フォーカスを試薬基板から5μm以内の培地に合わせてフェムト秒レーザー(チタンサファイアパルスレーザー:800 nm、150 fsパルス、1 mJ/パルス)の単パルスを照射する。パルス照射前後の心筋細胞の拍動状態を30Hzのフレームレートで画像を取得し、標的細胞塊の拍動状況をフレーム毎の輝度差分を測定することで解析する。その結果を図13と図14に示す。図13は試薬層43にアセチルコリンを含む試薬基板42を用いた結果であり、図14は試薬層43にエピネフリンを含む試薬基板42を用いた結果である。それぞれの図において、横軸は時間、縦軸は輝度差分に相当する任意値である。アセチルコリンを添加する前はほぼ一定の間隔で拍動しているが、レーザー照射(T2)後一定時間を置いた後は拍動が停止し、約20秒後に拍動が再開することがわかる。拍動再開後はレーザー照射前に比べて拍動間隔のバラつきが大きくなることがわかる。エピネフリンを添加するケースでは、レーザー照射(T2)後に拍動数が上昇することがわかる。図15は試薬層43にエピネフリンもアセチルコリンも含まない試薬基板42を用いた例で、レーザー照射(T2)しても心筋拍動に変化が無いことがわかる。
