Abstract
Description
(i)変異型BAP1遺伝子もしくは変異型BAP1タンパク質の存在、
(ii)DRL誘導性の細胞死に耐性がある、BAP1野生型細胞である参照細胞における発現レベルもしくはタンパク質濃度と比較した、野生型BAP1遺伝子の発現レベルの減少、もしくは野生型BAP1タンパク質濃度の減少、または
(iii)DRL誘導性の細胞死に耐性がある、BAP1野生型細胞である参照細胞における結合レベルと比較した、野生型BAP1タンパク質に対するASXLタンパク質の結合の減少または非結合、を検出することを含み、
試料中の、変異型BAP1遺伝子もしくは変異型BAP1タンパク質の、または野生型BAP1遺伝子の発現レベルの減少もしくはタンパク質濃度の減少の存在、あるいはASXLタンパク質の野生型BAP1タンパク質への結合の減少もしくは非結合が、個体の癌細胞がDRL誘導性の細胞死に対して感受性があることを示している、方法を提供する。

本発明者らは、BAP1腫瘍抑制遺伝子の変異が、ヒト癌におけるアポトーシス経路の治療的調節に対する感受性を付与することを発見した。本発明者らは、悪性胸膜中皮腫、膀胱癌、および乳癌におけるこの関連性を調査および検証し、腎細胞癌、子宮頸癌、およびブドウ膜黒色腫を含む、1〜36%のヒト癌にも及び得るという証拠を有している。本明細書に記載されるデータは、rTRAIL、TRAIL受容体1(TRAIL−R1、細胞死受容体4としても知られる;DR4)およびTRAIL−R2(DR5としても知られる)に結合することによってTRAIL経路を活性化する組換えタンパク質に焦点を合わせているが、BAP1は、FASリガンド経路もしくはTNF経路または内因性アポトーシス経路などの他のプロアポトーシス経路を調節することも見出されている(実施例6を参照のこと)。
全エクソーム配列判定
製造業者の指示(QIAGEN)に従って、カラム抽出技術を使用して、DNAを抽出した。製造業者の指示に従って、イルミナのペアエンド試料調製キット(Illumina paired end sample prep kit)を使用して、ゲノムライブラリーを調製した。製造業者の推奨プロトコルに従って、Agilent SureSelect Human All Exon 50Mbキットを使用して、エクソーム濃縮を行った。各エクソームを、Illumina HiSeq 2000 DNAアナライザーで75−bpペアエンドプロトコルを使用して配列判定し、エクソーム当たり約5〜10Gbの配列を生成した。デフォルト設定でBurrows−Wheelerアライナ(BWA)アルゴリズムを使用して、配列判定リードをヒトゲノム整列させた(NCBI build GrCh 37)(17)。マッピングされていないリードおよびPCR重複は、分析から除外された。20倍以上の細胞株エクソームの平均カバレッジは、80%であった。
上記のようにDNAを抽出した。SNP6.0アレイについては、DNAをAROSにアウトソーシングした(http://arosab/services/microarrays/genotyping/)。コピー数注釈を、PICNICアルゴリズムから導いた[18]。
単一ヌクレオチド置換を呼び出すために、CaVEManアルゴリズムを使用した[19]。このアルゴリズムは、単純ベイズ分類器(naive Bayesian classifier)を使用して、各塩基における可能性のある各遺伝子型(野生型、生殖系列、または体細胞変異)の事後確率(posterior probability)を推定する。挿入と欠失を呼び出すために、分割リードマッピング(split read mapping)を、Pindelアルゴリズムの修正として実装した[19]。Pindelは、ゲノム上に固定された一方のリードを、2つの部分にマッピングされた他方のリードにより検索し、推定の挿入/欠失に及ぶ。両方のアルゴリズムについて、利用可能な適合正常組織なしで研究された全ての細胞株において使用されている腫瘍のCGPパネルからの同一の推定正常を正規化した。可能な限り多くの生殖細胞系一塩基多型を排除するために、その後、正常の様々なパネルに対する有意な後処理フィルタリングを行った。これらには、1000ゲノムデータベース、DB SNP、およびCGP正常の内部パネルが含まれる。これらのステップに続いて、変異体の潜在的な機能的影響に関して、FATHMアルゴリズム(Cancer Genome Project)を使用して、ミスセンス変異に注釈を付けた。
手動の「単回投与」組み合わせスクリーニングを、96ウェルフォーマットを使用して行った。細胞を1日目に180μl培地中に予め最適化された播種密度で蒔いた。2日目に、薬物のストックから20μlの10倍の濃度の培地を加えた。次に、細胞を72時間または6日間増殖させ、アッセイ終了時に固定した。薬物ウェルを、DMSO処理済みの対照ウェルと比較した。

293T細胞を、10%ウシ胎児血清(FBS)、非必須アミノ酸、50U/mLペニシリン、50μg/mLストレプトマイシン、および1%ピルビン酸ナトリウムを添加した、ダルベッコ改変イーグル培地(DMEM)中で培養した。ヒト中皮腫細胞株を、10%FBS、1%ペニシリン−ストレプトマイシン、および1%ピルビン酸ナトリウムを添加した、RPMI−1640培地(H2369、H2373、H2461、H2591、H2595、H2722、H2731、H2795、H2803、H2804、H2869、H290、H513、IST−MES1、MPP−89、MSTO−211H、NCI−H2052、NCI−H2452、NCI−H226、NCI−H28)、または10%FBS、非必須アミノ酸、50U/mLペニシリン、50μg/mLストレプトマイシン、および1%ピルビン酸ナトリウムを添加した、ダルベッコの改変イーグル培地/栄養混合物F−12(DMEM/F−12)(H2818、H2810)中で培養した。細胞を、5%CO2で37℃に維持した。
細胞単層を、リン酸緩衝食塩水(PBS)中で洗浄し、氷上で放射性免疫沈降アッセイ(RIPA)緩衝液(Sigma−Aldrich)およびプロテアーゼ阻害剤(Complete−min;Roche)中に溶解させた。溶解物を、14000rpmで10分間遠心分離し、上清を吸引した。タンパク質濃度を、製造業者の指示に従って、BCAアッセイ(Calbiotech)を使用して、ウシ血清アルブミンの標準曲線から計算した。溶解物を、適切な濃度に調製し、4倍のLaemelli緩衝液および10倍の還元剤を添加した後、試料を70℃で10分間加熱した。溶解物を、プレキャスト4〜12%Bis−Trisゲル(Invitrogen)上で、200Vで1時間のSDS−PAGEに供した。製造業者の指示に従って、iBlotゲル転写装置(Invitrogen)を使用して、タンパク質をニトロセルロース膜に転写した。4℃で一晩、一次抗体を添加する前に(別段明記しない限り、TBS−T中に1:1000で)、膜をTween 20(TBS−T)を含むトリス緩衝食塩水中の5%のミルク(TBS−T)中でブロックした。翌日、膜を、TBS−Tで3回洗浄し、二次抗体を添加した(TBS−T中に1:2500で)。使用された抗体としては、BAP1(C−4;Santa Cruz sc−28383)、アルファチューブリン(細胞シグナル伝達#2125)、c−IAP1(細胞シグナル伝達#7065)、cIAP2(細胞シグナル伝達#3130)、リビン(Livin)(細胞シグナル伝達#5471)、サバイビン(Survivin)(細胞シグナル伝達#2803)、Alexa Fluor(登録商標)488(Invitrogen A−21202)がある。イムノブロットを、ImageQuant(商標)LAS 4000バイオモレキュラーイメージャー(biomolecular imager)を使用して画像化した。
接着性細胞株を、薬物投与の24時間前に播種した。細胞をトリプシン処理し、播種前にウェルのサイズ(96または384)およびアッセイ期間に最適な密度で計数した。薬物処理してから72時間後に、細胞を4%パラホルムアルデヒドで30分間固定した。dH2Oで2回洗浄した後、100μlのSyto60核酸染色剤(Invitrogen)を、1μMの最終濃度で添加し、プレートを室温で1時間固定した。630/695nMの励起/発光波長を使用して、蛍光シグナルの定量化を行った。
付着性中皮腫細胞株を、1ウェル当たり約10000個の細胞で96ウェルプレートに蒔いた。細胞を蒔いて、薬物を添加した時点で1日間付着させた。48時間後、浮遊細胞を含む培地を、各ウェルから集めた。残りの接着細胞をPBSで洗浄し、EDTA中0.05%トリプシンで動かした(mobilize)。全ての細胞を、以前に除去された培地を含むチューブに集め、遠心分離(300g、5分間)によりペレット化した。次に、細胞を、10μl/1ml濃度のアネキシンV−647抗体(Invitrogen)を含む1XアネキシンV結合緩衝液に再懸濁し、室温で15分間インキュベートした。次に、以下のようにフローサイトメトリー分析の前に、DAPI(2μg/ml)を各試料に添加した。アネキシンV−/DAPI細胞は、生存可能であると判断され、アネキシンV+/DAPI細胞は、アポトーシスを受けている(初期アポトーシス期)と考えられ、アネキシンV+/DAPI+細胞は、後期アポトーシスまたは壊死と考えられ、死滅と記録された。
細胞を、リン酸緩衝食塩水で洗浄し、4%パラホルムアルデヒド中、室温で20分間インキュベートすることにより固定した。細胞内BAP1染色のために、固定された細胞を、氷上で20分間PBS中0.1%トリトンX−100で透過処理し、PBSで2回洗浄し、氷上で20分間一次抗体(C−4;Santa Cruz sc−28383、1:100)と共にインキュベートし、再度2回洗浄し、蛍光二次抗体(Alexa Fluor(登録商標)488、1:200(Invitrogen A−21202))と共にインキュベートした。細胞をPBSで2回洗浄し、フローサイトメトリー分析のためにPBSに懸濁した。細胞死アッセイの一部として分析された細胞を、上記のように調製した。
触媒的に不活性なBAP1発現H226細胞(H226 C91A)およびWT BAP1発現H226細胞(H226 BAP1)からmRNAを抽出し、Illumina HT12アレイに流した(run)。
mRNAマイクロアレイから同定された有意に差次的に発現された遺伝子を、KEGG経路分析を使用して分析した。

レンチウイルスの生成および濃度
トランスフェクション試薬としてjetPEI(Polyplus Transfection)を使用して、パッケージングプラスミドpCMVdR8.2およびエンベローププラスミドpMDG.2と共に、293T細胞をレンチウイルスベクタープラスミドでトランスフェクションすることによって、レンチウイルスを生成した。293T細胞を、37℃でインキュベートし、レンチウイルスを含有する培地を、24時間および48時間で回収した。レンチウイルスを、4℃で2時間18000rpmで超遠心分離(SW28ローター、Optima LE80K Ultracentrifuge、Beckman)によって濃縮し、使用前は−80℃で保存した。
上記のレンチウイルス生成プロトコルに従って、BAP1を標的とするshRNAをコート゛するレンチウイルスを生成した。MPM細胞(H2818)を形質導入し、純粋な集団が達成されるまでピューロマイシン200μg/mLで処理した。shRNAノックダウンの有効性を評価するために、イムノブロッティングを行った。
全ての動物実験は、University College Londonの生物学的サービス倫理審査委員会によって承認され、英国内務省の規則および1986年の動物操作の証拠(科学的手続き)法(英国、ロンドンの本社)の下で認可された。マウスは、Charles Riverから購入し、特定の病原体のない条件下で個別に換気されたケージで飼育され、滅菌照射食品およびオートクレーブ処理された水を自由に摂取できるようにした。
8週齢のNOD.CB17−Prkdcscid/NcrCrl(NOD SCID)マウス(Charles River)の群の各脇腹に、マトリゲルおよび培地の1:1の混合物中の100万個の細胞のルシフェラーゼ形質導入中皮腫細胞株(H226 BAPおよびH226 C91A)を注射した。腫瘍細胞の注射後14日目に、生物発光画像化(IVIS)によって評価した場合に腫瘍が構築されたとき、ビヒクルまたはイソロイシンジッパーTRAIL(izTRAIL)のいずれかを用いて、処置を開始した[20]。実験期間中、ビヒクルまたはizTRAILのいずれかを、1日1回600mcgの用量で腹腔内投与した。腫瘍サイズを、生物発光画像化(IVIS)を使用して、0、13、19、26、および41日目に評価した。マウスを42日目に処分し、腫瘍を除去して秤量した。1群当たり6匹のマウスを処置した。研究者らは、これらの実験で予備知識を持っていなかった。
GraphPad Prism V.4(GraphPad Software)を使用して、統計分析を行った。反復測定ANOVAを使用して、複数の群を用いたインビボ実験を分析し、単一群データを、Student t検定を使用して評価した。別段明記しない限り、全てのインビトロ実験を、三連で行った。
本発明者らは、単独で、またはリガンド腫瘍壊死因子(TNF)関連の細胞死誘導リガンド(TRAIL)と組み合わせてのいずれかで、94個の小分子阻害剤および化学療法剤(表1を参照のこと)を使用して、15種の中皮腫細胞株(Met5a中皮正常対照株と共に)でコンビナトリアルケミカルスクリーニングを実施した。極端な薬物感受性の例を検出するために、本発明者らは、中皮腫における候補癌遺伝子であると最近同定された8個の遺伝子のセットに基づいて、反応とこれらの細胞株の変異状態との間の統計的関連について分析した(図1Aを参照のこと)。注目すべきことに、BAP1変異は、癌の種類わたってよく認識されている(図1BおよびCを参照のこと)。薬物反応に対する変異の最大の効果は、脱ユビキチン化酵素BAP1に変異を保有し、TRAILで処理された中皮腫細胞のそれであった(図2Aを参照のこと)。スクリーンに含まれる対照正常中皮細胞株MET−5Aにおいて観察される細胞生存率では有意な効果はなかった(図2Bを参照のこと)。BAP1変異細胞は、それらの野生型対応物よりもTRAILに対して有意に感受性が高かった(図2Bおよび図2Cを参照のこと)。さらに、これらの細胞株で検出されたBAP1変異は、切断型であると予測されるだろう(表2を参照のこと)。本発明者らは、BAP1変異が、通常、タンパク質発現の喪失と関連しており、変異細胞株は、一般に、TRAILに対して感受性があることをイムノブロット法によって確認した(図3Aを参照のこと)。
TRAILは、2つの活性な膜貫通型細胞死受容体、DR4およびDR5を介して結合し、カスパーゼカスケードおよびその後の細胞死を引き起こす。BAP1変異細胞において観察されたTRAILの生存率効果は、アポトーシスマーカーアネキシンVで染色された細胞の画分の増加と実際に関連していた(図3Bを参照のこと)。
H226中皮腫細胞株は、BAP1のホモ接合性欠失を有し、その結果、BAP1発現の完全な喪失をもたらす。本発明者らはさらに、以前に記載されたように(PMID 18990722)、示差的mRNA遺伝子発現に関するこの触媒的に不活性なBAP1の効果を調べ、さらにシグナル伝達経路影響分析(signalling pathway impact analysis)(SPIA)を実施した。野生型とc91a mt BAP1とを比較した場合に有意に改変されたこれらの経路の中には、細胞死経路の経路があった(図6Aを参照のこと)(http://www.genome.jp/dbget−bin/www_bget?path:map04210)。特に、触媒活性を示さない(catalytically dead)C91A変異体で安定的に形質導入されたH226細胞において、IAPファミリーのメンバーの発現が減少した(図6Bを参照のこと)。最大の効果がCIAP2で見られ、これはウエスタンブロットによって確認された(図6Cを参照のこと)。
rTRAILを、上記に記載される94個の単剤化合物のライブラリーと組み合わせて、アンカー薬として使用した。デルタAUC測定基準(Wesslesらを参照のこと)を使用して、相乗効果を記載し、これは、以前に記載されるゲノムサブ群と相関した。本発明者らは、SMAC模倣物LCL161、DNAヘリカーゼ阻害剤YK−4279、およびチロシンキナーゼ阻害剤ソラフェニブなどの薬物が、別様に耐性細胞におけるDRL誘導アポトーシスの有効性を増大させることを示した。このスクリーニングの最も相乗的な知見の1つは、BAP1野生型MPMにおけるSMAC模倣物LCL161およびrTRAILに対する感受性の関連性であった(図7Aを参照のこと)。これは、TRAIL耐性野生型BAP1発現細胞をLCL161とTRAILとの組み合わせで処理することによって検証された。TRAIL単独に耐性があることが以前に実証された野生型BAP1を安定的に発現するH226細胞株においても、さらなる検証を行った(図5および7B〜Dを参照のこと)。IAP阻害剤、LCL161、およびTRAILの組み合わせは、変異型および野生型の両方の株において細胞死の相乗的な増加を示し、これは、BAP1変異体および野生型細胞におけるDRL誘導性の細胞死が、他の薬剤との組み合わせによって向上され得ることを示す(図7B〜Dを参照のこと)。特に図7Dは、BAP1変異体および野生型細胞の両方が、TRAILとLCL161との組み合わせによる処理に反応して細胞死することを示す。内因性SMAC/Diabloは、IAPの特異的な天然の阻害剤である(14)。このデータは、BAP1コンピテント状態において、IAPに対するこの阻害効果を特異的に模倣することによってBAP1喪失を模写することができ、その結果、IAPの正味の不活性化およびrTRAILに対する感受性がもたらされることを示唆する。これは、見られたBAP1/外因性アポトーシス経路の混乱が、IAPの正味の活性の特異的な調整不全に関連しているという考えをさらに支持するであろう。
脱ユビキチン化酵素BAP1は、前述のような、胸膜中皮腫(36%)、ブドウ膜黒色腫(47%)、および肝内胆管癌(25%)で頻繁に変異している。この治療アプローチにも適している可能性がある追加のBAP1変異型腫瘍が発生するかどうかを判定するために、本発明者らは、The Cancer Genome Atlas(TCGA)(http://cancergenome.nih.gov/)の変異データを使用して、20種の癌における5180個の腫瘍試料のコホートにまでこの分析を拡張した。切断型BAP1変異は、最大6%の頻度で、多様な範囲の癌の種類においても観察された(図1bおよびcを参照のこと)Carbone、M.ら、Nature Reviews Cancer 13、153−159(2013年3月)。本発明者らが、以前に全エクソームおよびコピー数分析のために提出された1001個の癌細胞株のパネルにそれらの分析を拡張したとき、彼らは、BAP1における切断変異を有する17個の細胞株を同定した(http://cancer.sanger.ac.uk/cancergenome/projects/cell_lines/)。これらには、明細胞腎臓癌、膀胱癌、および乳癌細胞株が含まれた。TRAILでこれらの細胞株を処理すると、同じ癌の種類のBAP1野生型細胞株と比較して、顕著な生存率効果が得られた(図8Aを参照のこと)。本発明者らはまた、乳癌細胞株MDA−MB231中のレンチウイルスshRNAを使用して、BAP1を不活性化し、rTRAILに対する誇張されたアポトーシス反応を観察した(図8Bを参照のこと)。これは、TRAIL療法が(中皮腫に加えて)他の形態の癌において有効であり得ること、およびBAP1の阻害、またはBAP1がTRAIL耐性を誘導する経路を標的とすることによってTRAILに対して癌細胞を感受性をもつようにさせる(sensitise)ことが可能であることを示唆する。
本発明者らはまた、BAP1遺伝子のASXLタンパク質結合部位における変異が、BAP1誘導TRAIL耐性を損なうことを実証した(図11を参照のこと)。BAP1は、ASXL1、ASXL2、またはASXL3と複合体を形成することが示された。ASXLタンパク質に対する結合部位の変異は、BAP1−ASXL複合体の形成を阻害する。BAP1−ASXL複合体は、ヒストン2Aおよび他の基質を脱ユビキチン化することが示されており、BAP1およびASXL1、ASXL2、またはASXL3の両方が、この脱ユビキチン化機能に必要とされる。この複合体は、ポリコームレスプレッサー複合体(Polycomb Respressor Complex)および遺伝子転写の重要な調整因子である。本発明者らは、BAP1野生型およびASXL3変異体(切断型変異)細胞株H513がTRAILに感受性があることを示した。それゆえ、ASXL1、ASXL2、またはASXL3の機能喪失は、DRL誘導性の細胞死に対する細胞の感受性を増大させる。ASXL1、ASXL2、またはASXL3の変異はまた、DLRに対する感受性を予測するため、BAP1変異状態とは無関係に、細胞死のバイオマーカーとして使用することができる。
本願におけるデータは、DR4およびDR5受容体に結合することによってTRAIL経路を活性化する組換えタンパク質である、rTRAILに焦点を合わせているが、観察されたBAP1変異−感受性付与(mutation-sensitisation)は、FASリガンド経路およびTNFアルファ経路を含む、他の外因性アポトーシス経路に及ぶ(図10を参照のこと)。
本発明者らは、BAP1が、TRAIL、TNFアルファ(TNFα)、およびFASリガンド(FASL)などの、細胞死受容体リガンドによる細胞死受容体の活性化に反応して、細胞が細胞死を受けるかどうかの重要な調整因子であることを見出した。具体的には、野生型BAP1の非機能的発現または低発現は、細胞を細胞死受容体リガンド誘導性の細胞死に対して感受性にする。結果として、変異型BAP1遺伝子もしくは変異型BAP1タンパク質、または野生型BAP1タンパク質の発現が低い癌細胞が、DRL誘導性の細胞死に対する感受性のバイオマーカーとして使用され得ることが発見された。

●BAP1阻害剤および細胞死受容体リガンドを含む組成物、ならびに
●DRL誘導性の細胞死に対して感受性がない癌に罹患している個体を治療、予防、または改善する方法。

Claims (20)
- 個体の癌細胞が細胞死受容体リガンド(DRL)誘導性の細胞死に対して感受性があるかどうかを判定する方法であって、前記個体から採取された生物学的試料において、
(i)変異型BAP1遺伝子もしくは変異型BAP1タンパク質の存在;
(ii)DRL誘導性の細胞死に耐性がある、BAP1野生型細胞である参照細胞における発現レベルもしくはタンパク質濃度と比較した、野生型BAP1遺伝子の発現レベルの減少もしくは野生型BAP1タンパク質濃度の減少;または
(iii)DRL誘導性の細胞死に耐性がある、BAP1野生型細胞である参照細胞における結合レベルと比較した、野生型BAP1タンパク質に対するASXLタンパク質の結合の減少または非結合、
を検出することを含み、
前記試料中の、前記変異型BAP1遺伝子もしくは前記変異型BAP1タンパク質の、または前記野生型BAP1遺伝子の発現レベルの減少もしくはタンパク質濃度の減少の存在、あるいはASXLタンパク質の野生型BAP1タンパク質への結合の減少もしくは非結合が、前記個体の癌細胞がDRL誘導性の細胞死に対して感受性があることを示している、方法。 - 前記変異型BAP1遺伝子が、非機能的もしくは酵素的に不活性なBAP1タンパク質、またはDRL誘導性の細胞死に耐性がある、参照細胞における結合レベルと比較して、ASXLタンパク質への結合の減少を示すBAP1タンパク質をコードする遺伝子である、請求項1に記載の方法。
- 前記ASXLタンパク質が、ASXL1、ASXL2、またはASXL3である、請求項2に記載の方法。
- 変異型BAP1タンパク質が、非機能的であるまたは酵素的に不活性であるまたはASXLタンパク質に結合できないまたはDRL誘導性の細胞死に耐性がある、参照細胞における結合レベルと比較して、ASXLタンパク質への結合の減少を示す、請求項1に記載の方法。
- 前記発現レベルの減少が、前記参照細胞と比較して、少なくとも10%、15%、25%、35%、45%、50%、55%、60%、65%、70%、75%、80%、85%、90%、95%、または100%の減少である、請求項1〜4のいずれか一項に記載の方法。
- 前記野生型BAP1タンパク質の濃度の減少が、前記参照細胞と比較して、少なくとも10%、15%、25%、35%、45%、50%、55%、60%、65%、70%、75%、80%、85%、90%、95%、または100%の減少であり得る、請求項1〜5のいずれか一項に記載の方法。
- 前記変異型BAP1タンパク質のアミノ酸配列が、配列番号4またはその断片もしくは変異体によってコードされている、請求項1〜6のいずれか一項に記載の方法。
- 前記変異型BAP1遺伝子のヌクレオチド配列が、配列番号5またはその断片もしくは変異体によってコードされている、請求項1〜7のいずれか一項に記載の方法。
- 個体の癌細胞がDRL誘導性の細胞死に対して感受性があるかどうかを判定するためのキットであって、変異型BAP1遺伝子もしくは変異型BAP1タンパク質の発現を検出するための、またはDRL誘導性の細胞死に耐性がある参照細胞中の発現レベルもしくはタンパク質濃度と比較した、野生型BAP1遺伝子の発現レベルの減少もしくは野生型BAP1タンパク質濃度の減少を検出するための、またはDRL誘導性の細胞死に耐性があるBAP1野生型細胞である参照細胞における結合レベルと比較した、BAP1タンパク質へのASXLタンパク質の非結合もしくは結合の減少を検出するための、検出手段を備え、前記試料中の、前記変異型BAP1遺伝子もしくは前記BAP1タンパク質の、または前記野生型BAP1遺伝子の発現レベルの減少もしくは野生型BAP1タンパク質濃度の減少の存在、あるいは野生型BAP1タンパク質に対するASXLタンパク質の結合の減少もしくは非結合が、前記個体の癌細胞がDRL誘導性の細胞死に対して感受性があることを示している、キット。
- 前記キットが、少なくとも1つの対照または参照試料を含む、請求項9に記載のキット。
- 前記キットが、陰性対照および/または陽性対照を含む、請求項10に記載のキット。
- 前記陰性対照が、DRL誘導性の細胞死に耐性がある野生型BAP1タンパク質を含む、請求項11に記載のキット。
- 前記陽性対照が、変異型BAP1 mRNA、または変異型BAP1タンパク質、またはブランク試料を含む、請求項11に記載のキット。
- (i)変異型BAP1遺伝子もしくは変異型BAP1タンパク質、または(ii)BAP1野生型細胞である参照細胞における発現レベルもしくはタンパク質濃度と比較して、野生型BAP1遺伝子の発現レベルの減少もしくは野生型BAP1タンパク質濃度の減少を有する癌細胞の、DRL誘導性の細胞死に対して感受性があるバイオマーカーとしての、使用。
- (i)BAP1阻害剤またはBAP1阻害の効果を模倣する薬剤、および(ii)細胞死受容体リガンドを含む、組成物。
- 治療に使用するための、または医薬品として使用するための、(i)BAP1阻害剤もしくはBAP1阻害の効果を模倣する薬剤、および(ii)細胞死受容体リガンドを含む、組成物。
- 癌の治療、予防、または改善に使用するための、(i)BAP1阻害剤もしくはBAP1阻害の効果を模倣する薬剤、および(ii)細胞死受容体リガンドを含む、組成物。
- 請求項17に記載の使用のための、(i)BAP1阻害剤もしくはBAP1阻害の効果を模倣する薬剤、および(ii)細胞死受容体リガンドを含む、組成物であって、前記癌が、中皮腫、悪性胸膜中皮腫、ブドウ膜黒色腫、黒色腫、非黒色腫皮膚癌、腎臓癌、肺癌、胸膜癌、腹部癌、腹膜癌、心膜癌、頭頸部癌、脳腫瘍、乳癌、肝臓または胆道癌、上部管および下部管を含む消化器癌、尿路上皮癌、前立腺癌、精巣癌、精巣鞘膜癌、卵巣癌、子宮頸癌、肉腫、リンパ腫、または白血病である、組成物。
- 請求項17に記載の組成物と、薬学的に許容されるビヒクルと、を含む、薬学的組成物。
- 治療有効量のBAP1阻害剤またはBAP1阻害の効果を模倣する薬剤および細胞死受容体リガンド、ならびに薬学的に許容されるビヒクルを接触させることを含む、請求項19に記載の薬学的組成物を調製するための方法。
