Abstract
Description
The invention of the present application relates to a fluorimeter, and more particularly to a fluorimeter preferably used for sample identification and quantification.
As one field of light measurement, a fluorescence measurement technique for measuring fluorescence emitted from a substance is known. Material analysis by fluorescence measurement (fluorescence analysis method) is characterized by high sensitivity and high selectivity as compared with absorptiometry and the like, and is effective in, for example, identification and quantification of chemical substances.
Conventionally known general fluorometers are large measuring instruments and are installed in laboratories and measuring rooms. A material (hereinafter referred to as a sample) containing a target substance such as identification or quantification (hereinafter referred to as a sample) is collected outside and brought into a laboratory or measurement room for fluorescence measurement. .
In recent years, such fluorescence measurement techniques have been studied for application in various fields such as research and development of new drugs and new materials, process monitoring in plants, and environmental evaluation. In addition, active research is being conducted on techniques for labeling samples with fluorescent dyes and techniques for using immune reactions (antibody antigen reactions) for fluorescence measurements.
As the application field of fluorescence measurement expands in this way, fluorescence measurement is not performed in a special room such as a laboratory or measurement room, but is performed in various other places or at the site where a sample is collected. It is expected that there will be a need to measure and obtain results quickly. For example,
Methamphetamine is a typical stimulant and is a so-called prohibited drug. Therefore, the detection of methamphetamine is performed, for example, by baggage inspection at airport customs or drug enforcement by the police. Although the activities of so-called drug dogs are widely known for banned drug control at customs, there is a limit to inspecting a large amount of baggage without fail, and even if a substance suspected to be a banned drug is found, To detect and take legal action automatically, the discovered material must be scientifically analyzed and identified. For this purpose, it is necessary to temporarily hold the baggage and take measures such as sending the discovered substance to the inspection organization, and the customs clearance is temporarily put on hold.
If a discovered substance can be identified promptly at the site where prohibited drugs are controlled, there is no need to temporarily hold customs clearance and leave travelers for a long time, and they can be caught and arrested immediately. However, no testing equipment has been developed so far that can be used to identify prohibited drugs at the enforcement site.
In principle, methamphetamine can be detected by measuring fluorescence intensity by using an immune reaction as disclosed in
Slightly in the known literature, in
Thus, no practical portable fluorometer has been developed, and no practical teaching has been given for practical portable fluorometers, even within the scope of known literature. Absent. For this reason, it has not been clarified at all what kind of problems exist when making the fluorometer portable and practical. In this situation, if a practical portable fluorometer can be developed, the technology of fluorescence measurement can be widely applied in various fields such as plant monitoring and environmental evaluation, as well as the control of the above prohibited drugs. It is guessed.
One technical matter required for a portable fluorometer is, of course, the overall compactness. It is necessary to make the whole compact enough to be held by people. According to the research of the inventors of the present application, it has been found that if the entire fluorometer is made compact so as to be suitable for a portable type, a problem that cannot be ignored in terms of measurement accuracy occurs. Hereinafter, this point will be described.
In general, in various fluorescent materials, the range of wavelengths of light to be excited and the range of wavelengths of fluorescence generated by excitation are often close or partially overlapped. Therefore, when excitation light leaking from the excitation optical path enters the detector, it often becomes noise and is included in the output value (hereinafter, this excitation light is referred to as leakage light). A filter is arranged on the optical path in front of the detector so that only the fluorescence wavelength is transmitted and incident on the detector, but the filter does not exhibit sufficient wavelength selection characteristics for obliquely incident light. Sometimes. For this reason, it is possible that the leaked light is incident obliquely, passes through the filter, and enters the detector.
Such a problem is not so obvious in the case of a large-sized fluorometer such as used in a laboratory or a measurement room because a sufficient space can be provided in the optical system and the detection system. This is because the detection can be avoided by arranging the detector at a position sufficiently away from the light source and the excitation optical path. However, in the case of a portable fluorometer, in order to make the whole compact, it is necessary to arrange an optical system and a detection system in a limited narrow space. For this reason, the detector must be disposed at a position relatively close to the light source and the excitation optical path, and the problem of the leakage light is likely to become obvious.
This invention is made | formed in view of such a situation, and makes it a subject to provide the structure of leakage light prevention suitable for a portable fluorometer.

Next, modes for carrying out the present invention (hereinafter referred to as embodiments) will be described.
FIG. 1 is a schematic perspective view of a fluorometer according to the first embodiment of the present invention, and FIG. 2 is a schematic front sectional view of the fluorometer shown in FIG.
The fluorometer of the embodiment is assumed to be portable, that is, to be used while being carried by a person. As shown in FIG. 1, the fluorometer is a flat, substantially rectangular parallelepiped box-like thing as a whole. Since it is portable, its size is about the size of a person's palm or slightly larger.
As shown in FIG. 2, this fluorometer includes a
The fluorometer of this embodiment is configured to measure fluorescence by putting a sample in a dedicated container. Hereinafter, this container is referred to as a sample container. The sample container contains a test solution in advance, and is provided to the measurer as a fluorescence measurement kit. The “test liquid” means a liquid phase material used for the test. Further, in the present specification, the “object” means an object for which fluorescence measurement is performed. Normally, the sample in which the sample is dissolved in the test solution is the liquid phase object. However, if the sample is in the liquid phase and the target substance is a fluorescent substance, and the test solution is not used, the sample = liquid phase It becomes an object. In this specification, “fluorescence” is a broader concept and includes phosphorescence.

The

As shown in FIG. 2, the
Further, the
As the filter, an
The

The fluorometer of this embodiment having such a structure includes a leakage
FIG. 7 is a schematic front sectional view of the main part of the fluorometer according to the first embodiment, and shows a specific structure of the leakage
In the structure of this embodiment, what happens when there is no leakage
When there is no leakage
In the present embodiment, in consideration of this problem, as shown in FIG. 7, the leakage
The leak
More specifically, the leakage
As shown in FIG. 7, the optical path of the leaked light L reaches the leaked
The inner surface of the
About several percent of excitation light that passes through the
Even when the
Further, the light that enters the leaked
As shown in FIG. 4, the leaking
The fact that the leak
An example of the experimental results confirming the effect of reducing the leakage light by the leakage
When the experiment was performed under such conditions, the measurement result was 64.7 mV in the absence of the leakage
In the present embodiment, each leakage
To remove problematic light such as leakage light from the optical path, an ND filter (a neutral density filter) is often used. However, the use of a dedicated dimming element such as an ND filter causes an increase in cost, and a structure for holding it is required, and the optical system tends to be complicated. In addition, although it is an ND filter, there is a slight amount of light reflected and returned from the surface, which may cause noise. In the structure of the present embodiment, a cylindrical part surrounding the leaked light is provided and attenuated inside, so that the problem of the leaked light is effectively solved at a simple structure and at a low cost. Can do.
In the present embodiment, the
Next, how to use such a fluorometer will be described.
When performing fluorescence measurement using the fluorometer of the embodiment, a fluorescence measurement kit shown in FIGS. 3 to 4 is prepared. The
Next, the open /
In the above-described measurement, as in the case of the immunoassay using the quenching elimination effect described above, reference value data is acquired, and therefore measurement may be performed without first putting a sample. In this case, the
As an example of immunoassay, a more specific example of measurement will be described. Regarding methamphetamine, a technique for producing a monoclonal antibody as an anti-methamphetamine antibody by culturing a cell line obtained by immunizing an animal is disclosed. (JP-A-1-96198, JP-A-5-7497, JP-A-6-261784, etc.). Further, as fluorescent dyes for methamphetamine, those composed of a pentamethine cyanine derivative (JP-A-6-66725) and those composed of a merocyanine derivative (JP-A-8-92211) are known.
Therefore, as a test solution for detecting methamphetamine, an appropriately selected anti-methamphetamine antibody is labeled by binding an appropriately selected fluorescent labeling dye and dissolved in a PBS solution (phosphate buffer solution). Can be used as a test liquid (first test liquid).
When actually detecting methamphetamine, a fluorescence measurement kit in which a predetermined amount of such a first test solution is stored in the
If the sample is methamphetamine, the anti-methamphetamine antibody reacts with the antigen (methamphetamine) to quench the quenching. For this reason, the fluorescence intensity increases. Therefore, whether or not the sample is methamphetamine can be determined based on whether the intensity ratio is a certain level or more.
For example, suppose that white powder, which seems to be a prohibited drug, is attached to a package during customs inspection. The clerk decides that an inspection is necessary and temporarily collects the baggage before collecting the powder. And it measures as mentioned above and judges whether powder is methamphetamine. If it is determined to be methamphetamine, there is a high possibility that a large amount of methamphetamine is hidden somewhere in the baggage, so more detailed investigations and searches should be conducted. In addition to methamphetamine, prohibited drugs such as amphetamine and ***e can be similarly detected by preparing antibodies and fluorescent dyes, respectively.
Besides the control of prohibited drugs at customs, the fluorometer of this embodiment can be used. For example, when a prohibited drug is detected at a crime investigation site, a chemical substance left at the crime scene is identified and used as evidence. In addition to these, the fluorometer of the present embodiment can also be used in a doping test performed in, for example, a sports competition. In this case, a small amount of urine from the subject may be collected and used as a sample.
In such fluorescence measurement, the
In the case where the measurement is performed without first inserting the sample, and then the measurement is performed by inserting the sample, the noise of the leakage light can be removed by taking the difference between the two. However, in the case of fluorescence measurement using an antibody-antigen reaction, it is necessary to measure the rate of change in fluorescence intensity, and it is necessary to take a ratio between the two measurement results. When the ratio is taken, the noise of leaking light cannot be removed. If the amount of noise in the leaked light is always constant, it can be removed from the measurement results by measuring that amount in advance, but the intensity of the leaked light that has been measured in advance is always constant. Not limited to this, it easily fluctuates due to ambient temperature, light source deterioration, lens contamination, and the like. In addition, since the amount of leaked light is different for each photometer, the measurement result varies depending on the photometer even when measuring the same sample. Therefore, the significance of the structure of the embodiment for optically removing leakage light as described above is very large.
In addition, since the fluorometer of the present embodiment is portable, not only measurement in a limited place such as a laboratory or a measurement room, but also fluorescence measurement is performed in various other places, or samples are collected. Fluorescence measurement can be performed at the site and results can be obtained quickly.
The fluorometer of this embodiment is used for the control of prohibited drugs and criminal investigations described above, process monitoring in various plants, environmental investigations such as water quality inspection, research and development of new drugs, clinical diagnosis of various diseases, It can be used for the purpose of inspection of various foods and chemicals. Even in such surveys, research and development, various inspections, etc., there are many cases where fluorescence should be measured and identified and quantified quickly at the site where the sample is collected, and the fluorometer of this embodiment has utility value. Extremely high.
Next, the fluorometer of the second embodiment will be described. FIG. 9 is a schematic front sectional view of the main part of the fluorometer of the second embodiment.
In the second embodiment, the structure of the
