Abstract
Description
成熟神経細胞は増殖能力や失われた神経細胞を補填する能力が非常に低いため、成人CNSにおいて急性かつ/または慢性的に神経細胞が失われると、不可逆的な機能喪失につながる。したがって、機能を維持するためには、神経細胞の喪失を緩和するか抑制することが必須である。神経変性疾患の多くでは、その病因が明らかになっていないため治癒することができない。しかし、一次的な危険因子や二次的な危険因子がいくつかあり、これらの危険因子を標的として、神経細胞喪失の進行を抑制または緩和するための治療的介入がなされており、このような治療的介入を神経保護療法と総称している。危険因子のいくつかは疾患特異的であるが、例えば興奮性アミノ酸、フリーラジカル、および一酸化窒素などは神経変性障害すべてに共通する危険因子である。これらの危険因子は健常なCNSの構成に必須の自己成分であるが、変性組織中において過剰に蓄積すると細胞毒となり、神経細胞死が生じた領域からさらに損傷が広がる。
神経変性疾患の治癒方法は現在のところ存在せず、症状の緩和または管理に焦点を当てた治療が行われている。現在までになされた臨床研究のほとんどにおいて、神経保護療法の有効性は示されていない。AMDについては、抗血管新生剤または抗VEGF剤を眼の硝子体液中に直接注射することにより異常血管を退縮させて視力を改善できる可能性が示されている。
再生医療は、これまで回復不能とされていた臓器を刺激して自己治癒を促すことにより損傷した組織および臓器を修復、置換および/または再生させることを目的とした新たに台頭しつつある分野であり、組織工学、生体材料、細胞療法などが含まれる。細胞の再生は、末梢組織において一般的に見られる治癒プロセスであるが、成人の神経網膜でもそれ以外のCNSでも限定的にしか起こらない。しかし、網膜前駆細胞(RPC)の静止期細胞集団は、成人期を通して網膜毛様体(CB)に存在し、様々な網膜細胞に分化できる可能性を有しており、恐らく免疫調節物質や神経向性物質を産生できると考えられる。この静止期の前駆細胞集団は網膜損傷によって刺激を受けることが報告されたが、そのメカニズムは未だ明らかになっていない。損傷に応答して作動する治癒プロセスを解明し、このプロセスを増強できる方法を見出すことができれば、神経保護および細胞再生を促進するための新たな治療法の開発につながる可能性があり、これは当技術分野の研究目標の1つとなっている。

成体雄(8〜10週齢)C57BL/6Jマウスおよびヘテロ変異Cx3cr1GFP/+マウス(B6.129P-Cx3cr1tm1Litt/J、CX3CR1ケモカイン受容体対立遺伝子の一方が、GFPをコードする遺伝子で置換されたマウス;Jung, S., J. Aliberti, P. Graemmel, M. J. Sunshine, G. W. Kreutzberg, A. Sher, and D. R. Littman. 2000. Analysis of fractalkine receptor CX(3)CR1 function by targeted deletion and green fluorescent protein reporter gene insertion. Mol. Cell. Sparwasser, S. Wu, S. Vuthoori, K. Ko, F. Zavala, et al. 2002. In vivo depletion of CD11c+ dendritic cells abrogates priming of CD8+ T cells by exogenous cell-associated antigens. Immunity. 17:211-220)を用いる。FoxP3GFPマウスは、Memorial Sloan-Kettering Cancer CenterのAlexander Rudensky博士の厚意により入手した。ハンチンチンをコードするヒト遺伝子を過剰発現するトランスジェニックR6/2マウスは、ジャクソン研究所から入手した。
過去の報告(Rolls, A., R. Shechter, A. London, Y. Segev, J. Jacob-Hirsch, N. Amariglio, G. Rechavi, and M. Schwartz. 2008. Two faces of chondroitin sulfate proteoglycan in spinal cord repair: a role in microglia/macrophage activation. PLoS Med. 5:e171.; Shechter et al., 2009)に従って、[Cx3cr1GFP/+>WT]BMキメラを作製する。すなわち、WTレシピエントマウスの頭部を遮蔽して全身に致死量の放射線(950rad)を照射する。頭部を遮蔽することによって、網膜への直接的な損傷と、グルタミン酸塩の毒性暴露により誘導される骨髄系細胞以外の骨髄系細胞による浸潤を防ぐ。翌日、過去に報告されたプロトコル(Shechter et al., 2009)に従い、BM細胞5×106個を用いてマウスを新たに作製する。BM移植の8〜12週間後、キメラマウスをグルタミン酸塩による毒性暴露またはEAU誘導プロトコルに供する。
MC-21(CCR2に対する抗体;Mack, M., J. Cihak, C. Simonis, B. Luckow, A. E. Proudfoot, J. Plachy, H. Bruhl, M. Frink, H. J. Anders, V. Vielhauer, et al. 2001. Expression and characterization of the chemokine receptors CCR2 and CCR5 in mice. J. Immunol. 166:4697-4704)の腹腔内注射を損傷形成直後から開始し、実験期間を通して継続する。EAU誘導の場合、注射はEAUのピークの前後に行い、1日おきに合計5回行った(注射1回につき8μg投与)。
過去の報告(Varol, C., L. Landsman, D. K. Fogg, L. Greenshtein, B. Gildor, R. Margalit, V. Kalchenko, F. Geissmann, and S. Jung. 2007. Monocytes give rise to mucosal, but not splenic, conventional dendritic cells. J. Exp. Med. 204:171-180)に従って、CD115+単球を単離する。すなわち、マウスの大腿骨および脛骨からBM細胞を採取し、フィコール密度勾配により単核細胞を濃縮する。製造元のプロトコルに従ってビオチン化抗CD115抗体およびストレプトアビジン結合磁気ビーズ(ミルテニーバイオテク)を用いたMACSで濃縮することによってCD115+BM単球集団を単離する。次いで、損傷形成後1日目に単球(WT、Cx3cr1GFP/+、IL-10欠損、またはMHC-II欠損)を静脈内注射する(マウス1匹につき4〜5×106個の細胞を投与)。
Zhao et al., 2005(Growth factor responsive progenitors in the postnatal mammalian retina. Dev. Dyn. 232:349-358)に記載の方法を一部変更したプロトコルを用いて、BrdU(シグマアルドリッチ)をPBS中に溶解し、該溶液の硝子体内注射(眼球1つあたり1μg投与)を損傷形成直後から開始し、3日間継続して行う。この反復注射は、組織の他の部分を穿孔することなく、同じ孔から行う。これによって、網膜は、注射を1回しか受けていない場合と同等な形態を維持すると考えられる。最後の注射から1日後に動物を屠殺して増殖前駆細胞数の測定を行う。あるいは、最後の注射から1週間後に動物を屠殺して細胞の分化を検出する。コントロール動物には、グルタミン酸塩による毒性暴露やIOPの上昇を行うことなく、同じ投与レジメンに従って硝子体にBrdUを投与する。様々な単球操作を行った動物の網膜や毛様体を比較する際にこれと同じプロトコルが用いられることから、損傷とそれに次ぐBrdUの反復注射による増殖RPC数に対する相乗効果を否定することはできないが、こうした操作の後に見られる増殖前駆細胞数の変化は単球を介した効果に起因する可能性が高いと考えられることには注目されたい。
解剖学的に無傷の神経細胞を検出するため、損傷形成後3日目に、5%フルオロゴールド(蛍光色素)の生理食塩水溶液1μlをマウスの両側上丘に注射する。両側上丘における注射位置は、ブレグマの後方2.92mm、正中線の側方0.5mm、および頭蓋からの深さ2mmとする。72時間後にマウスを屠殺して眼球を取り出し、網膜を平坦にしながらPBS中4%パラホルムアルデヒド(PFA)を用いて全載標本とする。視神経円板からの距離がほぼ同じとなるように(0.3mm)視野を選択して、視神経円板からの距離に応じたRGC密度の変動を除外した。マウスに施した処置についての情報を持たない観察者によって蛍光顕微鏡(倍率800倍)下で視野をカウントした。各網膜について、1視野あたりの平均RGC数を算出した。詳細については、Schori et al., 2001(Vaccination for protection of retinal ganglion cells against death from glutamate cytotoxicity and ocular hypertension: implications for glaucoma. 過去の報告(Shechter, R., Y. Ziv, and M. Schwartz. 2007. New GABAergic interneurons supported by myelin-specific T cells are formed in intact adult spinal cord. Stem Cells 25:2277-2282)に従って、PBS灌流固定後に眼球を取り出し、2.5%PFA中で24時間かけて後固定して、70%エタノール中に移し、続いてパラフィン包埋した。病理組織学的検査を行うため、切片をいくつか選択してヘマトキシリンおよびエオシンで染色した。免疫標識に用いた抗体は、ウサギ抗GFP(1:100、MBL)およびマウス抗Brn3a(1:50、サンタクルーズバイオテクノロジー社)であった。M.O.M. Immunodetection Kit(ベクターラボラトリーズ)を使用して、マウス一次モノクローナル抗体の局在を特定した。活性化した骨髄系細胞を標識するため、FITC標識Bandeiraea simplicifoliaイソレクチンB4(IB-4;1:50;シグマアルドリッチ)を二次抗体溶液に加え、1時間インキュベートした。使用した二次抗体は、Cy2/Cy3標識ロバ抗マウス抗体またはCy2/Cy3標識ロバ抗ウサギ抗体(1:150〜1:200、いずれもJackson ImmunoResearch Laboratories社より入手)を含んでいた。スライドをヘキスト(1:2,000;インビトロジェン)で1分間染色した。顕微鏡分析には蛍光顕微鏡(E800、ニコン)を使用した。この蛍光顕微鏡は、デジタルカメラ(DXM 1200F;ニコン)、および20×、NA0.50の対物レンズまたは40×、NA0.75の対物レンズ(Plan Fluor;ニコン)を備えていた。画像取得ソフトウェアNIS-Elements F3(ニコン)を使用して、後固定後の組織の撮影を24℃で行った。撮影した画像は、フォトショップ9.0(アドビ)を使用してコントラストを微調整してトリミング、結合および最適化し、Canvas X(Deneba Software)を使用してレイアウトした。

J Exp Med 205:799-810)に従って、PBSで心臓内灌流を行い、解剖により網膜を取り出し、単一細胞懸濁液を調製した。蛍光色素標識モノクローナル抗体(mAb)をBD社、BioLegend社、eBioscience社またはAbDSerotec社から購入し、製造元のプロトコルに従って使用した。使用した蛍光色素標識モノクローナル抗体は、PEで標識した抗CD11b抗体、抗CD206(MMR)抗体および抗IL-4Rα抗体;PerCP-cy5.5で標識した抗Ly6C抗体および抗CD11b抗体;アロフィコシアニン(APC)で標識した抗CD115抗体、抗TCRβ抗体、抗FoxP3抗体および抗CD204抗体;Alexa647で標識した抗デクチン−1抗体;ならびにPacific Blue/Brilliant Violetで標識した抗TCRβ抗体、抗CD4抗体および抗CD45.2抗体であった。製造元のプロトコルに従ってFoxp3染色バッファーセット(eBioscience)を使用してFoxP3染色を行った。FACSDivaソフトウェアを使用してFACS LSRIIサイトメーター(いずれもBD社)により細胞を分析した。FlowJoソフトウェア(Tree Star社)で分析を行った。いずれの実験においても、関連する陰性コントロール群および陽性コントロール群を用いて、目的とする集団を決定し、それ以外の集団は除外した。
マウスを麻酔し、プラスチック器具を使用して軽く固定する。網膜を可視化するため、0.5%トロピカミド(Dr. Fischer)を1滴滴下して瞳孔を広げ、眼用潤滑剤(Celluspan;Dr. Fischer)を1滴滴下して眼球上にガラス製カバースリップを載せる。マウスにローダミンデキストラン(マウス1匹につき1mg;シグマアルドリッチ)を静脈内注射して血管を可視化した。蛍光照明装置およびPixelfly QE電荷結合素子カメラ(PCO)を備えたZoom Stereo Microscope SZX-RFL-2(オリンパス)の観察野にマウスを置く。赤色フィルターセットの励起波長は510〜550nmであり、発光波長は590nmである(ロングパス)。緑色フィルターセットの励起波長は460〜490nmであり、発光波長は510〜550nmである。蛍光露光時間は50ミリ秒とする。画像の取得はCamwareカメラ制御ソフトウェアプログラム(PCO)を使用して行う。画像分析はImageJ 1.43ソフトウェア(W. Rasband、アメリカ国立衛生研究所、メリーランド州ベセスダ)を使用して行う。
データ分析では、スチューデントのt検定を用いて2群を比較する。複数の群の比較には一元配置分散分析を用いる。帰無仮説が否定された場合、フィッシャーのLSD検定により各群を一対比較して追跡を行う(F<0.05)。結果は平均値±SEで示す。グラフ中、y軸のエラーバーはSEを表す。ぶどう膜炎に関する研究では、Levene検定により等分散性を調べた。等分散であった場合、2群の比較にはスチューデントのt検定を用い、複数の群の比較には一元配置分散分析を用いてデータの分析を行った。帰無仮説が否定された場合、TukeyのHSD検定により各群を一対比較して追跡を行った(p<0.05)。不等分散であった場合、データを対数変換して可能であれば等分散とし、等分散とすることができなかった場合、Kruskal-Wallis検定を用いて複数の群を比較し、続いてDunn検定を行った。結果は平均値±SEで示す。グラフ中、y軸のエラーバーはSEを表す。
ヒト単球は、CD14およびCD16の発現に基づいて3つの集団、すなわち、CD14+CD16-単球集団、CD14+CD16+単球集団、およびCD14dimCD16+単球集団に分けられる。CD14+CD16-単球は血中単球の80%〜90%を占め、ケモカイン受容体CCR2を高レベルで発現し、ケモカイン受容体CX3CR1(フラクタルカインの受容体)を低レベルで発現する。この主要なサブセットとは対照的に、ヒトCD16+単球はCX3CR1を高レベルで発現し、CCR2を低レベルで発現する(Cros et al., 2010)。Crosら(2010)によれば、遺伝子発現分析により、ヒトCD14dimCD16+単球とマウスパトロールGr1dim単球との類似性が示された。CD14dimCD16+細胞は局所組織の自然免疫監視および自己免疫疾患の病態形成に関与する真性単球である。
(i)ヒト末梢血からの単核細胞の単離:
健常なドナーから採取した新鮮血(8ml)をPBS中2.5%FCSで1:1に希釈し、フィコール勾配(Ficoll-Paque plus、アマシャムバイオサイエンス)上に重層した。チューブを1000g、20℃で20分間遠心分離した。単核細胞相を採取し、PBSで2回洗浄した。
FcRブロッキング試薬(細胞106個あたり2.5μl)(130-059-901、ミルテニーバイオテク)を使用して、単核細胞を室温で15分間処理することにより、Fc受容体をブロッキングした。次いで、洗浄せずに、抗ヒトCCR2モノクローナル−ビオチン試薬(FAB151B、R&Dシステムズ)(細胞106個あたり10μl)を添加し、2〜8℃で35分間インキュベートした。細胞を冷MACS(登録商標)バッファー(PBS中、1mM EDTAおよび2%FCS)で洗浄し、ストレプトアビジンマイクロビーズ(130-048-101、ミルテニーバイオテク)(細胞107個あたり20μl)を添加し、2〜8℃で20分間インキュベートした。細胞を洗浄し、MACSバッファー0.5ml中に再懸濁した。製造元のプロトコルに従ってLDカラム(130-042-901、ミルテニーバイオテク)を使用し、CCR2+細胞を除去した。
上記細胞をMACSバッファー(細胞107個あたり80μl)中に再懸濁し、CD14+マイクロビーズ(130-050-201、ミルテニーバイオテク)(細胞107個あたり20μl)を添加し、2〜8℃で15分間インキュベートした。次いで、細胞を洗浄し、MACSバッファー0.5ml中に再懸濁した。製造元のプロトコルに従って、LSカラム(130-042-401、ミルテニーバイオテク)を使用した磁気分離を行い、CD14+細胞をポジティブ選択した。
各試料を製造元のプロトコルに従って以下のように染色した。各試料を70μmナイロンメッシュに通し、FCRブロッキング試薬(細胞106個あたり30μl)(130-059-901、ミルテニーバイオテク)を用いて室温で15分間ブロッキングした。製造元のプロトコルに従って蛍光色素標識抗ヒトモノクローナル抗体を使用した。使用した抗体は、PerCP標識抗CD45抗体(345809、BD)、FITC標識抗CD115抗体(FAB329F、R&Dシステムズ)、Pacific Blue(登録商標)標識抗CD14抗体(BLG-325616)、Alexa Fluor(登録商標)700標識抗CD16抗体(BLG-302026)、PE標識抗CX3CR1抗体(MBL-D070-5)、およびPerCP標識抗CCR2抗体(BLG-335303)であった。
いくつかの網膜変性モデルを選択して使用する。使用する網膜変性モデルは、神経変性条件下において一般的に見られる毒性メディエーターであるグルタミン酸塩を用いて網膜を毒性暴露させたEAU誘導;視神経挫滅;高IOPモデル;および色素上皮変性病態を最もよく模倣するトランスジェニックマウスである。
EAU誘導はヒト後部ぶどう膜炎のモデルである。ぶどう膜炎は網膜変性を生じることが多いため、このモデルをCNS神経変性のモデルとみなす。2.5mg/mlの濃度(最終濃度:1.25mg/ml)の結核菌H37.RA株を含有するフロイントの完全アジュバント(CFA)との1:1エマルションとして、ヒト光受容体間レチノイド結合タンパク質(IRBP)に由来するペプチド(1〜20番目の残基)(22)(AnaSpec社、GL Biochem (Shanghai)社、またはワイツマン研究所のペプチド合成ユニット)500μgをマウスに皮下注射する。これと同時に、マウスの腹腔内に百日咳毒素(シグマ)1μgを注射する。
CNS障害の多くにおいてグルタミン酸塩の上昇が報告されている。グルタミン酸塩は、興奮毒性化合物として、急性および慢性の変性障害(緑内障の視神経変性を含む)において最も一般的に見られる毒性メディエーターの1つである(Doble, 1999)。したがって、グルタミン酸塩は、中枢神経系神経の神経変性の全般的なモデルとなる。このモデルにグルタミン酸塩を眼内注射すると、網膜変性障害(加齢黄斑変性、網膜色素変性など)、前部虚血性視神経症、緑内障、またはぶどう膜炎といった様々な眼疾患において生じるような網膜神経節細胞の神経変性を特に典型的に示す。過去の報告(Yoles E, Friedmann I, Barouch R, Shani Y, Schwartz M. Self-Protective mechanism awakened by glutamate in retinal ganglion cells. J Neurotrauma. 2001, Mar; 18(3):339-4; Schori, H., E. Yoles, L. A. Wheeler, T. Raveh, A. Kimchi, and M.Schwartz. 2002. Immune-related mechanisms participating in resistance and susceptibility to glutamate toxicity. Eur. J. Neurosci. 16:557-564)に従って、マウスを麻酔し、局所麻酔(Localin;Dr. Fischer)を眼に直接投与することにより処置し、l−グルタミン酸塩(シグマアルドリッチ)400nmolを含有する生理食塩水を総量1μlで硝子体内注射する。
様々な動物種において、MNUを単回全身投与すると網膜変性が引き起こされる。この網膜変性は再現性が高く、また、ヒト網膜色素変性と類似したアポトーシスを介して、MNU投与後7日以内に光受容体細胞の喪失が起こる(Tsubura A, Yoshizawa K, Kuwata M, Uehara N. Animal models for retinitis pigmentosa induced by MNU; disease progression, mechanisms and therapeutic trials. Histol Histopathol. 2010 Jul;25(7):933-44. Review)。
適切に設定された視神経挫滅により、軸索損傷の逆行性シグナル伝達が生じて進行性の神経節細胞死が誘導される(Levkovitch-Verbin H, Harris-Cerruti C, Groner Y, Wheeler LA, Schwartz M, Yoles E. RGC death in mice after optic nerve crush injury: oxidative stress and neuroprotection. Invest Ophthalmol Vis Sci. 2000 Dec;41(13):4169-74)。すなわち、マウスを麻酔し、以下のようにして眼球から1〜2mmの視神経眼窩内部位に重篤な挫滅創を形成させる。手術用双眼顕微鏡を用いて結膜を切開し、視神経を露出させる。調整されたクロスアクション鉗子を使用し、血液供給を妨げないよう十分注意しながら、神経を2秒間挫滅させる。
眼圧(IOP)は緑内障の危険因子であり、網膜や視神経の変性を引き起こす場合があるため、一般に神経変性モデルとして、特に緑内障モデルとして使用される。過去の報告(Da, T., and A. S. Verkman. 2004. Aquaporin-4 gene disruption in mice protects against impaired retinal function and cell death after ischemia. Invest. Ophthalmol. Vis. Sci. 45:4477-4483; Ben Simon, G. J., S. Bakalash, E. Ophthalmol. 141:1105-1111)に従って虚血性損傷を作製する。すなわち、麻酔を行った後、等張食塩溶液(生理食塩水)を入れた容器と連結したマイクロピペットを前房内に挿入することによって眼圧(IOP)を上昇させる。この容器を適切な高さに配置することにより、120mmHgの眼圧を60分間にわたって誘導する。

Molday, Shizuo Mukai, Glenn S. Cowley, Eliot L. Berson, Thaddeus P. Dryja. Transgenic mice with a rhodopsin mutation (Pro23His): A mouse model of autosomal dominant retinitis pigmentosa. Neuron (1992) 9:815-830)が挙げられる。
移植用の単球を同系マウスの骨髄から単離した。抗CD3抗体、抗CD19抗体および抗CD56抗体を結合させたマイクロビーズでPBMCを標識し、T細胞、B細胞およびNK細胞を除去した。磁気カラムを通過した非標識細胞を採取した。この画分を抗CD16マイクロビーズで標識し、再び上記磁気カラムに載せた。カラムを通過した細胞を採取し、CD14蛍光抗体およびCD16蛍光抗体で染色してFACS分析を行った。PBMCのうち、生細胞全体の19%は単球(CD14+)である。これらの単球のうち約10%はCD16+である。最終生成物中、生細胞の約90%は単球であり、そのうちCD16+は0.1%未満である。
12時間の周期で明期と暗期とを繰り返す3000ルクスの光にアルビノラットを暴露させ、これを1ヶ月間、3ヶ月間または6ヶ月間継続する。安楽死させる前に、眼底検査、眼底写真撮影、フルオレセインおよびインドシアニングリーンによる血管造影、ならびに光コヒーレンストモグラフィーを行う。光暴露させたラットを1ヶ月後、3ヶ月後または6ヶ月後に安楽死させ、病理組織学的評価を行う。4−ヒドロキシ−2−ノネナール修飾タンパク質およびニトロチロシン修飾タンパク質の存在について、免疫蛍光染色により網膜を調べる(Daniel M. Albert et al., 2010. Development of choroidal neovascularization in rats with advanced intense cyclic light-induced retinal degeneration. Arch Ophthalmol. 2010;128(2):212-222)。また、損傷発症後の様々な時点において、光暴露させたラットを自己由来単球の単回注射により上記と同様にして処置する。各動物モデルにつき、(i)硝子体内注射および(ii)網膜下注射の2種の投与経路ついて調査する。この研究は、形態学的評価基準または網膜断面の生存細胞成分のカウント数に基づいた網膜神経細胞の生存の評価をエンドポイントとする。
AMDおよびアルツハイマー病はいずれも、かなりの割合の高齢者が罹患する慢性神経変性疾患である。これらの疾患は不可逆的な機能喪失を特徴とし、これに対する治療法は存在しない。AMDおよびアルツハイマー病において生じる変性の病態形成にはある程度の共通点がある(Klaver et al.,1999. Is age-related maculopathy associated with Alzheimer’s disease? The Rotterdam study. Am J Epidemiol;150:963-8)。AMDおよびアルツハイマー病の病因はほとんど分かっていないが、これらの疾患の病態形成は驚くほど類似している。AMDの初期における組織病理学的所見は細胞外におけるドルーゼンの沈着および基底膜沈着である(Hageman, GS. & Mullins, RF. Molecular composition of drusen as related to substructural phenotype. Mol Vis 5, 28 (1999))。これらの病変は、脂質、糖タンパク質およびグリコサミノグリカンを含有し、変性しつつある神経網膜に由来すると考えられる。ドルーゼン沈着物の蓄積は、光受容体の喪失およびそれに続く黄斑機能の低下と関連している(Holz et al., Bilateral macular drusen in age-related macular degeneration. Neuron 6, 487-98)。老人斑は様々な成分から構成されており、そのなかには、アミロイド前駆体タンパク質として知られる膜貫通型ポリペプチドファミリーのタンパク質分解切断により生じる小さなペプチドが含まれる。2種のペプチドがアルツハイマー病の病態形成の主因として広く認められており、これらはアミロイド−β(Aβ)ペプチドとして知られている。アミロイド沈着物とドルーゼンとでいくつかの成分が共通しており、このような成分として例えばビトロネクチン、アミロイドP、アポリポタンパク質Eなどのタンパク質が挙げられ、さらには、アルツハイマー病のアミロイド斑に関連するAβペプチドおよびアミロイドオリゴマーもこのような成分に含まれる(Luibl et al., 2006. プロトコル:
マウスを麻酔し、L−グルタミン酸塩(GT)(生理食塩水1μl中)400nmolを右眼に硝子体内注射した。翌日、マウス由来のCD115+骨髄単核細胞(BMMC)を硝子体内注射用細胞として精製した。すなわち、(cx3cr1骨髄プロモーターの制御下にGFPレポーターを有する)ドナーマウスの大腿骨および脛骨から骨髄細胞を採取し、フィコール密度勾配により単核細胞を濃縮した。目的の集団をポジティブ選択するためのビオチン化抗CD115抗体およびストレプトアビジン結合磁気ビーズ(ミルテニーバイオテク)を使用した磁気細胞分離(MACS)システムにより、CD115+BM単球集団を単離した。GTの毒性に暴露させた眼にマウス1匹につき(PBS1μl中)105個の単球を(前日にGTを注入した孔から)硝子体内注射した。コントロール群にはPBSを注射した。
硝子体内に注射したマウス単球はRGCに対して保護効果を示した(図1A〜C)。単球は、注射した眼の硝子体内と神経節細胞層(損傷を与えたRGCが存在する部位)の近傍に検出された。また、いくつかの単球は網膜下腔でも検出できた(図2)。
損傷神経系部位への単球の動員を増強する方法が重要ではないこと、および網膜の損傷だけでなく視神経の損傷も治療できる可能性があることを示すため、CNS特異的抗原を用いた免疫化により内在性細胞の視神経損傷部位への遊走を促した。
[Cx3cr1GFP/+→WT]BMキメラを以下のようにして作製した。WTレシピエントマウスの全身に致死量の放射線(950rad)を照射した。この際、頭部を遮蔽して、網膜への直接的な損傷と、視神経挫滅(ONC)により誘導される骨髄系細胞以外の骨髄系細胞による浸潤を防いだ。翌日、Cx3cr1GFP/+トランスジェニックマウスに由来する骨髄細胞5×106個を尾静脈に注射することによって、マウスを新たに作製した。得られたマウスにおいては、血液から浸潤した単球のみがGFP標識を有する。
ONCにより網膜中の総白血球数(CD45+)が増加し、この白血球の増加は、45Dをワクチン接種した損傷マウスにおいて増強された(図3A)。
プロトコル:
損傷網膜における末梢単球数の増加に対するワクチン接種の効果と、視神経挫滅損傷後のRGCの生存率との相関関係を以下の実験により証明することができる。2.5mg/mlのフロイントの完全アジュバント(Difco)にMOG改変ペプチド45D(45番目のセリンがアスパラギン酸に改変されたミエリンオリゴデンドロサイト糖タンパク質(MOG)35−55)100μgを乳化させたエマルション100μlを1群のマウスに皮下注射する。
Claims (20)
- 網膜または視神経において、神経細胞変性の抑制、グルタミン酸塩の毒性からの神経細胞の保護または神経再生の促進に使用するための末梢血単核細胞(PBMC)由来単球亜集団であって、
該単球亜集団が、CD3+細胞、CD19+細胞およびCD56+細胞を相対的少量しか含んでいないか、またはこれらの細胞を実質的に含んでいないこと、ならびに
該網膜または視神経が、眼疾患、眼障害または眼病態による損傷を有することを特徴とするPBMC由来単球亜集団。 - CD16+細胞が実質的に含まれない、請求項1に記載のPBMC由来単球亜集団。
- 網膜または視神経において、神経細胞変性の抑制、グルタミン酸塩の毒性からの神経細胞の保護または神経再生の促進に使用するためのPBMC由来単球亜集団であって、
該単球亜集団が、CD3+細胞、CD19+細胞、CD56+細胞およびCD16+細胞を相対的少量しか含んでいないか、またはこれらの細胞を実質的に含んでいないこと、ならびに
該網膜または視神経が、眼疾患、眼障害または眼病態による損傷を有することを特徴とするPBMC由来単球亜集団。 - CD14+細胞が実質的に濃縮されている、請求項1または3に記載のPBMC由来単球亜集団。
- 前記疾患、障害または病態が、加齢黄斑変性または網膜色素変性から選択される網膜変性障害;前部虚血性視神経症;緑内障;およびぶどう膜炎から選択される、請求項1〜4のいずれか一項に記載のPBMC由来単球亜集団。
- 炎症性眼疾患、炎症性眼障害または炎症性眼病態の治療に使用するための末梢血単核細胞(PBMC)由来単球亜集団であって、
該単球亜集団が、CD3+細胞、CD19+細胞およびCD56+細胞を相対的少量しか含んでいないか、またはこれらの細胞を実質的に含んでいないことを特徴とするPBMC由来単球亜集団。 - CD16+細胞が実質的に含まれない、請求項6に記載のPBMC由来単球亜集団。

該単球亜集団が、CD3+細胞、CD19+細胞、CD56+細胞およびCD16+細胞を相対的少量しか含んでいないか、またはこれらの細胞を実質的に含んでいないことを特徴とするPBMC由来単球亜集団。 - CD14+細胞が実質的に濃縮されている、請求項6または8に記載のPBMC由来単球亜集団。
- 炎症性眼疾患、炎症性眼障害または炎症性眼病態による損傷を有する網膜または視神経において、神経細胞変性の抑制、グルタミン酸塩の毒性からの神経細胞の保護または神経再生の促進を行うための、請求項6〜9のいずれか一項に記載のPBMC由来単球亜集団。
- 前記疾患、障害または病態が、加齢黄斑変性または網膜色素変性から選択される網膜変性障害;前部虚血性視神経症;緑内障;およびぶどう膜炎から選択される、請求項10に記載のPBMC由来単球亜集団。
- 強膜炎、角膜炎、角膜潰瘍、雪眼炎、タイゲソン点状表層角膜症、角膜血管新生、フックスジストロフィー、円錐角膜、乾性角結膜炎および虹彩炎から選択される炎症性眼疾患を治療するための、請求項6〜9のいずれか一項に記載のPBMC由来単球亜集団。
- ヒトPBMCである、請求項1〜12のいずれか一項に記載のPBMC由来単球亜集団。
- 自己由来のPBMCである、請求項13に記載のPBMC由来単球亜集団。
- 同種異系のPBMCである、請求項13に記載のPBMC由来単球亜集団。
- 注射用製剤である、請求項14または15に記載のPBMC由来単球亜集団。
- 眼内注射用製剤である、請求項16に記載のPBMC由来単球亜集団。
- 請求項1〜17のいずれか一項に記載のPBMC由来単球亜集団を含む組成物。
- 網膜または視神経において、神経細胞変性の抑制、グルタミン酸塩の毒性からの神経細胞の保護または神経再生の促進を行うための方法であって、
該網膜または視神経が、眼疾患、眼障害または眼病態による損傷を有し、
該方法が、上記抑制、保護または促進を必要とする個体に請求項1〜17のいずれか一項に記載のPBMC由来単球亜集団を有効量で投与することを含む方法。 - 炎症性眼疾患、炎症性眼障害または炎症性眼病態を治療するための方法であって、
該治療を必要とする個体に請求項6〜17のいずれか一項に記載のPBMC由来単球亜集団を有効量で投与することを含む方法。
