Abstract
Description
The present invention relates to a sample storage device and a sample for which, for example, a sample that is a cell or a tissue thereof that is cultured for a long time in a culture device is temporarily stored in the vicinity of a safety cabinet that safely handles the sample taken out from the culture device. Regarding the storage system.
Conventionally, in the fields of pharmaceutical research and regenerative medicine, research is conducted by manipulating, processing, and culturing cells, and by developing it, sample culture is systematically performed, for example, commercialization of tissue regeneration from stem cells. In order to perform the above, using a safety cabinet installed in a laboratory (clean room) managed in a clean environment, medium exchange of sample cells for the above purpose, pipette stirring operation to cell colonies, etc. We are working on processing.
In the manufacture of regenerated tissue products and cell processing in regenerative medicine, while growing for a long time in a culture device such as a carbon dioxide gas incubator or a multi-gas incubator that constantly provides an environment suitable for growth of a sample that is a cell culture, as appropriate. Samples are taken out from the incubator to perform processing work and promote differentiation, and the cells are manipulated from the viewpoint of ensuring the safety of workers, preventing contamination from workers, preventing microbial contamination of cells, work efficiency, etc. Processing work such as is performed in a safety cabinet, which is a sterile operating device.
The safety cabinet is used for the purpose of primary isolation to protect workers from contaminated aerosols generated when handling pathogens, etc., and the inside that communicates the contaminated aerosols generated in the work space provided in the cabinet body to the work space. By drawing into the communication passage, the airflow direction inside the work space is secured, and the outside air (air in the clean room) flows in from the opening of the opening / closing door provided in front of the work space to the lower suction in front of the work space. It has a function of preventing the leakage of contaminated aerosol from the opening to the outside. In the safety cabinet, the contaminated aerosol drawn into the duct from the work space is sterilized and cleaned using, for example, a HEPA filter (High Effectivey Particulate Air Filter), and the sterilized and cleaned clean air is sterilized and cleaned outside the safety cabinet. In addition to the function of exhausting air to the air, it also has the function of circulating sterile and purified clean air inside the safety cabinet. When the safety cabinet has the above-mentioned function, it is possible to prevent not only the above-mentioned primary isolation but also the contamination in the clean room. In the cell culture operation, the primary isolation performance of this safety cabinet is utilized to perform aerosol isolation by aseptic cleaning by filtration with a HEPA filter between the operation environment in the work space and the external environment of the safety cabinet. It protects the cells to be cultured inside.
In performing the above-mentioned processing work, for example, between the work space of the biohazard countermeasure cabinet (corresponding to the safety cabinet) and the storage space provided in the culture device such as an incubator to store the container containing the sample. Being wary of microbial contamination during manual sample movement, the work space and storage space are communicated using a duct, and a container containing the cells to be cultured as a sample is used as an incubator by a transport device placed in the duct. It has been devised to deliver it to and from a biohazard countermeasure cabinet (see, for example, Patent Document 1).
In the sample storage system disclosed in Patent Document 1, the incubator is controlled in an environment suitable for cell growth (for example,
By the way, the work space of the safety cabinet (biohazard countermeasure cabinet) is in almost the same environment as the clean room, and gives a large temperature / humidity change when viewed from a sample taken out from a culture device such as an incubator, which is a long-term growth environment. Therefore, if the cells are placed in a temperature and humidity environment far from the culture environment for a long time, they are easily affected by heat shock stress, which is said to have a high possibility of affecting the quality of cells.
Further, in the arrangement configuration of each device in the clean room, the internal space of the incubator and the work space of the safety cabinet are often not communicated with each other through the duct due to the above-mentioned problem of maintaining temperature and humidity. Rather than slowly growing in an incubator for a long period of time, a polymerase chain reaction; when the gene DNA is amplified at once by using the synthase chain reaction method, equipment and equipment that are relatively positive pressure to increase reliability regarding foreign substance invasion. A pass box for delivery means between the material storage area (room) and the test DNA extraction area (there is a cabinet in the room) where the safety cabinet is installed is installed side by side with the safety cabinet, for example, inside the pass box. The space and the work space of the safety cabinet (corresponding to the biohazard countermeasure cabinet described in Patent Document 1) are communicated with each other through a duct for taking in and out of a sample, and the pass box and the safety cabinet are provided with external air having the same temperature and humidity. Is devised to be supplied via a HEPA filter (see, for example, Patent Document 2).
In the cleaning work apparatus disclosed in Patent Document 2, the work space of the pass box and the clean bench is passed by passing it through a HEPA filter immediately before the fresh air supplied from the outside is sent to the work space of the pass box and the clean bench. Purifying. However, in the case of the cleaning work apparatus disclosed in Patent Document 2, for example, the temperature of the internal space of the pass box or the working space of the clean bench is an external temperature (for example, the temperature in the clean room), and the sample held in these spaces. The cells are susceptible to the heat shock stress described above. In particular, the above-mentioned processing work is a work in which many samples are stored inside the pass box at one time, and the time from when the samples are stored inside the pass box until the processing work is completed and returned to the incubator. The longer the value, the greater the effect of heat shock stress, and there is a concern that the quality of the sample cells will deteriorate.
The present invention pays attention to a pass box for temporarily transferring a sample while preventing the internal space of the incubator and the work space of the cabinet from communicating with each other through a duct, and inside the pass box, for a work lot. A sample storage device that temporarily stores samples from multiple cultures so that the effects of heat shock stress on cells that are temporarily stored inside the pass box can be reduced. And to provide a sample storage system.
In order to solve the above-mentioned problems, the sample storage device of the present invention has at least a mounting table on which a culture container in which cells are cultured is placed, and a storage space provided above the mounting table. A first duct arranged between the sample storage device and sending air from the air conditioner to the sample storage device, and arranged between the return air side of the air conditioner and the sample storage device, said. Further, the sample storage device has a duct connection portion to which the first duct is connected, and the duct connection portion sucks air sent from the air conditioner and sends it toward the storage space. , The heating means and at least one of the humidifying means are arranged.

According to the present invention, it is possible to reduce the influence of heat shock stress on cells that are samples temporarily stored inside the pass box.
Hereinafter, this embodiment will be described with reference to the drawings.
As shown in FIG. 1, the
In the
The
The
The
Next, the configuration of the
As shown in FIGS. 2 and 3, the
The
Further, a
Here, the
The
The

In the above-mentioned
The air sucked into the
As shown in FIGS. 2 to 4, the above-mentioned
Although the case where the opening / closing door for opening / closing the
Further, in the present embodiment, when the
Next, the configurations of the
As shown in FIG. 5, the
At the front end of the mounting table 59, a
Here, between the mounting table 59 and the
Further, the
The
The
The
The
The front surface of the
An
An opening / closing
Here, as shown in FIG. 4, in the case of the
Further, an opening / closing
As shown in FIG. 6, the
The
The
The
The
It is not necessary to provide the
Hereinafter, the control in the
When the
As described above, the
The air sent into the
The air inside the
As described above, the temperature of the air flowing through the
In the present embodiment, the air from the
Further, the
In the present embodiment, the case where the
In this case, the management device that manages the temperature and humidity of the
In the present embodiment, the temperature and humidity of the
