Abstract
Description
The present invention relates to an automatic analyzer that analyzes the amount of components contained in blood or urine, for example, a program and a recording medium, and an automatic sample analysis method.
As a measure for eliminating a photometric error due to a positional deviation of a cell in which a measurement object is accommodated, there is one described in
In the invention described in
Irradiate light from a light source to a reaction mixture of a biological sample such as serum or urine and a reagent, calculate the absorbance from the change in the amount of transmitted light at a specific wavelength, and quantify the concentration of the measured substance according to the Lambert-Beer law Automatic analyzers are widely used.
There are two types of reactions measured by an automatic analyzer, mainly a color reaction due to a reaction between a substrate and an enzyme, and an immunoaggregation reaction between an antigen and an antibody. The analysis using the former reaction is called biochemical analysis, and there are enzymes, lipids, nitrogen compounds, etc. as test items. Analysis using the latter reaction is called an immunological analysis item, and among the test items, there are trace protein (CRP), tumor marker, hormone, blood drug and the like.
Among the measurement substances measured in the latter, there are test items that require highly sensitive detection in a low concentration region, and test items whose quantitative values are important for clinical diagnosis. In these items, a latex immunoturbidimetric method using latex particles sensitized (bound) with an antibody on the surface as a sensitizer is used.
In the latex immunoturbidimetry, light is irradiated to an agglomerate formed by aggregating latex particles that are scatterers with a measurement substance, and a change in the amount of transmitted light transmitted without being scattered is measured. Since the change in the amount of light after a lapse of a certain time increases as the concentration of the measurement substance increases, the concentration of the measurement substance is calculated from the change in the amount of transmitted light.
In recent years, the needs for measurement of immunological analysis items are increasing, and there is a further demand for improved performance in immunological analysis items. For this reason, not only a change in the amount of transmitted light but also a method of quantifying the concentration using a change in the amount of scattered light has been used.
Here, in the conventional automatic analyzer, in order to meet the need for reducing the amount of the reaction solution, a severe design has been required for the positional displacement of the reaction vessel in the height direction.
However, in recent years, measurement needs for immunological analysis items are increasing, and further improvement in performance is desired for immunological analysis items containing scatterers in the reaction solution.
In the immunological analysis item, scattered light is emitted from the scatterer when the reaction solution is irradiated with light. For this reason, in the scattered light measurement method in which the concentration is quantified from the change in the amount of scattered light, the position deviation in the optical axis direction of the reaction vessel (the light source generated due to the deviation in the distance from the central axis of the reaction disk to the center position of the reaction vessel) If there is a deviation in the distance from the center of the reaction vessel to the center position of the reaction vessel, the amount of scattered light changes greatly, which causes a problem that it has a very large effect.
The problem of the positional deviation in the optical axis direction may affect the performance when scattered light enters the light receiver even in the conventional absorbance measurement in which the concentration is quantified from the change in the amount of transmitted light.
In the automatic analyzer, a large number of reaction vessels are arranged in a circle, and the sample is reacted with the reagent in each reaction vessel to quantify the concentration in the sample. Then, before measuring a sample having an unknown concentration, a solution having a known measurement substance concentration is measured in advance, and a calibration curve is created in which the relationship between the measurement substance concentration and the change in light quantity is examined.
Therefore, there is no need to take into account variations between apparatuses, and the concentration of the measurement substance in the sample can be quantified.
However, if there are misalignments in the optical axis direction among a number of reaction vessels in the apparatus, measurement variations between reaction vessels will occur, causing deterioration in reproducibility and further improving analytical performance. I could not.
In the automatic analyzer described in
However, with this automatic analyzer, it is necessary to install multiple sensors and control each sensor, so it is necessary to make very fine adjustments to maintain high measurement accuracy. There was a problem.
Moreover, since it was not thought that the position shift of the reaction vessel in the optical axis direction would affect the measurement result, it has not been a technical problem until now.
Thus, until now, there has been almost no effective countermeasure against the displacement in the optical axis direction between the reaction vessels.
An object of the present invention is to easily realize high-accuracy measurement in an automatic analyzer having a mechanism for irradiating a reaction liquid obtained by reacting a specimen and a reagent with light and detecting light emitted from the reaction liquid. It is an object of the present invention to provide an automatic analyzer and a specimen automatic analysis method that can be used, and a program and a recording medium for realizing the same.
In order to solve the above problems, for example, the configuration described in the claims is adopted.

According to the present invention, in an automatic analyzer equipped with a mechanism for irradiating light to a reaction solution obtained by reacting a specimen and a reagent and detecting light emitted from the reaction solution, the reaction container is in the optical axis direction of the irradiation light. As a result, highly reliable measurement results can be provided to the clinical side.

Next, the sample analysis operation of the above-described automatic analyzer of the present invention will be described.

Next, the operator uses the operation function screen displayed on the
At this time, information such as a patient ID is also input from the keyboard 58. In order to analyze the inspection item designated for each sample, the
The
Thereafter, the sample and the reagent are agitated and mixed by the agitating
Here, in the
In addition, when the

Based on the above relationship, it is possible to easily realize highly accurate measurement by using the reaction container constant that corrects the positional deviation in the optical axis direction and that the positional deviation in the optical axis direction of the reaction container is correlated with the scattered light intensity. Hereinafter, the calculation flow of the reaction vessel constant using the scattered light intensity evaluation solution will be described with reference to FIG.

As the scattered light intensity evaluation solution, a latex solution in which latex particles are dispersed can be used in order to ensure a constant scattered light intensity during continuous measurement.

Details of step S6-6 will be described with reference to FIG.

Examples of the timing for executing the reaction vessel constant calculation processing procedure include the time of shipment of the automatic analyzer and the maintenance such as replacement of the
A detailed flow of the correction processing of the detected scattered light data using the reaction vessel constant will be described with reference to FIG.
First, the
I1’=(I1−b)/a … (1)

The length, position, etc. of this extraction section differ for each inspection item and are designated in advance as analysis parameters.

Programs for executing the procedures shown in FIGS. 6, 7, and 9 are recorded in the storage unit 50a of the
In the above-described embodiment of the automatic analyzer, the program and the recording medium, and the automatic specimen analysis method of the present invention, the scattered light intensity evaluation solution is dispensed to all the
Moreover, the
Furthermore, in comparison with the method in which integrating spheres are arranged before and after the reaction vessel as in
In addition, even if the diameter of the
Further, by calculating the reaction vessel constant at the time of apparatus maintenance, it can also be used to determine the necessity of adjusting the
In addition, the
Furthermore, by displaying the scattered light intensity and the reaction vessel constant for each of the
In addition, this invention is not restricted to said embodiment, A various deformation | transformation and application are possible.
For example, the positional deviation of the reaction vessel in the optical axis direction does not affect only the case of measuring the scattered light intensity described above, but also affects the absorbance measurement for measuring the change in the amount of transmitted light. That is, the present invention for correcting the difference in measured values between a large number of reaction vessels due to the positional deviation of the reaction vessel with respect to the optical axis direction is not only for measuring the scattered light intensity of a solution containing a scatterer. The present invention can also be applied when measuring the absorbance of a solution containing a scatterer with an absorptiometer.
For example, in FIG. 1, when calculating the concentration of the measurement target substance in the sample from the amount of transmitted light that is irradiated from the
Moreover, the substance for evaluation used when determining the reaction vessel constant is not limited to a solution containing a scatterer, and may be a solid containing a scatterer.
Furthermore, the reaction container used as a reference is reaction container No. The position is not limited to 1 and can be any position. However, it is desirable to use the
However, as described above, in the automatic analyzer, before measuring a sample with an unknown concentration, a calibration curve in which a solution with a known measurement substance concentration is measured in advance and the relationship between the measurement substance concentration and the change in light quantity is examined. Therefore, if the data from all the reaction vessels in the apparatus are prepared with the reaction vessel at an arbitrary position as a reference, the concentration of the measurement substance in the sample can be quantified.
