Description
[1]下記の工程(a)及び(b)を含む、イヌiPS細胞の製造方法。
(a)イヌ体細胞と核初期化因子とを接触させる工程
(b)該細胞を、マイトジェン活性化タンパク質キナーゼキナーゼ阻害剤、アクチビンレセプター様キナーゼ阻害剤、グリコーゲン合成酵素キナーゼ阻害剤、L型カルシウムチャンネルアゴニスト及びDNAメチル化阻害剤からなる群より選択される一以上の物質並びに白血病抑制因子を含む培地で培養する工程

なお、該4因子は、イヌ体細胞に導入することによってイヌiPS細胞を製造することができるという特徴を有する限り、前記アミノ酸配列において一又は数(2〜5)個のアミノ酸が欠失、置換、挿入又は付加されたアミノ酸配列、あるいは該アミノ酸配列をコードする塩基配列を有する、極めて高い相同性を有するものであってよい。
イヌから分離した体細胞は、細胞の種類に応じてその培養に適した自体公知の培地で前培養することができる。そのような培地としては、例えば、約5〜20%の胎仔ウシ血清を含む最小必須培地(MEM)、ダルベッコ改変イーグル培地(DMEM)、RPMI1640培地、199培地、F12培地等が挙げられるが、それらに限定されない。
そのような方法としては、例えば、タンパク質導入試薬を用いる方法、タンパク質導入ドメイン(PTD)もしくは細胞透過性ペプチド(CPP)融合タンパク質を用いる方法、マイクロインジェクション法等が挙げられる。タンパク質導入試薬としては、カチオン性脂質をベースとしたBioPOTER Protein Delivery Reagent(Gene Therapy Systems)、Pro−JectTM Protein Transfection Reagent(PIERCE)及びProVectin(IMGENEX)、脂質をベースとしたProfect−1(Targeting Systems)、膜透過性ペプチドをベースとしたPenetrain Peptide(Q biogene)及びChariot Kit(Active Motif)、HVJエンベロープ(不活化センダイウイルス)を利用したGenomONE(石原産業)等が市販されている。導入はこれらの試薬に添付のプロトコルに従って行うことができるが、一般的な手順は以下の通りである。核初期化因子を適当な溶媒(例えば、PBS、HEPES等の緩衝液)に希釈し、導入試薬を加えて室温で5〜15分程度インキュベートして複合体を形成させ、これを無血清培地に交換した細胞に添加して37℃で1ないし数時間インキュベートする。その後培地を除去して血清含有培地に交換する。
PTDとしては、ショウジョウバエ由来のAntP、HIV由来のTAT、HSV由来のVP22等のタンパク質の細胞通過ドメインを用いたものが開発されている。PTD由来のCPPとしては、11R(Cell Stem Cell, 4: 381-384 (2009))や9R(Cell Stem Cell, 4: 472-476 (2009))等のポリアルギニンが挙げられる。核初期化因子のcDNAとPTDもしくはCPP配列とを組み込んだ融合タンパク質発現ベクターを作製して組換え発現させ、融合タンパク質を回収して導入に用いる。導入は、タンパク質導入試薬を添加しない以外は上記と同様にして行うことができる。
マイクロインジェクションは、先端径1μm程度のガラス針にタンパク質溶液を入れ、細胞に穿刺導入する方法であり、確実に細胞内にタンパク質を導入することができる。
また、核初期化因子として核酸を遺伝子導入する場合には、上記に示したように同時に導入された薬剤耐性遺伝子やレポーター遺伝子等のマーカー遺伝子の発現を指標とすることもできる。
さらに、イヌiPS細胞から分化させた機能細胞(例、肝細胞)は、対応する既存の細胞株よりも実際の生体内での該機能細胞の状態をより反映していると考えられるので、医薬候補化合物の薬効や毒性のin vitroスクリーニング等にも好適に用いることができる。また、初代機能細胞を調製するために実験用イヌの臓器や組織を切除する必要がなく、動物愛護の観点からも望ましい。
大脳、網膜、胃粘膜、皮膚、骨格筋、肺、精巣から抽出したRNAから、公共のデータベースに登録されたcDNA配列情報に基づいてプライマーを設計し、イヌOct3/4、Klf4、Sox2、c−Mycの各遺伝子をRT−PCRにより増幅した。Sox2についてはうまく増幅できなかったので、データベース上の予測イヌSox2配列とヒトSox2配列とを比較したところ、イヌSox2にはN末に165アミノ酸の余分な配列が入っていることがわかった。そこで、この余計な部分を除いて再度プライマーを設計し、RT−PCRを行ったところ、首尾よくイヌSox2遺伝子を増幅することができた。PCRでの増幅の際に使用した各プライマーの配列を表1に示す。
増幅した各遺伝子をpMXs−IRES−GFP(図1c)のマルチクローニングサイトにそれぞれ挿入し、各遺伝子を搭載した4つのレトロウイルスベクターを得た。
あらかじめ6×104個のPlat−GP細胞を10cmシャーレに播いておき、そこにトランスフェクション試薬(Fugene6 transfection reagent、Roche)を100μl入れ、室温で5分間静置した。その後、各レトロウイルスベクター及びpCMV−VSVGを3μg加え、さらに室温で15分静置してからPlat−GP細胞の培養液に加えた。培養液はDME培地(invitrogen)(最終濃度がそれぞれ0.5%抗生剤、10%FBSになるように添加)を使用し、37℃、5%CO2で培養した。トランスフェクションして24時間後に培地を交換し、さらに48、60、72時間後に培養上清を回収し、それを混合した液をウイルス含有液とした。ウイルス含有液を孔径0.45μmのミリポアフィルターにて濾過し、そこに最終濃度が4μg/mlになるようにポリブレンを添加し、ウイルス液とした。
イヌ線維芽細胞は妊娠30日目のビーグル犬(オリエンタルバイオ株式会社から購入)から胎仔を摘出し、胎仔組織をせん断、酵素処理することにより得た。イヌ線維芽細胞にレトロウイルス感染により初期化遺伝子を導入させ、感染後のGFPの発現レベルをフローサイトメトリーにて解析した。感染後5日で、線維芽細胞の約60%がGFP陽性を示し、遺伝子が高い効率で導入されることが確認された(図2、3)。
Oct3/4、Sox2、Klf4、c−Mycの4遺伝子をレトロウイルスにてイヌ線維芽細胞に導入した。3回目の遺伝子導入が終了した時点で培養液を交換した(霊長類ES細胞用培地+ヒトLIF(1000U/ml)+bFGF(6ng/ml)+PD0325901(0.5μM)+A−83−01(0.25μM)+CHIR99021(3μM)+バルプロ酸(1mM))。遺伝子導入後2日目で線維芽細胞の約60%がGFP陽性を示し、導入後5日目ではGFP陽性を示す小さなコロニーも確認した(図4c)。導入後20日目に単層構造様のコロニーの形成が確認された。確認されたコロニーの形態はイヌES細胞に類似していた。遺伝子導入後、約1ヶ月でコロニーが機械的剥離可能な大きさに達した。以降はマウス胎仔線維芽細胞由来のフィーダー細胞上で継代培養を行った。マウス胎仔由来線維芽細胞は妊娠13日の妊娠マウスの胎仔組織をせん断、酵素処理することで得た。マウス胎仔由来線維芽細胞はフィーダー細胞として使用するためマイトマイシンCで処理し、イヌiPS細胞継代前日に10cmディッシュへ1.5×106個の細胞を播種した。
ES細胞マーカーの発現を確認するために免疫染色解析を行った。イヌiPS細胞は、4%のパラホルムアルデヒドで30分間反応させ固定した。固定した細胞はPBSで洗浄し、2%のFBS,0.2%トリトンX−100/PBSで前処理した。一次抗体として抗Oct3/4抗体(1:100に希釈、SANTA CRUZ)、抗SSEA−1抗体(1:50に希釈、ミリポア)、抗SSEA−4抗体(1:50に希釈、ミリポア)、抗TRA−1−60抗体(1:50に希釈、ミリポア)、抗TRA−1−81抗体(1:50に希釈、ミリポア)を使用した。二次抗体としてはTRITC標識抗マウス抗体を使用した。核は、1μg/mlのDAPI(Roche)により染色した。
免疫染色によりイヌiPS細胞は、Oct3/4、SSEA−4、TRA−1−60及びTRA−1−81について陽性を示し、イヌES細胞と同じ発現パターンを示すことが明らかになった(図6−1、6−2)。
アルカリフォスファターゼ染色は、Leukocyte Alkaline Phosphatase Kit(Sigma)を使用して行った。対照としてマウスiPS細胞株であるiPS−MEF−Ng−20D−17(京都大学 山中伸弥教授より提供)を用いた。染色の結果、イヌiPS細胞はアルカリフォスファターゼ陽性を示し、染色の程度はマウスiPS細胞と同程度を示した(図7)。
イヌiPS細胞の三胚葉分化能を確認するために以下の実験を行った。外胚葉への分化誘導は、イヌiPS細胞をGME(Glasgow Minimum Essential)培地(最終濃度が10% KSR、0.1mM non−essential amino acids、1mM pyruvate、0.2mM 2−mercaptoethanol、100nM Vitamin B12、33μg/ml heparin、0.5% penicillin/streptomycinとなるよう添加)にて14日間培養を行った。中胚葉への分化誘導は、α−ME(α−minimum essential)培地(最終濃度が10%ウシ胎仔血清、5×10−5mol/l 2−mercaptoethanol、0.5% penicillin/streptomycin、100ng/ml VEGFになるように添加)にてIV型コラーゲンでコートしたディッシュ上で10日間培養を行った。内胚葉への分化誘導は、DME(Dulbecco’s modified Eagle’s)培地(最終濃度が20% KSR、1mM non−essential amino acids、0.55mM 2−mercaptoethanol、0.5% penicillin/streptomycin、4ng bFGF、1% glutamine、100ng activin Aになるように添加)にて14日間培養を行った。
イヌiPS細胞とイヌiPS細胞作製のもとのイヌ線維芽細胞とで、遺伝子の発現パターンに違いがあるかどうかを調べるために、DNAマイクロアレイ解析を行った。解析は、イヌiPS細胞又はイヌ線維芽細胞由来のtotal RNAを用いて、Cell, 131: 861-872 (2007)に記載の手法にて行った。その結果を図9に示す。イヌiPS細胞において特異的に発現している遺伝子を検出したところ、ES細胞特異的遺伝子であるSox2(600倍)、Sall4(1600倍)の過剰発現を認めた。以上の結果より、イヌiPS細胞とイヌiPS細胞作製のもとのイヌ線維芽細胞とでは遺伝子の発現パターンが異なることが確認された(図9)。
イヌiPS細胞にコルセミド溶液を加え、数時間培養した。トリプシン処理により細胞を単一分散後、塩化カリウム溶液にて細胞及び核を膨潤し、カルノア溶液で固定した。Qバンド処理後、染色体を顕微鏡下で撮影し、カリオグラムを作製した。
本発明においてイヌiPS細胞にて確認された染色体数は78であったことから、今回樹立されたイヌiPS細胞はイヌ体細胞に由来することが確認された。また染色体の異常も認められなかった(図10)。
複数の低分子化合物を核初期化因子とともに添加することで、iPS細胞コロニー作製を高率に誘導させることができた。Oct3/4、Sox2、Klf4、c−Mycの4遺伝子をレトロウイルスにてイヌ線維芽細胞に導入した。3回目の遺伝子導入が終了した時点から培養液を交換した(霊長類ES細胞用培地+ヒトLIF(1000U/ml)+bFGF(6ng/ml)+バルプロ酸(0.5mM)+Bay K8644(1μM)+BIX01294(0.5μM)+RG108(0.02μM))。
遺伝子導入後4日目で確認できたコロニーのほぼすべてが、イヌES細胞様のコロニーであった。この結果から、複数の低分子化合物を初期化因子とともに細胞に導入することで、効率よくイヌiPS細胞を得ることができたことを確認した(図11)。
Nagataの報告(Methods in molecular biology, 577:109-20 (2009))に基づき、バキュロウイルスを用いて組換えイヌLIFを作製した。即ち、ケタミン塩酸塩及びキシラジンによる麻酔下で成犬ビーグル犬から摘出した組織(皮膚、筋肉、精巣)からtotal RNAを抽出した。該RNAを鋳型として、NCBIデータベースに登録されたcDNA配列情報(XM_534732)に基づいて設計したプライマーを用いてRT−PCRを行い、N末端に6×Hisタグを導入したイヌLIF cDNAをNested PCRにて増幅した。PCR増幅の際に使用したプライマーの配列を表2に示す。最初のPCRはF01-primerとR-primerを使用してPCRを行い、さらに該PCR産物を鋳型にして2回目のPCRをF02-primerとR-primerを用いて行った。
iPS細胞の製造において使用するイヌ脂肪由来細胞は、以下の方法にて採取した。
ケタミン塩酸塩及びキシラジンによる麻酔下で、成犬ビーグル犬から大網組織の一部を切除し、得られた大網組織を消化液(30mg コラゲナーゼ8型(Sigma)、1.3mg/mlグルコース、0.4g ウシアルブミン分画5(Sigma)を含む10mlのハンクス緩衝塩類溶液)にて処理し、脂肪組織のみを採取した。採取した脂肪組織をウシ胎児血清含有D−MEM培地にて中和し、さらに赤血球溶解溶液にて処理した後、遠心して、細胞塊を得た。この細胞塊が脂肪由来細胞となる。
脂肪由来細胞を用いたイヌiPS細胞の樹立は以下の方法にて行った。
導入する遺伝子は、マウス由来Oct3/4、Sox2、Klf4、c−Mycの各遺伝子をそれぞれ搭載した組み換えレトロウイルス4種(Addgene, Inc.から入手)を使用した。3回目の遺伝子導入が終わった時点で培養液(霊長類ES細胞用培地(リプロセル)+イヌLIF(20ng/ml)+CHIR99021(3μM)+PD0325901(0.5μM))を交換し、培養を行った。遺伝子導入を開始して、14日後にイヌES細胞様のコロニーを確認した(図13)。その後、あらかじめマウス胎仔由来線維芽細胞を播種しておいたシャーレ上にコロニーを移し、培養を継続した。
線維芽細胞由来のイヌiPS細胞と比較すると、コロニーの出現数が格段に増加したことから、脂肪由来細胞及びイヌ由来のLIFを用いることにより、イヌiPS細胞樹立効率が高くなることが示唆された。
実施例12で得られたイヌiPS細胞からquick gene 800(Fuji film)を用いてRNAを抽出し、High Capacity RNA-to-cDNA Master Mix(ABI)にてcDNAを合成した。得られたcDNAを鋳型にした定量的リアルタイムPCRにより、該イヌiPS細胞におけるES細胞特異的遺伝子の発現を解析した。PCRにて使用したプライマーのうちイヌOct3/4、イヌSox2、イヌEras、イヌTertはApplied Biosystems社から購入し、その他のプライマー(イヌNanog、イヌRex1)は公共のデータベースに登録されたcDNA配列情報(それぞれXM_543828(NCBIデータベース)、ENSCAFG00000025203(Ensemblデータベース))に基づいて設計した。イヌNanog及びイヌRex1に対するプライマーの配列を表3に示す。
実施例12で得られたイヌiPS細胞について、実施例6と同じ方法を用いてアルカリフォスファターゼ染色を行った。その結果、得られたイヌiPS細胞は、陰性コントロールと比較し、高いアルカリフォスファターゼ陽性率を示した(図15)。
Claims (16)
- 下記の工程(a)及び(b)を含む、イヌiPS細胞の製造方法。
(a)イヌ体細胞と核初期化因子とを接触させる工程
(b)該細胞を、マイトジェン活性化タンパク質キナーゼキナーゼ阻害剤、アクチビンレセプター様キナーゼ阻害剤、グリコーゲン合成酵素キナーゼ阻害剤、L型カルシウムチャンネルアゴニスト及びDNAメチル化阻害剤からなる群より選択される一以上の物質並びに白血病抑制因子を含む培地で培養する工程 - 前記一以上の物質が、マイトジェン活性化タンパク質キナーゼキナーゼ阻害剤、アクチビンレセプター様キナーゼ阻害剤及びグリコーゲン合成酵素キナーゼ阻害剤、あるいは、L型カルシウムチャンネルアゴニスト及びDNAメチル化阻害剤である、請求項1に記載の製造方法。
- 核初期化因子が、Oct3/4、Sox2及びKlf4である、請求項1又は2に記載の製造方法。
- 核初期化因子が、Oct3/4、Sox2、Klf4及びc−Mycである、請求項1又は2に記載の製造方法。
- アクチビンレセプター様キナーゼ阻害剤が、アクチビンレセプター様キナーゼ5阻害剤である、請求項1〜4のいずれか1項に記載の製造方法。
- グリコーゲン合成酵素キナーゼ阻害剤が、グリコーゲン合成酵素キナーゼ3β阻害剤である、請求項1〜5のいずれか1項に記載の製造方法。
- 工程(b)の培地が、さらにヒストン脱アセチル化酵素阻害剤を含む、請求項1〜6のいずれか1項に記載の製造方法。
- ヒストン脱アセチル化酵素阻害剤が、バルプロ酸又はその塩である、請求項7に記載の製造方法。
- 工程(b)の培地が、さらに塩基性線維芽細胞成長因子を含む、請求項1〜8のいずれか1項に記載の製造方法。
- 核初期化因子と接触後48時間以内に工程(b)の培養を行う、請求項1〜9のいずれか1項に記載の製造方法。
- 核初期化因子と接触後3〜5週間経過後にフィーダー細胞上で培養を行う、請求項1〜10のいずれか1項に記載の製造方法。
- 分化多能性及び自己複製能を有する、体細胞由来のイヌ多能性幹細胞。
- 初期化遺伝子が遺伝的に安定に存在する、請求項12に記載の細胞。
- 初期化遺伝子が、Oct3/4、Sox2、Klf4及びc−Mycからなる群より選択される一以上の遺伝子である、請求項13に記載の細胞。
- 請求項1〜11のいずれか1項に記載の方法により製造される、イヌiPS細胞。
- 請求項12〜15のいずれか1項に記載の細胞から分化した、イヌ体細胞。
