Abstract
Description
(a)アストロサイトの集団をこの薬剤と接触させるステップであって、これらのアストロサイトは、多能性幹細胞(PSC)からex−vivoで分化したものである、ステップ;
(b)ステップ(a)のアストロサイトの集団又はその馴化培地を、ニューロンの集団と共培養するステップ;及び
(c)ニューロンの集団の生存又は神経機能を増強するこの薬剤の効果を定量するステップ、
を含む。
アッセイに使用するアストロサイトの生成
ヒトES細胞系を、ヒト新生児***線維芽細胞(HEF)のフィーダー層上で5%CO2中で37℃で培養した。増殖培地(ES1)は、DMEM/F12(Sigma)、14%ノックアウト血清代用品(KSR)、非必須アミノ酸(1/100)、0.1mMベータ−メルカプトエタノール(3つ全て、Invitrogen/Gibcoから)、1mMピルビン酸Na、2μg/mlヘパリン(Sigma)及び8ng/mlヒト組換え塩基性FGF(FGF−2;Preprotech)からなった。播種の5日後、hES細胞コロニーを、1.2mg/mlコラゲナーゼIV(Worthington)で37℃で45〜90分間脱離させた。凝集物を、上記のようにさらに培養し、又は記載されたように(Izraelら、Mol Cell Neurosci 34、310〜323(2007)、分化に供した。
ステップ4:Matrigel接着1:スフェア/クラスターを、DMEM/F12中に1:30希釈したBD Matrigel(増殖因子低減Matrigel、BD Biosciences)で室温(RT)で1.5時間にわたって被覆した6ウェル組織培養プレートに移した。培養培地は、20ng/ml EGFを含むITTSPP/B27であり、1日後に、いずれの非接着性凝集物をも廃棄した。培地を、7〜9日間にわたって1日おきに交換した。
1.化合物は、成熟アストロサイトに向かうhAPC分化の間に添加され得、その後、化合物曝露したアストロサイト又はそれらの馴化培地が、ニューロンに添加され得る。
2.酸化ストレスを誘導する前の、24〜72時間にわたる最後の培地交換の時点。
3.酸化ストレスの間の共培養中。
4.組み合わせ、即ち1+2、1+3、2+3などが実施され得る。
ラット胚(E15)由来の脊髄を、各実験に使用した。髄膜を、全ての脊髄から除去した。これらの脊髄組織を、小さい断片へと切り刻み、15mlチューブへと分割した(チューブ1つ当たり5つの胚)。トリプシン(2ml、0.25%)(Gibco #250050014)を、各チューブに添加し、37℃で15分間インキュベートした。細胞を、NB培地(7ml)及びFCS(1ml)中に再懸濁し、300gで5分間遠心分離し、上清を吸引した。ペレットを、NB培地(2ml)中に再懸濁した。単一細胞を計数し、matrigel(BD #FAL354230)被覆96ウェルプレート(Costar #cc−3596)上に播種した(120K細胞/ウェル)。細胞を、2日の培地交換で、50ng/ml NGF(Alomone #n−100)を補充したNB培地中で培養した。運動ニューロンの特徴付けを、3日間の培養後に実施した。ニューロンを、チューブリン−b3、ニューロフィラメント及びHB9(運動ニューロン特異的転写因子)で染色した。ニューロンの少なくとも70%が、これらの抗体について陽性であった。
アストロサイトを、(脊髄手術後)8〜10日目に播種した。細胞を、50ng/ml NGF(Alomone #n−100)を補充したNB培地を用いて培養した。
hPSC由来アストロサイト(30〜150日齢のアストロサイト)由来の培地を収集し;ビタミンCを補充した新鮮なN2B27で1:1希釈した。この培地を、ACM(アストロサイト馴化培地)と命名した。
50μM又は150μMのH2O2(Sigma #216763)を、ビタミンCを補充したN2B27、即ちACM(150μl)中に希釈し、アストロサイトを播種したプレートの各ウェルに添加した。これらのプレートを、37℃で6時間インキュベートした。
染色:
細胞を、Tx−100 0.3%(sigma)を補充したブロッキング緩衝液と共に室温(RT)で1時間インキュベートし、PBS(−/−)で洗浄した。

細胞を、PFA 0.5%(希釈、EMC)で再懸濁し、4℃で30分間インキュベートした。細胞を、300gで5分間遠心分離し、上清を廃棄した。細胞を、1ml PBS中で洗浄し、300gで5分間遠心分離し、上清を廃棄した。
細胞を、Tx−100 0.5%と共に4℃で30分間インキュベートして、細胞膜を透過処理した。細胞を、300gで5分間遠心分離し、上清を廃棄した。細胞を、PBS中に再懸濁し、300gで5分間遠心分離し、上清を廃棄した。
細胞を、FACS緩衝液(FB)で再懸濁した(1×105細胞/チューブ、100ulの体積中)。
非コンジュゲート化一次抗体及び非コンジュゲート化アイソタイプ対照抗体を、FB中に別々に希釈した。一次抗体を、製造者の指示に従って、最適な希釈で希釈した。細胞を、希釈した一次抗体又は希釈したアイソタイプ対照抗体と共に再懸濁し、4℃で30分間インキュベートした。細胞を、300gで5分間遠心分離し、上清を廃棄した。細胞を、FBで洗浄し、300gで5分間遠心分離し、上清を廃棄した。対応する蛍光色素コンジュゲート化二次抗体を、FB中に希釈した。細胞を、希釈した二次抗体と共に再懸濁し、4℃で30分間インキュベートした。二次抗体を、製造者の指示に従って、最適な希釈で希釈したが、このインキュベーションは、暗所で実施しなければならない。細胞を、FBで再懸濁し、300gで5分間遠心分離し、上清を廃棄した。細胞を、0.5ml FB中に再懸濁し、フローサイトメーターで分析した。
蛍光色素コンジュゲート化一次抗体を、製造者の指示に従って、FB中に希釈した。細胞を、コンジュゲート化一次抗体と共に再懸濁し、暗所で4℃で15分間インキュベートした。細胞を、300gで5分間遠心分離し、上清を廃棄した。細胞を、FBで再懸濁し、300gで5分間遠心分離し、上清を廃棄した。細胞を、0.5ml FB中に再懸濁し、フローサイトメーターで分析した。

0.05%トリプシン(Invitrogen #25200−056)を、細胞サンプル(1×106〜10×106細胞)に添加した。細胞を、37℃で5分間インキュベートした。トリプシンを、2体積の2mg/ml BSA(Sigma)の添加によって不活性化した。細胞を、300rpmで遠心分離し、上清を除去した。ペレットを、1mlのRLT(Qiagen)及び10μlベータ−メルカプトエタノール中に再懸濁し、−80Cで貯蔵した。

ヒトアストロサイトを、ステップ1〜8を経て、上記のように(「アッセイに使用するアストロサイトの生成」)、生成した。移植のために、ヒトアストロサイト先駆体細胞を、増殖因子の除去によって分化させた。2つの細胞集団を実験に使用した:一方は、増殖因子の非存在下で7日間増殖させ(「7日目」)、他方は、42日間増殖させた(「42日目」)。「7日目」及び「42日目」のヒトアストロサイトを、2.85×105細胞/μLの濃度でDMEM/F12中に懸濁した。
実験設計
5つの移植実験群を試験した。群番号1では、67±2日齢のSOD1G93Aマウスに、「42日目」の分化したヒトアストロサイトを移植した(n=10匹のSOD1G93Aマウス)。群番号2では、67±2日齢のSOD1G93Aマウスに、「7日目」の分化したヒトアストロサイトを移植した(n=12匹のSOD1G93Aマウス)。群番号3では、「7日目」細胞の2回の注射を、67±2日齢のSOD1G93Aマウス及び97±2日目のSOD1G93Aマウス(n=13)に対して実施した。群番号4には、67±2日齢のSOD1G93Aマウス(n=10)にビヒクルのみを注射し(DMEM/F12偽注射群)、群番号5は、いずれの処置も受けなかった(インタクト群、n=4)。これらのマウスを、移植の3日前に開始して実験の最後までの、10mg/Kgシクロスポリン(Sandimmun、Novartis)の毎日のI.P.注射を介して免疫抑制し、さらに、15mg/kg CellCept(Roche)(O.S当たり)を、移植の3日前に開始して移植の7日後まで、1日2回与えた。
免疫抑制した動物は、67±2日(全ての群)及び97±2(群番号3)において移植を受けた。7μlの培地、又は7μlの総体積中の2.0×106細胞を、大槽を介してくも膜下腔内注射した。細胞を、30ゲージの45°斜角針(Hamilton;Reno、NV)を取り付けたHamilton Gastightシリンジ(10μL)を使用して送達した。移植セッションの完了後、細胞生存度を、Nucleocounterを使用して評価したところ、85%よりも高いことが見出された。
G93A変異を有するヒトSOD1遺伝子を保有するトランスジェニックマウスを使用した(B6SJL−Tg(SOD1*G93A)1Gur/J)。雄性及び雌性マウスを、実験群間で均等に分けた。マウスは、The Jackson Laboratory(Bar Harbor、ME)から取得し、社内コロニーとして維持した。
全ての手順を、イスラエルのガイドラインに厳格に従って実施した:任意の潜在的な疼痛又は動物の不快感を最小化するために、手段を講じた。マウスを、食物及び水を自由に摂取させ、標準的な温度(21℃)で光制御された環境に収容し、同じ部屋に位置付けられた換気ケージのラック中で維持した。脱水症を回避するために、ケージの水皿へのアクセスを、動物が疾患症状を示し始めた場合に提供した。
前肢握力
動物の計量及び全ての行動データ収集を、移植の1週間前に開始し、最終段階まで1週間に2回実施した。前肢筋握力を、「握力メーター」を使用して別々に決定した。握力試験を、動物に、力計測器に取り付けた細いバーを握らせることによって実施した。その後、後肢又は前肢がバーを放すまで、計測器から動物を引き離した。これにより、最大握力の力についての値が提供される。力の測定は、3つの別々の試験で記録し、分析において平均を使用した。
運動機能を、1週間に2回実施した。運動機能を、加速Rota−Rodデバイス(180秒間、Rota−Rod 7650;Ugo Basile、Comerio、Italy)を使用して試験した。マウスがロッドから落ちた時間を記録した。動物を、記録する前に1週間トレーニングした。記録は、インプランテーションの1週間前に開始した。
スコアリングは、以下の表に従って、0〜5の尺度である。マウスには、臨床像が2つの規定されたスコアの間にある場合、「中間」スコア(即ち、0.5、1.5、2.5、3.5)が与えられ得る。
体重を、1週間に2回測定した。
信頼性がある倫理的な方式で疾患の最終段階を決定するために、最終段階を、横向きに寝かせた場合にマウスが30秒以内に正しい位置に戻ることができないことによって規定した。
SOD1G93AマウスのKaplan−Meier分析を、統計ソフトウェアSigmastat(SAS Software)を使用して実施して、生存、疾患発症及び持続時間のデータを分析した。Weight Rotarod、BBB及び握力の結果を、反復測定ANOVAを介して分析した。一部の場合には、Student t検定を実施して、動物の群間のデータを比較した。全てのデータを、平均±S.E.M.として示し、有意性のレベルをp≦0.05に設定した。
組織処理
動物を、0.3%生理食塩水とその後の氷冷4%パラホルムアルデヒド(Fisher Scientific;Pittsburgh、PA)とによる経心腔的灌流によって、最終段階で屠殺した。脊髄を、動物から取り出し、その後、30%スクロース(Fisher)/.1Mホスファート塩緩衝液中で4℃で3日間、凍結保護した。この組織を、OCT(Fisher)中に包埋し、ドライアイスで素早く凍結させ、処理するまで−80℃で貯蔵した。脊髄組織ブロックを、30μmの厚さで、矢状面又は横断面において切断した。切片を、スライドガラス上に収集し、分析するまで−20℃で貯蔵した。脊髄スライスのサブセットを、自由浮遊組織化学のためにPBS中に収集した。
HuNA(ヒト核抗原;Millipore;Temecula、CA;モノクローナル;1:400)を使用して、移植片由来のヒト細胞を選択的に同定した(hGRP及びhFの両方)。
in−vitroでのヒト多能性幹細胞(PSC)由来アストロサイトの生成及び特徴付け。
a.リアルタイムPCR(qPCR)。以下のプローブ(mRNA)を試験した:BDNF、GDNF、PDGFR、GFAP、GLAST及びALDH1L1。図1は、hESC由来アストロサイトのアストロサイト系統遺伝子発現の動態を実証した。成熟表現型に向かうアストロサイト先駆体細胞の分化を促進するために、増殖因子を、培地から除去した。サンプルを、0日目(増殖因子あり)、及び増殖因子を除去した状況で1週間毎に、収集及び試験した。ヒト胎児脳を、陽性対照として機能したヒト成人脳と一緒に、全ての試験した遺伝子について参照サンプルとして使用した(相対定量(RQ)=1)。ヒト成人脳は、50%を超えるアストロサイトからなることに留意することが重要である。この研究では、有意な増加が、細胞分化に際して全ての試験したアストロサイト遺伝子において見出され、多くの場合には、アストロサイト遺伝子(即ち、GFAP、BDNF、PDGFRα及びGLAST)の発現は、ヒト胎児脳よりも有意に高かった(図1)。これらの結果は、このプロトコールが、ヒトアストロサイトの富化された集団を生じることを証明している。
hPSC由来アストロサイト生物学的機能性の試験
いくつかの実験を実施して、hPSC由来ヒトアストロサイトが、成熟健常アストロサイトの機能的特性を示すことを証明した。in−vitroでの結果は、作用の可能性のあるin−vivo機構に光明を投じた。その目的のために、以下の機能的機構を試験した:
hPSC由来アストロサイトの移植のための最適な条件の発見
異なる移植態様のための最適化実験を、SOD1動物に対する実験の前に実施した。
i.異なる移植位置を試験した(即ち、脊髄前角中に直接、くも膜下腔内に、又は側脳室内に)。
ii.hPSC由来アストロサイトの数を試験した(注射(単数又は複数)部位(単数又は複数)当たり)。
ALS動物モデル−SOD1マウスへのhPSC由来アストロサイトの移植
MNに対するhPSC由来アストロサイトの神経保護効果を、in vivoで、即ち、動物の運動改善及び生存に対して試験した。その目的のために、hSOD1 G93A C57BL6/SJLバックグラウンド(高コピー数)マウスモデルを、材料及び方法に記載したように、使用及び処置した。
Claims (18)
- 筋萎縮性側索硬化症(ALS)を予防又は処置するための薬剤をスクリーニングする方法であって、
(a)アストロサイトの集団を前記薬剤と接触させるステップであって、前記アストロサイトは、多能性幹細胞(PSC)からex−vivoで分化したものである、ステップ;
(b)ステップ(a)のアストロサイトの前記集団又はその馴化培地を、ニューロンの集団と共培養するステップ;及び
(c)ニューロンの前記集団の生存又は神経機能を増強する前記薬剤の効果を定量するステップ、
を含む、上記方法。 - ニューロンの前記集団が、低酸素、酸化ストレス下、グルタマート毒性下又はAMPA/カイナート毒性下にある、請求項1に記載の方法。
- ステップ(b)において、アストロサイトの前記集団対ニューロンの前記集団の比率が、1:1、10:1、100:1、1000:1又は10,000:1よりも高い、請求項1に記載の方法。
- 前記アストロサイトが、GFAP、GLAST、AQP4の各々、又はそれらの組み合わせを発現する、請求項1に記載の方法。
- 前記アストロサイトが、BDNF、GDNF及びVEGFからなる群より選択される神経栄養因子の分泌を示す、請求項1に記載の方法。
- 酸化ストレスが、活性酸素種(ROS)、H2O2、及びそれらの任意の誘導体からなる群より選択される、請求項1に記載の方法。
- 前記定量するステップが、アポトーシス下にあるニューロンの数を計数することによって実施される、請求項1に記載の方法。
- 前記アポトーシスが、カスパーゼ−3a標識、アネキシンV、チューブリン−B3、HB9又はDAPIによって検出される、請求項7に記載の方法。
- 前記薬剤が小分子である、請求項1に記載の方法。
- ALSの進行を処置又は予防することを必要とする対象において、ALSの進行を処置又は予防するための方法であって;それを必要とする前記対象への、治療有効量の、ヒト前駆体アストロサイト又はアストロサイトの細胞集団の投与を含み、前記ヒト前駆体アストロサイト及びヒトアストロサイトは、多能性幹細胞(PSC)からex−vivoで分化したものである、上記方法。
- 前記前駆体アストロサイト又はアストロサイトが、GFAP、GLAST、AQP4の各々、又はそれらの組み合わせを発現する、請求項10に記載の方法。
- 前記前駆体アストロサイト又はアストロサイトが、BDNF、GDNF及びVEGFからなる群より選択される神経栄養因子の分泌を示す、請求項10に記載の方法。
- 前記前駆体アストロサイト又はアストロサイトが、グルタマート取り込み能力を示す、請求項10に記載の方法。
- 前記投与が、それを必要とする前記対象の脳脊髄液、脳又は脊髄に指向される、請求項10に記載の方法。
- 前記ヒト前駆体アストロサイト又はアストロサイトが、前記対象にとって非自己である、請求項10に記載の方法。
- 前記ヒト前駆体アストロサイト又はアストロサイトが、前記対象にとって同種である、請求項10に記載の方法。
- 前記ヒト前駆体アストロサイト又はアストロサイトが、遺伝子改変されていない細胞である、請求項10に記載の方法。
- ALSを処置するために同定された医薬の製造のための、ヒト前駆体アストロサイト又はアストロサイトの細胞集団の使用であって、前記ヒト前駆体アストロサイト又はアストロサイトは、多能性幹細胞(PSC)からex−vivoで分化したものである、上記使用。
