Abstract
Description
The present invention relates to a cell capture chip capable of capturing a single cell from a cell suspension and suitable for analysis of cell properties and functions, a method for producing the same, and a cell capture method.
Conventionally, as disclosed in the following

Further, in
However, since the microarray chip disclosed in
Therefore, the present invention has been made to solve the above-described problems, captures cells without damaging the cells, and improves the passage speed of the dispersion medium in the cell suspension in the pores of the cell capture chip. The method for producing a cell trapping chip of the present invention includes a first surface and a second surface as main surfaces, and penetrating from the first surface toward the second surface, and in the inside thereof, an opening diameter of the first surface and A substrate having a through-hole capable of capturing a single cell having a constricted portion whose diameter is smaller than the opening diameter of the second surface, on a second surface side inner wall having the constricted portion of the through-hole as a boundary A hydrophilization treatment for promoting the passage of liquid from the first surface side to the second surface side is performed.
Another method for producing a cell-trapping chip of the present invention includes a first surface and a second surface as main surfaces, and penetrating from the first surface toward the second surface, and the first surface is opened inside the first surface and the second surface. The second surface side having the narrowed portion of the through-hole as a boundary with respect to the substrate having a diameter and a through-hole capable of capturing a single cell having a narrowed portion smaller than the opening diameter of the second surface The inner wall is subjected to a hydrophilization treatment in which the dynamic contact angle immediately after dropping pure water is 30 degrees or less.
The cell trapping method of the present invention is characterized in that a cell suspension is dropped onto the first surface of the cell trapping chip of the present invention so that a single cell is trapped in the through hole.
In the present specification, the “hydrophilic treatment surface” refers to a surface of the substrate that has been subjected to a hydrophilic treatment, and indicates a surface having improved hydrophilicity after treatment than before treatment. Such a hydrophilic treatment surface is provided, for example, in the through hole in order to promote the passage of the liquid from the first surface side to the second surface side of the substrate. Further, the hydrophilicity of the hydrophilized surface is evaluated by dynamic contact angle measurement. In addition, the dynamic contact angle measurement in this specification is based on the dynamic contact angle immediately after dropping pure water on the surface of a simple flat substrate that differs from the substrate of the cell trapping chip only in that there are no through holes. It is done by measuring.
The dynamic contact angle is measured by installing an imaging device such as a CCD camera on the side surface of the substrate and observing the state of pure water dropped on the main surface of the substrate over time. The dynamic contact angle in the said invention means the contact angle in the picked-up image immediately after dripping (0 second after dripping).
According to the cell trapping chip, the method for manufacturing a cell trapping chip, and the cell trapping method of the present invention, the dispersion medium in the cell suspension is passed through the through-hole without using an external force such as suction, and the target cell is trapped. Therefore, the cells can be captured in the through-hole without damaging the cells. Furthermore, since the cell capture chip of the present invention has an improved passage speed of the cell suspension in the through-hole, the cell capture operation can be performed efficiently.
Hereinafter, the present invention will be described in detail with reference to the drawings. In addition, this invention is not limited to the following embodiment.
<Cell capture chip>
As shown in FIGS. 1A and 1B, the
The
In order to be suitable for such applications, the perforated substrate constituting the
The
Both the
The through-
Here, FIG. 1C shows an enlarged view of the through
As shown to (a) of FIG. 1C, the through-
The through
In FIG. 1C (a) to FIG. 1C (c), the constricted portion 13c is provided at the center position of the substrate in the thickness direction of the
However, the constricted portion 13c is not limited to the center position, and may be either close to the
The diameter rc of the constricted portion can be appropriately set so that a single cell can be captured depending on the type of cell suspension, the type, size, deformability, and the like of cells to be captured. As the diameter rc of the constricted portion, for example, a range of 1 to 30 μm is preferable, and 5 to 20 μm is more preferable.
The opening diameter r1 of the first surface and the opening diameter r2 of the second surface of the through
Thus, by increasing the opening diameter r1 of the first surface with respect to the diameter rc of the constricted portion, it is possible to facilitate introduction of cells to be captured into the through-hole 13c and not to increase pressure loss. Further, by increasing the opening diameter r2 of the second surface with respect to the diameter rc of the constricted portion, it is possible not to increase the pressure loss, and to easily discharge the dispersion medium in the cell suspension.
Further, when the through
First, as shown in FIG. 1C (b), in the second surface side inner wall 13b from the constricted portion 13c having the diameter rc to the second surface opening having the diameter r2, the inclination angle of the wall surface with respect to the axis of the through
When the dispersion medium moves from the constricted portion 13c toward the opening of the
Here, in order to promote the discharge of the dispersion medium from the
Therefore, in light of the above inclination angle theta, and the microscopic contact angle alpha 1, the contact angle alpha 1 is configured for the inclined surface, the concave surface of radius of curvature R is increased larger inclination angle theta is. That is, when the micro contact angle α 1 is determined by the surface characteristics, the meniscus concave surface is formed by adding the contact angle α 1 to the inclination angle θ. For example, if the microscopic contact angle alpha 1 of glass which is hydrophilized and 15 degrees, in the straight pipe with no inclination angle theta, the meniscus concave is remarkable, concave liquid surface at the center position of the opening The dent displacement amount is large (that is, the curvature radius R of the liquid surface is small).
On the other hand, when the inclination angle θ is provided, a meniscus concave surface having a substantial (α 1 + θ) angle can be formed, so that the concave displacement at the center of the opening can be reduced. Therefore, the dispersion medium driven by the capillary force can be brought as close as possible to the surface position of the second surface, and the discharge of the dispersion medium from the second surface can be promoted.
However, if the inclination angle θ is increased too much, the area of the opening is increased, resulting in a decrease in the hole density, which is not suitable for a filter application. In view of these matters, the second surface side inner wall 13b is preferably formed to have an inclination angle θ of 20 to 60 degrees. The inclination angle θ is more preferably 30 to 55 degrees, and further preferably 35 to 50 degrees.
Moreover, you may provide the angle of the inclined surface of this 1st surface side
And in this embodiment, the 2nd surface side inner wall 13b of the through-
Here, the “hydrophilic treatment surface” in this specification refers to a surface of the substrate that has been subjected to a hydrophilic treatment, and indicates a surface having improved hydrophilicity after the treatment than before the treatment. According to one embodiment of the present invention, such a hydrophilic treatment surface is provided in order to promote the passage of the liquid from the
If a micro contact angle α is to be measured on a substrate having a through hole, the contact angle β becomes macro under the influence of the shape of the through hole and the through hole pitch. Therefore, in this specification, the property of a hydrophilization treatment surface is defined by the measurement result of the dynamic contact angle measurement when the same surface treatment is performed on a substrate having no through hole. That is, the micro contact angle α can be equated with a dynamic contact angle measured on a mere flat substrate surface having a surface state equivalent to the surface to be evaluated.
Thus, the 2nd surface side inner wall 13b has hydrophilicity, can promote the passage of the dispersion medium in a cell suspension, and can improve a passage speed.
Hereinafter, the relationship between the macro (apparent) contact angle β and the micro contact angle α will be described.
Since the
Therefore, the contact angle on the substrate surface provided with the holes will be described below.
First, as shown in FIG. 2, for example, the substrate is provided with holes having openings of φ80 μm opened at a pitch of 100 μm, and the surface state (hydrophilicity) of the substrate surface and the inner wall of the hole is the same, A macro contact angle β when a dispersion medium droplet is dropped on the surface with a diameter of 500 μm or more on the substrate will be described.
In describing the macro contact angle β, the micro contact angle α will be described first. In FIG. 2 (a), in the case of a hole (not penetrating) perpendicular to the same diameter as the opening diameter, the tapered hole (penetrating through) shown in FIG. In this case, the micro contact angle in each part is shown.
First, the case of the vertically opened holes shown in FIG. The (1 ') flat portion in FIG. 2A has a micro contact angle α. This can be equated with a dynamic contact angle on a flat substrate surface in which no hole is formed. 2 (a) and 2 (b), the arc-shaped solid line on the substrate surface represents the contour of the pure water droplet, and the tangent (broken line) of the contour represents the micro contact angle α. It is shown. Next, (2 ′) at the boundary portion between the flat portion and the hole, when the boundary is reached in the case of the hole opened vertically, the inclination angle (90 degrees) of the side surface of the hole is added to α, and (90 + α) In the state where it is in contact with the inner wall surface of (3 ′), it is (90 + α) degree like (2 ′). Further, on the opposite inner wall surface (4 '), since it becomes a meniscus concave surface, the opening is filled with liquid and again exhibits a contact angle on a flat surface.
In the same way in FIG. 2B, (1) the flat portion has a micro contact angle α. (2) Due to the inclination angle θ of the wall surface at the boundary of the tapered hole, the angle added here is (90−θ) degrees, and (90−θ + α) degrees is the apparent contact angle, and (3) On the wall surface (up to the boundary of the constricted portion), the angle is (90−θ + α) as in (2). Furthermore, (4) since the surface is concave before the constricted portion, the liquid is filled and the contact angle α is obtained again.
Therefore, it is only necessary to estimate how much the peripheral length of the liquid-substrate interface of the dispersion medium droplet and the contact line are affected by the opening. The macro contact angle β is estimated by the ratio of the length occupied by the flat portion, where p is the average pitch of the openings. Assuming that the micro contact angle α (α 1 ) is 10 degrees, the inclination angle θ is 30 degrees, the average pitch p is 120 μm, and half of the opening diameter r is (90−θ + α) degrees, cos (β) = ( 1−r / 2p) cos (α) + (r / 2p) cos (90−θ + α). Therefore, if it is approximated that a fine structure composed of through-holes is formed at an average pitch on the outer peripheral length around the contact surface, only p and the occupation ratio of the holes may be considered.
From this point of view, FIG. 3 shows how the macro contact angle β occurs when the micro contact angle α of the substrate surface is taken on the horizontal axis and the tilt angle θ is 30 degrees, 45 degrees, and 60 degrees. The graph which estimated was shown. As can be seen from FIG. 3, the macro contact angle β is measured as a contact angle strongly dependent on the tilt angle θ. However, if the micro contact angle α is not low, the measured macro contact angle β is not small. For this reason, the macro contact angle β preferable for significantly improving the passing speed of the dispersion medium as an object of the present invention is 55 degrees or less under the virtually estimated conditions.
At this time, the hydrophilicity of the second surface side inner wall 13b is preferably higher than the hydrophilicity of the first surface side
Furthermore, it is preferable that not only the second surface side inner wall 13b but also the

Examples of such a hydrophilic treatment surface include a plasma treatment surface whose surface has been modified by plasma irradiation, a silica-coated surface coated with a silica film having good hydrophilicity, and the like.
Such a
Here, in the
When the hydrophilized surface is a plasma-treated surface, the contact angle increases over time (hydrophilicity decreases), so the dynamic contact angle is 3 minutes after the plasma treatment. Determine by measuring within.

The shape of the cell trapping chip is not particularly limited as long as it is a plate shape. For example, the shape in plan view may be various shapes such as a polygon such as a triangle and a quadrangle, and a circle such as a perfect circle and an ellipse. it can.
Moreover, it is preferable that this board | substrate has heat resistance and chemical resistance. Even when heat treatment is performed before passing the cell suspension, or when the suspension to be used is acidic or alkaline, it is stable due to such good physical and chemical durability. Thus, separation and capture operations can be performed.
<Method for producing cell capture chip>
The cell trapping chip of the present invention, for example, forms a through-hole having a desired hole shape by laser irradiation or mechanical treatment on a flat substrate to form a perforated substrate, and then the surface of the perforated substrate is changed. It can be produced by a hydrophilic treatment. Hereinafter, a case where a glass substrate is used will be described as an example.
<Formation of through holes>
First, a glass substrate, for example, an alkali-free glass is prepared. And a through-hole is formed in this glass substrate by laser irradiation or a hydrofluoric acid etching after laser irradiation at a place where a through-hole is to be provided. Since the through-
The through-
Thereafter, the glass on which the modified line group is formed is etched with 1% to 45% hydrofluoric acid. By performing this etching, the modified line group is selectively etched, and holes having two-dimensionally arranged hourglass-shaped through holes are formed due to the difference in etching rate and etching amount in the vicinity of the substrate surface and inside the substrate. A board can be created. At this time, hydrochloric acid or nitric acid may be added to hydrofluoric acid as an additive.
In addition to the above, the laser device only needs to be able to obtain the same pulse width, wavelength, and output. In addition, if the type of the target substrate or the size of the through hole is changed, the laser device is changed accordingly. The laser irradiation conditions may be changed as appropriate so that a desired hourglass-shaped through hole can be formed, and the present invention is not limited to this. Further, etching may be performed by alkali etching (NaOH) or the like.
<Hydrophilic treatment>
In the present embodiment, the hydrophilic treatment is a treatment for increasing hydrophilicity on the surface of the perforated substrate obtained above. Here, as the hydrophilization treatment, a known treatment can be used without particular limitation, and examples thereof include plasma irradiation and silica coating. Further, at this time, the hydrophilic treatment is preferably a treatment in which the dynamic contact angle immediately after dropping pure water is 30 degrees or less, as described in the hydrophilic treatment surface.
In this embodiment, the hydrophilization treatment by plasma irradiation includes removal of hydrophobic groups from the surface by surface cleaning with radicals (oxygen radicals, etc.), removal of attached impurities, attachment of hydrophilic groups to the surface, surface modification, etc. It is a process that includes a concept. Therefore, although a glass substrate will be described as an example of the substrate, even a substrate made of other materials can be made hydrophilic.
As plasma irradiation conditions, for example, a hydrophilic treatment can be performed by performing a treatment for 10 minutes at atmospheric pressure, glow discharge,
The hydrophilic treatment by coating is a treatment for covering the surface of the perforated substrate so as to have hydrophilicity. The material used here is not particularly limited as long as it can cover the substrate, and examples thereof include inorganic coating materials and organic coating materials. As the inorganic coating material, silica, titania and the like are preferably mentioned, and silica is more preferable. If the coating material is silica, hydrophilicity can be imparted in the long term. The silica coat is preferably a hydrolyzate of a silane compound having a hydrolyzable group or a hydrolyzate of silicic acid, and a part of the alkali metal is removed from the hydrolyzate of an alkoxysilane compound or an alkali metal salt of silicic acid. A hydrolyzate of the demineralized silicic acid removed is more preferred. These hydrolysates may have an unreacted silanol group (Si—OH) group.
Examples of the organic coating material include hydrophilic polymer compounds such as polyethylene glycol, biological membrane mimic materials such as 2-methacryloyloxyethyl phosphorylcholine polymer (MPC polymer), polyethylene glycol-modified alkoxysilanes, and the like. Such a coating material may be dissolved in a solvent to prepare a coating solution, which is applied to the surface of the perforated substrate and then dried to form a coating film. The solution may be applied by using a known application method such as dip coating or spin coating on the surface of the perforated substrate.

The cell capturing method of the present embodiment is a method in which a cell suspension is dropped onto the
The cell suspension used here is not particularly limited as long as single cells are dispersed and contained in a dispersion medium. Examples of the single cells to be contained include blood cells and cancer cells, and those having a diameter of about 10 to 20 μm are preferable. Examples of the dispersion medium include water and physiological saline.

From the above, it can be seen that when the second surface is hydrophilized, the permeation is good regardless of the hydrophilization treatment of the first surface or the slide glass plate surface. Further, as shown in Examples 6 to 7, even when the same cell trapping chip is used, the surface subjected to hydrophilic treatment is the upper surface of the filter (first surface), and the surface not subjected to hydrophilic treatment is the lower surface of the filter (second surface). When used as a surface), it can be seen that penetration is difficult.
Moreover, in Test Examples 1 and 2, it permeated instantaneously after dropping pure water, and in Test Examples 3 and 4 it permeated within 1 second after dropping pure water, which was a very good permeation state. Moreover, in Test Example 5, the infiltration occurred in about 5 seconds after the addition of pure water, and was in a good infiltration state. On the other hand, in Test Examples 6 to 9 in which the lower surface (second surface) of the filter was not hydrophilized, it was difficult to penetrate.
Moreover, in the cell capture chip | tip which has the straight type through-hole of Test Examples 10-13, it did not osmose | permeate irrespective of the presence or absence of a hydrophilic treatment.
<Contact angle measurement>
The contact angle in the hydrophilization treatment used in Examples 1 to 7 was tested and evaluated as follows.
A non-alkali glass substrate manufactured by Asahi Glass Co., Ltd. having a size of 75 mm × 25 mm × 0.2 mm without a through hole was prepared as a target substrate for the test. For this substrate, three types were prepared: a substrate whose surface was subjected to hydrophilic treatment by plasma treatment and silica coating treatment, and a substrate which was not subjected to hydrophilic treatment. The hydrophilic treatment was performed under the same conditions as in the above example.
Subsequently, the contact angle when 1 μL of pure water was dropped onto the measurement target surface of the obtained substrate was measured. At this time, the contact angle was measured by photographing from the side surface of the target substrate with a CCD camera and calculating the contact angle by image processing. During the measurement, the contact angle tended to decrease over time after dropping pure water, and the contact angle (that is, the dynamic contact angle) immediately after dropping (0 s) is shown in Table 5. This contact angle is an average value measured three times. Moreover, since the hydrophilic treatment by plasma irradiation has a tendency that the hydrophilicity changes with time (in the direction in which the hydrophilicity deteriorates), the measurement was performed within 3 minutes after the plasma irradiation.
As described above, according to the present invention, the object can be obtained by simply dropping the cell suspension into the capture region of the first surface of the substrate without sucking it from the opening (discharge port) of the second surface of the through hole. Can be trapped in the through-hole for each unit cell. As a result, the cells are not damaged. In particular, the passage speed of the dispersion medium in the cell suspension was improved by improving the hydrophilicity of the inner wall of the second surface opposite to the surface on which the cell suspension was dropped in the through-hole having the constricted portion. It is assumed.
The cell capture chip of the present invention can be used for analysis of properties and functions of each cell.
