Abstract
Description
The present invention relates to a method for measuring the amount of angiotensin peptide in a sample and an angiotensin peptide quantification kit.
The renin-angiotensin system (RAS) plays an important role in the blood pressure regulation of the living body, and among them, the angiotensin peptide is a hormone that plays a central role in this system. Angiotensin peptides are produced from angiotensinogen (AGT) mainly produced in the liver by enzymes such as renin and angiotensin-converting enzyme (ACE). It has been pointed out that local and tissue RAS of organs such as the heart and kidneys are controlled by a mechanism different from that of whole body RAS, but there are still many unclear points. In particular, regarding the RAS of the kidney, there are several hypotheses regarding the production location and production mechanism of the angiotensin peptide, and the physiological significance is also discussed. Thus, accurate quantification of angiotensin peptides in organs such as kidneys and biological samples such as blood and urine is expected to help elucidate the involvement of tissues in the pathology of RAS.
At present, angiotensin peptides are quantified by immunoassays using antibodies such as radioimmunoassay (RIA) (see, for example, Non-Patent Document 1) and enzyme immunoassays (Enzyme-Linked immunosorbent assay; ELISA). There are many.
In addition, as another quantitative method, a measurement method combining liquid chromatography (LC) and mass spectrometry (MS) (hereinafter sometimes referred to as “LC / MS”) is used. In addition, a value obtained by quantifying angiotensin peptide in kidney, urine, plasma, or white adipose tissue has been reported (for example, see Non-Patent Document 2).
Furthermore, Patent Document 2 discloses a method for quantifying the amount of angiotensin peptide in plasma when the RAS angiotensin peptide production and degradation cascade reaches equilibrium in LC / MS.
In the quantification method of angiotensin peptide by immunoassay using an antibody, the quantitative value varies depending on the report. Although the antibody recognizes a specific amino acid sequence, it has been pointed out that each angiotensin peptide has high amino acid sequence homology, so that accurate quantification may be difficult due to cross-reaction of antibodies. In addition, it is difficult to quantify a plurality of types of angiotensin peptides at the same time by the quantification method of angiotensin peptides by immunoassay.
Moreover, the relative quantification is also included in the previous reports for quantifying the angiotensin peptide using LC / MS, and the quantification value varies depending on the report. In particular, only non-patent document 2 quantifies angiotensin peptides in the kidney using LC / MS using kidney, urine, plasma or white adipose tissue as a sample. However, the quantitative value is about 1000 times the quantitative value obtained by the conventional immunoassay, which is far from the same.
Furthermore, in

As the amino acid sequence shows, each angiotensin peptide has high amino acid homology. Therefore, in conventional immunoassay methods using antibodies, accurate quantification has been difficult due to cross-reaction of antibodies. In addition, in conventional immunoassay methods using antibodies, it has been difficult to quantify multiple types of angiotensin peptides simultaneously. In contrast, in the method of the present embodiment, since quantification is performed by mass spectrometry, each of these angiotensin peptides can be identified and accurately quantified by the difference in mass. Furthermore, it is also possible to quantify a plurality of types of angiotensin peptides at the same time.
Moreover, until now, in the method of quantifying angiotensin peptide using LC / MS, the quantified value is greatly different from the immunoassay method using an antibody. This is because the enzyme activity in the sample preparation stage was not sufficiently suppressed, the sample was not sufficiently purified, and the accuracy of the quantification method by mass spectrometry may be insufficient. It is done.
On the other hand, in this invention, an angiotensin peptide can be absolutely quantified by using the preparation method and purification method according to the kind of sample shown below, and the highly accurate quantification method using mass spectrometry.

When analyzing RAS of the whole body, it is preferable to use body fluid circulating throughout the body, such as blood, serum, and plasma, as a sample.
On the other hand, when analyzing RAS of a tissue, a tissue extract derived from an animal or a tissue extract derived from a cultured tissue, a cell extract derived from a cultured cell, or a body fluid derived from a specific tissue is used as a sample. It is preferable to do.
Specifically, urine (preferably, raw urine) may be used as a body fluid derived from a specific tissue, for example, in order to analyze RAS of the kidney.
For example, cerebrospinal fluid may be used to analyze RAS of the brain.
For example, in order to analyze RAS of the liver / biliary system, bile may be used.
Until now, in order to analyze RAS of tissues, it has been difficult to absolutely quantify angiotensin peptides in tissues. On the other hand, according to the method of this embodiment, a tissue extract derived from the above-mentioned test animal or a tissue extract derived from a cultured tissue, a cell extract derived from a cultured cell, or a body fluid derived from a specific tissue is sampled. And an angiotensin peptide in a tissue can be absolute quantified by using a preparation method and a purification method according to the type of the sample, and a highly accurate quantification method using mass spectrometry.
The animal from which the sample is derived is preferably a mammal. Examples of mammals include, but are not limited to, humans, monkeys, dogs, cats, rabbits, pigs, cows, mice, rats, and the like.
The isotope-labeled angiotensin peptide used in the method of the present embodiment may be any one having amino acid residues labeled with 13 C and 15 N isotopes. Specific examples of 13 C and 15 N isotope-labeled amino acid residues include, but are not limited to, valine, arginine, isoleucine, leucine, lysine, phenylalanine, proline, and the like. In the isotope-labeled angiotensin peptide, one type of amino acid residue may be substituted with 13 C and 15 N isotope-labeled amino acid residues, or two or more types may be substituted.
Among them, as the isotope-labeled angiotensin peptide used in the method of the present embodiment, the carbon atom of the second arginine residue from the N-terminus is substituted with a 13 C isotope, and the nitrogen atom is substituted with a 15 N isotope. It is preferable. This isotope-labeled angiotensin peptide results in a mass increase of about 10 Da compared to the native angiotensin peptide.
In
On the other hand, when quantifying two or more types of endogenous angiotensin peptides contained in a sample, two or more types of isotope-labeled angiotensin peptides corresponding to the endogenous angiotensin peptide to be quantified are mixed. do it. By mixing two or more types of isotope-labeled angiotensin peptides into the sample, the amount of two or more types of endogenous angiotensin peptides can be quantified simultaneously by mass spectrometry described later.
例えば、試料が動物から摘出された組織及び培養組織由来の組織抽出液、並びに培養細胞由来の細胞抽出液である場合、公知の方法(参考文献1:Navar L G et al., “Tubular Fluid Concentrations and Kidney Contents of Angiotensins I and II in Anesthetized Rats”, J. Am. Soc. Nephrol., vol.5, p1153-1158, 1994.)
[Sample preparation process]
Further, a sample may be appropriately prepared before the mixing step. The sample preparation method may be appropriately performed using a known method according to the type of the sample.
For example, in the case where the sample is a tissue extract derived from an animal, a tissue extract derived from a cultured tissue, and a cell extract derived from a cultured cell, a known method (Reference 1: Navar LG et al., “Tubular Fluid Concentrations and Kidney Contents of Angiotensins I and II in Anesthetized Rats ”, J. Am. Soc. Nephrol., Vol.5, p1153-1158, 1994.).
Specifically, first, the collected tissue, cultured cells, or cultured tissue are immediately homogenized in methanol that has been previously kept at -80 ° C. Isotope-labeled angiotensin peptide is mixed with the obtained homogenate. Thereafter, in order to remove excess proteins, cell membranes, and the like, centrifugation is performed to obtain the supernatant. Since this supernatant contains methanol, it is appropriately heated to evaporate the methanol. The obtained dried product can be dissolved in a sodium phosphate buffer or the like and used as a sample.
The preparation method of the tissue extract or cell extract has been used so far when quantifying angiotensin peptides by RIA. However, the present inventors, for the first time, performed the quantification of angiotensin peptide by mass spectrometry for the first time. As a preparation method, when the preparation method was applied, an angiotensin peptide could be quantified absolutely. This is because the activity of enzymes involved in RAS can be sufficiently suppressed by collecting tissue, cultured tissue, and cultured cells in methanol at a low temperature of −80 ° C. immediately, and angiotensin in vivo. This is presumably because the amount of peptide can be accurately quantified.
When the sample is a body fluid containing an enzyme such as protease (for example, blood, serum, plasma, raw urine, etc.), it is preferable to add a protease inhibitor immediately after collecting the body fluid such as blood. Thereby, the activity of the enzyme involved in RAS can be sufficiently suppressed, and the amount of angiotensin peptide in vivo can be accurately quantified.
Specific examples of the protease inhibitor include metal protease inhibitors such as EDTA; angiotensin-converting enzyme (ACE) inhibitors such as captopril and lisinopril; and renin inhibitors such as aliskiren. It is not limited to. These protease inhibitors may be added alone or in combination. Among them, in order to sufficiently suppress the activity of the enzyme involved in RAS, it is preferable to add a plurality of protease inhibitors in combination.
Further, after adding a protease inhibitor, an isotope-labeled angiotensin peptide may be mixed into a body fluid such as blood. When serum, plasma, or the like is used as a sample, a known method may be used to prepare protease-added and isotope-labeled angiotensin peptides from the mixed blood.
[Purification process]
Next, the endogenous angiotensin peptide and the isotope-labeled angiotensin peptide contained in the sample are purified from the sample mixed with the isotope-labeled angiotensin peptide.
As a purification method, a known method may be appropriately selected according to the type of sample.
Specific examples of the purification method include, but are not limited to, liquid chromatography (LC) and solid phase extraction. Among these, it is preferable to perform the solid phase extraction method and LC in this order for separation and purification.
For example, when the sample is a body fluid, it is preferable to perform separation and purification by a peptide extraction method in order to reduce proteins in the sample before solid phase extraction and separation and purification by LC.
Examples of the peptide extraction method include albumin removal treatment method, acetonitrile precipitation method, ultrafiltration method, DS method (Differential Solubilization method) and the like. Of these, the DS method is preferably used as the peptide extraction method.
Until now, the DS method has been used when searching and quantifying biomarkers (excluding angiotensin peptides) by LC / MS using a body fluid such as plasma as a sample. However, the present inventors have applied the DS method as a method for separating and purifying body fluids such as plasma and urine when quantifying angiotensin peptides by mass spectrometry for the first time this time. The absolute quantification of the angiotensin peptide contained in the sample could be performed. This is presumably because the angiotensin peptide was obtained at a higher recovery rate from the sample by using the DS method.
In particular, when performing separation and purification on a body fluid sample (for example, urine as shown in the examples described later), the ultrafiltration method is generally applied. . In contrast, the present inventors have found for the first time that a high recovery rate can be achieved by separating and purifying a body fluid sample containing almost no protein such as urine using the DS method. It is.
That is, in the method of this embodiment, when the sample is a body fluid, it is preferable to perform separation, purification by performing the DS method, the solid phase extraction method, and the LC in this order.
Specifically, the sample processing method using the DS method may be performed according to the following procedure.
First, a denaturing agent is added to the body fluid.
Examples of the denaturing agent include, but are not limited to, urea, thiourea, dithiothreitol (DTT), and the like. These modifiers may be used alone or in combination of two or more. Of these, urea, thiourea, and DTT are preferably used in combination because they can denature proteins more effectively.
Next, the body fluid to which the modifier is added is slowly dropped into an organic solvent such as acetone cooled to about 4 ° C. After dropping, the mixture is mixed at about 4 ° C. for about 30 minutes to 3 hours to denature and precipitate the protein in the sample. Subsequently, it isolate | separates by centrifugation etc., A supernatant is removed and a precipitate is obtained. The obtained precipitate is suspended in a strong acid such as hydrochloric acid and an organic solvent such as acetonitrile. After suspension, the mixture is mixed at about 4 ° C. for about 30 minutes to 3 hours to resolubilize the endogenous angiotensin peptide and isotope-labeled angiotensin peptide among the denatured proteins. Subsequently, separation is performed by centrifugation or the like, and the supernatant is obtained, whereby the endogenous angiotensin peptide and the isotope-labeled angiotensin peptide can be separated and purified. At this time, since the supernatant contains acetonitrile, the acetonitrile is evaporated by drying as necessary. The obtained dried product can be dissolved in an appropriate buffer to obtain a sample used for solid phase extraction or separation and purification by LC.
(Liquid Chromatography (LC))
As used herein, “liquid chromatography (LC)” refers to selective selection of one or more components of a fluid solution as the fluid uniformly permeates through the column or capillary channel of the micronized material. Means a delayed process. This delay results from the distribution of the components of the mixture between one or more stationary phases and the bulk fluid (ie, mobile phase) as the fluid moves relative to the stationary phase.
Specific examples of liquid chromatography include, for example, reverse phase liquid chromatography (RPLC), high performance liquid chromatography (HPLC), turbulent liquid chromatography (TFLC) (high turbulent liquid chromatography (HTLC) or high throughput. (Also called liquid chromatography).
In separation and purification by liquid chromatography (LC), a known LC device and column can be appropriately selected according to the type of angiotensin peptide. Chromatographic columns typically include a medium (ie, a packing component) to facilitate separation (ie, fractionation) of chemical moieties. The medium may contain fine particles.
Specifically, as a separation and purification method by LC, for example, a sample (or a sample purified in advance) is introduced into a column from an inflow port. It can then be eluted with a solvent or solvent mixture and discharged from the outflow port to obtain the produced sample.
Different elution modes may also be selected to elute the angiotensin peptide. Examples of the elution mode of liquid chromatography include a gradient mode, an isotonic mode, a polymorphic (ie, mixed) mode, and the like.
During chromatography, components (in this embodiment, angiotensin peptide) are separated according to various conditions such as eluent (also referred to as “mobile phase”), elution mode, gradient conditions, and temperature.
(Solid phase extraction method)
In the present specification, solid phase extraction (SPE) is one of analytical chemistry methods, and a target compound and impurities in a solution or suspension are based on physical or chemical properties. Means to separate. For example, as the mobile phase passes through or around the solid phase, undesirable components of the mobile phase are retained by the solid phase and the desired compound in the mobile phase is purified. Also, for example, the desired compound in the sample may be retained by the solid phase, and undesirable components of the mobile phase may be passed through or around the solid phase. In this case, the second mobile phase can then be used to elute the retained target compound from the solid phase for further processing or analysis.
A commercially available column may be used as the solid phase extraction column, and examples thereof include the Mono Spin (registered trademark) series (manufactured by GL Sciences).

<Process 2>
Subsequently, it is introduced into a mass spectrometer after separation and purification, and endogenous angiotensin peptide and isotope-labeled angiotensin peptide are detected and quantified.
In the present specification, “mass spectrometry (MS)” means an analysis technique in which a molecule is ionized and a compound is identified by its mass.
Specifically, as shown below, based on the mass-to-charge ratio (m / z) of the molecule, the ionization step, the detection step, and the quantification step are performed in this order.
[Ionization process]
In the method of the present embodiment, the analysis target needs to be ionized by the mass spectrometer. For this reason, the endogenous angiotensin peptide and isotope-labeled angiotensin peptide contained in the sample after separation and purification are ionized before being introduced into the mass spectrometer.
Examples of methods for ionizing endogenous angiotensin peptides and isotope-labeled angiotensin peptides include electron ionization, chemical ionization, electrospray ionization (ESI), photon ionization, atmospheric pressure chemical ionization (APCI), photoionization, and atmospheric pressure photoionization. (APPI), fast atom bombardment (FAB), liquid secondary ionization (LSI), matrix-assisted laser desorption ionization (MALDI), field ionization, field desorption, thermospray / plasma spray ionization, surface enhanced laser desorption ionization ( SELDI), inductively coupled plasma (ICP), particle beam ionization, and the like.
The ionization method can be appropriately selected based on the type of angiotensin peptide, the type of sample, the type of detector, the selection of positive / negative mode, and the like.
Among them, as a method for ionizing endogenous angiotensin peptide and isotope-labeled angiotensin peptide, electrospray ionization (ESI) or matrix-assisted laser desorption ionization (MALDI) is preferably used, and in the positive mode, electrospray ionization (ESI) is used. ) Is more preferable.
[Detection process]
The amount of endogenous angiotensin peptide and isotope-labeled angiotensin peptide ions are then detected.
Specifically, after the sample is ionized, the positively charged or negatively charged ions produced thereby can be analyzed to determine the mass to charge ratio. Examples of analyzers for determining the mass-to-charge ratio include triple quadrupole mass analyzers, quadrupole mass analyzers, ion trap analyzers, Fourier transform analyzers, orbitrap analyzers, and time-of-flight analysis. For example.
Ions can be detected using several detection modes. Specifically, for example, ions are detected using highly selective reaction monitoring (H-SRM), multiple reaction monitoring (MRM), selective reaction monitoring (SRM), selective ion monitoring mode (SIM), etc. May be.
Among these, the mass-to-charge ratio of ions of endogenous angiotensin peptide and isotope-labeled angiotensin peptide is preferably detected using multiple reaction monitoring using a triple quadrupole mass spectrometer.
Furthermore, it is preferable to detect two or more mass transitions when multiple reaction monitoring is used because it allows more accurate quantification.
Thereby, as shown in the below-mentioned quantitative process, two or more areas under the peak are obtained from a mass plot obtained from two or more mass transitions, and two or more quantitative values are obtained from two or more areas under the peak. By calculating the average value of these quantitative values, a more accurate quantitative value of the angiotensin peptide can be obtained.
Triple quadrupole mass analyzers are also called “tandem mass analyzers (MS / MS)” and can improve the selectivity of MS technology. In this technique, precursor ions (also referred to as parent ions) generated from the molecule of interest are filtered in an MS device, and then the precursor ions are fragmented to produce one or more product ions (daughter ions or fragments). This is also analyzed in a second MS procedure. By selecting a precursor ion, only ions generated by a specific molecule (in this embodiment an angiotensin peptide) pass through the fractionation chamber, where product ions are generated by collision with inert gas atoms. To do. Both precursor and product ions are produced reproducibly under a given set of ionization / fragmentation conditions. Therefore, MS / MS technology is a very powerful analytical tool. For example, a combination of filtration / fractionation can be used to eliminate interfering substances and is particularly useful in complex samples such as biological samples.
[Quantitative process]
In the quantification step, a signal is detected only when a daughter ion generated by dissociation of the ionized target peptide (parent ion, ie, angiotensin peptide) is detected. The target angiotensin peptide can be quantified from the peak of the signal. At this time, it is possible to calculate the recovery rate and the absolute quantification value by adding a known amount of a peptide obtained by isotopically labeling the angiotensin peptide to be quantified and using it as an internal standard.
More specifically, for example, the data on the amount of ions is relayed to a computer and generated as a plot of the number of ions versus time. From this plotted mass chromatogram, the area under the peak of endogenous angiotensin peptide (eg, angiotensin 1) and isotope labeled angiotensin peptide (eg, isotope labeled angiotensin 1) can be determined. Using these areas, the amount of endogenous angiotensin peptide can be calculated from the following formula [1].
Concentration of angiotensin peptide in sample = (area under sample peak / area under internal standard peak) × concentration of internal standard [1]
Moreover, the absolute amount of two or more types of angiotensin peptides can be obtained simultaneously by mixing two or more types of isotope-labeled angiotensin peptides.

For each sample, the first quadrupole (Q1) set so as to select a precursor ion having a mass-to-charge ratio as shown in Table 3 among the generated ions is passed through. It was. Next, ions flowing into the second quadrupole collided with the argon gas to generate an ion fraction. Next, among each ion fraction obtained in Q2, a third quadrupole (Q3) set to select a product ion (Production) whose mass-to-charge ratio is the value shown in Table 3 Was passed. Moreover, as shown in Table 3, as mass transition which is a combination of precursor ions and product ions,
At the same time, for the stable isotope-labeled angiotensin peptide that is an internal standard, the precursor ion and the product ion whose mass-to-charge ratio is the value shown in Table 3 are selected, and the triple quadrupole is similarly used. Mass spectrometry was performed.
In Table 3, “light” indicates an angiotensin peptide without labeling, and “heavy” indicates a stable isotope labeled angiotensin peptide.
さらに、MRM−HR(登録商標)法における定量メソッドの構築及びpeak area値の算出のためのソフトウェアとしてSkyline(MacCoss Lab Software、University of Washington)を用いた。
Further, Analyst (registered trademark) TF1.6 (manufactured by AB SCIEX) was used as LC-MS / MS data acquisition and analysis software for each sample. Furthermore, Skyline (MacCoss Lab Software, University of Washington) was used as software for constructing a quantitative method in the MRM-HR (registered trademark) method and calculating a peak area value.
The “MRM-HR (registered trademark) method” is a method that is based on almost the same principle as selective reaction monitoring (SRM). Specifically, in the MRM-HR (registered trademark) method, first, a peptide (target peptide) ion to be quantified is selected as a precursor ion in advance, and only the selected precursor ion is called a collision-induced dissociation method (CID). In this method, fragmentation is mainly caused by peptide bonds, and a product ion derived from the precursor ion is selectively detected.
As specific data analysis, first, a plot of the number of ions versus time was generated. The area under the peak was measured, and the following formula [1] was calculated based on the concentration of the internal standard stabilized isotope-labeled angiotensin peptide (25 fmol / on column (5 μL for all urine, plasma, and kidney samples)). Used to quantify the concentration of the sample.
Concentration of angiotensin peptide in the sample [fmol]
= (Area under the peak of the sample / Area under the peak of the internal standard) × Concentration of the internal standard ... [1]
In addition, a calibration curve was created using the following method, and since endogenous angiotensin peptide was included, only the slope of the straight line was used for calculation of the quantitative value.
First, a matrix that does not contain a stable isotope-labeled angiotensin peptide is prepared, a fixed amount of stable isotope-labeled angiotensin peptide (heavy peptides) is added as an internal standard, and an unlabeled angiotensin peptide (light peptides) with a further concentration is added. A standard solution was prepared by addition. The unlabeled angiotensin peptides (1, 2, 1-9 and 1-7) were manufactured by Sigma-Aldrich. Heavy peptides were added so that it might become 25 fmol / on column. Note that “on column” means the amount introduced, in this
Next, the ratio of the peak area value of light peptides and the peak area value of heavy peptides was taken (light / heavy), the horizontal axis was the concentration of light peptides, and the vertical axis was the peak area ratio (light amount). . The measurement was performed three times for each concentration standard solution. Moreover, the calibration curve was created using the method of small squares, and the linearity of the calibration curve was evaluated.
さらに、3回の測定のStandard deviation(SD)、精度(precision)及び真度(accuracy)を算出し、LLOQ(lower limit of quantification)を評価した。
また、Quality control sampleとして、低濃度(2.5fmol/on column)、中等濃度(50fmol/on column)、高濃度(75fmol/on column)の標準溶液を各5回ずつ測定し、precision及びaccuracyを評価した。
血漿試料でのアンジオテンシン1及び2の検量線を図1A(アンジオテンシン1)及び図1B(アンジオテンシン2)に示す。
Furthermore, the standard deviation (SD), accuracy (precision), and accuracy (accuracy) of three measurements were calculated, and LLOQ (lower limit of quantification) was evaluated. The evaluation item of LLOQ is “to satisfy precision ≦ 20% and accuracy ≦ 20%”.
In addition, as a quality control sample, a standard solution having a low concentration (2.5 fmol / on column), a medium concentration (50 fmol / on column), and a high concentration (75 fmol / on column) was measured 5 times each, and the precision and accuracy were measured. evaluated. The evaluation item was “to satisfy the precision ≦ 15% and accuracy ≦ 15%”.
Calibration curves for
またQuality control sampleとして、低濃度(2.5fmol/on column)、中等濃度(50fmol/on column)、高濃度(75fmol/on column)の標準溶液を各5回ずつ測定した結果、precision≦15%及びaccuracy≦15%を満たすことが確認できた。
From FIG. 1A (angiotensin 1) and FIG. 1B (angiotensin 2), LLOQ satisfying precision ≦ 20% and accuracy ≦ 20% is 0.5 fmol / on column for angiotensin 1 (AngI) and 0 for angiotensin 2 (AngII). It was 5 fmol / on column.
Further, as a quality control sample, a standard solution having a low concentration (2.5 fmol / on column), a medium concentration (50 fmol / on column), and a high concentration (75 fmol / on column) was measured five times, and precision ≦ 15% And accuracy ≦ 15% was confirmed.
