Abstract
Description
第一の制御性T細胞、第一の被抑制細胞、及び、第二の被抑制細胞、及び、必要に応じ第三番目以上の被抑制細胞、を準備する準備工程と、第一の抗原を第一の制御性T細胞に提示して、第一の抗原により第一の制御性T細胞を活性化させる抗原提示工程と、第一の制御性T細胞と、第一の被抑制細胞及び第二の被抑制細胞と、及び、必要に応じ第三番目以上の被抑制細胞、を共培養し、第二、及び、必要に応じ第三番目以上の被抑制細胞、の被抑制細胞の細胞***は低下されていないが、第一の被抑制細胞の細胞***は低下されている場合、第一の制御性T細胞は第一の被抑制細胞のみに対して特異的であると判定する判定工程と、を有する。
すなわち、従来の公知の検査法において、目的とする制御性T細胞の抑制能を評価する際、ポリクローナルな被抑制細胞をインビトロアッセイに投入する手法が広く知られている(非特許文献2、3,5、6,7)。
G: 制御性T細胞の有り条件における被抑制細胞の増殖細胞数
H: 制御性T細胞の無し条件における被抑制細胞の増殖細胞数
とおいた場合、抑制能= 1 - (G/H)の計算式により算出される。この算出法において前提とされてきたのは、被抑制細胞は、いずれの細胞も均等に抑制を受ける、という前提である。しかし本発明者らが明らかにしたことは、このような検査手法において、投入された被抑制細胞がポリクローナルな集団である場合は、抑制作用を受けるのは、投入された被抑制細胞のすべての細胞ではなく、あくまで、被抑制細胞のうち、そのアッセイに投入された制御性T細胞と同一の抗原を認識する被抑制細胞だけである、という点である。さらには、投入された被抑制細胞のうち、アッセイに投入された制御性T細胞と同一の抗原を認識しない細胞集団は、そもそも抑制されない、という観点である。
J: 利用された制御性T細胞と同一の抗原を認識する被抑制細胞の増殖細胞数
制御性T細胞の有り条件
K: 利用された制御性T細胞と同一の抗原を認識しない被抑制細胞の増殖細胞数
制御性T細胞の有り条件

とおいた場合、
G = J + K
H = L + M であり、
本発明における抑制能 = 1 - (J/L)
と計算される。
すなわち
G = J + K = 1 + 95 = 96
H = L + M = 5 + 95 = 100
である場合を仮定すると、
従来法によれば、
抑制能(従来法) = 1 - (G / H) = 1 -( 96 / 100) = 0.04 = 4 %
と過小評価され、抑制能があまり無いと判断されてしまうが、
本発明による計算法を用いることにより、
抑制能(本法) = 1 - (J / L) = 1 -(1 / 5) = 0.8 = 80 %
と強い抑制能を持つことが計算され、その抑制能を明瞭に示すことが可能になるのである。

の表現を用いて本発明を、数式を以って記述する。
Q = 第二の被抑制細胞の増殖細胞数:(いずれの制御性T細胞も無い条件)
R = 第一の被抑制細胞の増殖細胞数:(第一の制御性細胞の存在下)
S = 第二の被抑制細胞の増殖細胞数:(第一の制御性細胞の存在下)
T = 第一の被抑制細胞の増殖細胞数:(第二の制御性細胞の存在下)
U = 第二の被抑制細胞の増殖細胞数:(第二の制御性細胞の存在下)
において、従来法による抑制能は、
第一の制御性細胞 の 抑制能(従来法) = 1 - ((R+S)/(P+Q))
第二の制御性細胞 の 抑制能(従来法) = 1 - ((T+U)/(P+Q))
で記述されるが、本発明による抑制能は、
第一の制御性細胞 の 抑制能(本法) = 1 - ((R)/(P))
第二の制御性細胞 の 抑制能(本法) = 1 - ((U)/(Q))
と記述される。
例えば、
P = 100
Q = 1000
R = 5
S = 990
T = 95
U = 200
であった場合、
従来法による抑制能は、
PとQを識別せず、PとQの総和を求め、
RとSを識別せず、RとSの総和を求め、
TとUを識別せず、TとUの総和を求めてから抑制能を計算するので、
第一の制御性細胞 の 抑制能(従来法) = 1 - ((R + S) / (P + Q))
= 1 - ((5 + 990) / (100 + 1000))
= 1 - (995 / 1100)
= 0.09545
= 9.545 %
第二の制御性細胞 の 抑制能(従来法) = 1 - ((T + U) / (P + Q))
= 1 - ((95 + 200) / (100 + 1000))
= 1 - (295 / 1100)
= 0.731
= 73.2 %
と計算されるが、
本発明による抑制能は、
PとQを識別し、まずPとQの個々の値を求め、
RとSを識別し、まずRとSの個々の値を求め、
TとUを識別し、まずTとUの個々の値を求めてから抑制能を計算するので、
第一の制御性細胞 の 抑制能(本法) = 1 - ((R) / (P))
= 1 - ( 5 / 100 )
= 0.95
= 95 %
第二の制御性細胞 の 抑制能(本法) = 1 - ((U) / (Q))
= 1 - (200 / 1000 )
= 0.80
= 80 %
と計算される。
しかしMHCに依存しない抗原認識を用いる場合は、MHC分子を介さずTCR(T細胞レセプター)を認識し刺激することができるため、この場合は、目的の抗原を発現する任意の細胞を用いることが可能である。
[マウス]
C57BL/6およびBalb/cマウスは、CLEA Japan(東京、日本)から購入した。RAG-2欠損OTII TCRトランスジェニックマウス(以下OTII-マウス)、LCMV特異的マウス(以下SM-マウス)は動物施設で飼育した。全マウスは6ないし8週齢で使用し、施設の動物保護ガイドラインに従い、病原体の無い特別な条件で維持した。マウスB6 CD45.1 (Jax mice Stock No:002014)、B6.PL-Thy1(a)/CyJ (Jax406)はオリエンタル酵母工業株式会社より購入した。OTII-マウス、SM-マウスは、RAG2欠損マウスならびにJax002014, Jax406と交配し、各系統においてCD45.1,Thy1.1陽性のマウスを用意した。
次の試薬を、BD PharMingen(サンディエゴ、CA)から購入した。CD3ε(145-2C11)、CD28(37.51), CD16/32, APC-Cy7 Mouse Anti-Rat CD90/Mouse CD90.1(Clone OX-7),PerCP Mouse Anti-Rat CD90/Mouse CD90.1(Clone OX-7), Qdot605-CD4(RM4-5), PE-TCR Vb8.3(1B3.3)
以下をBiolegendより購入した。CD45.2-BV785 Brilliant Violet 785(TM) anti-mouse CD45.2 Antibody Clone 104, CD45.1 (A20)-PECy7,PerCP anti-mouse CD25 Antibody (Clone PC61 ), APC anti-mouse TCR Vβ5.1, 5.2 Antibody (clone MR9-4), Brilliant Violet 785(TM) anti-mouse CD45.2 Antibody Clone 104,
次の試薬を、Lonzaより購入した。RPMI 1640
次の試薬を、Peprotechより購入した。hIL-2 Recombinant Human IL-2,
Recombinant Human TGF-β1
被抑制細胞として、CD90.1(+)SM-ナイーブCD4 T細胞、CD45.1(+) OTII-ナイーブCD4 T細胞を用いた。
C57BL/6 CD45.1(+) CD45.2(+) OTII RAGKOマウス、ならびにC57BL/6 CD45.1(+)CD45.2(+) SM RAGKOマウスより、脾臓を取り出した。セルストレイナーを用いて脾組織を破砕し単細胞状態として浮遊細胞を分離した。細胞数を計算し、必要量を取り出し、CD4をマーカーとしてCD4T細胞をセルソーティングまたは、MACS磁気分離法を用いて単離した。単離したCD4T細胞を、TGFbeta,IL2と共にCD3e/CD28 コーティング培養器内にて、TFGβ、IL-2を添加し5日間培養しFoxp3陽性制御性T細胞を作成した。培養液にはこれらのほか、ATRA (all-trans-Retinoic Acid)を添加した。培養後の細胞は、細胞***検出用の色素Cell Trace Violet(ThermoFisher Scientific)で染色した。
被抑制細胞(エフェクター細胞)として、CD45.1(+) OTII-マウス、CD90.1(+) SM-マウスより、脾臓を取り出し、セルストレイナーを用いて浮遊細胞を分離した。浮遊細胞の一部は、必要量をCD4マーカーを用いて、CD4陽性T細胞をMACS磁気分離法を用いて処理し、単離した。その後、細胞***検出用の色素(Cell Trace Violet(ThermoFisher Scientific))で染色した。
野生型C57BL/6マウスより、脾臓を採取し、セルストレイナーを用いて浮遊細胞を分離した。浮遊細胞の一部は、CD11cマーカーを用いて、CD11c陽性T細胞をMACS磁気分離法を用いて処理し、抗原提示細胞として単離した。
50ml FBS (frozen at -20℃)
stock FBS are already HI(heat inactivated 56℃,30min)
5ml L-glutamine stock (200mM, 100x) (frozen at -20℃)
5ml NEAA stock (100x) in refrigerator
5ml sodium pyruvate stock (100mM, 100x) at refrigerator
5ml HEPES stock (1M, 100x) at room temperature
5ml streptomycine/ penicillin stock (100x) at -20℃
0.5ml 2-mercaptoethanol (1000x working stock;
37 ul 2ME in 10ml of PBS in refrigerator(4℃))
500ml RPMI-1640 with phenol red in cold room
Sterilize by filteration (0.2um)
結果、インビボで観察されたように、被抑制細胞であるSM-ナイーブCD4T細胞と、OTII-ナイーブCD4T細胞だけで培養した場合、それらはいずれも良好な細胞***を認めた。
実施例1においては、2種類のペプチド抗原はそれぞれ別々の抗原提示細胞により提示されたが、実施例2においては、2種類のペプチド抗原を、同一の抗原提示細胞により提示させた場合で行った。
実施例1,2では、2種類の外来性抗原ペプチド同士の共存状態における抗原特異性の解析を行った。即ち、実施例1,2では、2種類の抗原ともペプチド抗原であり、いずれもがMHC拘束性に提示されていた場合において実施例を示した。実施例3では、一方が外来性抗原ペプチドであるが、他方が細胞表面発現蛋白質の場合について解析した。細胞表面発現蛋白質として抗原性を有する場合として典型的なアロ抗原の場合について、例えば、H-2d MHCクラスII分子を取り上げ、免疫抑制が抗原特異的であるか検討を行った。
実施例3では、一方が外来性抗原ペプチドであるが、他方が細胞表面発現蛋白質の場合において、それぞれの抗原が別々の細胞に提示された場合について検証したが、実施例4では外来性抗原ペプチドとT細胞に応答性を有する細胞表面発現蛋白質(アロ抗原)が同一細胞に提示されている場合について検証した。
実施例3,4では、外来性ペプチド抗原、細胞表面発現蛋白質(アロ抗原)を用いた異なる二種類の抗原の共存環境における制御性T細胞による抗原特異的免疫抑制作用について検証を行ったが、実施例5では、2種類とも細胞表面発現蛋白質(アロ抗原)である場合について検証を行った。
本発明を用いることで抗原特異性調節因子のスクリーニングを行うことができる。
[制御性T細胞の作成と純化]
C57BL/6 CD45.1(+) CD45.2(+) OTII RAG-WTマウス、ならびにC57BL/6 CD45.1(+)CD45.2(+) SM RAG-WTマウスより、脾臓を取り出した。セルストレイナーを用いて脾組織を破砕し単細胞状態として浮遊細胞を分離した。細胞数を計算し、必要量を取り出し、CD25を標識し、MACS磁気分離法を用いてCD25+細胞を単離した。得られた細胞を、CD4,Foxp3GFPをマーカーとしてCD4T+Foxp3+細胞をセルソーティング法を用いて単離した。単離したCD4T+CD25+Foxp3+細胞を、CD3e/CD28 コーティング培養器内にて、IL-2を添加し5日間培養しFoxp3陽性制御性T細胞を作成した。培養されたリンパ球を、再びCD4,Foxp3GFPをマーカーとしてセルソーティング法を用いてCD4T+Foxp3+細胞を単離した。以上により得られた胸腺由来制御性T細胞を用いてアッセイを行った。
実施例7においては、2種類のペプチド抗原を、同一の抗原提示細胞により提示させアッセイを実施した。
Claims (4)
- 第一の制御性細胞、第一の被抑制細胞、及び、第二の被抑制細胞、及び、第三以上の被抑制細胞を準備する準備工程と、
第一の抗原を前記第一の制御性T細胞に提示して、前記第一の抗原により前記第一の制御性T細胞を活性化させる抗原提示工程と、
前記第一の制御性T細胞と、前記第一の被抑制細胞及び前記第二の被抑制細胞と、及び、第三以上の被抑制細胞を共培養し、前記第二の被抑制細胞,、ならびに第三以上の被抑制細胞の細胞***は低下されていないが、前記第一の被抑制細胞の細胞***は低下されている場合、前記第一の制御性T細胞は前記第一の被抑制細胞のみに対して特異的であると判定する判定工程を有することを特徴とする、インビトロでの制御性T細胞の特異性評価方法。 - 第一の抗原及び第二の抗原を前記第一の制御性T細胞に提示して、前記第一の抗原により前記第一の制御性T細胞を活性化させる抗原提示工程を有することを特徴とする請求項1に記載のインビトロでの制御性T細胞の特異性評価方法。
- 第一の制御性T細胞、第二の制御性T細胞、第一の被抑制細胞、及び、第二の被抑制細胞、を準備する準備工程と、
第一の抗原及び第二の抗原を前記第一の制御性T細胞に提示して、前記第一の抗原により前記第一の制御性T細胞を活性化させ、更に、第一の抗原及び第二の抗原を前記第二の制御性T細胞に提示して、前記第二の抗原により前記第二の制御性T細胞を活性化させる抗原提示工程と、
前記第一の制御性T細胞と、前記第一の被抑制細胞及び前記第二の被抑制細胞と、を共培養し、前記第二の被抑制細胞の細胞***は低下されていないが、前記第一の被抑制細胞の細胞***は低下されている場合、前記第一の制御性T細胞は前記第一の被抑制細胞のみに対して特異的であると判定し、
前記第二の制御性T細胞と、前記第一の被抑制細胞及び前記第二の被抑制細胞と、を共培養し、前記第一の被抑制細胞の細胞***は低下されていないが、前記第二の被抑制細胞の細胞***は低下されている場合、前記第二の制御性T細胞は前記第二の被抑制細胞のみに対して特異的であると判定する判定工程と、
を有することを特徴とする、インビトロでの制御性T細胞の特異性評価方法。 - 前記制御性T細胞は、Foxp3陽性の制御性T細胞又はIL-10産生性の制御性T細胞であることを特徴とする請求項1乃至3の何れか1項に記載のインビトロでの制御性T細胞の特異性評価方法。
