従属栄養菌株および培養の樹立
この実施例は、栄養成長に好ましい条件により培養される新規従属栄養細胞型および本発明の方法を使用して生成物を作製することを目的とする。ヘマトコッカス(Haematococcus)の細胞は、環境試料として単離するかまたはテキサス大学(Austin, Tex., USA)のCulture Collection of Algaeから入手したUTEX2505のような菌株保存機関から得る。細胞は、光合成ヘマトコッカス・プルビアリス(Haematococcus pluvialis)菌株BMIのようなより高い塩耐性を有する単離物を含みうる(Chekanovら、2014年)。細胞は、商業規模の光合成生産培養からも得られうる。存在する場合には、シストまたは胞子の発芽は、分画され、細分化されて、緑色運動性細胞を含有する培養を生成する運動性細胞を生成するのに好ましい条件下で起こる。倒立顕微鏡の使用は、生活環ステージを同定するために有効である(栄養性、未成熟シスト、シスト)。初期の混入物負荷を低減させるために、培養を200gで1分間遠心分離して、藻類および混入物を含む重い細胞を選択的にペレット化する。遠心分離直後に、培地を除去し、滅菌培地で交換し、1000gで1分間遠心分離し、運動性細胞を遠心管上部の光源にむかって泳動させる。運動性細胞を収集し、新しい管に移し、上清に含まれる標的生物以外の生物がわずかになるまで遠心分離を反復する。次いで、この混入物が低減された培養を、さらに細菌量を低減させるために抗生物質(アンピシリン50mg/Lおよびセフォタキサミン250mg/L)で24〜48時間処理し、次いで、希釈し、FCMを使用して標的単一細胞を、200μLの当分野で公知の基本成長培地、例えば、同じ抗生物質を含むギリアードF/2培地(尿素で置換した硝酸塩でも改変した新鮮な水を)を含有する96ウェルプレートにソーティングする。
従属栄養成長のための培地組成および温度
成長の最大の増加をもたらす培地構成要素は、多様な種について同定されている。これは、最初は、開始基本培地を用いてフラスコレベルで実行し、固定炭素源として酢酸塩を使用し、それは、頻繁な(時間ごと)手動の酢酸添加が過度に労働集約的であり、24時間ひっきりなしのケアを必要とすると予想されるからである。H.プルビアリス(H. pluvialis)KAS1601を一例として使用して、窒素源としての尿素の使用は、KNO3に対して40%の成長増加をもたらし、28℃の温度は、25℃に対して57%の増加をもたらし、静止期が開始する前の同じ時点の後に、全体として2.2倍のOD750の増加をもたらした。特に、好ましい窒素源を、25℃の酢酸ナトリウム1.6g/L、酵母抽出物0.16g/L、窒素1.76mM(尿素0.88mMまたはKNO31.76mM)、硫酸マグネシウム七水和物0.05g/L、塩化カルシウム二水和物0.05g/L、リン酸カリウム0.02g/L、鉄−EDTA0.01g/L、塩化鉄六水和物0.0063g/L、四ナトリウムEDTA22mg/L、硫酸コバルト0.3mg/L、硫酸マンガン6mg/L、硫酸亜鉛0.8mg/L、硫酸銅0.2mg/L、モリブデン酸アンモニウム0.7mg/L、ホウ酸0.4mg/L、塩酸チアミン0.4mg/L、ビオチン2μg/L、およびビタミンB12、2μg/Lのベースライン培地からなる従属栄養フラスコ成長培地を使用して決定する。分光光度計を使用してOD750測定値をとる。2日目には、尿素とKNO3成長培地のOD750に差はないが、4日目までに尿素を使用する培養はKNO3培養に比べて40%高いOD750を有する。好ましい温度は、25℃、28℃および31℃で好ましい窒素源(尿素)を使用して決定する。25℃から直接的にすすめ、適合を含まないこれらの条件下では、運動性細胞は、31℃で増殖しない。成長をモニターするために、分光光度計を使用してOD750測定値をとる。3日目には、25℃と28℃の成長した培養に差はないが、5日目までに、28℃の培養は25℃の培養に比べて57%高いOD750を有する。他の菌株の従属栄養成長に適した培地組成の決定のための同様の方法を、当分野で公知のとおりに行う。クラミドモナス(Chlamydomonas)について、同じ温度を、酢酸ナトリウム1.6g/Lを酢酸ナトリウム0.4g/Lおよび上記のとおりに決定した好ましい窒素源(尿素)に交換してフラスコ中で試験する。他の培地構成要素が、当分野で公知のとおりに標準的な多変量成長研究を使用して、修飾されまたは除外されまたは添加されうる。これは、炭素源としての多様な有機酸の使用を含み、有機酸は、1つのタイプまたはリグノセルロース誘導体において起こりうるように組み合わされた1つ超のタイプでありうる。
従属栄養培地組成およびマクロ独立細胞による色素生成
この実施例は、従属栄養条件下での好ましい成長のために選択した菌株を採用する。シードトレインを記載するための例としてヘマトコッカス・プルビアリス(Haematococcus pluvialis)KAS1601を使用して、9個の500mLのH.プルビアリス(H. pluvialis)の栄養従属栄養培養を、25℃で1Lフラスコのバッチ培養中、早期成長期に0.15のOD750を有し、後期成長期には約0.6のOD750まで増加する細胞密度により測定して、うまく成長期に入ってから静止期に入る前までの十分な時間、100rpmで振とうした。フラスコ成長培地は、酵母抽出物0.16g/L、尿素0.11g/L、硫酸マグネシウム七水和物0.05g/L、塩化カルシウム二水和物0.05g/L、リン酸カリウム0.02g/L、鉄−EDTA0.01g/L、塩化鉄六水和物0.0063g/L、四ナトリウムEDTA22mg/L、硫酸コバルト0.3mg/L、硫酸マンガン6mg/L、硫酸亜鉛0.8mg/L、硫酸銅0.2mg/L、モリブデン酸アンモニウム0.7mg/L、ホウ酸0.4mg/L、塩酸チアミン0.4mg/L、ビオチン2μg/L、およびビタミンB12、2μg/Lからなる。加えて、フラスコ培地は、酢酸1.15mL/Lを含むTris塩基2.78g/Lを含有するかまたは低密度のフラスコ培養の手間のかからない成長を可能とするために、最初に酢酸ナトリウム1.6g/Lを含有してもよい。培養は、3000gで5分間の遠心分離により濃縮されるかまたはそのまま使用される。微細藻類細胞(図1〜10)を、0.2g/Lの最初のバイオマス密度で、14Lの発酵容器(New Brunswick BioFlo3000;図1〜30)中の3Lのファーメンター従属栄養成長培地(Tris塩基0.7g/Lおよび最初の酢酸0.29mL/Lまたは任意選択により1回の酢酸ナトリウム0.4g/Lとともに供給したフラスコ従属栄養培地と同じ)に移す。3L発酵培養を、3L/分の注入空気および100〜150rpmの傾斜ブレードインペラーによりpH7.8でもたらされるガス交換とともに28℃でインキュベートする。BioCommand Software、ペリスタポンプおよびヘッドプレートポートを使用して、5%のポンプスピードで供給する10%酢酸(図1、実施形態20)のpHトリガー添加を含むフェドバッチ発酵の持続時間の間中、pHをH7.8〜7.3に維持し、窒素およびリン(mMベースで9:1の割合)を発酵の間中、10%のポンプスピードで頻繁に(4〜2時間ごと)供給して、ファーメンター従属栄養成長培地の開始濃度の近くに窒素およびリンのレベルを保つ。(酵母抽出物を除く)培地からの残りの栄養素を、10%スピードのペリスタポンプを使用して発酵の24時間ごとに1回、供給する。容器への注入空気の量を4L/分最大、撹拌を300rpmまで増加させることによって、溶解酸素を30%超に維持する。10L体積の非トリガー細胞からの細胞を、90L体積(Eppendorf BioFlo 610ファーメンター中の培養10L+ファーメンター従属栄養培地80L)に直接播種するために使用可能である。90Lの培養に、注入空気を50〜100LPMおよび350rpmまでの傾斜ブレードによる撹拌で上記のように供給する。この方法により、結果として生じるバイオマス(最初の0.2g/Lから6g/L)を、遅滞時間を含まず、0.7/日の高い比成長の96時間の延長された対数期を含む、120時間にわたって生産する。これらの構成要素の使用は、急速に成長する細胞の生理学により現行の実践よりも有利であり、窒素代謝からの培養pHシフトは有機酸の添加、炭素源としての倍加、により対処して、余分の塩を培地に添加することなく培養pHの釣り合いをとる。乾燥重量分析のために24時間ごとに試料(10mL)を収集して、比成長速度および藻類生成物の色素含有量を決定する(図1、実施形態40)。10mLの試料を直ちに3000gで5分間、遠心分離し、上清を除去し、細胞ペレットを−80℃で凍結し凍結乾燥して、乾燥重量を決定する。バイオマス1mgあたり50μLのアセトンを用いて粉砕した凍結乾燥バイオマスから、室温で5分間、色素を抽出する。分光光度計を使用して(BioRad Smart Spec)、清澄化した抽出物の吸収を476nmで読み取る。以下の式[A476/217]×[抽出体積(mL)]×[希釈因子]を使用して総色素(mg)を計算し、式中、217は、アセトン中のアスタキサンチンの吸光計数である。色素パーセントを、抽出中の1mgのバイオマス当量あたりの色素のmgから計算する。96時間で、バイオマスは赤色マクロ独立細胞中1.5%の色素含有量を有する2.1g/Lに達し(図2−右上);体積ベースで、これは31.5mg/Lに等しい。これは、Hataら(2001年)における、時間あたり0.063mg/Lに比べて顕著に高い速度である時間あたり0.33mg/Lの生成物形成(qp)に相当する。これは、Hataら(2001年)における、0.21/日(0.009/時間)に比べて、24時間において検出可能な遅滞期を含まない72時間にわたり、0.68/日(0.028/時間)の比成長速度に相当する。特に、細胞拡大よりも細胞***による、延長された持続時間にわたる高い比成長速度の従属栄養生産システムに関する顕著な改善を図4に示す。同様の比成長速度およびより高い内在性色素含有量を有する細胞株は、2.3%の色素含有量が、時間あたり0.5mg/Lの速度をもたらすように、本発明の方法によりより高いqp値をもたらす。生産性は、細胞型選択または適合、共培養、ファーメンター播種および操作、ならびに例示されたような他の手段により、さらに増加可能である。
改善された従属栄養培地組成およびクラミドモナス(Chlamydomonas)細胞によるバイオマス生産
2Lフラスコ中の1Lの培養物6個を、フラスコ従属栄養培地中で実施例3に記載のとおりに成長させた。実施例3のとおりに培養を濃縮し、14L発酵容器(Eppendorf−New Brunswick BioFlo 3000)中の(フラスコ従属栄養培地と同じであるが、最初に0.25g/Lだけ酢酸ナトリウムを供給する)3Lのファーメンター従属栄養成長培地中に0.05g/Lの最初のバイオマス密度で合わせた。3Lの発酵培養を、5L/分の注入空気および100〜250rpmの傾斜ブレードインペラーによりpH7.8でもたらされるガス交換とともに28℃でインキュベートする。発酵培養を維持し、実施例3のとおりに栄養素および固定炭素源としての有機酸をフェドバッチ様式で供給する。当分野で周知の方法を使用して、成長曲線を作成し、栄養培地中の尿素、リン酸塩およびアンモニウムのレベルを測定するために、試料(10mL)を24時間ごとに収集する。コナミドリムシ(C. reinhardtii)KAS1001を使用して、8.25g/Lのバイオマスに到達する、72時間にわたる1.7/日(0.07/時間)の比成長速度を達成し;これは、成長停止の前にわずか約1.4g/Lに到達するバイオマス収率を伴う40時間のみの持続時間についての先の最高速度1.7/日(0.07/時間)(Zhangら、1999年)に比べて、ほとんど2倍の発酵持続時間を伴う顕著に高い収率および延長した加速成長である。この約1.4g/Lのバイオマスは、塩毒性による成長阻害、よって比成長速度における短い持続時間と低い細胞密度のみをもたらす、少なくとも2.8g/Lまたは34mMの酢酸ナトリウムを必要とする。実施例3に記載のとおりに100Lのバイオリアクターに移したクラミドモナス(Chlamydomonas)培養は、4日間にわたって少なくとも1.0/日の比成長速度を維持する(生産サイクル:120時間)。0.2g/Lの最初の細胞密度は、溶解酸素を必要に応じて増加させるために加えた5psiの容器圧力とともに実施例3においてH.プルビアリス(H. pluvialis)について記載した条件下で成長させた場合、20〜30g/Lを超える最終細胞密度を生み出し、成長が阻害されない、高い速度の活性な細胞***の延長された期間を実証している。当分野で公知のとおりの温度および培地構成要素の調整とともに、同様のプロセスをクラミドカプサ(Chlamydocapsa)およびクロロモナス(Chloromonas)に適用する。
セネデスムス・オブリカス(Scenedesmus obliquus)を使用する、H.プルビアリス(H. pluvialis)の共培養におけるアンモニウム制御
2.5L体積のH.プルビアリス(H. pluvialis)KAS1601を実施例3のとおりに従属栄養フラスコ培地中で成長させた。1リッターのS.オブリカス(S. obliquus)KAS1003を非有機酸炭素源(グルコース3.6g/L)および代替窒素源(塩化アンモニウム0.09g/L)とともに、実施例3に記載のとおりに従属栄養フラスコ培地中で成長させた。実施例3に記載のとおりに培養を遠心分離し、3.6g/Lのグルコースを補充した実施例3に記載の従属栄養発酵培地中、3L体積のH.プルビアリス(H. pluvialis)とS.オブリカス(S. obliquus)の共培養に濃縮した。或いは、S.オブリカス(S. obliquus)を、当分野で公知のとおりにアルギン酸または多孔質ビーズ中に包埋し、ろ過、磁力または他の手段を使用するエンドポイント除去を可能とした。この3Lの発酵培養を実施例3に記載のとおりに維持した。試料を24時間間隔で採取して、バイオマス乾燥重量を得て、アンモニウム濃度を測定した。96時間で、アンモニウム濃度は、わずか1.4mMアンモニウムに到達したにもかかわらず、実施例3からのH.プルビアリス(H. pluvialis)の3L発酵培養中では45mMのアンモニウムを観察した。この3L発酵は、0.77/日(0.032/時間)の比成長速度をもたらす。最終バイオマスは、混合微生物を含む開放の池で自然に発生しうるものと同等かまたは優れる、約99%以上のH.プルビアリス(H. pluvialis)バイオマスからなる。細胞型または固定炭素源としてのグルコースの投与のような共培養パラメータの調整は、約2.5mMアンモニウムのカロテン生成トリガーに対して異なる標的成長速度および生産性に到達することを可能とする。適切なアンモニウム制御を伴う0.2g/LのH.プルビアリス(H. pluvialis)バイオマスで開始する発酵培養は、120時間で0.77/日の比成長速度および4.3g/Lのバイオマス収率を伴う少なくとも96時間の延長された対数期を経験する。適切なアンモニウム制御を伴う1g/LのH.プルビアリス(H. pluvialis)バイオマスで開始する発酵培養は、120時間で0.77/日の比成長速度および22g/Lのバイオマス収率を伴う96時間の延長された対数期を有する。最後の48時間は、アンモニウムストレスを伴い、その最初の24時間(72〜96時間)は、対数期としてバイオマスが同じ比成長速度でなお蓄積し、その2番目の24時間(96〜120時間)は、アスタキサンチンが運動性細胞の乾燥重量の1.5%まで蓄積した。この120時間の発酵ランは、時間あたり0.063mg/Lの顕著に低い収率を示したHataら、(2001年)に比べて、時間あたり5.5mg/Lのかなり優れた色素のqpをもたらした。バイオマスが蓄積し、色素生成中に細胞***が停止するために高い成長速度が短時間しか持続しない先行技術とは異なり、本発明の方法は、必要に応じて7日超を含む、何日にもわたり延長された高い比成長速度を可能とする。この実施例では、S.オブリカス(S. obliquus)が、多くのそのような細胞型について当分野で公知のとおり、それがなお有機酸でない固定炭素源を好み、窒素源として優先的にアンモニウムを消費する限り、従属栄養成長に適した異なる微生物細胞型で交換可能であると理解される。アスタキサンチンまたは他の色素生成細胞型または油生成細胞型の中の選択肢は、当分野で公知のとおり、セネデスムス(Scenedesmus)、クロレラ(Chlorella)、モノラフィディウム(Monoraphidium)、ロドトルラ(Rhodotorula)(赤色酵母)の他の種ならびにフェオダクチラム(Phaeodactylum)およびシクロテラ(Cyclotella)のような多くの様々な珪藻植物、ならびにスラウストキトリッド(thraustochytrids)およびスラウストキトリッド(thraustochytrid)様の細胞型である。共培養からのバイオマスは、特に、細胞の割合が好都合に操作される場合、1つの生物由来の価値ある生成物が第2の生物のそれを補完することに寄与できる。例えば、珪藻植物は、フコキサンチンを含有し、セネデスムス・オブリカス(Scenedesmus obliquus)は水溶性のカロテノタンパク質を含有し、スラウストキトリッド(thraustochytrids)は、DHA脂肪酸を含有する。
コナミドリムシ(Chlamydomonas reinhardtii)を使用するH.プルビアリス(H. pluvialis)の共培養におけるアンモニウム制御:およびカロテン生成に好都合な条件
3LのH.プルビアリス(H. pluvialis)KAS1601を実施例3のとおりに従属栄養フラスコ培地中で成長させた。1リッターのコナミドリムシ(C. reinhardtii)KAS1001を実施例3に記載のとおりに従属栄養フラスコ培地中で成長させた。培養を、5mMから20mMまでの間の酢酸ナトリウムとともに実施例3に記載の従属栄養フラスコ培地中のH.プルビアリス(H.pluvialis)とコナミドリムシ(C. reinhardtii)の共培養に混合した。3Lの発酵培養を実施例3に記載のとおりに維持した。試料を24時間間隔で採取して、バイオマス乾燥重量を得て、アンモニウム濃度を測定した。120時間の発酵の間、アンモニウム濃度は効果的に1mM未満に維持したが、実施例1におけるH.プルビアリス(H. pluvialis)の単独培養は、45mMアンモニウムに到達する。この3L発酵は、96時間にわたって0.90/日(0.038/時間)の比成長速度をもたらした。最終バイオマスは、混合微生物を含む開放の池で自然に発生しうるものと同様の約99%のH.プルビアリス(H. pluvialis)バイオマスからなるものであった。発酵システムに典型的な細胞型の投与のような共培養パラメータまたはrpmおよび酸素負荷のような操作パラメータの調整は、H.プルビアリス(H. pluvialis)のための約2.5mMアンモニウムのカロテン生成トリガーに対して異なる標的成長速度および生産性に到達することを可能とする。発明者らは、微細藻類培養物が時間あたり0.063mg/Lの先に報告された値をはるかに超える比生産性率でアスタキサンチンを生成することを観察し、すなわち、発明者らは、1.5%のアスタキサンチンにおける15g/Lのバイオマスにより、時間あたり1.0mg/L、好ましくはより高い時間あたり1.875mg/Lを、3%のアスタキサンチンにおける15g/Lのバイオマスにより、さらにより高い時間あたり3.75mg/Lを5日間で得ており;とりわけ、これは従属栄養成長における時間あたり0.063mg/L(Hata、2001年より)よりも30〜60倍高い。
突然変異体または遺伝子操作された生物における改変されたイソプレノイド生成を有する従属栄養細胞
この実施例は、フィトエン、ゲラニルゲラニルピロリン酸、フィトフルエンおよび他の上流の蓄積性色素または色素前駆体ならびにテルペンのような他の吸収源(sink)の改変された発現を有する従属栄養細胞に関する。そのような従属栄養的に成長した細胞は、本発明の方法において使用されうる。例えば、フィトエン(無色)およびフィトフルエン(色素性)は、化粧品におけるUV吸収において並外れた価値を有する。高い色素蓄積を含む従属栄養細胞は、もとのエンドポイント色素へのフラックスを停止または遅延させることにより、フィトエン(または他の化合物)の高い細胞および細胞株を作り出すための突然変異誘発または遺伝子操作による処理のための理想的な出発材料を作製する。実施例3におけるような従属栄養フラスコ培地中の5mLのH.プルビアリス(H. pluvialis)の従属栄養性の栄養培養を、それが0.15のOD750に達したときにCL−1000UV Crosslinker(254nmのUV−C光)を1分間、50%の細胞死が起こるように光源から13cmにおいて使用して、突然変異誘発した。振とうしない暗条件で室温における120時間の回収後、カロテン生成が開始するような十分のアンモニアが培地中に蓄積した。5mLの細胞を遠心分離し、4mLの培地を除去した。流動選別の前に50μmのメンブレンを通して細胞をろ過する直前に、DAPI(4’,6−ジアミジノ−2−フェニルインドール)を添加した(1μg/mL)。フローサイトメトリーを使用して、アスタキサンチンまたは他の有色色素を生成しない細胞を単離した。488nmの光源を使用した場合に、自己蛍光が530nmでは高く、695nmにおいて低い、微分蛍光分析に基づいて単離および選択を実行した。フィトエン含有量の定量化のために、単離物を実施例3のフラスコ従属栄養培地中、5mL体積まで120時間、成長させた。1mLのアセトンをバイオマス1mgあたりで使用して、凍結乾燥し、粉砕したバイオマスからフィトエンを抽出した。同じ突然変異体単離物からのフィトエン含有量分析を、異なる栄養成長物(vegetative grow out)およびカロテン生成シスト形成サイクルについて実行して、再現性を確実にした。フィトエン蓄積性突然変異体を単離するプロセスにおいて、(正常な野生型におけるような)アスタキサンチン以外の化合物を差次的に蓄積する細胞は、ルテイン、リコペン、ベータカロテン、カンタンキサンチン、またはフィトフルエンまたはさらに新しく異なる自己蛍光特性に基づいて同定可能であるような関連するゲラニルゲラニルピロリン酸を蓄積した。外因性の有機酸を供給される一方、暗条件で培養されるおかげで、この従属栄養細胞は、有害な成長効果のない別の表現型を発現する。これは、フィトエン突然変異体にとって特に有益である。この実施例は、ルテイン特性の変化を含む、葉緑素を欠く表現型の使用にもあてはまる。当分野で公知のとおり、色素をHPLCにより確認した。この従属栄養細胞由来のバイオマスは、ホスファターゼのような外因性酵素の導入のための、または化学修飾における使用のための蓄積された前駆体の単離のための標的となりうる。この細胞は、フィトエンデサチュラーゼノックアウトもしくはノックダウン、またはリコペンシクラーゼおよび下流のノックアウトもしくはノックダウンのような生合成経路中の酵素に影響を与える突然変異誘発されたまたは遺伝子操作された材料に起源を有しうる。
速い色素蓄積を伴う、アンモニウム耐性またはカロテン生成誘発された従属栄養細胞を単離すること
任意のカロテン生成トリガーまたはトリガーの組合せが、流動選別のための細胞を作成するために使用されうる。過剰のアンモニウムを使用する例として、実施例3におけるとおりの従属栄養フラスコ培地中のH.プルビアリス(H. pluvialis)の栄養培養5mLを、成長培地中のアンモニウムを例えば2.5mM以上まで蓄積させることにより、カロテノイド類を形成するように誘導した。アンモニウムストレスの8〜48時間後に、カロテノイド含有量が上昇した(低減された葉緑素)運動性細胞を、FACS Ariaフローソーターを使用するフローサイトメトリーにより単離する。運動性性細胞の単離を確実にするために、DAPIを分別培地に添加して、運動性細胞(DAPI陽性)およびシスト(DAPI陰性)を識別し;695nmからの低い自己蛍光を有するDAPI陽性細胞は低レベルの葉緑素を有し、530nmからの高い自己蛍光を有するDAPI陽性細胞は、ベータカロテンが上昇した(最終的なアスタキサンチン蓄積の代理)。実施例1に記載の方法を使用して、96ウェルプレートからの単一細胞単離物を実施例3のとおりに5mL体積まで成長させ、アスタキサンチン含有量定量化のためにカロテン生成形成条件に移した。1mgのバイオマスあたり1mLのアセトンを使用して、凍結乾燥し粉砕したバイオマスからアスタキサンチンを抽出した。同じ単離物からのアスタキサンチン含有量分析を、異なる栄養性成長物およびカロテン生成サイクルについても実行して、再現性を確実にした。アンモニウムストレスの期間を48時間を越えて延長することは、特に、ストレス状態が延長された後であっても、すぐに分化およびシスト形成せずに、カロテノイド類を蓄積してマクロ独立細胞のように運動性のままでいる亜集団を生成する1種または複数の細胞を選択する。この方法によって選択された細胞は、無色であること、またはアスタキサンチンに限らず前駆体または他のイソプレノイドの蓄積を示すことなどの様々な色素の特性を有する可能性がある。
藻類生成物の使用
多くの飼料、食品、栄養補給食品、医薬品、化粧品および作物保護への適用、ならびに多くの他の適用において、無処置のバイオマスまたは抽出した構成要素を利用するための多くの異なる方法がある。望ましい構成要素としてカロテノイド類を使用する、一部の非限定例を本明細書に提供する。例えば、本発明の方法により得られたアスタキサンチン含有生成物は、抽出されまたはバイオマス全体中に保持されるか、あるいは動物飼料、ヒトの栄養および栄養サプリメント、パーソナルケアおよび化粧品、ならびに着色剤のために使用される、脱脂した食物のような処理されたバイオマス中の残留物である。ヘマトコッカス(Haematococcus)藻類バイオマスまたは食物の一般的組成は、一般的なカロテノイド類、脂肪酸、タンパク質、炭化水素、およびミネラルからなる。ヘマトコッカス(Haematococcus)中のアスタキサンチンは、およそ70%の(16:0、18:1および18:2の脂肪酸に連結された)モノエステル、25%のジエステルおよび5%の遊離色素である。このエステル化された組成は、サケ科の魚類の天然の食餌供給源である甲殻類のそれに類似しており、容易に代謝される。1.5%のアスタキサンチン含有量において、約5.33kgのヘマトコッカス(Haematococcus)藻類バイオマスまたは食物が、この着色剤が飼料用に承認された典型である80ppmのアスタキサンチン濃度を達成するために、飼料1トンあたりに添加される。無処置のバイオマスのタンパク質および他の分画は、特に(化学合成され、持続可能でない)人工アスタキサンチンによってはもたらされない追加の飼料価値を提供する。本発明の方法により生成された壁の薄い藻類全体の相対的なもろさは、魚類の幼魚の健康および成長に影響を与えるための、消化性および栄養素利用可能性に関連する、当分野で公知の資格を与えられうる利益とともに、細胞破壊の容易さに関して、代わりの壁の厚い不動胞子よりも有益である。
クラミドモナス(Chlamydomonas)、クラミドカプサ(Clamydocapsa)およびクロロモナス(Chloromonas)由来のトランスジェニック従属栄養色素性細胞
この実施例は、細胞***せずに所望の成長速度において色素を生成する、クラミドモナス(Chlamydomonas)、クラミドカプサ(Clamydocapsa)およびクロロモナス(Chloromonas)を対象とする。コナミドリムシ(C. reinhardtii)を使用する1つの明示において、H.プルビアリス(H. pluvialis)由来のベータカロテンケトラーゼおよびカロテノイドヒドロキシラーゼを、米国特許出願公開第20090317878号に記載のようにCO2誘導性発現のためのプロモーターおよび当分野で公知のとおりH.プルビアリス(H. pluvialis)またはコナミドリムシ(C. reinhardtii)由来の任意選択の内因性制御エレメントを含有するプラスミドにクローニングした。さらに、例えば、米国特許第7,129,392号に記載のとおり、IPPイソメラーゼおよびメバロン酸経路の使用は、イソプレノイド生成を高める。プラスミドをコナミドリムシ(C. reinhardtii)の核に形質転換し、遺伝子導入系を当分野で公知のとおりに選択した。遺伝子導入系を色素含有量、実施例8および実施例4に記載の従属栄養成長についてそれぞれスクリーニングした。バイオ水素の副産物が、実施例11に記載の方法を使用する発酵の最後の数時間の間に生み出される。別の明示においては、通常は栄養枯渇およびハイライト(クラミドモナス目(Chlamydomonadales)の2段階プロセス)に曝露された後、細胞***を受けることによって、色素の蓄積が高まる、クラミドモナス(Chlamydomonas)、クラミドカプサ(Chlamydocapsa)またはクロロモナス(Chloromonas)の細胞を、実施例3に記載のとおりに培養して、光、栄養枯渇および細胞***の非存在下で色素性栄養細胞を得た。
バイオ水素生成物を生成するように改変されたH.プルビアリス(H. pluvialis)のトランスジェニック従属栄養細胞
この実施例は、独立型の生成物としてのまたは色素もしくは他の蓄積する化合物の副産物としてのバイオガスを記載する。コナミドリムシ(C. reinhardtii)由来のヒドロゲナーゼA1およびA2(HYDA1およびHYDA2)を(HYDEFとしてコナミドリムシ(C. reinhardtii)中で融合された)補助タンパク質HYDE、HYDFおよびHYDGとともにプラスミドにそれぞれの内因性制御エレメント(または任意選択のアンモニウム誘導性プロモーター)とともに当分野で公知のとおりにクローニングした。当分野で公知のとおり、プラスミドをH.プルビアリス(H. pluvialis)の核に形質転換した(Sharon-Gojmanら、2015年)。実施例8に記載のような好ましい菌株を選択するために、遺伝子導入系を色素含有量、従属栄養成長についてスクリーニングした。好気性条件下で高レベルの内部デンプンを含む十分な色素性バイオマスが生成した後に、副産物(バイオ水素)および導入遺伝子のmRNAレベルを、無酸素条件(嫌気性発酵)下でスクリーニングした。H2発生の持続時間は、デンプン異化が暗条件での発酵条件下でヒドロゲナーゼに電子を供与するときに細胞内に貯蔵された炭化水素の量に比例する。色素またはアスタキサンチンが、(形成したエタノール、乳酸塩、またはギ酸塩によるストレスとともに)溶解酸素が非常に低度である発酵によりストレス負荷された細胞中に蓄積するため、これは特に有利なプロセスである。こうして、発酵の最後の数時間は、必要に応じて、2つの生成物(1つは高価値および1つは低価値)を生成するために使用されうる。有利なことに、抽出により、このバイオマスは、内部貯蔵されたデンプン(重量)が細胞外生成物に転換されたため、より高い色素含有量を有するように見える。野生型または遺伝子改変されたクラミドモナス(Chlamydomonas)種も、当分野で公知のとおり、バイオガスのために使用されうる。本発明の方法は、体積基準でのより高いバイオマス収穫量、より高いバイオガス生産性とともに、以前にはこれらのクラミドモナス目(Chlamydomonadales)についての塩阻害により可能でなかった短期間でのより高いバイオマス密度を可能とする。4.3g/Lのバイオマス培養からの嫌気性条件下で時間あたり約0.96mmolH2/g乾燥重量の生産性率は、例えば、YuおよびTakahashi、2007年により示されたコナミドリムシ(C. reinhardtii)における時間あたり0.13mmolH2/Lからは著しい改善である、体積基準で時間あたり4.13mmolH2/Lまでの水素を0.135g/Lの計算された低い細胞密度で発生させる。
目的の組換え生成物を生成するために改変された従属栄養細胞
この実施例は、好ましい成長速度および比生産性により培養されて、本発明の方法を使用するフェドバッチ発酵を使用して組換え生成物を生成する、従属栄養細胞を対象とする。(色素、テルペン、バイオ水素のような)すでに例示した分子に加えて、本発明に有用な組換え細胞は、目的の(dsRNAを含む)異種RNAまたは異種タンパク質を生成する。そのような組換え細胞は、当分野で公知のとおりに遺伝子操作され、水産養殖および畜産における経口療法、作物保護、病害防除および健康増進、香味料および芳香剤が挙げられるが、これらに限定されない多くの分野における適用を有する(Somchaiら、2016年;Ceruttiら、2011年;Machadoら、2014年;Kumarら、2013年;Gimpelら、2015年)。本明細書では明示は、コナミドリムシ(C. reinhardtii)におけるプラスチドのタウマチン遺伝子発現について(タウマチンは、香味付けおよび香味マスキングのために使用されるタンパク質である)、およびdsRNA発現について与えられる。プラスチド発現は、複雑なタンパク質を発現することが周知であり、プラスチドとしてのdsRNAはDICER/RISC複合体のRNAiプロセシングを欠く。コナミドリムシ(C. reinhardtii)KAS1402のatpB欠損突然変異体を、rps14プロモーターにより制御された葉緑体コドンが最適化されたタウマチンとともにatpBおよび制御エレメントを含有するベクターK497でコーティングした0.6ミクロンの金微粒子を用いる微粒子銃(1100psi、標的距離6cm)により、形質転換した。固定炭素源を欠く培地において、光合成的に遺伝子導入系を選択した。3回の単一コロニー選択および酢酸を欠く培地における単離(光合成的)の後、遺伝子導入系を実施例3におけるとおりの従属栄養成長条件に移動した。発現解析を、製造者の指示に従ってプライマー585:CTGCTATTTCGACGACAGTGおよび586:ACGAGAACTCCGCTAAAGTGを使用して、qRT−PCRにより進める(iScript(商標)One−Step RT−PCR Kit With SYBR(登録商標) Green 170−8892)。遺伝子導入系におけるタウマチンmRNAレベルは、葉緑体ハウスキーピング遺伝子、Rpl14レベルと同様であったが、野生型において発現は見られなかった。120時間、10g/Lの細胞密度において0.1%で収穫した組換えタンパク質は、時間あたり0.08mg/Lのqpをもたらし、より高い収穫密度では、これは時間あたり0.08mg/Lを超えるであろう。コナミドリムシ(C. reinhardtii)KAS1402におけるdsRNA発現については、タウマチンを発現ベクター中で、GenScriptにより合成され、ベクターK588(図5)中に示される(Kumarら、2013年から改作した)3−HKT遺伝子断片のための789bp配列の逆方向反復コンストラクトで置換する。当分野で公知のとおり、対抗プロモーターを使用することにより、代わりのベクター構成が転写後に生成されるdsRNAについて可能である。選択されたコロニーを、実施例3および4に従って従属栄養的に成長させる。1日あたり0.7を超える72時間の最初の比成長速度の証明は、工業製造への規模拡張のための適合性を示している。実施例3および4に従って、4日間にわたる持続的な1.0/日の比成長速度のために、フェドバッチ発酵を使用して、100Lリアクター(Eppendorf BioFlo610)中で培養を50Lに規模拡張する。分析目的のためおよびキネティクスを確立するために、連続的遠心分離(WVO Designs Power Beast 3500rpm遠心分離機)を使用してバイオマスを水切りし、凍結乾燥した(12L Console 230−60 FreeZone LabConco凍結乾燥機アセンブリ)。
多様な表現型の従属栄養培養
この実施例は、栄養成長に好都合な条件により培養される新規従属栄養細胞型および本発明の方法を使用して生成物を作製することを対象とする。コナミドリムシ(C. reinhardtii)を例として用いて、菌株KAS1602(CC−125または137cに由来する変異体)を実施例3におけるとおりに従属栄養フラスコ培地中で成長させた。この細胞型は、無鞭毛性で葉緑素を欠き、栄養細胞としての緑色の概観よりも(一部、ルテイン/ゼアキサンチンに由来する)黄色であると特徴付けられる。これは、30μEの光とともに振とうフラスコ中で1週間、細胞を成長させ、次いで、(暗条件での静止フラスコとして)実施例3のフラスコ培地における従属栄養成長に資する条件で2週間、継代培養し、光源を容器上部に提供したときに走光性である能力のないことに基づいて鞭毛性細胞から単離するために、混合栄養条件下で蘇生された、先に凍結保存された細胞の個体群に由来した。これは、従属栄養下でロバストな成長により多くの世代の間、その表現型を保持した。この実施例は、例えば、Chlamydomonas Resource Centerから得た突然変異体菌株lts1−30mt−(CC−2359)または当分野で記載された(例えば、色素突然変異体に関するMcCarthyら、2004年および鞭毛突然変異体に関するMcVittie、1972年)とおりに生み出されたようなフィトエンシンターゼ遺伝子の突然変異によってカロテノイド類を欠くかまたは無鞭毛であるか、あるいは光合成または鞭毛突然変異体としての他の欠損を有する、UV光誘導性および化学的に誘導されたクラミドモナス(Chlamydomonas)突然変異体にも関する。さらに、この実施例は、暗条件での成長を可能とするための遺伝子操作を受ける藻類にもあてはまる。これは、制限なく、栄養転換のためまたは当分野で公知の好ましい炭素原料の利用のためのものを含む通性従属栄養生物に遺伝子操作された偏性光栄養生物を含みうる。これは、変異体、突然変異体および遺伝子工学を通じて達成されたような、偏性従属栄養生物にされたまたは光合成能力が弱められた通性従属栄養生物も制限なく含む。発酵においては、無鞭毛性細胞型は、それがインペラー(傾斜ブレードまたはRushtonインペラー)からの機械的損傷を受けにくく、改善されたガス交換および栄養素の混合のための350rpmよりも高いrpmでのファーメンター運転を可能とすることにおいて有利である。加えて、これは、暗条件において不必要な葉緑素を生成しないことにより低減された代謝負担からの利益を得る。結果として生じる、緑色または他の色素を欠くKAS1602および突然変異体菌株lts1−30mt−(CC−2359)のバイオマスは、化粧品、飼料などにおける使用に望ましい。
混合栄養培地組成およびマクロ独立細胞H.プルビアリス(H. pluvialis)による色素生成
この実施例は、従属栄養成長が30μEの光の下での混合栄養成長により置換されたような、有機酸および尿素を含む栄養培地を含む実施例3の変更である。H.プルビアリス(H. pluvialis)KAS1601の比成長速度は、0.05g/Lで開始した培養のバイオマスが96時間で9.0g/Lに達するような、1.5/日に達した。バイオマスの色素含有量は、2.2mg/L/時間のqpに相当する2.3%に達する。任意選択により、本発明の方法は、(話の中で議論されるような)当分野で公知のとおり、追加の仕上げステップにより藻類生成物を得るために実践されうる。例えば、加えられたストレスまたは光または誘導物質は、生成物収率を高めるためまたは高められた生成物含有量を有するシストを形成するために供給されうる(例えば、アスタキサンチン豊富なシストについては図2右下を参照されたい)。
非シスト形成微細藻類由来の着色補助剤および栄養素サプリメント
新しいプロセスは、ビタミンおよびミネラルとともに高タンパク質、低灰分を含み、栄養的、健康または着色の利益を付与する新しい生成物組成をもたらす、クラミドモナス目(Chlamydomonadales)からの色素性バイオマスの実質的に新しいプロフィールを提供する。家禽および魚類などのための飼料添加物のために、これらの利益は、魚類由来の成分により提供されるものに匹敵するが、着色のための色素沈着を提供する目的に役立つ。食物の代用となる飲料または栄養サプリメントのために、組成物は、栄養素添加の目的に役立つ。供給源である微細藻類次第で、食品としての組成物は、付随すると考えられる着色剤の任意の性質を有する、食事中のカロテノイド源であることができ、またはベータカロテンの場合のように、この材料は栄養素サプリメントおよび色添加物の両方として使用されうる。上記のように成長したバイオマスは、ファーメンターから収穫され、凍結乾燥され、次いで、New Jersey Feed Lab(Trenton NJ)などにより実行されるAssociation of Official Analytical Cheminsts(AOAC)の方法に従って、その主要構成要素について分析される。シスト形成していないバイオマスは次いで、当分野で公知のとおり、飼料配合物および着色または食品添加に適した形態に加工される。色素性の栄養細胞のタンパク質含有量は、本質的にシスト形成細胞または光誘導性細胞のそれの2倍であり、ヘマトコッカス属の種(Haematococcus spp.)またはドナリエラ属の種(Dunaliella spp.)のような飼料または食材において使用される商業的な色素性バイオマス全体における0.19〜0.24g/g DWタンパク質に比べて0.4〜0.5g/g DWタンパク質からなる(Lorenz、1999年;GRAS Notice grn000356;GRAS Notice grn000276;MuhaeminおよびKaswadji、2010年)。灰分は、0.16〜0.18g/g DW灰分から約0.03〜0.09g/g DW灰分まで約50〜80%低減される。よって、低灰分含有量とともに高タンパク質を含む新しい組成物は、重量ベースでより高い総合的な栄養価値を有し;魚類タンパク質は現在、価格が1500ドル/トンを超え、よって、一方で着色添加物(すなわち、着色剤)を送達する藻類タンパク質に置き換えられることが好ましい可能性のある飼料への実質的な投入原価を表す。これはまた、(アスタキサンチン抽出後に残るような)シスト形成バイオマス由来の脱脂ヘマトコッカス(Haematococcus)食物と比べて、かなり低量の繊維を有することにより異なることが好都合である:シスト形成バイオマス由来の脱脂された食物は、0.4g/g DWタンパク質により、タンパク質含有量はほぼ同様に等しいが、望ましくなく高い量の0.4g/g DW繊維も有する。アスタキサンチンは、サケ飼料1kgあたり80mgまでを構成する(WrolstadおよびCulver、2012年)が、タンパク質豊富なフィッシュミールは飼料1kgあたり200gを構成しうる(Hatlenら、2013年)。飼料1kgあたり80mgのアスタキサンチンでは、0.7%の色素含有量を有する11.4g藻類/kg飼料が必要であり;これは、収益化されうるタンパク質としての藻類バイオマスの50%を含み、3%のフィッシュミール置換率を提供する。

Claims (58)
- 目的の生成物を合成するための方法であって、
固定炭素源としての有機酸を含む培養培地を提供するステップと;
前記目的の生成物を生成する微細藻類細胞を提供するステップであって、前記微細藻類細胞が通性従属栄養生物である、ステップと;
前記微細藻類細胞を前記培養培地中、暗条件で培養して、前記微細藻類細胞から微細藻類培養物を生成するステップと;
前記微細藻類培養物中の前記微細藻類細胞が細胞分化を受ける前に、前記微細藻類培養物から前記細胞を単離するステップと;
前記微細藻類細胞から前記目的の生成物を精製するステップと
を含む、方法。 - 前記微細藻類細胞が、偏性従属栄養生物にされた通性従属栄養生物である、請求項1に記載の方法。
- 前記目的の生成物が、前記微細藻類細胞を含む微細藻類バイオマスである、請求項1に記載の方法。
- 前記目的の生成物が、色素、テルペン、組換え分子、バイオガス、またはその前駆体である、請求項1に記載の方法。
- 前記色素が、カロテノイド、イソプレノイド、またはその前駆体である、請求項4に記載の方法。
- 前記色素またはその前駆体が、アスタキサンチン、ルテイン、リコペン、ゼアキサンチン、カンタキサンチン、カロテン、フィトフルエン、フィトエン、またはその任意の組合せである、請求項4に記載の方法。
- 前記テルペンまたはその前駆体が、ピネン、リモネン、またはゲラニルゲラニルピロリン酸またはその任意の組合せである、請求項4に記載の方法。
- 前記組換え分子が、dsRNAまたは異種タンパク質である、請求項4に記載の方法。
- 前記微細藻類培養物が、少なくとも時間あたり0.063mg/Lの比生産性率、qpで前記色素を生成する、請求項5に記載の方法。
- 前記培養のステップが、1週間未満の間、行われる、請求項1に記載の方法。
- 前記目的の生成物の従属栄養成長および合成が、前記培養のステップの間に起こり、前記培養のステップが、フェドバッチ発酵下で実行される、請求項1に記載の方法。
- 前記目的の生成物の合成が、栄養豊富または枯渇条件下で起こる、請求項1に記載の方法。
- 前記有機酸が、酢酸、クエン酸、フマル酸、グリコール酸、乳酸、リンゴ酸、プロピオン酸、ピルビン酸、コハク酸、グルクロン酸、ガラクツロン酸、ウロン酸、クロロゲン酸、またはリグノセルロース酸である、請求項1に記載の方法。
- 前記培養培地が、窒素の主要供給源として尿素を含む、請求項1に記載の方法。
- 前記微細藻類細胞の乾燥、粉砕、溶解、または抽出のうちの1つまたは複数のステップをさらに含む、請求項1に記載の方法。
- 前記微細藻類細胞が、クラミドモナス目(Chlamydomonadales)に属する、請求項1に記載の方法。
- 前記微細藻類細胞が、ヘマトコッカス属の種(Haematococcus spp.)、クラミドモナス属の種(Chlamydomonas spp.)、クロロモナス属の種(Chloromonas spp.)、ドナリエラ属の種(Dunaliella spp.)、または、クラミドカプサ属の種(Chlamydocapsa spp.)である、請求項16に記載の方法。
- 前記微細藻類細胞が、優先的に有機酸および尿素を代謝する微細藻類細胞型で置き換えられる、請求項17に記載の方法。
- 前記微細藻類細胞が、第2の細胞と共培養される、請求項1に記載の方法。
- 前記第2の細胞が、その成長を支援するために、前記微細藻類細胞の成長を支援する有機酸と比べて異なる固定炭素源を使用する、請求項19に記載の方法。
- 前記第2の細胞が、窒素源としてアンモニウムを消費する、請求項20に記載の方法。
- 前記第2の細胞型が、セネデスムス属の種(Scenedesmus spp.)、クロレラ属の種(Chlorella spp.)、モノラフィディウム属の種(Monoraphidium spp.)、ロドトルラ属の種(Rhodotorula spp.)、珪藻植物、スラウストキトリッド(thraustochytrid)またはスラウストキトリッド(thraustochytrid)様微生物である、請求項21に記載の方法。
- 前記微細藻類細胞が、ヘマトコッカス種の属(Haematococcus spp.)に属し、前記第2の細胞がクラミドモナス種の属(Chlamydomonas spp.)に属する、請求項19に記載の方法。
- クラミドモナス目(Chlamydomonadales)由来の通性従属栄養微細藻類細胞であって、前記細胞が、炭素の主要供給源として有機酸のみを含有する培地中、暗条件で成長し、細胞分化が起こる前に、生成物を生成し;前記細胞が、ピネン、リモネン、アスタキサンチン、フィトエン、リコペン、ゲラニルゲラニルピロリン酸、バイオ水素、またはタウマチンを生成するための1つまたは複数のRNAまたはタンパク質に関する核酸を発現する組換え細胞であるか;または前記細胞が、dsRNA分子を発現する組換え細胞である、通性従属栄養微細藻類細胞。
- クラミドモナス目(Chlamydomonadales)由来の通性従属栄養微細藻類細胞であって、前記細胞が、炭素の主要供給源として有機酸のみを含有する培地中、暗条件で成長し、細胞分化が起こる前に、生成物を生成し;前記細胞が、イソプレノイドの改変された産生を伴う突然変異体細胞であるか、または鞭毛性突然変異体である、通性従属栄養微細藻類細胞。
- 前記細胞が、もとの菌株に対して改変されたイソプレノイドまたは葉緑素生成を有する、請求項24または25に記載の通性従属栄養微細藻類細胞。
- 前記細胞が、蛍光活性化細胞分取を使用して、カロテノイドの改変された生成を直接的に選択する、請求項24または25に記載の通性従属栄養微細藻類細胞。
- 前記細胞が、蛍光活性化細胞分取を使用する、カロテノイドでないテルペンの改変された生成を間接的に選択する、請求項24または25に記載の通性従属栄養微細藻類細胞。
- 目的の生成物の合成に好適な微細藻類細胞を同定し、単離する方法であって:
a.少なくとも部分的な従属栄養条件下で微細藻類菌株を培養して、微細藻類細胞を生成するステップと;
b.従属栄養条件下で成長させた場合、そこから前記微細藻類細胞が生成される前記微細藻類菌株に比べて好ましい特徴を有する非突然変異誘発微細藻類細胞を同定するステップであって、前記同定するステップが、蛍光活性化細胞分取技術および/または走光性反応を使用して実行される、ステップと;
c.前記好ましい特徴を有する前記非突然変異誘発微細藻類細胞を単離するステップと
を含む、方法。 - 前記培養のステップが、少なくとも前記培養ステップの一部については、混合栄養条件下で実行される、請求項29に記載の方法。
- 前記好ましい特徴が、目的の生成物の前記微細藻類細胞による合成の増加である、請求項29に記載の方法。
- 前記好ましい特徴が、前記微細藻類細胞における鞭毛の欠如である、請求項29に記載の方法。
- 前記非突然変異微細藻類細胞が、蛍光活性化細胞分取を使用して、カロテノイドでないテルペンの改変された生成を間接的に選択するか、またはカロテンの改変された生成を直接的に選択する、請求項29に記載の方法。
- 乾燥重量0.7%超のカロテノイド、乾燥重量40%超のタンパク質、乾燥重量10%未満の灰分、および2種以上のカロテノイドの混合物を含む組成を有する、クラミドモナス目(Chlamydomonadales)の従属栄養微細藻類バイオマスから得られた、着色剤、飼料添加物、ヒトの栄養素サプリメント、またはパーソナルケアもしくは化粧品成分。
- カロテノイド含有ライセートまたは抽出物である、請求項34に記載の着色剤、飼料添加物、ヒトの栄養素サプリメント、またはパーソナルケアもしくは化粧品成分。
- 微細藻類脂肪酸をさらに含有する、請求項34に記載の着色剤、飼料添加物、ヒトの栄養素サプリメント、またはパーソナルケアもしくは化粧品成分。
- 前記栄養豊富または枯渇条件が、硫酸塩、リン酸塩または尿素枯渇、または2.5mMを超えるアンモニウム蓄積のうちの1つまたは複数を含む、請求項12に記載の方法。
- 前記栄養豊富または枯渇条件が、2.6g/Lを超える浸透圧調節物質、45mMのNaCl、3g/Lを超える乳酸、成長温度をセ氏2度超える温度増加、またはその任意の組合せから選択される1つまたは複数の外因性因子と組み合わされる、請求項12に記載の方法。
- 前記微細藻類培養物が、100rpm以上の混合下で成長する、請求項1に記載の方法。
- サケ科魚類飼料中で使用される、請求項3および34に記載の微細藻類バイオマス。
- 幼虫駆除剤として使用される、請求項3に記載の微細藻類バイオマス。
- 香味料または香りの改変剤として使用される、請求項3に記載の微細藻類バイオマス。
- そのプロフィールが、微細藻類培養条件により変化する、請求項34に記載の着色剤、飼料添加物、ヒトの栄養素サプリメント、またはパーソナルケアもしくは化粧品成分。
- その中で1種のカロテノイドが総カロテノイドの50%超を構成する、請求項34に記載の着色剤、飼料添加物、ヒトの栄養素サプリメント、またはパーソナルケアもしくは化粧品成分。
- その中で1種のカロテノイドが総カロテノイドの85%超を構成する、請求項34に記載の着色剤、飼料添加物、ヒトの栄養素サプリメント、またはパーソナルケアもしくは化粧品成分。
- 1種のカロテノイドが、総カロテノイドの98%超を構成する、請求項34に記載の着色剤、飼料添加物、ヒトの栄養素サプリメント、またはパーソナルケアもしくは化粧品成分。
- 前記カロテノイドがアスタキサンチンである、請求項44、45および46に記載の1種のカロテノイド。
- 前記dsRNAが、少なくとも時間あたり0.04mg/Lの比生産性率、qpで生成される、請求項8に記載の方法。
- 前記異種タンパク質が、少なくとも時間あたり0.08mg/Lの比生産性率、qpで生成される、請求項8に記載の方法。
- シードトレインにおいて使用される、請求項3に記載の微細藻類バイオマス。
- 前記微細藻類培養物が、少なくとも0.24/日の比成長速度を有する、請求項1に記載の方法。
- 前記微細藻類培養物が、少なくとも0.77/日の比成長速度を有する、請求項51に記載の方法。
- 前記微細藻類培養物が、少なくとも1.0/日の比成長速度を有する、請求項51に記載の方法。
- 前記比成長速度が、96時間以上の期間にわたる、請求項51、52および53に記載の方法。
- 前記qpが、少なくとも時間あたり1mg/Lである、請求項9に記載の方法。
- 前記qpが、少なくとも時間あたり3.75mg/Lである、請求項9に記載の方法。
- 前記培養のステップが、約4日〜約1週間の間、行われる、請求項10に記載の方法。
- 前記比成長速度を有する微細藻類培養が、光の非存在下での細胞分化により、96時間以上の期間にわたって進行する、請求項1および54に記載の方法。
