Abstract
Description
a)上述のバイオリアクターを用意するステップと、
b)細胞培養の培地及び細胞を容器の内部空間に添加するステップと、
c)攪拌手段によって攪拌をもたらすステップと、
d)液状溶液及び/又はガスを添加管を介して内部空間へと添加するステップとを含む細胞培養の方法を開示する。
Claims (17)
- 内部空間(3;23)を画成する容器(2;22)と、攪拌手段(4;24)と、1以上の添加管(5;25)とを備えるバイオリアクターであって、添加管の送出オリフィス(6;26)が内部空間内に位置しており、逆止弁(7;27)が、添加管から内部空間(3;23)の方向に流体を流すが、逆方向の流れは阻止するように、送出オリフィスの近傍に配置されている、バイオリアクター(1;11;21)。
- 逆止弁(7;27)が送出オリフィス(6;26)から所定の距離に位置しており、その距離が、添加管(5;25)の直径の10倍以下、例えば2倍以下である、請求項1に記載のバイオリアクター。
- 添加管の送出オリフィス(6;26)が、当該バイオリアクターの使用時に液体中に浸漬されるように配置されている、請求項1又は請求項2に記載のバイオリアクター。
- 送出オリフィス(6;26)が、逆止弁(7;27)で覆われている、請求項1乃至請求項3のいずれか1項に記載のバイオリアクター。
- 逆止弁の開きの圧力が10kPa以下、例えば10Pa〜10kPa又は100Pa〜10kPaである、請求項1乃至請求項4のいずれか1項に記載のバイオリアクター。
- 逆止弁が、弾性閉鎖部材(38、48、58、68)を備える、請求項1乃至請求項5のいずれか1項に記載のバイオリアクター。
- 弾性閉鎖部材が、管、リップ、フラップ及びスプリットディスクからなる群から選択される、請求項6に記載のバイオリアクター。
- 容器が、可撓性プラスチックバッグ(2;22)を備える、請求項1乃至請求項7のいずれか1項に記載のバイオリアクター。
- 可撓性プラスチックバッグ(2)が、使用時に剛性支持構造(18)で支持されるように構成されている、請求項8に記載のバイオリアクター(11)。
- 攪拌手段がインペラ(4)を備える、請求項1乃至請求項9のいずれか1項に記載のバイオリアクター(1、11)。
- 可撓性プラスチックバッグ(22)が膨張可能であり、攪拌手段が可撓性プラスチックバッグのための可動支持プラットフォーム(24)を備える、請求項8に記載のバイオリアクター(21)。
- 放射線滅菌される、請求項1乃至請求項11のいずれか1項に記載のバイオリアクター。
- 添加管が、pH調節剤、栄養剤及び/又は成長因子を含む液状溶液を収容している供給容器と流体接続される、請求項1乃至請求項12のいずれか1項に記載のバイオリアクター。
- 容器の内部空間内に位置する細胞培養の培地などの液体をさらに含み、添加管の送出オリフィスが液体中に浸漬されている、請求項1乃至請求項13のいずれか1項に記載のバイオリアクター。
- 細胞培養の方法であって、
a)請求項1乃至請求項14のいずれか1項に記載のバイオリアクターを用意するステップと、
b)細胞培養の培地及び細胞を内部空間に添加するステップと、
c)攪拌手段によって攪拌をもたらすステップと、
d)液状溶液及び/又はガスを、添加管を介して内部空間へと添加するステップと
を含む方法。 - ステップb)において、細胞培養の培地の量は、添加管の送出オリフィスを浸漬するのに充分である、請求項15に記載の方法。
- 液状溶液が、pH調節剤、栄養剤及び/又は成長因子を含む、請求項15又は請求項16に記載の方法。
