Abstract
Description
重鎖可変領域(VH)アミノ酸配列(配列番号23)、及び軽鎖可変領域(VL)アミノ酸配列(配列番号24)、又は、
VHアミノ酸配列(配列番号27)、及びVLアミノ酸配列(配列番号28)を含む単離抗体と競合する。
本発明は、ヒトインターフェロンオメガ(IFNω)及び複数のヒトインターフェロンα(IFNα)のサブタイプ(IFNα/ω抗体)と結合し、その活性を中和するモノクローナル抗体を提供する。本発明は、少なくとも部分的に、本発明のIFNα/ω抗体が結合する、IFNωと、IFNαサブタイプと、が共有する最小限の中和性のエピトープの同定に基づく。本発明のIFNα/ω抗体は、IFNαサブタイプを中和するがIFNωを中和しない抗体よりも、I型IFN及びIFNシグネチャのSLE関連の調製物の中和においてより強力であり、したがって、IFNα及びIFNωの産生の増加に関連する例えば免疫媒介炎症性疾患のような任意の疾患の治療において、より効能が優れている可能性がある。本発明のIFNα/ω抗体はIFNβを中和しないので、全てのI型IFNを遮断することが予測される抗IFNAR療法と比較して、より好ましい安全性及びPK特性を有する可能性がある。本明細書で使用するとき、「IFNα/ω抗体は、本明細書に例示するINFω及び複数のIFNαサブタイプと結合し、中和する抗体を指す。

本発明のα/ω抗体を使用して、IFNα及びIFNωの産生の増加に関連する任意の疾病を治療又は予防することが可能である。本発明の方法において、本発明の任意のIFNα/ω抗体を使用することが可能である。あるいは、ヒトIFNω及びヒトIFNαサブタイプIFNαB2、IFNαF、IFNαG及び/又はIFNαJ1と結合するために、配列番号23の重鎖可変領域(VH)アミノ酸配列並びに配列番号24の軽鎖可変領域(VL)アミノ酸配列、又は配列番号27のVHアミノ酸配列並びに配列番号28のアミノ酸配列を含む単離された抗体と競合する任意の抗体を使用してもよい。更に、配列番号1の残基F27、L30、及びR33の1つ以上でIFN IFNωと結合し、配列番号19の残基F27、L30、及びR33の1つ以上でIFNα4aと結合する任意の抗体を使用してもよい。
IFNα及びIFNωの産生の増加に関連する状態の処置に有効な本発明のIFNα/ω抗体の「治療的有効量」は、標準的な研究手法によって決定することができる。例えば、SLEのような免疫媒介炎症性疾患の処置において有効であろう本発明のIFNα/ω抗体は、当該技術分野で周知の関連する動物モデルにIFNα/ω抗体を投与することによって決定することができる。
以下に、本明細書の他の箇所に記載した開示にしたがった本発明の番号付けした更なる特定の実施形態を列挙する。上記の本発明の実施形態の特徴は、これらの番号付けされた更なる実施形態の各々及び全てにもまた関係する。
1)(a)ヒトインターフェロンオメガ(IFNω)及び少なくとも4つ、5つ、6つ、7つ、8つ、9つ、又は10のヒトインターフェロンアルファ(IFNα)サブタイプと結合し、それらの活性を中和する、単離されたモノクローナル抗体。

以下を含む、12の個別の組換えヒトI型IFNα(αA(α2a)(配列番号5)、αB2(α8)(配列番号6)、αC(α10)(配列番号7)、αD(α1)(配列番号8)、αF(α21)(配列番号9)、αG(α5)(配列番号10)、αH2(α14)(配列番号11)、αI(α17)(配列番号12)、αJ1(α7)(配列番号13)、αK(α6)(配列番号14)、α4b(α4)(配列番号15)、αWA(α16)(配列番号16))、及びチンパンジーIFNω(chimp IFNω)(配列番号3)を、配列番号17〜21のようなシグナル配列を用い、標準的な方法を用いて、HEK 293細胞において発現した。発現レベル及び可溶性を改善するために、ヒトIFNωの80位のIFN−ω(T80E)で単一のアミノ酸変異体を生成し、HEK 293細胞において発現した。T80E IFNω変異体(配列番号2)は、野生型タンパク質と同等の活性を有していた。
マウスの接種
C2595の生成
ヒトIFN−α、チンパンジーIFN−ω、及びカニクイザルIFN−ωの混合物をBALB/cマウスに複数回腹腔内接種した。0日目にマウスにチンパンジーIFN−ωを接種した。14日目に、同じマウスにチンパンジー及びカニクイザルのIFNω、IFNαD、IFNαJ1、IFNαC、IFNαB2、IFNαH2、IFNαA、IFNα4a、IFNαG、IFNαF、IFNαWA及びIFNαIの混合物を接種した。208日目に、同じマウスにカニクイザルIFN−ω、ヒトIFNα4b、IFNαA、IFNαD、及びIFNαKの混合物を接種した。221日目に、同じマウスにカニクイザルIFN−ω、ヒトIFNαJ、IFNαI、IFNα4a、IFNαA及びIFNαF.の混合物を接種した。接種後、特異的IgG力価を評価した。十分な力価が得られた直後に、膵細胞を単離してFO細胞と融合した。得られたハイブリドーマを96ウェルプレートに播種して10日間培養した。まず、チンパンジーIFN−ωの結合及びヒトIFNαA、IFNαH2、IFNαD、及びIFNα4aの混合物の結合に関して、ELISAを用いて一次スクリーンによって抗原特異性クローンを同定した。IFN−α及び/又はIFN−ωへのハイブリドーマの結合は、Luminex多重アッセイによって更にスクリーニングした。更なる研究のために、IFNα及びヒト並びにカニクイザルのIFNωのほとんどに広域結合するクローンを選定した。
ヒトI型IFN結合性Fabは、de novo pIXファージディスプレイライブラリから選択した(Biol.397:385〜396、2010;国際特許出願公開第WO2009/085462号、米国特許公開第US2010/0021477号、米国特許公開第US2012/0108795号)。
pIXファージディスプレイライブラリは、ヒト野生型IFN−α配列C末端ポリヒスチジンタグの発現から生成され、固定化金属アフィニティークロマトグラフィーにより精製した、精製されたI型IFNαA(IFNα2)に対してパニングした。3ラウンドのパニングを用いた。第1、第2、及び第3ラウンドのパニングに、それぞれ、100nM、10nM及び1nMのビオチン化抗原を使用した。3ラウンドのパニング後に採取したファージミドクローン由来のモノクローナルFabについて、標準のELISAを用いて、それらが結合するチンパンジーIFNω、ヒトIFNα2、IFNα1、IFNαH2、IFNαG、IFNα F及びアビジンに関して一次スクリーニングした。ELISAにおいて、IFNα及びIFN−ωに特異的に結合したFabフラグメント(Fab)を配列決定し、それらが異なるV領域配列を有している場合には、一意のIFNバインダーとして同定した。それらのFabを、ヒトIgG1 mAbに変換し、抗炎症活性の同定と関係する細胞ベースの一連のアッセイにおいてそれらの中和活性を更に試験した後、IFNバインダーとして同定した。
pIXファージディスプレイライブラリは、ヒト野生型IFN−α配列C末端ポリヒスチジンタグの発現から生成され、固定化金属アフィニティークロマトグラフィーにより精製した、精製されたI型IFNαG(a5)に対してパニングした。3ラウンドのパニングを用いた。第1、第2、及び第3ラウンドのパニングに、それぞれ、100nM、10nM及び1nMのビオチン化抗原を使用した。3ラウンドのパニング後に採取したファージミドクローン由来のモノクローナルFabについて、標準のELISAを用いて、それらが結合するチンパンジーIFNω、ヒトIFNα2、IFNα1、IFNαH2、IFNαG、IFNα F及びアビジンに関して一次スクリーニングした。ELISAにおいて、IFNα及びIFN−ωに特異的に結合したFabフラグメント(Fab)を配列決定し、それらが異なるV領域配列を有している場合には、一意のIFNバインダーとして同定した。それらのFabを、ヒトIgG1 mAbに変換し、抗炎症活性の同定と関係する細胞ベースの一連のアッセイにおいてそれらの中和活性を更に試験した後、IFNバインダーとして同定した。
方法
親和性の測定
抗体の結合親和性は、ProteOn(Bio−Rad Hercules、カリフォルニア州)でSPR技術を用いて行われた。製造業者の推奨した標準NHS/EDC化学を用いて、ヤギの抗ヒトFc抗体(製造)をGLCチップ(Bio−Rad Hercules、カリフォルニア州)にアミンカップリングした。次に、抗IFN mAbを、抗体とカップリングしたチップに30μL/分の流量で2分間負荷した。50μL/mLの流量で2分間、ランニング緩衝液(緩衝液の組成)で洗浄した後、1:3希釈で100nM〜1.23nMまでの範囲の5つの異なる濃度で、組換えIFN抗原を3分間会合させ、10分間解離させた(どちらも50μL/mLの流量で)。チップは、異なる抗原の実行の間に各方向において100mMのリン酸を用いて生成した。ProteOnマネージャ(Bio−Rad Hercules、カリフォルニア州)を用いて、データ解析を行った。センサーグラムは、mAbによって分類した。(インタースポット又はブランクチャネル基準のいずれかを使用して)整列及び基準補正を適用した後、SPRデータを運動速度定数のラングミュアモデルにグローバルに適合させた(KD=koff/kon、式中、KD=平衡解離定数、kon=会合速度定数、koff=解離速度定数)。
完全に活性なI型IFNシグナル伝達経路を発現する(STAT2及びIRF9を安定に発現する)ように操作され、IFNα/β誘導性ISG54プロモーターの制御下でSEAPレポーター遺伝子でトランスフェクトされた、HEK−Blue(商標)IFN−α/β細胞(InvivoGen(カリフォルニア州San Diego)を使用した。細胞は、10%ウシ胎児血清、100ug/mLのブラストサイジン、及び30ug/mLのゼオシンでDulbeccoの改変イーグル培地中のコラーゲンI型コーティングされたT150フラスコにて、37℃、5%CO2で増殖した。細胞を採取し、384ウェルプレートに、1mLにつき50,000個の細胞で、各ウェルに50μLを播種した。播種した細胞を37℃、5%CO2で24時間インキュベートした。試験したインターフェロン試料は、調製し、使用済みのHEK ISRE無血清培地中で希釈し、IFN試料50μLを各ウェルに加えた。播種した細胞を37℃、5%CO2で20時間インキュベートした。室温で20分間インキュベートした後、60μL/ウェルのQUANTI−Blue(商標)を濾過水中に再縣濁した20μLの播種した細胞上清から、アルカリホスファターゼが検出された。Biotek Synergyプレートリーダーで650nmで光密度を読み取った。
健康なボランティアからのヘパリン化全血を、アイソタイプ対照と共に、いくつかの異なるI型IFN阻害剤を含有する96ウェルU底プレートに播種した。阻害剤及び適切なアイソタイプ対照を、10% FBSを用いたRPMI培地にて希釈した。IFN及び阻害剤又はアイソタイプ対照を、10% FBSを含有する体積30μLのRPMI培地中に希釈した。それらの試料を15〜20分間予めインキュベートした後、その希釈液を含有するプレートにヘパリン化した全血240μLを加えて、270μLの最終体積を得た。試料を混合し、37℃で20〜22時間インキュベートさせた。インキュベーション後、試料を400Xgで5分間遠心分離し、血漿を回収し、後の分析のために凍結した。IP−10プロファイリングは、Milliplexサイトカイン/ケモカインキット(Millipore、Premixed 39プレックスで行った。試料の調製及びアッセイは、BioRadモデル(Bioplex(商標)200)システム及びBioplexマネージャ(商標)ソフトウェア4.1を用いてデータを収集し、製造業者の推奨に従って行った。統計解析はGraphパッドPrism V.5ソフトウェアにより行った。場合によっては、IP−10の定量化を、Qiagenの単一検体ELISAキットを用いて行った。
SLEドナーの全血をAsterandから市販により入手した。全血遺伝子発現解析は、IFN刺激遺伝子(ISG)が濃縮されたプライマー及びプローブを含有するカスタムTaqMan低密度アレイカードを用いて行った。アレイに含まれていた遺伝子は以下の通り。ACTB、IL6、IL10、IL13、FAS、IL15、IL21、IL17A、EIF2AK2、OASL、18S、STAT1、LY6E、PLSCR1、MX1、IFIT1、IFI44、IFI44L、IFI27、ISG15、RSAD2、CXCL10、LAG3、及びTNFSF10。製造業者の使用説明書に従って、ABI Prism 7900 HT Sequence Detectionシステム(Applied Biosystems、米国カリフォルニア州)でRT−PCR増幅を行った。相対的発現値を、比較閾値サイクル(Ct)法を用いて算出した。端的に述べると、この手法は、2-ΔΔCtを用いて、較正群(健常な未処置の全血)に対して正規化した標的遺伝子の発現を算出する。ベータアクチン(ACTB)を内在性対照として選択した。閾値サイクル(Ct)は、その量の増幅した標的が一定の閾に達するサイクル数を示す。全てのインターフェロン誘導標的遺伝子及びACTBのCtデータを用いて、ΔCt値[ΔCt=Ct(標的遺伝子)−Ct(ACTB)]を得た。ΔΔCt値は、対照群(5人の健常な未処置の全血ドナー)の平均を各標的のΔCt値から減算して算出した。相対発現値は、等式2-ΔΔCを用いて算出した。本実験の3人のSLEドナーは、低密度アレイの9つのISGに関して少なくとも対照群の2倍の高さの遺伝子発現を有していた。3人の全てのドナーに対するI型IFN阻害剤の作用を比較するために、最初に、あらゆる治療/阻害剤について以下の式を用いて阻害率(%)を決定した。
(2-ΔΔCt SLE未処置血液−2-ΔΔCt阻害剤/-ΔΔCt SLE未処置血液)×100=阻害率(%)。次に、3人の全てのドナーに対する各処置についてのベースライン(%)を以下の等式によって算出した。100−阻害率(%)=ベースライン(%)。
SLE患者の血漿をSCIPAC(Kent、イギリス)から入手した。ISREに基づくレポーター遺伝子アッセイによって、I型IFN活性を有する血漿試料を更にIgG精製に使用した。IgGは、製造業者(Thermo SCIENTIFIC)の推奨するNAB(商標)タンパク質A/Gスピンカラムを用いて精製し、濃度を決定するためにタンパク質アッセイ(Pierce BCA)を実行した。1Xリン酸緩衝生理食塩水(PBS)中に5×107細胞/mLで懸濁したHEK293T細胞を用いて、自己抗原溶解物を調製した。Hek293−T細胞を破壊するために、−80℃で少なくとも10分間(ただし最初の凍結は少なくとも30分間)凍結させてから37℃で融解する凍結融解サイクルを4回行い、この凍結融解後、細胞の破片を遠心分離により除去し(5分間400g)、可溶性抗原の量をタンパク質アッセイによって定量した。1:1の比率で、精製したIgGと壊死細胞の溶解物とをいっしょに30分間RTでインキュベートして、免疫複合体を形成した。次いで、各ウェルにつき合計4mLのPBMC培地において、400μg/mLの最終濃度の免疫複合体を6ウェルプレートの3つのウェルに加えた。上述のように健康ドナーのIgGを精製及び「複合化」し、対照として使うためにPBMCを刺激するために用いた。これらの研究からの馴化培地をアリコートし、阻害実験のための内因性IFNのためのソースとして使用した。健康ボランティアから単離されたIgGから調製した製剤では、IFN活性は認められなかった。
組換えI型IFNの親和性及び中和
表2は、個々の組換えヒトI型インターフェロン(IFN)に対する抗体M43、M88、及びM3239の解離定数(KD)を示す。M43、M88、及びC2595は、少なくとも4つのIFNα分子(アルファB2、アルファF、アルファG及びアルファJ1)を中和した。アルファDについては、結合は認められなかった。
内因性I型IFNを中和する抗体の能力は、内因性白血球IFN(ウイルス誘導した)又は組換えIFNωで刺激したヒト全血からのケモカインIP−10(CXCL10)の放出を評価するアッセイで評価した。白血球IFN(図1A)及び組換えIFNω(図1B)の両方の用量依存性阻害が、M43及びC2595に認められ、ウイルス誘導された白血球によって精製されるような広範なスペクトルのI型の活性を中和する抗体の能力が示された。内因性白血球IFNはSigma(カタログ番号I4784−1MU)から購入した。
SLEに存在するI型IFNミリューをより正しく代表するために、刺激としてのSLE免疫複合体誘導したIFNを減少する抗体の能力について、抗体を試験した。SLE免疫複合体誘導したIFNは、SLE患者由来の免疫複合体でヒトPBMCを刺激することによって調製し、阻害剤mAb及び対照mAbの存在下でI型IFN誘導性のレポーター遺伝子のアッセイ(上記のISREレポーター遺伝子アッセイ)に、この条件付けした培地を使用した。選択的IFNω中和性のmAbを3つの異なるIFNαアンタゴニストとの組み合わせにおいて添加したところ、等量のアイソタイプ対照mAbの存在下での抗IFNα mAbと比べて、免疫複合体誘導したIFNの総活性は更に低下した(図2)。更に、抗IFNα/ω mAb M43は、IFNα阻害剤mAbと比べて、更なる活性の抑制を示し、IFNα及びIFNωの両方の遮断が、IFNαの中和のみの場合よりも総IFN活性を低下し得ることを示した(図2)。
上述のように、9つのIFN誘導した遺伝子の組み合わせを用いて、SLE遺伝子シグネチャのアッセイを開発した。遺伝子シグネチャを阻害する多様な抗体の能力を試験した。IFNω特異性アンタゴニストmAb(抗−ω)は同等の濃度のアイソタイプ対照mAbと比べてIFNシグネチャを下方調節し、IFN−ωがSLEにおいてIFNシグネチャを誘導する活性のI型IFNミリューの一部であることを示した。IFNω抗体とIFNα抗体との組み合わせは、IFNα又はIFNω阻害剤単独よりも、これらの患者の血液中で永続するIFNシグネチャのより顕著な抑制をもたらした(図3)。
エピトープ結合実験は、Octed(ForteBio、カリフォルニア州Menlo Park)を用いてリアルタイム無標識競合結合アッセイを使用して行った。イン・タンデムアッセイ形式及び古典的サンドイッチアッセイ形式の2つのアッセイ形式を使用した。
IFWM43(以下、mAbについてはM43、FabについてはFabM43)は、ヒトIFNα分子及びIFNωを広域に中和し、多くのIFNαサブタイプ及びヒトIFNωとの結合を示す。IFNαサブタイプ及びIFNωに対するその特異性に関する構造的ベースを明らかにするために、FabM43との複合体におけるIFN−ωの結晶構造を決定した。
タンパク質
Hisタグ付けしたFabM43(IgG1/κアイソタイプ)及びT80E変異(本例においてIFNω及びIFNωT80Eは同義)をHEK293F細胞にて発現し、アフィニティ及びサイズ排除クロマトグラフィーを用いて精製した。タンパク質は、20mMのトリス、pH 7.4、50mMのNaClにおける量である。
この複合体は、1.05:1.0(過剰のIFNω)のモル比でIFNωをFabM43と混合することによって調製し、20mMの酢酸Na(pH 5.5)、0.1M NaCL、及び10%グリコールで平衡したSuperdex 200カラムで精製した。精製した複合体を、Amicon−Ultra 10kDaカットオフを用いて10.24mg/mLに濃縮した。X回折に適した結晶は、記載されているように(Obmolovaら、Acta Crystallogr D Biol Crystallogr 66:927〜33、2010)、MMS播種した26%のPEG 3350、1MのLiCL、0.7%の1−ブタノールからのシッティングドロップにて得た。
X線データ収集については、結晶を、20%グリセロールを補充した合成母液(0.1MES(pH 6.5)、20% PEG 3350、1M LiCL)に数秒間浸し、液体窒素中でフラッシュ凍結させた。X線回折データはSwiss Light Sourceで収集した。X線データはプログラムXDS(Kabsch、Acta Crystallographica 66:125〜132、2010)で処理した。X線データ統計を表6に示す。

da*、b*及びc*における異方性の解像限界は、回折異方性スケールサーバによれば、3.0、2.5、及び2.5Åである(http://_services_mbi_ucla.edu/_anisoscale/)。回折データ統計は、これらの限界を用いた、異方性の切り捨て及びスケーリング後のデータセットに関する。
全体構造
非対称単位に6つのIFNω/FabM43複合体が存在する。これらの複合体の全てが非常に類似している。IFNω/FabM43複合体の全体的な分子構造を図4に示す。図面の説明Hの標識はVH、Lの標識はVL、左上はIFNωである。
M43は、ABループ(R22とQ40の間)の残基及び残基K134、M146、M149、K150、F153並びにへリックスEのL154で構成された立体配座エピトープを認識する(表7)。パラトープは6つのCDRのうち5つからの残基で構成されている。パラトープ残基は主に疎水性であり、一連のポケットを形成し、そこに、短いABへリックスの残基F27、L30、K31及びR33の側鎖がドッキングする。抗体と抗原の相互作用は、ほとんどがvdw及び疎水性パッキングのように思われる。抗体と抗原の間には少数のH結合しかなく、それらのほとんどにバックボーン−バックボーン又は側鎖−バックボーンの相互作用が関与している。Ab/Ag相互作用の大部分は、ABへリックスのいくつかの残基F27、L30、K31及びR33によるものである。したがって、IFNωのこの領域がエピトープの主な部分を構成しているように見える。
IFNAR1及び/又はIFNAR2との複合体中のIFNα/ωの結晶構造は最近報告されている(Thomasら、Cell 146:621〜632、2011)。M43/IFNα4の構造とIFNω/IFNAR1/IFNAR2複合体との比較は、M43重鎖とIFNAR2の明らかなオーバーラップを示している。したがって、M43はIFNAR2/IFN相互作用を遮断することによって中和する。
C2595(以下、mAbに関してはC2595、Fabに関してはFab357)は、複数のヒトIFN−α分子及びマウスハイブリドーマから得たIFNωを中和する抗体である。V領域をクローンし、ヒト重鎖及び軽鎖(IgG1κアイソタイプ)にキメラ化して、組換えFab357を生成した。IFN−ω/Fab357及びIFN−α4A/Fab357の結晶構造を決定した。
タンパク質
Hisタグ付けしたFab357(IgG1/κアイソタイプ)及びT80E変異ヒトIFNω(以下、IFNωT80E。IFNωと、T80Eを有するIFNωとは、本実施例では同義)をHEK293F細胞において発現させ、アフィニティ及びサイズ排除クロマトグラフィーを用いて精製した。タンパク質は、20mMトリス(pH 7.4)、50mM NaClにおける量である。)。IFNα4Aは、20mMトリス(pH 7.4)、50mM NaClにおいて、Crown Bioscience Inc.から入手した。
IFNα4A/Fab357複合体は、1.05:1.0のモル比でIFNα4AとFab357とを混合することによって調製し、0.1M NaCLを用いて、20mMのMES(pH 6.5)にて、SuperDex 200カラムで精製した。精製した複合体を5.5mg/mLに濃縮した。回折に適した結晶は、タンパク質溶液及び20% PEG 3350及び播種した0.2Mのクエン酸アンモニウムの等量の混合物を含むシッティングドロップ中で増殖した。
X線データ収集については、IFN−α4A/Fab357及びIFNω/Fab357の結晶を、20%グリセロールを補充した合成母液(それぞれに対応して、20% PEG 3350、0.2Mクエン酸アンモニウム、プレート10/20/11−MMS−A10;及び0.1MES(pH 6.5)、18% PEG 3350、0.2M LiCL、プレート12/21/2011−B11(R))中に数秒間浸漬し、液体窒素でフラッシュ凍結した。X線回折データは、それぞれ、Advance Photon Source of Argonne National Lab及びSwiss Light Sourceで収集した。X線データはXDSプログラムにより処理した。X線データ統計を表8に示す。

全体構造
両方の結晶構造中に、非対称単位に1つの抗原/抗体複合体が存在している。代表的な分子構造の全体を図5に示す。Fab分子は両方の結晶において非常に類似している。IFNα4Aの立体配座はIFNα4A/FabM88のそれと非常に類似している。IFNω/Fab357の構造については、IFNωのモデルは残基R22〜P39及びL118〜L154のみを含む。IFNωの不足している残基は、結晶パッキングがそれらの存在を排除するため、結晶の無秩序の結果ではない。明らかに、IFNωのタンパク質分解切断は、結晶化中に発生した。コールドボックス内でシッティングしている同じ複合体のその部分がいくらかのプロテアーゼ分解を受けていた証拠があるが、そのパターンは、結晶中で確認されたものと同一ではなかった(データは示されていない)。にもかかわらず、IFNωの結合領域はよく秩序だっている。

C2595はIFNAR2/IFN相互作用を遮断することによって中和する。
IFNα4Aとの複合体における抗−IFN抗体M88の結晶構造は2.5Åに対して決定した。主エピトープはIFNのABループのヘリカル要素A19〜D35である。M88の結合はIFNAR2の相互作用を防ぐであろう。したがって、M88はIFNAR2ブロッカーである。その構造はM88の結合の交差反応性を明らかにする。
タンパク質
Hisタグ付けしたFabM88(IgG1/カッパアイソタイプ)をクローンし、HEK293F細胞において発現し、アフィニティ及びサイズ排除クロマトグラフィーを用いて精製した。Fabは、20mMのトリス(pH 7.4)、50mMのNaClに受け取られた。IFNα4aは、(20mMのトリス(pH 7.4)、50mMのNaClにおいて)Crown Bioscience Inc.から入手した。
この複合体は、IFNα4AとFabM88とを、1.05:1.0(過剰のIFNα4A)のモル比で混合し、4℃で1時間インキュベートし、20mMのトリス(pH 8.0)、10%グリセロール、0.1MのNaClで20倍希釈してから、Amicon−Ultra 10kDaカットオフを用いて9.25mg/mLに濃縮することによって調製した。最初の結晶化は、IH1、IH2及びPEGスイート(Qiagen)で設定した。複合体の結晶化は、Oryx4ロボット(Douglas Instruments)を使用して、20℃で蒸気拡散法により行った。IH2#E12〜25% PEG 3K、0.2Mのクエン酸アンモニウムから結晶が現れた。これらの最初の結晶を用いて、結晶化シードを調製した。結晶の質を改善するために、IFNα4A/FabM88複合体を、20mmのMES(pH 6.5)、0.1MのNaCl、10%グリセロールで平衡したSuperdex 200カラム(GE Healthcare)で精製し、8.16mg/mLに濃縮した。X回折に適した結晶は、記載されているように(Obmolovaら、Acta Crystallogr D Biol Crystallogr 66:927〜935、2010)MMS播種した28%のPEG 3K、0.2Mのクエン酸アンモニウムから得た。
X線データ収集のために、1つの結晶を数秒間、20%グリコールを補充した母液に浸し、95Kで窒素のストリームにおいてフラッシュ凍結した。X線回折データは、Osmic(商標)VariMax(商標)共焦点光学系、Saturn 944 CCD検出器、及びX−stream(商標)2000極低温冷却システム(Rigaku、テキサス州)を備えたRIgAku MicroMax(商標)−007HFマイクロフォーカスX線ジェネレータを用いて収集した。回折強度は半分度画像で235度の結晶回転にわたって検出された。X線データをXDSプログラムにより処理した。X線データ統計を表10に示す。
全体構造
IFNα4A/FabM88複合体の全体的な分子構造を図6に示す。非対称単位にこれらの複合体は2つ存在する。2つの独立したIFNα4A分子の分子モデルは、一方に残基7〜102及び113〜160を、もう一方に12〜102及び113〜160を含む。両方の分子中の残基103と112との間の接続ループは無秩序である。2つのFab分子は軽鎖に関しては1〜212の残基、重鎖に関しては1〜221の残基を含む。C末端6xHisタグ及び鎖間ジスルフィド結合は無秩序である。
IFNα4A(図7A)の分子構造は、0.5〜0.7Åの平均Cα rmsdを有するIFNα2と非常に類似している。それはまた、IFNω(図7B、112の残基に関して0.61ÅのCα rmsd)及びIFNβ(図7C、94の残基に関して0.85ÅのCα rmsd)とも非常に類似している。IFNβのABループにより短い残基が1つあるために、IFNα/ωとIFNβの間にはいくつかの有意な違いがある。
M88は、ABループ(A19とD35の間)の残基及びへリックスEの残基V143、A146、E147及びR150で構成された立体配座エピトープを認識する(表11)。パラトープは6つの全てのCDRからの残基で構成されている。パラトープ残基は主に疎水性であり、一連のポケットを形成し、そこに、短いABへリックスの残基F27、L30、K31及びR33の側鎖がドッキングする。抗体と抗原の相互作用は、ほとんどがvdw及び疎水性パッキングのように思われる。抗体と抗原の間には少数のH結合しかなく、それらのほとんどにバックボーン−バックボーン又は側鎖−バックボーンの相互作用が関与している。Ab/Ag相互作用の大部分は、ABへリックスのいくつかの残基F27、L30、K31及びR33によるものである。したがって、IFNα4Aのこの領域がエピトープの主な部分を構成している。VLのF50は、構造の抗原と直接接していない。しかしながら、その側鎖は、結合に関与するY32(VL)及びP105(VH)の近くにある。この残基はおそらく、結合を有利にするために、CDR−H3の局所構造をそれが支持することから選択された。
M88はIFNαサブタイプの多くと強く結合するが、IFNωとの結合は弱い。現在の複合体の構造及びIFNωの構造を用いた分子モデリングに基づく、2つの戦略が可能である。1つの戦略は、IFNα4A/M88構造内の現在の接触の全てを維持する一方で追加のAb/Ag相互作用を生成することによりCDR−L1(extL1ライブラリ)を延長することである。構造及び配列の比較は、全てのIFNαサブタイプで5つの残基表面パッチ(D32、H34、D35、Y130及びK134)が100%保存されたことを示している(表12)。
IFNAR1及び/又はIFNAR2との複合体中のIFNα/ωの結晶構造は最近報告されている(Thomasら、Cell 146:621〜632、2011)。図8は、IFNω/IFNAR1/IFNΑ2複合体の上にM88/IFNα4を重ねたものである。HCとIFNAR2が重なり合うことは明らかである。したがって、M88はIFNAR2/IFN相互作用を遮断することによって中和する。
IFNωT80E/FabM43、IFNα4A/FabM88、IFNα4A/Fab357(C2595)及びIFNω/Fab357の結晶構造は、IFNω及び複数のIFNαサブタイプの広域中和に必要とされる最小限の共通エピトープを画定する(表14)。4つの結晶構造の抗体/抗原相互作用の解析は、IFNα4a(配列番号19)及びIFNω(配列番号1)のABループの3つの残基、F27、L30、及びR33がそれらの抗体と広範な接触を形成することを示している。これらの残基が抗体の結合に主に寄与する可能性が高い。したがって、F27、L30、及びR33は、IFNω/IFNαの交差中和エピトープの主要な要素である。
Claims (45)
- (a)ヒトインターフェロンオメガ(IFNω)、及び(b)少なくとも4、5、6、7、8、9、又は10個のヒトインターフェロンアルファ(IFNα)サブタイプと結合し、それらの活性を中和する、単離されたモノクローナル抗体。
- 前記抗体が、ヒトIFNαサブタイプIFNαB2、IFNαF、IFNαG及びIFNαJ1と結合し、それらの活性を中和する、請求項1に記載の抗体。
- 前記抗体が、ヒトIFNαサブタイプIFNαB2、IFNαC、IFNαF、IFNαG及びIFNαJ1と結合し、それらの活性を中和する、請求項1に記載の抗体。
- 前記抗体が、ヒトIFNαサブタイプIFNαB2、IFNαC、IFNαF、IFNαG、IFNαJ1及びIFNαAと結合し、それらの活性を中和する、請求項1に記載の抗体。
- 前記抗体が、ヒトIFNαサブタイプIFNαB2、IFNαC、IFNαF、IFNαG、IFNαJ1、IFNαA及びIFNαH2と結合し、それらの活性を中和する、請求項1に記載の抗体。
- 前記抗体が、ヒトIFNαサブタイプIFNαB2、IFNαC、IFNαF、IFNαG、IFNαJ1、IFNαA、IFNαH2及びIFNαKと結合し、それらの活性を中和する、請求項1に記載の抗体。
- 前記抗体が、ヒトIFNαサブタイプIFNαB2、IFNαC、IFNαF、IFNαG、IFNαJ1、IFNαA、IFNαH2、IFNαK及びIFNαWAと結合し、それらの活性を中和する、請求項1に記載の抗体。
- 前記抗体が、ヒトIFNαサブタイプIFNαB2、IFNαC、IFNαF、IFNαG、IFNαJ1、IFNαA、IFNαH2、IFNαK、IFNαWA及びIFNα4aと結合し、それらの活性を中和する、請求項1に記載の抗体。
- 前記ヒトIFNω及びヒトIFNαサブタイプの活性が、転写因子2(STAT2)、インターフェロン調節因子9(IRF9)、並びに分泌胚アルカリホスファターゼ(SEAP)のシグナル伝達因子及び活性化因子を安定に発現するHEK293細胞における、インターフェロン誘導性ISG54プロモーター下でのSEAPの阻害である、請求項1に記載の抗体。
- 前記抗体が、実施例3における「親和性の測定」の項で定義した条件下で、約5×10-8M未満、約1×10-8M未満、約1×10-9M未満、約1×10-10M未満、約1×10-11M未満、又は約1×10-12M未満のIC50値でヒトIFNωの活性を阻害し、且つ、5×10-8M未満、約1×10-8M未満、約1×10-9M未満、約1×10-10M未満、約1×10-11M未満、又は約1×10-12M未満のIC50値でIFNαサブタイプIFNαB2、IFNαF、IFNαG又はIFNαJ1の活性を阻害する、請求項9に記載の抗体。
- 前記抗体が、実施例3における「親和性の測定」の項で定義した条件下で、約5×10-9M未満、約1×10-9M未満、約5×10-10M未満、約1×10-10M未満、約5×10-11M未満、約1×10-11M未満、約5×10-12M未満又は約5×10-12M未満の解離定数(KD)のヒトIFNωと結合し、約5×10-9M未満、約1×10-9M未満、約5×10-10M未満、約1×10-10M未満、約5×10-11M未満、約1×10-11M未満、約5×10-12M未満、又は約5×10-12M未満のKDのヒトIFNαサブタイプIFNαB2、IFNαF、IFNαG又はIFNαJ1と結合する、請求項1に記載の抗体。
- 前記抗体が、ヒトIFNω、並びにヒトIFNαサブタイプIFNαB2、IFNαF、IFNαG及び/又はIFNαJ1との結合のために、
a)重鎖可変領域(VH)アミノ酸配列(配列番号23)、及び軽鎖可変領域(VL)アミノ酸配列(配列番号24)、又は、
b)VHアミノ酸配列(配列番号27)、及びVLアミノ酸配列(配列番号28)
を含む単離抗体と競合する、請求項1に記載の抗体。 - 前記抗体が、配列番号1の残基F27、L30、及びR33の1つ以上でIFNωと結合する、請求項12に記載の抗体。
- 前記抗体が、配列番号1の残基F27、L30、及びR33でIFNωと結合する、請求項13に記載の抗体。
- 前記抗体が、配列番号1の残基P26、K31、及びR34からなる群から選択される少なくとも1つのIFNω残基と更に結合する、請求項13に記載の抗体。
- 前記抗体が、配列番号1の残基R22、R23、I24、S25、P26、K31、D32、R34、D35、Q40、K134、M146、E147、M149、K150、F153及びL154からなる群から選択される少なくとも1つのIFNω残基と更に結合する、請求項13に記載の抗体。
- 前記抗体が、配列番号19の残基F27、L30、及びR33の1つ以上でIFNα4aと結合する、請求項13に記載の抗体。
- 前記抗体が、配列番号19の残基F27、L30、及びR33でIFNα4aと結合する、請求項17に記載の抗体。
- 前記抗体が、配列番号19の残基H26、K31、及びR34からなる群から選択される少なくとも1つのIFNα4a残基と更に結合する、請求項17に記載の抗体。
- 前記抗体が、配列番号19の残基A19、G22、R23、I24、S25、H26、C29、K31、D32、H34、D35、V143、A146、E147、M149、R150及びS153からなる群から選択される少なくとも1つのIFNα4a残基と更に結合する、請求項16に記載の抗体。
- 前記抗体がウィルス誘導性白血球インターフェロンの活性を阻害する、請求項12に記載の抗体。
- 前記ウィルス誘導性白血球インターフェロンの活性が、100U/mLのインターフェロンによって誘導された全血中のIP−10の放出である、請求項21に記載の抗体。
- 前記活性が、50μg/mLの抗体の存在下で50%超阻害される、請求項22に記載の抗体。
- 前記抗体が、全身性エリテマトーデス(SLE)免疫複合体によって誘導されるIFNの活性を阻害する、請求項12に記載の抗体。
- 前記活性が、約50%超阻害される、請求項24に記載の抗体。
- a)重鎖可変領域(VH)アミノ酸配列(配列番号23)、及び軽鎖可変領域(VL)アミノ酸配列(配列番号24)、又は、
b)重鎖可変領域(VH)アミノ酸配列(配列番号27)、及び軽鎖可変領域(VL)アミノ酸配列(配列番号28)
を含む、請求項12に記載の抗体。 - 前記抗体がヒトインターフェロン−β(IFNβ)と結合せず、中和しない、請求項12に記載の抗体。
- 前記抗体がヒトIFNαDと結合せず、中和しない、請求項12に記載の抗体。
- 前記抗体が、ヒト、ヒト化、又はヒト適応型である、請求項12に記載の抗体。
- 前記抗体が、IgG1型、IgG2型、IgG3型、又はIgG4型アイソタイプである、請求項29に記載の抗体。
- 前記抗体が二重特異性である、請求項29に記載の抗体。
- 前記抗体が、BLys、CD40L、IL−6、CD27、BDCA2、又はIL−12若しくはIL−23のp40サブユニットと結合する、請求項31に記載の抗体。
- 請求項1又は請求項12に記載の抗体と、製薬上許容された担体と、を含む、医薬組成物。
- IFNα及びIFNωの産生の増加に関連する疾病を治療又は予防する方法であって、請求項12に記載の単離された抗体の治療上の有効量を、それを必要とする患者に、その疾病の治療又は予防のために充分な時間にわたって投与することを含む、前記方法。
- 前記疾患が免疫媒介炎症性疾患である、請求項34に記載の方法。
- 前記免疫媒介炎症性疾患が全身性エリテマトーデス(SLE)、I型糖尿病、乾癬、原発性シェーグレン病、全身性硬化症、又は関節リウマチである、請求項35に記載の方法。
- 前記患者がI型インターフェロンシグネチャを示す、請求項34に記載の方法。
- 請求項12に記載の抗体が、二重特異性抗体である、請求項34に記載の方法。
- 前記二重特異性抗体が、BLys、CD40L、IL−6、CD27、BDCA2、又はIL−12若しくはIL−23のp40サブユニットと結合する、請求項38に記載の方法。
- IFNω、並びにIFNαサブタイプIFNαB2、IFNαC、IFNαF、IFNαG及び/又はIFNαJ1と、IFNARとの相互作用を阻害する方法であって、これを必要とする患者において、IFNω、並びにIFNαサブタイプIFNαB2、IFNαC、IFNαF、IFNαG及び/又はIFNαJ1と、IFNARとの相互作用を防ぐために十分な時間をかけて、請求項12に記載の単離された抗体を患者に投与することを含む、前記方法。
- 前記患者が免疫媒介炎症性疾患を有する、請求項40に記載の方法。
- 前記免疫媒介炎症性疾患が、SLE、I型糖尿病、乾癬、原発性シェーグレン病、全身性硬化症、又は関節リウマチである、請求項41に記載の方法。

- 前記二重特異性抗体が、BLys、CD40L、IL−6、CD27、BDCA2、又はIL−12若しくはIL−23のp40サブユニット、あるいはBDCA2と結合する、請求項44に記載の抗体。
