本出願は、2010年6月7日出願の欧州特許出願第10165108.1号に基づく優先権を伴う出願であり、この内容は参照され本明細書に援用される。
しかし、カーボンナノチューブの比較的コストが高いこと、および残留金属ナノ粒子及び/又はアモルファスカーボンなどの不純物が存在することにより、工業的規模で使用されることが妨げられている。
ことはできないことを観察した。
− 激しい酸化条件、
− カーボンナノホーンの酸化(凝集体及び個別の形で)、
− 個別化カーボンナノホーンを、前記プロセスで形成された種々の酸化種から分離するための手のかかるプロセス、
−非常に低い収率、などである。
(a) 原料カーボンナノホーンを、アルカリ金属により還元してカーボンナノホーンアルカリ塩とし;及び
(b) 前記カーボンナノホーンアルカリ塩を、極性非プロトン性溶媒中で、還元カーボンナノホーンの溶液を製造する、ことを含む。
1つの実施態様では、Xは、F、Cl、Br又はI又はトシレートを表す。
新たなかつ効果的な薬物輸送システムの開発は、多くの種類の治療薬の薬学的プロフィルを改良するために本質的に重要である。現在は、多くの異なる種類の薬物輸送システムが利用可能である。ナノ材料のうちでは、カーボンナノチューブ(CNT)が、治療分子を輸送するための新たな代替的効果的道具として出現してきた。
他の実施態様では、前記ナノホーンは機能化されてその表面に多数の生物分子の結合を可能にする。この実施態様では、「機能化」ナノホーンとは、その表面が、前記ナノホーン生物活性化合物の負荷を直接輸送及び/又は放出を制御することが可能となるように修飾されていることを意味する。
本発明の個別化されたカーボンナノホーンは多くの異なる生物活性化合物又は異なる生物活性化合物の組み合わせを輸送するために使用され得る。この領域でのカーボンナノホーンの応用についての議論は、例えば、Whitney et al.(2011)[62]及びそこで引用されている文献中に見出すことができる。本発明は、カーボンナノホーン内に含まれる少なくとも1つの生物活性化合物を輸送するために個別化されたカーボンナノホーンの使用を含む。例えば、2以上の生物活性剤の組み合わせを輸送するために、それらを同じカーボンナノホーン内に負荷することができる。
本発明のナノホーン輸送システムは医薬組成物に組み込まれ得る。かかる組成物は通常は、生物活性化合物及び医薬的に許容される担体を含むナノホーン輸送システムを含む。「医薬的に許容される担体」は、全ての種類の溶媒、分散媒体、コーティング、抗菌及び抗真菌剤、等張剤及び吸収遅延剤など、医薬投与と整合するものを含む(Gennaro、A.R.2000.Remington:The science and practice of pharmacy.Lippincott、Williams&Wilkins、Philadelphia、Pa.[22])。そのような担体の例は、限定されるものではないが、水、生理的食塩水、フィンガー液、デキストロース溶液及び5%ヒト血清アルブミンを含む。リポソーム及びオイルなどの非水溶性担体も使用され得る。従来の媒体又は試薬が活性化合物と適合性がない場合以外はこれらの組成物の使用が含まれる。補助的な活性化合物もまた、前記組成物に含まれ得る。
本発明は、次の1又はそれ以上を、1つのパッケージ又は容器に入れたキットを提供する:(1)本発明のナノホーン輸送システムに含まれた活性剤を含む組成物;(2)医薬的に許容される補助剤又は賦形剤;(3)注射器又はカテーテルなどの投与道具;(4)投与指示書である。2又はそれ以上の成分(1)〜(4)が同じ容器に入れられる実施態様もまた含まれる。
本発明の方法により得られ得る還元カーボンナノホーン溶液又は個別化されたカーボンナノホーンは、カーボンナノホーン上、好ましくは個別化されたカーボンナノホーン上に担持される触媒を調製するために使用され得る。
比較結果が図6にまとめられており、本発明の触媒は従来の触媒よりも優れていることが示される。
自動車用電源として水素系燃料電池の使用の欠点は、許容される軽量かつ安全な水素貯蔵媒体がないということである。水素貯蔵の4つの従来方法がこれまで使用されてきた:(a)液化水素、(b)圧縮ガス、(c)低温吸着及び(d)金属水素化物貯蔵システムである。これらの既存の方法の簡単なまとめを以下に与えられる。
(a) 容器;
(b)前記容器内に設けられた水素貯蔵床を含み、前記水素貯蔵床は還元カーボンナノホーン又は開口カーボンナノホーンを含む。
(c)ステップ(b)で得られた溶液を凍結するステップ;及び
(d)非プロトン性溶媒を昇華させて、個別化されたカーボンナノホーンのエアロゲルを得るステップ、を含む。
光電池は、光導電性材料内部で光子と電子の相互作用により太陽光を電気へ変換する。光電池を作るために光導電性材料、通常はシリコンが電子接触により結合されてジャンクションを形成する。現在、大抵のシリコン系光電池はシリコンp−nジャンクション装置である。光子が電池に衝突することで不整合電子をはじき出し前記ジャンクションを横切って動く電流を生成する。これらの電子接触の格子は、電流が集められる電池の配列を形成する。電池内で生成されたDC電流は、関与する材料及び電池に入射する放射線のエネルギー及び強度に依存する。
ここで、GはC1−12アルキル基;Xはハロゲン原子;及びLは金属イオンを表す。Lはポルフィリン基に配位可能な全ての金属である。Advanced inorganic chemistry、5th edition、P.354、1988 Cotton and Wilkinson([42])によれば、周期律表のほとんどの金属がポルフィリンに配位可能である。
− この合成は金属触媒を必要としない(カーボンナノチューブと異なる)。従って、カーボンナノホーンは本質的にカーボンナノチューブに比較してより高い純度を持つ。
− カーボンナノホーンの毒性はカーボンナノチューブに比較して低い。これはこの合成方法が金属を含まないことに部分的による(痕跡量金属を含まない)。加えて、最近の毒性学的報告では、カーボンナノチューブに最初に伴うほとんどの毒性は実際にその金属含有量に依存することを示している。さらに、マルチウオールカーボンナノチューブの場合には、その形状及び寸法から毒性が懸念され(長さが数ミクロンから数百ミクロンで、直径が数十ミクロンから数100ミクロン)、これらはあるアスベストの形状に類似する。カーボンナノホーンの寸法は非常に異なり(長さ約20から40nmで直径が2から5nm)、従ってこれらはアスベスト様の寸法、形状ではない。事実Miyawakiらはカーボンナノホーンは低毒性であることを確認した(ACS Nano、2008、2、213−226[38])。
− カーボンナノホーンは、カーボンナノチューブに比較してより優れた薬物輸送担体となり得る、というのはそのサイズが(a)前記粒子をカーボンナノチューブのように切断する必要がなく、及び(b)カーボンナノホーン(2−4nm直径)に比べてCNTの直径が約1.4nmである。CNHは従って、本質的により高い負荷容量を持つ。
− CNHsはDMSO中でCNTに比べて30倍位上溶解性がある。
− CNHsは、カーボンナノチューブ(CNT)に比べてより広い種類の溶媒に溶解され得る。
特に記載のない限り、全ての実施例は不活性雰囲気下、例えばアルゴン又は窒素雰囲気下で行った。具体的には、操作は、乾燥アルゴン雰囲気下でグローブボックス内(O2含有量10ppm未満、H2O含有量10ppm未満)で行われる。原料カーボンナノホーン及び化学試薬は高真空下で乾燥され、脱ガスされた後グローブボックス内に入れた。
第1の溶液:
ナフタレンカリウム塩は、0.040g(過剰量)のカリウム片を、0.0668gの昇華ナフタレンの乾燥THF140mL溶液中に、不活性雰囲気下で添加することで製造された。混合物を一晩還流させた。その後、前記混合物をろ過して過剰のカリウムを除去した。ナフタレンカリウム塩溶液(暗緑色)の重量が123.249g(THF比重=0.8892)であり、これから[K+]の濃度は3.75mMと計算された。
上の実験手順を続けて、第2のナフタレンカリウム塩溶液を製造した(K:0.100g、ナフタレン:0.1336g、THF:150mL)。前記新たなナフタレンカリウム塩溶液の重量は123.653g(THFの比重=0.8892)であり、これから[K+]の濃度を7.5mMと計算された。
第1の溶液:KC8_NHs_01
前記第1のTHF中のナフタレンカリウム塩溶液([K+]=3.75mM)をCNHs(48mg)上に加えて、室温で一晩不活性雰囲気下撹拌した。混合物をナイロン膜(孔サイズ0.22μm)でろ過し、残渣が無色になるまで蒸留THFで繰り返し洗浄した。残渣固体を真空下で30分間乾燥し、前記膜から集めて76mgの還元CNHsを得た。59mgのr−CNHsを59mLの蒸留DMSOに溶解した(1mg/mL)。前記還元CNHsの溶解は自発的であった。黒色溶液を室温で、不活性雰囲気下で一晩撹拌した。その後、還元CNHs溶液を、速度4000rpmで1時間遠心した。CNHsの沈殿はほとんど見られなかった。前記溶液をフラスコに入れて、不活性雰囲気下保存した。
THF中のナフタレンカリウム塩の前記第2の溶液([K+]=7.5mM)をCNHs(1mmg)上に入れて一晩室温で不活性雰囲気下に撹拌した。r−CNHsの前記新たな溶液の処理は前記のように行い、125mgの還元CNHsを得、それを蒸留DMSO125mL中に自発的に溶解し(1mg/1mL)、不活性雰囲気下で一晩撹拌した。遠心(1時間/速度:4000rpm)後、前記溶液を3つのフラスコに分けて、不活性雰囲気下で保存した。
1. CNHs−ベンジル生成物(KC8_NHs_01_ベンジル)の製造
次のようにして、前記r−CNHs(KC8_NHs_01)のDMSO溶液をヨードベンジルと処理してベンジル−CNHsを得た。1mLDMSOに希釈したヨードベンジル(300μL)を、不活性雰囲気下でr−CNHsの前記DMSO溶液中に添加した。前記反応混合物を室温で48時間以上撹拌した。r−CNHsの機能化後には沈殿も懸濁も見られなかった。前記DMSO溶液を一晩冷凍庫で凍結された。解凍するとCNHsが沈殿した。前記懸濁液をナイロン膜(孔サイズ0.2μm)で濾過し、DMSO及びメタノールで洗浄した。残留固体を蒸留水で洗浄してKIを除去し、その後メタノール及びCH2Cl2で洗浄した。前記残渣固体を室温で真空化で乾燥し、その後一晩凍結乾燥して19.5mgの機能化CNHsを得た。
次のようにして、前記r−CNHs(KC8_NHs_02)のDMSO溶液の前記第1の部分をヨードメタンと処理してメチル−CNHsを得た。1mLDMSOに希釈したヨードメタン(100μL)を、不活性雰囲気下でr−CNHsの前記DMSO溶液中に添加した。前記反応混合物を室温で48時間以上撹拌した。前記懸濁液をナイロン膜(孔サイズ0.2μm)で濾過し、DMSO及びメタノールで洗浄した。残留固体を蒸留水で洗浄してKIを除去し、その後メタノール及びCH2Cl2で洗浄した。前記残渣固体を室温で真空化で乾燥し、その後一晩凍結乾燥して26mgの機能化CNHsを得た。
次のようにして、前記r−CNHs(KC8_NHs_02)のDMSO溶液の前記第2の部分をヨードオクタンと処理してC8H17−CNHsを得た。1mLDMSOに希釈したヨードオクタン(200μL)を、不活性雰囲気下でr−CNHsの前記DMSO溶液中に添加した。前記反応混合物を室温で48時間以上撹拌した。前記懸濁液をナイロン膜(孔サイズ0.2μm)で濾過し、DMSO及びメタノールで洗浄した。残留固体を蒸留水で洗浄してKIを除去し、その後メタノール及びCH2Cl2で洗浄した。前記残渣固体を室温で真空化で乾燥し、その後一晩凍結乾燥して17.6mgの機能化CNHsを得た。
次のようにして、r−CNHs(KC8_NHs_02)のDMSOの最後の部分をメチルメタクリレートと処理し次のようにPMMA−CNHsを得た。10mLTHFに希釈したメチルメタクリレート(300μL)を、不活性雰囲気下でr−CNHsの前記DMSO溶液中に滴下した。前記反応混合物を室温で48時間以上撹拌した。前記反応終了後、メタノール10mLを混合物に加えた。r−CNHsの機能化後、沈殿も懸濁も見られず、そこでKC8_NHs_01_ベンジルの場合と同様に前記溶液を凍結した。前記懸濁液をナイロン膜(孔サイズ0.2μm)で濾過し、DMSO、メタノール及び最後にCH2Cl2で洗浄した。前記残渣固体を室温で真空化で乾燥し、その後一晩凍結乾燥して35.1mgの機能化CNHsを得た。
4.1mgの還元CNHsを5mLのDMSOに溶解し、その後一晩空気でバブリングしてCNHsへ酸化した。前記懸濁液をナイロン膜(孔サイズ0.2μm)で濾過し、メタノール及びCH2Cl2で洗浄した。前記残渣固体を室温で真空化で乾燥し、その後一晩凍結乾燥した。
40mLのDMSOを40mgのCNHの塩に注ぎ一晩撹拌した。弱遠心(2900g、1時間)を行い不溶解物質を除去した(少量)。この溶液に65mgのPd(Ac)2(40%過剰)を添加した。7分後、生成物(CNHsに担持されたPdナノ粒子)をPVDF膜(0.45μm孔サイズ)で濾過し、DMSOで洗浄した。
(a) Suzukiカップリング反応
CNH触媒:
5mgのナノPd−CNHs(CNHs上1%重量/重量Pd)をDMF(6ml)中に分散させ、そこに1−ヨードベンゼン(0.2mmol)、フェニルボロン酸(0.24mmol)及び炭酸ナトリウム(0.4mmol)を添加した。得られた混合物を110℃で3時間撹拌した。冷却後、反応混合物を0.1μmPTFE濾過膜で濾過した。前記ナノPd−CNHs触媒を回収し、有機不純物を除去するために十分洗浄して2回以上の触媒サイクルで再度使用した。
パラジウム触媒アリールハロゲン化物とアリールボロン酸の炭素−炭素カップリング反応は既に記載されている[43−48]。
CNH触媒:
3mgのナノPd−CNHs(CNHs上Pd1%重量/重量)をDMF(5ml)中に分散させ、そこにヨードベンゼン(0.1mmol)とトリブチルフェニルスズ(0.12mmol)を添加した。混合物を加熱して110℃で12時間撹拌した。冷却後、反応混合物を0.1μmPTFE濾過膜で濾過した。これによりナノPd−CNHs触媒を回収し、有機不純物を除去するために十分洗浄して2回以上位の触媒サイクルで再度使用した。濾液を50mL水に投入し、酢酸エチル(30ml)で3回抽出した。有機抽出層を集めて無水硫酸ナトリウムで乾燥し、減圧で蒸発させた。前記Stille反応の生成物、即ちビフェニルが得られ、これは1H−NMRスペクトルで確認された。
パラジウム触媒アリールハロゲン化物と有機スズの炭素−炭素カップリング反応は既に記載されている [43−45、49]。
CNH触媒
3mgのナノPd−CNHs(CNHs上Pd1%重量/重量)をDMF(5ml)中に分散させ、そこに4−ヨードアニソール(0.1mmol)とオレフィン基質(スチレン、0.2mmol)を添加した。得られた分散物を130℃に予熱されたオイルバスに入れ、5時間撹拌した。その後、冷却して反応混合物を0.1μmPTFE濾過膜で濾過した。これによりナノPd−CNHs触媒を回収し、有機不純物を除去するために十分洗浄して2回以上位の触媒サイクルで再度使用した。濾液を50mL水に投入し、酢酸エチル(30ml)で3回抽出した。有機抽出層を集めて無水硫酸ナトリウムで乾燥し、減圧で蒸発させた。前記Heck反応の生成物、即ち1−メトキシ−スチリルベンゼンが得られ、これは1H−NMRスペクトルで確認された。
パラジウム触媒芳香族とビニル系化合物の炭素−炭素カップリング反応は既に記載されている[43、51−54]。
ヨードベンゼン及び4−ヨードアニソールが前記カップリング反応の基質として使用され、カップリング反応をCNHs上の1%重量/重量のPdを一定として実施された。フェニルボロン酸とのSuzukiカップリング反応は加熱下で僅か3時間で起こり、n−トリブチルスズとのStilleカップリング反応は12時間、またスチレンとのHeck反応は5時間以内の生成物が得られた。全ての場合で、前記触媒は。テフロン膜(PTFE、0.2μm孔サイズ)で濾過され、DMF、メタノール、ジエチルエーテルで十分洗浄後乾燥させて次の触媒サイクルで再使用され、少なくとも3回の触媒サイクルでカップリング生成物の形成に良好な作用を示した。重要なことは、制御された実験では、不溶性のそのままのCNHsが使用された場合、反応は生じなかったことであり、このことは、試験された反応でC−C結合形成を触媒することにおいて、可溶化CNHs上に固定化されたPd−ナノ粒子の溶解性と同様に、Pd−ナノ粒子の重要性を明らかにしている。溶解性CNHs上に固定化されたPd−ナノ粒子について見出された優れた触媒活性は、従って、準均一系触媒反応で可溶化CNHsの表面に固定化されたPd−ナノ粒子の高い表面積ということで説明され得る。

Claims (17)
- カーボンナノホーンを安定化する方法であり、不活性雰囲気下で実施される次のステップ:
(a)原料カーボンナノホーンをアルカリ金属で還元してカーボンナノホーンアルカリ塩とするステップ;及び
(b)前記カーボンナノホーンアルカリ塩を非プロトン性極性溶媒に入れて還元カーボンナノホーンの溶液にするステップを含む、方法。 - 請求項1に記載の方法であり、前記カーボンナノホーンアルカリ塩が、式KC8の二元組成物の形である、方法。
- 請求項1又は2のいずれか一項に記載の方法であり、前記ステップ(a)が、非プロトン性であり、その構造が少なくとの1つの酸素原子を含む親核性溶媒の存在下で実施される、方法。
- 請求項1乃至3のいずれか一項に記載の方法であり、前記還元ステップ(a)が、不活性雰囲気下で式A+B−(A+がアルカリイオンのカチオンを表し、B−が多芳香族化合のアニオンを表す)のポリアリールアルカリ塩を原料カーボンナノホーンへ添加することを含む、方法。

- 請求項1乃至5のいずれか一項に記載の方法であり、前記極性非プロトン性溶媒が、ジメチルスルホキシド(DMSO)、N−メチル−2−ピロリジノン(NMP)、ジメチルホルムアミド(DMF)、アセトン、アセトニトリル、ベンゾニトリル、1、2−ジクロロベンゼン、クロロベンゼン、スルホラン、シクロペンタノン、シクロヘキサノン、N−ホルミルピペリジン(NFP)、ビニルピロリドン(NVP)、1、3−ジメチル−2−イミダゾリジノン(DMEU)、ブロモベンゼン又は安息香酸ベンジルである、方法。
- 請求項1乃至6のいずれか一項に記載の方法であり、さらに:
(c)ステップ(b)で得られる前記溶液を凍結するステップ;及び
(d)前記非プロトン性溶媒を昇華させて、個別化されたカーボンナノホーンのエアロゲルを形成するステップ、を含む方法。 - 請求項7により得られ得る個別化されたカーボンナノホーンのエアロゲル。
- 請求項1乃至6のいずれか一項に記載の方法で得られ得る還元カーボンナノホーンの溶液。
- 請求項1乃至6のいずれか一項に記載の方法がさらに:前記極性非プロトン性溶媒を蒸発させるステップを含む、方法。
- 請求項10に記載の方法で得られ得る個別化されたカーボンナノホーン。
- 請求項1に記載の方法であり、さらにステップ(b)で得られる還元カーボンナノホーンを式R−Xの化合物(ここで、Xは脱離基を表し、Rは、C1−10アルキル、C2−10アルケニル、C2−10アルキニル、C1−10アルキル、C1−10アルキルカルボニル、C6−10アルールカルボニル又はC1−10ヘテロアリールカルボニル基を表し、前記基は場合により置換されていてもよい)と反応させるステップを含む、方法。
- 請求項12に記載の方法により得られ得る機能化個別化されたカーボンナノホーン。
- 請求項11乃至13のいずれか一項に記載のカーボンナノホーンを薬物輸送システムとしての使用。
- 請求項11乃至13のいずれか一項に記載のカーボンナノホーンをカーボンナノホーン担持触媒のための使用。
- 請求項11乃至13のいずれか一項に記載のカーボンナノホーンを水素貯蔵システムの製造のための使用。
- 請求項11乃至13のいずれか一項に記載のカーボンナノホーンを光電池の製造のための使用。
