米国特許法規則1.53(b)に基づき出願した非仮出願は、米国特許法119(e)に基づき、2008年5月13日に出願の仮出願第61/052727号の優先権を主張し、出典明記によりその内容全体を援用するものである。
本発明は概して、抗体−薬剤コンジュゲートやその断片および代謝産物を含む抗体コンジュゲート化合物を、質量分析によって、捕獲し、検出し、分析し、スクリーニングし、特徴付けし、定量化するための方法に関する。また、本発明は、薬物動態学研究および毒物動態学研究のために質量分析用試料を調製するための方法にも関する。
様々な質量分析技術、例えば電子衝撃(EI)、化学イオン化(CI)、脱着化学イオン化(DCI)、高速原子衝撃(FAB)、エレクトロスプレーイオン化(ESI)、マトリックス支援レーザー脱離/イオン化(MALDI)、及び逆重畳積分のためのイオン選別の単イオンモニタリング(SIM)(Souppart et al (2002) Jour. of Chrom. 免疫親和性メンブラン(IAM)キャプチャおよび質量分析による抗体−薬剤コンジュゲートを検出及びスクリーニングするための方法は開示されている(米国公開特許2005/0232929)。
免疫親和性ビーズキャプチャは、(i) 強いストレプトアビジン−ビオチン相互作用、(ii) 対象タンパク質の分析のために十分な物質を捕獲する高い結合能、(iii) 低い非特異的結合、(iv) 質量分析適合溶剤による試料成分溶出、(v) 良好な試料回復、及び(vi) 自動化への従順さ、を利用するストレプトアビジンコート常磁性ビーズによって実施してよい。

Lが、共有結合的にAbに接着し、共有結合的にDに接着したリンカーであり、そして、 pが1、2、3、4、5、6、7又は8である、本発明の抗体コンジュゲート化合物。
(i) 式Iを有する抗体−薬剤コンジュゲート化合物を提供すること;
(ii) 抗体−薬剤コンジュゲート化合物と、場合によってpが0である式Iの抗体又はその抗体断片ないし代謝産物を、哺乳動物、組織、細胞培養物、血漿又は血清から選択される生物学的な供給源と接触させること;
(iii) 生物学的な供給源から生体試料を採取すること;

(i) 式Iを有する抗体−薬剤コンジュゲート化合物の混合物を提供すること、このとき該混合物は場合によって、pが0である抗体又はその断片ないし代謝物を含み;
(ii) 哺乳動物に混合物を投与すること;
(iii) 混合物が投与された哺乳動物から血液試料又は排出物を採取すること;

本明細書における「インタクトな抗体」は、抗原結合性可変領域VLおよびVHドメイン、並びに完全な軽鎖および重鎖定常ドメイン(CL)および重鎖定常ドメイン、CH1、CH2およびCH3を含むものである。定常ドメインは、天然の配列定常ドメイン(例えばヒト天然配列定常ドメイン)又はそのアミノ酸配列変異体であってもよい。インタクトな抗体は、抗体のFc領域(天然配列Fc領域又はアミノ酸配列変異体Fc領域)に起因する生物活性を意味する一又は複数の「エフェクター機能」を有してよい。抗体エフェクター機能の例には、C1q結合;補体依存性細胞障害作用;Fcレセプター結合;抗体依存性細胞媒介性細胞障害作用(ADCC);食作用;細胞表面レセプターの下方制御(例えばB細胞レセプター;BCR)などが含まれる。
「生体起源」は、(i) 哺乳動物、例えばマウス、ラット、ウサギ、イヌ、サル又はヒト;(ii) 哺乳類の組織;及び(iii) 培養細胞を意味する。
「標識」は、抗体に共有結合的に接着し得、(i) 検出可能なシグナルを生じる;(ii) 第二標識と相互作用して、第一又は第二標識、例えばFRET(蛍光共鳴エネルギー転移)によって生じる検出可能なシグナルを修飾する;(iii) 抗原又はリガンドとの相互作用を安定させるかまたは結合の親和性を増やす;(iv) 電荷、疎水性、形状又は他の物理学的パラメータによって、移動度、例えば電気泳動移動度又は細胞透過性に作用する、又は(v) キャプチャ分子を生じさせ、リガンド親和性、抗体/抗原結合又はイオン複合体調節するために機能する、任意の部分を意味する。
式Iの抗体ユニット(Ab−)は、所定の標的細胞集団と関連するレセプター、抗原又は他の受容性部分と結合するか又は反応的に結合するか又は複合体化する抗体(Ab)の任意のユニットをその範囲内に包含する。抗体は、治療上又は生物学上修飾される細胞集団の部分と結合する、複合体化する又は相互作用する任意のタンパク質ないしタンパク質様分子でありうる。一態様では、抗体ユニットは、抗体ユニットが反応する特定の標的細胞集団へ薬剤ユニットを運搬するように働く。このような抗体には、限定するものではないが、完全長抗体及び抗体断片といった大きな分子量タンパク質が含まれる。
式Iの抗体−薬剤コンジュゲート(ADC)にAbを含み、癌の治療に有用となりうる抗体には、腫瘍関連抗原(TAA)に対する抗体を含むが、これに限定されるものではない。このような腫瘍関連抗原は当分野で公知であり、当分野で周知である方法および情報を使用して抗体を生成するために調製されてよい。癌の診断及び治療のために有効な細胞標的を発見するために、研究者は、一又は複数の正常非癌性細胞と比較して、一又は複数の特定の種類の癌細胞の表面上に特異的に発現される膜貫通あるいは腫瘍関連のポリペプチドを同定することを目的としている。多くの場合、このような腫瘍関連ポリペプチドは、非癌性細胞の表面上と比較して癌細胞の表面上に多く発現される。このような腫瘍関連細胞表面抗原ポリペプチドの同定により、抗体ベースの治療法による破壊のために癌細胞を特異的に標的化する能力が生じる。

標識は、抗体と直接又は非共有的にコンジュゲートされてよい。例えば、抗体は、ビオチンとコンジュゲートさせることができ、前述した分類のうちの何れかはストレプトアビジンなどのアビジンとコンジュゲートさせることができ、その逆もまた可能である。ビオチンは選択的にアビジンと結合し、したがって、標識はこの間接的な方法でポリペプチド変異体にコンジュゲートさせることができる。
式Iの抗体−薬剤コンジュゲート(ADC)の薬剤部分(D)は、細胞障害効果又は細胞***停止効果を有する任意の化合物、部分又は基を含む。薬剤部分は、マイクロチューブリンインヒビター、有糸***インヒビター、トポイソメラーゼインヒビター又はDNA干渉物質として機能しうる化学療法剤と、癌療法のために用いられる特別なものとを含む。式Iの抗体−薬剤コンジュゲートの薬剤部分は、他の作用機構を有してもよく、いずれかのメカニズムに限定されるものではない。

リンカーは、式Iの抗体−薬剤コンジュゲートに従って抗体(Ab)を薬剤成分(D)に共有結合的に接着させる原子の鎖又は共有結合を含む、二官能性ないしは多機能性の化学部分である。抗体−薬剤コンジュゲート(ADC)は、薬剤及び抗体への結合について反応性機能を有するリンカー(L)を用いて都合良く調製されうる。リンカーは、チオール又はアミノといった抗体上に存在する求核基と反応する求電子基を有してもよい。抗体のシステインチオールは、リンカー上の求電子基と反応することができ、リンカーとの共有結合を形成する。有用な求電子基には、マレイミドおよびハロアセトアミド基を含むが、これらに限定されるものではない。また、リンカーは、アルキルジイルといった二価のラジカル、アリーレン、ヘテロアリーレン、アルキルオキシ(例としてポリエチレンオキシ、PEG、ポリメチレンオキシ)とアルキラミノ(例えばポリエチレンアミノ、JeffamineTM)の繰り返しユニットである−(CR2)nO(CR2)n−などの部分、及び、スクシナート、スクシンアミド、ジグリコレート、マロネート及びカプロアミドを含む二酸エステル及びアミドが含まれる。抗体上の有用な求核基には、スルフヒドリル、ヒドロキシルおよびアミノ基が含まれるがこれらに限定されるものではない。

式IのADCの実施態様には、米国公開特許2005/0238649にて開示されるように、モノメチルアウリスタチン薬剤部分(D)であるMMAEおよびMMAFと、マレイミドカプロイル(MC)、バリン-シトルリン(vc)およびパラ-アミノベンジルカルバモイル(PAB)サブユニットを含むリンカーとが含まれる。例示的なADCは以下を含む:

薬剤ローディング値は、式Iの分子において、抗体当たりの薬剤部分の数であるpによって表される。式IのADCの組成物には、1からおよそ8の薬剤とコンジュゲートされた抗体の混合物が含まれる。抗体と薬剤−リンカー試薬とのコンジュゲート又は抗体−リンカーと薬剤試薬とのコンジュゲートから生じる抗体−薬剤コンジュゲートの混合物は、コンジュゲート条件に応じて、およそ1からおよそ8の平均薬剤ローディング値を有するものとして特徴付けられる。抗体を薬剤部分にコンジュゲートすることによるADCの調製により、一又は複数の抗体に結合した薬剤を持つ生成分子の分布が生じ得、この抗体は薬剤部分に結合されておらず、p=0である。コンジュゲート反応からのADCの調製において抗体当たりの薬剤の平均数は、本発明の方法、すなわち親和性質量分析及びELISAアッセイによって特徴付けられうる。ELISAによって、あるADC調製でのpの平均値が決定されうる(Hamblett et al (2004) Clinical Cancer Res. 10: 7063-7070;Sanderson et al (2005) Clinical Cancer Res. 11: 843-852)。しかしながら、p(薬剤)値の分布は、ELISAの抗体−抗原結合および検出限界によって識別可能でない。また、抗体−薬剤コンジュゲートの検出のためのELISAアッセイは、薬剤部分が重鎖又は軽鎖断片又は特定のアミノ酸残基といった抗体に付着しているところを決定しない。この重要な分布パラメータは、ADC組成物の個々の分子の分離およびそれらの特徴づけと定量化を有する本発明の方法によって決定されてよい。試料の成分の分離は、方法の分離培地工程時、そして、質量分析工程の間に起こる。本発明の方法の分離培地工程の高い選択性により、複合した異質な生物学的試料から、それぞれのADC成分の分離と精製が達成される。本発明の方法の質量分析工程の解像度と精度が高いので、分離されるADC成分の検出と定量化が達成される。
あるADCでは、pは抗体上の付着部位の数によって限定されうる。例えば、付着がシステインチオールである場合、上記の例示的な実施態様のように、抗体はたった1つ又は複数のシステインチオール基を有してもよいし、付着しうるリンカーによってたった1つ又は複数の十分に活性なチオール基を有してもよい。リジンといった反応性の低いアミノ酸残基は、コンジュゲートさせる抗体に多くあるが、反応せず、薬剤部分又は薬剤−リンカー試薬との反応に利用されない。薬剤ローディングが高い、例えばp>5であると、特定の抗体−薬剤コンジュゲートの凝集、難溶性、毒性又は細胞透過性の喪失が起こりうる。

本発明の抗体薬剤コンジュゲート(ADC)は、治療される状態に適切な任意の手段によって生物学上の供給源と接触させるか又は投与してよい。典型的に、ADCは、非経口的、すなわち注入、皮下、筋肉内、静脈内、皮内、髄腔内および硬膜外に哺乳動物に投与される。式IのADCと接触、すなわち投与されうる生物源は、(i) 哺乳動物、例えばマウス、ラット、ウサギ、イヌ、サル又はヒト;(ii) 哺乳類の組織;及び(iii) 培養細胞を含む。生物学的試料は、1回、または、時限であるか、間欠的であるか、ランダムな間隔で、生物源から採取される。生物学的試料は、(i) 血液、胆汁、尿又は糞便;(ii) 組織抽出物;及び(iii) 細胞培養物、細胞溶解物又は細胞抽出物を含む。
本発明の親和性キャプチャLC−MS法は、抗体−薬剤コンジュゲート化合物の毒性のメカニズムを決定するために、組織分析法で使用されてよい。
本発明の治療上の抗体薬剤コンジュゲート(ADC)の薬学的製剤は、典型的に、薬学的に許容可能な非経口用ベヒクルと共に単位用量注入用形態で、非経口的投与、すなわちボーラス、静脈内、腫瘍内注入のために調製される。所望の純度を持つ抗体−薬剤コンジュゲート(ADC)は、場合によって、薬学的に許容される希釈剤、担体、賦形剤又は安定化剤と、凍結乾燥又は水溶液の形態に混合される(Remington's Pharmaceutical Sciences 16th edition, Osol, A. Ed. (1980))。
許容される希釈剤、担体、賦形剤及び安定化剤は、用いられる用量と濃度で生物源レシピエントに非毒性であり、ホスフェート、シトレート及び他の有機酸等のバッファー;アスコルビン酸及びメチオニンを含む酸化防止剤;保存料(例えばオクタデシルジメチルベンジルアンモニウムクロリド;ヘキサメトニウムクロリド;ベンザルコニウムクロリド、ベンズエトニウムクロリド;フェノール、ブチル又はベンジルアルコール;メチル又はプロピルパラベン等のアルキルパラベン;カテコール;レゾルシノール;シクロヘキサノール;3-ペンタノール;及びm-クレゾール);低分子量(約10残基未満)のポリペプチド;血清アルブミン、ゼラチン、又は免疫グロブリン等のタンパク質;ポリビニルピロリドン等の親水性ポリマー;グリシン、グルタミン、アスパラギン、ヒスチジン、アルギニン又はリシン等のアミノ酸;グアールゴムおよびデキストリンを含む、単糖、二糖および他の炭水化物;グルコース、マンノース、スクロース、マンニトール、トレハロース又はソルビトールといった糖質;EDTA等のキレート化剤;ナトリウム等の塩形成対イオン;金属錯体(例えば、Zn-タンパク質複合体);及び/又はTWEENTM、PLURONICSTM又はポリエチレングリコール(PEG)等の非イオン性界面活性剤を含む。例えば、凍結乾燥した抗ErbB2抗体製剤は国際公開97/04801に記載されており、出典明記によって本明細書中に特別に援用される。
徐放性調製物を調製してもよい。徐放性調製物の好ましい例は、ADCを含む疎水性固体ポリマーの半透性マトリクスを含み、そのマトリクスは成形物、例えばフィルム又はマイクロカプセルの形態である。徐放性マトリクスの例には、ポリエステル、ヒドロゲル(例えば、ポリ(2-ヒドロキシエチルメタクリレート)、又はポリ(ビニルアルコール))、ポリラクチド(米国特許第3773919号)、L-グルタミン酸とγエチル-L-グルタメートのコポリマー、非分解性エチレン-酢酸ビニル、分解性乳酸-グリコール酸コポリマー、例えばLUPRON DEPOTTM(乳酸-グリコール酸コポリマー及び酢酸ロイプロリドからなる注射可能なミクロスフィア)、及びポリ-D-(-)-3-ヒドロキシブチル酸が含まれる。
代謝産物が先行技術に対して新規性及び非自明性を有する限りにおいて、本明細書中に記載のADC化合物のインビボ代謝産物も、本発明の権利内である。このような生成物は、投与された化合物の、例えば酸化、還元、加水分解、アミド化、エステル化、酵素切断などから生じうる。したがって、本発明は、代謝産物が生じるために十分な時間、本発明の化合物を哺乳動物と接触させることを含む方法によって生成される新規の自明でない化合物を包含する。
典型的には、代謝産物は、ラット、マウス、モルモット、サルなどの動物又はヒトに検出可能な用量(例えば、およそ0.5mg/kg以上)で抗体−薬剤コンジュゲート混合物を投与し、代謝が生じるために十分な時間(典型的にはおよそ30秒から30時間)をおいて、尿、血液又は他の生体試料から代謝された生成物を単離することによって同定される。代謝産物の構造は本発明の質量分析法によって決定される。
半減期、クリアランス、曲線下面積(AUC)、及び分布容積を含む哺乳動物における薬物動態(PK)定量のための治療剤の循環量のモニタリングは、安全性/毒性限界及び適切な投薬計画を樹立するために必要である(Welling, P. (1997) Pharmacokinetics Processes, Mathematics, and Applications, 2版, American Chemical Society, Washington, DC)。生物学的利用能は、投与された化合物が投与投薬形態から全身循環に達する度合いであり、通常は投与された用量のパーセントとして表される。ある化合物の半減期は、該化合物のピーク血漿中濃度の50%が排出又は生体内分解(代謝)によって除去されるのに必要な時間である。該治療指数は、所望される治療活性と所望されない毒性副作用間の選択性を表す。本発明の方法による薬物動態測定値は、抗体及び抗体・薬剤コンジュゲート(ADC)の吸収、分布、代謝、及び排出(ADME)を明らかにするものである。
インビボ投与されると、抗体・薬剤コンジュゲートは、加水分解、薬剤部分切断、抗体変性、グルクロン酸抱合、酸化、又は他の代謝分解事象を被る場合がある。これらの事象を精確に特徴付け、その産物を測定するために、ビーズベースのアフィニティ捕獲及び質量分析法が開発される。
式Iの抗体・薬剤コンジュゲート(ADC)化合物と場合によっては式Iの抗体(ここで、pは0である)、又は抗体断片又はその代謝産物を、哺乳動物、組織、細胞培養物、血漿又は血清から選択される生物学的供給源に投与し、又はこれと接触させる。血清及び血漿試料からの分析は、その高いプロテオミクスバックグラウンド、つまり多くのタンパク質及び他の分析物のために問題があることは知られている。分、時間、日にわたる所定の時間の後に、式Iを有する抗体・薬剤コンジュゲート化合物、又はその断片もしくは代謝産物を含有する生物学的試料を集める。該生物学的試料は、シリンジ又はカニューレで液を抜き出すことを含む任意の手段によって集めることができる。該生物学的試料は、血液又は血液生成物、例えば血清、血漿等、脳脊髄液又は他の体液、例えば唾液、尿、リンパ液、胆汁、糞便、汗、又は呼気でありうる。
処方、固定、遠心分離、単離、消化、血球凝固の誘導又は防止、加水分解、又は精製を含む一般的な手順によって生物学的試料を処理して分析試料を形成する。
生物学的試料の処理は、試料成分の分離を妨げ、又はデータ収集又は解析を不明瞭にしうる不純物を除去し、試料不均一性を低減させるものである。あるいは、又は加えて、プロセシングは、試料の取り扱いを単純化し、分解から保存し、試料容積を最小化し、又は質量分析解析において興味ある試料成分(分析物)を選択する。あるいは、又は加えて、プロセシングは、生物学的試料を、薬物代謝又は薬物動態効果の測定において興味深い代謝産物、断片、又は誘導体に転換する。
式Iの抗体・薬剤コンジュゲート(ADC)化合物と場合によっては式Iの抗体(ここで、pは0である)、又は抗体断片又はその代謝産物を、ビーズがADCの抗体又は薬剤に特異的な固定化抗原を有する免疫親和性ビーズに捕捉せしめる。投与された抗体・薬剤コンジュゲートの抗体に特異的な抗原をビオチン化し、強いビオチン-ストレプトアビジン相互作用(KD=10−15M)によってストレプトアビジン被覆常磁性ビーズに結合させる。図1aはECD捕獲と称される一実施態様を例証している。抗体(MAb)及び抗体・薬剤コンジュゲート(ADC)は、磁石に接触したストレプトアビジンに結合しているビオチン化EDCタンパク質のECD(細胞外ドメイン)に結合する。図1bは、抗体(MAb)及び抗体・薬剤コンジュゲート(ADC)が、ビーズに共有的に結合しているECDタンパク質のECD(細胞外ドメイン)に結合するECD捕獲の他の実施態様を例証する。ビーズはカラム形式に又はウェル中に緩く設けられうる。図2は、抗体・薬剤コンジュゲート(ADC)が、磁石に接触したストレプトアビジン被覆常磁性ビーズに結合しているビオチン化抗薬剤モノクローナル抗体(ビオチン-抗薬剤MAb)に結合する抗薬剤部分抗体捕獲の他の実施態様を例証する。
免疫親和性ビーズは、多孔性ポリマーモノリスを含み得、捕集リザーバと流体が流れるように連通しているフロースルーチャネルに設けられうる。ビーズは、生物学的供給源からの試料が一端又はオリフィスに導入され、試料が他端又はオリフィスから溶出されるカラム又は漏斗のようなフロースルー容器に収容されてもよい。免疫親和性ビーズは、それぞれが別個の捕集リザーバに連通させられた複数のフロースルー容器に分布せしめられてもよい。容器及びリザーバは、12×8の行列の96マイクロタイターウェル形式、又は自動化及び結果の再現性のために24×16の行列の384マイクロタイターウェル形式に構成されうる。
ストレプトアビジン被覆常磁性ビーズを使用する理論的根拠は、(i)強いストレプトアビジン−ビオチン相互作用(KD=10−15M)、(ii)固定化ストレプトアビジン/ビオチン化分析物が立証された方法であること、(iii)高い結合能(インタクトなタンパク質に対して十分な材料)、(iv)低い非特異的結合、(v)質量分析に適合した溶媒を用いた試料の溶出、(vi)ビーズからの良好な試料回収、及び(vii)使用の容易性及び自動化への受け入れ性である。
例示的な実施態様では、生物学的試料はトリプシン消化で消化されうる。トリプシン消化では、試料はDDTで還元され、ヨード酢酸ナトリウムでS−カルボキシメチル化され、ついでトリプシンで消化されうる。消化された試料は、(i)逆相HPLC、例えばNucleosil C18カラム;(ii)サイズ排除クロマトグラフィー(SEC)、例えばTSK3000SWxLカラム;又は(iii)TSKボロン酸カラムを使用するボロン酸アフィニティクロマトグラフィーによって分析されうる。
分析試料を形成するために、生物学的試料を分離溶液に適用して、一を越える試料成分の分離をなさしめることができる。分離方法は、アフィニティ、クロマトグラフィー、及び電気泳動法を含む。アフィニティ法は、アフィニティクロマトグラフィー、吸着、及び固定化アフィニティマトリックスを含む。クロマトグラフィー法は、HPLC、疎水性相互作用(HIC)、アニオン交換、カチオン交換、逆相、順相、イオン対逆相、薄層、キャピラリーフロー、及びサイズ排除を含む。電気泳動法は、単一次元、スラブゲル、キャピラリー、ポリアクリルアミド、変性、天然、自由溶液、紙、2次元、等電点電気泳動、及び勾配電位を含む。他の分離方法は、透析、遠心分離、遠心沈降、浮選、沈降、免疫沈降、及びゲル濾過を含む。
分離方法は、限定しないが、溶出時間、疎水性、親水性、泳動時間、割合、速度、クロマトグラフィー保持時間、溶解度、分子容積又はサイズ、正味荷電、荷電状態、イオン電荷、等電点、解離定数(pKa)、抗体親和性、電気泳動移動度、イオン化ポテンシャル、双極子モーメント、水素結合能、及び気相中におけるイオン移動度を含む一又は複数の物理化学的性質による生物学的試料の成分の分離を行うことができる。
質量分析計入口装置中へのキャピラリーフロー注入による低い流量により、質量検出の感度を高め、例えばインタクトなタンパク質及び抗体・薬剤コンジュゲートのような低濃度分析物及び高分子量種を検出し特徴付けることができる。
質量分光分析のための抗体・薬剤コンジュゲート試料の調製は、一般に、知られている技術に従って実施されうる。“Modern Protein Chemistry: Practical Aspects”, Howard, G.C.及びBrown, W.E.編(2002) CRC Press, Boca Raton, Flを参照のこと。
本発明の方法は生物学的試料から由来する抗体混合物の分析に適しており、成分を連続的に又はバッチ形式で溶出せしめ、又は質量分析計によって直接検出せしめるアフィニティ又はクロマトグラフィーを含む一又は複数のプロセスにより、混合物の異なった化学成分が、最初に単離され、分離され、又は部分的に分離される。断片化、脱アミド、グリケーション、酸化、部分的配列情報、例えばN末端及びC末端、二量体及び凝集状態を含む抗体の様々な構造的特徴及び性質を、質量分光分析から解明することができる。生物学的試料中の一又は複数の化学成分は、投与される抗体・薬剤コンジュゲートが既知の配列、構造、及び分子量のものであるので、その精確な質量の測定によって高度に特異的な形で特徴付けることができる。
機能的システムでは、質量分析計は関心ある化学種の質量を、その正確な又は計算された質量の20ppm以内まで、典型的にはその正確な又は計算された質量の5ppm以下まで精確に測定する。市販の質量分析計は、質量スペクトルシグナル強度又はピーク面積が定量的に代表することを担保するために混合物中の複数の成分について十分なスペクトルを獲得することを可能にする頻度で同時に全質量スペクトルをサンプリングし記録することができる。これはまた全ての質量に対して観察される溶出時間が質量分析器によって変更又は歪められないことを担保し、定量的測定値が大量の一過性シグナルを測定する必要性によって損なわれないことを担保するのに役立つであろう。
高感度が、増加した分析物イオン化効率のために低流量で達成される(Gale等(1993) Rapid Commun. Mass Spectrom. 7:1017)。よって、分レンジ当たりナノリットルの流量で試料含有液のエレクトロスプレー注入を実施することによって、適切な検量後に精確な定量がもたらされ、質量分析と併用される場合、液中に含まれる分析物に対して高感度がもたらされる。高選択性及び感度をもたらし、MSに対する前工程としての精確な定量分析をもたらす小型化され統合されたマイクロカラム及びアフィニティクロマトグラフィー吸着剤を有するマイクロカラムアレイを含むシステム及び装置が報告されている(米国特許第6811689号;米国特許第6020208号;米国特許第6579719号)。
抗体のような比較的高分子量の化合物の質量は、殆どの質量分析計によって容易に決定される質量電荷比(m/z)(2000までから3000までの典型的なm/z範囲)で検出されうる。エレクトロスプレーイオン化質量分析ESI−MSは、荷電、極性又は塩基性化合物に、また多重に荷電した化合物を優れた検出限界で分析するために特に適している。ESIはよって大きな生体分子、例えば150000又はそれ以上の分子量(MW)の抗体及び抗体・薬剤コンジュゲートの検出及び特徴付けを可能にする。高分子量イオンを用いると、一連の多重に荷電した分子イオンが典型的には観察される。陽イオンの分子量は、測定されたm/z比に電荷数(n)を掛け、カチオン質量(C+)にイオン上の電荷数(n)を掛けたものを引くことによって決定される。
ESI−MSデータは、多数のスキャンを併せて平均化し、データを平滑化して良好なピーク強度及び形状をもたらすことによって獲得されうる。低分子量化合物では、観察される最も多いピークはしばしば陽イオンモードでは[M+H]+イオン、陰イオンモードでは[M−H]−である。二重及び三重に荷電したイオン並びに二量体がまた観察されうる。二重に荷電した陽イオンは質量(MW+2C+)÷2に観察され、ここで、MWは分子量であり、C+はイオン化陽イオン、例えばH+、Na+、又はNH4+である。非常に低分子量の化合物を除いて、検出されるイオンは多重に荷電される。ESIのソフトな(低イオン化電位)条件のため、典型的には分子イオンだけが観察される。ESIスペクトルは、イオンが有する様々な電荷数のために質量が異なる幾つかの分子イオンピークを有しうる。
以下の構造を有する抗体MUC16抗体−薬剤コンジュゲートである3A5−MC−vc−PAB−MMAE、「抗MUC16 ADC」:
(ここで、p(DAR)は1、2、3又は4であり、Valはバリンであり、Citはシトルリンであり、Abは3A5のシステイン改変したA118C重鎖変異体である抗MUC16モノクローナル抗体である)を、血漿および血清試料において分析した。3A5抗体変異体は、正常ヒト組織と比較してヒト卵巣上皮癌(EOC)において過剰に発現される細胞表面膜貫通タンパク質であるMUC16の細胞外ドメイン(ECD)上のエピトープを認識し、MUC16への結合により内部移行し、リソソームに輸送され、これによってアウリスタチン薬剤部分MMAEのMUC16陽性腫瘍細胞への標的輸送が達成される(国際公開2007/001851;2007年5月8日に出願の米国特許出願60/916657「CYSTEINE ENGINEERED ANTI-MUC16 ANTIBODIES AND ANTIBODY DRUG CONJUGATES」)。A118C(EU番号付け)変異体は、米国公開特許2007/0092940に従って、薬剤−リンカー試薬によりその最適化されたチオール反応性について選択した。
ビオチン化したヒトMUC16 ECDをストレプトアビジンコートビーズ上へ固定し、それを用いて室温で試験血漿又は血清の試料と共にインキュベートすることによって抗MUC16 ADCをキャプチャした。例えば、ビーズは、およそ10−100ミクロン直径のセファロース(登録商標)ビーズであってもよい。ビーズが常磁性である場合、試料成分の結合の後に、常磁性ビーズが磁石によって適当な状態に保たれ、ビーズに結合した試料成分の分離、単離および洗浄が可能となる。ビーズが常磁性でない場合、ビーズは移動相流量のための出入口を有するカラムで構成されてもよい。試料成分は、例えば高い酸及び有機濃度を有する溶出培地又はバッファにて被処理分析試料として溶出してよく、この溶出した試料は、分離培地に曝すために回収して試料成分を分離させた後に質量分析を行ってもよい。典型的な非特異的洗浄バッファは水溶性であり、およそpH7.4で酢酸ナトリウム及び塩化ナトリウムを含んでいてよい。典型的な抗体試料溶出バッファは水溶性であり、2〜4のpHで、イソプロパノール、アセトニトリル又は他の有機溶媒のような低分子性アルコールと蟻酸のような酸を含有してよい。溶出の後、固定したECDビーズは、回収しても、再利用しても、処分してもよい。
ビーズは、洗浄溶出溶液の出入口を有するカラム形式に構成されてよい。NHS−活性化セファロース(登録商標)ビーズの市販される実施態様にはNHS HiTrap HP1.0ml親和性カラム(Amersham)がある。
イオン化効率: 特定のp=0、1および2(それぞれDAR-0、DAR-1およびDAR-2)を有する抗MUC16 ADC(3A5−MC−vc−PAB−MMAE)標準物質を、異なる比率で混合した(例えば33:33:33および30:60:10)。次いで、脱グリコシル化のために混合物を37℃で終夜インキュベートした。脱グリコシル化混合物をおよそ30μg/mLに希釈し、10μlの一定量を分析のためにLC/MS上に直接注入した。
HBS−EP中30:60:10の比率の、p=0、1および2(DAR-0、DAR-1及びDAR-2)を有する抗MUC16 ADC(3A5−MC−vc−PAB−MMAE)試料成分の全イオンクロマトグラムを得て、ADCシグナルを含む代表的な時間ウインドウを選択した。時間ウインドウはLC条件の変動のために変化しうる。ADC試料成分について特徴的な電荷エンベロープを表す対応する質量スペクトルを抽出した。質量スペクトルの逆重畳積分により、対応する逆重畳積分した質量スペクトルを有するピーク面積表を生成した。抗MUC16 ADC試料成分の分子質量に基づいて、3つのメインピークは、それぞれおよそ144834Da、146033Daおよび147223DaのDAR−0、DAR−1およびDAR−2と同定した。内部較正なしで、機器の集団の精度はおよそ±50Daであった。他のマイナーピークは、主に標準物質のマトリックスバックグラウンド、添加物および/または不均一性によるものであった。それらは結果としてADC試料成分の相対量の算出に大きな影響を与えなかったので、その後の比率算出に用いなかった。3つの個々のピーク面積は総ピーク面積として合計し、各々の抗MUC16 ADC試料成分の相対的なパーセント比率を算出した。2つの添加混合物のそれぞれについて表1Aと1B(下記)にデータをまとめた。各添加組成物について3回試験した。明らかなように、平均精度は70%〜130%の範囲内であった。したがって、DAR−0、DAR−1およびDAR−2を有する抗MUC16 ADC試料成分は、陽性ターボイオンスプレーモードにおけるイオン化効率に有意差がなかったことが結論づけられた。

平均精度が、異なる組成物でDAR−0、DAR−1およびDAR−2の3つの抗MUC16 ADC試料成分について70%〜130%の範囲内であったことから、ECD改変親和性ビーズは血漿からの選択的な損失がなくADCを回収することができ、許容範囲内の精度を示したことが示された。
ラット、カニクイザルおよびヒトの血漿に添加した抗MUC16 ADC試料成分の代表的なTICクロマトグラムを、PBSバッファコントロールと比較した。ECD免疫親和性ビーズによってキャプチャされるこれら4つのマトリックスにおける抗MUC16 ADC試料成分のクロマトグラフィのパターンは非常に類似していた。代表的なクロマトグラフィの持続時間ウインドウから、3つの種の血漿マトリックスおよびPBS(バッファ)コントロール間で、同種のDAR分布パターンが観察された。試料成分は、DAR 0(+0、ネイキッド抗体)、DAR−1(+1 D、1のMC−vc−PAB−MMAE薬剤リンカーユニット)およびDAR−2(+2D、2のMC−vc−PAB−MMAE薬剤リンカーユニット)とした。DAR−0、DAR−1およびDAR−2構成成分の相対量の詳細な比較を表3に示す。全体の相対的標準偏差(RSD)は、試験した4つすべてのマトリックスについて30%以下で十分であった。したがって、親和性MS法は、異なる種全体にわたって最小限のマトリックス効果を示した。全体の精度は70〜130%の範囲内であった。

ECD免疫親和性ビーズキャプチャ効率を、ラット血漿中でインキュベートした後に、Thio Hu 抗MUC16(3A5) HC A118C−MC−vc−PAB−MMAEの抗薬剤Mab免疫親和性ビーズキャプチャと比較された。4つの異なる抗アウリスタチンモノクローナル抗体クローンをビオチン化し、ストレプトアビジンコート常磁性ビーズに固定した(図2)。これらの抗薬剤クローンは、1薬剤をロードしたADC(DAR+1)の非効率的なキャプチャを示した。
抗MUC16 抗体−薬剤コンジュゲートである3A5−MC−vc−PAB−MMAEを投与したカニクイザルの血清および血漿試料を、以下の工程によって処理した。
1. 試料、コントロール及びブランクのプレート位置を決定する(96深底ウェルプレート(2mlの正方形上部):Analytical Sales and Service Inc. カタログ番号59623-23、又は96ウェルプレート(500μlの円形上部): VWRカタログ番号47743-828)。一般的に、2つのブランクと1つのシステムコントロールを実行の初めに試験し、その後2つのブランクと2つのシステムコントロールを実行の最後に試験する。必要に応じて実行の全体にわたって更なるブランクを試験してよい。試料、コントロール又はブランクに用いるウェルに、後述する添加を行った。
2. 試料、コントロール又はブランクに用いる96深底ウェル正方形上部プレートの各ウェルに400μlのHBS−EPバッファ(Biacoreカタログ番号BR-1001-88)をピペットで移す。
3. ストレプトアビジンコートのダイナビーズM-280 (Dynabeads、M280ストレプトアビジン、10mg/mL、カタログ番号110029、Lot番号G74050、BioVeris)を緩やかに撹拌することによって再懸濁する。使用中のHBS−EPバッファプレートの各ウェルに100μlの懸濁したビーズ混合物をピペットで移す。およそ20秒間、室温で、KingFisher96の磁気粒子プロセッサ(Thermo Electron Corp.)によって混合する。
4. 400μLのHBS−EPバッファを含む新たな96深底ウェル正方形上部プレートへビーズを移し、およそ20秒間、室温で、KingFisherにより混合する。
5. 新たな96深底ウェル正方形上部プレートのブランク、試料又はコントロールの各ウェルに、400μlのHBS−EPバッファをピペットで移す。
6. HBS−EPバッファプレートのブランク、試料又はコントロールの各ウェルに、25μlのビオチン化抗MUC16-ECD(図1a)をピペットで移す。
7. HBS−EPバッファとビオチン化した抗MUC16-ECDを含む96深底ウェルプレートにビーズを移し、およそ20秒間穏やかに混ぜる。プレートをアルミニウムシールで被う。
8. プレートを攪拌機(スピード7に設定)に載せ、およそ120分間、室温でインキュベートする。
9. 400μlのHBS−EPバッファを含む96深底ウェル正方形上部プレートにビーズを移し、KingFisherを用いて2回洗浄する。
11. 96深底ウェル正方形上部プレートへ400μlのHBS−EPバッファをピペットで移し、次いで、各々希釈した50μlの血漿又は血清の試料、コントロール又は又はブランクを適切なウェルに添加する。
12. 希釈した血漿又は血清の試料にKingFisherを用いてビーズを移す。およそ20秒間穏やかに混ぜる。プレートをアルミニウムシールテープで被う。
13. プレートを攪拌機(速度7)に載せ、およそ120分間、室温でインキュベートする。
14. 500μlのHBS−EPバッファを含む96深底ウェル正方形上部プレートにビーズを移し、KingFisherを用いて2回洗浄する。
15. HBS−EPバッファ、80mMリン酸塩及びグリコN-グリカナーゼ(Prozyme glyko N-glycanase, Cat. No. GKE-5006D)をそれぞれ300パーツ:32パーツ:4パーツの比率で混合することによってHBS−EP−グリカナーゼバッファを調製する。
16. 336μlのHBS−EP−グリカナーゼバッファを96深底ウェル正方形上部プレートの各ウェルへピペットで移す。
17. KingFisherを用いてHBS−EP−グリカナーゼバッファにビーズを移す。およそ20秒間穏やかに混合する。プレートをアルミニウムシールテープで被う。
18. 37℃及び300rpmの撹拌速度に維持するように設定したインキュベーターにプレートを置き、終夜インキュベートする。
19. 500μlのHBS−EPバッファを含む96深底ウェル正方形上部プレートにビーズを移し、KingFisherを用いて2回洗浄する。
21. 水中10%アセトニトリルを500μl含む96深底ウェル正方形上部プレートにビーズを移し、KingFisherを用いて1回洗浄する。
22. 溶出溶媒として、1%の蟻酸を含む30%アセトニトリル水、50μlを、96深底ウェル正方形上部プレートにピペットで移す。
23. KingFisherを用いて溶出溶媒プレートにビーズを移す。プレートをアルミニウムシールテープで被う。
24. 速度7に設定した攪拌機にプレートを置き、およそ15分間振とうする。
25. KingFisherを用いて溶出プレートからビーズを取り除く。
26. マルチチャネルピペットを用いて、96ウェル注入プレート(VWR、500μl円形上部)に溶出プレートからの上清を移し、プレートをシリコン封入マットで被う。
27. 2〜8℃に維持するように設定した遠心機にて、3000rpmの設定でおよそ5分間遠心分離する。そうして、LC−MS上への注入用試料プレートとする。
以下の構造:
を有する、システイン改変抗HER2変異体V205CおよびA118Cのトラスツズマブ−MC−vc−PAB−MMAE(ここで、pが1、2、3又は4であり、Valはバリンであり、Citはシトルリンであり、Abはトラスツズマブのシステイン改変の、A118C重鎖変異体およびV205C軽鎖変異体、抗HER2モノクローナル抗体である)を血漿試料において分析した。
重鎖変異体(Thio Hu 抗HER2 4D5 HC A118C−MC−vc−PAB−MMAE、1.9MMAE/4D5 Ab)を、100μg/mlでラット血漿中でインキュベートした。特定の時点で試料を採取し、免疫親和性ECDビーズキャプチャにて処理した。図5は、0、8、24、48および96時間の時点で採取した試料の逆重畳積分質量分析データを示す。試料成分を、+0(ネイキッド抗体)、+1D(1のMC−vc−PAB−MMAE薬剤リンカーユニット)および+2D(2のMC−vc−PAB−MMAE薬剤リンカーユニット)のDARとした。およそ151000amuの小さいピークは、不完全に脱グリコシル化された試料成分である。
図10aは、ECDタンパク質が抗体又は抗体−薬剤コンジュゲート(ADC)への結合のために固体担体上に固定されている総ELISAアッセイフォーマットを示す。ADCは、化学発光検出のためにF(ab')2ヤギ抗ヒトFc−HRPに結合する。
図10bは、抗薬剤MAbが抗体−薬剤コンジュゲート(ADC)への結合のために固体担体上に固定されているコンジュゲートELISAアッセイフォーマットを示す。ADCは、溶液中のビオチン化ECDタンパク質に結合する。次いで、複合体は、化学発光検出のためにストレプトアビジン−西洋ワサビペルオキシダーゼ(HRP)に結合しうる。
図11は、Thio Hu 抗MUC16 (3A5) HC A118C-MC-vc-PAB-MMAEと共にインキュベートし、96時間まで経時的に分析したラット血漿試料中の、薬剤部分にコンジュゲートした残りの抗体をプロットすることによる、ELISA法、及び免疫親和性ビーズキャプチャ/質量分析(MS)法による試料成分の検出の比較を示す。

Claims (39)
- (i) 以下の式Iを有する抗体−薬剤コンジュゲート化合物を提供すること:

- 抗体−薬剤コンジュゲート化合物の抗体が、以下の(1)から(36)から選択される一又は複数の腫瘍関連抗原又は細胞表面レセプターに結合する、請求項26に記載の方法:

- LがAbのアミノ、カルボキシル又はチオールに共有結合的に付着している、請求項1に記載の方法。
- Lが、N-スクシンイミジル-4(2-ピリジルチオ)プロパノエート(SPDP)、N-スクシンイミジル-4-(N-マレイミドメチル)シクロヘキサン-1-カルボキシレート(SMCC)及びN-スクシンイミジル-4-(2-ピリジルチオ)ペンタノエート(SPP)から選択されるリンカー試薬から形成される、請求項1に記載の方法。
- Lが、マレイミドカプロイル(MC)、マレイミドプロパノイル(MP)、およびマレイミドカプロイル−バリン−シトルリン−パラ−アミノベンジルオキシカルボニル(MC−vc−PAB)から選択される、請求項1に記載の方法。
- Dがモノメチルアウリスタチンである、請求項1に記載の方法。
- (i) 以下の式Iを有する抗体−薬剤コンジュゲート化合物の混合物を提供すること、

このとき、
Abが抗体であり、
Dがメイタンシノイド又はモノメチルアウリスタチン薬剤部分であり、
Lが、Abに共有結合的に付着し、Dに共有結合的に付着したリンカーであり、
pが1、2、3、4、5、6、7又は8であり、そして、場合によって該混合物が、pが0である抗体又はその断片ないし代謝産物を含んでおり;
(ii) 哺乳動物に該混合物を投与すること;
(iii) 該混合物が投与された哺乳動物から血液試料又は排出物を採取すること;
(iv) 処方し、固定し、遠心し、単離し、消化し、血液細胞凝集を誘導ないし予防し、加水分解し、又は精製することによって分析試料を生成するために血液試料又は排出物を処理し、処理済みの分析試料を生成すること;
(v) 処理済みの分析試料に特異的な抗原を含む免疫親和性ビーズにて処理済みの分析試料を捕獲すること;
(vi) 処理済みの分析試料を溶出すること;
(vii) 分離培地に血液試料、排出物又は分析試料を置き、1より多くの試料成分を分離すること、このとき分離した試料成分は式Iを有する抗体−薬剤コンジュゲート化合物又はその抗体断片ないし代謝物を含み、pは0、1、2、3、4、5、6、7又は8であり;そして、
(viii) 一又は複数の分離した試料成分の質量電荷比を質量分析によって確認すること
を含む、哺乳動物において、抗体−薬剤コンジュゲート化合物の混合物をスクリーニングして、化合物又はその断片ないし代謝物のクリアランスを決定する方法。 - さらに、(iii)から(viii)の工程を1回又は複数回繰り返すことを含む、請求項37に記載の方法。
- さらに、質量分析を実行する前に、分離した試料成分を変性剤と接触させることを含む、請求項37に記載の方法。
