Abstract
Description
The present invention relates to a measurement chip for analyzing a sample by an interferometry using light and a method for manufacturing the same.
Conventionally, the measurement of bonds such as intermolecular interactions between biomolecules such as antigen-antibody reactions and intermolecular interactions between organic macromolecules is generally performed by using labels such as radioactive substances and phosphors. I have been. This labeling takes time, and in particular, labeling a protein may involve complicated methods or the property of the protein may change depending on the labeling. Therefore, in recent years, as a means for directly detecting a bond between a biomolecule and an organic polymer directly without using a label, the RIfS method (Reflectometric interference spectroscopy) using interference color change of an optical thin film is used. Spectroscopy) is known (see, for example, Patent Document 1).
According to the description in
Further, instead of putting the sensor chip into the reaction vessel, a transparent silicone rubber substrate with a groove is stacked on the sensor chip. By covering the groove of the silicone rubber substrate with the sensor chip, the groove is used as a channel, and the sample solution is circulated through the channel. Then, a biochemical substance layer in the sample solution is formed on the surface of the sensor chip, and the interference wavelength changes. In order to measure the interference wavelength, light is transmitted through the silicone rubber substrate, and the surface of the sensor chip is irradiated with the light. Then, since the reflected light on the surface of the sensor chip passes through the silicone rubber substrate, the spectral intensity of the light is measured.
However, the silicon substrate that is the base material of the sensor chip is hard and brittle. Therefore, if an impact or the like is given to the sensor chip, the sensor chip is damaged. It is necessary to handle the sensor chip carefully, making it difficult to handle the sensor chip.
Further, in order to improve the sealing performance between the sensor chip and the silicone rubber substrate stacked thereon, it is necessary to sandwich the silicone rubber substrate and the sensor chip with a certain load. As a result, the sensor chip may be damaged. Furthermore, since the groove depth of the silicone rubber substrate changes due to the compression of the silicone rubber substrate, the optical path length also changes, and the reproducibility of the spectral intensity measurement is not good. In particular, since the thickness of the bottom of the groove cannot be increased so as not to deteriorate the optical characteristics, if a three-dimensional shape such as a groove is formed on a substrate made of a soft material, the influence of deformation during compression is large.
Therefore, an object of the present invention is to provide a measuring chip capable of enhancing the reproducibility of spectral intensity measurement and a method for manufacturing the same. Furthermore, an object of the present invention is to provide a measuring chip with high impact resistance and a method for manufacturing the measuring chip.

According to a fourth aspect of the present invention, a sacrificial film is formed on a single crystal substrate, a single crystal high refractive index film is formed on the sacrificial film, and the high refractive index film is formed at the bottom of a groove formed in the substrate.
According to the present invention, since the single crystal high refractive index film is transferred to the bottom of the groove of the substrate, the substrate does not have to have a low impact resistance such as a single crystal silicon substrate. The impact resistance of the measuring tip can be increased. Therefore, it is easy to handle the measuring chip.
Furthermore, it is not necessary to make the substrate easily deformable, such as rubber, and the change in the depth of the groove can be suppressed. Therefore, when the interference film or high refractive index film is irradiated with light from above the groove, the optical path length does not change, and the reproducibility of the measurement of the spectral intensity of the reflected light reflected by the interference film and high refractive index film Will increase. Therefore, the spectral intensity can be measured accurately and stably.

FIG. 2 is a cross-sectional view of the
The

An
A high

The
3 to 5, the
As shown in FIG. 7, the

A method for manufacturing the
First, as shown in FIG. 10A, a
On the other hand, as shown in FIG. 10B, a
Next, as shown in FIG. 10C, a high refractive index film (for example, a single crystal silicon film or a single crystal germanium film) 23 is formed on the
Next, as shown in FIGS. 10D and 10E, the high
For joining the bottom 22a of the
After the transfer of the high

A method of using the
As shown in FIG. 11, the
The measuring
A
The
A
The
The
When the
When the
When the liquid sample flows in the
The
The
(1) Since the high
(2) The
(3) Since the high
(4) The
(5) Since the high
(6) If the high
(7) Since the high
(8) Since the high

The embodiment of the present invention has been described above. The technical scope of the present invention is not limited to the embodiment described above. The applicable embodiment of the present invention can be appropriately changed from the above-described embodiment without departing from the spirit of the present invention.
Hereinafter, an example in which the amount of change in the interference wavelength is changed when the depth of the
The
The
When the liquid sample is supplied to the
As is apparent from FIG. 12, it can be seen that the measurement can be performed regardless of the depth of the
