。
- 前記リンカーポリペプチドLは、長さが15〜20アミノ酸である、請求項1または2に記載の融合ポリペプチド。
- 前記リンカーポリペプチドLは、配列GGGGS(配列番号58)の3つ以上のリピートを含む、請求項1〜3のいずれか一項に記載の融合ポリペプチド。
- 前記リンカーポリペプチドLは、配列GGGGSGGGGSGGGGS(配列番号57)を含む、請求項1〜4のいずれか一項に記載の融合ポリペプチド。
- 前記リンカーポリペプチドLは、配列GGGGSGGGGSGGGGS(配列番号57)からなる、請求項1〜5のいずれか一項に記載の融合ポリペプチド。
- 前記半減期を延長する成分は、免疫グロブリンFc領域である、請求項1〜6のいずれか一項に記載の融合ポリペプチド。
- 前記Fc領域は、ヒトIgG1免疫グロブリンに由来する、請求項7に記載の融合ポリペプチド。
- 前記Fc領域は、アミノ酸修飾の以下の組み合わせ:
(i) Fc−YTE(M252Y,S254T,T256E);
(ii) Fc−FQQ(L234F,L235Q,K322Q);
(iii) Fc−TM(L234F,L235E,P331S);
(iv) Fc−YTE−FQQ(M252Y,S254T,T256E,L234F,L235Q,K322Q);
(v) Fc−YTE−TM((M252Y,S254T,T256E,L234F,L235E,P331S),
(vi) Fc−TM−ΔTHT(L234F,L235E,P331S,D221G,K222G,T223G,H224S,T225A),
(vii) Fc−TM−ΔTHTΔK(L234F,L235E,P331S,D221G,K222G,T223G,H224S,T225A,ΔK447),
の少なくとも1つを含むアミノ酸配列を有し、
アミノ酸のナンバリングは、KabatのEUインデックスによるものである、請求項7または8に記載の融合ポリペプチド。 - 前記半減期を延長する成分は、融合ポリペプチドのN−末端に付けられる、請求項1〜9のいずれか一項に記載の融合ポリペプチド。
- 前記半減期を延長する成分は、コネクターポリペプチドを介して前記融合ポリペプチドに付けられる、請求項1〜10のいずれか一項に記載の融合ポリペプチド。
- 前記コネクターポリペプチドは、アミノ酸配列GGSP(配列番号56)またはGGGGSGGGGSGGGGS(配列番号57)を含む、請求項11に記載の融合ポリペプチド。
- 前記コネクターポリペプチドは、アミノ酸配列GGGGSGGGGSGGGGSGGGGGS(配列番号69)、AAAGGGGSGGGGSGGGGSGGGGSA(配列番号70)を含む、請求項11に記載の融合ポリペプチド。
- リラキシン活性を有する融合ポリペプチドであって、前記融合ポリペプチドは、配列番号2、4、6、8、10、12、14、16、18、20、22、24、26、および28のいずれか1つにおいて示される配列を有する、融合ポリペプチド。
- 基準となるリラキシンタンパク質のリラキシン活性に対する前記融合ポリペプチドのリラキシン活性の比は、約10−5および約105の間である、請求項1〜14のいずれか一項に記載の融合ポリペプチド。
- 請求項1〜15のいずれか一項に記載の融合ポリペプチドをコードする核酸分子。
- 請求項16に記載の核酸分子を含むベクター。
- 請求項17に記載のベクターまたは請求項16に記載の核酸分子を含む宿主細胞。
- 請求項1〜15のいずれか一項に記載の融合ポリペプチドを産生するための方法であって、請求項18に記載の宿主細胞を培養するステップおよび前記融合ポリペプチドを収集するステップを含む方法。
- 請求項1〜15のいずれか一項に記載の融合ポリペプチドおよび薬学的に許容される賦形剤を含む医薬組成物。
- 療法における使用のための請求項1〜15のいずれか一項に記載の融合ポリペプチドまたは請求項20に記載の医薬組成物。
- 前記融合ポリペプチドまたは医薬組成物は、前記対象に投与される、心不全を有する対象の治療における使用のための、請求項1〜15のいずれか一項に記載の融合ポリペプチドまたは請求項20に記載の医薬組成物。
