Description
The present invention relates to a microarray substrate and a manufacturing method thereof, and more particularly to a microarray substrate including a carbonaceous film patterned on the substrate and a manufacturing method thereof.
In recent years, a technique for analyzing various liquids or gases with a single chip or device called a microchemical analysis system (μ-TAS) has attracted attention. Also in the field of biotechnology, the use of μ-TAS for analysis such as gene analysis has been studied. By analyzing a gene using μ-TAS, not only a plurality of gene expressions can be analyzed at once, but also an analysis can be performed with a very small amount of sample.
In order to analyze genes using μ-TAS, it is necessary to form a DNA microarray in which DNA is immobilized. A DNA microarray is a substrate on which tens of thousands to hundreds of thousands of partial DNA sequences are regularly arranged with high density. By using a DNA microarray, it is possible to analyze tens of thousands to hundreds of thousands of gene expressions at once.
In order to form a DNA microarray, a substrate capable of binding DNA regularly and firmly is required. In addition, the substrate is required to have a characteristic that the non-specific adsorption of DNA is small and the function of the bound DNA is not inhibited.
However, the conventional DNA microarray substrate using the DLC film has the following problems.
First, in the case where a chemical modification spot is merely formed on the surface of the DLC film, it is necessary to use a spotting technique for immobilizing DNA, which causes a problem that the operation becomes complicated. A method of removing unnecessary portions of the DLC film by reactive ion etching (RIE) is also known. However, since the selectivity is poor, not only the DLC film but also the lower substrate is damaged, or the surface of the DLC film is The problem arises that it becomes coarse and nonspecific adsorption occurs.
In addition, DNA is immobilized using an activated ester such as N-hydroxysuccinimide. Thereby, there is a merit that DNA is firmly fixed by a covalent bond, but it is impossible to recover the DNA without damaging the fixed DNA.
Such a problem occurs not only in DNA microarrays but also in cell microarrays or protein microarrays.
An object of the present invention is to solve the above-mentioned conventional problems, and to realize a microarray substrate that can be easily fixed and can be recovered without causing damage.
In order to achieve the above object, the present application has a microarray substrate including a target fixing region having a carbonaceous film and a plasma treatment layer formed on the surface of the carbonaceous film.
Specifically, the microarray substrate according to the present invention includes a base material and a plurality of target fixing regions regularly arranged on the base material, the target fixing region including a carbonaceous film and the carbonaceous material. A plasma treatment layer formed on the surface of the film, and the plasma treatment layer has an ionic functional group bonded to carbon constituting the carbonaceous film.
In the microarray substrate of the present invention, a plasma treatment layer having an ionic functional group is formed on the surface of the target fixing region. For this reason, a target of DNA, protein or cell can be fixed on the target fixing region by ionic bonding. Since the target is fixed by ionic bonding, the fixed target can be easily recovered by changing the ionic strength or pH of the atmosphere.
In the microarray substrate of the present invention, the ionic functional group may be at least one of an amino group and a carboxyl group.
In the microarray substrate of the present invention, the plurality of target fixing regions may be formed so that a voltage can be applied.
In this case, a bridge portion made of a carbonaceous film and connecting a plurality of target fixing regions to each other may be further provided.
In this case, it is preferable that the carbonaceous film in the bridge portion is thinner than the target fixing region.
In the microarray substrate of the present invention, the base material may be glass or silicon.
The method for manufacturing a microarray substrate according to the present invention includes a step (a) of forming a carbonaceous film on a substrate and a step of forming a plurality of target fixing regions by selectively removing the carbonaceous film ( b) and a step (c) of forming a plasma treatment layer having an ionic functional group on the surface of the carbonaceous film by irradiating the surface of the carbonaceous film with plasma. .
The method for producing a microarray substrate of the present invention includes a step of introducing an ionic functional group into the surface of a carbonaceous film by plasma irradiation. For this reason, an ionic functional group can be selectively introduced on the surface of the target fixing region. Therefore, it is possible to realize a microarray substrate in which the target can be easily fixed and collected.
In the method for producing a microarray substrate of the present invention, in step (c), plasma of a basic nitrogen-containing compound may be irradiated.
In the method for producing a microarray substrate of the present invention, the base material is made of silicon or silicon oxide, and in the step (b), etching is performed using plasma of a mixed gas containing fluorocarbon and oxygen and containing more oxygen than fluorocarbon. Just do it.
In the method for manufacturing a microarray substrate of the present invention, in the step (b), etching may be performed so as to leave a bridge portion connecting a plurality of target fixing regions.
In the method for manufacturing a microarray substrate of the present invention, the step (b) includes a step (b1) of removing a region excluding the target fixing region and the bridge portion in the carbonaceous film by etching, and a bridge portion after the step (b1). And a step (b2) in which the height of the bridge portion is made lower than that of the target fixing region.
According to the microarray substrate and the method for manufacturing the same according to the present invention, it is possible to realize a microarray substrate that allows DNA to be fixed easily and recovered without damage.
An embodiment of the present invention will be described with reference to the drawings. 1A and 1B are microarray substrates according to an embodiment, FIG. 1A shows a planar configuration, and FIG. 1B shows a cross-sectional configuration taken along line Ib-Ib in FIG. Yes. As shown in FIG. 1, a plurality of
The
The
Since the carbonaceous film is smooth and inactive, it is usually impossible to firmly fix targets such as DNA, protein and cells on the surface. For this reason, the target is fixed by introducing an activated ester such as N-hydroxysuccinimide. However, in this case, there is a problem that the target cannot be recovered because the functional group of the target reacts with the active ester to form a covalent bond.
In the microarray substrate of this embodiment, ionic functional groups are introduced on the surface of the
Below, the manufacturing method of the board | substrate for microarrays of this embodiment is demonstrated. First, as shown in FIG. 2A, a
First, the
Next, as shown in FIG. 2B, the
A method using RIE or the like is known for etching a carbonaceous film. However, when the carbonaceous film is etched by RIE, the surface of the carbonaceous film is damaged and the surface roughness increases. Moreover, there is a possibility that problems such as damage to the base material may occur. For this reason, it is preferable to use the following method of selectively etching a carbonaceous film.
First, a mask is formed on the base material on which the
Next, the
Oxygen may be used as the etching gas. Alternatively, a mixed gas of oxygen, fluorocarbon, and inert gas may be used. FIG. 3 shows the difference in etching rate between silicon oxide (SiO 2 ) and silicon (Si) and DLC, which is a carbonaceous film. The etching rate of SiO 2 and Si increases as the flow rate of carbon tetrafluoride (CF 4 ) increases, and is hardly etched with oxygen alone. On the other hand, the etching rate of DLC increases as the flow rate of oxygen increases. For this reason, it is possible to selectively etch the
Table 1 shows the measurement results of the surface roughness of the carbonaceous film surface after plasma irradiation. When the oxygen plasma is irradiated, the surface roughness becomes very large as compared with the case where the other plasma is irradiated. Therefore, in order to take advantage of the smoothness of the carbonaceous film, the etching rate is slightly reduced, but a mixed gas of oxygen and an inert gas such as argon or a mixture of oxygen and an inert gas and a fluorocarbon such as CF 4. It is preferable to use a gas.
When etching is performed using only oxygen, the surface roughness of the side surface portion of the spot increases, and the surface area increases. For this reason, a possibility that a functional group is also introduced into the side surface portion of the spot during the subsequent plasma treatment increases. By using a mixed gas of oxygen and an inert gas or a mixed gas of oxygen, an inert gas, and a fluorocarbon, an effect of reducing the introduction of functional groups into the side surface portion of the spot can be obtained.
In the case of adding a fluorocarbon, it is preferable that the ratio of oxygen is higher than that of the fluorocarbon in order to ensure etching selectivity with the substrate.
Next, as shown in FIG. 2C, the
First, the
As the reaction gas, ammonia (NH 3 ) or a mixed gas containing ammonia may be used. Thus, an amino group can be introduced by using ammonia. In addition, a carboxyl group can be introduced by using oxygen or a mixed gas containing oxygen. Even when an inert gas is used in place of oxygen, a carboxyl group is introduced by reaction with oxygen or moisture in the air. By combining ammonia or the like with oxygen or the like, both a carboxyl group and an amino group can be introduced. In addition, by setting the ultimate vacuum in the chamber before plasma irradiation to be low, not only amino groups but also carboxyl groups are introduced when ammonia is irradiated.
FIG. 4 shows the relationship between the surface potential and the abundance ratio of nitrogen to carboxyl groups (N / C (= O) O) in the carbonaceous film after plasma irradiation obtained by X-ray photoelectron spectroscopy (XPS). Yes.
For the XPS measurement, a photoelectron spectrometer JPS-9010MC manufactured by JEOL Ltd. was used. Al was used as the X-ray source, and X-rays were generated under the conditions of an acceleration voltage of 12.5 kV and an emission current of 17.5 mA. Measurement was performed on an area of 5 mm in diameter arbitrarily selected from the sample. Further, the composition up to a depth of about 5 nm is measured by making X-rays incident on the sample perpendicularly and setting the detection angle to 0 degree.
The binding energy measurement region was 274 eV to 294 eV, 389 eV to 409 eV, and 522 eV to 542 eV, and peaks of C1s, N1s, and O1s were obtained, respectively. By comparing the area ratios of the obtained peaks, the abundance ratio of oxygen to carbon O / C and the abundance ratio of nitrogen to carbon N / C were determined. The C1s peak was divided into a C—C component, a C—O component, a C═O component, and a C (═O) O component by curve fitting. The abundance ratio C (= O) O / C of the carboxyl group component C (= O) O to the total carbon was determined by determining the area ratio of the C (= O) O component to the C1s peak.
The state of nitrogen is not clear. However, it is considered that functional groups containing nitrogen (nitrogenous functional groups) such as amino groups, amide groups, and amine groups are formed. Although detailed analysis of the nitrogenous functional group is difficult, the N1s peak appeared at 398.9 eV in the XPS measurement. This is deviated from the binding energy (400 ± 1 eV) of N1s of amine and amide, and it is considered that at least a part of the nitrogenous functional groups is an amino group in this sample.
The surface potential was measured as follows. For the measurement, a zeta potential / particle size measurement system ELS-Z manufactured by Otsuka Electronics Co., Ltd. was used. After irradiating the carbonaceous film formed on the surface of the glass substrate with a predetermined plasma, the carbonaceous film was brought into close contact with the flat plate sample cell, and monitoring particles were injected into the cell. The monitoring particles used were those manufactured by Otsuka Electronics Co., Ltd. dispersed in a 10 mM sodium chloride (NaCl) solution. The monitor particles were electrophoresed at each level in the cell depth direction, and the apparent velocity distribution inside the cell was measured. Electrophoresis was performed under the conditions of an average electric field of 17.33 V / cm and an average current of 1.02 mA. By analyzing the apparent velocity distribution obtained based on the Mori-Okamoto equation, the surface potential of the carbonaceous film surface was obtained. The plate sample cell was used after being coated with polyacrylamide in order to suppress the influence of the charge on the cell surface.
As shown in FIG. 4, when the plasma of ammonia is irradiated in a state where the ultimate vacuum of the chamber is high (5 × 10 −3 Pa), the value of N / C (═O) O is increased and the surface potential is also increased. A positive value was shown. On the other hand, when oxygen or argon plasma was irradiated in a state where the ultimate vacuum was low (2 Pa), the value of N / C (= O) O was small, and the surface potential showed a large negative value. When the plasma of ammonia is irradiated after oxygen or argon, an intermediate value is shown.
Thus, by changing the plasma irradiation conditions when forming the
The strength of ionic bonds varies greatly depending on the concentration of ions in the solution. For this reason, the target fixed on the surface of the
The carbonaceous film is semi-insulating, and a weak current can be passed by adjusting the resistance value. Therefore, as shown in FIG. 5, if the
The resistance value of the carbonaceous film can be changed by the ratio of the SP2 bond and the SP3 bond. It can also be changed by introducing a functional group on the surface.
The height of the
FIG. 5 shows an example in which a plasma treatment layer is also formed on the
In the present embodiment, an example in which glass is used as the
A basic nitrogen-containing compound other than ammonia may be used for the plasma irradiation for forming the plasma treatment layer. Examples of basic nitrogen-containing compounds include organic amines represented by the general formula NR 1 R 2 R 3 (wherein R 1 , R 2 and R 3 are hydrogen, —CH 3 , —C 2 H 5 , —C 3 H 7 or —C 4 H 8 , and R 1 , R 2 and R 3 may be the same or different from each other.) Or benzylamine and its secondary and tertiary amines. However, ammonia is preferable because of cost and ease of handling.
In addition, although an example in which the plasma treatment layer is formed after the target fixing region is formed by etching has been shown, etching may be performed after the plasma treatment layer is formed.
The microarray substrate and the manufacturing method thereof according to the present invention can realize a microarray substrate that can be easily fixed without being damaged and can be recovered without damaging the substrate, and a carbonaceous film patterned on the substrate can be formed.
