以前のアプローチと対照的に、発明者の目的は、その変異が陽イオン透過性を改変するWT ChR2内の残基、を特定することであった。発明者は、3番目の膜貫通ドメインに、このドメイン内のいくつかの変異している残基が光サイクルを改変し、チャネルを駆動する(gating)ものとして、着目した(図1)5−7、12。Arg115からThr139までの各々の残基は、個別にシステインに置き換えられ、アフリカツメガエル卵母細胞中で機能的な変化をスクリーニングされた。
CatChは以前説明される5、13ように、ピチア・パストリス内で発現され、精製された。閃光光分解(Flash−photolysis)研究が実施され、吸収変化が、エキシマー励起染料レーザー(excimer pumped dye laser)(450nm、2−3mJ)13からの10nsのレーザー閃光の励起後に測定された。
C末端切断型のChR2(L132C)−YFP(ベクター:pcDNA3(−)−chop2−309(L132C)−EYFP)は、HEK293細胞にトランスフェクトされ、常にG418選択下(0.6mg/ml;PAA Germany,Colbe,Germany)で保持された。野生型WT ChR2のために、C末端切断型のChR2−YFPベクター:pcDNA4TO−chop2−309−EYFP)がHEK293−Trex細胞(Invitrogen)に安定してトランスフェクトされ、以前に記載されたように13、培養され、誘導された。定常関係(stationary relation)のピークは、測定の24時間前にヒトコドン最適化pcDNA3.1(−)−ChR2−YFP構成体(WT、H134RまたはL132C)で一時的にトランスフェクトされたHEK293細胞(Effectene、QIAGEN)から決定された。
2−4MΩの抵抗をもつパッチピペットは、水平なDMZユニバーサルプーラー(シリアルNo.5318904120B、Zeitz−Instruments,Augsburg,Germany)上で薄肉のホウケイ酸ガラス(GB150−8P,Science Products,Hofheim,Germany)から作成された。光電流は全細胞パッチ・クランプ法で記録され、400μmの光ファイバーに焦点を合わせられたダイオード励起固体レーザー(Pusch Opto Tech GmbH,Baden Baden,Germany;λ=473nm)からの光パルスで活性化された。光パルスは速いコンピュータ制御シャッター(Uniblitz LS6ZM2,Vincent Associates)によって適用された。与えられたすべての光強度が、光導体の端で測定される。異なる陽イオンでの透過率の見積りを得るために、我々は、光電流−電圧関係を測定し、逆転電位を測定した。細胞内液は、140mM NaCl、7mM EGTA、2mM MgCl2および10mM Tris(pH=9)を含み、細胞外液は140mM NaCl、2mM MgCl2および10mM Tris(pH=9)を含んだ。陽イオン透過率のために、外部の140mM NaClは、140mM KCl、90mM CaCl2または90mM MgCl2でそれぞれ交換された。プロトン透過性は、pHが9から7.4(または6)に減少したときに、電流−電圧関係の逆転電位のシフトによって測定された。透過率比は、Na+、K+、H+およびCa++のための条件を含む、ゴールドマン−ホジキン−カッツ(GHK)の式により計算された。
実験が、以前に記載された14とおりにHEK293細胞上で実施され、室温(23℃)で実施された。ピペット液は1mM グアニジン−HCl、199mM NMG−Cl(N−メチルグルカミン)、10mM EGTA、2mM MgCl2および20mM Hepes(pH7.4)を含み、浴溶液は200mM グアニジン−HCl、2mM MgCl2、2mM CaCl2、および20mM Hepes(pH7.4)を含んだ。青色光刺激への電流応答は、飽和光条件(saturating light condition)下および再度の、−60mVでの電流応答が最大電流の半分であるところの光条件(I0.5;2kHz low−pass Bessel filter;サンプリングレート:100kHz;細胞直径:15μm)下での電圧ステッププロトコル(voltage step protocol)の適用下で記録された。−60mVの保持電位での持続的な照射(2分)の間の定常I0.5の記録は、単独チャネルコンダクタンスを見積もるのに使用された(2kHz low−pass Bessel filter;サンプリングレート20kHz)。照射無し(コントロール、3個の記録)および有り(光刺激開始の30秒後、2個の記録)の交互の記録は、集められ、フーリエ変換され、単独チャネルコンダクタンスがローレンツ関数での近似から見積もられた(詳細は14参照)。より低い光強度は、光駆動性チャネルの開口と閉鎖の最大限度の変動を得るために選ばれた。
細胞外に露出したシステイン残基のないC末端切断型のChR2変異体(残基1−315)(C34AおよびC36A変異を含む)は、ベクターpTLN27にサブクローニングされた。単一のシステイン変異は、QuickChange Site−Directed Mutagenesis (Stratagene)によって導入されて、配列決定により確認された。mRNAは、SP6 mMessage mMachineキット(Ambion, Austin, TX)を使用して調製された。30ngのWT ChR2/CatCh mRNAを含む、50nl cRNAが各々のアフリカツメガエル卵母細胞に注入された。部分的な卵巣摘出後のコラゲナーゼ処理で卵母細胞を得た。cRNA注入後に、卵母細胞は、全トランスレチナール(1μM、エタノール中の1mMストックより)中で培養され、1mg/ml ゲンタマイシンを含むORIバッファー(90mM NaCl、2mM KCl、2mM CaCl2および5mM Mops、pH7.4)中で、18℃にて2〜4日間保持された。
光電流は、75−Wキセノンアーク灯および450±25nmの帯域フィルターで活性化され、その光は〜1018光子s−1cm−2の出力の1mm光導体と結合された。作用スペクトルは、各波長あたり〜1.4×1017光子s−1cm−2のファイバー出力を達成するために、中性密度フィルターと組み合わせて狭帯域幅フィルター(398−645 nm; ± 10 nm ; K−series Balzer)を使用することで記録された。作用スペクトルの生成のため、CaCC電流を抑制するためにORI溶液中のCa++をBa++に置き換えた。各波長における電流振幅は、等しい光子露出を表すために正規化された。分光法で決定された規定スペクトル(ground spectrum)は、その後平均データポイントに合わせられた。カルシウム依存性塩素チャネル(CaCC)の活性化を抑制するために、速いCa2+キレート剤1,2−ビス(2−アミノフェノキシ)エタン−N,N,N’,N’−テトラ酢酸(BAPTA)の20mM溶液の50nlが、各卵母細胞に注入された(卵母細胞において、〜1mMの最終濃度)。
CatChイオン透過率の見積りを得るために、異なる陽イオンのための光電流電圧関係および逆転電位が、HEK293細胞において測定された。これらの実験は、ナトリウム、カリウム、およびマグネシウムのための透過率がWT ChR21に匹敵していることを明らかにした(図2d)。WT ChR2(pH/pNa=2.5*106)と比較して、CatCh(pH/pNa=4*106)のプロトン透過性はわずかに増加した。Ca++透過率(pCa/pNa)は、細胞外液の140mM Na+を90mM Ca++に置き換えたときの逆転電位シフトにより測定された。CatChの相対的なCa++−透過率が、−30.7±2.7mV(WT ChR2、平均±標準偏差、n=5)から−21.6±3.8mV(CatCh、平均±標準偏差、n=5;図2e)への逆転電位(二イオン電位(bi−ionic potential))のシフトからもわかるように、WT ChR2における0.15から0.24まで増加した。CatChの増加するCa++透過率をさらに定量化するために、我々は、CatChを発現しているHEK293細胞のFura−2カルシウムイメージングを実施し、340/380の測定比率をWT ChR2発現細胞で測定された比率と比較した。
Fura−2AM(5mM;Invitrogen)は室温で30分から1時間ロードされた。ローディングの後に、細胞はCa++なしの140mM NaCl溶液(140mM NaCl、7mM EGTA、2mM MgCl2、および10mM HEPES)で回収された。黄色蛍光タンパク質はそのYFP−蛍光から各細胞の発現レベルを見積もるために、460/40nmのフィルター(Visitron Systems、Puchheim、Germany)を用いる光に500ms露出することにより励起された。溶液はその後、90mM CaCl2、7mM EGTA、2mM MgCl2および10mM HEPESから成る細胞外Ca++溶液に置き換えられた。暗闇において15分後、光駆動性チャネルは10秒間、青色光(460/40nm)で刺激された。Fura−2は、340nm(340/20)および380nm(380/20)、およびCCDカメラ(全てのフィルターはVisitron Systems、Puchheim、Germany)で検出された放射光(540/80nm)で、励起された。
ニューロンへの適用のためのCatChの適合性を試験するため、構成体は培養された海馬錐体細胞の中で発現された。
海馬は出生後P1 Sprague−Dawleyラット(Jackson Laboratory)から取り出され、パパイン(20Uml−1)で37℃にて20分間処理された。海馬は10%の牛胎児血清が補われたDMEM(Invitrogen/Gibco、高グルコース)で洗浄され、少量のこの溶液中で粉砕された。〜7万5000個の細胞が、24ウェルプレート中のポリ−D−リジン/ラミニンでコートされたカバーガラス上に播種(plate)された。3時間後に、プレート培地(plating medium)は、培養培地(2% B−27補足剤、2mM Glutamax−I、および100U/mlペニシリンおよび100μg/mlストレプトマイシンを含むNeurobasal A)に置き換えられた。ChR2(L132C)−YFPおよびChR2(WT)−YFPは、播種の5〜10日後にリポフェクタミン2000試薬(Invitrogen)を使用してトランスフェクトされた。あるいは、2〜5×109GC/mlのウイルス(AAV2/7−CAG−ChR2(L132C)−2A−EGFP−WPRE−bGH)が播種の4〜9日後に各々のウェルに加えられた。発現は、形質導入後5日間で目に見えるようになった。培養物の生存期間(〜5週間)について、神経毒症状は全く見られなかった。ここで記載されるいかなる実験においても、全トランスレチナールは培地または記録培地(recording medium)に全く添加されなかった。
サイトメガロウィルス初期エンハンサー/ニワトリβアクチン(CAG)プロモーターは、PCR増幅され、pAAV2−CAG−EGFPを入手するために、pAAV2−Rho−EGFP(Alberto Auricchioからの親切な贈り物28)に挿入された。pAAV2−CAG−EGFPウイルス発現プラスミドはさらに、ウッドチャック転写後調節要素(WPRE)および牛成長ホルモン(BGH)ポリアデニル化配列を含んだ。ChR2(L132C)−2A−EGFP(Volker Busskampoからの親切な贈り物−2A自己切断型ペプチド/CHYSEL29)はアダプターPCRによって構築され、Clontechのin fusion kitを使用して、EGFPの置換により、pAAV2−CAG−EGFPにサブクローニングされた。ウイルスベクター(pAAV2−CAG−ChR2(L132C)−2A−EGFP−WPRE−bGH)はパッケージ化され(血清型7)、親和性はペンシルバニア大学のGene Therapy Programで2.26×1011ゲノムコピー/mlの最終的な感染性ウイルス測定濃度で精製した。
海馬培養神経細胞の全細胞記録のために、5〜10Ωの抵抗をもつパッチピペットが、129mMグルコン酸カリウム、10mM KCl、4mM MgATPおよび0.3mM Na3GTPで満たされ、pH7.2に滴定された。タイロード溶液は細胞外溶液として用いられた(125mM NaCl、2mM KCl、2mM CaCl2、1mM MgCl2、30mMグルコースおよび25mM HEPES、pH7.4に滴定)。名目上はCa++無しの細胞外溶液は、0mM Ca++および3mM Mg++を有することを除いて、この同じ溶液を含んだ。記録は、興奮性シナプス伝達遮断剤、1,2,3,4−テトラヒドロ−6−ニトロ−2,3−ジオキソ−ベンゾ[f]キノキサリン−7−スルホンアミド(NBQX,10μM,Sigma)およびD(−)−2−アミノ−5−ホスホノペンタン酸(AP−5,50μM,Sigma)の存在下で行われた。電圧固定記録のため、1μMのテトロドトキシンが細胞外溶液に添加された。BKチャネル活性を阻害するため、1mM TEAが加えられた。記録は、蛍光ランプを備える倒立Zeiss Axiovert 25顕微鏡上で行われた。成功したタンパク質の発現はEGFPまたはYFPによってもたらされる蛍光によって立証された。ニューロンのアクセス抵抗は、15〜40MΩであり、実験全体にわたって、安定性のために監視された。電気生理学的信号は、Axopatch 200A増幅器(Axon Instruments,Union City,CA)を使用することで増幅され、10kHzでフィルターにかけられ、Axon Digidata1600(50Hz)でデジタル化され、かつpClamp9ソフトウェア(Axon Instruments)を用いて入手および分析された。光電流は、ダイオード励起固体レーザー(Pusch Opto Tech GmbH;λ1=473nm,P1=100mW,λ2=532nm,P2=50mW)からの様々な波長の光パルスまたはエキシマー励起色素レーザー(Coumarin 2,λ=450nm)からの10ns閃光により誘起された。特定の光強度は、図の注釈および文書に示され、細胞から〜500μmの距離にて、400μm直径の石英光ファイバー(STE−F100/400−Y−VIS/NIR; Laser 2000,Wessling,Germany)の末端での強度である。ChR2(L132C)−YFPおよびChR2(L132C)−2A−EGFPを発現するニューロンから測定された電流は同じである。
イメージングについて、海馬ニューロンがあるカバーガラスは、2%スクロースを含むPBSバッファー中の4%パラホルムアルデヒド中で10分間、4℃にて固定された。細胞は続いて、ウサギα−GFPIgG(Invitrogen、A11122)中で1.5時間培養され、その後Alexa Flour 488ロバ−α−ウサギIgG(Invitrogen、A21206)中で45分間培養された。取り付けられたカバーガラスの免疫蛍光はZeiss LSM510共焦点顕微鏡(Zeiss、Plan−Neofluar 40×/0.75)上で撮影された。
一見したところでは、CatCh、WT ChR2のL132C変異体、WT ChR2と比較してむしろ平凡な結果を示している:1)開口状態の存続期間の2倍の増加、2)光サイクル動態における、P520中間体の減速された減衰、3)変化しない単独のチャネルコンダクタンス、および4)わずかに赤にシフトした吸収極大(4nm)。2.5倍に増加した光電流は、発現レベルの増加と全く関連のないパラメータで容易に説明できる。しかしながら、もう一度見てみると、アフリカツメガエル卵母細胞を発現するCatChから得られた電圧固定法データのさらなる精査は、逆転電位の測定およびHEK293細胞上のカルシウムイメージング実験でその後確認された、上昇したCa++透過性を与えた。図1におけるモデルから判断すると、Ca++透過性の増加は、光駆動プロトンポンプ細菌ロドプシンのL94A変異体について示されるように(図1と比較)21、より柔軟な構造の構成およびその結果としての空洞の形成により容易にされ得る。この空洞は、C128から1らせん回転分だけ離れた、保存された膜貫通ヘリックス3(TM3)の一部として疎水性パッチ中に位置するであろう。C128(TM3)およびD156(TM4)の間の相互作用の操作は、ChR2の反応サイクルを劇的に減速し5、6、その効果は細菌ロドプシン変異体L93A22、23、すなわちL94の隣接残基でも観察された。ChR2では、TM3およびTM4の相互作用は、イオン孔への明反応のトランスデューサーとしての構造要素24を指す、開閉(gating)および選択性の両方に影響するように見える。より小さい、より親水性のシステインの挿入は螺旋状のセグメントの柔軟性を高め、Ca++のアクセスを容易にし得る。

- 配列番号1(CHOP−2)の1−309位置に示されるアミノ酸配列と少なくとも70%の相同性を有するアミノ酸配列を含み、かつ配列番号1においてL132に相当する位置での置換を含み、該置換がチャネルの極性を増加するものである、光誘起イオンチャネル。
- L132の置換を除いて、配列番号1(CHOP−2)の1−309位置に示されるアミノ酸配列を含む、好ましくはからなる、該置換がチャネルの極性を増加するものである、請求項1に記載の光誘起イオンチャネル。
- 置換が、L132C、L132S、L132E、L132DおよびL132Tから選択され、好ましくは置換がL132Cである、請求項1または2に記載の光誘起イオンチャネル。
- a)光感受性が、海馬ニューロンにおいて野生型CHOP−2と比較して5倍より多く、好ましくは10倍より多く、より好ましくは20倍より多く、例えば30倍より多く、さらにより好ましくは40倍より多く、例えば50倍より多く、もっとも好ましくは60倍より多く、または70倍より多く増加する;かつ/または
b)HEK293細胞でのFura−2−イメージングによって測定される、カルシウム伝導度が、野生型CHOP−2と比較して少なくとも2倍、好ましくは少なくとも3倍、より好ましくは少なくとも4倍増加される;かつ/または
c)海馬ニューロンにおいて全細胞電気生理学的記録により測定した野生型CHOP−2と比較して、少なくとも1.5倍、より好ましくは2倍、またはさらにより好ましくは2.5倍増加した刺激頻度を示す、
前記請求項のいずれかに記載の光誘起イオンチャネル。 - さらに、少なくとも1個の以下のアミノ酸残基:配列番号1の位置253に対応する位置のアスパラギン酸;配列番号1の位置257に対応する位置のリジン;配列番号1の位置260に対応する位置のトリプトファン;配列番号1の位置123に対応する位置のグルタミン酸;配列番号1の位置134に対応する位置のヒスチジンまたはアルギニン、好ましくはアルギニン;配列番号1の位置128に対応する位置のトレオニン、セリン、またはアラニン;および/または配列番号の位置156に対応する位置のアラニン、を含む、前記請求項のいずれかに記載の光誘起イオンチャネル。
- 共通モチーフ、L(I)DxxxKxxW(F,Y)を含む、前記請求項のいずれかに記載の光誘起イオンチャネル。
- 請求項1〜6のいずれか1項に記載の光誘起イオンチャネル、およびレチナールまたはレチナール誘導体を含む、チャネルロドプシンであって、レチナール誘導体は3,4−デヒドロレチナール、13−エチルレチナール、9−dm−レチナール、3−ヒドロキシレチナール、4−ヒドロキシレチナール、ナフチルレチナール;3,7,11−トリメチル−ドデカ−2,4,6,8,10ペンタエナール;3,7ジメチル−デカ−2,4,6,8−テトラエナール;3,7−ジメチル−オクタ−2,4,6−トリエナール;および6−7回転阻止レチナール、8−9回転阻止レチナール、および10−11回転阻止レチナールからなる群から選択される、チャネルロドプシン。
- 請求項1〜6のいずれか1項に記載の光誘起イオンチャネルをコードする核酸配列を含む、核酸構成体。
- 請求項1〜6のいずれか1項に記載の光誘起イオンチャネルをコードする核酸配列または請求項8に記載の核酸構成体を含む発現ベクターであって、好ましくは該ベクターは遺伝子治療、特にウイルス媒介性の遺伝子導入に好適である、発現ベクター。
- 請求項7に記載のチャネルロドプシン、請求項8に記載の核酸構成体または請求項9に記載の発現ベクターを含む、細胞。
- 哺乳動物細胞または昆虫細胞、または酵母細胞、好ましくはサッカロマイセス・セレビシエ、シゾサッカロミセス・ポンベまたはピチア・パストリスからの酵母細胞である、請求項10に記載の細胞。
- 哺乳動物細胞が、
(a)光受容細胞、網膜桿状体細胞、網膜錐体細胞、網膜神経節細胞、双極ニューロン、神経節細胞、偽単極ニューロン、多極ニューロン、ピラミッド状ニューロン、プルキンエ細胞、または、顆粒細胞;または
(b)メラノーマ細胞、COS細胞;BHK細胞;HEK293細胞;CHO細胞;骨髄腫細胞;またはMDCK細胞、
である、請求項11に記載の細胞 - 薬剤としての使用のための、請求項1〜6のいずれか1項に記載の光誘起イオンチャネル、または請求項7に記載のチャネルロドプシン、または請求項8に記載の核酸構成体、または請求項9に記載の発現ベクター、または請求項10に記載の細胞。
- 遺伝子治療における使用のための、請求項10に記載の発現ベクター。
- 失明または視力減少の治療における使用のための、請求項1〜6のいずれか1項に記載の光誘起イオンチャネル、または請求項7に記載のチャネルロドプシン、または請求項8に記載の核酸構成体、または請求項9に記載の発現ベクター、または請求項10に記載の細胞。
- がん細胞、好ましくは黒色腫のがん細胞の切除における使用のための、配列番号1の位置128に相当する位置にスレオニン、セリンまたはアラニンを有し、かつ/または配列番号1の位置156に相当する位置にアラニンを有する、請求項5に記載の光誘起イオンチャネル。
- 請求項1〜6のいずれか1項に記載の光誘起イオンチャネル、または請求項7に記載のチャネルロドプシン、または請求項10に記載の細胞の、高スループットのスクリーニングにおける使用。
