Abstract
Description
The present invention relates to pharmaceuticals, and more specifically to a transdermal drug administration system and a method for producing a transdermal drug administration system.
It is preferable that a drug that cannot exert its medicinal effect due to the first-pass effect on the liver by oral administration is administered transdermally. Here, in the stratum corneum, which is the outermost layer of the skin, the keratin made of ceramide, cholesterol, etc. around the epidermal keratinocytes covered with a cell membrane that is very stable against physical and chemical stimulation. Intercellular lipids are filled. This stratum corneum structure exerts a skin barrier function that prevents invasion of foreign substances from outside the body, and prevents evaporation of body water and leakage of body fluids. For this reason, even when a drug is applied to the skin surface, a water-soluble drug having a molecular weight of approximately 500 or more cannot be expected to be transdermally absorbed. Therefore, when a drug is administered transdermally, the skin is usually punctured by an injection needle and the drug is injected into the inside of the stratum corneum. However, since the needle reaches the nerve, the patient must endure the pain.
On the other hand, it has been proposed to transdermally administer a drug by piercing the skin with a microneedle preparation obtained by molding the drug into a microneedle shape having a height of several tens of μm to several mm (height). For example, see
However, the conventional microneedle preparation has a problem that a drying process is required and productivity is low. Therefore, an object of the present invention is to provide a transdermal drug administration system with high productivity.

(D) A method for producing a transdermal drug administration system is provided.
Further, according to the aspect of the present invention, (a) a mold provided with a plurality of recesses having at least a portion whose cross-sectional area decreases from the opening toward the bottom, and (b) each of the plurality of recesses is filled, A plurality of molding agents for piercing the skin having at least a portion having a cross-sectional area that decreases from the bottom toward the tip; and (c) a base material combined with the plurality of molding agents, and (d) a plurality of (E) A transdermal drug delivery system is provided in which the shaped drug is removable from the mold.

Embodiments of the present invention will be described below. In the following description of the drawings, the same or similar parts are denoted by the same or similar reference numerals. However, the drawings are schematic. Therefore, specific dimensions and the like should be determined in light of the following description. Moreover, it is a matter of course that portions having different dimensional relationships and ratios are included between the drawings.
(First embodiment)
As shown in FIGS. 1 and 2, the transdermal drug administration system according to the first embodiment has a cross-sectional area from the
The
When the
The active ingredients of the molded
Further, when the active ingredient is a substance that is desired to be continuously administered, such as hormones, the molded
Each of the plurality of
The volume of each of the plurality of

The keratinocytes that make up the epidermis of the skin are involved in the induction of innate immunity by sensing the invasion of foreign substances and producing inflammatory mediators such as cytokines and chemokines. In addition, antigen-presenting cells called Langerhans cells exist in the cell space of keratinocytes. Furthermore, dermal dendritic cells are resident in the dermis, and Langerhans cells and dermal dendritic cells that recognize or prey on foreign substances migrate to the regional lymph nodes and activate T cells and B cells in an antigen-specific manner. This induces an immune response against the foreign body. Therefore, for example, when the active ingredient of the
Each of the plurality of

The
Alternatively, the material of the
The
The area of the
Next, a manufacturing method of the transdermal drug administration system according to the first embodiment will be described. First, as shown in FIG. 3, a
Thereafter, as shown in FIG. 6, the
Next, as shown in FIG. 7, an adhesive 35 is applied to the surface of the
According to the manufacturing method of the transdermal drug administration system according to the first embodiment described above, when the
Moreover, the conventional microneedle formulation is manufactured by pouring an active ingredient-containing solution into a mold, and it is necessary to dry the solution poured into the mold. On the other hand, according to the manufacturing method of the transdermal drug administration system according to the first embodiment, the
Next, a method for using the transdermal drug administration system according to the first embodiment will be described. First, as shown in FIG. 1, a transdermal drug administration system is transported from a pharmaceutical factory to a hospital, a pharmacy, a patient's home, or the like while the molded
After that, as shown in FIG. 9, the microneedle array is pressed against the
In the method of using the transdermal drug administration system according to the first embodiment, when the transdermal drug administration system is transported from a pharmaceutical factory to a hospital, a pharmacy, a patient's home or the like, the molded
(Second Embodiment)
As shown in FIGS. 11 and 12, the transdermal drug administration system according to the second embodiment includes a plurality of

(Measurement method)
For the measurement of the compression modulus of the molded medicine, a Tribodenter compression tester manufactured by Hystron was used, and a spherical indenter having a tip radius of 100 μm was used as the indenter. The measurement was carried out at room temperature at a press-in speed of 30 μm / second, and the elastic modulus was calculated from the stress at 10% compression.
(Production example)
Spirulina dye as an active ingredient and crystalline cellulose as an excipient were mixed at a mass ratio of 3: 7 to prepare a solid drug having an average particle size of 20 μm. Next, using a round tableting machine having a diameter of 11.3 mm, the solid drug is filled into a mold made of an acrylic resin having a diameter of 10 mm and a thickness of 1 mm, and a force of 1,800 kgf is applied. Forty-nine conical molding agents having a bottom diameter of 80 μm, a tip diameter of 20 μm, and a height of 52 μm were formed inside the mold recess. Here, the drug was molded at room temperature, and the molded drug was not dried by heating. Further, a base material made of polylactic acid having 49 pillars having a bottom diameter of 80 μm, a tip diameter of 20 μm, and a height of 100 μm was prepared. Thereafter, the tip of the pillar provided on the base material and the bottom of the molded drug were adhered to each other through D-mannitol as an adhesive to obtain a transdermal drug administration system.
(result)
The compression molding modulus of the obtained molding was 1 GPa. The obtained transdermal drug administration system was pressed against the skin of a hairless mouse, then fixed with a medical tape, peeled off after 20 minutes, and visually observed. As a result, transfer of the pigment into the skin could be confirmed.
