Abstract
【解決手段】アミノ酸残基、糖残基、脂肪酸残基およびヌクレオチドからなる群から選択されるL1〜Lx〜Lmから成るリガンド決定基の構成要素単位を含む化合物であり、リガンド決定基にリンカー、及びレセプターと特異的に反応する共有結合反応性求電子基からなる共有結合反応性リガンドアナログ(CAL)。

Description
非触媒タンパク質は求電子性の低い活性部位特異的化合物に特異的結合する際に求核反応性を示すことが発見された。これらの活性化求核体はタンパク質中の通常の非共有結合部位を見定めて機能する。現在まで、そのように活性化した求核体は、セリンプロテアーゼにみられるような酵素の触媒部位にのみ存在すると考えられていた。本発明では、アルブミン、カルモジュリン、gp12O、ならびにgp120、EGFRおよび第VIII因子に対する抗体を含めたいくつかの非酵素タンパク質で求核部位が同定された。これらのタンパク質に活性化した求核体が存在することは、タンパク質のフォールディングならびに他の高分子および低分子とのタンパク質相互作用から生じる、本発明化合物との特異的非共有結合相互作用が形成されることを示している。

Lxはアミノ酸残基、糖残基、脂肪酸残基およびDNA残基からなる群から選択されるリガンド決定基の構成要素単位であり、
L'はLxの官能基であり、
Y"はリンカーまたは共有結合であり、
Y'は荷電した基または中性基であり、
Yは前記リガンド決定基に結合するレセプターと特異的に反応する共有結合反応性求電子基であり、
nは1から1000の整数であり、かつ
mは1から30の整数である] の共有結合反応性リガンドアナログ(CAL)を提供することである。
1. 抗体の求核反応性
タンパク質の求核反応性は、ある種のアミノ酸側鎖の活性化から生じる。セリンプロテアーゼでは、Ser-His-Asp三つ組構造の正確な空間配置によりSerの酸素に求核反応性を付与する水素結合ネットワークの形成が可能になる。ベンスジョーンズタンパク質のCDR1とセリンプロテアーゼの活性部位Ser残基周辺のペプチド領域との間の配列ホモロジーに基づいて、Abは酵素様活性部位を発現することが1973年に予測された(1)。Ser27a-His93-Asp1から構成されるセリンプロテアーゼ様触媒三つ組構造が、部位特異的突然変異誘発によってVIPに対する抗体(Ab)の軽鎖(L鎖)に存在することが同定された(2)。
実施例5に開示しているように、低分子量CAL(ハプテンCAL)に対する共有結合についての一連の精製タンパク質のスクリーニングは、求核反応性がHIV gp120、アルブミン、上皮増殖因子レセプターの可溶性細胞外ドメイン(sEGFR)、カゼインおよびCD4の可溶性細胞外ドメイン(sCD4)を含めた多様なタンパク質に広範囲に分布していることを示した。これらのタンパク質のいずれも公知の酵素活性を全く有さない。求核反応性のレベルは種々のタンパク質で多様であり、このことはそれぞれのタンパク質の独自の分子性質を示唆している。
同一タンパク質の個別の分子を伴う分子間ホモ会合反応は、異なるタンパク質間のヘテロ複合体形成反応に使用される力と類似した力を必要とする。HIV gp120-CAL調製物を含めるがそれに限定されないタンパク質の共有結合ホモ集合の自然形成が本発明に開示されている。LaCALとNuRとの間のヘテロ複合体化の場合と同様に、集合はgp120の自然集合の形成に必要な非共有結合相互作用により誘導される。gp120は非共有結合オリゴマーとしてHIV表面に発現することがよく知られている。gp120オリゴマーの間の接触領域が我々のモデルでは部分共有結合力を介して結合する天然求核体-求電子体対を含むことから、非共有結合gp120オリゴマーの天然構造を刺激するgp120-CALの共有結合性集合の形成が起こる。本発明に開示されているgp120-CAL調製物は、天然求電子体よりも強い共有結合反応性を発現する人工求電子体を含有する。したがって、天然求核体と安定共有結合が形成し、天然gp120オリゴマーに類似したコンホメーションを有するgp120-CALオリゴマーの集合が生じる。
種々の種類の生体現象への有用性を開拓するために、多様なハプテンCAL構造およびLaCAL構造が本発明に開示される。共有結合反応性求電子体および弱結合基が空間的に見当し合ってNuRの活性部位に対する特異的共有結合を達成することが可能になるという考えがCALの設計を推進した。設計特性の例は以下のように開示されている。

標的NuRに共有結合するCALは以下の目的に使用できる:
・ 可溶性NuRの永続的不活性化、
・ 微生物NuRの永続的不活性化、
・ 細胞表面に発現したNuRの強力な拮抗作用および作動作用、
・ 様々な細胞活性および機能の不活性化または活性化
・ 表面にNuRを発現している細胞の死滅の成長停止の誘導、
・ 生体分子の自然自己集合アレイの共有結合模倣体の発生、
・ ワクチン候補としての共有結合性gp120オリゴマー模倣体の使用、
・ 表面にNuRを発現している細胞および微生物の高感度画像化
・ 抗原特異的な抗体の単離、誘導および阻害、
・ 診断イムノアッセイ応用における抗体および抗原の高感度検出。
・ NuRとの結合速度について一連のハプテンCALおよびpCALをスクリーニングすることによって、これらの化合物のうち最良のものを同定する。これには可逆結合ステップに対するKdおよび共有結合ステップに対する一次速度定数k3の大きさの測定がある。
多くの酵素は、基質との共有結合的な相互作用を利用して化学的変換を触媒する。一方、Ab触媒に関するほとんどの研究は、反応物と生成物との間のエネルギー障壁を下げる機構としての非共有的結合力、例えば、エステル加水分解の負に荷電したオキシアニオン遷移状態を安定化する静電力に注目してきた(1、2の総説)。元となる仮説は、Abは専ら非共有的手段によってリガンドと相互作用するということである。天然のAbは酵素とは見分けのつかない化学反応性を表すという当初の指摘は、自己抗体のタンパク質分解活性およびヌクレアーゼ活性の報告(3、4)から得られた。同様の活性が後に多発骨髄腫の患者のAb軽鎖(5)、輸血によって誘発された血友病の患者の同種異系抗体(6)、ポリペプチドによる通常免疫で生じたAb(7、8)および抗酵素Abに対する抗イディオタイプAb(9)で発見された。変異誘発および阻害剤の研究から、天然Abのタンパク質分解活性は、従来のセリンプロテアーゼで使用されるものと同様の求核機構によって生じることが明らかになった(10、11)。天然Abの触媒活性は、B細胞クローン選択理論の原理に反するものと解釈することができた。V領域をコードする生殖細胞系遺伝子の配列多様化、その後の最も親和性の高いB細胞レセプターによる選択的な抗原結合、それによるAbのクローン増殖の刺激という免疫過程の中で抗原特異的Abが生成される。触媒には、抗原の化学的変換およびB細胞選択の中断を引き起こすことが予測される生成物(IgαおよびIgβサブユニットに関連した表面イムノグロブリン)の遊離が必要である。したがって、Ab触媒活性の適応選択は、敬遠される現象である。この理由のため、天然に生じるAbによる触媒は、機能と密接な関係のある一般的現象とは対照的に、V領域の配列多様性によって生じる分子の偶発的な出来事であると考えられることが多い。
Ab。 ヒトポリクローナルIgGは、6人の健常人被験者の血清(研究室認識番号、1086、1087、1088、1091、1092、1518)からプロテインG-セファロース(Amersham Pharmacia)のアフィニティークロマトグラフィーによって調製した。8匹のBALB/cマウス(4〜5週齢)から収集した血清のIgGも同様に得た。KLHに結合させた合成Cys-gp120(421〜436)(KQIINMWQEVGKAMYA;gp120 HIV SF2種の421〜436残基)で過免疫することによるポリクローナルAbの調製は、(13)に説明されている。雌BALB/cマウス(5〜6週齢)は、0日目、27日目および41日目にRIBIアジュバントに入れてexEGFR(10μg/注射)を、14日目にはA431腫瘍細胞(生理食塩水に溶かした107個の細胞)を注射して免疫することによって、exEGFRに対するポリクローナルAbを得た。exEGFRに対するモノクローナルAb(クローンC225、H11およびC116)は、Labvision(Fremont、CA)から購入した。対照の抗BSAモノクローナルIgG(クローンBGN/H8)は、Biogenesis(Kingston、NH)から入手した。単鎖Fv構造物(N=15)は、(11)に記載された狼瘡患者から得られたヒトFvライブラリーから無作為に選択した。(MM系クローン、12、14、18、20、24、F1、F2、F4、F5、F6、F7、F11、F12、F14、F17、F18)。scFVタンパク質は、Ni-NTAカラムの金属アフィニティークロマトグラフィーによって電気泳動的に均一(27kDaのバンド)になるまで精製した(11)。発現濃度は、細菌培養液1リットル当り0.3〜5.7mgであった。該ライブラリーには、ヌクレオチド配列決定によって決定された異なるscFvクローンが含まれており(11)、多様なAbのV領域を確実にサンプリングすることが可能である。この研究で調べたscFvクローンの1つ、MM-F4の配列決定を行い(GenBank番号AF522073)、そのVLおよびVH領域がそれぞれXおよびIファミリーに属することが決定され、生殖細胞系遺伝子相対物はそれぞれ、V1-13およびVH1-2であった。SDS電気泳動ゲルにおけるscFvバンドの確認は、c-mycに対するモノクローナルAbを使用したイムノブロッティングによって行った(10)。
ハプテンCALで確認したAb求核性。 ホスホネートハプテンCAL I〜III(図8)は、セリンプロテアーゼの公知の活性部位特異的阻害剤のアナログである(19)。セリンプロテアーゼトリプシンのように、健常人被験者から得られたIgGは煮沸した変性SDSに耐性があるCAL Iアダクトを形成した(図9、IgG、150kDアダクト、トリプシン、21KDアダクト)。免疫学的に操作されていないBALB/cマウスから得られた収集IgGは同様のIアダクトを形成した。CAL I内の正に荷電したアミジノ基は元来、化合物に取り込まれて、P1部位の塩基性残基に対する選択性を示すトリプシンの選択的認識を可能にした(該残基はペプチド基質の切断部位のすぐ側にある、文献20)。CAL IIは、共有結合反応性リン原子に隣接した正に荷電したアミジノ基を持たない。IgGは、IよりもIIに対する反応性が240倍少なかったので、Abのトリプシン様P1特異性が示唆される。Iよりも弱い遊離基を含有するIIIは、IgGとは検出可能なアダクトを形成しなかった(メトキシ遊離基が存在すると、リン原子の求核性が減少する、メトキシ含有ホスホン酸ジエステルはある種のセリンプロテアーゼと弱く結合することが報告されている、文献21)。IgGおよびトリプシンとの共有結合Iアダクトの形成増加は、反応時間の関数であることが明白であった(図9B)。IgGの反応速度は、同一条件下で測定したトリプシンよりも14.5倍速かった(それぞれ、172.7±14.2および11.9±0.6 AAU/分、図9Bデータの直線回帰より)。ホスホニル化されたタンパク質複合体の加水分解は同等であると推定されたので(図1の反応スキームを参照すること)、IgGの求核効果はトリプシンよりも優れていると結論づけることが可能である。
従来のセリンプロテアーゼの活性化された求核残基は、ホスホン酸ジエステルプローブ、例えばセリンプロテアーゼの触媒性Ser-His-Asp三つ組構造の水素結合によって活性化されたSer残基と共有結合的に反応する。タンパク質分解性およびエステル分解性のAbにおけるこのような求核体の存在は、変異誘発およびホスホネート共有結合の研究から推理されてきた(10〜12)。基質への求核攻撃は、ある種の酵素による触媒における速度制限段階である(24)。Abで報告されている触媒速度定数(kcat)の大きさは一般的に酵素よりも小さいので、一般的にAbの求核反応性には不完全性が存在すると思われる。本明細書で報告した研究は他の場合を示す。ハプテンホスホン酸ジエステルとの共有結合アダクトの形成速度から測定したところ、タンパク質分解活性が低いにもかかわらず、IgG調製物はトリプシンよりも強力な求核反応性を示した。ポリクローナルIgGおよび個々のscFvクローンの研究は、見かけ上共通の求核活性を示した。対照実験では、この反応性は熱変性で失われたので、求核体の活性化はタンパク質の天然構造に左右されるという予測と一致する。ホスホン酸ジエステルに結合する共有結合Abは、確立されたセリンプロテアーゼ反応試薬DFPによって阻害された。さらに、Abのタンパク質分解活性は、ホスホネートならびにDFPによって阻害されたので、ホスホネートと反応する求核体はセリンプロテアーゼ様の特性であることが裏付けられた。これらの研究によって、Abに本来備わっている特性としての求核反応性は、免疫応答中に生じた非共有型抗原結合力と独立して発現したことが示唆される。この結論は、タンパク質分解性Ab軽鎖の触媒的三つ組構造は生殖細胞系V遺伝子によってコードされるという以前の我々の報告と一致する(25)。
自己免疫、アレルギーおよび移植拒絶反応の結果として産生されるある種のAbの病原性効果の元となるのは、可変領域による特異的抗原認識である。例えば、重症筋無力症(文献1の概説)および血友病(文献2の概説)に見いだされるAbは、アセチルコリンレセプターおよび第VIII因子それぞれの重要なエピトープに結合し、立体的な妨害機構によってこれらのタンパク質の生物学的活性を妨害する。その他のAbは、Fc領域を利用して病原作用を媒介するが、Ab可変領域による抗原認識はこれらの効果を開始する刺激であり、例えば、赤血球抗原のAb認識は、自己免疫溶血性貧血および不適合輸血におけるFc領域による補体活性化を刺激する。同様に、肥満細胞表面のFcレセプターに結合したIgEによるアレルゲン認識は脱顆粒を刺激する。他の疾患では、Ab病原性の機構ははっきりしない。例えば、狼瘡(文献3の概説)および橋本甲状腺炎(文献4の概説)では核酸に対するAbは明白に疾患関連性があるが、他の免疫異常もまた、これらの疾患で明白で、Abの正確な機能的効果には議論の余地が残されている。最近、Ab病原性の基礎となる新規可変領域機構、すなわち抗原の触媒的切断が明らかになった。ポリペプチド(5〜8)および核酸(9)に対するAbなどの加水分解触媒は、永久的に抗原を不活性化する能力を備えていた。さらに、触媒は交代能力を備えており、すなわち、1個のAb分子が複数の抗原分子を加水分解することができるので、このようなAbは化学量論的な抗原認識に左右されるAbよりも能力が高い機能的効果を発揮できることが示唆される。
CAL。 ジフェニルN-(6-ビオチンアミドヘキサノイル)アミノ(4-アミジノフェニル)メタンホスホネート(1)は、PyBOP(Novabiochem、San Diego、CA)を使用してジフェニルアミノ(4-アミジノフェニル)メタンホスホネート(19、20)および6-ビオチンアミドヘキサン酸(Anaspec、San Jose、CA)から調製した。HPLC精製物質[保持時間20.76分、純度95%(220nm)、YMC ODS-AMカラム(4.6x250nm)、TFA 0.05% 水溶液(A)、アセトニトリルによるTFA 0.05%溶液(B)、90:10から20:80で45分(1.0ml/min)]は、ESI-MSによって特徴付けし[測定m/z 721.3(MH+、C36H45N6O6PSの算出MH+、721.3)]、DMFに溶かした10mM溶液として-70℃で保存した。活性化エステル2は、同前駆体アミンをジスベリン酸スクシニジル(Pierce、Rockford、IL)でアシル化することによって調製し、同様の方法で特徴付けした[測定m/z、635.3(MH+、C32H35N4O8Pの算出MH+、635.2)]。VIP-CAL(3)は以下の通りに合成した。N末端にビオチンを有するVIP配列は、4-メチルトリチル(22)をLys20の側鎖保護として使用すること以外は、標準的9-フルオレニルメトキシカルボニル方法(21)によってRinkアミドMBHA樹脂(0.72nmol/g、Novabiochem)で作製した。該ペプチド樹脂は、ジクロロメタンに溶かした1%TFA溶液(5分x10)で処理して、4-メチルトリチル基を除去し、Lys20の脱保護したアミノ基はN,N-ジイソプロピルエチルアミン0.1mMを含有する1-メチル-2-ピロリジノンに溶かした2でアシル化した。ペプチド樹脂は、TFA-エタンジチオール-チオアニソール-フェノール(90:1:1:8)で室温で2時間処理した。濾過によって樹脂を除去した後、ジエチルエーテルを溶液に添加して沈殿を生成し、遠心によって収集してジエチルエーテルで洗浄した。HPLC精製物質[保持時間50.25分、純度96%(220nm)、Vydac 214TP C4カラム(4.6x250mm)、A:B、90:10から60:40で60分(1.0ml/min)]は、ESI-MSによって特徴付けし[測定m/z 4071.4(MH+、C185H282NO49O49PS2の算出MH+ 4072.0)]、DMSOに溶かした10mM溶液として-70℃で保存した。
VIP-CAL。 VIP-CAL(化合物3、図14A)の設計における重要な点は、(a)セリンプロテアーゼで見いだされるような活性化求核体と選択的に反応することができる求電子ホスホン酸ジエステル基が含まれていること(16)、(b)塩基性アミノ酸のC末端側鎖のペプチド結合(Arg/Lys)を選択的に切断するタンパク質分解性IgGモデルクローンc23.5による認識を可能にするために、ホスホネートの近隣に正に荷電したアミジノ基が配置されていること(23、25)、および(c)VIP配列のLys20の側鎖にこれらの基を取り込むことである。ハプテンCAL 1は、ホスホン酸ジエステル基およびアミジノ基を含有するが、VIP配列を持たない。Lys20に共有結合性反応部分が位置するという根拠は、Lys20-Lys21ペプチド結合はモノクローナルAbクローンc23.5(23)およびVIPに対するAbを含有するヒトポリクローナルIgG調製物(24)によって切断される結合の1つであることが観察されたことである。プロテアーゼのペプチド阻害剤は、ペプチド主鎖内またはペプチド末端に位置する共有結合性反応基を常に含有する(例えば、26、27)。本研究では、異なるAb活性部位に含まれる求核体の共有結合距離内にホスホネート基が接近する機会を最大限にすることが目的である。この理由のため、(該ホスホネートをペプチド主鎖に含めると、この基の接近しやすさにより立体的な制限をもたらし得るのとは対照的に)該ホスホネート基は、いくつかのC-C結合での回転を可能にさせる柔軟なリンカーを使用してLys20の側鎖に配置された。
これらのデータから以下の結論が導かれる。(a)非共有結合および共有結合相互作用を機能的に協調させることによって、求核性抗VIP AbがVIP-CALと特異的かつ共有的なアダクトを形成することが可能になり、(b)VIP-CALは、いくつかのペプチド結合でVIPの切断に関与する反応それぞれを阻害し、様々な抗VIP触媒Abの汎用阻害剤としての能力が示唆される。VIP-CALがAb求核体を導くために非共有結合Abパラトープ-抗原エピトープ結合が重要であることは、以下の観察、VIP配列を持たないハプテンCALとの抗VIPモノクローナルAbの反応性は低いこと、関係のないアイソタイプの一致したAbとVIP-CALを有する血漿タンパク質との反応は限られていること、およびCAL部分のないVIPによってVIP-CALと抗VIP Abとの共有結合反応が阻害されることから明らかである。最近、その他のポリペプチド抗原(HIV gp120および上皮増殖因子レセプター)のCAL誘導体はまた、これらの抗原に特異的な特異的Abと共有結合アダクトを形成することが報告されているが、関係のないAbに特異的なAbでは反応がほんわずかな程度であることが明らかであった(31、32)。これらをもとにすると、これらの考察によって、抗原特異性に基づいて個々のAb亜集団が永久的阻害を行う過程が明らかにされる。

最初に、CAL反応は、抗原特異性に基づいて分泌型AbおよびAb産生細胞を不可逆的に不活性化する機会を提供する。この目的の実現可能性は、Lys20側鎖にホスホン酸ジエステル基を含有する合成VIP-CALによって血管作用性腸管ペプチド(VIP)に対するAbが特異的に共有結合によって遮断されることに関する観察によって支持される(1)。原則として、この取り組みによって、有害なAb作用に関係する疾患、例えば、自己免疫疾患、アレルギー疾患および移植片拒絶を治療するための基本的な技術を導くことができた。非触媒Abはまた、非共有結合抗原認識と協調して求核反応性を発現するので、CALの能力的に改良された効果は、タンパク質分解性Abとの反応に限定されない。以下の抗原、VIP(1)、完全長HIV gp120(2)およびペプチドgp120フラグメント、上皮増殖因子レセプター(細胞外ドメイン、3)および第VIII因子に特異的なAbは、適切なLaCALに共有結合することが以前の研究で観察された。
LaCALの2種類の標的、DNAおよびVIPに対する抗体をこの実施例で検討する。この考察では、どちらの抗体集団が触媒活性を有するかということは想定していない。実施例1で説明したように、適切な抗原エピトープをLaCALに組み入れるならば、このように調べた抗原特異的抗体は全て、LaCALにはるかに特異的な共有結合を示す。
求核性抗ペプチドAbの研究から得られた概念は、以下の理由、ある種のDNA切断酵素の求核中心によるタンパク質-DNA共有結合中間体の形成はよく知られた現象であること、ハプテンホスホネートプローブによって、Ab求核反応性がパラトープ特異性とは独立していることが確認されたこと、およびAb求核活性は、生殖細胞系によってコードされており、抗DNA Abが成熟するとき該活性は失われると予測する理由はないことによって、抗DNA Abに適用することができる。DNA-CALおよびCIP-CALの設計は、Ab求核体が非共有結合抗原結合部位から空間的に分離されている分割部位モデルをベースとしている。図1の反応スキームでは、Abに対する可逆的抗原結合を完全に模倣したCALのKiは低く(平衡解離定数)、k3は大きい(共有結合反応の強度)。これらは望ましいCALの動力学的特性である。
H-ホスホネート官能基の1個を4-ニトロフェニルホスホネートに変換する。次に、未反応のH-ホスホネートをヨウ素で酸化して、トリエステルを散在させた通常のリン酸ジエステル結合を得る。フルオロホスフェートCALを合成するために、該生成物を通例のDNA合成法(I酸化/NH3水溶液)によって、次に18-クラウン-6塩化チオニル(iii)の存在下でKFで処理する。これによって、リン酸ジエステルがフルオロリン酸ジエステルに変換される(フルオロホスフェート基1個/塩基5個になるようにKFを滴定する)。誘導体化は、アルカリ性加水分解後に4-ニトロフェノールを光度的に測定することによって、または元素分析によってフッ素を定量することによって評価する。オリゴ-CALの長さは、塩基30個に維持することができる。ビオチンは、合成中にオリゴ-CALの5'末端に結合させる。CAL部分を無作為に配置するが、その濃度はAb結合をもたらすために十分である(エピトープの最小限の長さはヌクレオチド5個である)。狼瘡の抗ssDNA Abは、ポリ(dT)鎖に選択性を示すが(5、24)、個々の配列への特異性は比較的に限定されていない。したがって、オリゴ-CAIの中心領域は、両側に7マーのランダム配列が隣接した16マー[(N)7-(T)16-(N)7]であることができ、合成中にA/T/G/C/の混合物を使用して隣接部に縮重が導入されている。
Ab細胞毒性のAb DNA-CALによる阻害。 狼瘡の抗DNA Abは、膜を貫通して、アポトーシスを引き起こす。DNA-CALで処理した抗DNA Abの細胞毒性作用は、尿細管細胞系(LLC-PK)およびMTT測定を使用して評価することができる(37)。該IgGの原料は、狼瘡患者、健常人被験者、8週齢のMRL/lprマウス、20週齢のMRL/lprマウスおよび同齢の対照MRL/++マウスである。さらに、少なくとも1本のモノクローナルL鎖を分析して、Ab不均一性による予期しない影響を排除する(例えば、KM38クローン)。IgGは、希釈した、または様々な濃度のDNA-CALおよび(CAL部分のない)対照DNAとインキュベートして、遊離のリガンドをAim1のように除去して、該混合物を細胞と混合して24ウェルプレートで培養した。細胞毒性は、24時間後に定量する。対照ウェルにはAbを入れない。ヨウ化プロピジウム(死細胞)およびHoechst33342(死細胞+生細胞)で染色した後、顕微鏡検査によって確認する。他の研究は、遊離CALを除去せずに、AbおよびCALで細胞を処理することによって行う。CAL自体は細胞毒性作用を生じず、CALおよび細胞が互いにAb結合を競合するとき有益な効果が得られることを示すために、これらが必要である。腎臓における狼瘡のAbの直接的な細胞毒性作用は一般的に、DNA含有抗原および/または交差反応性タンパク質抗原へのAb結合に関与し、その後、完全には定義されていない経路によって細胞/核膜を貫通すると考えられる。CALによる共有結合Abの機序は、永久に抗原結合機能を破壊するだろう。したがって、Fcエフェクター機能が関与しなければ、CAL結合Abは細胞毒性作用がないと予測される(培養物中に補体またはADCC競合細胞のもととなるものがなく、Fc関与の機会が最小限に抑えられていることに注意されたい)。

抗原特異的共有結合レセプターの機序は、LaCALの開発によって実現可能になった。共有結合BCRおよびTCRを占有することによって、これらの細胞をアポトーシスさせることによってB細胞およびT細胞の抗原特異的耐性(免疫寛容)を誘導する能力が維持された。必要であれば、LaCALの効果はこれらの化合物内に毒素を含めることによって補強され、前進的な副作用を引き起こすことなく標的細胞を選択的に殺す。シュードモナス内毒素およびリシンなどの毒素を様々なタンパク質に結合させることは、文献に詳細に記載されており、タンパク質化学結合または遺伝子融合技術によって実現することができる(43)。
第VIII因子(FVIII)に対するAbは、FVIIIIで治療したある種の血友病患者における出血増加の明らかな原因である(文献14の概説)。血友病患者が産生するAbの重要なFVIIIエピトープの1つは、A2ドメインの残基484〜508で構成される(15)。FVIIIは、血液凝固の固有経路の1成分で、第10因子から第10a因子への変換を触媒する第9a因子の補因子としての役割を担う。第VIII因子(FVIII-CAL)のCAL誘導体は、分泌型抗FVIII Abを選択的な共有結合阻害を実現する候補試薬で、抗FVIII産生B細胞の死を引き起こす。
(iii) ELISAプレートにコーティングしたFVIIIの結合。AbとCALとの不可逆的な複合体化は、凝固パラメータを生理学的濃度に回復することが予測される。ELISA実験を実施して、Ab-CAL複合体がもはやFVIIIと反応しないことを確認する。血液における効果を測定するために、CALはAb陽性の後天性血友病患者の分画していない血液と混合して、血液の凝固パラメータが生理学的濃度まで回復することを測定する。
この実施例では、ハプテンCALおよびLaCALによって標的化するために適したNuRの発見方法を説明する。
抗レトロウイルス治療の進歩にもかかわらず、HIV-1感染患者の多くは検出不可能なウイルス負荷を克服することができない。ほとんどの治療の失敗および抵抗の原因である耐性変異は既に、最新世代のペプチドをベースとした融合阻害剤に認められた[1]。逃れられる見込みが少ないHIVの治療の新しい試みが必要とされている。親和性の高い宿主細胞CD4レセプターに対するgp120結合がHIV感染を開始する。研究に適した種では早期の結果が見込まれたにもかかわらず、可溶性CD4治療では今まで一次単離物を中和することはできなかった(2、3)。これは、利用できるgp120部位全てをsCD4で飽和することができないことによると思われた(4)。この実施例では、gp120に対するsCD4模倣物の共有結合が従来のCD4に見られる化学量論的問題を補うHIV不活性化の新しい試みの概略を述べる。
この実施例では、生物学的に自己集合した共有結合模倣物をLaCAL技術を使用して調製する用途を例示する。弱い求電子体-求核体対が生物学的分子を必要とするホモ対合およびヘテロ対合反応に多大な寄与をすることが前提である。自己集合NuRの場合、1分子内の求核部位は、NuR複合体を順番に集合させる要因として他の分子の求電子部位と組み合わさることができる。したがって、人工的な求電子プローブをNuR内に配置すると、天然NuRが集合した構造に類似した、安定かつ正確に集合したNuR複合体を生成する能力が確保される。gp120モノマー間の共有結合相互反応は、天然のオリゴマーを模倣した安定なオリゴマーの形成に必要な求核体および求電子体の正確な組み合わせをもたらすので、該反応は順番通りに着々と進行することが期待される。
gp120-CALおよびビオチン化gp120-CALの調製は、最近の報告に記載した(2)。簡単に説明すると、gp120-CALは、組換えgp120およびアシル化剤Iから調製され(図29)、これは、ジフェニルアミノ(4-アミジノフェニル)メタン-ホスホネート[3]および市販のホモ二機能性架橋結合剤、ビス(スルホスクシニミジル)スベリン酸二ナトリウム塩から調製される。ビオチン化gp120-CAL(Bt-gp120-CAL)は、同様の方法でビオチン化gp120から調製され、これは組換えgp120および市販のビオチン化試薬、6-ビオチンアミドヘキサン酸N-ヒドロキシ-3-スルホスクシニミジルエステルから調製した。導入されたビオチンおよび2-(4'-ヒドロキシアゾベンゼン)安息香酸[4]およびフルオレスカミン[5]を使用して消費されたアミンの定量によって、ビオチン/gp120およびホスホネート/gp120モル比は、それぞれ、0.6〜0.8および5〜27であることが明らかになった。ホスホネート含量の多様さは、反応の再現性のなさを示しているのではないことに注意されたい。オリゴマー化過程におけるそれらの役割を研究するために、様々なホスホネート含量のBt-gp-120-CALを得るよう試みた。gp120に関係のないタンパク質、上皮増殖因子レセプター(exEGFR)の細胞外ドメインは、オリゴマー化反応の対照として使用した。exEGFR-CALは、gp120-CALと同様の方法を使用して調製した。
Abの広範な中和を誘導することは、HIV-1ワクチンの開発に対する重要な挑戦であった[7〜10]。実験的なワクチン接種によってgp120単量体に対して誘発されたAbのほとんどは、1次単離物のHIV-1感染を中和することができなかった[11〜14]。これらの抗体は一般的に、免疫学的に優勢なgp120のV3ループに対して特異的で、これはまた、高率で変異を受ける領域であり、Abによって制御することができないウイルス変種の出現を可能にする。モノマーgp120のCD4-結合部位(CD4bs)を認識する多くのAbはまた、HIV-1を中和することができない。これは、モノマーgp120のCDbsおよびウイルス表面上に発現する天然のgp120トリマーとの構造の違いが原因であった。オリゴマーgp120に対するAb結合は、Abの中和活性を予測すると一般に考えられている[15〜17]。したがって、天然gp120と構造的に同じ免疫原は、予防ワクチン研究並びに受動的免疫治療研究の両方のために必要である。したがって、その後は、天然のトリマー型のHIVエンベロープ(env)糖タンパク質に類似したアナログを開発することに労力が払われた。
以下の戦略は、オリゴマーgp120-CALワクチン候補を得るために有用である。
