Abstract
Description
本発明の実施形態は、サイズベース慣性分離を用いて血液および他の体液から標的細胞を富化するマイクロ流体デバイスに関する。
マイクロフルイディクスチップなどのマイクロ流体デバイスは、典型的には、材料(たとえば、ガラス、シリコン、またはポリジメチルシロキサン(PDMS)などのポリマー)中にエッチングまたは成形された一群のマイクロチャネルを含む。かかるマイクロフルイディクスチップの用途は、不均一サンプルから希薄濃度の比較的大きな粒子/細胞の富化である。典型的な例は、非常に高いスループットで血液細胞から循環腫瘍細胞(CTC)を富化することである。患者の血液サンプルからCTCを富化することは、癌の診断および治療に不可欠である。難題は、患者の血液サンプル中のCTCが希薄なことである。すなわち、CTCは、転移疾患の患者では全血1mL当たりCTC1〜10個程度の頻度で見いだされるが、同一体積の血液中には数百万個の白血球(WBC)および十億個の赤血球(RBC)が存在する。
本発明の第1の態様によれば、マイクロ流体デバイスが提供される。マイクロ流体デバイスは、標的細胞と非標的細胞とを含むサンプルを受容するための少なくとも1つの入口と、中心領域の上流端と周辺領域の下流端とを有する第1のスパイラルチャネル部分であって、上流端が入口に結合され、第1のスパイラルチャネル部分が、標的細胞と非標的細胞とが下流端で異なるストリームを占有するように構成される、第1のスパイラルチャネル部分と、第1のスパイラルチャネル部分の下流端で非標的細胞のストリームに結合するように配置された第1の廃棄物出口と、第1のスパイラルチャネル部分の下流端で標的細胞のストリームに結合するように配置されたリンクチャネル部分と、周辺領域の上流端と中心領域の下流端とを有する第2のスパイラルチャネル部分であって、第2のチャネル部分の上流端がリンクチャネル部分に結合され、第2のスパイラルチャネル部分が、標的細胞と非標的細胞とが下流端で異なるストリームを占有するように構成される、第2のスパイラルチャネル部分と、第2のスパイラルチャネル部分の下流端で非標的細胞のストリームに結合するように配置された第2の廃棄物出口と、第2のスパイラルチャネル部分の下流端で標的細胞のストリームに結合するように配置されたサンプル出口と、を含む。
以下では、限定されるものではないが例として添付の図面を参照して本発明の実施形態を説明する。

中心領域の上流端と周辺領域の下流端とを有する第1のスパイラルチャネル部分であって、前記上流端が前記入口に結合され、前記第1のスパイラルチャネル部分が、前記標的細胞と前記非標的細胞とが前記下流端で異なるストリームを占有するように構成される、第1のスパイラルチャネル部分と、
前記第1のスパイラルチャネル部分の前記下流端で非標的細胞のストリームに結合するように配置された第1の廃棄物出口と、
前記第1のスパイラルチャネル部分の前記下流端で標的細胞のストリームに結合するように配置されたリンクチャネル部分と、
