470nmのメラニン吸収を測定した。無色素細胞XB2中のタンパク質ライセートは較正物質として機能する(Abs470nm=0)。試料miCtrl(cel−miR−239b)は参照試料として使用される(Abs470nm=1)。メラニンの相対レベルは、hsa−miR−330−5p(mi−330)およびhsa−miR−137(mi−137)のミメティックで処理された色素沈着MNT−1細胞がトランスフェクションコントロール(cel−miR−239b)に対して減少していることを示す。メラニン吸収も各ライセート試料中のタンパク質の量に対して正規化されている。
Tyr、TYRP1、MLANAおよびMITF−MのmRNA発現をPCR−qRT分析により測定した(相対定量法ΔΔCt)。陰性対照(cel−miR−239b)で処理されたMNT−1細胞を較正物質試料として使用した(相対量、RQ=1)。hsa−miR−330−5p(mi−330)、hsa−miR−7(mi−7)およびhsa−miR−137(mi−137)のミメティックで処理されたMNT−1細胞のmRNA発現は、較正物質試料に対する相対値として記録されている。エラーバーは3つ組の測定の標準偏差を表す。MNT−1細胞のmRNA TYRレベルは、hsa−miR−330−5p、hsa−miR−7およびhsa−miR−137のミメティックで処理すると低下する。TYRP1、MLANAおよびMITF−MのmRNAレベルは、miR−7によって若干低下し、miR−137によってかなり低下するのが明らかである。
対照に比べて、化合物12,085で処理した後のMNT−1細胞におけるmiR−330−5pの発現は48時間後に20%超増加する。値は平均値で表され、エラーバーは標準誤差を表す。*P=0.0233(対応のないスチューデントt検定)
対照に比べて、化合物12,080で処理した後のMNT−1細胞におけるmiR−137の発現は48時間後に20%超増加する。値は平均値で表され、エラーバーは標準誤差を表す。*P=0.0056(対応のないスチューデントt検定)
対照に比べて、化合物12,092で処理した後のMNT−1細胞におけるmiR−7の発現は48時間後に20%超増加する。
A)チロシナーゼ(TYR)の3’UTR部分におけるmiR−330−5pの結合部位の模式図。miR−330−5pの結合部位の野生型配列(WT)を示す。変異型配列(MUT)には、5つのヌクレオチド変化が認められる(赤色で示す)。
B)ルシフェラーゼレポーターの活性の試験は、miR−330−5pのTYR 3’UTRとの結合を明らかにする。Lu1205ヒトメラノーマ細胞に(インサートなし、WTおよびMUT)様々なルシフェラーゼレポーターベクターをトランスフェクトし、miR−330−5pのミメティックを24時間コトランスフェクトする。溶解した細胞から得られたタンパク質抽出物について、ルシフェラーゼ活性を測定する。インサートのないコンストラクトを参照として使用した。値は平均値の形態で表す。エラーバーは標準偏差を表す。(**P<0.005 マンホイットニーの検定)。
細胞
ヒトメラノーマ色素沈着細胞MNT−1(Reishら、1995)を、10%ウシ胎仔血清(#10270、Gibco)、1%ペニシリン−ストレプトマイシン(#15140、Gibco)および1%L−グルタミン(#25030、Gibco)を含むRPMI1640培地(#21875、Gibco)中、37℃および5%CO2で高湿度雰囲気下にて培養した。XB2マウスケラチノサイト(RheinwaldおよびGreen、1975)を、10%ウシ胎仔血清(#10270、Gibco)、1%ペニシリン−ストレプトマイシン(#15140、Gibco)および1%L−グルタミン(#25030、Gibco)を含むDMEM(#41965、Gibco)中、37℃および5%CO2で高湿度雰囲気下にて培養した。
−1日目に、MNT−1細胞を6ウェルプレート(#353046、Falcon)に1ウェル当たり4×105個播種した。0日目に、5μLのリポフェクタミン2000(#1668−019、Invitrogen)を使用して、細胞に50nMのmiRNA(hsa−mir−137 #C−300604−07−005、hsa−mir−7 #C−300547−05−0005、hsa−mir−330−5p #C−301082−01−0005、陰性対照#NC−002000−01−05、Dharmacon)のミメティックをトランスフェクトした。1日目に、培地を交換した。3日目に、細胞をトリプシン処理し、1ウェル当たり4×105個の細胞を播種した。4日目に、細胞に50nMのmiRNAのミメティックを再びトランスフェクトした(0日目を参照のこと)。5日目に、培地を交換した。7日目に、細胞をトリプシン処理し、1ウェル当たり4×105個の細胞を播種した。8日目に、細胞に50nMのmiRNAのミメティックをトランスフェクトした(0日目を参照のこと)。9日目に、培地を交換した。10日目に、細胞をトリプシン処理し、2つに分けた(1枚の6ウェルから2枚の6ウェル)。12日目に、一方の6ウェルプレートをRNAの溶解に使用し、他方をタンパク質の溶解に使用した。色素脱失の陽性対照として、MNT−1細胞を3.5mMのコウジ酸(Sigma K3125−5G)と共に12日間インキュベートした。細胞は記載のように分布し、コウジ酸を培地と共に4日ごとに新しくした。コウジ酸の阻害率(%)は10%程度である。
−1日目に、hsa−miR−7、hsa−miR−137およびhsa−miR−330−5pの阻害による潜在的色素沈着促進効果を調査するために、Lu1205細胞(ヒト無色素メラノーマ細胞株)を6ウェルプレート(#353046、Falcon)に1ウェル当たり4×105個播種した。0日目に、5μLのリポフェクタミン2000(#1668−019、Invitrogen)を使用して、細胞に50nMのmiRNAの各インヒビター(=アンタゴmiR)(hsa−miR−137 #IH−300604−08−0005、hsa−miR−7 #IH−300546−08−0005、hsa−miR−330−5P #IH−301082−02−0005、陰性対照#EN−002005−01−05、Dharmacon)をトランスフェクトした。1日目に、培地を交換した。3日目に、細胞をトリプシン処理し、1ウェル当たり4×105個の細胞を播種した。4日目に、細胞に50nMのmiRNAのインヒビターをもう一度トランスフェクトした(0日目を参照のこと)。5日目に、培地を交換した。7日目に、細胞をトリプシン処理し、1ウェル当たり4×105個の細胞を播種した。8日目に、細胞に50nMのmiRNAのインヒビターをトランスフェクトした(0日目を参照のこと)。9日目に、培地を交換した。10日目に、細胞をトリプシン処理し、2つに分けた(1枚の6ウェルから2枚の6ウェル)。12日目に、一方の6ウェルプレートをRNAの溶解に使用し、他方をタンパク質の溶解に使用した。
細胞をPBSで2回洗浄した後、6ウェルディッシュの1ウェル当たり700μLのQiazol溶解バッファー(Qiagen)を添加した。miRNeasy Miniキット(Qiagen)を製造業者のプロトコルに従って使用して、全RNAを単離した。RNAseを使用することなく、全RNAを水30μlで溶離した。NanoVue分光光度計(GE Healthcare)を使用して、RNAの濃度を測定した。MLV RT−酵素系(Invitrogen)を60μLの反応中で使用して、1μgの全RNAを逆転写して、対応するcDNAを産生した。
IQ(商標)SYBR(登録商標)Green supermix(Biorad)を使用して、リアルタイムPCRを行った(最終反応体積 25μL:Mix Master 23μL、cDNA 2μL)。サーモサイクリング条件(Icycler、Biorad)は以下の通りであった:95℃で90秒、さらに95℃で30秒、60℃で60秒および95℃で10秒を40サイクル。プライマーの濃度および配列を以下の表5に示す:
12日目に、細胞をPBS中で2回洗浄し、6ウェルディッシュのRIPAタンパク質用溶解バッファー75μL/ウェルに溶解した。タンパク質ライセートを1.5mlのEppendorf試験管に移し、軽くボルテックスして、溶液をホモジナイズし、その後試験管を氷中に入れ、または−80℃で貯蔵した。2μLのタンパク質ライセートを使用して、メラニンの470nmにおける吸光度をNanoVue分光光度計(GE Healthcare)で測定した。無色素XB2角化細胞のタンパク質ライセートを較正物質試料として使用した(参照=0)。メラニンの相対量を検出するために、すべての試料をトランスフェクションコントロール試料(miCtrl)と比較した。試料miCtrlの470nmにおける吸光度を参照として1と定めた。さらに、miCtrlに対する各試料のタンパク質の濃度を考慮して、470nmにおけるメラニン吸収を正規化した。
プロテアーゼインヒビター(Roche)を含む75μLの放射性免疫沈降用バッファー(RIPA)(PBS中1%NP40、0.5%デオキシコール酸ナトリウム、0.1%SDS)を使用して、6ウェルプレートからタンパク質を4℃で30分間抽出した。タンパク質ライセートを14000rpm、4℃で30分間遠心した。予冷し、氷中に入れておいた1.5mlのEppendorf試験管に上澄液を移した。タンパク質の濃度をBCA Protein Assay(ThermoFisher)で決定した。タンパク質(50μg)を10%SDS−ポリアクリルアミドゲルにて分離し、ProTronニトロセルロースメンブラン(Whatman)に100V、室温で1時間移した。メンブランを緩衝Tris食塩水(SCT)中の5の脱脂乳粉末で飽和させた。メンブランを0.5%(v/v)のTween/SCT(TTBS)に希釈した一次抗体(下記を参照のこと)で一晩かけてプローブ付し、次いでTTBS中で1回5分間の洗浄を3回行った。二次抗体をTTBSで1:10,000に希釈し、室温で1時間適用した。次いで、メンブランを、TTBS中で1回10分間を3回洗浄し、Amersham Hyperfilm(GE Healthcare)を使用して、SuperSignal WestPico化学発光溶液(Thermoscientific)で発像させた。

hsa−miR−330−5pおよびhsa−miR−137は色素沈着のレベルに影響を及ぼす
hsa−miR−330−5p、hsa−miR−137のミメティックで12日間処理したMNT−1細胞は、陰性対照で処理した細胞に比べて色素沈着レベル全体で有意な低下を示す(写真は示さない)。XB2ケラチノサイトは無色素細胞の対照として使用する。12日目のタンパク質ライセートの写真から、miR−7で処理したMNT−1細胞は、色素沈着全体に対して有意な効果を何ら示さないことが明らかである。色素脱失の陽性対照として、MNT−1細胞をコウジ酸で処理した。
hsa−miR−7のミメティックで12日間処理したMNT−1細胞は、陰性対照細胞に比べて色素沈着レベル全体で有意な低下を示さない(写真は示さない)。メラニンの470nmにおける吸光度の測定による相対レベルの定量によって、hsa−miR−7のミメティックで処理したMNT−1細胞中においてメラニンが若干増加していることが示唆されている(図1)。しかし、ウェスタンブロット分析によって明らかなように、分子レベルでは、hsa−miR−7のミメティックによる処理によってTYRおよびMITFのタンパク質発現が低下する(写真は示さない)。さらに、miR−7はTYR、TYRP1、MLANAおよびmRNA MITF−Mのレベルを低減するようである(図2)。
1.化合物
0日目(D0)に、150000個のMNT−1ヒトメラノーマ細胞を24ウェルプレートに播種した。D1に、細胞に以下の化合物で生物学的処理を3つ組にて48時間行った:

12,092 DMSO 16.10−5
12,093 エタノール 4.10−4
ここで、
12,080=アスコルビルグルコシド
12,085=国際公開第2011/033207号に記載のように調製されたロイコドパクロム
12,092=国際公開第2006/134282号に記載のように調製されたチオクト酸ジアセチルレスベラトリル
12,093=トラネキサム酸セチル(TXC)。
隣接する塩基を含むTYR(NM_000372)の3’UTR(全長393ntの3’UTRの68〜75の位置)中のmiR−330−5pの予測結合部位は、以下のプライマーを使用して設計され、pmirGLOレポーターベクター(Promega)のホタルルシフェラーゼをコードする領域の下流にPmel部位およびXbaI部位を使用してクローニングした。
1.化合物
色素沈着MNT−1ヒトメラノーマ細胞を化合物で処理し、miRNA(miR−330−5p、miR−137およびmiR−7)の発現を48時間後に分析した。
チロシナーゼの3’UTRにおけるmiR−330−5pの予測結合部位(WT)を、ルシフェラーゼレポーターベクターの下流にクローニングした。さらに、miR−330−5pの変異型結合部位(MUT)をクローニングした(図6A)。
Claims (10)
- hsa−miR−330、hsa−miR−7、hsa−miR−137、それらの成熟型およびそれらの前駆体から選択される少なくとも1つのマイクロRNAの、色素脱失活性成分としての使用。
- 色素沈着過度障害を予防および/または治療するために使用するための、hsa−miR−330、hsa−miR−7、hsa−miR−137、それらの成熟型およびそれらの前駆体から選択されるマイクロRNA。
- 色素沈着過度障害が、黒皮症、肝斑、黒子、老人性黒子、光老化に伴う不規則な色素沈着過度、そばかす、擦過または熱傷または瘢痕または皮膚病または接触アレルギーによる炎症後色素沈着過度、母斑、遺伝性色素沈着過度、代謝由来または薬剤起因性の色素沈着過度、およびメラノーマから選択される、請求項2に記載のマイクロRNA。
- マイクロRNAが、hsa−miR−330、hsa−miR−330−5p、hsa−miR−330−3p、hsa−mir−7−1、hsa−miR−7−2およびhsa−miR−7−3、hsa−miR−7−5p、hsa−miR−7−1−3p、hsa−miR−7−2−3p、hsa−miR−137、ならびにそれらの前駆体から選択されることを特徴とする、請求項1に記載の使用または請求項2もしくは3に記載のマイクロRNA。
- 色素脱失化合物を同定するためのインビトロの方法であって、
a.少なくとも1つの被験化合物をメラノサイト試料と接触させるステップ、
b.前記メラノサイトにおいてhsa−mir−330、hsa−mir−7、hsa−mir−137、それらの成熟型およびそれらの前駆体から選択される少なくとも1つのマイクロRNAの発現または活性を測定するステップ、
c.ステップaで処理された前記メラノサイトにおいて測定される前記マイクロRNA、その成熟型および前駆体のうち少なくとも1つの発現または活性の活性化が、無処置のメラノサイトと比べて少なくとも20%である化合物を選択するステップ
を含む方法。 - ステップbが、ステップaの前および後に実施されることを特徴とする、請求項5に記載の方法。
- a’.少なくとも2つのメラノサイト試料を調製するステップと、
a.前記試料の1つを少なくとも1つの被験化合物と接触させるステップと、次いで
b.前記試料において、hsa−mir−330、hsa−mir−7およびhsa−mir−137、それらの成熟型およびそれらの前駆体から選択される少なくとも1つのマイクロRNAの発現または活性を測定するステップと、
c.ステップaで処理されたメラノサイトにおいて測定される、前記マイクロRNA、その成熟型および前駆体のうちの少なくとも1つの発現または活性の活性化が、無処置のメラノサイトと比べて少なくとも20%である化合物を選択するステップ
とを含むことを特徴とする、請求項5に記載の方法。 - 前記hsa−mir−330、hsa−mir−7、hsa−mir−137、その成熟型および前駆体から選択される少なくとも1つのマイクロRNAの発現または活性が、対応するマイクロRNAの定量で測定されることを特徴とする、請求項5〜7のいずれか一項に記載の方法。
- 前記被験化合物が植物抽出物から選択されることを特徴とする、請求項5〜8のいずれか一項に記載の方法。
- 前記メラノサイトが、メラノーマ系統、好ましくはヒトメラノーマから得られることを特徴とする、請求項5〜9のいずれか一項に記載の方法。
