Abstract
Description
本発明は、匹敵し得る量のドコサヘキサエン酸を産生する、ゴア(Goa)の入江から花粉釣り餌法(Pollen baiting method)によって単離された、新規な未報告のスラウストキトリウム(Thraustochytrid)菌株‐SC1のスクリーニングに関する。著しい量のオメガ‐3脂肪酸の産生を示す、単離生物の脂肪酸プロファイルが開示されている。また、それらの分子系統樹を示す単離生物の18S rRNAシークエンスも明らかにされている。
ここ10年間、多価不飽和脂肪酸(PUFAs)の栄養上の重要性に対する関心は顕著に増加している。多価不飽和脂肪酸は、2または3以上の二重結合を含む長鎖の脂肪酸である。それらに対する興味は、その治療的、食料および栄養上の適用における潜在力に起因する。それらは、選択された種子植物およびいくつかの海産の供給源から商業的に産生される。しかし、現在の供給源からの精製されたPUFAsの産生は、拡大した市場を充足させるのには不十分になる(Gill and Valivety 1977)。予測される需要の拡大に適合し、魚油の欠点を回避するために、PUFAsの代替的産生プロセスが現在開発されている。

を産生する能力を持つスラウストキトリウム菌株のカタログが作られた。
1.PUFA産生株のさらなる単離、スクリーニングおよび維持:DHAの豊富な油脂の商業的産生のための潜在能力を持つ数種の株は既に単離されている。しかしながらより高いPUFA産生量および/またはより魅力的なプロファイルのPUFAを産生するスラウストキトリウムの単離および最適化が可能である。それらの単離物と複合物に対する要求は増加する。
Huangらは、新規に単離されたドコサヘキサエン酸産生スラウストキトリウムの、多価不飽和脂肪酸プロファイルと18S rRNA遺伝子の比較解析に基づく分類法について記述した。特徴的な量のDHAを産生する7種の海洋微生物の菌株が、日本とフィジーの沿岸部で採取された海水からスクリーニングされた。それらはDHAに加えて中間脂肪酸もその呼吸の途中で蓄積する。それらの単離菌株は、18S rRNA遺伝子中のそれら固有の挿入配列によって、新規のスラウストキトリウムであることが証明された。単離菌株と典型的なスラウストキトリウムの18S rRNA遺伝子の分子解析によって作られた系統発生樹は、同一のPUFAプロファイルを持つ菌株は単一系統群を形成することを示している。この結果は、C20〜22のPUFAプロファイルがスラウストキトリウムの分類のための効果的な特徴として感知可能であることを示唆している。

スラウストキトリウム菌株はマングローブと呼ばれる海洋維管束植物の葉や樹状突起の死骸に生息する。これらの植物の葉の死骸はゴアの入江から採取し、花粉釣り餌法によって単離した。葉は、抗生物質(ストレプトマイシンとペニシリン)および少量のマツ花粉粒を入れた滅菌した海水で3日間培養した。この花粉を、新たな抗生物質入りの滅菌した海水に再接種した。3日間のインキュベーションの後、花粉を顕微鏡下で観察した。10種の異なるスラウストキトリウム菌株が発見され、それらは抗生物質とともにMV(岩塩‐3.4%、ペプトン‐0.15%、酵母エキス‐0.1%、グルコース‐2%、KH2PO4‐0.025%、pH7)アガープレートに5日間プレート培養した。スラウストキトリウムはプレートの上にコロニー/層を形成する。これらのスラウストキトリウムを新たな抗生物質入りMV培地へと接種した。3日間の増殖の後、純粋培養物が得られた。スラウストキトリウムは5日間培養され、総脂肪酸含有量を決定するために細胞をGC‐MS分析にかけた。
総脂質を、2:1体積/体積のクロロホルム:メタノールでのフォルク抽出法(Folch method of extracting)を用いて、培養物から抽出した。総脂質ならびに各脂肪酸の定量のために、内部標準(ヘプタデカン酸またはペンタデカン酸)を抽出前に加えた。脂肪酸を抽出し、エステル化して、Agilent6890NガスクロマトグラフィをAgilent5973マススペクトロメーターならびにHP6850シリーズガスクロマトグラフィにFID探知機を装着したものに連結し、GC‐MS分析を行った。GC‐MS検出はキャピラリーカラム(30m、HP‐5ms、WCOT、内径0.25mm、フィルム厚0.25μm、150℃で2分、300℃まで6℃/分、300℃で20分加熱、ヘリウムキャリアガス流1.0ml/分、分離比50:1)を用いてEI法により70eV(m/z50〜550、発射口部(source)230℃、四重極部(quadruple)150℃)で行った。GC‐FIDには、キャピラリーカラムDB‐23(30m、WCOT、内径0.25mm、フィルム厚0.5μm)を用いた。加熱炉の温度は160℃で2分間、180℃まで6℃/分、180℃で2分間、230℃まで4℃/分、230℃で10分間となるように設定し、N2キャリアガス流1.5ml/分、導入部温度が230℃ならびに検出部温度が250℃、分離比50:1で行った。
高DHA含有量を持つSC‐1、AVE5およびAVE7の1.7kb18S rRNA配列を遺伝子からプライマーを用いて保護領域まで増幅した。増幅された断片を配列分析した。配列は、配列表に表されている。上述のように3種の菌株−SC‐1、AVE5、AVE7はDHAの産生者であると見受けられるので、これらの菌株はMicrobial Type Culture Collection and Gene Bank(MTCC),Institute of Microbial Technology, Chandigarh, Indiaに寄託した。

- スラウストキトリウム属を含む群から選択される、ドコサヘキサエン酸(DHA)および他のオメガ‐3脂肪酸/脂肪酸中間体を産生する微生物。
- 配列番号1で示される18S RNA配列を含む、請求項1に記載のオメガ‐3脂肪酸産生微生物。
- 30%を超えるドコサヘキサエン酸を産生する、請求項2に記載のオメガ‐3脂肪酸産生微生物。
- 25%を超えるドコサヘキサエン酸を産生する、請求項2に記載のオメガ‐3脂肪酸産生微生物。
- 20%を超えるドコサヘキサエン酸を産生する、請求項2に記載のオメガ‐3脂肪酸産生微生物。
- 15%を超えるドコサヘキサエン酸を産生する、請求項2に記載のオメガ‐3脂肪酸産生微生物。
- 10%を超えるドコサヘキサエン酸を産生する、請求項2に記載のオメガ‐3脂肪酸産生微生物。
